The stability of psychotropic substances representing various drug groups important from the perspective of forensic chemistry, including benzodiazepines, antidepressants, carbamazepine, cocaine, and their selected metabolites, was investigated for 1 month in two alternative biological matrices, vitreous humor and liver homogenate. Three different thermal storage conditions (−20, 4, and 20 °C) were tested. Liquid chromatography-mass spectrometry (LC-MS) analysis was preceded by an effective solid-phase microextraction (SPME) procedure. The results were statistically analyzed using one-way ANOVA to find significant concentration variations over time. The results obtained allowed for dividing the analytes into four groups: stable under all tested conditions, only at −20 and 4 °C, only at 20 °C, and overall unstable. Nordiazepam, venlafaxine, and cocaine and its metabolites turned out to be the most unstable substances, while fluoxetine showed the highest storage stability in both matrices. The SPME/LC-MS method was comprehensively evaluated according to the principles of white analytical chemistry (WAC), which reconcile the greenness and functionality of the method. A close to 100% whiteness score proves its sustainability and suitability for the intended application. Graphical abstract

Glucocorticoids (GCs) are known to play an important role in maintaining basal and stress-related homeostasis by interacting with endocrine mediators and prostaglandins (PGs). Although a growing body of evidence shows that GCs exert their regulatory action at a multitude of sites in the reproductive axis through corticosteroid receptors, little is known about the direct role of cortisol, an active form of GCs, in the equine endometrium. Thus, the study aimed to determine the effect of cortisol on PGF2α synthesis in the endometrial tissue and cells in vitro. In Exp.1, the immunolocalization and the expression of the glucocorticoid receptor (GCR) in the endometrium throughout the estrous cycle were established. In Exp. 2 and 3, the effects of cortisol on PGF2α secretion and transcripts associated with the arachidonic acid (AA) cascade in endometrial tissues, and cells were defined. Endometrial tissues obtained from the early, mid, and late luteal phases and the follicular phase of the estrous cycle were exposed to cortisol (100, 200, and 400 nM) for 24 h. Endometrial epithelial and stromal cells (early phase of estrous cycle) were exposed to cortisol (100 nM) for 24 h. Then, PGF2α secretion and transcripts associated with the AA cascade (PLA2G2A, PLA2G4A, PTGS2, and PGFS) were assessed. GCR was expressed in the cytoplasm and the nucleus in the luminal and glandular epithelium as well as in the stroma. Endometrial GCR protein abundance was up-regulated at the late luteal phase compared to the mid-luteal phase of the estrous cycle. Cortisol dose-dependently decreased PGF2α secretion, PLA2G2A and PLA2G4A transcripts in endometrial tissues. Additionally, cortisol treatment decreased PGF2α secretion from endometrial epithelial and stromal cells. Moreover, it affected PLA2G2A, PLA2G4A, and PTGS2 transcripts in endometrial stromal cells. These findings suggest that cortisol suppresses the synthesis of PGF2α by affecting the AA cascade in the equine endometrium during the estrous cycle.

We define a model for computing probabilities of right-nested conditionals in terms of graphs representing Markov chains. This is an extension of the model for simple conditionals from Wójtowicz and Wójtowicz (Erkenntnis, 1–35. 10.1007/s10670-019-00144-z, 2019). The model makes it possible to give a formal yet simple description of different interpretations of right-nested conditionals and to compute their probabilities in a mathematically rigorous way. In this study we focus on the problem of the probabilities of conditionals; we do not discuss questions concerning logical and metalogical issues such as setting up an axiomatic framework, inference rules, defining semantics, proving completeness, soundness etc. Our theory is motivated by the possible-worlds approach (the direct formal inspiration is the Stalnaker Bernoulli models); however, our model is generally more flexible. In the paper we focus on right-nested conditionals, discussing them in detail. The graph model makes it possible to account in a unified way for both shallow and deep interpretations of right-nested conditionals (the former being typical of Stalnaker Bernoulli spaces, the latter of McGee’s and Kaufmann’s causal Stalnaker Bernoulli models). In particular, we discuss the status of the Import-Export Principle and PCCP. We briefly discuss some methodological constraints on admissible models and analyze our model with respect to them. The study also illustrates the general problem of finding formal explications of philosophically important notions and applying mathematical methods in analyzing philosophical issues.

This retrospective cohort study comprehensively evaluates cardiovascular anomalies (CVAs) and associated extracardiac structural malformations (ECMs) among 1005 fetuses undergoing invasive prenatal testing at a single tertiary Polish center in the context of chromosomal aberrations detected in them by array comparative genomic hybridization (aCGH) and G-band karyotyping. The results of our study show that CVAs are among the most common malformations detected in fetuses undergoing invasive prenatal testing, as they affected 20% of all cases seen in our department. Septal defects predominated among fetuses with numerical aberrations, while conotruncal defects were the most common findings among fetuses with pathogenic copy number variants (CNVs). In 61% of cases, CVAs were associated with ECMs (the diagnosis was confirmed postnatally or in cases of pregnancy termination by means of autopsy). The most common ECMs were anomalies of the face and neck, followed by skeletal defects. In total, pathogenic chromosomal aberrations were found in 47.5% of CVAs cases, including 38.6% with numerical chromosomal aberrations. Pathogenic CNVs accounted for 14.5% of cases with CVAs and normal karyotype. Thus, our study highlights the importance of assessing the anatomy of the fetus, and of the genetic testing (preferably aCGH) that should be offered in all CVA and ECM cases.

Vitreous humor (VH) is an alternative biological matrix with a great advantage of longer availability for analysis due to the lack of many enzymes. The use of VH in forensic toxicology may have an added benefit, however, this application requires rapid, simple, non-destructive, and relatively portable analytical analysis methods. These requirements may be met by Fourier transform infrared spectroscopy technique (FT-IR) equipped with attenuated total reflection accessory (ATR). FT-IR spectra of vitreous humor samples, deposited on glass slides, were collected and subsequent chemometric data analysis by means of Hierarchical Cluster Analysis and Principal Component Analysis was conducted. Differences between animal and human VH samples and human VH samples stored for diverse periods of time were detected. A kinetic study of changes in the VH composition up to 2 weeks showed the distinction of FT-IR spectra collected on the 1st and 14th day of storage. In addition, data obtained for the most recent human vitreous humor samples—collected 3 and 2 years before the study, presented successful discrimination of all time points studied. According to the PCCP thesis, the probability of a conditional A → C is the conditional probability P(C|A). This claim is undermined by Lewis’ triviality results, which purport to show that apart from trivial cases, PCCP is not true. In the present article we show that the only rational, “Dutch Book-resistant” extension of the agent’s beliefs concerning non-conditional sentences A and C to the conditional A → C is by assuming that P(A → C) = P(C|A) (i.e., in accord with PCCP). In other cases a diachronic Dutch Book against the agent can be constructed. There is a tension between our findings and Lewis’ results, which needs to be explained. Therefore, we present a probability space which corresponds in a natural way to the diachronic Dutch Book—and which allows the conditional A → C to be interpreted as an event in a mathematically sound way. It also allows to formalize the notion of conditionalizing A → C on ¬C which plays a crucial role in Lewis’ proof. Our conclusion is that Lewis’ proof is circular, so it cannot be considered to be a sound argument against PCCP.

We analyze radio continuum emission of early-type galaxies with dynamical measurements of central super-massive black hole (SMBH) masses, and well-characterized large-scale environments, but regardless on the exact level of the nuclear activity. The 1.4 GHz radio fluxes collected with $\sim$arcmin resolution for 62 nearby targets (distances $\lesssim$ 153 Mpc), correspond to low and very low monochromatic luminosities $L_{\rm r} \sim 10^{35} - 10^{41}$ erg s$^{-1}$. We quantify possible correlations between the radio properties with the main parameters of supermassive black holes, host galaxies, and hot gaseous halos, finding a general bimodality in the radio luminosity distribution, with the borderline between ``radio-bright'' and ``radio-dim'' populations $\log L_{\rm r} / L_{\rm Edd} \simeq -8.5$. We analyze the far-infrared data for the targets, finding that all radio-bright sources, and over a half of radio-dim ones, are over-luminous in radio with respect to the far-infrared--radio correlation. High-resolution radio maps reveal that the overwhelming majority of radio-dim sources are unresolved on arcsecond scale, while the bulk of radio-bright sources display extended jets and lobes of low- and intermediate-power radio galaxies; those jets dominate radio emission of radio-bright objects. Regarding the origin of the radio emission of radio-dim sources, we discuss the two main possibilities. One is the ADAF model, in which the radio and the nuclear X-ray radiative outputs at very low accretion rates, are both dominated by unresolved jets. The other possibility is that the radio-dim sources, unlike the radio-bright ones, are characterized by low values of SMBH spins, so that their radio emission is not related to the jets, but instead is due to a combination of starforming processes and past nuclear outbursts., Comment: revised version accepted for publication in the Astrophysical Journal

OBJECTIVES To analyze near-term cerebroplacental Doppler, heart morphology, and neonatal biometry in isolated hypoplastic left heart syndrome (HLHS) relative to healthy controls. METHODS This retrospective study included 55 fetuses with HLHS (29 with mitral valve stenosis [MS]/aortic valve atresia [AA], 14 with MS/aortic valve stenosis, and 12 with mitral valve atresia [MA]/[AA]) diagnosed prenatally between 2010 and 2019 at 2 referral centers and 101 healthy controls. Ultrasound assessment included umbilical artery (UA), middle cerebral artery (MCA) pulsatility index (PI), and cerebroplacental ratio (CPR), with neonatal weight, length, head circumference (HC), Apgar score, and UA pH measured at birth. RESULTS In total, 32.7% of HLHS fetuses had abnormal MCA-PI and UA-PI, and 38.2% had CPRs below the fifth percentile before birth. All tested Doppler parameters differed from those of the healthy controls (P ≤ .01). Birth weight and length were comparable between HLHS and control fetuses, whereas birth HCs were smaller in the HLHS group than in the control group (P = .018). In both groups, increased UA-PI correlated with lower birth weight, but only HLHS fetuses with UA-PI > the 95th percentile had a lower median HC at birth than those with normal UA-PI (P = .045). The median UA-PI percentile was higher in fetuses with MA than in fetuses with MS (P = .015). The ascending aortic diameter correlated with birth weight (P = .036) and birth length (P = .039). CONCLUSION Abnormal cerebroplacental hemodynamics are evident in a high percentage of near-term fetuses with HLHS, and increased placental resistance may contribute to birth weight and HC. Moreover, heart morphology may impact placental circulation and neonatal biometry.

Objectives: Local and international organizations recommend folic acid (FA) supplementation in the periconceptional period. This study aimed to analyse the prevalence of periconceptional supplementation with FA in women at high risk of fetal anomalies refferred for first trimester screening. Material and methods: Our analysis involved 1,455 women at high risk of fetal anomalies refferred for first trimester screening. FA supplementation was assessed by face-to-face interviews conducted by doctors performing first trimester screening for aneuploidy. Results: FA supplementation before pregnancy was reported by 46.8% of the women and during the first trimester by 57.2% of those studied. Women used FA supplementation more frequently if they had a history of at least one miscarriage (OR 2.2, 95% CI 1.70–2.83; p < 0.001), a history of assissted reproductive techniques (OR 2.25, 95% CI 1.18–4.31; p = 0.014), or were aged between 30 and 34 (OR 2.87, 95% CI 1.47–5.58; p = 0.002). Among 122 women with a history of fetal defects only 50% confirmed FA supplementation before pregnancy and 62.2% during pregnancy (p = 0.488). A similar frequency of FA supplementation was noted among women with epilepsy, diabetes, and hypertension. Less frequent taking of FA was noted among women at least third and subsequent pregnancies (p < 0.001). In the current pregnancy, neural tube defects (NTDs) were less frequent by 86% in the group of women with FA supplementation than in the non-supplementation group (1 case vs 6 cases, respectively) and for other fetal defects by 62.5% (24 vs 40 cases, respectively). Conclusions: We found an unsatisfactory compliance with recommendations for the use of folic acid supplementation during periconceptional period among women at high risk of fetal defects and folate deficiency, that could have negative effects on the health of child and mother. The study results show the need to increase the awareness of FA supplementation during periconceptional period especially in women with high risk of fetal anomalies.

Analyzing ultrasound images of relatively small fetal structures is a difficult task. This difficulty partic- ularly applies to diagnosing congenital defects of the fetus, including one of the most common defects, which is the cleft palate. To date, no methods have been developed for visualizing the fetal palate. There- fore, there is a need to improve the effectiveness of diagnostics in this area by developing appropriate image analysis and recognition methods using computer-based techniques. At the same time, relatively fast algorithms are being sought that can be a part of the software of an ultrasound device. The contribu- tion of the paper consists in defining a new computer method satisfying all the requirements mentioned above. The method applies syntactic pattern recognition approach to the analysis of the image of the fetus’s palate. It is based on extracting of a sequence of images on multiple binarization thresholds (in the preprocessing phase), defining picture primitives on the basis of the histogram analysis, and applying a parser for a GDPLL( k ) string grammar (Generalized Dynamically Programmed LL( k ) grammar) for clas- sifying abnormalities (in the recognition phase). The implementation of the method is computationally efficient and it can be a helpful tool for supporting doctors in the diagnostic process (as it has been veri- fied in practice). The computer recognition of fetal defects on basis of the analysis of the structure of the fetus’s palate is a novel achievement. Such results have never been reported before.

The binding legal provisions related to the spending of funds from the EU budget regulate issues related to the management of EU finances, taking into account the specific nature of the spending process. Sound financial management of the European Union as well as shared management are principles strictly related to the proper management of funds coming from the EU budget by creating rules on which the system of their monitoring, control, audit and management was built. The primary purpose of the publication is to define the scope of meaning of the principle of sound management, and the principles of shared management in the management and control system of operational programs and attempt to set a framework for countries and EU bodies to create a system of spending funds from the EU budget. While discussing the subject and subject scope of the indicated principles, the dogmatic and legal method was used, as well as the legal and comparative degree. In addition, the analysis of selected judgments of the European Court of Justice has been used to verify the correctness of the theses.

We define a semantics for conditionals in terms of stochastic graphs which gives a straightforward and simple method of evaluating the probabilities of conditionals. It seems to be a good and useful method in the cases already discussed in the literature, and it can easily be extended to cover more complex situations. In particular, it allows us to describe several possible interpretations of the conditional (the global and the local interpretation, and generalizations of them) and to formalize some intuitively valid but formally incorrect considerations concerning the probabilities of conditionals under these two interpretations. It also yields a powerful method of handling more complex issues (such as nested conditionals). The stochastic graph semantics provides a satisfactory answer to Lewis’s arguments against the PC = CP principle, and defends important intuitions which connect the notion of probability of a conditional with the (standard) notion of conditional probability. It also illustrates the general problem of finding formal explications of philosophically important notions and applying mathematical methods in analyzing philosophical issues.

Tissue fibrosis is characterized by excessive deposition of extracellular matrix (ECM) components that result from the disruption of regulatory processes responsible for ECM synthesis, deposition, and remodeling. Fibrosis develops in response to a trigger or injury and can occur in nearly all organs of the body. Thus, fibrosis leads to severe pathological conditions that disrupt organ architecture and cause loss of function. It has been estimated that severe fibrotic disorders are responsible for up to one-third of deaths worldwide. Although intensive research on the development of new strategies for fibrosis treatment has been carried out, therapeutic approaches remain limited. Since stem cells, especially mesenchymal stem cells (MSCs), show remarkable self-renewal, differentiation, and immunomodulatory capacity, they have been intensively tested in preclinical studies and clinical trials as a potential tool to slow down the progression of fibrosis and improve the quality of life of patients with fibrotic disorders. In this review, we summarize in vitro studies, preclinical studies performed on animal models of human fibrotic diseases, and recent clinical trials on the efficacy of allogeneic and autologous stem cell applications in severe types of fibrosis that develop in lungs, liver, heart, kidney, uterus, and skin. Although the results of the studies seem to be encouraging, there are many aspects of cell-based therapy, including the cell source, dose, administration route and frequency, timing of delivery, and long-term safety, that remain open areas for future investigation. We also discuss the contemporary status, challenges, and future perspectives of stem cell transplantation for therapeutic options in fibrotic diseases as well as we present recent patents for stem cell-based therapies in organ fibrosis.

Elastin is one of the main structural matrix proteins of the arteries, lung, cartilage, elastic ligaments, brain vessels, and skin. These elastin fibers display incredible resilience and structural stability with long half-life. However, during some physiological and pathophysiological conditions, elastin is prone to proteolytic degradation and, due to the extremely low turnover rate, its degradation is practically an irreversible and irreparable phenomenon. As a result of elastin degradation, new peptides called elastin-derived peptides (EDPs) are formed. A growing body of evidence suggests that these peptides play an important role in the development of age-related vascular disease. They are also detected in the cerebrospinal fluid of healthy people, and their amount increases in patients after ischemic stroke. Recently, elastin-like polypeptides have been reported to induce overproduction of beta-amyloid in a model of Alzheimer's disease. Nevertheless, the role and mechanism of action of EDPs in the nervous system is largely unknown and limited to only a few studies. The article summarizes the current state of knowledge on the role of EDPs in the nervous system.

Blood is one of the most common body fluids discovered at crime scenes involving violent actions. It is one of the most important types of forensic evidence since it allows for the identification of the individual providing that there is a match with a known DNA profile. Determining the time since deposition (TSD) can assist investigators in establishing when the crime occurred or if a bloodstain present is actually related to the investigated event. To develop a forensically sound method for determining the TSD of a bloodstain, it is necessary to understand the underlying biochemical mechanisms occurring during aging. As biochemical processes occurring in blood are necessary for the continued survival of living organisms, they are important subjects of human biology and biomedicine and are well understood. However, the biochemistry of bloodstain aging ex vivo is primarily of interest to forensic scientists and has not yet been thoroughly researched. This preliminary study utilizes steady-state fluorescence spectroscopy to probe the changes in fluorescence properties of peripheral and menstrual blood up to 24-h post deposition. Peripheral and menstrual blood exhibited similar kinetic changes over time, assigned to the presence of the fluorophores: tryptophan, nicotinamide adenine dinucleotide (NADH), and flavins in both biological fluids. The biochemical mechanism of blood aging ex vivo is discussed.

It was the second pregnancy of an otherwise healthy married couple. The fetus (male) had detailed echocardiography monitoring in the second half of the pregnancy due to progression of cardiomegaly, and echocardiographic features of congestive heart failure. Marfan syndrome was suspected based on cardiac anomalies. For the first time, the rupture of aneurysm of aortic sinus Valsalva was documented. Despite transplacental treatment with digoxin there was fetal demise at the 34th week of gestation and postmortem newborn phenotype confirmed prenatal diagnosis. Marfan Syndrome is a rare genetic anomaly which can be diagnosed prenatally by detailed echocardiography, usually with bad prognosis (just opposite to “benign”case diagnosed later on in life span). The most common prenatal cardiac manifestations are cardiomegaly with signs of cardiac insufficiency. We present the case with new echocardiographic features.

Triclosan (TCS) is a well-known compound that can be found in disinfectants, personal care products. There is one publication concerning the involvement of PPARγ in the mechanism of action of TCS. It is known that activation of PPARγ regulates the expression of the NF-κB mediated inflammation by acting on nitric oxide synthase (NOS) genes. However, there are no studies demonstrating a relationship between the effects of TCS on the PPARγ signaling pathway, changes in NF-κB expression, and NOS isoform synthesis. Therefore, the aim of this study was to evaluate the effect of TCS on the expression of PPARγ, NF-κB, nNOS, iNOS, and eNOS in mouse neocortical neurons. In addition, the effects of co-administration of synthetic alpha-naphthoflavone (αNF) or beta-naphthoflavone (βNF) flavonoids and triclosan were investigated. Our results show that TCS alters PPARγ, NF-κB, iNOS, and eNOS expression in mouse neurons in vitro. After 48 h of exposure, TCS increased PPARγ expression and decreased NF-κB expression. Moreover, under the influence of TCS, the expression of iNOS was increased and at the same time the expression of nNOS was decreased, which was probably caused by high levels of ROS. The experiments have shown that both αNF and βNF are able to modulate the effects of TCS in primary cultures of mouse cortical neurons.

We present an analysis of the mid-infrared properties of the youngest radio galaxies, based on low-resolution data provided by the WISE satellite. We restrict our analysis to sources with available X-ray data that constitute the earliest phase of radio galaxy evolution, that is, those classified as gigahertz peaked spectrum and/or compact symmetric objects. We compare the MIR color distribution in our sample to that in the general population of local active galactic nuclei (AGN), in the population of evolved FR II radio galaxies, and also in the population of radio galaxies with recurrent jet activity. We conclude that the triggering of radio jets in AGN does not differentiate between elliptical hosts with substantially different fractions of young stars; instead there is a relationship between the jet duty cycle and the ongoing star formation.

The dynamical evolution and radiative properties of luminous radio galaxies and quasars of the FRII type, are well understood. As a result, through the use of detailed modeling of the observed radio emission of such sources, one can estimate various physical parameters of the systems, including the density of the ambient medium into which the radio structure evolves. This, however, requires rather comprehensive observational information, i.e. sampling the broad-band radio continua of the targets at several frequencies, and imaging their radio structures with high resolution. Such observations are, on the other hand, not always available, especially for high-redshift objects. Here we analyze the best-fit values of the source physical parameters, derived from an extensive modeling of the largest currently available sample of FRII radio sources, for which good-quality multi-wavelength radio flux measurements could be collected. In the analyzed dataset, we notice a significant and non-obvious correlation between the spectral index of the non-thermal radio emission continuum, and density of the ambient medium. We derive the corresponding correlation parameters, and quantify the intrinsic scatter by means of Bayesian analysis. We propose that the discovered correlation could be used as a cosmological tool to estimate the density of ambient medium for large samples of distant radio galaxies. Our method does not require any detailed modeling of individual sources, and relies on limited observational information, namely the slope of the radio continuum between the rest-frame frequencies 0.4GHz and 5GHz, possibly combined with the total linear size of the radio structure., Accepted for publication in the Astrophysical Journal

Objectives: To analyse a population of foetuses with prenatally diagnosed hypoplastic left heart syndrome (HLHS). Material and methods: Retrospective study of foetuses diagnosed with HLHS between 2013 and 2017 in a referral centre. Results: HLHS was found in 9.7% (65/665) of foetuses with cardiovascular abnormalities (CVA). As an isolated anomaly, HLHS was present in 40% of cases; in 24.5% other CVA were detected; in 14%, CVA and extracardiac anomalies; and in 21.5% only extracardiac malformations. Genetic disorders were present in 18.4% (12/65) of foetuses. 42% of cardiovascular and 25% of extracardiac anomalies were diagnosed postnatally. There were 10 (15.4%) elective terminations, 1 (1.5%) spontaneous foetal demise. Two newborns died after birth before surgery. Of the 52 children who underwent Norwood surgery, 13 (25%) died (9 with additional anomalies, and 4 with isolated HLHS). Of the 38 children who underwent stage II surgery, 2 (5.2%) with isolated HLHS died, and 1 (2.6%) with CVA. Conclusions: A diagnosis of HLHS is an indication for a detailed examination of cardiac and noncardiac structures. It is advisable to consider genetic testing, together with the microarray assessment. The prognosis depends on underlying cardiac and extracardiac anomalies and coexisting genetic defects.

DDE (dichlorodiphenyldichloroethylene) is an environmental metabolite of the pesticide DDT, which is still present in the environment, and its insecticidal properties are used to fight malaria and the Zika virus disease. We showed for the first time that the neurotoxic effects of DDE involve autophagy, as demonstrated by elevated levels of Becn1, Map1lc3a/MAP1LC3A, Map1lc3b, and Nup62/NUP62 and an increase in autophagosome formation. The suggestion that the aryl hydrocarbon receptor (AHR) and the constitutive androstane receptor (CAR) are involved in the neurotoxic effect of DDE was supported by increases in the mRNA and protein expression of these receptors, as detected by qPCR, ELISA, immunofluorescence labeling and confocal microscopy. Selective antagonists of the receptors, including alpha-naphthoflavone, CH223191, and CINPA 1, inhibited p,p'-DDE- and o,p'-DDE-induced LDH release and caspase-3 activity, while specific siRNAs (Ahr and Car siRNA) reduced the levels of p,p'-DDE- and o,p'-DDE-induced autophagosome formation. Although the neurotoxic effects of DDE were isomer independent, the mechanisms of p,p'- and o,p'-DDE were isomer specific. Therefore, we identified previously unknown mechanisms of the neurotoxic actions of DDE that, in addition to inducing apoptosis, stimulate autophagy in mouse neocortical cultures and induce AHR and CAR signaling.

We present a systematic analysis of the mid-infrared (MIR) properties of the youngest radio galaxies, based on low-resolution data provided by the {\it WISE} and {\it IRAS} satellites. We restrict our analysis to sources with available X-ray data that constitute the earliest phase of radio galaxy evolution, i.e. those classified as Gigahertz Peaked Spectrum (GPS) and/or Compact Symmetric Objects (CSOs). In our sample of 29 objects, we find that the host galaxies are predominantly red/yellow ellipticals, with some of them displaying distorted morphology. We find a variety of MIR colors, and observe that the sources in which the MIR emission is dominated by the ISM component uniformly populate the region occupied by galaxies with a wide range of pronounced ($\geq 0.5 M_{\odot}$\,yr$^{-1}$) star formation activity. We compare the MIR color distribution in our sample to that in the general population of local AGN, in the population of evolved FR\,II radio galaxies, and also in the population of radio galaxies with recurrent jet activity. We conclude that the triggering of radio jets in AGN does not differentiate between elliptical hosts with substantially different fractions of young stars; instead there is a relationship between the jet duty cycle and the ongoing star formation. Fetal cardiac assessment is an integral part of the obstetric ultrasound. The inclusion of the outflow tracts and the three-vessel and tracheal view into the ultrasound screening enhances the detection rate for cardiovascular anomalies. Both, international and Polish guidelines recommend routine evaluation of the upper mediastinum. The aim of the study was to present the principles for assessing the structures of the upper mediastinum in normal conditions and to draw attention to the pathologies which may be visible in this plane.

Here we discuss the jet production efficiency in a sample of 17 young radio galaxies with measured redshifts, kinematic ages, and nuclear X-ray fluxes, for which the observed luminosities of compact jets/lobes and accretion disks correspond to the same episode of the AGN activity. For the targets, we analyze the available optical data, estimating the bolometric luminosities of the accretion disks $L_{\rm bol}$, and the black hole masses; we also derive the minimum jet kinetic luminosities, $P_{\rm j}$. With such, we investigate the distribution of our sample in the three-dimensional space of the accretion rate $\lambda_{\rm Edd} \equiv L_{\rm bol}/L_{\rm Edd}$, the nuclear X-ray luminosity $L_{\rm X}$ considered here as a limit for the emission of the disk coronae, and $P_{\rm j}$, expressing the latter two parameters either in the Eddington units, or in the units of the disk luminosity. We find that (i) the accretion rate $\lambda_{\rm Edd}$ in our sample is distributed within a narrow range $\lambda_{\rm Edd} \sim 0.01 - 0.2$; (ii) the normalized jet power $P_{\rm j}/L_{\rm Edd}$ formally correlates with the accretion rate $\lambda_{\rm Edd}$, with some saturation at the largest values $\lambda_{\rm Edd}> 0.05$; (iii) the jet production efficiency $\eta_{\rm jet} \equiv P_{\rm j}/\dot{M}_{\rm acc} c^2$ spans a range from $\eta_{\rm jet} \lesssim 10^{-3}$ up to $\sim 0.2$ at maximum, which is below the level expected for magnetically arrested disks around maximally spinning black holes; and (iv) there is a diversification in $\eta_{\rm jet}$ on the hardness--intensity diagram $L_{\rm X}/L_{\rm bol} - \lambda_{\rm Edd}$, with the jets being produced most efficiently during the high/hard states, and suppressed during the soft states., Comment: the Astrophysical Journal, in press

Triclocarban is a phenyl ether that has recently been classified as a contaminant of emerging concern. Evidence shows that triclocarban is present in human tissues, but little is known about the impact of triclocarban on the nervous system, particularly at early developmental stages. This study demonstrated that triclocarban that was used at environmentally relevant concentrations induced apoptosis in mouse embryonic neurons, inhibited sumoylation, and changed the epigenetic status, as evidenced by impaired activities of HDAC, sirtuins, and DNMT, global DNA hypomethylation, and alterations of methylation levels of bax, bcl2, Ahr, and Car genes. The use of selective antagonists and specific siRNAs, which was followed by the co-localization of aryl hydrocarbon receptor (AHR) and constitutive androstane receptor (CAR) in mouse neurons, points to the involvement of AHR and CAR in triclocarban-induced neurotoxicity. A 24-h treatment with triclocarban enhanced protein levels of the receptors which was paralleled by Car hypomethylation and Ahr hypermethylation. Car hypomethylation is in line with global DNA hypomethylation and explains the increased mRNA and protein levels of CAR in response to triclocarban. Ahr hypermethylation could reflect reduced Ahr mRNA expression and corresponds to lowered protein levels after 3- and 6-h exposures to triclocarban that is likely related to proteasomal degradation of activated AHR. We hypothesize that the triclocarban-induced apoptosis in mouse neurons and the disruption of epigenetic status involve both AHR- and CAR-mediated effects, which may substantiate a fetal basis of the adult onset of neurological diseases; however, the expression of the receptors is regulated in different ways.

Empirical data suggest, that in many situations people are not able to estimate the probability of events on the base of available information in a way consistent with the normative model. This fact infl uences the choice of conclusions which are considered to be rational. The article outlines the factors that may affect the assumed value of a priori probability and – indirectly – the value of a posteriori probability. All these factors will be collectively referred to as the parameter j. Its value depends on the context in which the reasoning is made. In the article I show, that a critical assessment of our reasoning is not always justified.

Several lines of evidence suggest that exposures to Endocrine Disrupting Chemicals (EDCs) such as pesticides increase the risks of neuropsychiatric disorders. Despite extended residual persistence of dichlorodiphenyltrichloroethane (DDT) in the environment, the mechanisms of perinatal actions of DDT that could account for adult-onset of depression are largely unknown. This study demonstrated the isomer-specific induction of depressive-like behavior and impairment of Htr1a/serotonin signaling in one-month-old mice that were prenatally exposed to DDT. The effects were reversed by the antidepressant citalopram as evidenced in the forced swimming (FST) and tail suspension (TST) tests in the male and female mice. Prenatally administered DDT accumulated in mouse brain as determined with gas chromatography and tandem mass spectrometry, led to global DNA hypomethylation, and altered the levels of methylated DNA in specific genes. The induction of depressive-like behavior and impairment of Htr1a/serotonin signaling were accompanied by p,p'-DDT-specific decrease in the levels of estrogen receptors i.e. ESR1 and/or GPER1 depending on sex. In contrast, o,p'-DDT did not induce depressive-like effects and exhibited quite distinct pattern of biochemical alterations that was related to aryl hydrocarbon receptor (AHR), its nuclear translocator ARNT, and ESR2. Exposure to o,p'-DDT increased AHR expression in male and female brains, and reduced expression levels of ARNT and ESR2 in the female brains. The evolution of p,p'-DDT-induced depressive-like behavior was preceded by attenuation of Htr1a and Gper1/GPER1 expression as observed in the 7-day-old mouse pups. Because p,p'-DDT caused sex- and age-independent attenuation of GPER1, we suggest that impairment of GPER1 signaling plays a key role in the propagation of DDT-induced depressive-like symptoms.

Objectives To analyze a population of fetuses with prenatally diagnosed right aortic arch (RAA). Methods Retrospective study of fetuses with RAA diagnosed prenatally between 2011 and 2015 in two referral centers. Results Right aortic arch was found in 4.4% (46/1036) of fetuses with cardiovascular abnormalities (CVA). As an isolated anomaly, RAA was present in 30.4% of cases; in 32.6%, other CVA were detected; in 23.9%, CVA and extracardiac anomalies; and in 13.1%, only extracardiac malformations. The most common noncardiac abnormalities were thymus hypoplasia/aplasia (7/17), of which six had deletion 22q.11.2. In another three fetuses, trisomy 21 was present. One intrauterine fetal death occurred at 41 weeks of pregnancy, and two fetuses died after birth. In six of 18 infants with known follow-up, symptoms of dysphagia were reported, of which four infants underwent surgical intervention. In 12 infants, an isolated RAA was clinically silent. Conclusions The diagnosis of RAA is an indication for a detailed examination of cardiac and noncardiac structures, including the thymus. It is advisable to consider genetic testing, together with the assessment of deletion 22q11.2, especially in the case of accompanying defects. The prognosis depends on underlying cardiac and extracardiac anomalies and possibly coexisting genetic defects. Isolated anomalies are asymptomatic. © 2017 John Wiley & Sons, Ltd.

Dibutyl phthalate (di-n-butyl phthalate, DBP) is one of the most commonly used phthalate esters. DBP is widely used as a plasticizer in a variety of household industries and consumer products. Because phthalates are not chemically bound to products, they can easily leak out to enter the environment. DBP can pass through the placental and blood–brain barriers due to its chemical structure, but little is known about its mechanism of action in neuronal cells. This study demonstrated the toxic and apoptotic effects of DBP in mouse neocortical neurons in primary cultures. DBP stimulated caspase-3 and LDH activities as well as ROS formation in a concentration (10 nM–100 µM) and time-dependent (3–48 h) manner. DBP induced ROS formation at nanomolar concentrations, while it activated caspase-3 and LDH activities at micromolar concentrations. The biochemical effects of DBP were accompanied by decreased cell viability and induction of apoptotic bodies. Exposure to DBP reduced Erα and Pparγ mRNA expression levels, which were inversely correlated with protein expression of the receptors. Treatment with DBP enhanced Ahr mRNA expression, which was reflected by the increased AhR protein level observed at 3 h after exposure. ERα, ERβ, and PPARγ antagonists stimulated DBP-induced caspase-3 and LDH activities. AhR silencing demonstrated that DBP-induced apoptosis and neurotoxicity are mediated by AhR, which is consistent with the results from DBP-induced enhancement of AhR mRNA and protein expression. Our study showed that AhR is involved in DBP-induced apoptosis and neurotoxicity, while the ERs and PPARγ signaling pathways are impaired by the phthalate.
