Abstract Vitamin B2 (riboflavin) is essential for cellular growth, energy production, and redox potential. Certain lactic acid bacteria (LAB) can synthesize B2 in low levels in fermented products, however it is mostly retained inside the cell. This study aimed to develop B2-enriched soymilk by fermenting with B2-producing probiotic Lactiplantibacillus plantarum strains and traditional starter culture Lactobacillus acidophilus NCIM2902. Using the central composite design approach, processing parameters were optimized for enhanced B2 content and probiotic count. Six independent variables were assessed: temperature (A: 35–45 °C), pH (B: 4–6), time (C: 3–18 h), and inoculum size for strains L. plantarum MTCC 25432 (D: 1–2%), L. plantarum MTCC 25433 (E: 1–2%), and L. acidophilus NCIM 2902 (F: 1–2%). The second-order model effectively predicted responses, identifying optimal fermentation conditions for developing vitamin B2-enriched soymilk: temperature (A) 36 °C, pH (B) 5.5, fermentation time (C): 11 h, inoculum size for L. plantarum MTCC 25432 (D): 2%, MTCC 25433 (E): 2%, and L acidophilus NCIM (F): 0.43%. These conditions resulted in a threefold increase in B2 concentration (481 µg/L) while maintaining a probiotic count of 9 logs CFU/mL. Additionally, techno-functional characterization, including rheology and texture profile analysis, showed that enhanced protease activity of co-cultured LAB improved protein hydrolysis (6259 nm), positively impacting the water holding capacity (WHC) and overall acceptability of the fermented soymilk. This optimized fermentation process represents a novel approach to developing nutritionally enhanced dairy-free soy products with high riboflavin content, utilizing the synergistic benefits of co-fermentation by two riboflavin-producing L. plantarum strains and traditional starter culture of L. acidophilus. This advancement is particularly significant for lactose-intolerant and vegan consumers who may lack sufficient dietary sources of Vitamin B2.

Blood is a life saver in many emergencies like accidents or for the patients suffering from deadly diseases such as cancer and thalassemia. Conventionally, blood collection is done in five steps, i.e., recruitment, screening, physical examination, collection, and postdonation care. The huge demand of blood requires the implementation of smart approaches in the field of blood donation, which allows specific, measurable, achievable, and timely outcomes. This paper throws light on various smart approaches to motivate and attract people for blood donation. Gamification, social media, short message service, mobile applications, and other web-based applications are some of these approaches reviewed in this article. A number of platforms are also highlighted where one can seek potential donors or blood banks easily during urgency, avoiding the complex steps followed in traditional blood donation systems.

Abstract An investigation was carried out to evaluate the impact of pretreatments viz. wax coating, hot water dipping, and fungicide treatment on quality characteristics of kinnow. In the present study, a trial was conducted to extend the shelf life of kinnow fruits by maintaining the initial quality characteristics with three wax-to-water dilution ratios (1:1, 1:2, 1:3), hot water dipping at 56 °C with varying time (40, 60, 80 s) along with fungicide treatment at various concentration (150, 200, 250 ppm). Both treated and untreated (experimental control) samples were kept at refrigerated conditions (5–8 °C) and 90–95% RH for 60 days. The effect of the storage period on quality parameters was recorded regarding weight loss, TSS, titrable acidity, ascorbic acid, and total color difference (TCD). The process parameters i.e. wax dilution, hot water dipping time, and fungicide concentration were optimized using response surface methodology (RSM). Optimized process parameters were 1:2.66 waxing dilution, 80 s hot water dipping time, and 250 ppm fungicide concentration. It could be concluded from the results that a combination of wax coating, hot water dipping, and fungicide treatment can help to prolong the storage period of kinnow up to 60 days with retained quality, as compared to 30 days in untreated control kinnow fruits when stored at 5–8 °C and 90–95% relative humidity.

Abstract Insecticidal potential of seven commonly available alkaloids against melon thrips (Thrips palmi Karny) was investigated through in vivo experiments and the bioactivity was explained via in silico approaches. In vivo screening showed highest mortality of T. plami larvae for reserpine (43%), closely followed by tropinone (41%) after 24 h of incubation. After 48 h, tropinone surpassed reserpine with 83% mortality, indicating its prolonged insecticidal activity. A detailed bioassay of tropinone revealed its LC50 values as 1187.9 and 686.9 µg mL−1 after 24 and 48 h, respectively. While studying the molecular interactions between the alkaloids and four physiologically important target proteins of T. palmi, tropinone demonstrated the highest ligand efficiency and lowest predicted inhibitory constant, particularly when forming complexes with CathB protein. However, binding energy calculations of the docked complexes showed most favorable binding of reserpine with CathB. To clear the ambiguity, considering both binding energy and ligand efficiency as the evaluation parameters, a molecular dynamics study was carried out, which predicted higher stability of CathB-tropinone complex than CathB-reserpine complex in terms of the total energy of the system. These in silico findings aligned well with the in vivo results, confirming tropinone as a promising candidate for effective thrips management programs.

Background: Development of secondary esophageal cancer after hematopoietic stem cell transplantation has been described; however, there is little consensus on treatment and surveillance for these patients. The objective of this study was to describe our experience treating patients with secondary esophageal cancer. Methods: A retrospective chart review of prospectively collected data was performed to identify patients who underwent hematopoietic stem cell transplantation from 1997 to 2012 and in whom esophageal cancer developed later. Results: A total of 5066 patients underwent hematopoietic stem cell transplantation, and esophageal cancer developed in 11 (0.2%) of these patients. The median time to diagnosis of esophageal cancer after hematopoietic stem cell transplantation was 11 years (interquartile range, 8.5 to 14 years). Four patients received a diagnosis of stage III or IV disease. Seven patients underwent esophagectomy, 6 patients after neoadjuvant treatment. Three patients experienced adverse events postoperatively, all grades II and IIIa. Two surgical patients died of distant recurrence 2 years and 3 years, respectively, after their esophageal cancer diagnosis. The other 5 surgical patients have not experienced recurrence of their esophageal cancer. Conclusions: For patients with secondary esophageal cancer, esophagectomy after neoadjuvant treatment has acceptable morbidity and may be a viable option for this cohort.

Background: Dysmenorrhea is a very common problem and a large number of females in our society are suffering from it. This study aims to determine the prevalence of dysmenorrhea and its associated lifestyle and other factors. Methodology: This was a cross-sectional study which was conducted among females aged between 15 to 45 years in urban population of district Ghaziabad through a house-to-house survey, using a pre-designed, semi-structured questionnaire. Results: The prevalence of dysmenorrhea was 67.8%. Out of the participants having dysmenorrhea, 36.5% females were experiencing mild pain, 44.3% were experiencing moderate pain and 19.1% were experiencing severe pain. The chances of dysmenorrhea were significantly more in participants of 15 to 25 years of age, participants who had attained early menarche, those having pre-menstrual symptoms and those who had passage of clots during menstruation, participants consuming refined flour, fried food, preservatives, soda and those who were skipping meals. Participants doing physical exercises showed less chance of menstrual pain. Conclusions: The prevalence is dysmenorrhea is quite high in our society. It may be related to factors which are modifiable, such as diet and exercise. An improvement of lifestyle is recommended.

Background: Reported advantages to robotic thoracic surgery include shorter length of stay (LOS), improved lymphadenectomy, and decreased complications. It is uncertain if these benefits occur when introducing robotics into a well-established video-assisted thoracoscopy (VATS) practice. We compared the two approaches to investigate these advantages. Materials and methods: IRB approval was obtained for this project. Patients who underwent segmentectomy or lobectomy from May 2016–December 2018 were propensity-matched 2: 1 (VATS: robotic) and compared using weighted logistic regression with age, gender, Charlson Comorbidity Index, surgery type, stage, Exparel, and epidural as covariates. Complication rates, operation times, number of sampled lymph nodes, pain level, disposition, and LOS were compared using Wilcoxon rank-sum and with Rao-Scott Chi-squared tests. Results: 213 patients (142 VATS and 71 robot) were matched. Duration of robotic cases was longer than VATS (median 186 min (IQR 78) vs. 164 min (IQR 78.75); p 0.05). Conclusions: The robotic approach facilitates better lymph node sampling, even in an established VATS practice.

Abstract For the application of biochar in restoring pesticide-contaminated soils and minimizing the risk associated with their uptake in plants, it is crucial to understand the biochar impact on soil biological activities and dissipation and accumulation of pesticides in plant and soil systems. In this study, the effect of Mentha-distilled waste-derived biochar was investigated on chlorpyrifos and atrazine contaminated sandy loam soil. The four application rates of atrazine (2, 4, 6, and 8 mg kg−1) and chlorpyrifos (2, 4, 6, and 12 mg kg−1) and a single application rate of biochar (4%) were used in this study. The degradation of pesticides, the diversity of the bacterial community, and enzymatic activities (alkaline phosphatase, dehydrogenase, arylsulfatase, phenol oxidase, urease activity and N-acetyl glucosaminidase) were examined in soil. The uptake of two pesticides and their effect on growth and stress parameters were also investigated in plants (A. paniculata). The dissipation of chlorpyrifos and atrazine followed simple first-order kinetics with a half-life of 6.6–74.6 and 21–145 days, respectively. The presence of deisopropyl atrazine desethyl atrazine (metabolites of atrazine) and 3,5,6-trichloro-2-pyridinol (a metabolite of chlorpyrifos) was observed in soil and plant tissues. Biochar application significantly (p = 0.001) enhanced the degradation rate of chlorpyrifos and atrazine leading to the lower half-life of chlorpyrifos and atrazine in soil. A significant reduction (p = 0.001) in the uptake of chlorpyrifos and atrazine and alteration in their binding affinity and uptake rate in plant tissues was observed in biochar treatments. The incorporation of biochar improved chlorpyrifos/atrazine degrader and plant growth-promoting bacterial genera such as Balneimonas, Kaistobacter, Rubrobacter, Ammoniphilus, and Bacillus. The upregulation of functional genes associated with nucleotide, energy, carbohydrate, amino acid metabolism, xenobiotic biodegradation, and metabolism: atrazine degradation was observed in biochar treatments. The biochar amendments significantly (p = 0.001) reduced the plant’s uptake velocity (Vmax) and affinity (Km) of chlorpyrifos and atrazine. These results delineated that Mentha-distilled waste-derived biochar can potentially remediate chlorpyrifos and atrazine contaminated soils and ensure the safety of plants for consumption. Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), a threat to maize production systems, is a polyphagous pest of global significance. There is no registered bioinsecticide of botanical origin to provide green remedy against this pest of concern. The present study reports for the first time the potency of the polar and non-polar bioinsecticidal leads sourced from Lippia alba (Mill.) N.E. Br. leaves. Shade-dried leaves of L. alba were extracted and evaluated; based on preliminary bioassay, the ethyl acetate leaf extract of L. alba (LEAE) was found to be the most potent against FAW in the in vitro and in vivo studies. Ultraperformance liquid chromatography–quadrupole time-of-flight–mass spectrometric (UPLC-QToF-MS) analysis of LEAE revealed the rich chemical profile of 28 compounds, dominated by flavones, namely, naringenin, trihydroxy-dimethoxy flavone, and dihydroxy-trimethoxy flavone. Among others, glycosides, such as clerodendrin, calceolarioside E, forsythoside B, geniposide, and martynoside, and glucuronides, such as luteolin-7-diglucuronide, tricin-7-O-glucuronide, and luteolin-7-O-glucuronide, were also identified. LEAE exhibited exceptionally high in vitro [LC50 = 6,900 parts per million (ppm)] and in vivo (computed as damage score on a scale of 1–9) insecticidal activity against S. frugiperda, with no phytotoxicity at a dose as high as 20 times of LC50. LEAE also exhibited significant antifeedant, ovicidal, and growth regulatory activity at the 70–16,000 ppm (w/v) concentration range. In silico assessment revealed strong binding of martynoside, calceolarioside E, and forsythoside B with acetylcholinesterase-, sodium-, and chloride-dependent γ-aminobutyric acid (GABA) receptor and ryanodine receptor, respectively, facilitated by hydrogen bonds (conventional and C–H bonds) stabilized by hydrophobic pi–sigma, pi–pi stacked, pi–alkyl, and alkyl interactions. The present study established L. alba as a potential bioresource and secondary metabolite enriched LEAE as bioinsecticide for further product development.

Abstract Scaling up the synthesis of fluorescent silica nanoparticles to meet the current demand in diverse applications involves technological limitations. The present study relates to the hydrothermal synthesis of water-soluble, crystalline, blue-emitting amine-functionalized silica nanoparticles from coal fly ash sustainably and economically. This study used tertiary amine (trimethylamine) to prepare amine-functionalized fluorescent silica nanoparticles, enhancing fluorescence quantum yield and nitrogen content for nanofertilizer application. The TEM and FESEM studies show that the silica nanoparticles have a spherical morphology with an average diameter of 4.0 nm. The x-ray photoelectron and Fourier transform infrared spectroscopy studies reveal the presence of the amine group at the surface of silica nanoparticles. The silica nanoparticles exhibit blue fluorescence with an emission maximum of 454 nm at 370 nm excitation and show excitation-dependent emission properties in the aqueous medium. With the perfect spectral overlap between silica nanoparticle emission (donor) and chlorophyll absorption (acceptor), fluorescent silica nanoparticles enhance plant photosynthesis rate by resonance energy transfer. This process accelerates the photosynthesis rate to improve the individual plant’s quality and growth. These findings suggested that the fly ash-derived functionalized silica nanoparticles could be employed as nanofertilizers and novel delivery agents.

Indian Railway Catering and Tourism Corporation (IRCTC) is among the busiest railways reservation systems since the Indian Railways (IR) is the vital and economical mode of transportation in India. Hence, rating of the trains seems to be critical aspect for selecting an appropriate train for travelling. In this study, we have considered 7 vital attributes of 500 popular trains and rate their performance based on 7 important related attributes. For this purpose, we have employed 2 different approaches to analyse of the train attributes, which eventually contribute to the overall performance of the trains. Here, we have developed a rule based rough set decision support system to analyse the criticality of the train attributes while rating the train performance. Furthermore, we have also used 3 Machine Learning (ML) model estimators: Extra Trees Classifier (ETC), Support Vector Machine Classifier (SVMC) and Multinomial Naive Bayes Classifier (MNBC) and perform their comparative analysis with respect to 7 performance metrics while predicting the overall train rating based.

The water issue has emerged as a significant barrier to agricultural productivity, particularly in dry and semiarid agro-ecologies, particularly during the winter season. Superabsorbent hydrogels have been identified as effective tools for improving crop and water productivity in arid locations by enhancing moisture retention. The application of hydrogel to soil is commonly done as a basal application, and its effectiveness is heavily influenced by the moisture level present in the soil. Therefore, a two-year field study was carried out to study the effects of various hydrogels and their application techniques and to assess the efficacy of recently developed cross–linked hydrogels (SPG 1118 and Pusa hydrogel) on soil water distribution (both spatial and temporal), water productivity and profitability of red cabbage. The experiment consisted of two irrigation regimes [full irrigation (100% ETc) and limited irrigation (75% ETc)] assisted in main plots, with two type of hydrogels (Pusa Hydrogel and SPG 1118) and two methods of application [root dipping and soil application of hydrogels and Control with no application of hydrogel] assisted in sub plots. The field experiment was conducted using spilt plot design with three replications. The analysis of soil water distribution patterns reveals that the utilization of hydrogels has resulted in a notable increase of 3–6% in soil water content (SWC) under full irrigation conditions, as compared to plots subjected to limited water conditions. In comparison to plots without hydrogel application, the plots treated with SPG 1118 and Pusa hydrogel exhibited enhanced soil water content of 5–6% and 2–4%, respectively. Additionally, the application of hydrogels in the root dipping showed increased water availability in the crop's root zone compared to soil application in throughout the crop season. Thus, hydrogels with reduced irrigation application resulted in significantly higher crop water productivity (14%) and irrigation water productivity (8 – 14%) compared to full irrigation levels. Likewise, root dipping of SPG 1118 led in considerable improvement in CWP (12–14%) and IWP (11–12%) over no hydrogel applied plots. In relation to profitability, the adoption of full irrigation in conjunction with SPG 1118 hydrogel resulted in an 8% increase in the benefit cost ratio compared to the control plots. Hence, the use of the unique SPG 1118 hydrogel through direct application to the roots, whether under conditions of full or limited irrigation, presents an inventive approach to address the growing issues of water shortages and sustainability in countries experiencing water stress.

Introduction: The measurement of bite force is increasingly gaining importance in dentistry. This preliminary report evaluates the maximum voluntary bite force (MVBF) of young healthy individuals with normal occlusion and correlates the effect of age and gender with the maximum bite force (MBF). Materials and Methods: 405 participants in the age group of 18 to 40 years were selected from the outpatient department meeting inclusion and exclusion criteria. MVBF was measured with the help of a digital device called “Byte” at incisors, right first molar, and left first molar region. Results: There was a strong correlation between age with bite force. Bite force increased with age. Males had more bite force than females. The posterior region had higher bite force than the anterior region. Conclusion: Measurement of bite force is very important for every patient undergoing restorative treatment. The “Byte” device was found to be very efficient.

The field of thoracic surgery has seen tremendous advances over the last decade with substantial progress over just the last several years. In this review, we discuss changes to lung cancer screening guidelines in the United States, briefly review ongoing trials and current screening programs in Europe and discuss the promising role of artificial intelligence (AI) in lung cancer screening. Additionally, we discuss new diagnostic techniques, including image-guided video-assisted thoracoscopic surgery (iVATS), robotic bronchoscopy, and the use of biomarkers. We review current trends towards thoracic surgery becoming increasingly minimally invasive with the evolution of uniportal VATS (U-VATS) and uniportal robotic surgery (U-RATS). Lastly, we summarize the latest immunotherapy trials, which have revolutionized both neoadjuvant and adjuvant treatment for lung cancer and have demonstrated promising potential to improve the lives of our patients. These advancements have dramatically changed the landscape of thoracic surgery in patients with lung cancer.

In mango pickle industry, a significant quantity of mango seed kernels is discarded as solid wastes. These seed kernels can be an ideal source for obtaining extracts rich in bioactive polyphenolic compounds with good antioxidant properties. The potential of mango kernel phenolic extract (MKPE) was investigated as a natural and effective antimicrobial agent for controlling major postharvest fungal pathogen infections, a significant threat to global food supply chains. Fungal pathogens contribute to the deterioration of fruits, vegetables, and grains during storage and transportation, leading to economic losses and compromised food safety. MKPE was obtained from pickling variety 'Ramkela' raw mango kernels, and its phenolic composition was characterized using LC–MS. The in vitro antifungal activity of MKPE against Botrytis cinerea, Colletotrichum gloeosporoides, and Rhizopus stolonifer was evaluated in vitro. A concentration-dependent inhibition of fungal radial growth against all three pathogens was observed, exhibiting the potential of MKPE as a valuable natural resource for addressing postharvest losses caused by fungal pathogens. The extraction process yielded a total phenolic content of 2128 mg GAE/100 g. Major polyphenolic bioactive compounds present were mangiferin, quercetin, and rhamnetin. The in-vitro antimicrobial assay showed reduction in the radial growth and inhibition percent of the pathogens. EC50 values of MKPE for B. cineria, C. gloeosporoides, and R. stolonifer was found to 364.17, 963.8 and 926 ppm, respectively. Our results demonstrate an economical, sustainable, and eco-friendly approach to manage post-harvest diseases rendered by fungi using mango MKPE from pickling industry waste.

The present study aimed at the numerical optimization of process variables to analyze the effect of hypochlorous acid (HOCl) treatment on postharvest quality characteristics of tomatoes. Box Behnken design was used to optimize the process variables, including concentration (40, 60, and 80 ppm), pH (4, 5, and 6), and dipping time (2, 4, and 6 min), and their effect on responses, i.e., weight loss (%), redness (a*), and microbial load (log cfu/ml). The optimized values of HOCl treatment process variables were found to be concentration (80 ppm), pH (4), and dipping time (6 min), and the corresponding responses were 11.69% (weight loss), 21.09 (redness), and 4.79 log cfu/ml (microbial load). The effects of different treatments, i.e., HOCl treatment (optimum values), ClO2 treatment (concentration 60 ppm, dipping time 15 min), and control (tap water), on weight loss (%), firmness (N), TSS (%), redness (a*), and microbial load (log cfu/ml) were compared for 10 days of storage (ambient conditions). Results revealed that decay rates for HOCl-treated tomatoes were significantly lower (54.86%) in comparison to ClO2 treatment (79.16%) and control samples (97.22%) at the end of the storage period, indicating the effectiveness of HOCl treatment. However, the microbial load in the HOCl-treated samples was significantly lower (7.61 ± 0.01 log cfu/ml), followed by ClO2 (8.39 ± 0.04 log cfu/ml) in comparison to control samples (10.14 ± 0.02 log cfu/ml) during the storage period. It could be concluded that HOCl could be used as a disinfectant for enhancing the shelf life of tomatoes and ensuring microbial safety.

Introduction: Bone and joint infections are very common in eastern India. Due to the lack of authentic data on the type of bacteriology in our region, this study was planned. Materials and Methods: After ethical clearance, retrospective data of 2 years from 2019 to 2021 were collected. Out of 115 patients, 77 patients were included in the study whereas 38 were excluded due to nonbacterial infections and incomplete data. Results: The average age of patients was 21.14 years. Pediatric patients were 58.66%. The common clinical condition was chronic osteomyelitis, followed by septic arthritis. The most common bone affected was femur, followed by tibia. The most common organism noted was methicillin-resistant Staphylococcus aureus (MRSA) and coagulase-negative Staphylococcus aureus (CNSA). Gram-negative infections were also noted. Conclusions: Eastern India has a high incidence of bone and joint infection. MRSA followed by CNSA is the most common isolate affecting large bones such as femur and tibia.

A robust method was developed using LC-ESI-MS/MS-based identification and quantification of 103 fortified pesticides in a mango fruit drink. Variations in QuEChERS extraction (without buffer, citrate, and/or acetate buffered) coupled with dispersive clean-up combinations were evaluated. Results showed 5 mL dilution and citrate buffered QuEChERS extraction with anhydrous (anhy) MgSO4 clean-up gave acceptable recovery for 100 pesticides @ 1 μg mL−1 fortification. The method was validated as per SANTE guidelines (SANTE/11813/2021). 95, 91, and 77 pesticides were satisfactorily recovered at 0.1, 0.05, and 0.01 μg mL−1 fortification with HorRat values ranging from 0.2–0.8 for the majority. The method showed matrix enhancement for 77 pesticides with a global uncertainty of 4.72%–23.89%. The reliability of the method was confirmed by real sample analysis of different brands of mango drinks available in the market. The greenness assessment by GAPI (Green Analytical Procedure Index) indicated the method was much greener than other contemporary methods.

To achieve higher crop yields and maintain environmental conservation, it is imperative to adopt novel agricultural methods that improve both the quantity and quality of produce. The following study focused on investigating the effectiveness of integrated use of biogas slurry and inorganic nutrigation on spinach growth and nutrient uptake through drip irrigation. A field experiment was conducted using a split-plot design; spinach was cultivated with seven different treatments: biogas slurry nutrigation (BSN) (T1), integrated inorganic + organic nutrigation: 40% Recommended dose of fertilizer (RDF) + BSN (T2), 60% RDF + BSN (T3), 80% RDF + BSN (T4), 100% RDF (T5), slurry broadcasting (SB) (T6), and control (T7). The results showed that spinach grown with (T4) 80% RDF + BSN exhibited optimum plant height and leaf count compared to spinach under other treatments and was at par with T5 100% RDF for crop parameters. T5 treated plants demonstrated the longest roots, followed by T4 treated plants. The highest recorded yield of 5.72 tons ha−1 was achieved in treatment T5 with 100% RDF, followed closely by T4 with 80% RDF + BSN at 5.52 tons ha−1 and T3 with 60% RDF + BSN at 5.36 tons ha−1. These three treatments exhibited comparable yields, showcasing the effectiveness of incorporating biogas slurry nutrigation in conjunction with inorganic fertilizer for achieving high spinach yields. In terms of macronutrient content in spinach leaves, significant differences were found for nitrogen (N), phosphorus (P), and potassium (K) contents. Incorporating biogas slurry into the soil modified microbial enzyme activities, specifically dehydrogenase and phosphatase. Normally, alkaline phosphatase shows greater activity than acidic phosphatase, but the addition of biogas slurry equalized the enzymatic activity of both, establishing a harmonized enzymatic profile. Our results suggest that fertilizing spinach with integrated biogas slurry nutrigation (if properly filtered) + inorganic nutrigation through drip nutrigation is a viable strategy for improving the morphological parameters and productivity of spinach while also contributing to environmental preservation and a reduction in production costs.

Loss of HLA-I gene expression in prostate cancer is associated with repressive chromatin states, which can be reversed by pharmacological DNMT and HDAC inhibition leading to increased activation of co-cultured tumor-specific CD8+ T-cells.

Water is the main constraint on agricultural output in arid- and semi-arid agroecologies, especially in winter. Hydrogels reduce percolation and evaporation losses in the soil root zone and provide a more continuous stream of moisture to the crop, enhancing water retention. Therefore, the impact of recently developed cross-linked hydrogels (SPG 1118 and Pusa Jal Nidhi) on the performance of red cabbage (Brassica oleracea L.) was studied during 2020–21 and 2021–22 using a spilt-plot design with three replications under drip fertigation. The study results revealed that application of full irrigation (100% ETc) exhibited significantly higher head yield of ~16% compared to limited irrigation (75% ETc). Content of NPK in leaf and head were significantly higher under full irrigation (1.31–1.38% of N in head and 2.49–2.62% of N in leaf) compared to limited irrigation. Among modified application of hydrogels, seedling root dipping with hydrogel SPG 1118 exhibited ~12% higher head yield over control plots. Application of SPG 1118 recorded ~4–5% and 9–14% of higher head yield over Pusa Jal Nidhi and control plots. Likewise, rootapplied SPG 1118 hydrogel had higher plant height, number of leaves, and NPK uptake in head and leaf than soilapplied Pusa hydrogel and control. Due to vertical root development, control plots had longer roots (19.0–20.8 cm) and smaller root volumes (51.8–53.3 g) than hydrogel plots. Thus, root application of SPG 1118 with drip irrigation and fertigation increases hydrogel efficacy and reduces water and nutrient losses from evaporation and deep percolation.

Abstract Environmental crises, declining factor productivity, and shrinking natural resource is a threat to global agricultural sustainability. The task is much more daunting in the Indo-Gangetic northern plains of India, where depletion of the underground water table and erratic rains due to the changing climate pose a major challenge to agriculture. To address these challenges a field investigation was carried out during 2016–2018 to test the efficacy of biopolymeric superabsorbent hydrogels namely Pusa Hydrogel (P-hydrogel: a semi-synthetic cellulose derivative-based product) and kaolin derivative of Pusa Hydrogel (K-hydrogel: semi-synthetic cellulose derivative) to assess their effect on crop and water productivity, soil moisture, root dynamics, and economics of soybean (Glycine max L.)–wheat (Triticum aestivum L.) system under three irrigation regimes namely full irrigation, limited irrigation and rainfed. The results revealed that the full irrigation along with P-hydrogel led to enhanced grain yield, biomass yield, and water productivity (WP) of soybean (1.61–10.5%, 2.2–9.5%, and 2.15–21.8%, respectively) and wheat (11.1–18.3%, 12–54% and 11.1–13.1%, respectively) over control plots. Likewise, under water stressed plots of rainfed conditions with P-hydrogel exhibited 52.7 and 20.6% higher system yields (in terms of wheat equivalent yield) over control and other combinations during the respective study years. Whereas the magnitude of increase in system yield under limited irrigation with P-hydrogel was ~ 15.1% and under full irrigation with P-hydrogel was 8.0–19.4%. Plots treated with P-hydrogel retained 3.0–5.0% higher soil moisture compared to no-hydrogel plots, while K-hydrogel treated plots held the lower moisture (4.0–6.0%) than the control. In terms of profitability, full irrigation along with P-hydrogel plots registered 12.97% higher economic returns over control. The results suggested that P-hydrogel (2.5 kg ha−1) reduces runoff water loss in full irrigation applied plots and retained more water, where loss of water is more thus reduces number of irrigations. Hence P-hydrogel with irrigation water is a viable option for sustainable production of soybean-wheat systems in the Indo-Gangetic plains of India and other similar eco-regions of the world.

Abstract Introduction PD-L1 expression in non-small cell lung cancer (NSCLC) predicts response to immune checkpoint blockade, however is an imperfect biomarker given tumor heterogeneity, and the antigen presentation pathway requiring other components including HLA I expression. HLA I downregulation may contribute to resistance, warranting its evaluation in attempts to guide patient selection. In addition, earlier detection of acquired resistance could prompt earlier change in treatment and prolong patient survival. Analysis of circulating tumor cells (CTCs) captures heterogeneity across multiple sites of metastases, enables detection of changes in tumor burden that precede radiographic response, and can be obtained in serial fashion. Methods To quantify the expression of both PD-L1 and HLA I on CTCs, we developed exclusion-based sample preparation technology, achieving high-yield with gentle magnetic movement of antibody-labeled cells through virtual barriers of surface tension. To achieve clinical-grade quantification of rare cells, we employ high quality fluorescence microscopy image acquisition and automated image analysis together termed quantitative microscopy. Results In preparation for clinical laboratory implementation, we demonstrate high precision and accuracy of these methodologies using a diverse set of control materials. Preliminary testing of CTCs isolated from patients with NSCLC demonstrate heterogeneity in PD-L1 and HLA I expression and promising clinical value in predicting PFS in response to PD-L1 targeted therapies. Conclusions By confirming high performance, we ensure compatibility for clinical laboratory implementation and future application to better predict and detect resistance to PD-L1 targeted therapy in patients with NSCLC.

Although therapeutic options for patients with advanced renal cell carcinoma (RCC) have increased in the past decade, no biomarkers are yet available for patient stratification or evaluation of therapy resistance. Given the dynamic and heterogeneous nature of clear cell RCC (ccRCC), tumor biopsies provide limited clinical utility, but liquid biopsies could overcome these limitations. Prior liquid biopsy approaches have lacked clinically relevant detection rates for patients with ccRCC. This study employed ccRCC‐specific markers, CAIX and CAXII, to identify circulating tumor cells (CTC) from patients with metastatic ccRCC. Distinct subtypes of ccRCC CTCs were evaluated for PD‐L1 and HLA‐I expression and correlated with patient response to therapy. CTC enumeration and expression of PD‐L1 and HLA‐I correlated with disease progression and treatment response, respectively. Longitudinal evaluation of a subset of patients demonstrated potential for CTC enumeration to serve as a pharmacodynamic biomarker. Further evaluation of phenotypic heterogeneity among CTCs is needed to better understand the clinical utility of this new biomarker.

BackgroundAntioxidants detain the development and proliferation of various non-communicable diseases (NCDs). γ-oryzanol, a group of steryl ferulates and caffeates, is a major antioxidant present in rice grain with proven health benefits. The present study evaluated the distribution and dynamics of γ-oryzanol and its components in spatial and temporal scales and also delineated the effect of processing and cooking on its retention.MethodsSix rice varieties (four Basmati and two non-Basmati) belonging to indica group were analyzed at spatial scale in four different tissues (leaf blades, leaf sheaths, peduncle and spikelets) and temporal scale at three developmental stages (booting, milky and dough). Additionally, the matured grains were fractioned into husk, embryo, bran, and endosperm to assess differential accumulation in these tissues. Further, milling and cooking of the samples was done to assess the retention upon processing. After extraction of γ-oryzanol by solvent extraction method, individual components were identified by UPLC-QToF-ESI-MS and quantified by RP-HPLC.ResultsThe non-seed tissues were significantly different from the seed tissues for composition and quantitative variation of γ-oryzanol. Cycloartenyl caffeate was predominant in all the non-seed tissues during the three developmental stages while it showed significant reduction during the growth progression toward maturity and was totally absent in the matured grains. In contrary, the 24-methylenecycloartanyl ferulate, campesteryl ferulate and β-sitosteryl ferulate showed significant increment toward the growth progression to maturity. Milling caused significant reduction, retaining only an average of 58.77% γ-oryzanol. Cooking of brown rice in excess water showed relatively lower average retention (43.31%) to samples cooked in minimal water (54.42%). Cooked milled rice showed least mean retention of 21.66%.ConclusionThe results demonstrate prominent compositional variation of γ-oryzanol during different growth stages. For the first time, the study demonstrated that ferulate esters of γ-oryzanol were predominant in the seed tissues while caffeate esters were dominant in non-seed tissues. Basmati cultivars show differential expression of γ-oryzanol and its components compared to non-Basmati cultivars. Cooking in excess water causes maximum degradation of γ-oryzanol. Post-harvest losses due to milling and cooking indicate the necessity of biofortification for γ-oryzanol content in rice grain.

Background Neuroendocrine prostate cancer (NEPC) is an aggressive subtype, the presence of which changes the prognosis and management of metastatic prostate cancer.Methods We performed analytical validation of a Circulating Tumor Cell (CTC) multiplex RNA qPCR assay to identify the limit of quantification (LOQ) in cell lines, synthetic cDNA, and patient samples. We next profiled 116 longitudinal samples from a prospectively collected institutional cohort of 17 patients with metastatic prostate cancer (7 NEPC, 10 adenocarcinoma) as well as 265 samples from 139 patients enrolled in 3 adenocarcinoma phase II trials of androgen receptor signaling inhibitors (ARSIs). We assessed a NEPC liquid biomarker via the presence of neuroendocrine markers and the absence of androgen receptor (AR) target genes.Results Using the analytical validation LOQ, liquid biomarker NEPC detection in the longitudinal cohort had a per-sample sensitivity of 51.35% and a specificity of 91.14%. However, when we incorporated the serial information from multiple liquid biopsies per patient, a unique aspect of this study, the per-patient predictions were 100% accurate, with a receiver-operating-curve (ROC) AUC of 1. In the adenocarcinoma ARSI trials, the presence of neuroendocrine markers, even while AR target gene expression was retained, was a strong negative prognostic factor.Conclusion Our analytically validated CTC biomarker can detect NEPC with high diagnostic accuracy when leveraging serial samples that are only feasible using liquid biopsies. In developing a Trichoderma viride-based biocontrol program for Fusarium wilt disease in chickpea, the choice of the quality formulation is imperative. In the present study, two types of formulations i.e. powder for seed treatment (TvP) and tablet for direct application (TvT), employing T. viride as the biocontrol agent, were evaluated for their ability to control chickpea wilt under field conditions at three dosages i.e. recommended (RD), double of recommended (DD) and half of recommended (1/2 RD). A screening study for the antagonistic fungi strains based on volatile and non-volatile bioassays revealed that T. viride ITCC 7764 has the most potential among the five strains tested (ITCC 6889, ITCC 7204, ITCC 7764, ITCC 7847, ITCC 8276), which was then used to develop the TvP and TvT formulations. Gas Chromatography-Mass Spectrometry (GC-MS) analysis of volatile organic compounds (VOCs) of T. viride strain confirmed the highest abundance of compositions comprising octan-3-one (13.92%), 3-octanol (10.57%), and 1-octen-3-ol (9.40%) in the most potential T. viride 7764. Further Physico-chemical characterization by standard Collaborative International Pesticides Analytical Council (CIPAC) methods revealed the optimized TvP formulation to be free flowing at pH 6.50, with a density of 0.732 g cm-3. The TvT formulation showed a pH value of 7.16 and density of 0.0017 g cm-3 for a complete disintegration time of 22.5 min. The biocontrol potential of TvP formulation was found to be superior to that of TvT formulation in terms of both seed germination and wilt incidence in chickpea under field conditions. However, both the developed formulations (TvP and TvT) expressed greater bioefficacy compared to the synthetic fungicide (Carbendazim 50% WP) and the conventional talc-based formulation. Further research should be carried out on the compatibility of the developed products with other agrochemicals of synthetic or natural origin to develop an integrated disease management (IDM) schedule in chickpea.

Citrus fruits fall in the category of those commercially grown fruits that constitute an excellent repository of phytochemicals and biologically active compounds, with health-promoting properties. Processing of fruit results in generation of large amounts of waste, which are fed to animals or disposed of, increasing the burden on the environment. However, due to its richness in valuable compounds, citrus fruit waste viz. peels (flavedo and albedo), seeds, and pomace are considered potent bio-resource materials for various uses in the food and non-food sectors. The inherent bioactive compounds present in citrus waste can be used as food additive, encapsulant, nanoparticle, prebiotic, pectin source, essential oil, polyphenol, carotenoid, or dietary fiber. It can also be used as a natural ingredient for cosmetics, medicines, packaging materials, and synthetic fuels. Use as bio-absorbents, biofertilizers, biodiesel, biogas, and bioethanol are some other non-food applications of citrus waste. Irrespective, citrus waste is considered as an ecological risk, alongside other types of waste. Considering this risk, some strategies have recently been developed to reduce its adverse effects. This review on the same lines covers all possible effective and economical ways of valorization of citrus waste in the food and non-food sectors.

Xanthomas are the subcutaneous lipid deposits which can be seen in normolipedemic or hyperlipidemic conditions. Xanathomas are usually of single type and presence of polymorphic xanthomas arise suspicion of dyslipidemia. We report a case of young girl presenting with pleomorphic xanthomas (Xanthelasma palpebrarum, intertriginous Xanthomas, tendon Xanthomas, and eruptive xanthomas), hypothyroidism and multiple cardiac valvular involvements with very high serum total cholesterol and low-density lipoprotein along with deranged lipid profile in both parents. Pleomorphic xanthoma arouses a suspicion and helped in making a diagnosis of rare homozygous familial hypercholesterolemia.

Introduction: Periodontal disease is considered to be one of the most common oral conditions in human population sharing an equal prevalence rate in both developed and developing countries. It is observed that in India, more than 800 million beedis are sold every year; hence, beedi making is a big industry involving approximately 4.4 million workers, and 76% of these workers are women. Objectives: The objectives of this study were to assess the frequency of gingival and periodontal disease and to evaluate the treatment needs in labors working in a beedi factory. Materials and Methods: A cross-sectional study was conducted on 1000 daily wage labors working in a beedi factory resided at Patna, Bihar. Subjects aged between 19 and 60 years were randomly selected for the study. The community periodontal index of treatment needs (CPITN) index with CPTIN probe along with the demographic data was recorded in daylight. Results: None of the study subjects was found to fall in category community periodontal index-1 (CPI-1), 67.2% of the study population was found to be in CPI-2, 12.3% were found to be in CPI-3, and 9.7% of the study subjects scored CPI-4. For the treatment needs, majority of the subjects were in TN2 category; however, with advancing age, there was increase in the percentage of population requiring TN3 treatment needs. Conclusion: CPITN index along with the demographic data was recorded in daylight. The high prevalence rate of periodontal disease in these beedi factory workers was found, which increased with advancing age. The treatment need index also showed rise in the requirement of complex procedures with advancing age, suggesting the role of dental education and regular checkups in these population groups.

Background: Stroke can broadly be categorized into ischemic or hemorrhagic. Ischemic stroke accounts for 85% of cerebrovascular accidents (CVAs), whereas hemorrhagic stroke accounts for 15% of CVAs. Stroke is broadly associated with loss of sensation or unilateral paralysis of orofacial structures. Objectives: The present study was conducted to evaluate the prevalence of various oral features in patients with ischemic and hemorrhagic stroke. Materials and Methods: One hundred patients diagnosed with stroke admitted in the intensive care unit were included in the study. The evaluation of oral manifestations and their prevalence was done by a well-experienced oral medicine expert deputed in the dental department of the hospital. A single examiner performed all oral evaluations. Results: The mean and median for the age were 60.8 and 59. Sixty of 100 patients were male, whereas 38 were female. Forty patients had hemorrhagic stroke, whereas 60 had ischemic stroke. Senenty-eight patients of 100 had features of periodontitis, 90 of 100 patients presented with halitosis, 79 presented with caries, 83 patients had positive signs of tongue hypermobility, and 75 patients had dysphagia. Conclusion: Oral hygiene is the most neglected aspect during rehabilitation in stroke patients. It is critical for stroke patients to receive thorough oral care, as it can prevent other systemic ailments and potentially life-threatening complications like aspiration pneumonia.

Metabolic reprogramming during Dietary Restriction (DR) activates cytoprotective gene expression. Here the authors show that PUFAs generated during DR signal via the p38-MAPK pathway to enhance cytoprotective gene expression, contributing to increased longevity.

Animals integrate changes in external and internal environments to generate behavior. While neural circuits detecting external cues have been mapped, less is known about how internal states like hunger are integrated into behavioral outputs. Here, we use the nematode C. elegans to examine how changes in internal nutritional status affect chemosensory behaviors. We show that acute food deprivation leads to a reversible decline in repellent, but not attractant, sensitivity. This behavioral change requires two conserved transcription factors MML-1 (MondoA) and HLH-30 (TFEB), both of which translocate from the intestinal nuclei to the cytoplasm during food deprivation. Next, we identify the insulin-like peptide INS-31 as a candidate ligand relaying food-status signals from the intestine to other tissues. Further, we show that neurons likely use the DAF-2 insulin receptor and AGE-1/PI-3 Kinase, but not DAF-16/FOXO to integrate these intestine-released peptides. Altogether, our study shows how internal food status signals are integrated by transcription factors and intestine-neuron signaling to generate flexible behaviors via the gut-brain axis.

ABSTRACT OBJECTIVE To present the overall survival rate for lung cancer and identify the factors associated with early diagnosis of stage I and II lung cancer. METHODS This is a retrospective cohort study including individuals diagnosed with lung cancer, from January 2009 to December 2017, according to the cancer registry at UMass Memorial Medical Center. Five-year overall survival and its associated factors were identified by Kaplan–Meier curves and Cox’s proportional hazards model. Factors associated with diagnosing clinical stage I and II lung cancer were identified by bivariate and multivariate backward stepwise logistic regression (Log-likelihood ratio (LR)) at 95% confidence interval (CI). RESULTS The study was conducted with data on 2730 individuals aged 67.9 years on average, 51.5% of whom female, 92.3% white, and 6.6% never smoked. Five-year overall survival was 21%. Individuals diagnosed with early-stage disease had a 43% five-year survival rate compared to 8% for those diagnosed at late stages. Stage at diagnosis was the main factor associated with overall survival [HR = 4.08 (95%CI: 3.62–4.59)]. Factors associated with early diagnosis included patients older than 68 years [OR = 1.23 (95%CI: 1.04–1.45)], of the female gender [OR = 1.47 (95%CI: 1.24–1.73)], white [OR = 1.63 (95%CI: 1.16–2.30)], and never-smokers [OR = 1.37 (95%CI: 1.01–1.86)]; as well as tumors affecting the upper lobe [OR = 1.46 (95%CI: 1.24–1.73)]; adenocarcinoma [OR = 1.43 (95%CI: 1.21–1.69)]; and diagnosis after 2014 [OR = 1.61 (95%CI: 1.37–1.90)]. CONCLUSIONS Stage at diagnosis was the most decisive predictor for survival. Non-white and male individuals were more likely to be diagnosed at a late stage. Thus, promoting lung cancer early diagnosis by improving access to health care is vital to enhance overall survival for individuals with lung cancer.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) began churning out incredulous terror in December 2019. Within several months from its first detection in Wuhan, SARS-CoV-2 spread to the rest of the world through droplet infection, making it a pandemic situation and a healthcare emergency across the globe. The available treatment of COVID-19 was only symptomatic as the disease was new and no approved drug or vaccine was available. Another challenge with COVID-19 was the continuous mutation of the SARS-CoV-2 virus. Some repurposed drugs, such as hydroxychloroquine, chloroquine, and remdesivir, received emergency use authorization in various countries, but their clinical use is compromised with either severe and fatal adverse effects or nonavailability of sufficient clinical data. Molnupiravir was the first molecule approved for the treatment of COVID-19, followed by Paxlovid™, monoclonal antibodies (MAbs), and others. New molecules have variable therapeutic efficacy against different variants or strains of SARS-CoV-2, which require further investigations. The aim of this review is to provide in-depth information on new molecules and repurposed drugs with emphasis on their general description, mechanism of action (MOA), correlates of protection, dose and dosage form, route of administration, clinical trials, regulatory approval, and marketing authorizations.

Global warming is a serious threat to food security and severely affects plant growth, developmental processes, and, eventually, crop productivity. Respiratory metabolism plays a critical role in the adaptation of diverse stress in plants. Aconitase (ACO) is the main enzyme, which catalyzes the revocable isomerization of citrate to isocitrate in the Krebs cycle. The function of ACO gene family members has been extensively studied in model plants, for instance Arabidopsis. However, their role in plant developmental processes and various stress conditions largely remained unknown in other plant species. Thus, we identified 15 ACO genes in wheat to elucidate their function in plant developmental processes and different stress environments. The phylogenetic tree revealed that TaACO genes were classified into six groups. Further, gene structure analysis of TaACOs has shown a distinctive evolutionary path. Synteny analysis showed the 84 orthologous gene pairs in Brachypodium distachyon, Aegilops tauschii, Triticum dicoccoides, Oryza sativa, and Arabidopsis thaliana. Furthermore, Ka/Ks ratio revealed that most TaACO genes experienced strong purifying selection during evolution. Numerous cis-acting regulatory elements were detected in the TaACO promoters, which play a crucial role in plant development processes, phytohormone signaling, and are related to defense and stress. To understand the function of TaACO genes, the expression profiling of TaACO genes were investigated in different tissues, developmental stages, and stress conditions. The transcript per million values of TaACOs genes were retrieved from the Wheat Expression Browser Database. We noticed the differential expression of the TaACO genes in different tissues and various stress conditions. Moreover, gene ontology analysis has shown enrichment in the tricarboxylic acid metabolic process (GO:0072350), citrate metabolic process (GO:0006101), isocitrate metabolic process GO:0006102, carbohydrate metabolic (GO:0005975), and glyoxylate metabolic process (GO:0046487). Therefore, this study provided valuable insight into the ACO gene family in wheat and contributed to the further functional characterization of TaACO during different plant development processes and various stress conditions.

Introduction: One of the most common burn sequels is postburn contracture (PBC) neck. These contractures affect the patient significantly causing both functional limitations and distorted aesthetic appearance. Therefore, the reconstruction of the head-and-neck area is a challenge for surgeons to restore not only the function but also to improve the esthetic appearance. Materials and Methods: We conducted a tertiary hospital-based prospective study in 12 patients with cases of severe PBC that underwent split-thickness skin graft (SSG) cover as a treatment modality and evaluated the results after the operative procedure in terms of function and appearance of the neck and its complications. Result of the function evaluated with respect to the range of extension movement of the neck –good 120°or more, fair 90°–120°, and bad 120° in nine cases and fair esthetic appearance in eight cases and good in four cases. Conclusions: The excision of all scar tissue is possible only in few cases, but the incision releases were required in the majority of severe neck contractures. The STS grafting is a simple, reliable, and safe operation. Sheets of unmeshed SSG in the neck and lower face resulted in better postoperative neck function, the cosmetic appearance in terms of color match, and cervicomental angle.

Summary: The presence of nonhuman RNAs in man has been questioned and it is unclear if food-derived miRNAs cross into the circulation. In a large population study, we found nonhuman miRNAs in plasma by RNA sequencing and validated a small number of pine-pollen miRNAs by RT-qPCR in 2,776 people. The presence of these pine-pollen miRNAs associated with hay fever and not with overt cardiovascular or pulmonary disease. Using in vivo and in vitro models, we found that transmission of pollen-miRNAs into the circulation occurs via pulmonary transfer and this transfer was mediated by platelet-pulmonary vascular cell interactions and platelet pollen-DNA uptake. These data demonstrate that pollen-derived plant miRNAs can be horizontally transferred into the circulation via the pulmonary system in humans. Hexavalent chromium is a highly soluble environmental contaminant. It is a widespread anthropogenic chromium species that is 100 times more toxic than trivalent chromium. Leather, chrome plating, coal mining and paint industries are the major sources of hexavalent chromium in water. Hexavalent chromium is widely recognised as a carcinogen and mutagen in humans and other animals. It is also responsible for multiorgan damage, such as kidney damage, liver failure, heart failure, skin disease and lung dysfunction. The fate of the toxicity of hexavalent chromium depends on its oxidation state. The reduction of Cr (VI) to Cr (III) is responsible for the generation of reactive oxygen species (ROS) and chromium intermediate species, such as Cr (V) and Cr (IV). Reactive oxygen species (ROS) are responsible for oxidative tissue damage and the disruption of cell organelles, such as mitochondria, DNA, RNA and protein molecules. Cr (VI)-induced oxidative stress can be neutralised by the antioxidant system in human and animal cells. In this review, the authors summarise the Cr (VI) source, toxicity and antioxidant defence mechanism against Cr (VI)-induced reactive oxygen species (ROS).

Rheumatoid arthritis (RA) is a major global public health challenge, and novel therapies are required to combat it. Silver nanoparticles (AgNPs) have been employed as delivery vehicles of anti-inflammatory drugs for RA therapy, and it has been recently realized that AgNPs have anti-inflammatory action on their own. However, their conventional synthesis processes might result in cytotoxicity and environmental hazards. Instead, the use of natural products as a reducing and stabilizing agent in the biosynthesis of silver nanoparticles has arisen as an option to decrease the cytotoxic and environmental concerns associated with chemical synthesis of AgNPs. In this study, we challenged the efficacy of Commiphora mukul (guggul) aqueous extract as a reducing and/or capping agent for the biosynthesis of AgNPs. Guggul-mediated biosynthesized silver nanoparticles (G-AgNPs) were characterized via UV-vis spectroscopy, dynamic light scattering, and scanning electron microscopy. In addition, their anti-arthritic potential was evaluated in an adjuvant-induced arthritis (AIA) model. The fabricated NPs showed an absorption peak at 412 nm, corresponding to the typical surface plasmon resonance band of AgNPs. The synthesized G-AgNPs were nearly spherical, with a particle size of 337.6 ± 12.1 nm and a negative surface charge (−18.9 ± 1.8 mV). In AIA rat model, synthesized G-AgNPs exerted a potent anti-inflammatory action, as manifested by a remarkable reduction in paw volume (>40%) along with elicitation of a minimal arthritic score, compared to control rats. In addition, when compared to arthritic rats, treatment with G-AgNPs efficiently restored the activity of antioxidant enzyme, superoxide dismutase, and catalase, indicating the efficiency of synthesized G-AgNPs in alleviating the oxidative stress associated with RA. Finally, histological examination revealed comparatively lower inflammatory cells infiltration in ankle joint tissue upon treatment with G-AgNPs. Collectively, biosynthesized G-AgNPs might represent a plausible therapeutic option for the management of RA.

Environmental concerns related to synthetic pesticides and the emphasis on the adoption of an integrated pest management concept as a cardinal principle have strengthened the focus of global research and development on botanical pesticides. A scientific understanding of the mode of action of biomolecules over a range of pests is key to the successful development of biopesticides. The present investigation focuses on the in silico protein-ligand interactions of allyl isothiocyanate (AITC), a major constituent of black mustard (Brassica nigra) essential oil (MEO) against two pests, namely, Meloidogyne incognita (Mi) and Fusarium oxysporum f. sp. lycopersici (Fol), that cause severe yield losses in agricultural crops, especially in vegetables. The in vitro bioassay results of MEO against Mi exhibited an exposure time dependent on the lethal concentration causing 50% mortality (LC50) values of 47.7, 30.3, and 20.4 μg ml−1 at 24, 48, and 72 h of exposure, respectively. The study revealed short-term nematostatic activity at lower concentrations, with nematicidal activity at higher concentrations upon prolonged exposure. Black mustard essential oil displayed excellent in vitro Fol mycelial growth inhibition, with an effective concentration to cause 50% inhibition (EC50) value of 6.42 μg ml−1. In order to decipher the mechanism of action of MEO, its major component, AITC (87.6%), which was identified by gas chromatography–mass spectrometry (GC-MS), was subjected to in silico docking and simulation studies against seven and eight putative target proteins of Mi and Fol, respectively. Allyl isothiocyanate exhibited the highest binding affinity with the binding sites of acetyl cholinesterase (AChE), followed by odorant response gene-1 (ODR1) and neuropeptide G-protein coupled receptor (nGPCR) in Mi, suggesting the possible suppression of neurotransmission and chemosensing functions. Among the target proteins of Fol, AITC was the most effective protein in blocking chitin synthase (CS), followed by 2,3-dihydroxy benzoic acid decarboxylase (6m53) and trypsinase (1try), thus inferring these as the principal molecular targets of fungal growth. Taken together, the study establishes the potential of MEO as a novel biopesticide lead, which will be utilized further to manage the Mi–Fol disease complex.

Kinnow mandarin (Citrus reticulate L.) peels are a storehouse of well-known bioactive compounds, viz., polyphenols, flavonoids, carotenoids, limonoids, and tocopherol, which exhibit an effective antioxidant capacity. However, naringin is the most predominant bitter flavanone compound found in Kinnow peels that causes their bitterness. It prohibits the effective utilization of peels in food-based products. In the present study, a novel approach for the debittering of Kinnow peels has been established to tackle this problem. A comparative evaluation of the different debittering methods (chemical, microbial, and ultrasound-assisted microbial treatments) used on Kinnow peel naringin and bioactive compounds was conducted. Among the chemical and microbial method; solid-state fermentation with A. niger led to greater extraction of naringin content (7.08 mg/g) from kinnow peels. Moreover, the numerical process optimization of ultrasound-assisted microbial debittering was performed by the Box–Behnken design (BBD) of a response surface methodology to maximize naringin hydrolysis. Among all three debittering methods, ultrasound-assisted microbial debittering led to a greater hydrolysis of naringin content and reduced processing time. The optimum conditions were ultrasound temperature (40 °C), time (30 min), and A. niger koji extract (1.45%) for the maximum extraction rate of naringin (11.91 mg/g). These debittered Kinnow peels can be utilized as raw material to develop therapeutic food products having a high phytochemical composition without any off-flavors or bitterness.

Nematicidal potential of essential oils (EOs) has been widely reported. Terpenoids present in most of the essential oils have been reported responsible for their bioactivity though very less is known about their modes of action. In the present study, an in vitro screening of nine Eos, namely, Citrus sinensis (OEO), Myrtus communis (MTEO), Eucalyptus citriodora (CEO), Melaleuca alternifolia (TEO), Acorus calamus (AEO), Commiphora myrrha (MREO), Cymbopogon nardus (CNEO), Artemisia absinthium (WEO), and Pogostemon cablin (PEO) against Meloidogyne incognita revealed OEO, CNEO, and TEO as most effective with LC50 39.37, 43.22, and 76.28 μg ml–1 respectively. EOs had varying compositions of mono- and sesquiterpenes determined by gas chromatography-mass spectrometry (GC-MS) analysis. The in silico molecular interactions screening of major EO constituents and the seven selected target proteins of the nematode indicated highest binding affinity of geraniol-ODR1 (odorant response gene 1) complex (ΔG = -36.9 kcal mol–1), due to extensive H-bonding, hydrophobic and π-alkyl interactions. The relative binding affinity followed the order: geraniol-ODR1 > β-terpineol-ODR1 > citronellal-ODR1 > l-limonene-ODR1 > γ-terpinene-ODR1. Taken together, the cumulative in vitro and computational bioefficacy analysis related to the chemoprofiles of EOs provides useful leads on harnessing the potential of EOs as bionematicides. The insight on biochemical ligand–target protein interactions described in the present work will be helpful in logical selection of biomolecules and essential oils for development of practically viable bionematicidal products.
