Silicon phosphides with a new structure have been synthesized. These salt inclusion materials contain exotic polycations such as [Ba6Cl]11+. The structure and properties are investigated.

Ultraviolet radiation (UVR) increases the incidence of cutaneous melanoma1–4. The ageing, sun-exposed dermis accumulates UVR damage5, and older patients develop more melanomas at UVR-exposed sites4,6,7. As fibroblasts are functionally heterogeneous and play key roles in the stromal contribution to cancer8,9, we asked whether UVR modifies dermal fibroblast function. Here we confirmed the expression of collagen-cleaving matrix metalloprotein-1 (MMP1) by UVR-damaged fibroblasts was persistently upregulated to reduce local levels of collagen 1 (COL1A1), and found dermal COL1A1 degradation by MMP1 decreased melanoma invasion. Conversely, we show inhibiting extracellular matrix degradation and MMP1 expression restored melanoma invasion to UVR damaged dermis. We confirmed in vitro findings in a cohort of primary cutaneous melanomas of aged humans, showing more cancer cells invade as single cells at the invasive front of melanomas expressing and depositing more collagen. We found collagen and single melanoma cell invasion are robust predictors of poor melanoma-specific survival. These data indicate melanomas arising over UVR-damaged, collagen-poor skin of the elderly are less invasive, and this reduced invasion improves survival. Consequently, although UVR increases tumour incidence, it delays primary melanoma invasion by degrading collagen. However, we show melanoma-associated fibroblasts can restore invasion in low-collagen primary tumours by increasing collagen synthesis. Finally, we demonstrate high COL1A1 gene expression is a biomarker of poor outcome across a broad range of primary cancers.

Centriole duplication is tightly controlled to maintain correct centriole number through the cell cycle. Key to this is the regulated degradation of PLK4, the master regulator of centriole duplication. Here, we show that the Rac1 guanine nucleotide exchange factor (GEF) Tiam1 localises to centrosomes during S-phase, where it is required for the maintenance of normal centriole number. Depletion of Tiam1 leads to an increase in centrosomal PLK4 and centriole overduplication, whereas overexpression of Tiam1 can restrict centriole overduplication. Ultimately, Tiam1 depletion leads to lagging chromosomes at anaphase and aneuploidy, which are potential drivers of malignant progression. The effects of Tiam1 depletion on centrosomal PLK4 levels and centriole overduplication can be rescued by re-expression of both wild-type Tiam1 and catalytically inactive (GEF*) Tiam1, but not by Tiam1 mutants unable to bind to the F-box protein βTRCP (also known as F-box/WD repeat-containing protein 1A) implying that Tiam1 regulates PLK4 levels through promoting βTRCP-mediated degradation independently of Rac1 activation., Summary: Tiam1 maintains centriole number and ensures correct chromosome segregation via interaction with βTRCP, part of the E3 ligase complex, which degrades PLK4, the master regulator of centriole duplication.

The Tiam family proteins — Tiam1 and Tiam2/STEF — are Rac1-specific Guanine Nucleotide Exchange Factors (GEFs) with important functions in epithelial, neuronal, immune and other cell types. Tiam GEFs regulate cellular migration, proliferation and survival, mainly through activating and directing Rac1 signalling. Dysregulation of the Tiam GEFs is significantly associated with human diseases including cancer, immunological and neurological disorders. Uncovering the mechanisms and consequences of dysregulation is therefore imperative to improving the diagnosis and treatment of diseases. Here we compare and contrast the subcellular localisation and function of Tiam1 and Tiam2/STEF, and review the evidence for their dysregulation in disease.

Andrew Porter is first author on ‘The interaction between CASK and the tumour suppressor Dlg1 regulates mitotic spindle orientation in mammalian epithelia’, published in JCS. Andrew is a postdoc in the lab of Prof. Angeliki Malliri at the Cancer Research UK Manchester Institute, investigating how abnormal mitotic events – spindle misorientation, centriole defects and chromosomal instability – contribute to tumourigenesis.

Oriented cell divisions are important for the formation of normal epithelial structures. Dlg1, a tumour suppressor, is required for mitotic spindle orientation in Drosophila epithelia and chick neuroepithelia, but how Dlg1 is localised to the membrane and its importance in mammalian epithelia are unknown. We show that Dlg1 is required in non-transformed mammalian epithelial cells for oriented cell divisions and normal lumen formation. We demonstrate that the MAGUK protein CASK, a membrane-associated scaffold, is the factor responsible for Dlg1 membrane localisation during spindle orientation, thereby identifying a new cellular function for CASK. Depletion of CASK leads to misoriented divisions in 3D, and to the formation of multilumen structures in cultured kidney and breast epithelial cells. Blocking the CASK–Dlg1 interaction with an interfering peptide, or by deletion of the CASK-interaction domain of Dlg1, disrupts spindle orientation and causes multilumen formation. We show that the CASK–Dlg1 interaction is important for localisation of the canonical LGN–NuMA complex known to be required for spindle orientation. These results establish the importance of the CASK–Dlg1 interaction in oriented cell division and epithelial integrity. The new cover of Experimental Biology and Medicine features the hermeneutic circle of biology, a concept we have adapted from the hermeneutic principle that one understands the whole only in terms of each part and the parts only in terms of the whole. Our hermeneutic circle summarizes the course of experimental biology through 2500 years of the achievements of reductionist research (understanding the parts), which culminates in our ability to rapidly sequence the genome. Rather than returning along the same path in a constructionist approach that simply builds upon this knowledge, but in reverse, an alternative is to close the circle with synthetic constructions that seek to integrate the full complexity of biological and physiological systems (understanding the whole), of which organs-on-chips are one example. This closing of the circle cannot be a comprehensively accurate representation of biology, but it can be a synthetic one that effectively defines particular biological subsystems. The illustration of the hermeneutic circle of biology is also intended to suggest both the multiple cycles that may be required to reach such a synthesis and the expansion of the circle in an outward spiral as knowledge increases. Our commentary explains the symbolism of the new cover in a philosophical and scientific discussion.

In vitro and in vivo studies and evidence from human tumors have long implicated Rho GTPase signaling in the formation and dissemination of a range of cancers. Recently next generation sequencing has identified direct mutations of Rho GTPases in human cancers. Moreover, the effects of ablating genes encoding Rho GTPases and their regulators in mouse models, or through pharmacological inhibition, strongly suggests that targeting Rho GTPase signaling could constitute an effective treatment. In this review we will explore the various ways in which Rho signaling can be deregulated in human cancers.

This paper is about the relation of historical relativity and religious certainty. To live in history and to live with historical relativity is to admit that we are subject to the limitations of creaturehood: we know right and wrong, truth and falsity, relative to our own time and place, history and culture, but we do not know any truth or right that is non-relative to time and place. The differences between historical and naturalistic discourses of explanation are radical, and usually overlooked by scientists.

People encountering the disabled often reveal in their reactions a distaste for disability that comes from pity rather than solidarity; when that pity is traced to its roots, such people are capable of urging the disabled toward a “merciful death,” rather than offering any solidarity. The result is acceptance of euthanasia for the disabled. At stake is the difference between grace and works: for a disabled person by his or her mere presence reveals one's dependence on grace. But if one can “help” the disabled one, then one gains a sense of control that comes with works.

The paper finds at least nine material differences between acts in history and entities in nature. (1) Nature rules out intentional structures essential to human acts. (2) Material trajectories in nature are unique, but acts in history are open to multiple interpretations. (3) In terms of set theory, history is bigger than nature. (4) Historical acts cannot be demarcated from the rest of the world by interactions with the world at a boundary. What happens far off-stage can transform human acts in focal view. (5) Human acts can be revised after the fact. Material trajectories in nature cannot be. (6) Meaning and interpretation are constitutive of human acts. (7) Human acts may involve physical motions; they do not always do so. (8) Human acts always involve final causes. (9) Efficient causes in nature determine their effects; efficient causes in history may not.

Alvin Plantinga, Phillip Johnson, and many others, following C. S. Lewis, take their definition of naturalism to mean rejection of the supernatural (and with it, theism). A better definition of contemporary naturalism is proposed: naturalism thinks in terms of reality as understood by the sciences, and the natural is composed of systems demarcated from the rest of the world; those systems have states, and a state is an unambiguous function of time. Naturalism abstracts these from all other features of the things it knows. Action is interpreted as an efficient cause making a change in the trajectory of a natural system. Methodological naturalism merely applies these definitions to this world for purposes of science. Comprehensive naturalism insists that all things whatsoever are exhaustively intelligible on naturalistic terms. Although Rac and its activator Tiam1 are known to stimulate cell-cell adhesion, the mechanisms regulating their activity in cell-cell junction formation are poorly understood. Here, we identify β2-syntrophin as a Tiam1 interactor required for optimal cell-cell adhesion. We show that during tight-junction (TJ) assembly β2-syntrophin promotes Tiam1-Rac activity, in contrast to the function of the apical determinant Par-3 whose inhibition of Tiam1-Rac activity is necessary for TJ assembly. We further demonstrate that β2-syntrophin localizes more basally than Par-3 at cell-cell junctions, thus generating an apicobasal Rac activity gradient at developing cell-cell junctions. Targeting active Rac to TJs shows that this gradient is required for optimal TJ assembly and apical lumen formation. Consistently, β2-syntrophin depletion perturbs Tiam1 and Rac localization at cell-cell junctions and causes defects in apical lumen formation. We conclude that β2-syntrophin and Par-3 fine-tune Rac activity along cell-cell junctions controlling TJ assembly and the establishment of apicobasal polarity.
