Abstract

AbstractVenous thromboembolism (VTE) is a potentially fatal vascular disease that can be successfully managed to reduce the risk of mortality and impairment. Anticoagulation therapy is frequently recommended for the treatment and prevention of venous thromboembolism. Despite breakthroughs in anticoagulant therapy using parenteral anticoagulant medications and vitamin K antagonists, limits in their usage persist, prompting research into alternative anticoagulants such as factor Xa inhibitors. Of the agents under development, edoxaban shows the most promise due to strong data confirming its therapeutic benefit for VTE. Nanoparticle drug delivery systems (NDDS) are designed technologies that employ nanoparticles to convey therapeutic drugs in a targeted and regulated manner. Intraendothelial transport of therapies in VTE is facilitated by the induction of cell adhesion molecule-mediated endocytosis upon interaction of intercellular adhesion molecule 1 (ICAM-1) on endothelium cells by ICAM-1-targeted carriers. The potential application of NDDS as nanoparticles as anticoagulant agents has been emphasized. There is proof of outstanding efficacy in preventing blood coagulation, dissolving blood clots, and improving in vivo detection of thrombus. These nanoplatforms’ growing evaluations, strong action, enhanced biocompatibility, and lack of problems from excessive bleeding are all positive signs of their possible integration in the clinical treatment of blood coagulation diseases in the future. Additionally, by avoiding biological barriers and enhancing a medication’s therapeutic index, NDDSs offer superior drug delivery to targeted treatment or even directly to the injured site as compared to standard anticoagulants. Emerging scientific and clinical data suggests that NDDS may be a safe, efficacious, and attractive treatment option for a variety of cardiovascular disorders, including thromboembolic syndromes. This study presents an updated knowledge of nanocarriers for thrombosis as well as an outline of recent significant breakthroughs in targeted thrombosis therapy. [ABSTRACT FROM AUTHOR]

18. Antiviral Activity of Cinchona officinalis , a Homeopathic Medicine, against COVID-19.

Abstract

Background Coronavirus disease 2019 (COVID-19) is a potentially fatal disease caused by the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Several studies have shown that hydroxychloroquine (HCQ) significantly inhibits SARS-CoV-2 infections in vitro. Objective Since the phytoconstituents of Cinchona officinalis (CO) are similar to those of HCQ, the objective of this study was to test the antiviral potential of different homeopathic formulations of CO. Methods An analysis of the molecular composition of CO was carried out using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry, followed by a detailed docking study. The constituents of CO were docked against various targets of SARS-CoV-2, and the binding potential of the phytoconstituents was compared and quantified. The ligand with the lowest Glide docking score is considered to have the best binding affinity. The cytotoxicity of several homeopathic formulations, including CO mother tincture (CO-MT), was also checked on VeroE6 cells. A known antiviral, remdesivir, was used as a positive control for the in vitro assays to evaluate the effects of CO-MT against SARS-CoV-2-infected VeroE6 cells. Results Molecular docking studies showed that constituents of CO exhibited binding potential to various targets of SARS-CoV-2, including Mpro, PLpro, RdRp, nucleocapsid protein, ACE2 (in host) and spike protein. Quinoline, one of the constituents of CO, can potentially bind the spike protein of SARS-CoV-2. Quinic acid showed better binding capabilities with Mpro, PLpro RdRp, nucleocapsid protein and ACE2 (allosteric site) than other constituents. Quinidine exhibited better binding to ACE2. Compared to HCQ, other phytoconstituents of CO had the equivalent potential to bind the RNA-dependent RNA polymerase, nucleocapsid protein, Mpro, PLpro and spike protein of SARS-CoV-2. In vitro assays showed that homeopathic CO-MT was not cytotoxic and that CO-MT and remdesivir respectively caused 89% and 99% inhibition of SARS-CoV-2 infection in VeroE6 cells. Conclusion Based on this in silico and in vitro evidence, we propose CO-MT as a promising antiviral medicine candidate for treating COVID-19. Abstract

Salinity is one of the environmental stresses that alter protein expression. To evaluate that, two ICRISAT Pennisetum glaucum (pearl millet) genotypes (IP 19586 and IP 22269) were subjected to 0.3 dSm-1 and 10 dSm-1 salinities. The agronomic parameters and ion content were assessed to find the tolerance level of the genotype to salinity. Differential expression of proteins on salt stress in both genotypes were analyzed by 2-dimensional gel electrophoresis. MALDI–MS/MS-TOF-TOF analysis helped in ascertain the amino acid sequences and the MASCOT search suggested the biological process of the predicted proteins. IP 22269 exclude the accumulation of toxic level of sodium in leaf. Based on physiological responses, IP 22269 was proposed to be salt-sensitive, and IP 19586 salt-tolerant genotypes. 67 proteins were responsive either less or more abundant to salinity. As per findings, the proteins responsive to salt stress are granule-bound starch synthase 1, alpha-amylase, Meiotic recombination protein SPO11, flavonoid 3'- monooxygenase, ethylene receptor 4, protein transcription initiation factor Y 11g, and auxin-responsive protein IAA16. Further, based on the STRING analysis these proteins form a protein-protein interaction network with 171 proteins. This research elucidates few salt stress tolerance traits and network between the salt stress-responsive mechanisms in pearl millet.
