MyStoriesBag is a storytelling and game creation tool that aims to facilitate the sharing of cultural heritage by promoting a collaborative and participatory experience through user-generated content. The guided visual online editor allows users to actively create content and play the games they create. This emphasises the concept of sharing experiences, creating a sense of community, and strengthening the connection between users and their cultural heritage.

The proposed method involves analyzing historical culinary recipes through topic modeling and data visualization. It utilizes an unsupervised pipeline, pre-trained language models, and visualization techniques. The method was thoroughly tested on English-language cookbooks and provides information on the evolution of culinary practices.

Chromatin organization, which is under the control of histone deacetylases (HDACs), is frequently deregulated in cancer cells. Amongst HDACs, HDAC8 plays an oncogenic role in different neoplasias by acting on both histone and non-histone substrates. Promising anti-cancer strategies have exploited dual-targeting drugs that inhibit both HDAC8 and tubulin. These drugs have shown the potential to enhance the outcome of anti-cancer treatments by simultaneously targeting multiple pathways critical to disease onset and progression. In this study, a structure-based virtual screening (SBVS) of 96403 natural compounds was performed towards the four Class I HDAC isoforms and tubulin. Using molecular docking and molecular dynamics simulations (MDs), we identified two molecules that could selectively interact with HDAC8 and tubulin. CNP0112925 (arundinin), bearing a polyphenolic structure, was confirmed to inhibit HDAC8 activity and tubulin organization, affecting breast cancer cell viability and triggering mitochondrial superoxide production and apoptosis.

In pediatrics, hematuria usually responds to benign etiologies. A rare cause is nutcracker syndrome, defined as compression of the left renal vein between the aorta and the superior mesenteric artery, resulting in elevated pressure in the left renal vein with the development of collateral veins and varicose dilatation. The prevalence of this syndrome is unknown. When symptomatic, it causes hematuria, proteinuria, and chronic pelvic pain. At puberty, the rapid growth and development of the vertebral bodies can produce a narrowing of the angle between the aorta and the superior mesenteric artery. We describe the case of a teenage girl diagnosed with multiple metastatic mucoepithelial carcinoma of bone who presented anemia-producing hematuria. The diagnosis of nutcracker syndrome was arrived at with consideration of the therapeutic options. Finally, with shoe enhancement for scoliosis correction, hematuria was significantly lessened.

Histone deacetylases (HDACs) are crucial in gene transcription, removing acetyl groups from histones. They also influence the deacetylation of non-histone proteins, contributing to the regulation of various biological processes. Thus, HDACs play pivotal roles in various diseases, including cancer, neurodegenerative disorders, and inflammatory conditions, highlighting their potential as therapeutic targets. This paper reviews the structure and function of the four classes of human HDACs. While four HDAC inhibitors are currently available for treating hematological malignancies, numerous others are undergoing clinical trials. However, their non-selective toxicity necessitates ongoing research into safer and more efficient class-selective or isoform-selective inhibitors. Computational methods have aided the discovery of HDAC inhibitors with the desired potency and/or selectivity. These methods include ligand-based approaches, such as scaffold hopping, pharmacophore modeling, three-dimensional quantitative structure–activity relationships, and structure-based virtual screening (molecular docking). Moreover, recent developments in the field of molecular dynamics simulations, combined with Poisson–Boltzmann/molecular mechanics generalized Born surface area techniques, have improved the prediction of ligand binding affinity. In this review, we delve into the ways in which these methods have contributed to designing and identifying HDAC inhibitors.

Central figures in the phenomenological tradition, such as Aron Gurwitsch, Jean-Paul Sartre, and Maurice Merleau-Ponty, drew extensively on gestalt psychology in their writings. The dialogue between phenomenology and psychology they began continues today in the field of embodied cognitive science. We take up this conversation starting from Aron Gurwitsch’s rich phenomenological analysis of the perception of the cultural world. Gurwitsch’s phenomenological descriptions of the perception of the cultural world bear a striking resemblance to work in embodied cognitive science that takes its inspiration from Gibson’s ecological psychology. Gibson coined the term “affordance” to refer to the possibilities for action that can be directly perceived by persons [Gibson 1979]. However, Gibson from his earliest writings made a distinction between a universal, strictly individual and nonsocial form of perception and a perception of the world that was subject to social and cultural influences. We use Gurwitsch to argue against Gibson’s individualist understanding of direct perception. Each affordance that can be selected as an object of perception refers to a wider sociocultural context, which Gurwitsch called an “order of existence”. We end our paper by taking up the question of the relation of phenomenological description of the perceptual world and explanations of perceptual experience provided by embodied cognitive science.

To reduce the spread of COVID-19, in March of 2020, the state of Virginia issued a stay-at-home order requiring fitness center closures for 2.4 months. The purpose of this study was to explore how the fitness center closures influenced physical activity (PA) in older adults who previously participated in the centers' exercise classes. Eleven older adults (69.0 ± 6.6 years) completed semistructured interviews, which were transcribed and analyzed to identify emergent themes. Ten participants reported engaging in PA during the stay-at-home order; four participants maintained/increased PA compared to prepandemic levels. Four themes emerged regarding PA are as follows: recognition of the value of PA, high self-efficacy in regard to PA, adaptation to circumstances, and functional limitations. Findings suggest that prior exercise class participation positively influenced PA. While most participants did not maintain prepandemic PA levels, they remained active because they recognized the benefits of PA, had high self-efficacy for exercise, and adapted to their circumstances. Transmission and preservation are key aspects of intangible heritage, so new communication strategies are crucial to reach a wider audience. The aim of this work is to apply digital storytelling tools and games to disseminate heritage and help people to discover it, by developing challenges and adventures based on the resources retrieved from Querylab's ICH Discovery section.

The success of cultural heritage archives depends on their ease of use and navigation. A simple and intuitive user interface can improve accessibility and inclusiveness. This paper presents a new approach to create semantic graphs from archive contents. The resulting graphical representation allows users to explore and navigate the data in new and intuitive ways.

In multilingual textual archives, the availability of textual annotation, that is keywords either manually or automatically associated with texts, is something worth exploiting to improve user experience and successful navigation, search and visualization. It is therefore necessary to study and develop tools for this exploitation. The paper aims to define models and tools for handling textual annotations, in our case keywords of a scientific library. With the background of NLP, machine learning and deep learning approaches are presented. They allow us, in supervised and unsupervised ways, to increase the quality of keywords. The different steps of the pipeline are addressed, and different solutions are analyzed, implemented, evaluated and compared, using statistical methods, machine learning and artificial neural networks as appropriate. If possible, off-the-shelf solutions will also be compared. The models are trained on different datasets already available or created ad hoc with common characteristics with the starting dataset. The results obtained are presented, commented and compared with each other.

La hematuria en pediatría responde habitualmente a etiologías benignas. Una causa poco frecuente es el síndrome de cascanueces, que se define como la compresión de la vena renal izquierda entre la aorta y la arteria mesentérica superior, que deriva en una presión elevada en la vena renal izquierda con el desarrollo de venas colaterales y dilataciones varicosas. La prevalencia de este síndrome se desconoce Cuando es sintomático ocasiona hematuria, proteinuria y dolor pélvico crónico. En la pubertad, el crecimiento rápido y el desarrollo de los cuerpos vertebrales pueden producir un estrechamiento del ángulo entre la aorta y la arteria mesentérica superior. Se describe el caso de una adolescente con diagnóstico de carcinoma mucoepitelial metastásico óseo múltiple que presenta hematuria anemizante. Se arribó al diagnóstico de síndrome de cascanueces y se discutieron las opciones de tratamiento. Finalmente, con el uso de realce del calzado para corrección de la escoliosis, se atenuó significativamente la hematuria.

El artículo presenta, mediante una muestra de las noticias del diario La Nación, un análisis en base al discurso de diversos sujetos involucrados a lo largo del conflicto establecido entre la empresa agroindustrial “Vicentin” y el Gobierno nacional durante la primera mitad del año 2020. Mediante una propuesta cuali-cuantitativa pretendemos adentrarnos en las caracterizaciones difundidas por dicho medio gráfico hegemónico, al tiempo que también se intentará dar cuenta del contenido de los dichos, las alianzas pergeñadas y las frecuencias de las caracterizaciones emitidas. Así, el objetivo será elucidar el conflicto en un plano discursivo –ideológico planteado por el mencionado diario.

IntroductionAlthough allogeneic hematopoietic stem cell transplantation (HCT) can be a curative therapy for hematologic disorders, it is associated with treatment-related complications and losses in cardiorespiratory fitness and physical function. High-intensity interval training (HIIT) may be a practical way to rapidly improve cardiorespiratory fitness and physical function in the weeks prior to HCT. The primary aim of this study was to assess the feasibility of implementing a pre-HCT home-based HIIT intervention. The secondary aim was to evaluate pre to post changes in cardiorespiratory fitness and physical function following the intervention.MethodsThis was a single-arm pilot study with patients who were scheduled to undergo allogeneic HCT within six months. Patients were instructed to complete three 30-minute home-based HIIT sessions/week between the time of study enrollment and sign-off for HCT. Sessions consisted of a 5-minute warm-up, 10 high and low intervals performed for one minute each, and a 5-minute cool-down. Prescribed target heart rates (HR) for the high- and low-intensity intervals were 80-90% and 50-60% of HR reserve, respectively. Heart rates during HIIT were captured via an Apple Watch and were remotely monitored. Feasibility was assessed via retention, session adherence, and adherence to prescribed interval number and intensities. Paired t-tests were used to compare changes in fitness (VO2peak) and physical function [Short Physical Performance Battery (SPPB), 30-second sit to stand, and six-minute walk test (6MWT)] between baseline and sign-off. Pearson correlations were used to determine the relationship between intervention length and changes in cardiorespiratory fitness or functional measures.ResultsThirteen patients (58.8±11.6 years) participated in the study, and nine (69.2%) recorded their training sessions throughout the study. Median session adherence for those nine participants was 100% (IQR: 87-107). Adherence to intervals was 92% and participants met or exceeded prescribed high-intensity HR on 68.8±34.8% of intervals. VO2peak improved from baseline to sign-off (14.6±3.1 mL/kg/min to 17.9±3.3 mL/kg/min; pConclusionsFindings demonstrate that implementing a pre-HCT home-based remotely monitored HIIT program is feasible and may provide benefits to cardiorespiratory fitness and physical function.

Abstract Background Studies on the application of developmental care initiatives in Italian NICUs are rather scarce. We aimed to assess parental access to the NICUs and facilities offered to the family members and to test “the state of art” regarding kangaroo mother care (KMC) and breastfeeding policies in level III Italian NICUs. Methods A questionnaire both in paper and in electronic format was sent to all 106 Italian level III NICUs; 86 NICUs (i.e., 80% of NICUs) were completed and returned. The collected data were analysed. In addition, a comparison between the 2017 survey results and those of two previous surveys conducted from 2001 to 2006 was performed. Results In total, 53 NICUs (62%) reported 24-h open access for both parents (vs. 35% in 2001 and 32% in 2006). Parents were requested to temporarily leave the unit during shift changes, emergencies and medical rounds in 55 NICUs (64%). Some parental amenities, such as an armchair next to the crib (81 units (94%)), a room for pumping milk and a waiting room, were common, but others, such as family rooms (19 units (22%)) and adjoining accommodation (30 units (35%)), were not. KMC was practised in 81 (94%) units, but in 72 (62%), i.e., the majority of units, KMC was limited to specific times. In 11 (13%) NICUs, KMC was not offered to the father. The average duration of a KMC session, based on unit staff estimation, was longer in 24-h access NICUs than in limited-access NICUs. KMC documentation in medical records was reported in only 59% of questionnaires. Breastfeeding was successful in a small proportion of preterm infants staying in the NICU. Conclusion The number of 24-h access NICUs doubled over a period of 13 years. Some basic family facilities, such as a dedicated kitchen, rooms with dedicated beds and showers for the parents, remain uncommon. KMC and breastfeeding have become routine practices; however, the frequency and duration of KMC sessions reported by NICU professionals still do not meet the WHO recommendations.

Abstract Background Severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) infection and the resulting coronavirus disease 2019 (COVID‐19) have afflicted hundreds of millions of people in a worldwide pandemic. During this pandemic, otolaryngologists have sought to better understand risk factors associated with COVID‐19 contamination during surgical procedures involving the airways such as tracheostomies. Objective This study provides a standardized technique of performing an ultrasound (US)‐guided percutaneous dilatational tracheostomy (PDT) on COVID‐19 patients in the intensive care unit (ICU). It also outlines safety strategies for health care providers that includes proper use of personal protective equipment (PPE) and regular testing of otolaryngologists for COVID‐19 contamination. Methods This study analyzed data from 44 PDT procedures performed on COVID‐19 patients in the ICU of hospitals in Sao Paulo and Santos, Brazil. The PDT procedures were conducted between April 2020 and August 2020, which coincided with a peak of the COVID‐19 pandemic in São Paulo, Brazil. Surgeons were tested for COVID‐19 using a two‐stage serological enzyme‐linked immunosorbent assay specific for SARS‐CoV‐2 antigens. Conclusion This study describes a safe standardized technique of US‐guided PDT for COVID‐19 patients in the ICU using a method that also decreases the risk of surgeon contamination.

When integrating data from different sources, there are problems of synonymy, different languages, and concepts of different granularity. This paper proposes a simple yet effective approach to evaluate the semantic similarity of short texts, especially keywords. The method is capable of matching keywords from different sources and languages by exploiting transformers and WordNet-based methods. Key features of the approach include its unsupervised pipeline, mitigation of the lack of context in keywords, scalability for large archives, support for multiple languages and real-world scenarios adaptation capabilities. The work aims to provide a versatile tool for different cultural heritage archives without requiring complex customization. The paper aims to explore different approaches to identifying similarities in 1- or n-gram tags, evaluate and compare different pre-trained language models, and define integrated methods to overcome limitations. Tests to validate the approach have been conducted using the QueryLab portal, a search engine for cultural heritage archives, to evaluate the proposed pipeline.

(1) Background: SARS-CoV-2 RNA-dependent RNA polymerase (RdRp) is a promising therapeutic target to fight COVID-19, and many RdRp inhibitors nucleotide/nucleoside analogs, such as remdesivir, have been identified or are in clinical studies. However, the appearance of resistant mutations could reduce their efficacy. In the present work, we structurally evaluated the impact of RdRp mutations found at baseline in 39 patients treated with remdesivir and associated with a different degree of antiviral response in vivo. (2) Methods: A refined bioinformatics approach was applied to assign SARS-CoV-2 clade and lineage, and to define RdRp mutational profiles. In line with such a method, the same mutations were built and analyzed by combining docking and thermodynamics evaluations with both molecular dynamics and representative pharmacophore models. (3) Results: Clinical studies revealed that patients bearing the most prevalent triple mutant P323L+671S+M899I, which was present in 41% of patients, or the more complex mutational profile P323L+G671S+L838I+D738Y+K91E, which was found with a prevalence of 2.6%, showed a delayed reduced response to remdesivir, as confirmed by the increase in SARS-CoV-2 viral load and by a reduced theoretical binding affinity versus RdRp (ΔGbindWT = −122.70 kcal/mol; ΔGbindP323L+671S+M899I = −84.78 kcal/mol; ΔGbindP323L+G671S+L838I+D738Y+K91E = −96.74 kcal/mol). Combined computational approaches helped to rationalize such clinical observations, offering a mechanistic understanding of the allosteric effects of mutants on the global motions of the viral RNA synthesis machine and in the changes of the interactions patterns of remdesivir during its binding.
