The rate of reproduction and lifetime productivity of the animals determine the success of a herd. As these features significantly impact sow productivity, welfare, and profitability, there is considerable interest in global swine breeding programmes to genetically select better sows for these attributes. In this scenario, reliable heritability estimates and genetic correlations between traits are essential for the success of genetic selection for such traits. The goals of the current study were to examine the reproductive and longevity traits and to estimate the genetic and phenotypic parameters for these traits in Landlly sows kept at a swine production farm, Izatnagar, Uttar Pradesh, India. The effect of non-genetic factors on different reproductive traits was estimated using SPSS version 16.0 and Bayesian approach which were utilized to estimate the genetic parameters for reproductive and longevity traits. The current study’s finding that the year and season of birth significantly affect reproductive traits generally suggests that there is potential for boosting productivity by optimising managemental practices. The heritability estimates were low for the reproductive and longevity traits, indicating a very little additive genetic variance in these traits, and individual selection will not be helpful for improving them. As some traits, such as LSB_FF & LSW_FF and LSB _SF & LSW_SF, have a moderate genetic correlation, indirect selection can be used to improve these two pairs of features.

Abstract Enhancement of nanoscale confinement in the subwavelength waveguide is a concern for advancing future photonic interconnects. Rigorous innovation of plasmonic waveguide-based structure is crucial in designing a reliable on-chip optical waveguide beyond the diffraction limit. Despite several structural modifications and architectural improvements, the plasmonic waveguide technology is far from reaching its maximum potential for mass-scale applications due to persistence issues such as insufficient confined energy and short propagation length. This work proposes a new method to amplify the propagating plasmons through an external on-chip surface acoustic signal. The gold–silicon dioxide (Au-SiO2) interface, over Lithium Niobate (LN) substrate, is used to excite propagating surface plasmons. The voltage-varying surface acoustic wave (SAW) can tune the plasmonic confinement to a desired signal energy level, enhancing and modulating the plasmonic intensity. From our experimental results, we can increase the plasmonic intensity gain of 1.08 dB by providing an external excitation in the form of SAW at a peak-to-peak potential swing of 3 V, utilizing a single chip.

Livestock infectious diseases pose a significant threat to animal health and welfare on a worldwide scale, and efficient management of these diseases is essential for maintaining agronomic health, securing national and international food supply, and reducing rural poverty in developing nations. It is challenging to eliminate the negative effects of disease effectively, economically, and permanently in livestock because of the shortcomings and constraints of current non-selection disease management approaches (e.g., vaccination, treatment, eradication strategy, and genome editing). Due to these shortcomings and restrictions, breeders are more interested and committed to addressing livestock health issues by selecting animals with favorable health traits. Selecting animals with specific health traits (such as disease tolerance, disease resistance, and immune response) can help to improve the health of livestock. Variability in host immune responses to infection plays a major role in host genetic variation in disease resistance. An approach can be employed as a strategy to fight infections by finding animals that are relatively immune or tolerant to diseases and revealing the inherent genetic variations in immune response at the molecular level. The accelerated and expanding understanding of genes and genomes in livestock, such as the evaluation of a broad number of DNA markers in phenotypic registered populations, could be used to identify and classify candidate genes responsible for variation in the immune response against major infectious diseases which would undoubtedly improve animal selection processes. Advancements in marker research could influence the selection programs if incorporated with the herd's genetic variant details as well as production traits. In this review, we have highlighted the genetic variation observed in host immune response to infectious diseases in bovines and its use in animal breeding for disease resistance.

Age-related macular degeneration (AMD) is a multifactorial degenerative disease characterised by the gradual loss of central vision in individuals aged more than 50 years. There is currently no cure for this disease, but treatment can delay its progression. Consequently, there is an urgent need for the development of both new and cost-effective therapeutics. In this study, a novel combination of a corticosteroid and flavonoid was investigated on human retinal pigment epithelial cell lines to explore its potential pharmacological effect on AMD. Combination therapies, such as anti-VEGF (vascular endothelial growth factor) agents combined with photodynamic therapy and anti-VEGF agents in conjunction with corticosteroids, have been utilized previously and are known to be effective. However anti-VEGF injections are associated with serious side effects and are costly. Various disease conditions associated with AMD were stimulated on human retinal cells, which were then exposed to different concentrations of triamcinolone acetonide (TA) and quercetin (QCN) individually and in combination. This investigation aimed to assess their potential for the treatment of AMD. The combination of TA and QCN demonstrated a superior anti-inflammatory effect, as TA and QCN primarily act on different inflammatory signaling pathways. Furthermore, in terms of anti-VEGF activity, both drugs exert their effects through different mechanisms: QCN inhibits kinase pathways leading to the deactivation of VEGF receptors, whereas TA destabilises VEGF mRNA, resulting in increased suppression of VEGF-C with combination treatments. The anti-oxidant assay yielded similar outcomes, demonstrating a synergetic effect when treated with combination drugs. These findings collectively suggest TA and QCN as a promising combination therapy for targeting AMD with multiple pathological conditions.

An inbred strain limits the segregating variance and provides more power and requires fewer experimental animals to produce good reproducibility. This study was undertaken with the aim to assess the impact of sex of offsprings and sex specific microsatellite variants on performance and fitness traits in F4 inbred Swiss albino mice. The phenomics of different performance and fitness traits were estimated on 506 F4 inbred mice population. Swiss albino mice have been widely utilized in various biological researches worldwide. Phenotypic and fitness related traits of F0 and F1 inbred mice were estimated on 918 and 707 individual offsprings, respectively. The influence of fixed effects (litter size and sex) on birth weight (BW), weaning weight (WW) and adult body weight (ABW) in both the generations were found to be statistically significant. Genetic characterization of F0 outbred and the F1 inbred strain of Swiss albino mice were evaluated by using 10 microsatellites markers. The results indicated that total number of alleles per locus ranged from 3 (D2Mit61, D3Mit55, D8Mit14, D9Mit27, D10Mit180, D11Mit167) to 4 (D1Mit15, D2Mit51, D5Mit18, D7Mit323) in F0 and F1 inbred population, with a mean value of 3.4 indicating polymorphism in all 10 loci. The mean of effective number of alleles was 2.935 and 2.733 in F0 and F1 population, respectively. Estimates of the FIS ranged from 0.139 (D10Mit180) to 0.999 (D9Mit27); and from 0.109 (D3Mit55) to 0.679 (D2Mit51) in F0 and F1 inbred population, respectively. The estimated mean markerbased FIS was 0.294 and 0.372 in F0 and F1 populations, respectively. The mean values of observed heterozygosity (Ho) and expected heterozygosity (He) were 0.460 and 0.654, respectively for F0 and 0.390 and 0.627, respectively for F1 inbred mice population. Slight reduction in heterozygosity and 7.8% increase in inbreeding coefficient were observed in F1 inbred in comparison to F0 population. The results suggested that genome wide microsatellite genotyping might be more useful for accurate measuring and reliable estimation of population genetic parameters and inbreeding coefficient.

Posterior segment eye diseases (PSEDs) including age macular degeneration (AMD) and diabetic retinopathy (DR) are amongst the major causes of irreversible blindness worldwide. Due to the numerous barriers encountered, highly invasive intravitreal (IVT) injections represent the primary route to deliver drugs to the posterior eye tissues. Thus, the potential of a more patient friendly topical route has been widely investigated. Mucoadhesive formulations can decrease precorneal clearance while prolonging precorneal residence. Thus, they are expected to enhance the chances of adherence to corneal and conjunctival surfaces and as such, enable increased delivery to the posterior eye segment. Among the mucoadhesive polymers available, chitosan is the most widely explored due to its outstanding mucoadhesive characteristics. In this review, the major PSEDs, their treatments, barriers to topical delivery, and routes of topical drug absorption to the posterior eye are presented. To enable the successful design of mucoadhesive ophthalmic drug delivery systems (DDSs), an overview of mucoadhesion, its theory, characterization, and considerations for ocular mucoadhesion is given. Furthermore, chitosan-based DDs that have been explored to promote topical drug delivery to the posterior eye segment are reviewed. Finally, challenges of successful preclinical to clinical translation of these DDSs for posterior eye drug delivery are discussed.

The current treatment for the acquired retinal vasculopathies involves lifelong repeated intravitreal injections of either anti-vascular endothelial growth factor (VEGF) therapy or modulation of inflammation with steroids. Consequently, any treatment modification that decreases this treatment burden for patients and doctors alike would be a welcome intervention. To that end, this research aims to develop a topically applied nanoparticulate system encapsulating a corticosteroid for extended drug release. Poly (lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) supports the controlled release of the encapsulated drug, while surface modification of these NPs with chitosan might prolong the mucoadhesion ability leading to improved bioavailability of the drug. Triamcinolone acetonide (TA)-loaded chitosan-coated PLGA NPs were fabricated using the oil-in-water emulsion technique. The optimized surface-modified NPs obtained using Box-Behnken response surface statistical design were reproducible with a particle diameter of 334 ± 67.95 to 386 ± 15.14 nm and PDI between 0.09 and 0.15. These NPs encapsulated 55–57% of TA and displayed a controlled release of the drug reaching a plateau in 27 h. Fourier-transform infrared spectroscopic (FTIR) analysis demonstrated characteristic peaks for chitosan (C-H, CONH2 and C-O at 2935, 1631 and 1087 cm−1, respectively) in chitosan-coated PLGA NPs. This result data, coupled with positive zeta potential values (ranged between +26 and +33 mV), suggests the successful coating of chitosan onto PLGA NPs. Upon coating of the NPs, the thermal stability of the drug, polymer, surfactant and PLGA NPs have been enhanced. The characteristics of the surface-modified NPs supports their use as potential candidates for topical ocular drug delivery for acquired retinal vasculopathies.

In vitro screening and testing of drugs and devices is necessary, but in vitro conditions differ greatly from those found in vivo. These differences can lead to false promises of efficacy, or can hide problems of tissue compatibility. Models with ex vivo tissues can be highly valuable bridges which provide relevant matrices for testing [1–9]. Ex vivo tissue models which are closer both biochemically and biophysically can provide useful feedback in a more time- and cost-efficient manner. Herein we describe an ex vivo corneal model for use in drug delivery testing and corneal infection modeling [10]. The protocol covers the tissue harvesting, sterilization, inoculation, and bacterial load quantification. We envision that the model can be used to study bacterial physiology on metabolizable matrices and to study the direct effects of microbial colonization on the cornea's integrity and clarity. • Devitalized cornea. • Non-submersed conditions. • Contact lens compatible.

Mastitis is a complex disease responsible for huge economic losses to the dairy sector. The causal organisms include a wide variety of micro-organisms including several species of bacteria. Escherichia coli has been identified as one of the most common gram-negative bacteria causing clinical mastitis in cattle. The immune system, of different species and/or breeds, tries to combat these pathogens in an inconsistent manner with differential mode and intensity of immune response, eventually producing contradicting outcomes of this disease. Several reports suggest the existence of variability among different animal breeds/species, resulting in a dissimilar outcome of this disease among them. In order to evaluate the variation among different breeds/species, the present study was undertaken to examine the stimulant effect of E. coli lipopolysaccharide (LPS) on peripheral blood mononuclear cells (PBMCs). The PBMCs were harvested from blood samples of crossbred cattle, Tharparkar cattle and Murrah buffaloes. After 6 h of in vitro stimulation, qRT-PCR was employed to measure the relative mRNA expression levels of CCL5, IL-1β, IL-12β, IFN-γ and IL-10 genes in stimulated and unstimulated PBMCs. The selected genes revealed significant differences in the pattern of innate immune response among crossbred cattle, Tharparkar cattle and Murrah buffaloes. The results clearly indicate the presence of variation in the outcome of immune response even when the immunocytes were stimulated with the same dose of the antigen.

The permeability through the cornea determines the ability of a drug or any topically applied compound to cross the tissue and reach the intraocular area. Most of the permeability values found in the literature are obtained considering topical drug formulations, and therefore, refer to the drug permeability inward the eye. However, due to the asymmetry of the corneal tissue, outward drug permeability constitutes a more meaningful parameter when dealing with intraocular drug-delivery systems (i.e., drug-loaded intraocular lenses, intraocular implants or injections). Herein, the permeability coefficients of two commonly administered anti-inflammatory drugs (i.e., bromfenac sodium and dexamethasone sodium) were determined ex vivo using Franz diffusion cells and porcine corneas in both inward and outward configurations. A significantly higher drug accumulation in the cornea was detected in the outward direction, which is consistent with the different characteristics of the corneal layers. Coherently, a higher permeability coefficient was obtained for bromfenac sodium in the outward direction, but no differences were detected for dexamethasone sodium in the two directions. Drug accumulation in the cornea can prolong the therapeutic effect of intraocular drug-release systems.

Aim: Toll-like receptor 2 (TLR2) and TLR4 genes play critical roles in host recognition of Mycobacterium bovis infection and initiation of innate and adaptive immune response. The present study was aimed at exploring the association of seven single nucleotide polymorphisms (SNPs) in TLR2 and TLR4 genes with susceptibility/resistance against bovine tuberculosis (bTB) infection in cattle. Materials and Methods: A case-control resource population of 35 positive and 45 negative animals was developed after screening with single intradermal tuberculin test for bTB. Resource population was screened for SNPs in TLR2 and TLR4 genes using polymerase chain reaction-restriction fragment length polymorphism. The PROC LOGISTIC procedure of SAS 9.3 was used to find an association of allelic and genotypic frequencies with bTB. Results: In TLR2 gene, two of SNPs under study (rs55617172 and rs68268253) revealed polymorphism while in the case of TLR4 gene all four SNPs under investigation (rs8193041, rs207836014, rs8193060, and rs8193069) were found to be polymorphic in case-control population. Gold nanoparticles (GNPs) are commonly synthesized using the Turkevich method, but there are limitations on the maximum concentration of gold nanoparticles that can be achieved using this method (often < 1 mM (=0.34 mg/mL) gold precursor loading). Here, we report an inverse Turkevich method which significantly increases the concentration of gold nanoparticles (up to 5-fold) in the aqueous phase by introducing poly (vinyl alcohol) (PVA) to the synthesis system for stabilization. The aim of this study is to understand the effect of PVA and other synthesis parameters, such as trisodium citrate and tetrachloroauric acid concentration, with the goal of maximizing concentration while maintaining gold nanoparticle morphology, stability, and narrow size distribution. The size distribution of GNPs is investigated for a range of parameters by dynamic light scattering and electron microscopy, and ultraviolet-visible (UV–vis) spectroscopy is also utilized to explore the localized surface plasmon resonance (LSPR). Further, the interaction between GNPs and PVA is investigated by Fourier-transform infrared spectroscopy. In addition to increasing the gold loading by varying synthesis parameters, we also develop a novel anti-solvent precipitation method for the PVA-coated GNPs, which enables continuous condensation and purification of GNPs by forming a gold/PVA nanocomposite.

Aim: Out of various members of heat shock protein (HSP) superfamily which act a molecular chaperon by binding to the denaturing protein thus stabilizing them and preserving their activity, HSP70 are of major importance in thermotolerance development. Thus, present investigation aimed at a screening of HSP70 gene for polymorphisms and possible differences in thermotolerance in Tharparkar breed of cattle. Materials and Methods: A 295 bp fragment of HSP70 gene was subjected to polymerase chain reaction-singlestrand conformation polymorphism (SSCP) followed by sequencing of different SSCP patterns in 64 Tharparkar cattle. A comparative thermotolerance of identified genotypes was analyzed using heat tolerance coefficients (HTCs) of animals for different seasons. Results: Three SSCP patterns and consequently two alleles namely A and B were documented in one fragment of HSP70 gene. On sequencing, one single-nucleotide polymorphism with G > T substitution was found at a position that led to a change of amino acid aspartate to tyrosine in allele A. It was found that in maintaining near normal average rectal temperature, genotype AA was superior (p≤0.01). Genotype AA, thus, was found to be most thermotolerant genotype with the highest HTC (p≤0.01). Conclusion: The polymorphism at HSP70 is expected to be a potent determinant for heat tolerance in cattle, which may aid in selection for thermotolerance in cattle.

Aim: One of the major biochemical aspects of thermoregulation is equilibrium of ion gradient across biological membranes. Na+/K+-ATPase, a member of P type-ATPase family, is a major contributor to the mechanism that actively controls crossmembrane ion gradient. Thus, we examined ATP1A1 gene that encodes alpha-1 chain of Na+/K+-ATPase, for genetic polymorphisms. Materials and Methods: A total of 100 Vrindavani (composite cross strain of Hariana x Holstein-Friesian/Brown Swiss/Jersey) and 64 Tharparkar (indigenous) cattle were screened for genetic polymorphism in ATP1A1 gene, using polymerase chain reaction single-strand conformation polymorphism and DNA sequencing. For association studies, rectal temperature (RT) and respiration rate (RR) of all animals were recorded twice daily for 3 seasons. Results: A SNP (C2789A) was identified in exon 17 of ATP1A1 gene. Three genotypes namely CC, CA, and AA were observed in both, Vrindavani and Tharparkar cattle. The gene frequencies in Tharparkar and Vrindavani for allele A were 0.51 and 0.48, and for allele C were 0.49 and 0.52, respectively, which remained at intermediate range. Association study of genotypes with RT and RR in both cattle population revealed that the animals with genotype CC exhibited significantly lower RT and higher heat tolerance coefficient than CA and AA genotypes. Conclusion: Differential thermoregulation between different genotypes of ATP1A1 gene indicate that the ATP1A1 gene could be potentially contributing to thermotolerance in both, Tharparkar, an indigenous breed and Vrindavani, a composite crossbred cattle.

Leptin is a 16 kD protein, synthesized by adipose tissue and is involved in regulation of feed intake, energy balance, fertility and immune functions. Present study was undertaken with the objectives of sequence characterization and studying the nucleotide variation in leptin gene in Murrah buffalo. The leptin gene consists of three exons and two introns which spans about 18.9kb, of which the first exon is not transcribed into protein. In buffaloes, the leptin gene is located on chromosome eight and maps to BBU 8q32. The leptin gene was amplified by PCR using oligonucleotide primers to obtain 289 bp fragment comprising of exon 2 and 405 bp fragment containing exon 3 of leptin gene. The amplicons were sequenced to identify variation at nucleotide level. Sequence comparison of buffalo with cattle reveals variation at five nucleotide sequences at positions 983, 1083, 1147, 1152, 1221 and all the SNPs are synonymous resulting no in change in amino acids. Three of these eight nucleotide variations have been reported for the first time in buffalo. The results indicate conservation of DNA sequence between cattle and buffalo. Nucleotide sequence variations observed at leptin gene between Bubalus bubalis and Bos taurus species revealed 97% nucleotide identity.
