3. Mechanochemical Reactivity of Ribonucleosides Mediated by Inorganic Species: Implications for Extraterrestrial Organic Matter Interpretation.

Abstract

Metal species and carbonate are often found as minerals in extraterrestrial rocky bodies. Based on this, the mechanochemical-induced degradation of canonical purine and pyrimidine ribonucleosides into their corresponding nucleobases mediated by some of the main constituents of those materials (iron, nickel, or aluminum) was accomplished. In some cases, the previous heating of the samples intensified mechanochemical degradation. Additionally, carbonate acts as an activator for ribonucleoside degradation with a catalyst (a Lewis acid ion); however, it has almost no effect on ribonucleoside degradation in the absence of a catalyst. These results can contribute to the hypothesis that organic matter in extraterrestrial samples could have undergone mechanochemical reactions (i.e., shock/impact events), from its formation until its journey to Earth. Mechanochemical energy could occur in planetesimal accretion, asteroid formation (i.e., through planetesimal disintegration), and meteoroid atmospheric entry. 7. Valorizing sardine scales: a circular approach to sustainable collagen for cosmetics and nutrition applications.

Abstract

Background and Objective: In recent years, the consumption of fish products has led to a worrying trend where approximately two-thirds of the total amount of fish is discarded as waste. At the same time, scientific interest in exploring natural collagen sources for cosmetics and dietary supplements has increased. This study explores the potential of valorizing sardine scales (Sardina pilchardus), a by-product of the canning industry, through the extraction of collagen for potential use in dermocosmetic formulations and food supplements. Methods: Collagen from sardine scales was obtained though acid and enzymatic extraction. The collagen extracts were characterized by UV-Vis, FTIR spectroscopy, SDS-PAGE, powder X-ray diffraction (PXRD) and scanning electron microscopy (SEM). The collagen was hydrolysed with papain to small peptides. Subsequently, the biological activities of acid-soluble collagen as well as the collagen peptides in terms of antioxidant and antimicrobial activity were evaluated. Furthermore, the capacity of collagen peptides to permeate the intestinal barrier, simulated with caco-2 cells, was evaluated. Results: Purified collagen extracts were obtained from sardine scales, with enzymatic extraction method having a yield three times higher than the acid method. The SDS-PAGE analysis confirmed the extraction of type I collagen as well as its hydrolysis into small fragments (25–12 kDa). In terms of biological activities, collagen and collagen peptides have not demonstrated antimicrobial activity. However, regarding antioxidant activity, collagen peptides showed three times more capacity compared to non-hydrolyzed collagen. Meanwhile, in 6 h, about 6.37% of collagen peptides could permeate the intestinal barrier. Conclusion: This work represents a continuous effort to advance our understanding and utilization of Portuguese marine waste resources, with focus on the valorization of sardine co-products for the development of food supplement or cosmetic formulations, contributing to the sustainable evolution of the circular blue economy. 23. Degradable copper(ii)-doped starch-based biopolymeric films with antibacterial activity

Abstract

The search for new bioactive molecules and sustainable materials to address antimicrobial resistance continues to be of significant attention in many research areas. In this work, new copper(II) coordination polymers and complexes containing ammonia and aromatic carboxylate ligands were self-assembled, characterized, and applied as bioactive dopants to produce starch-based biopolymeric films. The structures of [Cu(NH3)2(nca)2] (1) (Hnca = 2-naphthoic acid), [Cu(NH3)2(μ-ndca)]n (2) (H2ndca = 2,6-naphthalenedicarboxylic acid), and [Cu(NH3)2(μ-obba)]n (3) (H2obba = 4,4′-oxybis(benzoic acid)) reveal discrete monocopper(II) units in 1 or 1D coordination polymer chains in 2 and 3. In all compounds, the hexacoordinate Cu(II) centers feature an octahedral {CuN2O4} environment with mutually trans ammonia ligands. The compounds 1–3 were used as bioactive Cu-dopants (5%) to prepare biopolymeric films, 1–3@[PS]n and 1–3@[PS-MCC]n, based on sustainable and low-cost biofeedstocks such as potato starch (PS) or its mixture with microcrystalline cellulose (PS-MCC), respectively. Due to the importance in biomaterial-related infections, the growth inhibition of two clinically significant Gram-positive bacteria species, S. epidermidis and S. aureus, was studied in the presence of the prepared biopolymeric films. The Cu(NH3)2-carboxylates and derived biopolymeric materials showed a pronounced antibacterial activity, with doped films being able to inhibit the growth of 7 out of 8 strains tested, revealing a particularly high performance against the clinical isolates of S. epidermidis. Abstract

LA/P/0140/2020 DL 57/2016 Norma transitória. In Portugal, publications with mechanochemical methods date back to 2009, with the report on mechanochemical strategies for the synthesis of metallopharmaceuticals. Since then, mechanochemical applications have grown in Portugal, spanning several fields, mainly crystal engineering and supramolecular chemistry, catalysis, and organic and inorganic chemistry. The area with the most increased development is the synthesis of multicomponent crystal forms, with several groups synthesizing solvates, salts, and cocrystals in which the main objective was to improve physical properties of the active pharmaceutical ingredients. Recently, non-crystalline materials, such as ionic liquids and amorphous solid dispersions, have also been studied using mechanochemical methods. An area that is in expansion is the use of mechanochemical synthesis of bioinspired metal-organic frameworks with an emphasis in antibiotic coordination frameworks. The use of mechanochemistry for catalysis and organic and inorganic synthesis has also grown due to the synthetic advantages, ease of synthesis, scalability, sustainability, and, in the majority of cases, the superior properties of the synthesized materials. It can be easily concluded that mechanochemistry is expanding in Portugal in diverse research areas. publishersversion published
