Abstract Fluorescence polarization (Fpol) imaging of methylene blue (MB) is a promising quantitative approach to thyroid cancer detection. Clinical translation of MB Fpol technology requires reduction of the data analysis time that can be achieved via deep learning-based automated cell segmentation with a 2D U-Net convolutional neural network. The model was trained and tested using images of pathologically diverse human thyroid cells and evaluated by comparing the number of cells selected, segmented areas, and Fpol values obtained using automated (AU) and manual (MA) data processing methods. Overall, the model segmented 15.8% more cells than the human operator. Differences in AU and MA segmented cell areas varied between − 55.2 and + 31.0%, whereas differences in Fpol values varied from − 20.7 and + 10.7%. No statistically significant differences between AU and MA derived Fpol data were observed. The largest differences in Fpol values correlated with greatest discrepancies in AU versus MA segmented cell areas. Time required for auto-processing was reduced to 10 s versus one hour required for MA data processing. Implementation of the automated cell analysis makes quantitative fluorescence polarization-based diagnosis clinically feasible.

Abstract Background The life history and predation rate of the ladybird beetle, Harmonia dimidiata Fabricius (Coleoptera: Coccinellidae) were compared at four different temperatures (16, 20, 24, and 28 °C). The beetles were fed on the bird cherry-oat aphid, Rhopalosiphum padi Linnaeus (Hemiptera: Aphididae) and investigated at 70 ± 10% RH with a photoperiod of 14:10 h (L:D). Results The reproductive rates (R 0) were 20.07, 51.37, 66.95, and 14.54 beetle offspring at 16, 20, 24, and 28 °C, respectively. Results indicated that temperature had good impacts on the feeding potential, development, survivorship and fecundity, especially at 24 and 28 C compared with the other tested temperatures. In addition, the jackknife and bootstrap techniques were employed to estimate the population parameters' means. The obtained means of R 0 and other population parameters, using the bootstrap technique fit a normal distribution. Meanwhile, the jackknife technique generated biologically meaningless zero values for R 0. Both finite and predation rates were incorporated into limited predation rates for comparison of predation potential. Conclusion Both of the growth and predation rates indicated that H. dimidiata is more effective biocontrol agent for R. padi at 24 and 28 °C than at 20, 16 °C.

Purpose: The Islamic banking literature has been growing rapidly in the last decade. The aim of this study is to carry out a retrospective hybrid review to reveal this literature’s influential scientific actors (countries, institutions, journals, authors and documents), identify and discuss its most important streams, and finally, present a future research agenda. Methodology: We use a bibliometric approach, performing a review and objective analysis of 1,304 articles dealing with Islamic banking published during 1983–2021. We apply citation, keyword, and coauthorship analysis, as well as bibliographic coupling via VOSviewer software and Biblioshiny (an R package). Findings: We identify the influential aspects in the literature and discuss four important research streams: (1) overview, growth, and legal framework of Islamic banks; (2) Islamic banks’ performance and risk management practices; (3) customer and marketing perspectives of Islamic banking; and (4) the dynamics of efficiency in Islamic banks. Originality: This is one of the first studies to apply state-of-the-art methodology to review the literature related to Islamic banking and to highlight the dynamics of Islamic banks while presenting an extensive future research agenda.

Abstract Background The southeastern US is an epicenter for incident HIV in the US with high prevalence of human papillomavirus (HPV) co-infections. However, epidemiologies of HPV-associated clinical conditions (CC) among people living with HIV-1 infection (PLWH) are not fully known. Methods Electronic medical records (EMR) of PLWH attending one of the leading HIV clinics in the southeastern US between 2006 and 2018 were reviewed and analyzed. The retrospective study was nested within the University of Alabama at Birmingham HIV clinical cohort, which has electronically collected over 7000 PLWH’s clinical and sociobehavioral data since 1999. Incidence rates of HPV-related CC including anogenital warts, penile, anal, cervical, and vaginal/vulvar low- and high-grade squamous intraepithelial lesions (LSIL and HSIL) were estimated per 10,000 person years. Joinpoint regressions were performed to examine temporal changes in the trends of incident CC. All rates and trends were stratified by gender and race. Results Of the 4484 PLWH included in the study (3429 men, 1031 women, and 24 transgender), we observed 1038 patients with HPV-related CC. Abstract Background Atypical breast hyperplasias (AH) have a 10-year risk of progression to invasive cancer estimated at 4–7%, with the overall risk of developing breast cancer increased by ~ 4-fold. AH lesions are estrogen receptor alpha positive (ERα+) and represent risk indicators and/or precursor lesions to low grade ERα+ tumors. Therefore, molecular profiles of AH lesions offer insights into the earliest changes in the breast epithelium, rendering it susceptible to oncogenic transformation. Methods In this study, women were selected who were diagnosed with ductal or lobular AH, but no breast cancer prior to or within the 2-year follow-up. Paired AH and histologically normal benign (HNB) tissues from patients were microdissected. RNA was isolated, amplified linearly, labeled, and hybridized to whole transcriptome microarrays to determine gene expression profiles. Genes that were differentially expressed between AH and HNB were identified using a paired analysis. Gene expression signatures distinguishing AH and HNB were defined using AGNES and PAM methods. Regulation of gene networks was investigated using breast epithelial cell lines, explant cultures of normal breast tissue and mouse tissues. Results A 99-gene signature discriminated the histologically normal and AH tissues in 81% of the cases. Network analysis identified coordinated alterations in signaling through ERα, epidermal growth factor receptors, and androgen receptor which were associated with the development of both lobular and ductal AH. Decreased expression of SFRP1 was also consistently lower in AH. Knockdown of SFRP1 in 76N-Tert cells resulted altered expression of 13 genes similarly to that observed in AH. An SFRP1-regulated network was also observed in tissues from mice lacking Sfrp1. Re-expression of SFRP1 in MCF7 cells provided further support for the SFRP1-regulated network. Treatment of breast explant cultures with rSFRP1 dampened estrogen-induced progesterone receptor levels. Conclusions The alterations in gene expression were observed in both ductal and lobular AH suggesting shared underlying mechanisms predisposing to AH. Loss of SFRP1 expression is a significant regulator of AH transcriptional profiles driving previously unidentified changes affecting responses to estrogen and possibly other pathways. The gene signature and pathways provide insights into alterations contributing to AH breast lesions.

Salmonella is estimated to cause over a million infections and ~400 deaths annually in the U.S. Salmonella enterica serotype Javiana strains (n = 409) that predominantly originated from the State of Arkansas over a six-year period (2003 to 2008) were studied. This period coincided with a rapid rise in the incidence of S. Javiana infections in the U.S. Children under the age of 10 displayed the highest prevalence of S. Javiana infections, regardless of sex or year of detection. Antimicrobial susceptibility to 15 different antimicrobials was assessed and 92% (n = 375) were resistant to at least one of the antimicrobials. Approximately 89% of the isolates were resistant to sulfisoxazole alone and 3% (n = 11) were resistant to different antimicrobials, including gentamicin, ciprofloxacin or ceftiofur. The pulsed-field gel electrophoresis (PFGE) analyses assessed the genotypic diversity and distribution of S. Javiana strains using XbaI restriction. Nine major clusters were identified and isolates from each group were digested with the restriction enzyme AvrII. Isolates with identical profiles of XbaI and AvrII were found to be disseminated in human populations. These distinct “types” of S. Javiana were persistent in human populations for multiple years. A subset of isolates (n = 19) with unique resistance phenotypes underwent plasmid and incompatibility (Inc) type analyses and the isolates resistant to more than one antimicrobial harbored multiple plasmids (pagC, cdtB, and iroN. The whole genome sequences (WGS) of 18 isolates that mostly originated from Arkansas from 2003 to 2011 were compared with isolates collected from different areas in the U.S. in 1999, indicating the perseverance of S. Javiana in disseminating antimicrobial resistance and virulence genes.

Primary peritoneal serous carcinomas (PPSC) are exceedingly rare in male patients. Only a few cases were reported, and mostly with the limited immunophenotypical characterization. No molecular analysis of PPSC in males has been previously performed. We here describe another case of PPSC in a male patient. A comprehensive molecular analysis of the tumor revealed SF3B1 gene mutation as a possible driver.

Sulconazole has been reported to degrade into sulconazole sulfoxide via sulfur oxidation; however, structural characterization data was lacking and the potential formation of an N-oxide or sulfone could not be excluded. To clarify the degradation pathways and incorporate the impurity profile of sulconazole into the United States Pharmacopeia–National Formulary (USP–NF) monographs, a multifaceted approach was utilized to confirm the identity of the degradant. The approach combines stress testing of sulconazole nitrate, chemical synthesis of the degradant via a hydrogen peroxide-mediated oxidation reaction, semi-preparative HPLC purification, and structural elucidation by LC–MS/MS and NMR spectroscopy. Structural determination was primarily based on the comparison of spectroscopic data of sulconazole and the oxidative degradant. The mass spectrometric data have revealed a McLafferty-type rearrangement as the characteristic fragmentation pathway for alkyl sulfoxides with a β-hydrogen atom, and was used to distinguish the sulfoxide from N-oxide or sulfone derivatives. Moreover, the generated sulconazole sulfoxide was utilized as reference material for compendial procedure development and validation, which provides support for USP monograph modernization. Introduction There is very limited literature describing the association of lateral condyle mass (LCM) fracture of the distal humerus associated concurrently with olecranon fracture. Herein, a case is reported of a displaced LCM fracture with displaced olecranon fracture, due to complex trauma while getting out of a vehicle, which was managed by open reduction and internal fixation. Case Presentation A 4.5- year- old boy suffered severe pain and swelling around his right elbow due to trauma which he suffered while trying to get out of a vehicle which was in motion. Plain radiographs of the anteroposterior, lateral, and oblique views showed a displaced lateral mass fracture associated with a displaced olecranon fracture. Open reduction and internal fixation was carried out and near normal function was achieved. Conclusions In view of the paucity of literature and rare incidence of this injury, this case report highlights the need to be aware of other injuries which can occur in association with LCM fractures in children.

Summary The integrin α6β4 is defined as an adhesion receptor for laminins. Referred to as ‘β4’, this integrin plays a key role in the progression of various carcinomas through its ability to orchestrate key signal transduction events and promote cell motility. To identify novel downstream effectors of β4 function in breast cancer, microRNAs (miRNAs) were examined because of their extensive links to tumorigenesis and their ability to regulate gene expression globally. Two breast carcinoma cell lines and a collection of invasive breast carcinomas with varying β4 expression were used to assess the effect of this integrin on miRNA expression. A novel miRNA microarray analysis termed quantitative Nuclease Protection Assay (qNPA) revealed that β4 expression can significantly alter miRNA expression and identified two miRNA families, miR-25/32/92abc/363/363-3p/367 and miR-99ab/100, that are consistently downregulated by expression of this integrin. Analysis of published Affymetrix GeneChip data identified 54 common targets of miR-92ab and miR-99ab/100 within the subset of β4-regulated mRNAs, revealing several genes known to be key components of β4-regulated signaling cascades and effectors of cell motility. Gene ontology classification identified an enrichment in genes associated with cell migration within this population. Finally, gene set enrichment analysis of all β4-regulated mRNAs revealed an enrichment in targets belonging to distinct miRNA families, including miR-92ab and others identified by our initial array analyses. The results obtained in this study provide the first example of an integrin globally impacting miRNA expression and provide evidence that select miRNA families collectively target genes important in executing β4-mediated cell motility.

This paper presents an enhanced approach for classifying Bengali songs into different genres by leveraging feature importance analysis and deep learning techniques. The research addresses the challenge of limited data points in the Bengali Song Dataset by employing strategies, including audio segmentation and feature importance analysis, to enhance model performance. Multiple machine learning and deep learning architectures are evaluated to identify the most effective models for Bengali song classification. Additionally, this research conducts feature importance analysis to identify significant audio features contributing to classification accuracy. The best-performing deep learning model achieves an impressive validation accuracy of 94.17%, showcasing the project efficacy of the proposed methodology. Our findings highlight the effectiveness of our proposed methodology, demonstrating significant improvements in classification accuracy and contributing to advancements in Bengali music classification research. The tax reform has been instrumental in creating a common national market, enhancing the competitiveness of Indian businesses on a global scale. Moreover, the simplicity and transparency of GST have contributed to improving tax compliance, broadened the tax base, and boosted government revenues. This paper also delves into the economic implications of GST, particularly its impact on inflation, consumption patterns, and overall economic growth. By reducing the tax burden on both consumers and businesses, GST has stimulated economic demand and fostered investment. In recent years amid 2013-2024, Sino-Pak relations witnessed a qualitative transformation as bilateral ties expanded in strategic perspective. Pakistan has been forced to pursue a strategic collaboration with China in order to preserve the balance of power in South Asia as a result of the dynamic post 9/11 global political changes and expanding strategic partnership between the US and India. Under the auspices of the Indo-US strategic partnership, India is making most of its resources and collaborating with US in a number of areas, including energy, defense and security, so as to fulfill its ambitions of becoming a dominating force in the region and building blue water navy. India regional aspirations are too great for Pakistan to overcome on its own. In order to oppose India's imperial aspirations in South Asian area, the Sino-Pak formed a strategic alliance. This study will pose some queries about most recent iterations of this partnership. In this research, strategic partnership amid two countries is described using a Rational Actor Model. The research goes on to say that Sino-Pak will not be deterred from pursuing the project despite India's assertion that corridor route goes through disputed Northern Pakistani territory. This paper dissects prospects and difficulties for Pakistan at public and global levels for the making of mechanism to defeat the considerations of opponents. The researcher discusses outstanding features of the China-Pakistan Economic Corridor (CPEC) and efforts to analyses the opportunities and challenges coming along its way. The project was launched after Chinese President's visit to Pakistan in 2015 and project has so far reaped significant economic, external and strategic benefits for Pakistan, but with the CPEC faces a number of challenges. In this research highlights the significance of the CPEC in China's Belt and Road Initiative (BRI) and focuses on the strategic significance of Gwadar port as well as the significance of the CPEC for both China-Pakistan that is providing the shortest connectivity to China with Central Asia, West Asia, South Asia and onward to Europe. Moreover, it emphasizes on how distance will be abbreviated for oil transportation from Gulf to China and will empower Beijing to assume a significant part as a significant import and export corridor in the world. It additionally resolves the inquiry concerning the import/export imbalance among China and Pakistan and features that the CPEC will also boost the bilateral trade. In this research has utilized a descriptive, comparative, analytical, and qualitative, approaches, through Graham Allison "Rational Actor Model". Secondary data has been analyzed through document analysis. Pakistan has to face both challenges and benefits at the same time. Furthermore, the study examines various segments of the development and prospects of the CPEC and questions the threat perception for the game changer project the CPEC and how Pakistan can handle these threats. The role of international players to destabilize CPEC is also the burning question of the time that needs to be dealt with iron hands.
