1. Human values, civic participation, and wellbeing: analysis on their relationship among older Europeans

Abstract: IntroductionSo far, both for the general and older population, research on human values and wellbeing mainly shows correlational associations but does not inquire about the direction of this relationship. This is also true for values and civic participation. Therefore, our objective is to identify the directional association between civic participation and Schwartz values, and between values and wellbeing, among older Europeans.MethodsA pseudo-panel was created from the cross-sectional data of the European Social Survey (ESS 2002-2018), controlling for gender, age-group, country and level of studies (n = 3926). The data analysis was performed using a cross-lagged model, applying both random-effects and fixed-effects models.ResultsOn the one hand, the relationship between participation and values is bidirectional, but the effect of civic participation on values is more significant since participating stimulates the development of certain values. On the other hand, although the relationship between values and wellbeing is also bidirectional, the effect of wellbeing on values is stronger since a given level of wellbeing favors the development of particular values.DiscussionWe conclude that civic participation should be promoted within the older population since it directly increases wellbeing, and moreover reinforces those (Growth-oriented) values that positively influence the health, happiness and life satisfaction of older people.

2. The Characterization of a Novel PrMADS11 Transcription Factor from Pinus radiata Induced Early in Bent Pine Stem

Abstract: A novel MADS-box transcription factor from Pinus radiata D. Don was characterized. PrMADS11 encodes a protein of 165 amino acids for a MADS-box transcription factor belonging to group II, related to the MIKC protein structure. PrMADS11 was differentially expressed in the stems of pine trees in response to 45° inclination at early times (1 h). Arabidopsis thaliana was stably transformed with a 35S::PrMADS11 construct in an effort to identify the putative targets of PrMADS11. A massive transcriptome analysis revealed 947 differentially expressed genes: 498 genes were up-regulated, and 449 genes were down-regulated due to the over-expression of PrMADS11. The gene ontology analysis highlighted a cell wall remodeling function among the differentially expressed genes, suggesting the active participation of cell wall modification required during the response to vertical stem loss. In addition, the phenylpropanoid pathway was also indicated as a PrMADS11 target, displaying a marked increment in the expression of the genes driven to the biosynthesis of monolignols. The EMSA assays confirmed that PrMADS11 interacts with CArG-box sequences. This TF modulates the gene expression of several molecular pathways, including other TFs, as well as the genes involved in cell wall remodeling. The increment in the lignin content and the genes involved in cell wall dynamics could be an indication of the key role of PrMADS11 in the response to trunk inclination.

3. Are you ready for retirement? The influence of values on membership in voluntary organizations in midlife and old age

Abstract: Membership in voluntary organizations is associated with individual and social benefits. Due to the negative consequences of the global pandemic on older people, and the governmental challenges posed by population aging, voluntary membership is of great importance to society. To effectively promote volunteering among older people, it is necessary to understand the determinants of voluntary membership. This study analyses the influence of individual values—secular/traditional and survival/self-expression–on voluntary membership among European adults (N = 31,985). Specifically, it examines which values orient two age groups (middle age: 50–64 and old age: 65–79), as well as men and women toward a certain type of association (Social Awareness; Professional and Political; Education and Leisure; Religion). The sample of 31,985 comprises 60% of adults aged 50–64 and 40% aged 65–79; of which 56% are women and 44% men. The empirical estimation considers different levels of data aggregation: individual, national and welfare system, therefore multilevel analysis is used as an analytical strategy. Individual-level variables from the Integrated Values Survey (2005/09, 2010/14, and 2017/20) and national-level variables (Gini Index and Gross Domestic Product Per Capita) from the World Bank and Eurostat are used. The results indicate that traditional and self-expression values promote membership in voluntary organizations in general more than secular and survival values. However, there are differences according to the type of organization. Furthermore, values are found to moderate the effect of age and gender on voluntary membership.

4. Correlation between mobility in mass transport and mortality due to COVID-19: A comparison of Mexico City, New York, and Madrid from a data science perspective

Abstract: In most big cities, public transports are enclosed and crowded spaces. Therefore, they are considered as one of the most important triggers of COVID-19 spread. Most of the existing research related to the mobility of people and COVID-19 spread is focused on investigating highly frequented paths by analyzing data collected from mobile devices, which mainly refer to geo-positioning records. In contrast, this paper tackles the problem by studying mass mobility. The relations between daily mobility on public transport (subway or metro) in three big cities and mortality due to COVID-19 are investigated. Data collected for these purposes come from official sources, such as the web pages of the cities’ local governments. To provide a systematic framework, we applied the IBM Foundational Methodology for Data Science to the epidemiological domain of this paper. Our analysis consists of moving averages with a moving window equal to seven days so as to avoid bias due to weekly tendencies. Among the main findings of this work are: a) New York City and Madrid show similar distribution on studied variables, which resemble a Gauss bell, in contrast to Mexico City, and b) Non-pharmaceutical interventions don’t bring immediate results, and reductions to the number of deaths due to COVID are observed after a certain number of days. This paper yields partial evidence for assessing the effectiveness of public policies in mitigating the COVID-19 pandemic.

5. The Botrytis cinerea Gene Expression Browser

Abstract: For comprehensive gene expression analyses of the phytopathogenic fungus Botrytis cinerea, which infects a number of plant taxa and is a cause of substantial agricultural losses worldwide, we developed BEB, a web-based B. cinerea gene Expression Browser. This computationally inexpensive web-based application and its associated database contain manually curated RNA-Seq data for B. cinerea. BEB enables expression analyses of genes of interest under different culture conditions by providing publication-ready heatmaps depicting transcript levels, without requiring advanced computational skills. BEB also provides details of each experiment and user-defined gene expression clustering and visualization options. If needed, tables of gene expression values can be downloaded for further exploration, including, for instance, the determination of differentially expressed genes. The BEB implementation is based on open-source computational technologies that can be deployed for other organisms. In this case, the new implementation will be limited only by the number of transcriptomic experiments that are incorporated into the platform. To demonstrate the usability and value of BEB, we analyzed gene expression patterns across different conditions, with a focus on secondary metabolite gene clusters, chromosome-wide gene expression, previously described virulence factors, and reference genes, providing the first comprehensive expression overview of these groups of genes in this relevant fungal phytopathogen. We expect this tool to be broadly useful in B. cinerea research, providing a basis for comparative transcriptomics and candidate gene identification for functional assays.

6. Prácticas de mujeres en sus trabajos productivos y reproductivos

Abstract: En este artículo nos proponemos desarrollar un primer análisis de datos cuantitativos para describir modalidades de trabajos productivos y características del trabajo reproductivo de mujeres asalariadas y trabajadoras por cuenta propia de la ciudad de Rafaela (Santa Fe, Argentina). Dichos datos provienen de una herramienta estructurada, la Encuesta sobre mujeres y trabajos (EMT), centrada en la descripción del hogar, sus integrantes y su vivienda, las características del trabajo productivo de la respondiente, el uso del tiempo en el hogar y emociones a ellos asociadas. En una muestra de 103 casos, correspondiente a los datos obtenidos en 2018, se destaca la coexistencia de ocupaciones diversas en el ámbito productivo, el sostenimiento de gran parte de las tareas reproductivas y el uso habitual de redes sociales para la realización de actividades comerciales. Así, es posible identificar un conjunto de procesos vinculados a la estructuración social a partir del análisis de estos datos. La feminización del trabajo impone sus tiempos y modalidades, en hogares con diferentes tipologías y la redistribución prioritaria de aquellas tareas que hay que resolver indefectiblemente. Además, el tiempo destinado para sí mismas da cuenta de reconfiguraciones subjetivas con procesos de individualización propios de la sociedad actual. Dichas prácticas indican reconfiguraciones para resolver el dilema de la doble presencia, en el marco de los condicionamientos del trabajo productivo.

7. Ectopic Expression of Arabidopsis thaliana zDof1.3 in Tomato (Solanum lycopersicum L.) Is Associated with Improved Greenhouse Productivity and Enhanced Carbon and Nitrogen Use

Abstract: A large collection of transgenic tomato lines, each ectopically expressing a different Arabidopsis thaliana transcription factor, was screened for variants with alterations in leaf starch. Such lines may be affected in carbon partitioning, and in allocation to the sinks. We focused on ‘L4080’, which harbored an A. thaliana zDof (DNA-binding one zinc finger) isoform 1.3 (AtzDof1.3) gene, and which had a 2–4-fold higher starch-to-sucrose ratio in source leaves over the diel (p < 0.05). Our aim was to determine whether there were associated effects on productivity. L4080 plants were altered in nitrogen (N) and carbon (C) metabolism. The N-to-C ratio was higher in six-week-old L4080, and when treated with 1/10 N, L4080 growth was less inhibited compared to the wild-type and this was accompanied by faster root elongation (p < 0.05). The six-week-old L4080 acquired 42% more dry matter at 720 ppm CO2, compared to ambient CO2 (p < 0.05), while the wild-type (WT) remained unchanged. GC-MS-TOF data showed that L4080 source leaves were enriched in amino acids compared to the WT, and at 49 DPA, fruit had 25% greater mass, higher sucrose, and increased yield (25%; p < 0.05) compared to the WT. An Affymetrix cDNA array analysis suggested that only 0.39% of the 9000 cDNAs were altered by 1.5-fold (p < 0.01) in L4080 source leaves. 14C-labeling of fruit disks identified potential differences in 14-DPA fruit metabolism suggesting that post-transcriptional regulation was important. We conclude that AtzDof1.3 and the germplasm derived therefrom, should be investigated for their ‘climate-change adaptive’ potential.

8. Increasing Heavy Metal Tolerance by the Exogenous Application of Organic Acids

Abstract: Several metals belong to a group of non-biodegradable inorganic constituents that, at low concentrations, play fundamental roles as essential micronutrients for the growth and development of plants. However, in high concentrations they can have toxic and/or mutagenic effects, which can be counteracted by natural chemical compounds called chelators. Chelators have a diversity of chemical structures; many are organic acids, including carboxylic acids and cyclic phenolic acids. The exogenous application of such compounds is a non-genetic approach, which is proving to be a successful strategy to reduce damage caused by heavy metal toxicity. In this review, we will present the latest literature on the exogenous addition of both carboxylic acids, including the Kreb’s Cycle intermediates citric and malic acid, as well as oxalic acid, lipoic acid, and phenolic acids (gallic and caffeic acid). The use of two non-traditional organic acids, the phytohormones jasmonic and salicylic acids, is also discussed. We place particular emphasis on physiological and molecular responses, and their impact in increasing heavy metal tolerance, especially in crop species.

9. Salinity impairs photosynthetic capacity and enhances carotenoid-related gene expression and biosynthesis in tomato (Solanum lycopersicum L. cv. Micro-Tom)

Abstract: Carotenoids are essential components of the photosynthetic antenna and reaction center complexes, being also responsible for antioxidant defense, coloration, and many other functions in multiple plant tissues. In tomato, salinity negatively affects the development of vegetative organs and productivity, but according to previous studies it might also increase fruit color and taste, improving its quality, which is a current agricultural challenge. The fruit quality parameters that are increased by salinity are cultivar-specific and include carotenoid, sugar, and organic acid contents. However, the relationship between vegetative and reproductive organs and response to salinity is still poorly understood. Considering this, Solanum lycopersicum cv. Micro-Tom plants were grown in the absence of salt supplementation as well as with increasing concentrations of NaCl for 14 weeks, evaluating plant performance from vegetative to reproductive stages. In response to salinity, plants showed a significant reduction in net photosynthesis, stomatal conductance, PSII quantum yield, and electron transport rate, in addition to an increase in non-photochemical quenching. In line with these responses the number of tomato clusters decreased, and smaller fruits with higher soluble solids content were obtained. Mature-green fruits also displayed a salt-dependent higher induction in the expression of PSY1, PDS, ZDS, and LYCB, key genes of the carotenoid biosynthesis pathway, in correlation with increased lycopene, lutein, β-carotene, and violaxanthin levels. These results suggest a key relationship between photosynthetic plant response and yield, involving impaired photosynthetic capacity, increased carotenoid-related gene expression, and carotenoid biosynthesis.

10. Defects in the Ferroxidase That Participates in the Reductive Iron Assimilation System Results in Hypervirulence in Botrytis Cinerea

Abstract: ABSTRACT The plant pathogen Botrytis cinerea is responsible for gray-mold disease, which infects a wide variety of species. The outcome of this host-pathogen interaction, a result of the interplay between plant defense and fungal virulence pathways, can be modulated by various environmental factors. Among these, iron availability and acquisition play a crucial role in diverse biological functions. How B. cinerea obtains iron, an essential micronutrient, during infection is unknown. We set out to determine the role of the reductive iron assimilation (RIA) system during B. cinerea infection. This system comprises the BcFET1 ferroxidase, which belongs to the multicopper oxidase (MCO) family of proteins, and the BcFTR1 membrane-bound iron permease. Gene knockout and complementation studies revealed that, compared to the wild type, the bcfet1 mutant displays delayed conidiation, iron-dependent sclerotium production, and significantly reduced whole-cell iron content. Remarkably, this mutant exhibited a hypervirulence phenotype, whereas the bcftr1 mutant presents normal virulence and unaffected whole-cell iron levels and developmental programs. Interestingly, while in iron-starved plants wild-type B. cinerea produced slightly reduced necrotic lesions, the hypervirulence phenotype of the bcfet1 mutant is no longer observed in iron-deprived plants. This suggests that B. cinerea bcfet1 knockout mutants require plant-derived iron to achieve larger necrotic lesions, whereas in planta analyses of reactive oxygen species (ROS) revealed increased ROS levels only for infections caused by the bcfet1 mutant. These results suggest that increased ROS production, under an iron sufficiency environment, at least partly underlie the observed infection phenotype in this mutant. IMPORTANCE The plant-pathogenic fungus B. cinerea causes enormous economic losses, estimated at anywhere between $10 billion and $100 billion worldwide, under both pre- and postharvest conditions. Here, we present the characterization of a loss-of-function mutant in a component involved in iron acquisition that displays hypervirulence. While in different microbial systems iron uptake mechanisms appear to be critical to achieve full pathogenic potential, we found that the absence of the ferroxidase that is part of the reductive iron assimilation system leads to hypervirulence in this fungus. This is an unusual and rather underrepresented phenotype, which can be modulated by iron levels in the plant and provides an unexpected link between iron acquisition, reactive oxygen species (ROS) production, and pathogenesis in the Botrytis-plant interaction.

11. Gene networks underlying the early regulation of Paraburkholderia phytofirmans PsJN induced systemic resistance in Arabidopsis.

Abstract: Plant defense responses to biotic stresses are complex biological processes, all governed by sophisticated molecular regulations. Induced systemic resistance (ISR) is one of these defense mechanisms where beneficial bacteria or fungi prime plants to resist pathogens or pest attacks. In ISR, the defense arsenal in plants remains dormant and it is only triggered by an infection, allowing a better allocation of plant resources. Our group recently described that the well-known beneficial bacterium Paraburkholderia phytofirmans PsJN is able to induce Arabidopsis thaliana resistance to Pseudomonas syringae pv. tomato (Pst) DC3000 through ISR, and that ethylene, jasmonate and salicylic acid are involved in this protection. Nevertheless, the molecular networks governing this beneficial interaction remain unknown. To tackle this issue, we analyzed the temporal changes in the transcriptome of PsJN-inoculated plants before and after being infected with Pst DC3000. These data were used to perform a gene network analysis to identify highly connected transcription factors. Before the pathogen challenge, the strain PsJN regulated 405 genes (corresponding to 1.8% of the analyzed genome). PsJN-inoculated plants presented a faster and stronger transcriptional response at 1-hour post infection (hpi) compared with the non-inoculated plants, which presented the highest transcriptional changes at 24 hpi. A principal component analysis showed that PsJN-induced plant responses to the pathogen could be differentiated from those induced by the pathogen itself. Forty-eight transcription factors were regulated by PsJN at 1 hpi, and a system biology analysis revealed a network with four clusters. Within these clusters LHY, WRKY28, MYB31 and RRTF1 are highly connected transcription factors, which could act as hub regulators in this interaction. Concordantly with our previous results, these clusters are related to jasmonate, ethylene, salicylic, acid and ROS pathways. These results indicate that a rapid and specific response of PsJN-inoculated plants to the virulent DC3000 strain could be the pivotal element in the protection mechanism.

Abstract: One of the functions of the Center of Distance Education of the School of Engineering of the University of Buenos Aires is the design of educational materials using various technologies, so as to facilitate the teaching and learning of science. In this context, a proposal of basic physics simulations design is presented. Previously, simulations were developed in other languages such as Java and Flash. The objective of the initial search was oriented to find open source simulations that can be easily customized or authoring tools such as GeoGebra or EJS. It is intended to avoid implementations from scratch since it is time consuming and difficult to implement in the short term. In particular, a process of development of simulations with HTML5 was started. The reason leading the change was the need to have educational materials that could be used from any terminal, particularly from mobile terminals so that students can make use of the simulator anytime and anywhere. An example of a simulation of a pendulum is provided which main objective is that the student could manipulate variables, observe the behavior of the pendulum and make inferences that help them understand and apply the physical model under study.

13. Inspection of the grapevine BURP superfamily highlights an expansion of RD22 genes with distinctive expression features in berry development and ABA-mediated stress responses.

Abstract: The RESPONSIVE TO DEHYDRATION 22 (RD22) gene is a molecular link between abscisic acid (ABA) signalling and abiotic stress responses. Its expression has been used as a reliable ABA early response marker. In Arabidopsis, the single copy RD22 gene possesses a BURP domain also located at the C-terminus of USP embryonic proteins and the beta subunit of polygalacturonases. In grapevine, a RD22 gene has been identified but putative paralogs are also found in the grape genome, possibly forming a large RD22 family in this species. In this work, we searched for annotations containing BURP domains in the Vitis vinifera genome. Nineteen proteins were defined by a comparative analysis between the two genome predictions and RNA-Seq data. These sequences were compared to other plant BURPs identified in previous genome surveys allowing us to reconceive group classifications based on phylogenetic relationships and protein motif occurrence. We observed a lineage-specific evolution of the RD22 family, with the biggest expansion in grapevine and poplar. In contrast, rice, sorghum and maize presented highly expanded monocot-specific groups. The Vitis RD22 group may have expanded from segmental duplications as most of its members are confined to a region in chromosome 4. The inspection of transcriptomic data revealed variable expression of BURP genes in vegetative and reproductive organs. Many genes were induced in specific tissues or by abiotic and biotic stresses. Three RD22 genes were further studied showing that they responded oppositely to ABA and to stress conditions. Our results show that the inclusion of RNA-Seq data is essential while describing gene families and improving gene annotations. Robust phylogenetic analyses including all BURP members from other sequenced species helped us redefine previous relationships that were erroneously established. This work provides additional evidence for RD22 genes serving as marker genes for different organs or stresses in grapevine.

14. Effects of the plant growth-promoting bacterium Burkholderia phytofirmans PsJN throughout the life cycle of Arabidopsis thaliana.

Abstract: Plant growth-promoting rhizobacteria (PGPR) induce positive effects in plants, such as increased growth or reduced stress susceptibility. The mechanisms behind PGPR/plant interaction are poorly understood, as most studies have described short-term responses on plants and only a few studies have analyzed plant molecular responses under PGPR colonization. Here, we studied the effects of the PGPR bacterial model Burkholderiaphytofirmans PsJN on the whole life cycle of Arabidopsis thaliana plants. We reported that at different plant developmental points, strain PsJN can be found in the rhizosphere and also colonizing their internal tissues. In early ontogeny, strain PsJN increased several growth parameters and accelerated growth rate of the plants. Also, an Arabidopsis transcriptome analysis revealed that 408 genes showed differential expression in PsJN-inoculated plants; some of these genes are involved in stress response and hormone pathways. Specifically, genes implicated in auxin and gibberellin pathways were induced. Quantitative transcriptional analyses of selected genes in different developmental stages revealed that the beginning of these changes could be evidenced early in development, especially among the down-regulated genes. The inoculation with heat-killed bacteria provoked a more severe transcriptional response in plants, but was not able to induce plant growth-promotion. Later in ontogeny, the growth rates of inoculated plants decreased with respect to the non-inoculated group and, interestingly, the inoculation accelerated the flowering time and the appearance of senescence signs in plants; these modifications correlate with the early up-regulation of flowering control genes. Then, we show that a single inoculation with a PGPR could affect the whole life cycle of a plant, accelerating its growth rate and shortening its vegetative period, both effects relevant for most crops. Thus, these findings provide novel and interesting aspects of these relevant biological interactions.

Abstract: Resumen La investigación en la que se funda este artículo tuvo como objetivo comprender cómo se construye la identidad sexual en hombres gay jóvenes, entre 18 y 29 años de edad, de la ciudad de Antofagasta, en el norte de Chile. El enfoque de la investigación fue cualitativo y la selección de los participantes fue de carácter intencional. La técnica de producción de información fue la entrevista semi-estructurada. Se identificaron tres categorías que inciden en la construcción de la identidad sexual de los jóvenes entrevistados: a) la socialización de género; b) la discriminación; y c) la "salida del armario" (revelación de la orientación sexual a otros). Los ejes mencionados son importantes en cómo los entrevistados perciben, viven y aceptan (o no) su orientación sexual. Igualmente, un hallazgo importante es la significación otorgada por los jóvenes entrevistados a la homofobia internalizada, proceso que se sustenta en la construcción e internalización de rígidos roles de género, siendo este tipo de homofobia una de las causas de la discriminación y auto-discriminación que existe entre homosexuales.

Abstract: The main objective of this research is to understand how civic participation influences the well-being of older people, thus being a key activity of active aging. In addition, it recognizes differences by gender, age group and generational cohort. The data are drawn from the European Social Survey (2002–2016), considering individuals born before 1965 in 14 European countries. A pseudo-panel has been constructed from the cross-sectional data and a cross-lagged model applied using random effects (n = 1412). In addition to demonstrating that there is a two-way association between civic participation and the indicators of well-being, this study provides empirical evidence that the relationship from past civic participation to present health and happiness is stronger than vice versa. Therefore, this result confirms the causal effect of civic participation on well-being. Consequently, civic participation and its activities promote the well-being of the elderly and should be encouraged through public and social policies.

Abstract: Undergraduate students participating in the UCLA Undergraduate Research Consortium for Functional Genomics (URCFG) have conducted a two-phased screen using RNA interference (RNAi) in combination with fluorescent reporter proteins to identify genes important for hematopoiesis in Drosophila. This screen disrupted the function of approximately 3500 genes and identified 137 candidate genes for which loss of function leads to observable changes in the hematopoietic development. Targeting RNAi to maturing, progenitor, and regulatory cell types identified key subsets that either limit or promote blood cell maturation. Bioinformatic analysis reveals gene enrichment in several previously uncharacterized areas, including RNA processing and export and vesicular trafficking. Lastly, the participation of students in this course-based undergraduate research experience (CURE) correlated with increased learning gains across several areas, as well as increased STEM retention, indicating that authentic, student-driven research in the form of a CURE represents an impactful and enriching pedagogical approach.

Abstract: Al analizar los aportes sectoriales sobre la recaudación de impuestos, en este caso el análisis de la patente municipal evidenció que las organizaciones poseen escasos conocimientos sobre este tributo. Por tal razón, el objetivo del estudio es determinar su incidencia en el presupuesto de los Gobiernos Autónomos Descentralizados. La investigación posee un orden cuantitativo de alcance correlacional, pues se determinó la relación de la recaudación presupuestada frente la recaudada e ingresos totales en el año 2017. Mediante el coeficiente de correlación de Pearson se midió la relación de las variables y se identificó una relación lineal directa. Se concluye que, si existe incidencia entre la recaudación del impuesto de patentes y el presupuesto de los municipios. Abstract: Consumers of climate model information face difficulty in assessing which models and projections are best for their particular needs. This difficulty stems from the abundance of climate information, as well as the relative inaccessibility or unavailability of information concerning a given model’s quality, trade-offs, and suitability for a particular geographic region or decision-making application. Consumer reports have traditionally provided potential consumers with background knowledge and a review of available products and services to help to make decisions. As a knowledge broker for climate information in the Great Lakes region, the Great Lakes Integrated Sciences and Assessments (GLISA) team has developed a suite of climate model consumer-report-style documents to help climate information consumers make decisions when selecting models and projections for their work. To develop the reports, GLISA reviewed examples of consumer reports from other sectors, relied on the feedback and advice of our ongoing Practitioner Working Group composed of real-world consumers, and incorporated otherwise-unavailable information from model developers. Due to close, continuing partnership with our Practitioner Working Group, the content and the formatting of our climate model consumer reports respond directly to the needs of consumers. Our reports therefore evolve based on needs of the practitioners as well as with the knowledge generated by our research in usability of climate knowledge. We pose that climate model consumer reports, especially when developed in the context of trusted user–knowledge broker relationships, contribute to making climate information more relevant to and usable by practitioners.

29. Civic participation as a promoter of well-being: comparative analysis among european countries

Abstract: Previous research on the direction of the relationship between civic participation and well-being has evidenced that civic participation is a promoter of well-being among older adults in Europe. Accordingly, the objective of the present study is to identify and analyze the differences between European welfare systems regarding both civic participation and well-being. For this purpose, a logistic multilevel regression analysis was performed as an empirical strategy, using the cross-sectional data from the 9 waves of the European Social Survey (2002–2018). Significant differences in well-being were observed, with Anglo-Saxon elders being the healthiest, and the Nordic the happiest and most satisfied with their lives. In contrast, Eastern European seniors reported the lowest levels of well-being. Also, Nordic countries are the most civically engaged, followed by the Continental and Anglo-Saxon, while Mediterranean and Eastern countries engage the least. However, the impact of civic participation on well-being is strongest for the Mediterranean countries, while its impact on satisfaction and happiness is weakest for the Nordic countries. The 3 models of the multilevel analysis indicate that civic participation has a positive impact on health, happiness and life satisfaction, and that this effect is quite robust. Also, by adding country-level macro variables to the model, it is possible to reduce the random effects and hence to better explain these international differences. Concisely, the impact of civic participation on the well-being of the elderly differs across nations and should therefore be considered by policy makers.

Abstract: Nitrate commands genome‐wide gene expression changes that impact metabolism, physiology, plant growth, and development. In an effort to identify new components involved in nitrate responses in plants, we analyze the Arabidopsis thaliana root phosphoproteome in response to nitrate treatments via liquid chromatography coupled to tandem mass spectrometry. 176 phosphoproteins show significant changes at 5 or 20 min after nitrate treatments. Proteins identified by 5 min include signaling components such as kinases or transcription factors. In contrast, by 20 min, proteins identified were associated with transporter activity or hormone metabolism functions, among others. The phosphorylation profile of NITRATE TRANSPORTER 1.1 (NRT1.1) mutant plants was significantly altered as compared to wild‐type plants, confirming its key role in nitrate signaling pathways that involves phosphorylation changes. Integrative bioinformatics analysis highlights auxin transport as an important mechanism modulated by nitrate signaling at the post‐translational level. We validated a new phosphorylation site in PIN2 and provide evidence that it functions in primary and lateral root growth responses to nitrate., Phosphoproteomic analysis in response to nitrate treatment reveals early nitrate‐induced changes in Arabidopsis thaliana root phosphoproteins associated to signaling while late changes are associated to transport and hormone metabolism. Moreover, nitrate promotes the dephosphorylation of PIN2, impacting PIN2 subcellular localization and root development.

Abstract: In most big cities, public transports are enclosed and crowded spaces. Therefore, they are considered as one of the most important triggers of COVID-19 spread. Most of the existing research related to the mobility of people and COVID-19 spread is focused on investigating highly frequented paths by analyzing data collected from mobile devices, which mainly refer to geo-positioning records. In contrast, this paper tackles the problem by studying mass mobility. The relations between daily mobility on public transport (subway or metro) in three big cities and mortality due to COVID-19 are investigated. Data collected for these purposes come from official sources, such as the web pages of the cities’ local governments. To provide a systematic framework, we applied the IBM Foundational Methodology for Data Science to the epidemiological domain of this paper. Our analysis consists of moving averages with a moving window equal to seven days so as to avoid bias due to weekly tendencies. Among the main findings of this work are: a) New York City and Madrid show similar distribution on studied variables, which resemble a Gauss bell, in contrast to Mexico City, and b) Non-pharmaceutical interventions don’t bring immediate results, and reductions to the number of deaths due to COVID are observed after a certain number of days. This paper yields partial evidence for assessing the effectiveness of public policies in mitigating the COVID-19 pandemic.

Abstract: We present a case of a woman affected by Systemic Lupus Erythematosus (SLE) and distal renal tubular acidosis (DRTA) that during pregnancy presented an exacerbation of SLE together with a renal colic with spontaneous stone passage. Radiological exam revealed diffuse calcifications in both kidneys which suggests, in a context of DRTA, a nephrocalcinosis. With the stabilization of SLE and medical treatment directed to correct metabolic alterations we achieved radiological and clinical stability of lithiasic disease.Se presenta el caso de una mujer afecta de Lupus Eritematoso Sistémico (LES) y acidosis tubular renal distal (ATRD) que en el embarazo presenta una exacerbacióndel LES junto con un cuadro de cólico nefrítico con expulsión espontánea de cálculos. Las pruebas radiológicas muestran calcificaciones difusas en ambos parénquimas renales lo cual sugiere, en el contexto de una ATRD, una nefrocalcinosis. Junto con la estabilización del LES y un tratamiento médico dirigido a corregir las alteraciones metabólicas se alcanza una estabilidad radiológica y clínica de la enfermedad litiásica.

Abstract: Availability of the essential macronutrient nitrogen in soil plays a critical role in plant growth, development, and impacts agricultural productivity. Plants have evolved different strategies for sensing and responding to heterogeneous nitrogen distribution. Modulation of root system architecture, including primary root growth and branching, is among the most essential plant adaptions to ensure adequate nitrogen acquisition. However, the immediate molecular pathways coordinating the adjustment of root growth in response to distinct nitrogen sources, such as nitrate or ammonium, are poorly understood. Here, we show that growth as manifested by cell division and elongation is synchronized by coordinated auxin flux between two adjacent outer tissue layers of the root. This coordination is achieved by nitrate‐dependent dephosphorylation of the PIN2 auxin efflux carrier at a previously uncharacterized phosphorylation site, leading to subsequent PIN2 lateralization and thereby regulating auxin flow between adjacent tissues. A dynamic computer model based on our experimental data successfully recapitulates experimental observations. Our study provides mechanistic insights broadening our understanding of root growth mechanisms in dynamic environments., Arabidopsis roots adjust their growth to the type of nitrogen source via phosphorylation of auxin carrier PIN2 and adjustment of auxin distribution between neighboring tissues.

Abstract: The RESPONSIVE TO DEHYDRATION 22 (RD22) gene is a molecular link between abscisic acid (ABA) signalling and abiotic stress responses. Its expression has been used as a reliable ABA early response marker. In Arabidopsis, the single copy RD22 gene possesses a BURP domain also located at the C-terminus of USP embryonic proteins and the beta subunit of polygalacturonases. In grapevine, a RD22 gene has been identified but putative paralogs are also found in the grape genome, possibly forming a large RD22 family in this species. In this work, we searched for annotations containing BURP domains in the Vitis vinifera genome. Nineteen proteins were defined by a comparative analysis between the two genome predictions and RNA-Seq data. These sequences were compared to other plant BURPs identified in previous genome surveys allowing us to reconceive group classifications based on phylogenetic relationships and protein motif occurrence. We observed a lineage-specific evolution of the RD22 family, with the biggest expansion in grapevine and poplar. In contrast, rice, sorghum and maize presented highly expanded monocot-specific groups. The Vitis RD22 group may have expanded from segmental duplications as most of its members are confined to a region in chromosome 4. The inspection of transcriptomic data revealed variable expression of BURP genes in vegetative and reproductive organs. Many genes were induced in specific tissues or by abiotic and biotic stresses. Three RD22 genes were further studied showing that they responded oppositely to ABA and to stress conditions. Our results show that the inclusion of RNA-Seq data is essential while describing gene families and improving gene annotations. Robust phylogenetic analyses including all BURP members from other sequenced species helped us redefine previous relationships that were erroneously established. Abstract: In recent years, the amount of texts in natural language, in digital format, has had an impressive increase. To obtain useful information from a large volume of data, new specialized techniques and efficient algorithms are required. Text mining consists of extracting meaningful patterns from texts; one of the basic approaches is clustering. The most used clustering algorithm is k-means. This chapter proposes an improvement of the k-means algorithm in the convergence step; the process stops whenever the number of objects that change their assigned cluster in the current iteration is bigger than the ones that changed in the previous iteration. Experimental results showed a reduction in execution time up to 93%. It is remarkable that, in general, better results are obtained when the volume of the text increase, particularly in those texts within big data environments.

Abstract: Nitrogen is an essential macronutrient and its availability in soil plays a critical role in plant growth, development and impacts agricultural productivity. Plants have evolved different strategies to sense and respond to heterogeneous nitrogen distribution. Modulating root system architecture, including primary root growth and branching, is among the most essential plant adaptions to ensure adequate nitrogen acquisition. However, the immediate molecular pathways coordinating the adjustment of root growth in response to varying nitrogen sources are poorly understood. Here, using a combination of physiological, live in vivo high- and super resolution imaging, we describe a novel adaptation strategy of root growth on available nitrogen source. We show that growth, i.e. tissue-specific cell division and elongation rates are fine-tuned by modulating auxin flux within and between tissues. Changes in auxin redistribution are achieved by nitrogen source dependent post-translational modification of PIN2, a major auxin efflux carrier, at an uncharacterized, evolutionary conserved phosphosite. Further, we generate a computer model based on our results which successfully recapitulate our experimental observations and creates new predictions that could broaden our understanding of root growth mechanisms in the dynamic environment.

Abstract: The banana passionfruit (Passiflora mollissima Bailey) is a fruit from the Andean region and is used as raw material for the preparation of various food products. Tests were carried out for its in vitro multiplication using somatic embryos obtained from its leaves. A disinfection using sodium hypochlorite for 5 minutes and Murashige and Skoog (MS) growth medium protocols with 1 mg/L of gibberellins was designed in order to induce germination. Vitro explants were selected for the multiplication in the MS medium supplemented with 2 mg/L of benzyl aminopurine and 1 mg/L of naphthenic acid. The stronger leaves were selected and inoculated in the MS medium and Woody Plant (WPM). The formation of embryos was observed since the third week. The formed plants were inoculated in a WPM medium with 1 mg/L of benzyl aminopurine. A 2-ip WPM 1 mg/L medium was used for the rooting period. Finally, they were acclimatized in peat with earth or pearlite, and then planted in pots with earth for further studies. Seed disinfection using sodium hypochlorite presented 20% contamination and 80% of plants germination, proving to be the best disinfectant (p < 0.05). From this last procedure, somatic embryos of leaves in a 2-ip WPM with 1 mg/L were obtained. The ideal acclimatization occurred in the medium with peat and earth, in which case the survival level obtained was 100% by comparison with earth and pearlite, in which no plant growth was observed. Micropropagation represents an economic and effective technique in the breeding of pathogen-free plants.

Abstract: Objetivo:Describir características del consumo de bebidas alcohólicas e identificar patrónde consumo en estudiantes de odontología de la Universidad San Sebastián, sede Concepción.Materiales y métodos:Estudio exploratorio, descriptivo, transversal, no experimental. Lamuestra estuvo constituida por 601 alumnos (65% del total). Se aplicaron dos cuestionariosdurante el segundo semestre del año 2015. El primero: cuestionario estructurado diseñado pararecolectar datos sobre el consumo de alcohol y sus características. El segundo: cuestionarioautoadministrado de”Identificación de Trastornos Relacionados con el Consumo de Alcohol”.Los datos fueron analizados mediante estadígrafos descriptivos. La asociación de la variabledependiente “alcohol” con las variables independientes “edad”, ”género”, “ubicación geográficade origen” y “semestre académico” fueron evaluados mediante una prueba de regresión logísticabinomial. Sólo los datos de los sujetos que consumían alcohol fueron evaluados. El nivel designificación empleado en todos los casos fue deα≤0,05.Resultados:El 15% de los estudiantes es consumidor de riesgo y 18% presenta consumoperjudicial o posible dependencia. En estudiantes que consumen alcohol, la variable ingresofamiliar, en relación con las variables época del año en que consumen y la presencia de problemasderivados del alcohol, están asociadas.Conclusión:Existe mayor prevalencia de consumo en el género masculino y un alto consumode bebidas alcohólicas en ambos géneros en cualquier época del año. El consumo de bebidasalcohólicas es independiente del ingreso familiar.

Abstract: Patients with rheumatoid arthritis (RA) may display atypical CD21-/lo B cells in their blood, but the implication of this observation remains unclear. We report here that the group of patients with RA and elevated frequencies of CD21-/lo B cells shows decreased ataxia telangiectasia-mutated (ATM) expression and activation in B cells compared with other patients with RA and healthy donor controls. In agreement with ATM involvement in the regulation of V(D)J recombination, patients with RA who show defective ATM function displayed a skewed B cell receptor (BCR) Igκ repertoire, which resembled that of patients with ataxia telangiectasia (AT). This repertoire was characterized by increased Jκ1 and decreased upstream Vκ gene segment usage, suggesting improper secondary recombination processes and selection. In addition, altered ATM function in B cells was associated with decreased osteoprotegerin and increased receptor activator of nuclear factor κB ligand (RANKL) production. These changes favor bone loss and correlated with a higher prevalence of erosive disease in patients with RA who show impaired ATM function. Using a humanized mouse model, we also show that ATM inhibition in vivo induces an altered Igκ repertoire and RANKL production by immature B cells in the bone marrow, leading to decreased bone density. We conclude that dysregulated ATM function in B cells promotes bone erosion and the emergence of circulating CD21-/lo B cells, thereby contributing to RA pathophysiology.

Abstract: Nitrate is an essential macronutrient for plants, a primary nitrogen source in natural and human-made ecosystems. Nitrate can also act as a signaling molecule that directs genome-wide gene expression changes with an impact on plant metabolism, physiology, growth and development. Nitrate and phytohormone signaling pathways crosstalk to modulate growth and developmental programs in a multifactorial manner. Nitrate-signaling controls plant growth and development using molecular mechanisms that involve phytohormone-signaling pathways. In contrast, many phytohormones modulate or impact nitrate signaling in interconnected pathways. In this review, we explore recent progress in our understanding of well-documented connections between nitrate and phytohormones such as auxin, cytokinin and abscisic acid. We also discuss recent studies connecting nitrate to other phytohormones such as ethylene, salicylic acid, gibberellins and brassinosteroids. While many molecular details remain to be elucidated, a number of core signaling components at the intersection between nitrate and the major hormonal pathways have been described. We focus on established interactions of nitrate and different hormonal pathways to bring about cellular, growth and developmental processes in Arabidopsis thaliana.

Abstract: Plant defense responses to biotic stresses are complex biological processes, all governed by sophisticated molecular regulations. Induced systemic resistance (ISR) is one of these defense mechanisms where beneficial bacteria or fungi prime plants to resist pathogens or pest attacks. In ISR, the defense arsenal in plants remains dormant and it is only triggered by an infection, allowing a better allocation of plant resources. Our group recently described that the well-known beneficial bacterium Paraburkholderia phytofirmans PsJN is able to induce Arabidopsis thaliana resistance to Pseudomonas syringae pv. tomato (Pst) DC3000 through ISR, and that ethylene, jasmonate and salicylic acid are involved in this protection. Nevertheless, the molecular networks governing this beneficial interaction remain unknown. To tackle this issue, we analyzed the temporal changes in the transcriptome of PsJN-inoculated plants before and after being infected with Pst DC3000. These data were used to perform a gene network analysis to identify highly connected transcription factors. Before the pathogen challenge, the strain PsJN regulated 405 genes (corresponding to 1.8% of the analyzed genome). PsJN-inoculated plants presented a faster and stronger transcriptional response at 1-hour post infection (hpi) compared with the non-inoculated plants, which presented the highest transcriptional changes at 24 hpi. A principal component analysis showed that PsJN-induced plant responses to the pathogen could be differentiated from those induced by the pathogen itself. Forty-eight transcription factors were regulated by PsJN at 1 hpi, and a system biology analysis revealed a network with four clusters. Within these clusters LHY, WRKY28, MYB31 and RRTF1 are highly connected transcription factors, which could act as hub regulators in this interaction. Concordantly with our previous results, these clusters are related to jasmonate, ethylene, salicylic, acid and ROS pathways. These results indicate that a rapid and specific response of PsJN-inoculated plants to the virulent DC3000 strain could be the pivotal element in the protection mechanism.
