Buffalo is a dominant dairy animal in many agriculture-based economies. However, the poor reproductive efficiency (low conception rate) of the buffalo bulls constrains the realization of its full production potential. This in turn leads to economic and welfare issues, especially for the marginal farmers in such economies. The mammalian sperm surface proteins have been implicated in the regulation of survival and function of the spermatozoa in the female reproductive tract (FRT). Nonetheless, the lack of specific studies on buffalo sperm surface makes it difficult for researchers to explore and investigate the role of these proteins in the regulation of mechanisms associated with sperm protection, survival, and function. Abstract In pregnant animals, communication between the mother and conceptus occurs via extracellular vesicles (EVs) that carry several biomolecules such as nucleic acids (miRNAs, mRNAs), proteins, and lipids. At the time of implantation, the endometrium undergoes several morphological and physiological changes, such as angiogenesis, apoptosis, and cell proliferation regulation at the implantation site, to attain a receptive state. Abstract Quantifiable decline in the maternal body temperature during the pre-calving offers the possibilities for predicting the calving that can improve the calving management. As infrared thermography (IRT) is a simple non-contact tool for precise measurement of surface temperature, we investigated the use of IRT to establish thermal signatures around calving in the Murrah buffalo. The IRT of eye, right lateral, left lateral and rear side of udder skin surface temperature (USST) were recorded at 6 h interval from 96 h before the expected date of calving, at the time of calving and 24 h post-calving in Murrah buffaloes (n = 28). In parallel, blood samples were collected for progesterone (P4) assay. The results revealed that the IRT of the eye, right and left lateral and rear side of USST showed a significant decrease in the temperature from 48 h pre-calving till the onset of calving with a ΔT (°C) of 0.56, 0.91, 0.70, and 0.90, respectively. Mean USST significantly declined from 48 h pre-calving with a ΔT of 0.85 °C. The residual temperature of both eye and various ROI of the udder also followed a similar and significant declining trend from 48 to 0 h of calving indicating that circadian influence on the USST was minimum. Plasma P4 concentration significantly decreased from 72 h pre-calving till calving. It is concluded that a marked reduction in the IRT of the USST at 6–12 h pre-calving would be useful in predicting the onset of calving in the Murrah buffalo.

Abstract Agrichemicals such as organophosphorus pesticides’ metabolites (OPPMs) are more hazardous and pervasive than their parent pesticides. Parental germline exposure to such xenobiotics leads to an elevated susceptibility towards reproductive failures e.g. sub- or in-fertility. Abstract Bull fertility is of paramount importance in bovine industry because semen from a single bull is used to breed several thousands of cows; however, so far, no reliable test is available for bull fertility prediction. In the present study, spermatozoa from high- and low-fertility bulls were subjected to high-throughput transcriptomic, proteomic and metabolomic analysis. Using an integrated multi-omics approach the molecular differences between high- and low-fertility bulls were identified. We identified a total of 18,068 transcripts, 5041 proteins and 3704 metabolites in bull spermatozoa, of which the expression of 4766 transcripts, 785 proteins and 33 metabolites were dysregulated between high- and low-fertility bulls. At transcript level, several genes involved in oxidative phosphorylation pathway were found to be downregulated, while at protein level genes involved in metabolic pathways were significantly downregulated in low-fertility bulls. We found that metabolites involved in Taurine and hypotaurine metabolism were significantly downregulated in low-fertility bulls. Integrated multi-omics analysis revealed the interaction of dysregulated transcripts, proteins and metabolites in major metabolic pathways, including Butanoate metabolism, Glycolysis and gluconeogenesis, Methionine and cysteine metabolism, Phosphatidyl inositol phosphate, pyrimidine metabolism and saturated fatty acid beta oxidation. These findings collectively indicate that molecules governing sperm metabolism potentially influence bull fertility.

The water buffalo (Bubalus bubalis) is an indispensable part of the Indian dairy sector and in several instances, the farmers incur economic losses due to failed pregnancy after artificial insemination (AI). One of the key factors for the failure of conception is the use of semen from the bulls of low fertilizing potential and hence, it becomes important to predict the fertility status before performing AI. In this study, the global proteomic profile of high fertile (HF) and low fertile (LF) buffalo bull spermatozoa was established using a high-throughput LC-MS/MS technique. A total of 1,385 proteins (≥1 high-quality PSM/s, ≥1 unique peptides, p < 0.05, FDR < 0.01) were identified out of which, 1,002 were common between both the HF and LF groups while 288 and 95 proteins were unique to HF and LF groups respectively. We observed 211 and 342 proteins were significantly high (log Fc ≥ 2) and low abundant (log Fc ≤ 0.5) in HF spermatozoa (p < 0.05). Gene ontology analysis revealed that the fertility associated high abundant proteins in HF were involved in spermatogenesis, sperm motility, acrosome integrity, zona pellucida binding and other associated sperm functions. Besides this, the low abundant proteins in HF were involved in glycolysis, fatty acid degradation and inflammation. Furthermore, fertility related differentially abundant proteins (DAPs) on sperm viz., AKAP3, Sp17, and DLD were validated through Western blotting and immunocytochemistry which was in coherence with the LC-MS/MS data. The DAPs identified in this study may be used as potential protein candidates for predicting fertility in buffaloes. Our findings provide an opportunity in mitigating the economic losses that farmers incur due to male infertility.

The present study quantitatively characterized the proteomic changes in bull spermatozoa induced by the cryopreservation process. We performed high-throughput comparative global proteomic profiling of freshly ejaculated (before cryopreservation), equilibrated (refrigerated storage; during cryopreservation), and frozen (ultralow temperature; after cryopreservation) bull spermatozoa. Using the liquid chromatography–mass spectrometry (LC-MS/MS) technique, a total of 1,692, 1,415, and 1,286 proteins were identified in fresh, equilibrated, and cryopreserved spermatozoa, respectively. When the proteome of fresh spermatozoa was compared with equilibrated spermatozoa, we found that 166 proteins were differentially expressed. When equilibrated spermatozoa were compared with cryopreserved spermatozoa, we found that 147 proteins were differentially expressed between them. Similarly, we found that 156 proteins were differentially expressed between fresh and cryopreserved spermatozoa. Among these proteins, the abundance of 105 proteins was lowered during the equilibration process itself, while the abundance of 43 proteins was lowered during ultralow temperature preservation. Remarkably, the equilibration process lowered the abundance of sperm proteins involved in energy metabolism, structural integrity, and DNA repair and increased the abundance of proteins associated with proteolysis and protein degradation. The abundance of sperm proteins associated with metabolism, cGMP-PKG (cyclic guanosine 3′,5′-monophosphate-dependent protein kinase G) signaling, and regulation of the actin cytoskeleton was also altered during the equilibration process. Collectively, the present study showed that the equilibration step in the bull sperm cryopreservation process was the critical point for sperm proteome, during which a majority of proteomic alterations in sperm occurred. These findings are valuable for developing efficient protocols to minimize protein damage and to improve the quality and fertility of cryopreserved bull spermatozoa.

In bovines, cryopreserved semen is used for artificial insemination; however, the fertility of cryopreserved semen is far lower than that of fresh semen. Although cryopreservation alters sperm phenotypic characteristics, its effect on sperm molecular health is not thoroughly understood. The present study applied next-generation sequencing to investigate the effect of cryopreservation on the sperm transcriptomic composition of bull spermatozoa. While freshly ejaculated bull spermatozoa showed 14,280 transcripts, cryopreserved spermatozoa showed only 12,375 transcripts. Comparative analysis revealed that 241 genes were upregulated, 662 genes were downregulated, and 215 genes showed neutral expression in cryopreserved spermatozoa compared to fresh spermatozoa. Gene ontology analysis indicated that the dysregulated transcripts were involved in nucleic acid binding, transcription-specific activity, and protein kinase binding involving protein autophosphorylation, ventricular septum morphogenesis, and organ development. Moreover, the dysregulated genes in cryopreserved spermatozoa were involved in pathways associated with glycogen metabolism, MAPK signalling, embryonic organ morphogenesis, ectodermal placode formation, and regulation of protein auto-phosphorylation. These findings suggest that the cryopreservation process induced alterations in the abundance of sperm transcripts related to potential fertility-associated functions and pathways, which might partly explain the reduced fertility observed with cryopreserved bull spermatozoa.

Acrosome intactness of spermatozoa is the critical factor for establishing sperm reservoir in oviduct and for fertilizing an oocyte. However, frozen thawed spermatozoa tend to show higher proportion of acrosome reacted spermatozoa thereby compromising the fertility. Conventional staining techniques identify only sperm acrosome integrity and not precisely the acrosome reaction status. In this context, the current study was conducted to assess the acrosome status of cryopreserved spermatozoa using Fluorescein isothiocyanate conjugated peanut agglutinin and propidium iodide (FITC-PNA+PI) in stallions with varying sperm motility. Stallions were classified into high- (≥45%) and low-motile group (≤30%) based on their post-thaw sperm motility. The proportion of live acrosome intact (LAI) spermatozoa was significantly higher in high-motile group as compared to low-motile group. A significant positive correlation was observed between LAI and post-thaw sperm motility. In conclusion, the present study showed that FITC-PNA+PI combination could be used for rapid and accurate assessment of acrosome reaction status of stallion spermatozoa, and the proportion of LAI population in cryopreserved stallion semen had a strong correlation with sperm motility.

Abstract Background Low conception rate (CR) despite insemination with morphologically normal spermatozoa is a common reproductive restraint that limits buffalo productivity. This accounts for a significant loss to the farmers and the dairy industry, especially in agriculture-based economies. The immune-related proteins on the sperm surface are known to regulate fertility by assisting the spermatozoa in their survival and performance in the female reproductive tract (FRT). Regardless of their importance, very few studies have specifically catalogued the buffalo sperm surface proteome. The study was designed to determine the identity of sperm surface proteins and to ascertain if the epididymal expressed beta-defensins (BDs), implicated in male fertility, are translated and applied onto buffalo sperm surface along with other immune-related proteins. Results The raw mass spectra data searched against an in-house generated proteome database from UniProt using Comet search engine identified more than 300 proteins on the ejaculated buffalo sperm surface which were bound either by non-covalent (ionic) interactions or by a glycosylphosphatidylinositol (GPI) anchor. The singular enrichment analysis (SEA) revealed that most of these proteins were extracellular with varied binding activities and were involved in either immune or reproductive processes. Flow cytometry using six FITC-labelled lectins confirmed the prediction of glycosylation of these proteins. Several beta-defensins (BDs), the anti-microbial peptides including the BuBD-129 and 126 were also identified amongst other buffalo sperm surface proteins. The presence of these proteins was subsequently confirmed by RT-qPCR, immunofluorescence and in vitro fertilization (IVF) experiments. Conclusions The surface of the buffalo spermatozoa is heavily glycosylated because of the epididymal secreted (glyco) proteins like BDs and the GPI-anchored proteins (GPI-APs). The glycosylation pattern of buffalo sperm-surface, however, could be perturbed in the presence of elevated salt concentration or incubation with PI-PLC. The identification of numerous BDs on the sperm surface strengthens our hypothesis that the buffalo BDs (BuBDs) assist the spermatozoa either in their survival or in performance in the FRT. Our results suggest that BuBD-129 is a sperm-surface BD that could have a role in buffalo sperm function. Further studies elucidating its exact physiological function are required to better understand its role in the regulation of male fertility.

This study aimed to estimate the changes in the milk yield, milk fat, energy indicators [NEFA, BHBA, Dry Matter Intake (DMI) and Body Condition Score (BCS)] and concentration of innate immune molecules (Haptoglobin: Hp, Serum Amyloid A: SAA, TLR-4, TNF-α, IL-1β, IL-6, and IL-8), during the transition period in primiparous and multiparous dual-purpose zebu (Deoni) cows. The blood sample was collected at weekly intervals during pre-partum (-21±2, -14±1, -7±1, d), date of calving (day 0), and postpartum period (3±1, 7±1, 14±1, 21±2 d) for estimation of the above plasma variables using commercially available bovine specific ELISA kits. DMI and BCS during the corresponding period were also recorded. Data were analyzed using a linear mixed model considering group, time and their interaction as fixed effects. Group, time and their interaction had significant effect on DMI where primiparous cows consumed higher DMI during early postpartum period as compared to multiparous cows. Group alone had significant effect on milk yield, milk fat per cent and BHBA level while time alone influenced BCS. The interaction of group and time had significant effects on plasma TLR-4 and IL-8 concentration. Group also had significant effect on Hp and TNF-α levels. It was concluded that parity had significant effect on metabolic and immune indicators where higher DMI during transition period resulted in more milk yield in primiparous than multiparous indigenous (Deoni) cows.

Abstract Background The incidence of poor semen quality and sub-fertility/infertility is higher in crossbred as compared to Zebu males. Several attempts have been made to understand the possible reasons for higher incidence of fertility problems in crossbred males, at sperm phenotype, proteome and genome level but with variable results. Since the quality of the ejaculated spermatozoa is determined by the testicular environment, assessing the testicular transcriptome between these breeds would help in identifying the possible mechanisms associated with infertility in crossbred bulls. However, such information is not available. We performed global transcriptomic profiling of testicular tissue from crossbred and Zebu bulls using Agilent Bos taurus GXP 8X60k AMADID: 29411 array. To the best of our knowledge, this is the first study comparing the testicular mRNAs between crossbred and Zebu bulls. Results Out of the 14,419 transcripts detected in bovine testis, 1466 were differentially expressed between crossbred and Zebu bulls, in which 1038 were upregulated and 428 were downregulated in crossbred bulls. PI4KB and DPY19L2 genes, reported to be involved in sperm capacitation and acrosome formation respectively, were among the top 10 downregulated transcripts in crossbred testis. Genes involved in ubiquitination and proteolysis were upregulated, while genes involved in cell proliferation, stem cell differentiation, stem cell population maintenance, steroidogenesis, WNT signalling, protein localization to plasma membrane, endocannabinoid signalling, heparin binding, cAMP metabolism and GABA receptor activity were downregulated in crossbred testis. Among the 10 genes validated using qPCR, expression of CCNYL, SOX2, MSMB, SPATA7, TNP1, TNP2 and CRISP2 followed the same trend as observed in microarray analysis with SPATA7 being significantly downregulated and transition proteins (TNP1, TNP2) being significantly upregulated in crossbred bulls. Conclusions Abundant proteolysis by ubiquitination and downregulation of WNT signaling, cell proliferation, differentiation and steroidogenesis might be associated with higher incidence of poor semen quality and/or sub-fertility/infertility in crossbred bulls as compared to Zebu bulls. Downregulation of SPATA7 (Spermatogenesis Associated 7) and upregulation of transition proteins (TNP1 and TNP2) in crossbred bull testis might be associated with impaired spermatogenesis processes including improper chromatin compaction in crossbred bulls.

Spermatozoa carries a reservoir of mRNAs regulating sperm functions and fertilizing potential. Although it is well recognized that a considerable proportion of high genetic merit breeding bulls produce poor-quality semen, the transcriptomic alterations in spermatozoa from such bulls are not understood. In the present study, comparative high-throughput transcriptomic profiling of spermatozoa from good and poor-quality semen-producing bulls was carried out to identify the transcripts associated with semen quality. Using next-generation sequencing (NGS), we identified 11,632 transcripts in Holstein Friesian bull spermatozoa; after total hit normalization, a total of 544 transcripts were detected, of which 185 transcripts were common to both good and poor-quality semen, while 181 sperm transcripts were unique to good quality semen, and 178 transcripts were unique to poor-quality semen. Among the co-expressed transcripts, 31 were upregulated, while 108 were downregulated, and 46 were neutrally expressed in poor-quality semen. Bioinformatics analysis revealed that the dysregulated transcripts were predominantly involved in molecular function, such as olfactory receptor activity and odor binding, and in biological process, such as detection of chemical stimulus involved in sensory perception, sensory perception of smell, signal transduction, and signal synaptic transmission. Since a majority of the dysregulated transcripts were involved in the olfactory pathway (85% of enriched dysregulated genes were involved in this pathway), the expression of selected five transcripts associated with this pathway (OR2T11, OR10S1, ORIL3, OR5M11, and PRRX1) were validated using real-time qPCR, and it was found that their transcriptional abundance followed the same trend as observed in NGS; the sperm transcriptional abundance of OR2T11 and OR10S1 differed significantly (p < 0.05) between good and poor-quality semen. It is concluded that poor-quality semen showed altered expression of transcripts associated with olfactory receptors and pathways indicating the relationship between olfactory pathway and semen quality in bulls.

Male fertility is extremely important in dairy animals because semen from a single bull is used to inseminate several thousand females. Asthenozoospermia (reduced sperm motility) and oligozoospermia (reduced sperm concentration) are the two important reasons cited for idiopathic infertility in crossbred bulls; however, the etiology remains elusive. In this study, using a non-targeted liquid chromatography with tandem mass spectrometry-based approach, we carried out a deep metabolomic analysis of spermatozoa and seminal plasma derived from normozoospermic and astheno-oligozoospermic bulls. Using bioinformatics tools, alterations in metabolites and metabolic pathways between normozoospermia and astheno-oligozoospermia were elucidated. A total of 299 and 167 metabolites in spermatozoa and 183 and 147 metabolites in seminal plasma were detected in astheno-oligozoospermic and normozoospermic bulls, respectively. Among the mapped metabolites, 75 sperm metabolites were common to both the groups, whereas 166 and 50 sperm metabolites were unique to astheno-oligozoospermic and normozoospermic bulls, respectively. Similarly, 86 metabolites were common to both the groups, whereas 45 and 37 seminal plasma metabolites were unique to astheno-oligozoospermic and normozoospermic bulls, respectively. Among the differentially expressed metabolites, 62 sperm metabolites and 56 seminal plasma metabolites were significantly dysregulated in astheno-oligozoospermic bulls. In spermatozoa, selenocysteine, deoxyuridine triphosphate, and nitroprusside showed significant enrichment in astheno-oligozoospermic bulls. In seminal plasma, malonic acid, 5-diphosphoinositol pentakisphosphate, D-cysteine, and nicotinamide adenine dinucleotide phosphate were significantly upregulated, whereas tetradecanoyl-CoA was significantly downregulated in the astheno-oligozoospermia. Spermatozoa from astheno-oligozoospermic bulls showed alterations in the metabolism of fatty acid and fatty acid elongation in mitochondria pathways, whereas seminal plasma from astheno-oligozoospermic bulls showed alterations in synthesis and degradation of ketone bodies, pyruvate metabolism, and inositol phosphate metabolism pathways. The present study revealed vital information related to semen metabolomic differences between astheno-oligozoospermic and normospermic crossbred breeding bulls. It is inferred that fatty acid synthesis and ketone body degradations are altered in the spermatozoa and seminal plasma of astheno-oligozoospermic crossbred bulls. These results open up new avenues for further research, and current findings can be applied for the modulation of identified pathways to restore sperm motility and concentration in astheno-oligozoospermic bulls.

In this study, we evaluated the peripheral concentrations of Toll-like receptors (TLR)-4 during transition period in relation to postpartum productive and reproductive performances of Deoni (zebu) cattle. Accuracy and threshold values of TLR-4 to predict the postpartum performance, were also estimated using receiver operating characteristics (ROC) analysis. Blood samples were collected at weekly intervals during transition period (from 21 days before to 21 days after calving) and TLR-4 concentration was estimated using bovine specific ELISA kits. Plasma TLR-4 concentration was significantly higher on third day of postpartum in cows that became pregnant within breeding period than the cows that remained non-pregnant (4.48 vs 1.80 ng/mL). ROC analysis revealed that the accuracy of TLR-4 for predication of ability to become pregnant within breeding period was acceptable (AUC: 0.75) with a threshold value of 2.13 ng/mL. It is concluded that concentrations of TLR-4, during transition period, could be used for predicting the possibilities of Deoni cows getting pregnant within the breeding period with moderate accuracy.

Receiver Operating Characteristics (ROC) analysis is a popular method to discriminate between the two conditions of tested animals. In this study, we estimated accuracy and threshold values of metabolic (Dry matter Intake; DMI and Body Condition Score: BCS, NEFA and BHBA) and immune indicators (Haptoglobin: Hp, Serum Amyloid A: SAA, IL-6, TNF-a, IL-1b, and IL-8) during transition period (–21, –14, –7, 0, +3, +7, +14 and +21 days) to predict the high yielding (HY) and pregnant Deoni cows. ROC analysis revealed that SAA (–21 d), IL-6 (–21 and –7 d), BCS (–7 d) and BHBA (–7 d) during pre-partum period, differentiated HY from low or medium yielder (LY/MY) cows with moderate to excellent accuracy (AUC >0.8). During postpartum period, IL-6 (+7 d), TNF-a (+21 d), DMI (+21 d), NEFA (+14 d and +21 d) and BHBA (+21 d) levels had moderate to excellent accuracy to differentiate HY from LY or MY cows. IL-6 (–14 d and –7 d), TNF-a (–14 d) and DMI (–21 d; above 2 kg/100 kg BW) during pre-partum period while, SAA (+3 d and +7 d), IL-6 (+3 and +21 d) and TNF-a (+7 and +21 d) during postpartum period were significantly predicted the pregnant cows with moderate to excellent accuracy. Altogether, it is concluded that SAA, IL-6 and TNF-a levels had higher accuracy in discrimination of HY and pregnant cows from LY or MY and non-pregnant cows, respectively. Therefore, their corresponding threshold values could be used for predicting HY and pregnant Zebu (Deoni) cows.

Objective: To assess the stability of 10 candidate internal control genes (ICGs), namely GAPDH, ACTB, RPL23, RPS15A, ATPSF1, GLUT5, HMBS, ATP2B4, PPIA, and BRP to normalize the transcriptional data from testes samples of Zebu and crossbred bulls. Methods: Total RNA was isolated from testicular tissue of Zebu and crossbred bulls (n=6 each) between 2-8 years of age. cDNA was synthesized, and the quantitative real-time polymerase chain reaction (PCR) was performed. The cycle threshold values were used for the analysis of the stability of ICGs. Four different statistical algorithms: geNorm, Normfinder, BestKeeper, and RefFinder, were used to assess the stability of these genes. Results: ATPSF1, HMBS, PPIA, and RPS15A were the most reliable and stable ICGs for Zebu testes, and ATPSF1, RPL23, and PPIA for crossbred testes. Conclusions: A panel of stable ICGs (ATPSF1, HMBS, PPIA, RPS15A for Zebu and ATPSF1, RPL23, and PPIA for crossbred) for normalization of gene expression data in testes samples can be helpful for researchers to conduct functional genomics studies at the testicular level in cattle bulls.

Understanding the changes in the spermatozoa during cryopreservation is indispensable for tailoring and increasing the efficiency of cryopreservation process success. However, the dynamics of damage to sperm organelles during different stages of cryopreservation is underexplored. This study assessed the mitochondrial membrane potential (MMP) and DNA damage during different stages of cryopreservation, viz. immediately after ejaculation, after equilibration and after freezing and thawing in cattle and buffalo spermatozoa using flow cytometry. Proportion of spermatozoa with high MMP decreased significantly after equilibration (from 66.06±4.59 to 42.58±6.30 in Holstein bulls and from 60.32±5.51 to 39.98±7.58 in buffalo bulls). Sperm DNA integrity [DNA fragmentation index (DFI %)] in Holstein Friesian (HF) bulls did not differ significantly between fresh and equilibrated samples but a significantly higher % DFI was observed in frozen-thawed semen samples as compared to both fresh and equilibrated samples. In contrast, % DFI in buffalo spermatozoa did not differ among the three stages of cryopreservation. It was concluded that mitochondrial damages occur during equilibration while chromatin damages occur during freezing and thawing of cattle bull spermatozoa; whereas buffalo bull spermatozoa were lesser susceptible to DNA damage during cryopreservation as compared to cattle spermatozoa.

Crossbreeding of indigenous cattle (Bos indicus) with improved (Bos taurus) breeds gained momentum and economic relevance in several countries to increase milk production. While production performance of the crossbred offspring is high due to hybrid vigor, they suffer from a high incidence of reproductive problems. Specifically, the crossbred males suffer from serious forms of subfertility/infertility, which can have a significant effect because semen from a single male is used to breed several thousand females. During the last two decades, attempts have been made to understand the probable reasons for infertility in crossbred bulls. Published evidence indicates that testicular cytology indices, hormonal concentrations, sperm phenotypic characteristics and seminal plasma composition were altered in crossbred compared to purebred males. A few recent studies compared crossbred bull semen with purebred bull semen using genomics, transcriptomics, proteomics and metabolomics; molecules potentially associated with subfertility/infertility in crossbred bulls were identified. Nevertheless, the precise reason behind the poor quality of semen and high incidence of sub-fertility/infertility in crossbred bulls are not yet well defined. To identify the underlying etiology for infertility in crossbred bulls, a thorough understanding of the magnitude of the problem and an overview of the prior art is needed; however, such systematically reviewed information is not available. Therefore, the primary focus of this review is to compile and analyze earlier findings on crossbred bull fertility/infertility. In addition, the differences between purebred and crossbred males in terms of testicular composition, sperm phenotypic characteristics, molecular composition, environmental influence and other details are described; future prospects for research on crossbred males are also outlined.

Crossbred bulls produced by crossing Bos taurus and Bos indicus suffer with high incidence of infertility/subfertility problems; however, the etiology remains poorly understood. The uncertain predictability and the inability of semen evaluation techniques to maintain constant correlation with fertility demand for alternate methods for bull fertility prediction. Therefore, in this study, the global differential gene expression between high- and low-fertile crossbred bull sperm was assessed using a high-throughput RNA sequencing technique with the aim to identify transcripts associated with crossbred bull fertility. Crossbred bull sperm contained transcripts for 13,563 genes, in which 2,093 were unique to high-fertile and 5,454 were unique to low-fertile bulls. After normalization of data, a total of 776 transcripts were detected, in which 84 and 168 transcripts were unique to high-fertile and low-fertile bulls, respectively. The aim of this study was to estimate the prevalence of subclinical mastitis (SCM), its innate immunity and to compare the efficacy of indirect diagnostic tests [California mastitis test (CMT), differential electrical conductivity (DEC), absolute EC, enzyme based color detection method (Tanucheck kit) and brothymol blue strip (BTB) tests] to milk somatic cell count (SCC) method using 200×103 cells/ml as a cut off value in Deoni (N=27) and HF crossbred (N=32) cows. Cumulative prevalence of SCM was 38% and 63% in Deoni and HF crossbred cows, respectively on milk SCC basis (N=215). Breed, udder health status and its interaction had significant effect on SCC level. The overall sensitivity and specificity of above indirect tests were 38 and 99%; 52 and 72%; 52 and 69%; 76 and 31%, and 1 and 100%, respectively. mRNA expression of serum amyloid A (SAA), IL-1β and TNF- α genes were significantly up-regulated while, TNF-α gene was significantly down-regulated in PBMC of SCM affected HF crossbred and Deoni cows, respectively. It is concluded that, DEC is more suitable indirect method to detect SCM and mRNA expression of SAA and TNF-α was strongly related to SCM in HF crossbred and Deoni cows, respectively.

Although, it is known that spermatozoa harbor a variety of RNAs that may influence embryonic development, little is understood about sperm transcriptomic differences in relation to fertility, especially in buffaloes. In the present study, we compared the differences in sperm functional attributes and transcriptomic profile between high- and low-fertile buffalo bulls. Sperm membrane and acrosomal integrity were lower (P < 0.05), while protamine deficiency and lipid peroxidation were higher (P < 0.05) in low- compared to high-fertile bulls. Transcriptomic analysis using mRNA microarray technology detected a total of 51,282 transcripts in buffalo spermatozoa, of which 4,050 transcripts were differentially expressed, and 709 transcripts were found to be significantly dysregulated (P < 0.05 and fold change >1) between high- and low-fertile bulls. Majority of the dysregulated transcripts were related to binding activity, transcription, translation, and metabolic processes with primary localization in the cell nucleus, nucleoplasm, and in cytosol. Pathways related to MAPK signaling, ribosome pathway, and oxidative phosphorylation were dysregulated in low-fertile bull spermatozoa. Using bioinformatics analysis, we observed that several genes related to sperm functional attributes were significantly downregulated in low-fertile bull spermatozoa. Validation of the results of microarray analysis was carried out using real-time qPCR expression analysis of selected genes (YBX1, ORAI3, and TFAP2C). The relative expression of these genes followed the same trend in both the techniques. Collectively, this is the first study to report the transcriptomic profile of buffalo spermatozoa and to demonstrate the dysregulation of functionally relevant transcripts in low-fertile bull spermatozoa. The results of the present study open up new avenues for understanding the etiology for poor fertility in buffalo bulls and to identify fertility biomarkers.

The glycans on the plasma membrane of cells manifest as the glycocalyx, which serves as an information-rich frontier that is directly in contact with its immediate milieu. The glycoconjugates (GCs) that adorn most of the mammalian cells are also abundant in gametes, especially the spermatozoa where they perform unique reproduction-specific functions e.g., inter-cellular recognition and communication. This study aimed to implicate the sperm glycosylation pattern as one of the factors responsible for low conception rates observed in buffalo bulls. We hypothesized that a differential abundance of glycans exists on the spermatozoa from bulls of contrasting fertilizing abilities endowing them with differential immune evasion abilities. Therefore, we investigated the role of glycan abundance in the phagocytosis and NETosis rates exhibited by female neutrophils (PMNs) upon exposure to such spermatozoa. Our results indicated that the spermatozoa from high fertile (HF) bulls possessed a higher abundance of O-linked glycans e.g., galactosyl (β-1,3)N-acetylgalactosamine and N-linked glycans like [GlcNAc]1-3, N-acetylglucosamine than the low fertile (LF) bull spermatozoa. This differential glycomic endowment appeared to affect the spermiophagy and NETosis rates exhibited by the female neutrophil cells (PMNs). The mean percentage of phagocytizing PMNs was significantly different (P < 0.0001) for HF and LF bulls, 28.44 and 59.59%, respectively. Furthermore, any introduced perturbations in the inherent sperm glycan arrangements promoted phagocytosis by PMNs. For example, after in vitro capacitation the mean phagocytosis rate (MPR) rate in spermatozoa from HF bulls significantly increased to 66.49% (P < 0.01). Likewise, the MPR increased to 70.63% (p < 0.01) after O-glycosidase & α2-3,6,8,9 Neuraminidase A treatment of spermatozoa from HF bulls. Moreover, the percentage of PMNs forming neutrophil extracellular traps (NETs) was significantly higher, 41.47% when exposed to spermatozoa from LF bulls vis-à-vis the spermatozoa from HF bulls, 15.46% (P < 0.0001). This is a pioneer report specifically demonstrating the role of O-linked glycans in the immune responses mounted against spermatozoa. Nevertheless, further studies are warranted to provide the measures to diagnose the sub-fertile phenotype thus preventing the losses incurred by incorrect selection of morphologically normal sperm in the AI/IVF reproduction techniques.

The accurate prediction of male fertility is of major economic importance in the animal breeding industry. However, the results of conventional semen analysis do not always correlate with field fertility outcomes. There is evidence to indicate that mammalian fertilization and subsequent embryo development depend, in part, on the inherent integrity of the sperm DNA. Understanding the complex packaging of mammalian sperm chromatin and assessment of DNA integrity could potentially provide a benchmark in clinical infertility. In the era of assisted reproduction, especially when in-vitro fertilization or gamete intrafallopian transfer or intracytoplasmic sperm injection is used, assessment of sperm DNA integrity is important because spermatozoa are not subjected to the selection process occurring naturally in the female reproductive tract. Although sperm DNA integrity testing measures a significant biological parameter, its precise role in the infertility evaluation in farm animals remains unclear. In this review, the earlier findings on sperm DNA integrity in relation to male fertility are compiled and analyzed. Furthermore, the causes and consequences of sperm DNA damage are described, together with a review of advances in methods for detection of sperm DNA damage, and the prognostic value of sperm DNA quality on male fertility.

The aim of this study was to investigate the peripheral concentration of metabolic and inflammatory indicators during transition period and their relationship in cows that developed clinical endometritis (CE) and remained nonendometritis during later postpartum period. A total of 70 cows were selected and blood was collected from 21 days before calving to 21 days after calving. After adjusting the sampling date based on the actual calving date (–21±2, –7±1, 0, 7±1, 14±1 and 21±2 days) and health status, a minimum of 8 samples each from the CE and nonendometritis cows were used at each time point of transition period for the estimation of acute phase proteins (APPs) (haptoglobin, Hp; serum amyloid A, SAA; and alpha-1 acid glycoprotein, AGP), pro-inflammatory cytokines (IL-1ô€…, TNF-ô€„, IL-6 and IL-8) and energy indicators (NEFA, BHBA, Leptin and IGF-I) using bovine specific ELISA kits. APPs (Hp, SAA and AGP), cytokines (IL-1ô€…, TNF-ô€„, IL-6) and energy indicators (NEFA, BHBA, Leptin) levels significantly increased, while IL-8 and IGF-I levels significantly decreased in transition cows that eventually developed to CE compared to the cows which remained non-endometritis during 22–47 days postpartum. It is concluded that cows that diagnosed as CE during postpartum period were experienced negative energy balance and compromised their immune status during transition period.

Among the uterine diseases, clinical endometritis (CE) is a major challenge to livestock farming as it causes sub- or infertility problems in dairy animals. The aim of this study was to evaluate the expression of cytokines (IL- 1β, TNF-α, IL-6, and IL-8), acute phase proteins [APPs; haptoglobin (Hp), serum amyloid A (SAA) and alpha-1 acid glycoprotein (AGP)] and energy indicators [leptin and insulin-like growth factor (IGF)-1] genes in uterine tissue of CE affected cows. The uterine biopsy from CE cows (4) and non-endometritis cows (4) was processed for quantitative real-time PCR to study the mRNA expression of these innate immune molecules. We observed that mRNA expression of SAA, IL-1β, IL-8 and leptin genes were significantly up-regulated while, TNF-α and IGF-I genes were significantly down-regulated in CE cows. It can be concluded that bovine APPs, cytokines and energy indicators genes are differentially expressed in CE affected cows.

Objective: To study the effects of different levels of exotic inheritance on ejaculate quality in bulls and its passage through different generations. Methods: Data on semen production traits and ejaculate quality were obtained for 38 crossbred bulls and grandsire-sire-progeny relationship in relation to semen quality was studied. The bulls were classified into three groups based on the level of exotic inheritance viz. F1, 50.0%-62.5% exotic germplasm and >75% exotic germplasm. Results: Results of the present study indicated that about 40% of the ejaculates obtained from the crossbred bulls were rejected from further processing due to poor ejaculate quality. The F1 bulls produced significantly higher proportions (57.00±10.00) of poor quality ejaculates compared to the interse mated bulls. The age at first semen collection in crossbred bulls ranged from 567 to 1 010 days with an average of 738.89±18.18 days while the mean age at first semen freezing was 865.72±34.60 days. Conclusions: It may be inferred that the “acceptable quality semen producing ability” decreased from grandsire through sire to male progeny and among the increasing exotic genetic levels of CB cattle, F1 bulls produced significantly higher “low grade ejaculates” that were unfit for cryopreservation.

Four crossbred cows with mummified fetus were utilized for the study. The cows weresubjected togynecologicalexamination and based on the findings the cases were diagnosed asmummified fetus. The cows were treated with 2 mg estradiol valerate and 500 μg cloprostenoland were examined every 12 hr after 24 hr of the treatment for cervical dilatation and othersigns related to fetal expulsion. The time duration between treatment and starting of cervicaldilatation ranged from 48 to 58h (53.00 ± 2.08 hr). Complete dilatation of cervix was observedafter 70.00 ± 2.94 hr post treatment (Range = 64-76 hr). The mean fetalcrown-rump length(CRL) was 31.5 cm, which ranged from 27.5 to 38 cm. The number of cotyledons in pregnanthorn also showed wide variation (Range 24-38 numbers) with mean ± SE of 30.3 ± 3.07numbers. In the placenta of three animals irregular shaped large adventitious cotyledons wereobserved in the inter-cotyledonary areas. Out of the four animals treated, three animals wereconceived within three estrous cycles and one animal had cystic ovary in the next cycle and wasnot conceived even after four cycles. It was concluded that the estradiolandprostaglandin F2α(PGF2α)combinationtherapy was effective for expulsion of mummified fetus in crossbred cowswithout affecting much on future fertility.
