Abstract

For the first time in the history of high energy astronomy, a large CdTe gamma-ray camera is operating in space. ISGRI is the low-energy camera of the IBIS telescope on board the INTEGRAL satellite. This paper details its design and its in-flight behavior and performances. Having a sensitive area of 2621 cm$^2$ with a spatial resolution of 4.6 mm, a low threshold around 12 keV and an energy resolution of $\sim$ 8% at 60 keV, ISGRI shows absolutely no signs of degradation after 9 months in orbit. All aspects of its in-flight behavior and scientific performance are fully nominal, and in particular the observed background level confirms the expected sensitivity of 1 milliCrab for a 10$^6$s observation., Comment: INTEGRAL A&A special issue

Abstract

A basic component, called Polycell, has been developed for the ISGRI (INTEGRAL Soft Gamma Ray Imager) CdTe camera on board the INTEGRAL (INTErnational Gamma-Ray Astrophysics Laboratory) satellite. Operating at room temperature, it covers the 20 keV - 1 MeV energy range. It features a sub-ensemble of 16 CdTe detectors and their associated front end electronics. This electronics is based on 4-channel analog-digital ASICs. Their analog part features a low noise preamplifier, allowing a threshold below 20 keV and a pulse rise-time measurement which permits a charge loss correction. The digital part ensures the internal acquisition timing sequence as well as the dialogue with external electronics. Two versions of the ISGRI ASIC have been developed in a collaboration of two CEA microelectronics teams from CEA/DTA/LETI/DSYS and CEA/DSM/DAPNIA/SEI, respectively on a standard CMOS AMS process hardened against radiation by lay-out, and on a Silicon On Insulator process (DMILL MHS), the latter being latch-up free. This paper presents the ASIC and polycell architecture as well as experimental results obtained with polycells equipped with AMS ASICs.

Abstract

Thanks to recent developments in photon counting detectors, X-Ray spectral imaging has received increasing attention. This technique permits the quantication of the chemical components in an object. The energy selectivity of the detector is known to have a strong impact in the material decomposition capabilities of an imaging device. Although spectral imaging devices are widely used in conjunction to polychromatic illuminations, detectors are typically characterized for one or few incident energies. To the best of our knowledge, no criterion for the overall energy resolution of a photon counting detector is available yet. In this paper, we propose a criterion for energy selectivity over a range of incident energies. Our criterion is based on the analysis of the response of the detector to multiple monochromatic illuminations. In particular, it can be easily computed from the Radon transform of the energy response function of the detector. After a simulation study, we show how this criterion is related to the quality of material decompositions obtained from photon counting measurements.

27. Synovial fluid examination for the diagnosis of synovial amyloidosis in patients with chronic renal failure undergoing haemodialysis.

Abstract

The diagnosis of synovial amyloidosis is based upon synovial biopsy. Synovial fluid (SF) in seven patients with amyloid arthropathy associated with chronic renal failure undergoing haemodialysis were studied. The SF and synovial samples of 10 consecutive patients with seronegative mono- or oligoarthritis served as controls. Six of the seven patients with amyloid positive synovial biopsy specimens showed amyloid in their SF. No amyloid was found in the synovial tissue or fluid of the 10 patients in the control group, the sensitivity being 87.7%. The finding of amyloid in SF was highly reproducible, showing its presence in the same joint on several occasions. The deposits were Congophilia resistant to potassium permanganate pretreatment, and the immunohistochemical analysis proved that they contained beta 2 microglobulin. The high sensitivity and good reproducibility of the method shows that the finding of amyloid in SF is sufficient for the diagnosis of synovial amyloidosis. It is possible to perform immunohistochemical analysis on the SF sediment. Amyloid arthropathy can therefore be added to the list of conditions in which synovial fluid examination can be clinically helpful. [ABSTRACT FROM PUBLISHER]

Abstract

International audience, Sphinx1 is a novel pixel architecture adapted for X-ray imaging that can detect radiation by photon counting and by charge integration. In photon counting mode, each photon is compensated by one or more counter-charge packets which can be dimensioned at a level as low as 100 electrons and the number of injected counter-charge packets indicates the incoming photon energy, thus allowing a spectrometric detection. The pixel is also able to detect radiation by integrating the charges deposited by all incoming photons and converting this analog value into a digital data with a least significant bit (LSB) of 100 electrons through the use of the counter-charge concept. The pixel was simulated in both modes, and simulation results indicate an equivalent noise charge (ENC) of 48 electrons-rms, an integral nonlinearity factor (INL) < 0.15% and a static power consumption < 1 μW.

Abstract

Bowman–Birk inhibitors (BBI) from legumes, such as soyabean, pea, lentil and chickpea, are naturally occurring plant protease inhibitors which have potential health-promoting properties within the mammalian gastrointestinal tract. BBI can survive both acidic conditions and the action of proteolytic enzymes within the stomach and small intestine, permitting significant amounts to reach the large intestine in active form to exert their reported anti-carcinogenic and anti-inflammatory properties. In a previous study, we reported the ability of a recombinant form of TI1B (rTI1B), representing a major BBI isoinhibitor from pea, to influence negatively the growth of human colorectal adenocarcinoma HT29 cells in vitro. In the present study, we investigate if this effect is related directly to the intrinsic ability of BBI to inhibit serine proteases. rTI1B and a novel engineered mutant, having amino acid substitutions at the P1 positions in the two inhibitory domains, were expressed in the yeast Pichia pastoris. The rTI1B proved to be active against trypsin and chymotrypsin, showing Ki values at nanomolar concentrations, whereas the related mutant protein was inactive against both serine proteases. The proliferation of HT29 colon cancer cells was significantly affected by rTI1B in a dose-dependent manner (IC50 = 31 (sd 7) μm), whereas the inactive mutant did not show any significant effect on colon cancer cell growth. In addition, neither recombinant protein affected the growth of non-malignant colonic fibroblast CCD-18Co cells. These findings suggest that serine proteases should be considered as important targets in investigating the potential chemopreventive role of BBI during the early stages of colorectal carcinogenesis. Abstract

Increasing evidence implicates serine proteinases in the proteolytic cascades leading to the pathological destruction of extracellular matrices such as cartilage in osteoarthritis (OA). We have previously demonstrated that the type II transmembrane serine proteinase (TTSP) matriptase acts as a novel initiator of cartilage destruction via the induction and activation of matrix metalloproteinases (MMPs). Hepsin is another TTSP expressed in OA cartilage such that we hypothesized this proteinase may also contribute to matrix turnover. Herein, we demonstrate that addition of hepsin to OA cartilage in explant culture induced significant collagen and aggrecan release and activated proMMP-1 and proMMP-3. Furthermore, hepsin directly cleaved the aggrecan core protein at a novel cleavage site within the interglobular domain. Hepsin expression correlated with synovitis as well as tumour necrosis factor α expression, and was induced in cartilage by a pro-inflammatory stimulus. However, a major difference compared to matriptase was that hepsin demonstrated markedly reduced capacity to activate proteinase-activated receptor-2. Overall, our data suggest that hepsin, like matriptase, induces potent destruction of the extracellular matrix whilst displaying distinct efficiencies for the cleavage of specific substrates.
