Abstract

Simple Summary: This study explored the protein expression levels of UCHL1, SNRNP200, and PAK4 in clear cell renal cell carcinoma (CCRCC) using advanced proteomic techniques. We found that UCHL1 and SNRNP200 are significantly upregulated, and PAK4 is downregulated in high-grade CCRCC, and all are involved in critical cellular pathways linked to cancer progression and poor clinical outcomes. Specifically, UCHL1 is implicated in the Akt signaling pathway, SNRNP200 plays a role in RNA splicing and cell cycle regulation, and PAK4 is associated with cell proliferation and invasion. The enhanced expression of UCHL1 and SNRNP200, and decreased expression of PAK4 correlate with shorter progression-free survival in CCRCC patients, suggesting their potential as prognostic markers and targets for therapeutic intervention. This research contributes to the understanding of CCRCC's molecular dynamics and opens new avenues for targeted treatments. Recent advancements in proteomics have enhanced our understanding of clear cell renal cell carcinoma (CCRCC). Utilizing a combination of liquid chromatography-tandem mass spectrometry (LC-MS/MS) followed by immunohistochemical validation, we investigated the expression levels of UCHL1, PAK4, and SNRNP200 in high-grade CCRCC samples. Our analysis also integrated Reactome pathway enrichment to elucidate the roles of these proteins in cancer-related pathways. Our results revealed significant upregulation of UCHL1 and SNRNP200 and downregulation of PAK4 in high-grade CCRCC tissues compared to non-cancerous tissues. UCHL1, a member of the ubiquitin carboxy-terminal hydrolase family, showed variable expression across different tissues and was notably involved in the Akt signaling pathway, which plays a critical role in cellular survival in various cancers. SNRNP200, a key component of the RNA splicing machinery, was found to be essential for proper cell cycle progression and possibly linked to autosomal dominant retinitis pigmentosa. PAK4's role was noted as critical in RCC cell proliferation and invasion and its expression correlated significantly with poor progression-free survival in CCRCC. Additionally, the expression patterns of these proteins suggested potential as prognostic markers for aggressive disease phenotypes. This study confirms the upregulation of UCHL1, SNRNP200, and PAK4 as significant factors in the progression of high-grade CCRCC, linking their enhanced expression to poor clinical outcomes. These findings propose these proteins as potential prognostic markers and therapeutic targets in CCRCC, offering novel insights into the molecular landscape of this malignancy and highlighting the importance of targeted therapeutic interventions. Abstract

Introduction. This publication aims to present the results of a retrospective analysis of the treatment outcomes of patients with metastatic renal-cell carcinoma (RCC) who underwent cytoreductive nephrectomy (CN) followed by systemic treatment with sunitinib. Material and methods. The retrospective analysis includes the results of 67 patients treated and followed up at the Institute of Oncology in Poznan University of Medical Sciences. Results. Among the 67 patients included in the study, 24 were female (35.82%) and 43 were male (64.18%). The patients treated with sunitinib experienced several adverse effects, including weight loss, anaemia, neutropenia, hypokalemia, and thyroid dysfunction. For these reasons, some patients (n = 32, 47.76%) required a reduction in the dose of sunitinib. The most common reason for sunitinib discontinuation was disease progression (n = 52,77.61%). Conclusions. Treatment with sunitinib requires regular clinical and laboratory monitoring to appropriately reduce the drug dose or increase the interval between drug cycles in the event of adverse effects. 20. Preoperative cell-free DNA concentration in plasma as a diagnostic and prognostic biomarker of clear cell renal cell carcinoma.

Abstract

Introduction: Assessment of renal tumour masses is based on conventional imaging studies (computer tomography or magnetic resonance), which does not allow characterisation of the histopathological type. Moreover, the prediction of prognosis in localised and metastatic renal cell carcinoma requires improvement as well. Analysis of circulating free DNA (cfDNA) in blood is one of the variants of liquid biopsy that may improve diagnostics and prognosis issues of patients with renal tumour masses suspected to be renal cell carcinoma. The aim of the study was to assess the diagnostic and prognostic role of preoperative cfDNA concentration in the plasma samples of clear cell renal cell carcinoma (ccRCC) patients. Material and methods: The preoperative plasma cfDNA concentration was assessed in ccRCC patients (n = 46) and healthy individuals (control group) (n = 17). The circulating free DNA concentration was reflected by the 90 bp DNA fragments determined by real-time polymerase chain reaction. Results: The median cfDNA concentration was significantly higher in ccRCC patients (n = 46) compared to the control g roup (n = 17) (2588 ±2554 copies/ml vs. 960 ±490 copies/ml, p < 0.01). In multivariate analysis, the preoperative plasma cfDNA concentration was the significant factor increasing the probability of ccRCC detection (OR: 1.003; 95% CI: 1.001-1.005). The median cfDNA concentration depended on the stage of ccRCC; it was higher in metastatic ccRCC patients (n = 11) compared to non-metastatic ccRCC patients (n = 35) (3619 ±4059 copies/ml vs. 2473 ±1378 copies/ml, p < 0.03). Kaplan-Meier survival analysis demonstrated that patients with high cfDNA values (above 2913 copies/ml) had significantly worse cancer-specific survi-val (HR: 4.5; 95% CI: 1.3-16.9, logrank Mantel-Cox test p = 0.015). 26. Late prehistoric economy and society of the islands off the coast of Venezuela : a contextual interpretation of the non-ceramic evidence

Abstract

Archaeologists have portrayed north-central Venezuela as the seat of the Valencioid 'chiefdom' (a.d. 900-1500) and assumed socio-cultural continuity between the Valencioid culture and the historic Caraca Indians. These assumptions have neither been constructed nor tested on socially meaningful archaeological contexts. My research formulates and tests the cognitive value of the non-ceramic evidence recovered from socially meaningful archaeological contexts on the islands of the Central Coast of Venezuela as sources for understanding the developmental trajectory of the Valencioid polity(ies). The data come from archaeological surveys and excavations on 55 offshore islands and at 47 sites. Through horizontal excavation, off-site control units and statistical control over sample size it is established that the economic purpose for the occupation of a large Valencioid campsite at Dos Mosquises Island (a.d. 1400-1500) was primarily to exploit Conch Shell (Strombus gigas) for food and raw material. Other local resources, such as fishes, lobsters, turtles and birds, were complementary. The conceptual polarity food/artefact, often applied to the archaeofaunal analyses, is replaced by contextual discrimination between food, non-food remains and natural objects, which leads to inferences on social group composition, labour division, specialisation, differential access to food and the exportation of shell raw material outside the islands. The results of the contextual analyses of allochthonous mammal and special purpose artefacts indicate the presence of prominent members of the society (chief, shaman and/or warriors) and the ceremonial character of the core locus at the Dos Mosquises site. It is suggested that the organisation of the insular enterprise is most likely controlled from this core locus. From a macro-regional perspective, the resulting analyses challenge the notion of a 600 yearlong unilinear evolution toward social complexity of the Valencioid polity. The previous view of a straightforward, hegemonic character of this polity is replaced by perspective of recurrent long and short-term changes in the nature and intensity of regional interactions between several polities. These interactions were based on a changing multilateral negotiations of power through trade, co-operativev entures,r esourcee xploitation, intermarriage,c eremonial assistancew, arfare and peace. The purported continuity between the insular Valencioids and the Caraca Indians is not supported by the archaeological data.
