Abstract

Monitoring populations is critical to understanding habitat use, especially for endangered species and is important for determining the effectiveness of land management strategies. The Florida Grasshopper Sparrow ( Ammodramus savannarum floridanus ) is a critically endangered non-migratory grassland bird that has been monitored since the 1990s. It resides primarily in dry prairie habitat managed by frequent (2–3 years) prescribed fires. Monitoring is confounded by the presence of the migratory and wintering eastern Grasshopper Sparrow ( A. s. pratensis ), that has vocal and morphological similarities. These similarities could lead to misclassifications and erroneous conclusions about land management. Our goal was to determine the impact of fires on Florida Grasshopper Sparrow habitat use at two spatial scales, 100 m and 400 m buffers surrounding point count locations, while controlling for the presence of the eastern Grasshopper Sparrow. We combined point count data (1996–2011), external sources of data (eBird), and Bayesian dynamic occupancy models that accounted for misclassifications to evaluate habitat use and dynamics in response to prescribed fires. The probability of misclassifying a Florida Grasshopper Sparrow peaked in April and then decreased to near zero after May 1. The probability of persistence at point count stations decreased as years-since-fire (maximum of pixels within 400 m buffer) increased and was greatest for recent burns (< 1 year). Seasonality of the most recent burn (i.e., ordinal date of burn for pixels within 100 m buffer) also influenced probability of persistence with Florida Grasshopper Sparrows having greater-than-average persistence when burns occurred during January and July, peaking in early April. The probability of colonization peaked approximately two months after a fire and rapidly decreased to negligible values. This is the first long-term study to examine the effect of prescribed burn on Florida Grasshopper Sparrow occupancy that accounts for the presence of eastern Grasshopper Sparrows. This modeling framework may also provide utility for other species and subspecies that are difficult to distinguish during surveys.

Vegetation characteristics can influence nest-site selection and nest survival of birds. The Florida Grasshopper Sparrow (Ammodramus savannarum floridanus) is a critically endangered ground nesting grassland bird endemic to central Florida. Currently, the two largest remaining populations are found on sites with differently managed habitats. One site is burned regularly to maintain native dry prairie habitat and the other is a cattle pasture that is mowed and burned to optimize cattle forage. Little is known about how vegetation influences Florida Grasshopper Sparrow nest-site selection and nest success in these different habitats. We measured microhabitat characteristics (percent vegetation cover and grass height) at Florida Grasshopper Sparrow nests and paired random plots at both sites for three breeding seasons (2014-2016). Percent cover differed significantly between the sites for seven of eight vegetation types, with the cattle pasture characterized primarily by grasses and dry prairie characterized by more diverse native vegetation. Despite these vegetative differences, grass height did not differ at the two locations, suggesting that plant height, rather than plant species composition, may be more important for nest site selection in this bird. Microhabitats around nests at the dry prairie site had 31% less bare ground and 32% more grass than non-nest plots. No variables predicted the placement of nests at the cattle pasture, possibly because of the more homogenous habitat at that site. We did not find a vegetative component of nest success, which suggests that other nonvegetative factors may influence nest predation. Understanding the vegetation characteristics associated with Florida Grasshopper Sparrow nests will help inform habitat management strategies for maintaining vegetation height for nest-site habitat of this critically endangered subspecies.

Abstract

Nest microclimate can be affected by wind and solar radiation, which can influence offspring survival. The endangered Florida Grasshopper Sparrow (Ammodramus savannarum floridanus) constructs a domed nest on the ground in dry prairie and pasture habitats of central Florida. We measured the nest opening orientation for 116 Florida Grasshopper Sparrow nests across 2 sites with different habitats (2014-2016). Nest openings were primarily oriented to the northeast and orientation did not differ by site. Nest opening orientation did not influence the probability that a nest would successfully fledge at least 1 nestling. It is possible that Florida Grasshopper Sparrows may orient nests to the northeast to avoid wind-blown rain from summer storms and solar radiation during the hottest part of the day, but high local rates of nest predation documented at both sites mask the selective advantage of nest orientation on nest survival. 12. DISCOVERY AND EARLY MONITORING OF A NEW FLORIDA GRASSHOPPER SPARROW POPULATION ON RANCHLANDS IN OSCEOLA COUNTY: EVIDENCE OF METAPOPULATION CONNECTIVITY.

Abstract

In 2012, the critically endangered Florida Grasshopper Sparrow (Ammodramus savannarum floridanus) was known to exist only on three public conservation areas with native dry prairie habitat. In 2013, an additional site was discovered on a grazed, semi-improved cattle ranch in Osceola County, Florida, USA (hereafter the Ranch). We describe initial results from an effort to establish a long-term demographic monitoring program at the Ranch (2014-2016). Band-resighting data revealed that the Ranch supported a minimum population of 29 territorial males in 2015 and 19 in 2016, representing 39% and 34% of the known wild male population, respectively. The weighted mean for apparent annual survival was 8% for fledged nestlings, 38% for independent fledglings, and 58% for adult males. High rates of nest loss to red imported fire ants (Solenopsis invicta) and other predators resulted in very low reproductive success and an estimated productivity of 1.66 young per pair in 2015. During 2013-2016, we documented 10 long-distance dispersal events (range = 12-25 km) for five adult males between the Ranch and other populations. All dispersing males appeared to be unpaired, and several males returned to the Ranch after their initial dispersal, suggesting the movements may have been forays by unpaired individuals searching for females. More extensive monitoring on the Ranch could help determine whether unbanded sparrows detected at the site each spring are locally produced or immigrants from other populations. Regardless, because of the size of the population and its central location, the Ranch supports species recovery goals of redundancy, resiliency, and connectivity. Abstract

The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.

Abstract

There is an increasing concern of land owners to protect and maintain healthy and sustainable agroecosystems through the implementation of best management practices (BMP). The objectives of this study were: (1) To develop and evaluate the use of a Geographic Information System (GIS) technology for enhancing field-scale management practices; (2) evaluate the use of 2-dimensional displays of the landscape and (3) define spatial classes of variables from interpretation of geostatistical parameters. Soil samples were collected to a depth of 2 m at 15 cm increments. Existing data from topographic, land use, and soil survey maps of the Winfred Thomas Agricultural Research Station were converted to digital format. Additional soils data which included texture, pH, and organic matter were also generated. The digitized parameters were used to create a multilayered field-scale GIS. Two dimensional (2-D) displays of the parameters were generated using the ARC/INFO software. The spatial distribution of the parameters evaluated in both fields were similar which could be attributed to the similarity in vegetation and surface elevation. The ratio of the nugget to total semivariance, expressed as a percentage, was used to assess the degree of spatial variability. The results indicated that most of the parameters were moderate spatially dependent Biophysical constraint maps were generated from the database layers, and used in multiple combination to visualize results of the BMP. Understanding the spatial relationships of physical and chemical parameters that exists within a field should enable land managers to more effectively implement BMP to ensure a safe and sustainable environment.

Abstract

The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles ("MISEV") guidelines for the field in 2014. We now update these "MISEV2014" guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.

Abstract

Many aspects of blue whale biology are poorly understood. Some of the gaps in our knowledge, such as those regarding their basic taxonomy and seasonal movements, directly affect our ability to monitor and manage blue whale populations. As a step towards filling in some of these gaps, microsatellite and mtDNA sequence analyses were conducted on blue whale samples from the Southern Hemisphere, the eastern tropical Pacific (ETP) and the northeast Pacific. The results indicate that the ETP is differentially used by blue whales from the northern and southern eastern Pacific, with the former showing stronger affinity to the region off Central America known as the Costa Rican Dome, and the latter favouring the waters of Peru and Ecuador. Although the pattern of genetic variation throughout the Southern Hemisphere is compatible with the recently proposed subspecies status of Chilean blue whales, some discrepancies remain between catch lengths and lengths from aerial photography, and not all blue whales in Chilean waters can be assumed to be of this type. Also, the range of the proposed Chilean subspecies, which extends to the Galapagos region of the ETP, at least seasonally, perhaps should include the Costa Rican Dome and the eastern North Pacific as well.
