The ecosystem services assessment plays one of the key role in the modern concept of sustainable development, including combat climate change and achievement carbon neutrality, since the value assessment of all benefits and risks from ecosystem services is the most visible for decision makers (business and government). Mangrove forests in developing countries, such as Sri Lanka, are important factor in achieving carbon neutrality. The purpose of present investigation was the economic assessment of climate regulation services on the example of the Madu Ganga wetlands (Sri Lanka). The carbon stock was calculated as the marginal cost of reducing carbon emissions, and the sequestration was assessed through the calculation of the carbon social cost or the marginal cost of damage. As a result, the high cost of carbon storage by the Madu Ganga wetlands was revealed - approximately, it amounted to $153,341,221. The cost of the ES for carbon sequestration was $2,153,424. The total cost of services to regulate the carbon cycle of the Madu Ganga wetlands was $155,494,645, or 0.18% of the nominal GDP of the island of Sri Lanka for 2021. The high value of the mangrove forests of Madu Ganga shows their crucial role in achieving carbon neutrality within the framework of the concept of sustainable development.

The article discusses the ecological significance of the phototrophic communities developing in entrance zone of grotto type caves as ecotones and refugiums. Species intrazonality and the dependence of cave species composition on geographic location were considered. The aim of present study was to identify similar caves of different regions based on the species composition of phototrophic communities using own data and data from literature. The empiric basis for investigation was present by phototropic species lists of own studies conducted by standard methods for phototropic species identification, as well as species lists from literature. The most frequently encountered phototrophs were species Chroococcus minutus and Chlorella vulgaris. Their occurrence in all the studied caves was revealed based on the taxonomic analysis. Thus, the intrazonal nature of the flora developed in grotto type caves is partially confirmed. Cluster analysis applying Euclidean distance was used to estimate similarity of the phototrophic communities developed in grotto caves of various genesis. Four clusters were identified: cretaceous caves of the Voronezh region; limestone grottoes of Ukraine and Abkhazia; monastic cells of Moldova and Ukraine; caves of Italy and Hungary. The dependence of the phototrophs biodiversity on the composition of the rocks and the genesis of the cavities was established. The geographical trend of photosynthetic species distribution in the grotto type caves was revealed.

Unique natural objects, such as the caves of the Gobustan National Historical and Artistic Preserve, are also of great cultural and historical value due to rock art and sites of ancient people. A favorable microclimate makes these habitats convenient for colonization by microbiota, including phototrophs. In arid regions with intense seasonal fluctuations of microclimatic parameters, the conditions for survival are the least favorable; therefore, it becomes especially important to determine the composition of communities that are the most adapted to specific conditions. This work aimed to identify the biodiversity of communities of caves and grottoes of the Gobustan Reserve. The studies were carried out in July 2019. Samples were analyzed for cyanobacteria and algae by microscopy and cultivation methods, microfungi were isolated by soil dilution, and the fouling glass method was also used. In total, 29 taxa of cyanobacteria and algae, 18 taxa of fungi, and 3 species of mosses were identified. The studied habitats were dominated by the algae Chlorella vulgaris, Aphanocapsa sp., and Stichococcus bacillaris; the subdominants were Jaaginema subtilissimum, Leptolyngbya tenuis, Chlorococcum minutum, and Humidophila contenta. Microfungi had the highest occurrence of Aspergillus niger, Aureobasidium pullulans, Alternaria alternata, and Talaromyces ruber. It was noted that cyanobacteria dominated in morphologically differentiated biofilms and green algae on the rocks. The greatest number of microfungi was found in the aphotic zone and bryophyte tufts. The dominance of green algae is atypical for most caves of other regions and may be associated with intense lighting of habitats. The absence of protonema is a consequence of the aridity and low moisture content of the substrates.

The present study aims to investigate the effectiveness of bioformulations based on endophytic fungi to control apple scab and Valsa canker disease in two orchards in the Aurès region (Algeria). In both orchards, the results showed that the treatment of senescent apple leaves by invert emulsions containing Trichoderma longibrachiatum and Chaetomium globosum harmed the ascogenesis of winter forms of Venturia inaequalis by reducing the number of ascospore-ejecting asci, the number of morphologically mature asci, and a considerable increase in the immature asci number. This antifungal activity was more essential in soil-incorporated leaves, showing the importance of the combination of treatments with cultural practices to efficiently control the apple scab disease. Furthermore, the disease incidence decreased by 52.63% and 50.68% in R’haouat and Bouhmama orchards, respectively. Moreover, the treatment of Valsa ceratosperma cankers with a biogel containing the endophytic yeast Metschnikowia sp. led to wound healing varying from 43.52% and 87.97% after 120 days but remained more considerable than conventional treatment with Folicur (tebuconazol). The current results open real opportunities concerning the implementation of eco-friendly and potent apple protection systems.

SnS2 and SnSe2 have recently been shown to have a wide range of applications in photonic and optoelectronic devices. However, because of incomplete knowledge about their optical characteristics, the use of SnS2 and SnSe2 in optical engineering remains challenging. Here, we addressed this problem by establishing SnS2 and SnSe2 linear and nonlinear optical properties in the broad (300–3300 nm) spectral range. Coupled with the first-principle calculations, our experimental study unveiled the full dielectric tensor of SnS2 and SnSe2. Furthermore, we established that SnS2 is a promising material for visible high refractive index nanophotonics. Meanwhile, SnSe2 demonstrates a stronger nonlinear response compared with SnS2. Our results create a solid ground for current and next-generation SnS2- and SnSe2-based devices.

Abstract. Aim. An analysis of species composition (phototrophic organisms and micromycetes) of epibioses communities of entrance area of monastic cells of the historical and archaeological complex of Old Orhei. Comparison of species composition of epibioses communities on the surface areas and in the cells. Comparison of phototrophic species composition of abovementioned communities with similar communities of the cult chalk caves of the Voronezh region and limestone grottoes of Podolsky Tovtry. Objects. The object of the investigation was the entrance area of the monastic cells of the rocky monastery "Dormition of God’s Mother", the reserve of Old Orhei (Moldova). Methods. Phototrophic species composition was determined by the methods of microscopy and inoculation in the culture medium. Mosses and lichens from epibioses communities were studied by traditional botanical methods. Micromycetes were studied by isolation methods on the Chapec-Dox selective medium. The abundance and occurrence of species in communities was determined. Results. Biodiversity of epibioses communities of hypogean habitats was revealed. It is shown that the species diversity on the surface is similar to the species composition in caves. The dominant species in the underground and surface epibioses communities were determined. The similarity of the species composition of the flora with caves of the Voronezh region and Ukraine was not revealed. Conclusions. Cyanobacteria predominate in the flora structure of the monastic cells of the rocky monastery "Dormition of God’s Mother". Micromycetes of epibioses communities are represented by widespread soil species. The species composition of cells is similar to the species composition on the surface. It can be assumed that the species composition of hypogean habitats is determined by a complex of conditions, among which the most important is the illumination, temperature and humidity, substrate quality and species composition on the surface.

Epithelial furrowing is a fundamental morphogenetic process during gastrulation, neurulation, and body shaping. A furrow often results from a fold that propagates along a line. How fold formation and propagation are controlled and driven is poorly understood. To shed light on this, we study the formation of the cephalic furrow, a fold that runs along the embryo dorsal-ventral axis during Drosophila gastrulation and the developmental role of which is still unknown. We provide evidence of its function and show that epithelial furrowing is initiated by a group of cells. This cellular cluster works as a pacemaker, triggering a bidirectional morphogenetic wave powered by actomyosin contractions and sustained by de novo medial apex-to-apex cell adhesion. The pacemaker's Cartesian position is under the crossed control of the anterior-posterior and dorsal-ventral gene patterning systems. Cell apical constriction driven by actomyosin contraction forces is a conserved mechanism during tissue folding in embryo development. While much is now understood of the molecular mechanism responsible for apical constriction and of the tissue-scale integration of the ensuing in-plane deformations, it is still not clear if apical actomyosin contraction forces are necessary or sufficient per se to drive tissue folding. To tackle this question, we use the Drosophila embryo model system that forms a furrow on the ventral side, initiating mesoderm internalization. Past computational models support the idea that cell apical contraction forces may not be sufficient and that active or passive cell apico-basal forces may be necessary to drive cell wedging leading to tissue furrowing. By using 3D computational modelling and in toto embryo image analysis and manipulation, we now challenge this idea and show that embryo-scale force balance at the tissue surface, rather than cell-autonomous shape changes, is necessary and sufficient to drive a buckling of the epithelial surface forming a furrow which propagates and initiates embryo gastrulation., (© 2022. Tissue elongation is known to be controlled by oriented cell division, elongation, migration and rearrangement. While these cellular processes have been extensively studied, new emerging supracellular mechanisms driving tissue extension have recently been unveiled. Tissue rotation and actomyosin contractions have been shown to be key processes driving Drosophila egg chamber elongation. First, egg chamber rotation facilitates the dorsal-ventral alignment of the extracellular matrix and of the cell basal actin fibers. Both fiber-like structures form supracellular networks constraining the egg growth in a polarized fashion thus working as 'molecular corsets'. Second, the supracellular actin fiber network, powered by myosin periodic oscillation, contracts anisotropically driving tissue extension along the egg anterior-posterior axis. During both processes, cellular and supracellular planar polarity provide a critical cue to control Drosophila egg chamber elongation. Actomyosin supracellular networks emerge during development and tissue repair. These cytoskeletal structures are able to generate large scale forces that can extensively remodel epithelia driving tissue buckling, closure and extension. How supracellular networks emerge, are controlled and mechanically work still remain elusive. During Drosophila oogenesis, the egg chamber elongates along the anterior-posterior axis. Here we show that a dorsal-ventral polarized supracellular F-actin network, running around the egg chamber on the basal side of follicle cells, emerges from polarized intercellular filopodia that radiate from basal stress fibers and extend penetrating neighboring cell cortexes. Filopodia can be mechanosensitive and function as cell-cell anchoring sites. The small GTPase Cdc42 governs the formation and distribution of intercellular filopodia and stress fibers in follicle cells. Finally, our study shows that a Cdc42-dependent supracellular cytoskeletal network provides a scaffold integrating local oscillatory actomyosin contractions at the tissue scale to drive global polarized forces and tissue elongation.

High-throughput screens allow us to understand how transcription factors trigger developmental processes, including cell specification. A major challenge is identification of their binding sites because feedback loops and homeostatic interactions may mask the direct impact of those factors in transcriptome analyses. Moreover, this approach dissects the downstream signaling cascades and facilitates identification of conserved transcriptional programs. Here we show the results and the validation of a DNA adenine methyltransferase identification (DamID) genome-wide screen that identifies the direct targets of Glide/Gcm, a potent transcription factor that controls glia, hemocyte, and tendon cell differentiation in Drosophila. The screen identifies many genes that had not been previously associated with Glide/Gcm and highlights three major signaling pathways interacting with Glide/Gcm: Notch, Hedgehog, and JAK/STAT, which all involve feedback loops. Furthermore, the screen identifies effector molecules that are necessary for cell-cell interactions during late developmental processes and/or in ontogeny. Typically, immunoglobulin (Ig) domain-containing proteins control cell adhesion and axonal navigation. This shows that early and transiently expressed fate determinants not only control other transcription factors that, in turn, implement a specific developmental program but also directly affect late developmental events and cell function. Finally, while the mammalian genome contains two orthologous Gcm genes, their function has been demonstrated in vertebrate-specific tissues, placenta, and parathyroid glands, begging questions on the evolutionary conservation of the Gcm cascade in higher organisms. Here we provide the first evidence for the conservation of Gcm direct targets in humans. In Drosophila, the transcription factor Gcm/Glide plays a key role in cell fate determination and cellular differentiation. In light of its crucial biological impact, major efforts have been put for analyzing its properties as master regulator, from both structural and functional points of view. However, the lack of efficient antibodies specific to the Gcm/Glide protein precluded thorough analyses of its regulation and activity in vivo. In order to relieve such restraints, we designed an epitope-tagging approach to "FLAG"-recognize and analyze the functional protein both in vitro (exogenous Gcm/Glide) and in vivo (endogenous protein). We here (i) reveal a tight interconnection between the small RNA and the Gcm/Glide pathways. AGO1 and miR-1 are Gcm/Glide targets whereas miR-279 negatively controls Gcm/Glide expression (ii) identify a novel cell population, peritracheal cells, expressing and requiring Gcm/Glide. Peritracheal cells are non-neuronal neurosecretory cells that are essential in ecdysis. In addition to emphasizing the importance of following the distribution and the activity of endogenous proteins in vivo, this study provides new insights and a novel frame to understand the Gcm/Glide biology., (Copyright © 2012 Elsevier Inc. The Gcm/Glide transcription factor is transiently expressed and required in the Drosophila nervous system. Threshold Gcm/Glide levels control the glial versus neuronal fate choice, and its perdurance triggers excessive gliogenesis, showing that its tight and dynamic regulation ensures the proper balance between neurons and glia. Here, we present a genetic screen for potential gcm/glide interactors and identify genes encoding chromatin factors of the Trithorax and of the Polycomb groups. These proteins maintain the heritable epigenetic state, among others, of HOX genes throughout development, but their regulatory role on transiently expressed genes remains elusive. Here we show that Polycomb negatively affects Gcm/Glide autoregulation, a positive feedback loop that allows timely accumulation of Gcm/Glide threshold levels. Such temporal fine-tuning of gene expression tightly controls gliogenesis. This work performed at the levels of individual cells reveals an undescribed mode of Polycomb action in the modulation of transiently expressed fate determinants and hence in the acquisition of specific cell identity in the nervous system., Competing Interests: The authors have declared that no competing interests exist.

The neural stem cells that give rise to the neural lineages of the brain can generate their progeny directly or through transit amplifying intermediate neural progenitor cells (INPs). The INP-producing neural stem cells in Drosophila are called type II neuroblasts, and their neural progeny innervate the central complex, a prominent integrative brain center. Here we use genetic lineage tracing and clonal analysis to show that the INPs of these type II neuroblast lineages give rise to glial cells as well as neurons during postembryonic brain development. Our data indicate that two main types of INP lineages are generated, namely mixed neuronal/glial lineages and neuronal lineages. Genetic loss-of-function and gain-of-function experiments show that the gcm gene is necessary and sufficient for gliogenesis in these lineages. The INP-derived glial cells, like the INP-derived neuronal cells, make major contributions to the central complex. In postembryonic development, these INP-derived glial cells surround the entire developing central complex neuropile, and once the major compartments of the central complex are formed, they also delimit each of these compartments. During this process, the number of these glial cells in the central complex is increased markedly through local proliferation based on glial cell mitosis. Taken together, these findings uncover a novel and complex form of neurogliogenesis in Drosophila involving transit amplifying intermediate progenitors. Germline silencing of transposable elements is essential for the maintenance of genome integrity. Recent results indicate that this repression is largely achieved through a RNA silencing pathway that involves Piwi-interacting RNAs (piRNAs). However the repressive mechanisms are not well understood. To address this question, we used the possibility to disrupt the repression of the Drosophila I element retrotransposon by hybrid dysgenesis. We show here that the repression of the functional I elements that are located in euchromatin requires proteins of the piRNA pathway, and that the amount of ovarian I element piRNAs correlates with the strength of the repression in the female germline. Antisense RNAs, which are likely used to produce antisense piRNAs, are transcribed by heterochromatic defective I elements, but efficient production of these antisense small RNAs requires the presence in the genome of euchromatic functional I elements. Finally, we demonstrate that the piRNA-induced silencing of the functional I elements is at least partially posttranscriptional. In a repressive background, these elements are still transcribed, but some of their sense transcripts are kept in nurse cell nuclear foci together with those of the Doc retrotransposon. In the absence of I element piRNAs, either in dysgenic females or in mutants of the piRNA silencing pathway, sense I element transcripts are transported toward the oocyte where retrotransposition occurs. Our results indicate that piRNAs are involved in a posttranscriptional gene-silencing mechanism resulting in RNA nuclear accumulation.
