1. Baker Grade IV Capsular Contracture Is Correlated with an Increased Amount of Silicone Material: An Intrapatient Study.

Contains fulltext : 299835.pdf (Publisher’s version ) (Open Access), BACKGROUND: Breast implant surgery is one of the most frequently performed procedures by plastic surgeons worldwide. However, the relationship between silicone leakage and the most common complication, capsular contracture, is far from understood. This study aimed to compare Baker grade I with Baker grade IV capsules regarding their silicone content in an intradonor setting, using two previously validated imaging techniques. METHODS: Twenty-two donor-matched capsules from 11 patients experiencing unilateral complaints were included after bilateral explantation surgery. All capsules were examined using both stimulated Raman scattering (SRS) imaging and staining with modified oil red O (MORO). Evaluation was done visually for qualitative and semiquantitative assessment and automated for quantitative analysis. RESULTS: Using both SRS and MORO techniques, silicone was found in more Baker grade IV capsules (eight of 11 and 11 of 11, respectively) than in Baker grade I capsules (three of 11 and five of 11, respectively). Baker grade IV capsules also showed significantly more silicone content compared with the Baker grade I capsules. This was true for semiquantitative assessment for both SRS and MORO techniques ( P = 0.019 and P = 0.006, respectively), whereas quantitative analysis proved to be significant for MORO alone ( P = 0.026 versus P = 0.248 for SRS, respectively). CONCLUSIONS: In this study, a significant correlation between capsule silicone content and capsular contracture is shown. An extensive and continued foreign body response to silicone particles is likely to be responsible. Considering the widespread use of silicone breast implants, these results affect many women worldwide and warrant a more focused research effort. CLINICAL QUESTION/LEVEL OF EVIDENCE: Risk, III.

Abstract

A novel method for applying cavity ring-down spectroscopy in the liquid phase, compatible with LC analyses, is presented. The core of the setup is a home-built cavity ring-down flow cell (cell volume 12 [micro]L) that is constructed using a silicon rubber spacer, which is clamped leak-tight between two high-reflectivity mirrors. The mirrors are in direct contact with the liquid flow, which provides for a small path length and short ring-down times. Inside the cavity there are no windows, reflection losses, or Brewster angles to be considered. Due to the small size of the presented cavity geometry, the setup can be implemented in conventional-size LC apparatuses. With a flow injection setup, a detection limit of 2.5 nM was obtained for Crystal Violet in ethanol, and the linear dynamic range of the system is at least 2 orders of magnitude. The method has the potential to become a powerful alternative for commercial LC UV/visible absorbance detectors.

Abstract

We describe our set-up for Stimulated Raman Scattering (SRS) microscopy with shot noise limited detection for a broad window of biologically relevant laser powers. This set-up is used to demonstrate that the highest signal-to-noise ratio (SNR) in SRS with shot noise limited detection is achieved with a time-averaged laser power ratio of 1:2 of the unmodulated and modulated beam. In SRS, two different coloured laser beams are incident on a sample. If the energy difference between them matches a molecular vibration of a molecule, energy can be transferred from one beam to the other. By applying amplitude modulation to one of the beams, the modulation transfer to the other beam can be measured. The efficiency of this process is a direct measure for the number of molecules of interest in the focal volume. Combined with laser scanning microscopy, this technique allows for fast and sensitive imaging with sub-micrometre resolution. Recent technological advances have resulted in an improvement of the sensitivity of SRS applications, but few show shot noise limited detection. The dominant noise source in this SRS microscope is the shot noise of the unmodulated, detected beam. Under the assumption that photo damage is linear with the total laser power, the optimal SNR shifts away from equal beam powers, where the most signal is generated, to a 1:2 power ratio. Under these conditions the SNR is maximized and the total laser power that could induce photodamage is minimized. Compared to using a 1:1 laser power ratio, we show improved image quality and a signal-to-noise ratio improvement of 8 % in polystyrene beads and C. Elegans worms. Including a non-linear damage mechanism in the analysis, we find that the optimal power ratio converges to a 1:1 ratio with increasing order of the non-linear damage mechanism.

Abstract

Dibenzo(a,l)pyrene (DB(a,l)P) and the 11,12-dihydrodiol of DB(a,l)P form depurinating adducts at the N-3 and N-7 sites of the adenine residue and the reaction requires the presence of a double stranded DNA. Both DB(a,l)P and the dihydrodiol are carcinogenic, the former being one of the most potent polycyclic aromatic hydrocarbons. DB(a,l)P is present in cigarette smoke and smoke from combustion of substances such as coal. The intermediates formed by the activation of DB(a,l)P can bind to DNA and start tumors.

Abstract

Carotenoids are among the key biomarkers in the search for life on other planets, and non-destructive Raman spectroscopy on future rover missions is a potential sensitive detection method, especially under resonant conditions. In this research, reflectance spectra of minerals and microorganisms were measured using ultraviolet/visible diffuse reflectance spectroscopy in order to evaluate potential resonance Raman conditions and the possible degree of sample damage during laser irradiation. We report a photodegradation and semi-quantitative Raman study of β-carotene and the carotenoid-containing extremophile Deinococcus radiodurans mixed with calcite at excitation wavelengths of 440nm, 532nm and 785nm. A different type of carotenoid was detected in a culture of Chroococcidiopsis. Carotenoids embedded in bacterial membranes were found to be less sensitive to photodegradation than in a mineral matrix. Corrections for self-absorption effects were performed using the 1085cm

Abstract

In conventional Raman spectroscopic measurements of liquids or surfaces the preferred geometry for detection of the Raman signal is the backscattering (or reflection) mode. For non-transparent layered materials, sub-surface Raman signals have been retrieved using spatially offset Raman spectroscopy (SORS), usually with light collection in the same plane as the point of excitation. However, as a result of multiple scattering in a turbid medium, Raman photons will be emitted in all directions. In this study, Monte Carlo simulations for a three-dimensional layered sample with finite geometry have been performed to confirm the detectability of Raman signals at all angles and at all sides of the object. We considered a non-transparent cuboid container (high density polyethylene) with explosive material (ammonium nitrate) inside. The simulation results were validated with experimental Raman intensities. Monte Carlo simulation results reveal that the ratio of sub-surface to surface signals improves at geometries other than backscattering. In addition, we demonstrate through simulations the effects of the absorption and scattering coefficients of the layers, and that of the diameter of the excitation beam. The advantage of collecting light from all possible 4 angles, over other collection modes, is that this technique is not geometry specific and molecular identification of layers underneath non-transparent surfaces can be obtained with minimal interference from the surface layer. To what extent all sides of the object will contribute to the total signal will depend on the absorption and scattering coefficients and the physical dimensions. Abstract

Time resolved Raman spectroscopy (TRRS) can provide subsurface information from multi-layered samples of transparent and translucent evaporative and silicate minerals up to several centimetres thick. Depth information was obtained using 3-ps pulsed laser excitation at 720 nm and a gated intensified charge-coupled device detector with stepwise increasing delay times. Blocks of different minerals were used as first, second or third layers, and Raman spectra from deeper layers could be detected through 10 mm of translucent calcite and up to 40 mm of transparent halite crystals. Measurements by conventional confocal Raman, as well as spatially offset Raman spectroscopy were also successful in distinguishing different mineral layers. This study establishes the great potential for the use of Raman spectroscopy in future planetary exploration, where TRRS could be used as a non-invasive tool for profiling the (sub-)surface at millimetre-depth resolution. Abstract

Raman spectroscopy is one of the major characterization methods employed over the last few decades as a nondestructive technique for the study of heterogeneous catalysts and related catalytic reactions. However, the promise of practical applicability on millimeter-sized catalyst bodies, such as extrudates, has not been fulfilled completely. Large fluorescence signals and the highly scattering nature of the extrudates often hamper its practical usage. Different approaches to overcome this problem were examined, including the use of time-resolved Raman spectroscopy (TRRS), spatially offset Raman spectroscopy (SORS), surface-enhanced Raman spectroscopy (SERS), and combinations of these techniques. This paper demonstrates that especially TRRS can provide chemical information at depth within catalyst bodies, overcoming fluorescence background signals and allowing for visualization of analytes at different depths. It also examines the application of time-resolved SERS within catalyst bodies to gain insight into localized activity. With these options a wider applicability of Raman spectroscopy for industrial catalysis research becomes within reach. © 2012 Society for Applied Spectroscopy.

Abstract

The interaction of the nonsteroidal anti-inflammatory drug flurbiprofen (FBP) with human serum albumin (HSA) hardly influences the fluorescence of the protein's single tryptophan (Trp). Therefore, in addition to fluorescence, heavy atom-induced room-temperature phosphorescence is used to study the stereoselective binding of FBP enantiomers and their methyl esters to HSA. Maximal HSA phosphorescence intensities were obtained at a KI concentration of 0.2 M. The quenching of the Trp phosphorescence by FBP is mainly dynamic and based on Dexter energy transfer. The Stern-Volmer plots based on the phosphorescence lifetimes indicate that (R)-FBP causes a stronger Trp quenching than (S)-FBP. For the methyl esters of FBP, the opposite is observed: (S)-(FBPMe) quenches more than (R)-FBPMe. The Stern-Volmer plots of (R)-FBP and (R)-FBPMe are similar although their high-affinity binding sites are different. Abstract

Archaeological bone undergoes alterations after burial (diagenesis) that constitute a problem for the survival of archaeological information. A common method to assess this alteration is Fourier transform infrared spectrometry (FTIR). However, the commonly applied method (FTIR-KBr) is destructive and sample preparation may influence the results. This paper tests the suitability of FTIR attenuated total reflection (FTIR-ATR), a method not commonly used to investigate bone diagenesis. FTIR-ATR requires less sample preparation and can be non-destructive, allowing analysis of bone cross-sections. Modern and archaeological bones were analysed using both methods and different sample preparation methods were tested. The results show that FTIR-ATR has advantages for the rapid assessment of bone diagenesis. © University of Oxford, 2012.

39. Flanking base effects on the structural conformation of the (+)-trans-anti-benzo[α]pyrene diolepoxide adduct to N-dg in sequence-defined oligonucleotides

Abstract

Conformations of the trans adduct of (+)-anti-benzo[a]8- pyrene -7,8-dihydrodiol-9,10-epoxide (BPDE) to N2-guanine, the major stable DNA adduct of the environmental carcinogen benzo[a]pyrene, were studied as a function of flanking bases in single-stranded and in double-stranded oligonucleotides. Three llmer oligonucleotides d(CTATG1G2G3TATC) were synthesized containing the (+)-transanti-BPDE adduct at one specific guanine of the GGG sequence (a known mutational hot spot). Polyacrylamide gel electrophoresis of the three single-stranded oligonucleotides showed that the adduct bound to G2 or G3 (5'-flanking base guanine) caused significantly stronger retardation than the same adduct bound to Gt (5'-flanking base thymine). The strength of the carcinogen-base interaction was reflected in the spectroscopic properties of the pyrenyl moiety. Low temperature fluorescence measurements under line-narrowing (FLN) or non-line-narrowing (NLN) conditions showed that in single-stranded form the adduct at G2 or G3 (5'-flanking base guanine) adopts a conformation with strong interaction with the bases. This was also observed for the same adduct at the sequence AGA. In contrast, the (+)-trans-anti-BPDE adduct with a 5' -flanking thymine exists in a primarily helix-external conformation. Similar differences were observed in the double-stranded oligonucleotides: the adducts at G2 and G3 were found to exist in similar conformational equilibria, again with significant carcinogen-base interactions, while the adduct at G1 showed a predominantly external conformation. The nature of the 3'-flanking base appeared to have little influence on the conformational equilibrium of the (+)-trans-anti-BTDE-guanine adduct The results could provide insight into the mutational specificity and flanking base effects observed for (+)-anti-BPDE.

Raman and MIR spectroscopy techniques provide bond-specific vibrational frequencies to characterize molecular species without external labelling. In addition, these techniques are rapid, non-invasive and provide multiplex advantage. In this review we discuss the instrumentation, applications and latest advancements of Raman and MIR spectroscopic imaging in various fields, ranging from materials science to medicine and illustrate with specific examples from our recent research.

Abstract

Fluorescence line-narrowing spectroscopy (FLNS) is a cryogenic technique that can be used to obtain high-resolution fluorescence spectra. The technique has mainly been used for fingerprint identification. However, this review focuses on the trend to use FLNS for probing purposes: obtaining information on interactions with the local environment or determining conformations. We briefly discuss the basics of FLNS, solute-matrix interactions and important instrumental aspects. We describe examples from various bioanalytical fields: DNA adducts of the aromatic carcinogen benzo[a]pyrene (BP); binding of BP metabolites to monoclonal antibodies or the estrogen receptor; FLNS studies of porphyrin-containing proteins; and, photosynthetic systems. As an illustration from the field of environmental chemistry, we discuss the use of luminescence line-narrowing spectroscopy - a special mode of FLNS - to study the complexation of lanthanides to humic substances.

In an extended introduction, key aspects of resonance Raman spectroscopy (RRS) such as enhanced sensitivity and selectivity are briefly discussed in comparison with normal RS. The analytical potential is outlined. Then achievements in different fields of research are highlighted in four sections, with emphasis on recent breakthroughs: (1) The use of visible RRS for analyzing carotenoids in biological matrices, for pigments and dyes as dealt with in art and forensics, and for characterizing carbon nanotubes. (2) The use of RRS in the deep UV (excitation below 260 nm) in the bioanalytical and life sciences fields, including nucleic acids, proteins and protein-drug interactions. Metalloproteins can be studied by visible RRS in resonance with their chromophoric absorption. (3) Progress in theoretical calculations of RRS excitation profiles and enhancement factors, which ultimately might facilitate analytical RRS. (4) Instrumental and methodological achievements including fiber-optic UV-RRS, coupling of RRS to liquid chromatography and capillary electrophoresis. Sensitivities can approach the single-molecule level with surface-enhanced RRS or tip-enhanced RRS. Last but not least, promising fluorescence background rejection techniques based on time-gated detection will be presented. Vibrationally resolved fluorescence spectra of four angular [N]phenylenes were recorded with laser excited Shpol’skii spectroscopy (LESS) in an n-octane matrix at 10 K. In general, the same vibrational frequencies were observed in the fluorescence excitation and emission spectra, indicating that the geometries of ground and electronically excited state are very similar. Because of intensity borrowing from the S2 state, vibrations of two different symmetries were observed in the fluorescence excitation spectra of angular [3]phenylene and zig-zag[5]phenylene. This finding allowed the location of the S2 state for these compounds. DFT calculations(RB3LYP/6-31G*) of the ground state vibrational frequencies were made. The calculated vibrational modes were in reasonably good agreement with the experimental data. A new very low-frequency vibration of approximately 100 cm-1 was predicted and experimentally confirmed for all [N]phenylenes investigated. This vibration seems to be unique for [N]phenylenes and is attributed to an in-plane movement of the carbon backbone.

Abstract

Deuteration effects on the vibronic structure of the emission and excitation spectra of triangular [4]phenylene (D3h[4]phenylene) were studied using laser-excited Shpolskii spectroscopy (LESS) in an octane matrix at 4.2 K. For correct assignment of the vibrational modes, the experimental results were compared with calculated frequencies (B3LYP/6-31G*). CH vibrations were identified by their characteristic isotopic shifts in the spectra of deuterated triangular [4]phenylenes. Two CC stretching modes, at 100 cm–1 and 1176 cm–1, suitable as probes for bond strength changes in the excited state, were identified. The isotope effect on the internal conversion rates of triangular [4]phenylene was evaluated from measurements of temperature dependent lifetime. Isotope dependency and the magnitude of the internal conversion rates indicate that internal conversion in triangular [4]phenylene is most likely induced by CH vibrations. The results obtained by LESS and lifetime measurements were compared with PM3 PECI calculations of the excited state structure. The theoretical results and the relation between ground and excited state vibration energies of the 1176 cm–1 probe vibration indicate a reduction of bond alternation of the central cyclohexatriene ring in the excited state.
