Plant extracellular vesicles (EVs) are membrane bound organelles involved mainly in intercellular communications and defense responses against pathogens. Recent studies have demonstrated the presence of proteins, nucleic acids including small RNAs, and lipids along with other metabolites in plant EVs. In this paper, we described the isolation and characterization of extracellular vesicles from Sorghum bicolor. Nanoparticle tracking analysis, dynamic light scattering, and cryo-electron tomography showed the presence of a heterogeneous population of EVs isolated from the apoplastic wash of sorghum leaves. Cryo-electron microscopy revealed that EVs had a median size of 110 nm and distinct populations of vesicles with single or multiple lipid bilayers and low or high amounts of contents. The heterogeneity was further supported by data showing that only a subset of EVs that were stained with a membrane dye, Potomac Gold, were also stained with the membrane-permeant esterase-dependent dye, Calcein-acetoxymethyl ester. Proteomic analysis identified 437 proteins that were enriched in multiple EV isolations, with the majority of these also being found in the EV proteome of Arabidopsis. These data suggest a partial conservation of EV contents and function between the monocot, sorghum, and a distantly related eudicot, Arabidopsis.

Energy production from fossil fuels is gradually phased out as many countries aim to transition to a low-carbon society. As society and technology are intertwined, phasing out fossil fuels impacts people and communities. Especially those who heavily rely on the fossil fuel industry will be worse off. Therefore, calls are being made for ajust transitionthat ensures the rehabilitation of workers, regions, and communities negatively affected by fossil fuel industry closures. We argue that spatial justice can help inform just transition’s theoretical and practical aspects. Therefore, a spatial justice approach should be a prerequisite for a just transition. The concept of spatial justice is intertwined with the social justice principles of procedural, distributive, and restorative justice, which are central to the current conceptual understanding of just transition. We use the case of the closure of peat-based electricity production in rural Ireland to demonstrate how a spatial justice approach can underpin a just transition and how it can help with practicalities like identifying and addressing the issues and concerns in local communities. To ensure a just transition, a spatial justice approach is needed to identify and address the deeper problems affecting the resiliency of rural and mono-industrial regions dependent on fossil fuels.

This chapter considers aspects of energy justice and spatial justice in large-scale plantation agriculture for bioenergy crops in rural Yucatan, Mexico. The government promoted bioenergy projects for rural revitalization benefits while simultaneously producing feedstock for renewable biofuels. This research reveals how local communities were disenfranchised from their access to common property resources. We show how unsuccessful renewable energy projects further aggravated precarity conditions in rural communities as they bore the unequal burden of the projects’ failure. Additionally, affected communities were unable to participate in the projects’ planning to express their opinions on the impacts of the projects. Moreover, their energy poverty challenges remained unmitigated. Therefore, we argue that attaining the tenets of energy justice and just transition, especially in the rural areas of the global South, remains unmet.

Packaging is an indispensable tool for maintaining the quality of perishable food products. Petroleum-based packaging has been used as the material of choice for a long time. However, because these materials are nonbiodegradable they pose serious environmental risks. A suitable alternative is the use of biodegradable polymers. In recent years, biodegradable polymers have been found to have innumerous applications in packaging, agriculture, medicine, and other areas. In the food industry these polymers can be used as edible films and coatings, packaging materials, carriers of antimicrobial and antioxidative materials, etc. Nanocomposites can also be applied to food packaging in the future. A novel use of biopolymers is as a nanodelivery vehicle for delivering nutraceuticals and drugs orally. These are made from polysaccharides, lipids, and proteins. This chapter aims to study different aspects of biopolymers, including their classification, properties, and applications with special emphasis on industrial applications.

Radon, a naturally occurring radioactive gas that can accumulate inside dwellings, represents the second biggest cause of lung cancer globally. In Ireland, radon is linked to approximately 300 lung cancer cases every year, equating to 12% of all lung cancer deaths. Despite the health risks posed by radon air pollution, Ireland lacks well-defined and universally applicable air pollution-related public health policies. Through purposive literature sampling, we critically examine the case of indoor radon policy development in Ireland. Specifically, we analyse the evidence-based policymaking process relating to indoor radon pollution from three different knowledge dimensions, namely political, scientific, and practical knowledge. In doing so, we identify various challenges inherent to pollution-related public policymaking. We highlight the difficulties of balancing and integrating information from multiple disciplines and perspectives and argue that input from multiple scientific areas is crucial, but can only be achieved through continued, dialogic communication between stakeholders. On the basis of our analysis, we suggest that a transdisciplinary perspective, defined as a holistic approach which subordinates disciplines and looks at the dynamics of whole systems, will allow evidence-based policymaking to be effective. We end with recommendations for evidence-based policymaking when it comes to public health hazards such as radon, which are applicable to sustainable air pollution management beyond Ireland.

Modular assembly and heterologous expression in the moss Physcomitrella patens of pairs of diterpene synthases results in accumulation of modern land plant diterpenoids. Physcomitrella patens is a representative of the ancient bryophyte plant lineage with a genome size of 511 Mb, dominant haploid life cycle and limited chemical and metabolic complexity. For these plants, exceptional capacity for genome editing through homologous recombination is met with recently demonstrated in vivo assembly of multiple heterologous DNA fragments. These traits earlier made P. patens an attractive choice as a biotechnological chassis for photosynthesis-driven production of recombinant peptides. The lack of diterpene gibberellic acid phytohormones in P. patens combined with the recent targeted disruption of the single bifunctional diterpene synthase yielded lines devoid of endogenous diterpenoid metabolites and well-suited for engineering of terpenoid production. Here, we mimicked the modular nature of diterpene biosynthetic pathways found in modern land plants by developing a flexible pipeline to install three combinations of class II and class I diterpene synthases in P. patens to access industrially relevant diterpene biomaterials. In addition to a well-established neutral locus for targeted integration, we also explored loci created by a class of Long Terminal Repeat Retrotransposon present at moderate number in the genome of P. patens. Assembly of the pathways and production of the enzymes from the neutral locus led to accumulation of diterpenes matching the reported activities in the angiosperm sources. In contrast, insights gained with the retrotransposon loci indicate their suitability for targeting, but reveal potentially inherent complications which may require adaptation of the experimental design.

Fresh-cut fruits and vegetables are highly perishable commodities. Packaging forms an important tool to maintain the shelf life of packed fresh-cut agricultural produce. Petroleum-based films are conventionally used for fresh fruits and vegetables. However, being nonbiodegradable and derivability from nonrenewable resources, these films lead towards serious ecological problems. To address this issue, various efforts have been focused on renewable and biodegradable films obtained from biopolymers. Widely studied biopolymers for film preparation are derived from biomass (gelatin, starch, cellulose, etc.), microbes (polyhydroxyalkanoates), and bio-derived monomers (polylactic acid). However, such films possess poor mechanical and barrier properties as compared to their commercial counterparts. Incorporation of various additives has been proposed to improve the film characteristics. In the present review, comprehensive information has been provided on different methodologies for film fabrication, properties, and applications of stand-alone bio-based films for packaging of fresh-cut produce. Furthermore, successful commercial implementation of such film is also summarized.

Catalyzing stereo- and regio-specific oxidation of inert hydrocarbon backbones, and a range of more exotic reactions inherently difficult in formal chemical synthesis, cytochromes P450 (P450s) offer outstanding potential for biotechnological engineering. Plants and their dazzling diversity of specialized metabolites have emerged as rich repository for functional P450s with the advances of deep transcriptomics and genome wide discovery. P450s are of outstanding interest for understanding chemical diversification throughout evolution, for gaining mechanistic insights through the study of their structure–function relationship, and for exploitation in Synthetic Biology. In this review, we highlight recent developments and examples in the discovery of plant P450s involved in the biosynthesis of industrially relevant monoterpenoids, sesquiterpenoids, diterpenoids and triterpenoids, throughout 2016 and early 2017. Examples were selected to illustrate the spectrum of value from commodity chemicals, flavor and fragrance compounds to pharmacologically active terpenoids. We focus on a recently emerging theme, where P450s control metabolic bifurcations and chemical diversity of the final product profile, either within a pathway, or through neo-functionalization in related species. The implications may inform approaches for rational assembly of recombinant pathways, biotechnological production of high value terpenoids and generation of novel chemical entities. Various studies have been focused on seaweeds derived polysaccharides based composites because of its renewability and sustainability for food packaging and pharmaceutical applications including tissue engineering, drug delivery, and wound dressing. Alginate, carrageenan, and agar are widely used for this purpose due to their biocompatibility, availability, gelling capacity, and encapsulation efficiency. Essential oils (like oregano, clove, lemongrass, etc.) as antimicrobial and antioxidant agent, biopolymer (like starch, cellulose, chitosan, etc.), and nanoparticles (organically modified and unmodified inorganic nanoclays, nano-cellulose, carbon nanotubes) as reinforcing material are frequently used for the fabrication of seaweed based materials. Composites have an edge over pure polymer based material in terms of mechanical and barrier properties, controlled release of drugs, and adsorption efficiency. This review comprehensively addresses different types of additives and their impact on various functional properties of seaweed based composites, their methods of incorporation, and applications with special emphasis on food and pharmaceutical usage.

BACKGROUND Ultrasound biomicroscopy (UBM) is a non-invasive procedure to visualise the structures of the anterior segment using high frequency ultrasound. Any abnormalities in the structures like the iris, ciliary body and zonules can be easily detected. In case of corneal oedema, the Descemet’s membrane can be detected with UBM than the slit-lamp biomicroscopy. It plays a useful diagnostic tool in glaucoma. It differentiates between open and closed angle closure. It also helps in the diagnosis of plateau iris configuration and malignant glaucoma. Bleb status and tenon’s cyst can be visualized by the UBM. It plays a major role to rule out angle recession or cyclodialysis cleft in cases of trauma. Zonular dialysis and iridodialysis as well as the integrity of the posterior capsule can be assessed with UBM. It is the only modality which can easily diagnose pars plannits or cyclitic membranes noninvasively. In cases of unexplained hypotony, it helps to diagnose ciliary body atrophy or traction on the ciliary body. This article is a short overview describing the indications and uses of anterior segment pathologies.

Seaweed and cellulose are promising natural polymers. This article reviews the basic information and recent developments of both seaweed and cellulose biopolymer materials as well as analyses the feasible formation of seaweed/cellulose composite films. Seaweed and cellulose both exhibit interesting film-forming properties. Nevertheless, seaweed has poor water vapour barrier and mechanical properties, whereas cellulose is neither meltable nor soluble in water or common organic solvents due to its highly crystalline structure. Therefore, modification of these hydrocolloids has been done to exploit their useful properties. Blending of biopolymers is a must recommended approach to improve the desired characteristics. From the review, seaweed is well compatible with cellulose, which possesses excellent mechanical strength and water resistance properties. Moreover, seaweed/cellulose composite films can prolong a product’s shelf life while maintaining its biodegradability. Additionally, the films show potential in contributing to the bioeconomy. In order to widen seaweed and cellulose in biocomposite application across various industries, some of the viewpoints are highlighted to be focused for future developments and applications.

In the last decade, jatropha-based bioenergy projects have gotten significant attention as a solution to various social, economic, and environmental problems. Jatropha's popularity stemmed out from different discourses, some real and some perceived, in scientific and non-scientific literature. These discourses positioned jatropha as a crop helpful in producing biodiesel and protecting sustainability by reducing greenhouse gas emissions compared to fossil fuels and increasing local, rural development by creating jobs. Consequently, many countries established national policies that incentivized the establishment of jatropha as a bioenergy feedstock crop. In this paper, we explore the case of jatropha bioenergy development in Yucatan, Mexico and argue that the popular discourse around jatropha as a sustainability and rural development tool is flawed. Analyzing our results from 70 semi-structured interviews with community members belonging to a region where plantation-scale jatropha projects were introduced, we found that these projects did not have many significant social sustainability benefits. We conclude from our case that by just adding bioenergy projects cannot help achieve social sustainability in rural areas alone. In ensuring social sustainability of bioenergy projects, future policymaking processes should have a more comprehensive understanding of the rural socioeconomic problems where such projects are promoted and use bioenergy projects as one of the many solutions to local problems rather than creating such policies based just on popular discourses.

Public support is critical to renewable energy sector growth, an important element of reducing fossil fuel dependence and mitigating climate change. Prevalent understandings of public support for renewable energy projects often work within a binary framework of acceptance and non-acceptance, arguably unable to capture the nuances of localized public responses to specific projects. Taking a place-based approach and insights from social representation theory, we report on public responses to wood-based electricity production in Wisconsin, USA. Findings indicate that public responses are tied to social and cultural contexts, varying in relation to community histories and identities shaped by other community resources. These results suggest that public perceptions of renewable energy technologies are shaped by representations formed in socio-spatial context, offering insight to inform future decisions in the renewable energy policy process.

Glucose-6-phosphate dehydrogenase (G6PDH) can initiate the glucose-6-phosphate (G6P) shunt around the Calvin–Benson cycle. To understand the regulation of flux through this pathway, we have characterized the biochemical parameters and redox regulation of the three functional plastidic isoforms of Arabidopsis G6PDH. When purified, recombinant proteins were measured, all three exhibited significant substrate inhibition by G6P but not NADP+, making the determination of enzyme kinetic parameters complex. We found that the half-saturation concentration of G6PDH isoform 1 is increased under reducing conditions. The other two isoforms exhibit less redox regulation, however, isoform 2 is strongly inhibited by NADPH. Redox regulation of G6PDH1 can be partially reversed by hydrogen peroxide or protected against by the presence of its substrate, G6P. Overall, our results support the conclusion that G6PDH can have significant activity throughout the day and can be dynamically regulated to allow or prevent flux through the glucose-6-phosphate shunt.

Cytosolic lipid droplets are endoplasmic reticulum-derived organelles typically found in seeds as reservoirs for physiological energy and carbon to fuel germination. Here, we report synthetic biology approaches to co-produce high-value sesqui- or diterpenoids together with lipid droplets in plant leaves. The formation of cytosolic lipid droplets is enhanced in the transient Nicotiana benthamiana system through ectopic production of WRINKLED1, a key regulator of plastid fatty acid biosynthesis, and a microalgal lipid droplet surface protein. Engineering of the pathways providing the universal C5-building blocks for terpenoids and installation of terpenoid biosynthetic pathways through direction of the enzymes to native and non-native compartments boost the production of target terpenoids. We show that anchoring of distinct biosynthetic steps onto the surface of lipid droplets leads to efficient production of terpenoid scaffolds and functionalized terpenoids. The co-produced lipid droplets “trap” the terpenoids in the cells., Cytosolic lipid droplets are ER-derived organelles that accumulate in seeds to fuel germination. Here, Sadre et al. show that by boosting lipid production and anchoring key terpenoid biosynthetic enzymes to the droplet surface, an efficient terpenoid production platform can be established in leaves.

In 2015, the adoption of resolution A/RES/70/1 2030, Transforming our world: the 2030 Agenda for Sustainable Development (UN Agenda 2030) was an important statement of future intent for UN member states. The Agenda touches the entire geography of the globe covering potentially all aspects of how society, economy, the environment and the polity are organized. The working philosophy of our approach is based on the existing open governance adopted by the UN HLPF at a global level. We wish to explore a form of interoperability of public policy dialogues at national and global levels. By exploring HLPF structures in national parliaments and making them interoperable with the UN HLPF we are proposing how a body like the UN HLPF can induce ‘whole-of-society’ participation at different levels, but particularly at the binding level of the nation-state. This has the potential to create very effective global ‘whole-of-society’ partnerships, led by governments that set, implement and review sustainable development public policies around the globe. We will use Ireland as a case study to show how our proposals might be integrated to the existing policymaking processes without being overtly disruptive. There are available reports on the barrier properties of composites; however, with highly focused research in this field, there is a need to reevaluate the findings and current trends. Hybrid composites are a mixture of inorganic and organic components at the nanometer or molecular level, whereas, biocomposites are formed by reinforcing natural fibers into a matrix to improve their properties, including barrier characteristics. Such composites can be used for many applications, including packaging, to maintain the quality of products. Thus, the barrier properties of films are important characteristics. This chapter discusses the parameters that influence the barrier properties of composites such as the pore size, chemical structure, free volume, and crystallinity of the polymer. The aims of this chapter are to provide comprehensive knowledge about factors affecting the permeability and preparation methodology, the types of composites, and the types of barrier characteristics including oxygen, carbon dioxide, water vapor, and aroma, and to present the potential applications of such hybrid films.

Invasive sino-orbital aspergillosis is a rare cause of orbital apex syndrome (OAS) in immunocompetent patients and often misdiagnosed as tumour because of its aggressive nature and invasive patterns. We report a 23-year-old immunocompetent man presenting with painful progressive loss of vision, ophthalmoplegia and proptosis of the right eye suggestive of OAS. MRI with gadolinium contrast showed an enhancing heterogeneous mass filling the paranasal sinuses, extraconal space and extending up to the right orbital apex. A functional endoscopic biopsy reported as invasive sino-orbital aspergillosis. He was started on intravenous voriconazole and maximal surgical debridement was done. He gradually regained his vision to 20/30 in the right eye. A review of literature reported several such cases which were managed medically or surgically but with poor visual recovery. This case highlights the need for awareness among clinicians for early diagnosis and treatment to prevent vision loss and better survival.

The United Nations 2030 Agenda emphasizes the importance of multistakeholder partnerships for achieving the 17 UN Sustainable Development Goals (SDGs). Indeed, Goal 17 includes a target for national governments to promote multistakeholder partnerships between state and non-state actors. In this paper, we explore how members of civil society organizations and the private sector perceive both the possibilities and challenges of multistakeholder partnerships evolving in Ireland for achieving the SDGs. The research uses data gathered during 2018 and includes documentary research, participant observations of stakeholder forums in Ireland and the United Nations, and semi-structured interviews to address related questions. The results demonstrate that numerous challenges exist for forming multistakeholder partnerships for the SDGs, including a fragmented understanding of the Goals. They also note previous examples of successful multistakeholder partnership models, the need for more leadership from government, and an overly goal-based focus on SDG implementation by organizations as major impediments to following a multistakeholder partnership approach in the country. These findings suggest that although Goal 17 identifies multistakeholder partnerships as essential for the SDGs, they are challenging to form and require concerted actions from all state and non-state actors for SDG implementation.

A 64-year-old hypertensive woman presented with complaints of a painless, progressive decrease in vision, headache, dizziness for the last month. She was a known case of diabetes mellitus on insulin therapy. On examination, best-corrected visual acuity was 6/12 in the right eye and 6/18 in the left eye. The fundus examination in both eyes revealed features of grade IV hypertensive retinopathy. Systemic examination revealed the raised blood pressure of 200/110 mm Hg. Diagnosis of pheochromocytoma was made on the basis of increased urinary norepinephrine (892.8 mg/dL) and mass in the left adrenal gland (measuring 31×28 mm) at contrast-enhanced CT. Medical management to control hypertension was done and ultimately, she underwent left laparoscopic adrenalectomy. After 10 months of surgery, the patient was asymptomatic, blood pressure was within normal limit and her vision improved to 6/6 in both the eyes. The retinal features of hypertensive retinopathy had completely disappeared.

Cerebral sagittal sinus thrombosis is a very rare condition and presents as diagnostic challenge due to its varied presentations. Most common signs and symptoms are headache, blurring of vision, diplopia, and papilledema. Pregnancy and puerperium constitute one of its rare etiological factors. A high index of clinical suspicion of the disease is important when a pregnant female presents with stroke like symptoms of headache, hemiparesis, diplopia, and seizures in the early stages of pregnancy because early neuroimaging can clinch the diagnosis. Management with adequate dosage of anticoagulants can be life-saving and can prevent severe visual loss. Here, we report and discuss the presentation and management of a case of cerebral superior sagittal sinus thrombosis presenting as diplopia in a young primigravid in the first trimester of pregnancy.

Fructose 6-phosphate is an intermediate in the Calvin-Benson cycle and can be acted on by phosphoglucoisomerase to make glucose 6-phosphate (G6P) for starch synthesis. A high concentration of G6P is favorable for starch synthesis but can also stimulate G6P dehydrogenase initiating the glucose-6-phosphate shunt an alternative pathway around the Calvin-Benson cycle. A low concentration of glucose 6-phosphate will limit this futile cycle. In order to understand the biochemical regulation of plastidic glucose 6-phosphate supply and consumption, we characterized biochemical parameters of two key enzymes, phosphoglucoisomerase (PGI) and G6P dehydrogenase (G6PDH). We have found that the plastidic PGI in has a higherKmfor G6P compared to that for fructose 6-phosphate. TheKmof G6PDH isoform 1 is increased under reducing conditions. The other two isoforms exhibit less redox regulation; isoform 2 is the most inhibited by NADPH. Our results support the conclusion that PGI restricts stromal G6P synthesis limiting futile cycling via G6PDH. It also acts like a one-way valve, allowing carbon to leave the Calvin-Benson cycle but not reenter. We found flexible redox regulation of G6PDH that could regulate the glucose-6-phosphate shunt.HighlightGlucose 6-phosphate stimulates glucose-6-phosphate dehydrogenase. This enzyme is less active during the day but retains significant activity that is very sensitive to the concentration of glucose 6-phopshate.

Bioenergy holds significant promise to mitigate the climate-related problems associated with fossil fuel use in heat, electricity, and transportation fuel production. Many governments are encouraging bioeconomy growth with new policies. International trade between bioenergy producing and consuming nations has increased over the years. Developed countries with significant greenhouse gas emission (GHG) emission reduction goals are replacing fossil fuels with bioenergy, creating new export commodities for developing nations. However, increased bioeconomy development can put local social, economic, and environmental conditions in bioenergy producing areas at risk. To minimize the potentially adverse impacts of bioenergy development on existing socioeconomic and environmental conditions, several sustainability certification programs have recently been developed. However, there may be significant differences in how actors across multiple scales, including international non-governmental organizations, state and national governments, and local community members perceive a sustainable bioeconomy. In this chapter, we look specifically at two bioenergy development cases, one in the context of economic development in Latin America (jatropha-based bioenergy development in Yucatan, Mexico) and another in the context of a post-industrialized nation (wood-based bioenergy development in Wisconsin, USA) to understand how different actors view sustainability. Our conclusions suggest that, first, developing a sustainable bioeconomy requires addressing sustainability in all stages in the supply chain, and that, second, community perceptions matter in developing a sustainable bioeconomy, thus there is value in a bottom-up approach to policymaking.

The effect of allyl isothiocyanate (AITC), in combination with low temperature (10°C) storage on post harvest quality of minimally processed shredded cabbage was investigated. An optimum concentration of 0.05μL/mL AITC was found to be effective in maintaining the microbial and sensory quality of the product for a period of 12days. Inhibition of browning was shown to result from a down-regulation (1.4-fold) of phenylalanine ammonia lyase (PAL) gene expression and a consequent decrease in PAL enzyme activity and o-quinone content. In the untreated control samples, PAL activity increased following up-regulation in PAL gene expression that could be linearly correlated with enhanced o-quinone formation and browning. The efficacy of AITC in extending the shelf life of minimally processed shredded cabbage and its role in down-regulation of PAL gene expression resulting in browning inhibition in the product is reported here for the first time.

Energy justice is increasingly being used as a framework to conceptualize the impacts of energy decision making in more holistic ways and to consider the social implications in terms of existing ethical values. Similarly, renewable energy technologies are increasingly being promoted for their environmental and social benefits. However, little work has been done to systematically examine the extent to which, in what ways and in what contexts, renewable energy technologies can contribute to achieving energy justice. This paper assesses the potential of renewable electricity technologies to address energy justice in various global contexts via a systematic review of existing studies analyzed in terms of the principles and dimensions of energy justice. Based on publications including peer reviewed academic literature, books, and in some cases reports by government or international organizations, we assess renewable electricity technologies in both grid integrated and off-grid use contexts. We conduct our investigation through the rubric of the affirmative and prohibitive principles of energy justice and in terms of its temporal, geographic, socio-political, economic, and technological dimensions. Renewable electricity technology development has and continue to have different impacts in different social contexts, and by considering the different impacts explicitly across global contexts, including differences between rural and urban contexts, this paper contributes to identifying and understanding how, in what ways, and in what particular conditions and circumstances renewable electricity technologies may correspond with or work to promote energy justice.

Effect of radiation processing (0.5–2 kGy) and storage on the volatile oil constituents and glucosinolateprofile of cabbage was investigated. Among the volatile oil constituents, an enhancement in trans-hex-2-enal was noted on irradiation that was attributed to the increased liberation of precursor linolenic acidmainly from monogalactosyl diacyl glycerol (MGDG). Irradiation also enhanced sinigrin, the major gluc-osinolate of cabbage that accounted for the enhanced allyl isothiocyanate (AITC) in the volatile oils of theirradiated vegetable. During storage the content of trans-hex-2-enal increased immediately after irradi-ation and then returned to the basal value within 24 h while the content of sinigrin and AITC increasedpost irradiation and thereafter remained constant during storage. Our findings on the enhancement inpotentially important health promoting compounds such as sinigrin and AITC demonstrates that besidesextending shelf life and safety, radiation processing can have an additional advantage in improving thenutritional quality of cabbage. 2013 Elsevier Ltd. Deoxyxylulose 5-phosphate synthase (DXS), a thiamine diphosphate (ThDP) dependent enzyme, plays a regulatory role in the methylerythritol 4-phosphate (MEP) pathway. Isopentenyl diphosphate (IDP) and dimethylallyl diphosphate (DMADP), the end products of this pathway, inhibit DXS by competing with ThDP. Feedback inhibition of DXS by IDP and DMADP constitutes a significant metabolic regulation of this pathway. The aim of this work was to experimentally test the effect of key residues of recombinant poplar DXS (PtDXS) in binding both ThDP and IDP. This work also described the engineering of PtDXS to improve the enzymatic activity by reducing its inhibition by IDP and DMADP. We have designed and tested modifications of PtDXS in an attempt to reduce inhibition by IDP. This could possibly be valuable by removing a feedback that limits the usefulness of the MEP pathway in biotechnological applications. Both ThDP and IDP use similar interactions for binding at the active site of the enzyme, however, ThDP being a larger molecule has more anchoring sites at the active site of the enzyme as compared to the inhibitors. A predicted enzyme structure was examined to find ligand-enzyme interactions, which are relatively more important for inhibitor-enzyme binding than ThDP-enzyme binding, followed by their modifications so that the binding of the inhibitors can be selectively affected compared to ThDP. Two alanine residues important for binding ThDP and the inhibitors were mutated to glycine. In two of the cases, both the IDP inhibition and the overall activity were increased. In another case, both the IDP inhibition and the overall activity were reduced. This provides proof of concept that it is possible to reduce the feedback from IDP on DXS activity.

Glucosinolates, found principally in the plant order Brassicales, are modulated by different post-harvest processing operations. Among these, ionizing radiation, a non-thermal process, has gained considerable interest for ensuring food security and safety. In gamma-irradiated cabbage, enhanced sinigrin, a major glucosinolate, has been reported. However, the molecular basis of such a radiation induced effect is not known. Herein, the effect of radiation processing on the expression of glucosinolate biosynthetic genes was investigated. RT-PCR based expression analysis of seven glucosinolate biosynthetic pathway genes (MYB28, CYP79F1, CYP83A1, SUR1, UGT74B1, SOT18 and TGG1) showed that CYP83A1, MYB28, UGT74B1, CYP79F1 and SUR1 were up-regulated in irradiated cabbage. The content of jasmonates, signalling molecules involved in glucosinolate induction was, however, unaffected in irradiated cabbage suggesting their non-involvement in glucosinolate induction during radiation processing. This is the first report on the effect of gamma irradiation on the expression of glucosinolate biosynthetic genes in vegetables.

Over the past decade Spiking Neural Networks (SNN) have emerged as one of the popular architectures to emulate the brain. In SNN, information is temporally encoded and communication between neurons is accomplished by means of spikes. In such networks, spike-timing dependent plasticity mechanisms require the online programming of synapses based on the temporal information of spikes transmitted by spiking neurons. In this work, we propose a spintronic synapse with decoupled spike transmission and programming current paths. The spintronic synapse consists of a ferromagnet-heavy metal heterostructure where programming current through the heavy metal generates spin-orbit torque to modulate the device conductance. Low programming energy and fast programming times demonstrate the efficacy of the proposed device as a nanoelectronic synapse. We perform a simulation study based on an experimentally benchmarked device-simulation framework to demonstrate the interfacing of such spintronic synapses with CMOS neurons and learning circuits operating in transistor sub-threshold region to form a network of spiking neurons that can be utilized for pattern recognition problems., The article will appear in a future issue of Physical Review Applied

Gamma irradiation (0.5-2.5 kGy) in combination with low temperature (4-15 °C) storage was attempted to increase shelf life of ready-to-cook shredded cabbage wrapped in cling films. A maximum extension in shelf life of 8 days, while retaining the microbial and sensory quality, was obtained with an irradiation dose of 2 kGy and storage at 10 °C. Gamma irradiation also inhibited browning of shredded cabbage at their cut edges resulting in enhanced visual appeal. An increase in total antioxidant activity was observed with respect to DPPH and hydroxyl radical scavenging ability while the nitric oxide radical scavenging activity and ferric reducing property remained unaffected with irradiation. Total phenolic, flavonoid and vitamin C content remained unchanged due to irradiation. No significant migration of additives from cling films into stimulant water was observed up to a radiation dose of 2 kGy thus demonstrating the feasibility of such films for above applications.
