Abstract

The UCNA experiment was designed to measure the neutron $\beta$-asymmetry parameter $A_0$ using polarized ultracold neutrons (UCN). UCN produced via downscattering in solid deuterium were polarized via transport through a 7 T magnetic field, and then directed to a 1 T solenoidal electron spectrometer, where the decay electrons were detected in electron detector packages located on the two ends of the spectrometer. A value for $A_0$ was then extracted from the asymmetry in the numbers of counts in the two detector packages. We summarize all of the results from the UCNA experiment, obtained during run periods in 2007, 2008--2009, 2010, and 2011--2013, which ultimately culminated in a 0.67\% precision result for $A_0$., Comment: 6 pages, 7 figures, to appear in proceedings of the International Workshop on Particle Physics at Neutron Sources 2018

Abstract

The evaluation of the performance of adiabatic annealers is hindered by lack of efficient algorithms for simulating their behaviour. We exploit the analyticity of the standard model for the adiabatic quantum process to develop an efficient recursive method for its numerical simulation in case of both unitary and non-unitary evolution. Abstract

A classic T-cell phenotype in systemic lupus erythematosus (SLE) is the downregulation and replacement of the CD3ζ chain that alters T-cell receptor signaling. However, genetic associations with SLE in the human CD247 locus that encodes CD3ζ are not well established and require replication in independent cohorts. Our aim was therefore to examine, localize and validate CD247-SLE association in a large multiethnic population. We typed 44 contiguous CD247 single-nucleotide polymorphisms (SNPs) in 8922 SLE patients and 8077 controls from four ethnically distinct populations. The strongest associations were found in the Asian population (11 SNPs in intron 1, 4.99 × 10(-4) < P < 4.15 × 10(-2)), where we further identified a five-marker haplotype (rs12141731-rs2949655-rs16859085-rs12144621-rs858554; G-G-A-G-A; P(hap) = 2.12 × 10(-5)) that exceeded the most associated single SNP rs858554 (minor allele frequency in controls = 13%; P = 4.99 × 10(-4), odds ratio = 1.32) in significance. Imputation and subsequent association analysis showed evidence of association (P < 0.05) at 27 additional SNPs within intron 1. Cross-ethnic meta-analysis, assuming an additive genetic model adjusted for population proportions, showed five SNPs with significant P-values (1.40 × 10(-3) < P< 3.97 × 10(-2)), with one (rs704848) remaining significant after Bonferroni correction (P(meta) = 2.66 × 10(-2)). Our study independently confirms and extends the association of SLE with CD247, which is shared by various autoimmune disorders and supports a common T-cell-mediated mechanism.

Abstract

This paper details the solar neutrino analysis of the 385.17-day Phase-III data set acquired by the Sudbury Neutrino Observatory (SNO). An array of $^3$He proportional counters was installed in the heavy-water target to measure precisely the rate of neutrino-deuteron neutral-current interactions. This technique to determine the total active $^8$B solar neutrino flux was largely independent of the methods employed in previous phases. The total flux of active neutrinos was measured to be $5.54^{+0.33}_{-0.31}(stat.)^{+0.36}_{-0.34}(syst.)\times 10^{6}$ cm$^{-2}$ s$^{-1}$, consistent with previous measurements and standard solar models. A global analysis of solar and reactor neutrino mixing parameters yielded the best-fit values of $\Delta m^2 = 7.59^{+0.19}_{-0.21}\times 10^{-5}{eV}^2$ and $\theta = 34.4^{+1.3}_{-1.2}$ degrees., Comment: 104 pages, 26 figures, 20 tables, submitted to Phys. Abstract

An array of Neutral-Current Detectors (NCDs) has been built in order to make a unique measurement of the total active flux of solar neutrinos in the Sudbury Neutrino Observatory (SNO). Data in the third phase of the SNO experiment were collected between November 2004 and November 2006, after the NCD array was added to improve the neutral-current sensitivity of the SNO detector. This array consisted of 36 strings of proportional counters filled with a mixture of $^3$He and CF$_4$ gas capable of detecting the neutrons liberated by the neutrino-deuteron neutral current reaction in the D$_2$O, and four strings filled with a mixture of $^4$He and CF$_4$ gas for background measurements. The proportional counter diameter is 5 cm. The total deployed array length was 398 m. The SNO NCD array is the lowest-radioactivity large array of proportional counters ever produced. 11. Investigation of the role of tau gene transcription in neurodegeneration

Abstract

Although the tau gene (MAPT) is not mutated in the majority of tauopathies, there are pathological disturbances in tau-isoform homeostasis. Investigation into MAPT linkage disequilibrium and haplotype structure, and the common variation of MAPT associated with increased risk of progressive supranuclear palsy (PSP) and corticobasal degeneration (CBD) identified a possible basis for the disturbances in tau-isoform homeostasis. Of multiple haplotypes in the H1 clade, the H1c sub-haplotype drives the association with PSP and CBD, suggesting that it carries the pathogenic variation leading to increased risk of these largely sporadic disorders. The aim of this project was to investigate the regulation of the transcription of MAPT by first identifying transcription factors (TFs) and TF complexes that bind to the MAPT core promoter and its conserved regulatory domains. The specific aim was to determine if differential binding of TFs and TF complexes to the allelic variants of the SNPs in the MAPT promoter region forms the basis of the allele-specific differences in MAPT transcription and splicing that we have observed in vitro and in vivo. Results from electrophoretic mobility shift assays, pull-down experiments and mass spectrometry had shown potentially novel proteins binding to the disease-associated SNP under investigation. Bioinformatic analyses helped to stratify these proteins according to possible relevance. While immunoblotting alongside co-transfection of siRNA and luciferase promoter construct experiments had shown initial evidence of a complicated relationship between cis and trans factors within the tau promoter region. These findings were novel and they provide new insight into the regulation of one of the most important genes in neurodegeneration. The identification of MAPT transcriptional machinery would provide further insight into the role of MAPT in neurodegeneration and the basis for therapeutic intervention.

Abstract

Ultra-cold neutrons (UCN), neutrons with energies low enough to be confined by the Fermi potential in material bottles, are playing an increasing role in measurements of fundamental properties of the neutron. The ability to manipulate UCN with material guides and bottles, magnetic fields, and gravity can lead to experiments with lower systematic errors than have been obtained in experiments with cold neutron beams. The UCN densities provided by existing reactor sources limit these experiments. The promise of much higher densities from solid deuterium sources has led to proposed facilities coupled to both reactor and spallation neutron sources. In this paper we report on the performance of a prototype spallation neutron-driven solid deuterium source. This source produced bottled UCN densities of 145 +/-7 UCN/cm3, about three times greater than the largest bottled UCN densities previously reported. These results indicate that a production UCN source with substantially higher densities should be possible.

Abstract

We present the first measurements of the survival time of ultracold neutrons (UCNs) in solid deuterium SD2. This critical parameter provides a fundamental limitation to the effectiveness of superthermal UCN sources that utilize solid ortho-deuterium as the source material. Superthermal UCN sources offer orders of magnitude improvement in the available densities of UCNs, and are of great importance to fundamental particle-physics experiments such as searches for a static electric dipole moment and lifetime measurements of the free neutron. 15. Changes in the ‘electromagnetic’ summer seasons as inferred from Schumann resonance frequency records at Nagycenk, Hungary, during 1994-2016

Abstract

A change in the length of meteorological summer seasons has been pointed out recently in connection with global warming: longer and hotter summers in the North Hemisphere (Wang et al., 2021). It has been demonstrated that the diurnal variation of Schumann resonance peak frequencies (SRPFs) in the vertical electric component, mainly driven by the changing distance of dominant lightning sources relative to the observer, is characteristic to the season (Sátori, 1996). It is therefore possible to define ‘electromagnetic’ (EM) seasons based on SRPF variations. SRPFs measured at Nagycenk, Hungary were used to study the variation of the length, start, and end of the North and South Hemisphere (NH/SH) summer seasons, in an electromagnetic sense, in the time period 1994-2016. In each year, diurnal SRPF variations were correlated with the all-year average diurnal curve of the most characteristic month of summer: July (NH) and January (SH). When the trend in the correlation coefficient approached (underwent) the limit 0.85, the start (end) of the corresponding season was registered.In agreement with earlier findings, our results show that the SH summer (140-170 days) is longer than the NH summer (100-140 days). However, in contrast to the findings based on temperature variation-associated thresholds for determining the start and end of the seasons, the EM NH summer did not exhibit a clear elongation in the examined time period. On the other hand, the SH summer season was lengthened by 10-15 days mostly due to the delay of its retreat.

Abstract

Monitoring water level changes is necessary to manage, conserve and restore natural, and anthropogenic lake systems. However, the in-situ monitoring of lake systems is unfeasible due to limitations of costs and access. Furthermore, current remote sensing methods are restricted to large lakes and low spatial resolutions. We develop a novel approach using subsequential pixel-wise observations of the Sentinel-1B sensor based on interferometric synthetic aperture radar to detect water level changes in small lakes. We used 24 small ungauged lakes of the Cajas Massif lake system in Ecuador for development and validation. We found Differential Interferometric Synthetic Aperture Radar (DInSAR)-derived water level changes across lakes to be consistent with precipitation, capturing the peak of the wet seasons. Furthermore, accumulated water level changes could be explained by differences in lake area among lakes. Although with limitations, this study shows the underutilized potential of DInSAR to understand water level changes in small lakes with current radar data availability.

IntroductionRecent outbreaks of Zika virus (ZIKV) in South and Central America have highlighted significant neurological side effects. Concurrence with the inflammatory neuropathy Guillain-Barré syndrome (GBS) is observed in 1:4000 ZIKV cases. Whether the neurological symptoms of ZIKV infection are a consequence of autoimmunity or direct neurotoxicity is unclear.MethodsWe employed rat dorsal root ganglion (DRG) neurons, Schwann cells (SCs), and human stem cell-derived sensory neurons myelinated with rat SCs as cellular models to screen for IgG and IgM autoantibodies reactive to peripheral nerve in sera of ZIKV patients with and without GBS. In this study, 52 ZIKV-GBS patients were compared with 134 ZIKV-infected patients, and 91 non-ZIKV controls. Positive sera were taken forward for target identification by immunoprecipitation and mass spectrometry, and candidate antigens validated by ELISA and cell-based assays. Autoantibody reactions against glycolipid antigens were also screened on an array.ResultsOverall, IgG antibody reactivity to rat SCs (6.5%) and myelinated co-cultures (9.6%) were significantly higher, albeit infrequently, in the ZIKV-GBS group compared to all controls. IgM antibody immunoreactivity to DRGs (32.3%) and SCs (19.4%) was more frequently observed in the ZIKV-GBS group compared to other controls, while IgM reactivity to co-cultures was as common in ZIKV and non-ZIKV sera. Strong axonal-binding ZIKV-GBS serum IgG antibodies from one patient were confirmed to react with neurofascin-155 and 186. Serum from a ZIKV non-GBS patient displayed strong myelin-binding and anti-lipid antigen reaction characteristics. There was no significant association of ZIKV-GBS with any anti-glycolipid antibodies.ConclusionAutoantibodies in ZIKV associated GBS patients’ sera target heterogeneous peripheral nerve antigens suggesting heterogeneity of the humoral immune response despite a common prodromal infection.

Abstract

Circulating autoantibodies (auto-Abs) neutralizing high concentrations (10 ng/ml; in plasma diluted 1:10) of IFN-alpha and/or IFN-omega are found in about 10% of patients with critical COVID-19 (coronavirus disease 2019) pneumonia but not in individuals with asymptomatic infections. We detect auto-Abs neutralizing 100-fold lower, more physiological, concentrations of IFN-alpha and/or IFN-omega (100 pg/ml; in 1:10 dilutions of plasma) in 13.6% of 3595 patients with critical COVID-19, including 21% of 374 patients >80 years, and 6.5% of 522 patients with severe COVID-19. These antibodies are also detected in 18% of the 1124 deceased patients (aged 20 days to 99 years; mean: 70 years). Moreover, another 1.3% of patients with critical COVID-19 and 0.9% of the deceased patients have auto-Abs neutralizing high concentrations of IFN-beta. We also show, in a sample of 34,159 uninfected individuals from the general population, that auto-Abs neutralizing high concentrations of IFN-alpha and/or IFN-omega are present in 0.18% of individuals between 18 and 69 years, 1.1% between 70 and 79 years, and 3.4% >80 years. Moreover, the proportion of individuals carrying auto-Abs neutralizing lower concentrations is greater in a subsample of 10,778 uninfected individuals: 1% of individuals 80 years. By contrast, auto-Abs neutralizing IFN-beta do not become more frequent with age. Auto-Abs neutralizing type I IFNs predate SARS-CoV-2 infection and sharply increase in prevalence after the age of 70 years. They account for about 20% of both critical COVID-19 cases in the over 80s and total fatal COVID-19 cases.

Abstract

Circulating autoantibodies (auto-Abs) neutralizing high concentrations (10 ng/ml; in plasma diluted 1:10) of IFN-α and/or IFN-ω are found in about 10% of patients with critical COVID-19 (coronavirus disease 2019) pneumonia but not in individuals with asymptomatic infections. We detect auto-Abs neutralizing 100-fold lower, more physiological, concentrations of IFN-α and/or IFN-ω (100 pg/ml; in 1:10 dilutions of plasma) in 13.6% of 3595 patients with critical COVID-19, including 21% of 374 patients >80 years, and 6.5% of 522 patients with severe COVID-19. These antibodies are also detected in 18% of the 1124 deceased patients (aged 20 days to 99 years; mean: 70 years). Moreover, another 1.3% of patients with critical COVID-19 and 0.9% of the deceased patients have auto-Abs neutralizing high concentrations of IFN-β. We also show, in a sample of 34,159 uninfected individuals from the general population, that auto-Abs neutralizing high concentrations of IFN-α and/or IFN-ω are present in 0.18% of individuals between 18 and 69 years, 1.1% between 70 and 79 years, and 3.4% >80 years. Moreover, the proportion of individuals carrying auto-Abs neutralizing lower concentrations is greater in a subsample of 10,778 uninfected individuals: 1% of individuals 80 years. By contrast, auto-Abs neutralizing IFN-β do not become more frequent with age. Auto-Abs neutralizing type I IFNs predate SARS-CoV-2 infection and sharply increase in prevalence after the age of 70 years. They account for about 20% of both critical COVID-19 cases in the over 80s and total fatal COVID-19 cases. © 2021 The Authors, some rights reserved.

Abstract

Background: Convalescent plasma has been widely used to treat COVID-19 and is under investigation in numerous randomized clinical trials, but results are publicly available only for a small number of trials. The objective of this study was to assess the benefits of convalescent plasma treatment compared to placebo or no treatment and all-cause mortality in patients with COVID-19, using data from all available randomized clinical trials, including unpublished and ongoing trials (Open Science Framework, https://doi.org/10.17605/OSF.IO/GEHFX). Method(s): In this collaborative systematic review and meta-analysis, clinical trial registries (ClinicalTrials.gov, WHO International Clinical Trials Registry Platform), the Cochrane COVID-19 register, the LOVE database, and PubMed were searched until April 8, 2021. Investigators of trials registered by March 1, 2021, without published results were contacted via email. Eligible were ongoing, discontinued and completed randomized clinical trials that compared convalescent plasma with placebo or no treatment in COVID-19 patients, regardless of setting or treatment schedule. Aggregated mortality data were extracted from publications or provided by investigators of unpublished trials and combined using the Hartung-Knapp-Sidik-Jonkman random effects model. We investigated the contribution of unpublished trials to the overall evidence. Result(s): A total of 16,477 patients were included in 33 trials (20 unpublished with 3190 patients, 13 published with 13,287 patients). 32 trials enrolled only hospitalized patients (including 3 with only intensive care unit patients). Risk of bias was low for 29/33 trials. Of 8495 patients who received convalescent plasma, 1997 died (23%), and of 7982 control patients, 1952 died (24%). The combined risk ratio for all-cause mortality was 0.97 (95% confidence interval: 0.92; 1.02) with between-study heterogeneity not beyond chance (I2 = 0%). The objective of this study was to assess the benefits of convalescent plasma treatment compared to placebo or no treatment and all-cause mortality in patients with COVID-19, using data from all available randomized clinical trials, including unpublished and ongoing trials (Open Science Framework, https://doi.org/10.17605/OSF.IO/GEHFX ). METHODS: In this collaborative systematic review and meta-analysis, clinical trial registries (ClinicalTrials.gov, WHO International Clinical Trials Registry Platform), the Cochrane COVID-19 register, the LOVE database, and PubMed were searched until April 8, 2021. Investigators of trials registered by March 1, 2021, without published results were contacted via email. Eligible were ongoing, discontinued and completed randomized clinical trials that compared convalescent plasma with placebo or no treatment in COVID-19 patients, regardless of setting or treatment schedule. Aggregated mortality data were extracted from publications or provided by investigators of unpublished trials and combined using the Hartung-Knapp-Sidik-Jonkman random effects model. We investigated the contribution of unpublished trials to the overall evidence. RESULTS: A total of 16,477 patients were included in 33 trials (20 unpublished with 3190 patients, 13 published with 13,287 patients). 32 trials enrolled only hospitalized patients (including 3 with only intensive care unit patients). Risk of bias was low for 29/33 trials. Of 8495 patients who received convalescent plasma, 1997 died (23%), and of 7982 control patients, 1952 died (24%). The combined risk ratio for all-cause mortality was 0.97 (95% confidence interval: 0.92; 1.02) with between-study heterogeneity not beyond chance (I2 = 0%). Abstract

Assessments of ecosystem service and function losses of wetlandscapes (i.e., wetlands and their hydrological catchments) suffer from knowledge gaps regarding impacts of ongoing hydro-climatic change. This study investigates hydro-climatic changes during 1976–2015 in 25 wetlandscapes distributed across the world’s tropical, arid, temperate and cold climate zones. Results show that the wetlandscapes were subject to precipitation (P) and temperature (T) changes consistent with mean changes over the world’s land area. However, arid and cold wetlandscapes experienced higher T increases than their respective climate zone. Also, average P decreased in arid and cold wetlandscapes, contrarily to P of arid and cold climate zones, suggesting that these wetlandscapes are located in regions of elevated climate pressures. For most wetlandscapes with available runoff (R) data, the decreases were larger in R than in P, which was attributed to aggravation of climate change impacts by enhanced evapotranspiration losses, e.g. caused by land-use changes.
