Magnetic solid-phase extraction coupled to high-performance liquid chromatography (HPLC)-diode array detection method has been developed for the sensitive analysis of Anastrozole (ANA) and Letrozole (LET) in human urine samples. Characterization studies of the newly synthesized material (Fe3O4@TEPA) were carried out by Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction, Raman spectroscopy, and scanning electron microscope techniques. The method has provided linearity for both the analytes ( r2 > 0.9900) in the range of 4.00–160.00 ng mL−1. The limit of detections for ANA and LET was found to be 1.14 and 1.26 ng mL−1, respectively. The limit of quantification for ANA and LET was obtained 3.78 and 3.86 ng mL−1, respectively. The new method was simple, convenient, fast, and suitable for the direct analysis of ANA and LET in urine samples. Finally, the developed method was applied to the spiked human urine samples to test accuracy and repeatability. Recovery values for each of the molecules were calculated in the range of 93.4–108.0% (n = 3) at two concentration levels, while % RSDs were lower than 6.2%.

The current research investigated the chemical profiling and biological activities of Emex australis Steinh. aerial and fruit parts (methanol and dichloromethane-DCM extracts). Chemical composition was established by determining the amounts of phenolic and flavonoid contents, HPLC-PDA polyphenolic quantification, and UHPLC-MS secondary metabolites profiling to understand the observed biological activities. Antioxidant evaluation was performed utilizing six different assays (DPPH, ABTS, FRAP, CUPRAC, phosphomolybdenum, and metal chelation). Moreover, enzyme inhibition abilities against cholinesterases, α-amylase, α-glucosidase, and tyrosinase were also determined. The fruit methanol extract contained the highest phenolic (157.96 mg GAE/ g extract) and flavonoid contents (41.43 mg QE/g extract), which could justify the observed significant antioxidant (except metal chelating), α-amylase, α-glucosidase, and tyrosinase inhibitory potentials. The other extracts were active against cholinesterases and tyrosinase. The HPLC-PDA polyphenolic analysis quantified a number of 11 phenolic secondary metabolites. The fruit-MeOH extract was found to quantify the maximum phenolics with catechin (4.27 μg/g extract) and gallic acid (4.84 μg/g extract) in higher amounts. Similarly, rutin present in maximum quantity in the aerial-MeOH extract (2.93 μg/g extract). Furthermore, the UHPLC-MS study of aerial and fruit methanol extracts reveals the existence of 22 secondary metabolites, most of which were flavonoid derivatives. Besides that, Pearson coefficient analysis (PCA) was also carried to determine a possible correlation between bioactive contents and observed biological assays. Thus, it is concluded that E. australis extracts contain important classes of secondary metabolites and also exhibited considerable antioxidant and inhibition potential against clinically relevant enzymes.

Narcotic and psychotropic substances are natural, synthetic, or semisynthetic compounds that are present in both solid and liquid illicit products. The alterations effects on the central nervous system related to their use can be psycholeptic, psychoanaleptic, or psychodiseptic and are able to generate tolerance, addiction, or dependence phenomena, creating social and public order problems. In this scenario, the analytical evaluations that aim to determine these analytes in seized nonbiological samples, and which assume the character of judicial evidence, must meet high analytical requirements of reliability, transparency, and procedures uniformity at a national level. For the first time in the literature, the herein validated method is able to provide the simultaneous quantitative determination of 37 of the most common narcotic substances as well as the most commonly used excipients/adulterants found in seized illicit material. Nowadays, the challenges in analytical chemistry, and mostly in quantitative analysis, include the development and validation of new materials, strategies and procedures to meet the growing need for rapid, sensitive, selective and green methods. In this context, considering the constantly updated International Guidelines, constant innovation is mandatory both in the pre-treatment procedures and in the instrumental configurations to obtain reliable, true, and reproducible information. In this context, additionally to the classic plasma (or serum) matrices, biopsies, whole blood, and urine have seen an increase in the works that also consider non-conventional matrices. Obviously, all these studies have shown that there is a correlation between the blood levels and those found in the new matrix, in order to be able to correlate and compare the results in a robust way and reduce any bias problems. This review provides an update of the most recent developments currently in use in the sample pre-treatment and instrument configurations in the biological/clinical fields. Furthermore, the review concludes with a series of considerations regarding the role and future developments of Analytical Chemistry in light of the forthcoming challenges and new goals to be achieved.

Background: Biopharmaceuticals are biological drugs consisting of a complex compound that can be produced by a living organism or derive from it. Biopharmaceuticals are very complicated compounds from structural point of view and for this reason, they cannot be fully characterized in terms of their structure with current analytical methods as it happens instead of low molecular weight chemicals drugs. Introduction: The regulatory guidelines require the characterization of the primary or higher sequence of these molecules and the characterization of any post-translational modifications. The use of biopharmaceuticals has really grown in the last few years: in 2016, the number of biopharmaceuticals approved by the Food and Drug Administration (FDA) and European Medicines Agency (EMA) for use in humans’ diseases was 1357. From 2013 to 2016, 73 of these compounds were approved for the treatment of cancer, inflammation, immune disorders, infections, anemia and cardiovascular diseases. Aim/Conclusion: The aim of the present review is to provide an overview of recent approaches for the characterization of biopharmaceutical products in HPLC that have been presented in the literature in the last years.

Background: Recently, in all fields of analytical chemistry, increased attention has been paid to extraction procedures and instrumental methods, which are easily scalable and are able to automate in order to improve the “high-throughput” capability. Introduction: The main goal of these applications relates to an improvement of the precision in the quantitative analysis, reduction of different sources of errors, decrease the analysis time and, in general, improve the analytical performances. Often these points can be in contrast to each other, not allowing to achieve the expected result but forcing a compromise between the objectives of the method and the analytical performance. Methods: In this review, following the evolution of the (micro)extraction procedures and instrument configurations, the recent procedures used in bioanalytical chemistry are critically evaluated. The aim of this paper is providing an overview of the approaches available in order to perform on-line coupling of various extraction techniques with chromatographic methods for the analysis of different compounds in various samples. Furthermore, a comparison between off-line and on-line systems, advantages of on-line systems applied on major extractive techniques and future perspectives are described. Result: The extraction methods suitable for on-line coupling covered in this review are: liquid-liquid extraction (LLE), solid phase extraction (SPE), solid phase microextraction (SPME), dispersive liquid- liquid microextraction (DLLME), microextraction by packed sorbent (MEPS), supercritical fluid extraction (SFE) and fabric phase sorptive extraction (FPSE). Conclusion: An overview of the micro-extraction techniques mentioned above was provided, making a comparison between them and focusing attention on future perspectives.

A new analyte separation and preconcentration method for the trace determination of antidepressant drugs, Fluoxetine (FLU) and Citalopram (CIT) in urine and wastewaters, was developed based on HPLC-DAD analysis after magnetic solid phase extraction (MSPE). In the proposed method, FLU and CIT were retained on the newly synthetized magnetic sorbent (Fe3O4@PPy-GO) in the presence of buffer (pH 10.0) and then were desorbed into a lower volume of acetonitrile prior to the chromatographic determinations. Before HPLC analysis, all samples were filtered through a 0.45 µm PTFE filter. Experimental parameters such as interaction time, desorption solvent and volume, and pH were studied and optimized in order to establish the detection limit, linearity, enrichment factor and other analytical figures of merit under optimum operation conditions. In the developed method, FLU and CIT were analyzed by diode array detector at the corresponding maximum wavelengths of 227 and 238 nm, respectively, by using an isocratic elution of 60% pH 3.0 buffer, 30% acetonitrile, and 10% methanol. By using the optimum conditions, limit of detections for FLU and CIT were 1.58 and 1.43 ng mL−1, respectively, while the limit of quantifications was 4.82 and 4.71 ng mL−1, respectively. Relative standard deviations (RSD%) for triplicate analyses of model solutions containing 100 ng mL−1 target molecules were found to be less than 5.0 %. Finally, the method was successfully applied to urine (both simulated and real healthy human) and wastewater samples, and quantitative results were obtained in recovery experiments.

Background: Ionic liquids (ILs) are a unique class of compounds consisting exclusively of cations and anions that possess distinctive properties such as low volatility, high thermal stability, miscibility with water and organic solvents, electrolytic conductivity and non-flammability. Ionic liquids have been defined as "design solvents", because it is possible to modify their physical and chemical properties by appropriately choosing cations and anions, in order to meet the specific characteristics based on their potential application. Introduction: Due of their tunable nature and properties, ILs are considered as the perfect candidates for numerous applications in analytical chemistry including sample preparation, stationary phases in liquid or gas chromatography, additives in capillary electrophoresis, or in mass spectrometry for spectral and electrochemical analysis. In the last years, the number of publications regarding ILs has rapidly increased, highlighting the broad applications of these compounds in various fields of analytical chemistry. Results: This review first described the main physico-chemical characteristics of ionic liquids, and subsequently reported the various applications in different subdisciplines of analytical chemistry, including the extraction procedure and separation techniques. Furthermore, in each paragraph the most recent applications of ionic liquids in the food, environmental, biological, etc. fields have been described. Conclusion: Overall, the topic discussed highlights the key role of ionic liquids in analytical chemistry, giving hints for their future applications in chemistry but also in biology and medicine.

Therapeutic drug monitoring (TDM) of antiseizure medications (ASMs) represents a valuable tool to establish an appropriate patient therapy, to collect important information about drugs’ interactions and to evaluate patient’s metabolic capabilities. In recent years, a new volumetric absorptive microsampling technique using VAMS® technology and Mitra® devices, consisting of a sampling technique for the collection of fixed-volume capillary blood, was developed. These new devices provide a new home-sampling technique for whole blood that has been spread out to simplify sample collection from finger-pricks. This review is aimed to compare published articles concerning the application of VAMS® in epilepsy and to identify the strengths and improvement points for the TDM of antiseizure medications. VAMS® allowed a minimally invasive blood sampling even in the absence of trained personnel. Good stability data have indicated that storage and delivery can be facilitated only for specific ASMs. Trueness and precision parameters have been evaluated, and the hematocrit (HCT) effect was minimized.

Currently, forensic research is multidisciplinary with new methods and parameters useful to define the cause and time of death as well as survival/agony times. The identification of biochemical markers able to estimate agonal period has been studied by many forensic researchers. It is known that the estimation of agonal time in different types of death is not always easy, hence our interest in literature’s data. The studies analyzed in this review confirm the important role of thanatobiochemistry for the estimation of survival times. Regardless of the death cause, the survival/agony time between the primary event and death influences markers concentrations in biological samples (e.g., blood, urine, cerebrospinal fluid). Different biomarkers can be used for qualitative evaluations in deaths with short and long agony (e.g., C-reactive protein, ferritin, GFAP, etc.). Instead, the quantitative interpretation showed limits due to the lack of reference cut-offs. Thanatobiochemistry is a useful tool to confirm what emerged from autopsies findings (macroscopic and histological analysis), but further studies are desirable to confirm the evidence emerging from our review of the literature.

© 2021 Elsevier B.V.A magnetic solid phase extraction (MSPE) coupled with high-performance liquid chromatography-diode array detection (HPLC-DAD) methodology was developed for the determination of chloramphenicol (CP) and tetracycline (TET) antibiotic residues in milk samples. As a solid phase sorbent, C-nanofiber coated magnetic nanoparticles were synthesized and extensively characterized using Field Emission Scanning Electron Microscopy (FE-SEM), Raman Spectroscopy and X-ray Powder Diffraction (XRD) analysis. Experimental variables of MSPE method for both antibiotic analytes were investigated and optimized systematically. After MSPE, the linear range for both the analytes (r2 > 0.9954) were obtained in a range 10.0–600.0 ng mL−1. The limit of detections (LODs) for CP and TET were 3.02 and 3.52 ng mL−1, respectively while RSDs % were below than 4.0%. Finally, the developed method based on MPSE-HPLC-DAD was applied to real milk samples to quantify the antibiotic residues. Recovery values for each antibiotic compound were found in the range of 94.6–105.4% (n = 3) by using spiked model solution.

This study introduces an innovative device for the noninvasive sampling and chromatographic analysis of different compounds present in exhaled breath aerosol (EBA). The new sampling device, especially in light of the recent COVID-19 pandemic that forced many countries to impose mandatory facemasks, allows an easy monitoring of the subject’s exposure to different compounds they may come in contact with, actively or passively. The project combines the advantages of a fabric-phase sorptive membrane (FPSM) as an in vivo sampling device with a validated LC-MS/MS screening procedure able to monitor more than 739 chemicals with an overall analysis time of 18 min. The project involves the noninvasive in vivo sampling of the EBA using an FPSM array inserted inside an FFP2 mask. The study involved 15 healthy volunteers, and no restrictions were imposed during or prior to the sampling process regarding the consumption of drinks, food, or drugs. The FPSM array-LC-MS/MS approach allowed us to effectively exploit the advantages of the two complementary procedures (the convenient sampling by an FPSM array and the rapid analysis by LC-MS/MS), obtaining a powerful and green tool to carry out rapid screening analyses for human exposure to different compounds. The flexible fabric substrate, the sponge-like porous architecture of the high-efficiency sol–gel sorbent coating, the availability of a large cache of sorbent coatings, including polar, nonpolar, mixed mode, and zwitterionic phases, the easy installation into the facemask, and the possibility of sampling without interrupting regular activities provide FPSMs unparalleled advantages over other sampling techniques, and their applications are expected to expand to many other clinical or toxicological studies.

Following the convenient, yet very powerful pathway to create designer extraction sorbent using sol-gel chemistry, a novel sol-gel phenyl/methyl/poly(dimethylsiloxane) sorbent coating was created on polyester fabric substrate for fabric phase sorptive extraction (FPSE) and was subsequently applied to monitor human exposure to selected polycyclic aromatic hydrocarbons (PAHs) including pyrene, chrysene, and benzo[a]pyrene in plasma samples obtained from tobacco smoker volunteers using high performance liquid chromatography-fluorescence detector (HPLC-FLD). A rapid FPSE-HPLC-FLD method was developed that adequately resolved the PAHs chromatographically, after their successful extraction from human plasma using fabric phase absorption extraction (FPSE) and subsequently analysed in the liquid chromatographic system by means of an analytical column (InterSustain C-18 column 150 × 4.6 mm, 5 μm) using acetonitrile (ACN) and water as mobile phases in gradient elution mode. With the optimized conditions, the retention times were found to be 6.168, 7.214, and 10.404 min for pyrene, chrysene, and benzo[a]pyrene, respectively. The total chromatographic runtime was limited to 12.5 min. The method, validated through the calculation of all the analytical parameters according to the International Guidelines, was applied to the analysis of real samples collected from informed volunteers. The proposed approach which included the use of sol-gel phenyl/methyl/poly(dimethylsiloxane) immobilized on hydrophobic polyester substrate and C18 stationary phase used in HPLC, has shown a high potential as a rapid tool for future clinical, forensic and toxicological applications, also in the light of the LOD and LOQ values comparable to those normally obtainable with more sophisticated, and expensive instruments that often require highly trained personnel. The results reported here further consolidate the application of FPSE in the analysis of biological samples for both diagnostic and analytical-clinical purposes.

Ethanol (ethylic alcohol) represents the most commonly used drug worldwide and is often involved in clinical and forensic toxicology. Based on several reports, excessive alcohol consumption is the main contributing factor in traffic accidents, drownings, suicides, and other crimes. For these reasons, it becomes essential to analyze the alcohol concentration during autopsy. Although blood is usually used for alcohol analysis in post-mortem cases, it could suffer alterations, putrefaction, and microbial contaminations. As an alternative to whole blood, vitreous humor has been successfully used in medico-legal studies. In this work, post-mortem specimens were analyzed for ethanol determination. The analysis of blood and vitreous humor were carried-out using gas chromatography-flame ionized detector (GC-FID) with a total run time of 6 min. The method was validated in terms of limit of detection, limit of quantification, dynamic range, sensibility, recovery, precision and trueness. A linear regression analysis indicated a coefficient of determination (R2) of 0.9981. The study confirmed no statistically differences between alcohol concentration in blood and vitreous humor, leading vitreous humor as an excellent matrix that could be used as an alternative to whole blood in toxicological analysis in cases where blood is not available.

Recently, high demand of high-throughput analyses with high sensitivity and selectivity to molecules and drugs in different classes with different physical-chemical properties—and a reduction in analysis time—is a principal milestone for novel methodologies that researchers are trying to achieve—especially when analytical procedures are applied to clinical purposes. In addition, to avoid high doses of a single drug that could cause serious side effects, multi-drug therapies are often used to treat numerous diseases. For these reasons, the demand for methods that allow the rapid analysis of mixed compounds has increased in recent years. In order to respond to these needs, new methods and instruments have been developed. However, often the complexity of a matrix can require a long time for the preparation and processing of the samples. Different problems in terms of components, types of matrices, compounds and physical-chemical complexity are encountered when considering drugs association profiles for quantitative analyses. This review addresses not only recently optimized procedures such as chromatographic separation, but also methods that have allowed us to obtain accuracy (precision and trueness), sensitivity and selectivity in quantitative analyses for cases of drug associations.

A new sample preparation and chromatographic separation method for methyl paraben (MP), propyl paraben (PP) and butyl paraben (BP) in cosmetic and environmental samples has been developed, based on fabric phase sorptive extraction (FPSE) and high performance liquid chromatography-photodiode array (HPLC-PDA) detection. In the proposed method, MP, PP and BP molecules were efficiently retained on a sol–gel Carbowax-20M sorbent-coated FPSE membrane when the matrix pH was adjusted to 5. Subsequently, the extracted analytes were desorbed from the FPSE membrane with methanol prior to chromatographic separation and determinations. The samples were transferred to HPLC vials after filtration with a 0.45 μm porous PTFE filter. Experimental conditions were studied to optimize variables such as pH, adsorption time and desorption solvent. Using the optimal conditions, analytical parameters such as linearity ranges, detection limits, and pre-concentration factors for each of the selected parabens were calculated from experimental data. HPLC analyses of MP, PP and BP were carried out in isocratic elution mode using acetic acid (3%) : acetonitrile : methanol in the ratio 70 : 20 : 10. Limit of detection (LOD) values for MP, PP and BP were calculated as 2.85, 2.98 and 2.75 ng mL−1, respectively. Relative standard deviations (RSDs) were below 3.8% for determinations of model solutions including 100 ng mL−1 of each paraben. Finally, the developed method was applied to cosmetic and environmental samples and quantitative results were obtained in recovery experiments.

Phenolic compounds are secondary metabolites that are found ubiquitously in plants, fruits, and vegetables. Many studies have shown that regular consumption of these compounds could have a positive effect on our health. The aim of this study was to compare the phytochemical contents of the water extracts from three different plants used as folk remedies in Turkey: Aesculus hippocastanum, Olea europaea, and Hypericum perforatum. A liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS) analysis was performed to explore the phenolic profiles. The biological activities of these extracts were also evaluated in terms of their antioxidant activities (2,2-diphenyl-1-picrylhydrazyl DPPH, 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid ABTS, Ferric Reducing Antioxidant Power Assay FRAP, cupric ion reducing antioxidant capacity CUPRAC, &beta, carotene, phosphomolybdenum, and metal chelating) and enzyme inhibitory properties (against acetylcholinesterase, butyrylcholinesterase, and tyrosinase). The aqueous extract of H. perforatum showed the highest levels of total phenolic, flavonoid, and saponin contents. Protocatechuic acid, vanillic acid, verbascoside, hesperidin, hyperoside, apigenin 7-hexosides, and quercetin were the most common compounds found in this species. The results confirm that A. hippocastanum, O. europaea, and H. perforatum represent a potential source of natural-derived molecules with positive properties that could be used as valid starting point for new food supplements, and drugs in the pharmaceutical, cosmetic, and food industries.

This paper reports a novel fabric phase sorptive extraction-high performance liquid chromatography-photodiode array detection (FPSE-HPLC-PDA) method for the simultaneous extraction and analysis of three drug residues (ciprofloxacin, sulfasalazine, and cortisone) in human whole blood, plasma, and urine samples, generally administered in human patients to treat inflammatory bowel disease (IBD). The drugs of interest were well resolved using a Luna C18 column (250 mm × 4.6 mm; 5 μm particle size) in gradient elution mode within 20 min. The analytical method was optimized and validated in the range 0.05–10 μg/mL for whole blood, 0.25–10 μg/mL for human plasma, and 0.10–10 μg/mL for human urine. Blank human whole blood, plasma, and urine were used as the sample matrix for the method development and validation; while methyl-p-hydroxybenzoate was used as the internal standard (IS). Weighted-matrix matched standard calibration curves showed a good linearity up to a concentration of 10 μg/mL. The intra- and inter-day accuracy values (precision and trueness) were found in the range from −10.9% to 12.3%, and the performances of the validated FPSE-HPLC-PDA were further tested on real IBD patient samples. This is the first FPSE procedure applied simultaneously to whole blood, plasma, and urine samples for the determination of residual IBD drugs, which possess a wide range of polarity (logP values ranging from 2.30 for Ciprofloxacin, to 1.66 for Cortisone, and 2.92 for Sulfasalazine). The new approach exhibits high potential for immediate adoptation as a rapid, robust and green analytical tool for future clinical and pharmaceutical applications.

Capparis spinose L. also known as Caper is of great significance as a traditional medicinal food plant. The present work was targeted on the determination of chemical composition, pharmacological properties, and in-vitro toxicity of methanol and dichloromethane (DCM) extracts of different parts of C. spinosa. Chemical composition was established by determining total bioactive contents and via UHPLC-MS secondary metabolites profiling. For determination of biological activities, antioxidant capacity was determined through DPPH, ABTS, CUPRAC, FRAP, phosphomolybdenum, and metal chelating assays while enzyme inhibition against cholinesterase, tyrosinase, α-amylase and α-glucosidase were also tested. All the extracts were also tested for toxicity against two breast cell lines. The methanolic extracts were found to contain highest total phenolic and flavonoids which is correlated with their significant radical scavenging, cholinesterase, tyrosinase and glucosidase inhibition potential. Whereas DCM extracts showed significant activity for reducing power, phosphomolybdenum, metal chelation, tyrosinase, and α-amylase inhibition activities. The secondary metabolites profiling of both methanolic extracts exposed the presence of 21 different secondary metabolites belonging to glucosinolate, alkaloid, flavonoid, phenol, triterpene, and alkaloid derivatives. The present results tend to validate folklore uses of C. spinose and indicate this plant to be used as a potent source of designing novel bioactive compounds.

The reported method aims to be a powerful aid for the simultaneous determination of tetrahydrocannabinol (THC), cannabidiol (CBD), cannabinol (CBN), cannabigerol (CBG), tetrahydrocannabinolic acid (THCA), cannabidiolic acid (CBDA), and tetrahydrocannabivarin (THCV) in oily based preparations. The chromatographic separation was carried out using an Hypersil Gold PFP (50 × 2.1 mm, 1.9 μm) column, using H2O + 2 mM ammonium formate + 0.2 % formic acid (M1) and Methanol + 2 mM ammonium formate + 0.2 % formic acid (M2) as mobile phases. The flow rate was set 0.4 mL/min. Specifically, this method was validated in terms of linearity, limit of detections and quantifications (LODs and LOQs), accuracy (precision and trueness, both intra and interday), selectivity, and matrix effects. This procedure allowed quantifying seven phytocannabinoids in less than 10 min. The validated method shows a good linearity within the range 0.25-1000 ng/mL, while precision and trueness (intra- and inter-day) were below13.25 % and 7.59 %, respectively. Regarding the matrix effect, the method satisfies all the requirements, except for the THC and THCV, where it reaches about 120 %. This element does not affect the method performances as it has been observed that this value is constant and reproducible and therefore does not involve errors in the quantitative analysis. The method was tested and applied on more 70 different oily based preparations. Furthermore, starting from four different cannabis cultivar (FM2, Bedrolite, Bedrocan, and Bediol), it allowed to evaluate the reproducibility of the magistrali preparations. The real samples, in fact, derive from different local pharmacies, and were analyzed by the accredited UNI CEI EN ISO/IEC 17025:2018, Pharmatoxicology Laboratory (ACCREDIA, lab n. 2274 ASLPE, accreditation number 1822 L), accordingly to the current regulations.

Introduction The analysis of plant and herbal samples is a challenging task for analytical chemists due to the complexity of the matrix combined with the low concentration of analytes. In recent years different liquid-phase microextraction (LPME) techniques coupled with a variety of analytical equipment have been developed for the determination of both organic and inorganic analytes. Objective Over the past few years, the number of research papers in this field has shown a markedly growing tendency. Therefore, the purpose of this review paper is to summarise and critically evaluate research articles focused on the application of LPME techniques for the analysis of plant and herbal samples. Results Due to the complex nature of the samples, the direct application of LPME techniques to the analysis of plants has not often been done. LPME techniques as well as their modalities have been commonly applied in combination with other pretreatment techniques, including a solid-liquid extraction technique supported by mechanical agitation or auxiliary energies for plant analysis. Applications and the most important parameters are summarised in the tables. Conclusion This review summarises the application of the LPME procedure and shows the major benefits of LPME, such as the low volume of solvents used, high enrichment factor, simplicity of operation and wide selection of applicable detection techniques. We can expect further development of microextraction analytical methods that focus on direct sample analysis with the application of green extraction solvents while fully automating procedures for the analysis of plant materials.

A new liquid chromatography-tandem mass spectrometry (LC-MS/MS) confirmation method for the direct analysis of 17 drugs starting from 200μL of diluted oral fluid (OF), in a single chromatographic run, was developed and validated. Cocaine, benzoylecgonine (BEG), cocaethylene, Δ-9-tetrahydrocannabinol (Δ-9-THC), buprenorphine, 6-acetylmorphine (6AM), morphine, codeine, methadone, 2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine (EDDP), amphetamine, methamphetamine, 3,4-methylenedioxymethamphetamine (MDMA), methylenedioxyamphetamine (MDA), 3,4-Methylenedioxy-N-ethylamphetamine (MDE), ketamine, N-methyl-1-(1,3-benzodioxol-5-yl)-2-butanamine (MBDB) were determined in a chromatographic run of 12min only with no sample pre-treatment, after the addition of 15 different internal standards (ISs). The method met all requirements in terms of linearity, accuracy (precision and trueness), recovery, and stability requested by FDA guidelines. Carry-over and interferences were negligible, as well as the matrix effects. LLOQs are below the limits defined by European guidelines and Italian national laws. The original oral fluid collections are stable at least six months at -20°C and one week at +4°C.

A fast off-line FPSE-HPLC-PDA method has been reported that allows simultaneous clean up and determination of six non-steroidal anti-inflammatory drugs (NSAIDs) in saliva samples from healthy volunteers. Particularly, furprofen, indoprofen, ketoprofen, fenbufen, flurbiprofen, and ibuprofen were chromatographically resolved. Benzyl paraben was chosen as the internal standard (BzPB, IS). These target compounds were successfully extracted from human saliva using fabric phase sorptive extraction (FPSE) and then analysed in the liquid chromatographic system by means of a short analytical column (Symmetry C18, 75 × 4.6 mm, 3.5 µm) using acetonitrile (AcN) and phosphate buffer (PBS, 30 mM; pH = 2.5) as the mobile phases. The method, validated through the calculation of all analytical parameters in accordance of International Guidelines, was applied to real saliva sample analysis collected from informed volunteers. The proposed approach that included the use of sol–gel polytetrahydrofuran (sol–gel PTHF) sorbent immobilized on cellulose support and C18 stationary phase used in HPLC, showed high potential as a fast tool for future clinical and forensic applications. The herein reported results encourage potential future application of FPSE in the forensic field. Furthermore, the FPSE membrane was tested in dried saliva spot mode (DSS) in order to check its potential use as a sampling device, also for forensic applications.

Monophenols are widely present in a variety of foods including dairy products, fruits, coffee, rice, cereals, wines, and beers. They are primarily originated from plant materials and they are found either in the form of monophenol or their precursor. One major source of monophenols is wood. Monophenols exhibit a number of important physiological activities such as antioxidant, antimicrobial, antiviral, and antiinflammatory. In addition to their biological activities, monophenols play key roles in the flavor of foods. Some monophenols (thymol, eugenol, guaiacol, vanillin, 4-hydroxybenzaldehyde, etc.) are also known as the contributors to off-flavor. However, monophenols have not received much attention yet in assessing their exact roles in terms of biological activity as well as their role as flavor in foods and beverages compared to other plant-based natural compounds.

Monoterpenes and monoterpenoids represent a set of chemicals widely diffused in plants whose basic structure consists of two linked isoprene units. In addition to their abundance in plants, monoterpenes and monoterpenoids constitute approximately 90% of essential oils and demonstrate a wide diversity in their chemical structures and biological activities. Many members of monoterpenes and monoterpenoids family exert interesting pharmacological function such as antibacterial, hypotensive, antiinflammatory, and antipruritic characteristics. Their abundance and low cost have favorably positioned them as flavors and fragrances. One major consumption of these compounds is in nutraceutical formulations. The current chapters describe the phytochemistry of representative family members. The biological activities and interactions have been discussed. The techniques of extraction, purification, and fractionation of these compounds have also been covered. Analytical instruments primarily used for identification and quantification of monoterpenes and monoterpenoids are also briefly reviewed.
