Arsenic (As) is a toxic metalloid present in high levels in diverse regions of Argentina. The aim of this study was to determine acute As-mediated toxicity in two different populations of autochthonous Hyalella curvispina amphipods from a reference site (LB) and an agricultural one (FO) within North Patagonia Argentina. Previously, both populations exhibited significant differences in pesticide susceptibility. Lab assays were performed to determine acute lethal concentrations, as well as some biochemical parameters. Lethal concentration (LC 50 ) values obtained after 48 and 96 hr As exposure were not significantly different between these populations, although FO amphipods appeared slightly less susceptible. LC 50 -48 hr values were 3.33 and 3.92 mg/L As, while LC 50 -96 hr values were 1.76 and 2.14 mg/L As for LB and FO amphipods. The no observed effect concentration (NOEC) values were 0.5 mg/L As. Cholinesterase (ChE) activity was significantly diminished by As acute exposure (0.5-1.5 mg/L As), indicative of a significant neurotoxic action for this metalloid in both amphipod populations. Activities of catalase (CAT) and glutathione S-transferase (GST) and levels of reduced glutathione (GSH) were differentially altered following As exposure. CAT activity was increased after 96 hr As exposure. GST activity and GSH levels were significantly elevated followed by either a decrease or a return to control values after 96 hr treatment. However, additional studies are necessary to understand the mechanisms underlying the As-mediated oxidative effects in H. curvispina . Our findings suggest that measurement of ChE activity in H. curvispina amphipods might serve as a useful biomarker of As exposure and effect.

During the last years, a carbaryl insecticide was extensively applied in the valley of Río Negro and Neuquén, North Patagonia Argentina, to manage codling moths (Cydia pomonella), the main pest of pear and apple trees. In this study carbaryl susceptibility and B-esterase activity from both insecticide-exposed and non-exposed field populations of amphipods Hyalella curvispina were studied. Two subpopulations, one susceptible to carbaryl (LC 50 =213±7.5μg/L carbaryl) and one resistant to it (LC 50 =14,663±2379μg/L carbaryl), were found in the agricultural area selected in this study. Both populations were, in turn, more resistant to carbaryl than the population from a pristine area (LC 50 =11.31±2.27μg/L carbaryl). The in vivo 48h-IC 50 values for cholinesterase (ChE) were close to the corresponding 48h-LC 50 values as determined for the non-exposed population (IC 50 =7.16±0.86μg/L carbaryl) and for the susceptible subpopulation from the insecticide-exposed site (IC 50 =193±99μg/L carbaryl). Carbaryl exposure of the amphipods from the agricultural area mentioned above produced a significant decrease of carboxylesterase (CabE) activity, at a sublethal concentration (10μg/L) that was not able to significantly inhibit ChE, thereby showing a protective role of CabE and its usefulness as early biomarker. However, at lethal concentrations the inhibition of ChE activity was higher than that of CabE. On the other hand, CabE of amphipods from the pristine site was less sensitive to carbaryl than ChE, suggesting a different participation of CabE in ChE protection in the susceptible population of H. curvispina. Pulse exposure to carbaryl for 2h caused a significant inhibition of ChE in amphipods from both populations, with a fast recovery as expected for a carbamate insecticide. In conclusion, we proved that amphipods from the said agricultural area have developed resistance to carbaryl and showed the presence of two subpopulations with a different response to the insecticide. We evaluated the acute toxicity and biochemical effects of the organophosphorus pesticide azinphos methyl (AM) in the amphipod Hyalella curvispina that inhabits ponds and irrigation channels of an intensive fruit-producing region in Rio Negro and Neuquén valley, North Patagonia, Argentina. The analysis by nonlinear regression of data from the 96 h-acute toxicity tests indicated the coexistence of two subpopulations of H. curvispina with different susceptibilities to AM. The 96 h-LC₅₀ for the resistant subpopulation (166 ± 56 μg/L) was 216-fold higher than the 96h-LC₅₀ value for the susceptible one (0.77 ± 1.33 μg/L).The two subpopulations could not be distinguished based on the biochemical measurements in control amphipods. Cholinesterase activity was significantly inhibited in AM-exposed amphipods in a concentration-dependent manner. The IC₅₀ value obtained after 96 h of exposure (2.18 ± 1.95 μg/L) was significantly lower than the 48 h-IC₅₀ value (29.6 ± 17.4 μg/L). Carboxylesterase activity was significantly inhibited after 48 h of exposure to 12.5 and 62.5 μg/L AM (inhibition, 51%). This enzyme was thus able to protect cholinesterase from inhibition at 12.5 μg/L AM. Reduced glutathione and catalase showed a significant increase after 24 h of exposure as an adaptive response to AM, whereas glutathione S-transferase activity was not significantly modified. The analysis of species sensitivity distribution showed that both subpopulations of H. curvispina were more tolerant to AM than most amphipod species, and that the susceptible subpopulation was more sensitive to AM than the other local aquatic species analyzed. The maximum concentration of AM in drainage water within the fruit-producing area reported by other studies would affect most of the amphipod species (99%) and also a 44% of local aquatic ones. The Río Negro and Neuquén Valley is the most important apple and pear growing region in Argentina. Cydia pomonella L. (Lepidoptera: Tortricidae), the main fruit-tree pest is being controlled by azinphosmethyl (AzMe), acetamiprid (Acet), and thiacloprid (Thia) among other insecticides. The objective of this study was to evaluate the response of neonate larvae of codling moth to these three insecticides and on the role of cytochrome P450 monooxygenase in their toxicity. All field populations presented significantly lower mortality to a discriminating concentration (DC) of Acet and AzMe. In addition, 13 of the 14 populations showed significantly lower mortality to DC of Thia. Most of the field populations (71%) showed significantly higher 7-ethoxycoumarine O-deethylase activity compared with the laboratory-susceptible strain. While positive significant correlation (gamma = 0.59) was found between Thia and AzMe mortalities at the DC level, no significant correlations were detected between Acet and Thia (gamma = 0.35) or Acet and AzMe (gamma = 0.12). However, Acet and Thia mortalities were significantly correlated to the percentage of individuals exhibiting 7-ethoxy-coumarine O-deethylase activity activities higher than the mean upper 95% confidence limit of the susceptible strain (gamma = -0.52 and gamma = -0.63, respectively).

Background: Resistance to insecticides has been related to application history, genetic factors of the pest and the dynamic within the treated area. The aim of this study was to assess the geographic variation in azinphos-methyl response and the role of esterase and cytochrome P450 monooxygenase enzymes in codling moth populations collected within different areas of the Río Negro and Neuquén Valley, Argentina., Results: Diapausing field-collected populations showed resistance ratios at the LC(50) that were 0.7-8.7 times higher than that of the susceptible strain. Mean esterase (EST) and cytochrome P450 monooxygenase activities (expressed as α-N min(-1) mg(-1) prot(-1) and pg 7-OHC insect(-1) min(-1) respectively) were significantly correlated with LD(50) values from the field-collected populations. In addition, azinphos-methyl response was associated with the geographic area where the insect population was collected: populations from isolated and more recent productive areas presented significantly lower resistance ratios in comparison with populations from older and more intensive productive areas., Conclusion: The populations assayed presented different resistance levels to azinphos-methyl. The response was highly correlated with the orchard's geographic location. Black flies, a non-target species of the insecticides used in fruit production, represent a severe medical and veterinary problem. Large increases in the level of resistance to the pyrethroids fenvalerate (more than 355-fold) and deltamethrin (162-fold) and a small increase in resistance to the organophosphate azinphos methyl (2-fold) were observed between 1996-2008 in black fly larvae under insecticide pressure. Eventually, no change or a slight variation in insecticide resistance was followed by a subsequent increase in resistance. The evolution of pesticide resistance in a field population is a complex and stepwise process that is influenced by several factors, the most significant of which is the insecticide selection pressure, such as the dose and frequency of application. The variation in insecticide susceptibility within a black fly population in the productive area may be related to changes in fruit-pest control. The frequency of individuals with esterase activities higher than the maximum value determined in the susceptible population increased consistently over the sampling period. However, the insecticide resistance was not attributed to glutathione S-transferase activity. In conclusion, esterase activity in black flies from the productive area is one mechanism underlying the high levels of resistance to pyrethroids, which have been recently used infrequently. These enzymes may be reselected by currently used pesticides and enhance the resistance to these insecticides.

Amphibian embryos are naturally exposed to prooxidant conditions throughout their development. Environmental exposure to contaminants may affect their capacity to respond to challenging conditions, to progress in a normal ontogenesis, and finally to survive and succeed in completing metamorphosis. We studied the effects of the exposure to two anticholinesterase agents, the carbamate carbaryl and the organophosphate azinphos methyl, on the antioxidant defenses of developing embryos of the toad Rhinella (Bufo) arenarum. Reduced glutathione (GSH) levels were increased early by carbaryl, but were decreased by both pesticides at the end of embryonic development. The GSH-dependent enzymes glutathione reductase and glutathione peroxidases showed oscillating activity patterns that could be attributed to an induction of activity in response to oxidative stress and inactivation by excess of reactive oxygen species. Glutathione-S-transferases, which may participate in the conjugation of lipid peroxide products in addition to pesticide detoxification, showed an increase of activity at the beginning and at the end of development. Catalase also showed variations in the activity suggesting, successively, induction and inactivation in response to pesticide exposure-induced oxidative stress. Superoxide dismutase activity was increased by carbaryl and transiently decreased by azinphos methyl exposure. Judging from the depletion in GSH levels and glutathione reductase inhibition at the end of embryonic development, the oxidative stress caused by azinphos methyl seemed to be greater than that caused by carbaryl, which might be in turn related with a higher number of developmental alterations caused by the organophosphate. GSH content is a good biomarker of oxidative stress in the developing embryos exposed to pesticides. The antioxidant enzymes are in turn revealing the balance between their protective capacity and the oxidative damage to the enzyme molecules, decreasing their activity.

The organophosphorous compound azinphosmethyl (AzMe) is applied extensively in northern Patagonia (southern Argentina) to manage codling moths (Cydia pomonella). This area is irrigated by fast-flowing channels that provide a favorable habitat for many species, including amphipods (Hyalella curvispina) and a field-mixed population of black flies (Simulium bonaerense, Simulium wolffhuegeli, and Simulium nigristrigatum). In the present study, AzMe susceptibility and carboxylesterase (CarbE) activity from both insecticide-exposed and nonexposed field populations were studied. The median lethal dose determined in codling moths from an insecticide-treated orchard was significantly higher (3.48 microg/insect) than that observed in those from an untreated orchard (0.69 microg/insect). Similarly, the median lethal concentration (LC50) determined in black flies collected from the treated area (0.021 mg/L) was significantly higher than that recorded in those from the untreated site (0.011 mg/L). For amphipods, both a subpopulation susceptible to AzMe (LC50, 1.83 microg/L) and a resistant one (LC50, 390 microg/L) were found in the treated area. Both subpopulations were more resistant to AzMe than the population from the untreated site (LC50, 0.43 microg/L). Significant differences (p < 0.001) in CarbE activities were observed between populations from pesticide-treated and untreated areas. Mean activities +/- standard deviation from treated and untreated sites were 0.21 +/- 0.16 and 0.016 +/- 0.008 micromol/min/mg protein, respectively, for codling moths; 2.17 +/- 1.71 and 0.81 +/- 0.35 micromol/min/mg protein, respectively, for black flies; and 0.27 +/- 0.10 and 0.14 +/- 0.07 micromol/min/mg protein, respectively, for amphipods. The results suggest that enhanced CarbE activity is one of the mechanisms that provide AzMe resistance in H. curvispina, Simulium spp., and C. pomonella populations from the insecticide-treated areas.

We studied the effect of two insecticides azinphos methyl and carbaryl on two resident aquatic species (Oncorhynchus mykiss and Bufo arenarum). Juvenile trout and larval stages of toad were used for exposure and recovery assays. O. mykiss was more sensitive to azinphos methyl exposure than B. arenarum larvae, with a mean 96-h LC50 of 0.007 mg/l. Carbaryl is markedly less toxic than the organophosphate and the differences in potency, expressed as LC50, for both species varies only by five-fold. The relationship between cholinesterase (ChE) inhibition and lethality is not straightforward: O. mykiss survives with an almost complete inhibition of the brain enzyme when exposed to azinphos methyl and carbaryl. Their IC50 values are one or two orders of magnitude lower than the corresponding 96-h LC50 value. In B. arenarum larvae, the IC50 values for azinphos methyl and carbaryl are one half and one third of their 96-h LC50s, respectively. The time courses of enzyme inhibition and recovery also points out differences between both types of pesticides and species. Identifying the key features conferring species selectivity can be exploited to minimize the incidence and severity of intoxication of non-target organism. The data presented here highlight the necessity of including several species and endpoint analyses in the pesticide risk evaluations of aquatic ecosystems.

Amphibians are good bioindicators of environmental pollution due to their susceptibility to chemicals during their freshwater cycles. The effects of environmental pollution, together with changes in human activity and climate, have contributed to the reduction in the amphibian population over recent decades. However, toxicological research on amphibians has been rather scarce compared with that on other vertebrates. In this article we review the biochemical alterations underlying xenobiotic action and/or the detoxifying responses described for anuran species, with the aim of establishing possible biomarkers of effect. During the embryonic development of anurans, morphological and behavioural alterations are the effects most frequently cited in connection with chemical exposures. However, such biomarkers have a low sensitivity and are unspecific compared with biochemical alterations. Some primary pesticide targets, in particular cholinesterases for organophosphates and carbamates, have been evaluated. Esterases change seasonally and with the stage of development, and their sensitivity to anticholinesterase agents varies between species. Thus their use as biomarkers in anurans must be carefully analysed. Enzymes and endogenous compounds related to oxidative metabolism may also be used as biomarkers of effect. Glutathione pool, glutathione-S-transferases and metallothioneins respond in different ways to pesticides and heavy metals in anuran embryos and tadpoles. Mixed-function oxidases, in turn, are less developed in amphibians, and show a reduced induction in response to pesticide exposures. Endogenous polyamine levels are also proposed as good age-related biomarkers of damage. Finally, molecular biomarkers related to receptor binding, signal transduction and genetic response have gained increasing relevance, as they have been implicated in the fertilisation process and the earliest events in anuran development. The identification of transcription factors associated with the exposure of amphibians to xenobiotics as well as other alterations in hormone signalling appears highly promising. However, these techniques are likely to complement other methods. In conclusion, the use of several biomarkers with multiple endpoints is needed to link exposure to response and to provide better predictive tools for the environmental protection of endangered anuran species.

Mixed populations of the pest blackflies Simulium bonaerense Coscarón & Wygodzinsky, S. wolffhuegeli (Enderlein) and S. nigristrigatum Wygodzinsky & Coscarón (Diptera: Simuliidae) are highly resistant to DDT and pyrethroids in the Neuquén Valley, a fruit-growing area of northern Patagonia, Argentina. As these insecticides have not been used for blackfly control, resistance is attributed to exposure to agricultural insecticides. Pre-treatment with the synergist piperonyl butoxide (PBO) reduced both DDT and fenvalerate resistance, indicating that resistance was partly due to monooxygenase inhibition. Pre-treatment with the synergist tribufos to inhibit esterases slightly increased fenvalerate toxicity in the resistant population. Even so, biochemical studies indicated almost three-fold higher esterase activity in the resistant population, compared to the susceptible. Starch gel electrophoresis confirmed higher frequency and staining intensity of esterase electromorphs in the resistant population. Incomplete synergism against metabolic resistance indicates additional involvement of a non-metabolic resistance mechanism, such as target site insensitivity, assumed to be kdr-like in this case. Glutathione S-transferase activities were low and inconsistent, indicating no role in Simulium resistance. Knowing these spectra of insecticide activity and resistance mechanisms facilitates the choice of more effective products for Simulium control and permits better coordination with agrochemical operations.

Treatment with exogenous spermidine enhanced acute malathion toxicity during larval development of the toad Bufo arenarum Hensel. The polyamine was rapidly incorporated in the larvae with a subsequent metabolization to putrescine and spermine, which were excreted to the media. Endogenous polyamine levels were not changed by either spermidine or malathion treatments. However, 0.5-mM spermidine modified malathion uptake and bioavailability increasing the concentration of the xenobiotic in the larvae. The amount of reduced thiols was decreased by both compounds, but the depletion was insufficient to induce cytotoxicity. The oxidative degradation of polyamines competes for the pool of reduced glutathione used in the conjugation of malathion in the larvae, thus leading to the reported potentiation of toxicity. Our results suggest that exposure to thiols-depleting agents may induce alteration of organophosphate degradation in amphibian larvae.

Reduced glutathion (GSH) content and glutathione S-transferase (GSH S-transferase) activity were investigated in developing toad embryos exposed to parathion, malathion, lindane and dieldrin. The embryonic GSH content was reduced after 96 h of incubation with 20.00 ppm malathion and 2.00 ppm lindane. Parathion and dieldrin did not produce any change. A similar effect was obtained in advanced stages of development (6-days larvae), but only with malathion. No correlation between the decrease in GSH level and mortality or morphologic abnormalities was observed. The four pesticides increased the activity of GSH S-transferase indicating that the enzyme is susceptible to induction during early development. The higher effect depicted by malathion may be related with an enhanced conjugation of the pesticide. Both GSH decrease and GSHS-transferase induction modifies the cell redox status and may indirectly influence transcription and translation. The early expression of GST genes provides the embryo with a useful mechanism for the regulation of tolerance against chemical stress.
