Abstract In the realm of disease vectors and agricultural pest management, insecticides play a crucial role in preserving global health and ensuring food security. The pervasive use, particularly of organophosphates (OPs), has given rise to a substantial challenge in the form of insecticide resistance. Carboxylesterases emerge as key contributors to OP resistance, owing to their ability to sequester or hydrolyze these chemicals. Consequently, carboxylesterase enzymes become attractive targets for the development of novel insecticides. Inhibiting these enzymes holds the potential to restore the efficacy of OPs against which resistance has developed. This study aimed to screen the FooDB library to identify potent inhibitory compounds targeting carboxylesterase, Ha006a from the agricultural pest Helicoverpa armigera. The ultimate objective is to develop effective interventions for pest control. The compounds with the highest scores underwent evaluation through docking studies and pharmacophore analysis. Among them, four phytochemicals—donepezil, protopine, 3′,4′,5,7-tetramethoxyflavone, and piperine—demonstrated favorable binding affinity. The Ha006a-ligand complexes were subsequently validated through molecular dynamics simulations. Biochemical analysis, encompassing determination of IC50 values, complemented by analysis of thermostability through Differential Scanning Calorimetry and interaction kinetics through Isothermal Titration Calorimetry was conducted. This study comprehensively characterizes Ha006a-ligand complexes through bioinformatics, biochemical, and biophysical methods. This investigation highlights 3′,4′,5,7-tetramethoxyflavone as the most effective inhibitor, suggesting its potential for synergistic testing with OPs. Consequently, these inhibitors offer a promising solution to OP resistance and address environmental concerns associated with excessive insecticide usage, enabling a significant reduction in their overuse.

Abstract Electrospinning stands out as a flexible and viable method, presenting designed nanoscale materials with customized properties. This research demonstrates the immobilization of carboxylesterase protein Ha006a, reported for its adequacy in pesticide bioremediation by utilizing the electrospinning strategy. This strategy was utilized to create nanofibers by incorporating variable mixtures of biodegradable and cost-effective polyvinyl alcohol (PVA)-chitosan (CS) nanofiber solution (PVA100, PVA96, PVA94, PVA92 and PVA90). All the mixtures were electrospun at a reliable voltage of 21 kV, maintaining a gap of 12 cm from the nozzle. The Ha006a, sourced from Helicoverpa armigera, was consolidated into the optimized PVA90 polymer mixture. The electrospun nanofibers experienced comprehensive characterization utilizing distinctive microscopy and spectroscopy procedures counting FESEM, TGA, XRD and FTIR. The comparative investigation of the esterase property, ideal parameters and stability of the unbound and bound/immobilized Ha006a was scrutinized. The results uncovered an essential elevation in the ideal conditions of enzyme activity post-immobilization. The PVA-CS control nanofiber and Ha006a-PVA-CS showed a smooth structure, including an average breadth of around 170.5 ± 44.2 and 222.5 ± 66.5 nm, respectively. The enzyme-immobilized nanofibers displayed upgraded stability and comprehensive characterization of the nanofiber, which guaranteed genuineness and reproducibility, contributing to its potential as a potent device for bioremediation applications. This investigation opens the way for the manufacture of pesticide-resistant insect enzyme-based nanofibers, unlocking their potential for assorted applications, counting pesticide remediation and ensuring environmental sustainability.

Abstract This study focused on strategically employing the carboxylesterase enzyme Ha006a, derived from the pesticide-resistant microorganism Helicoverpa armigera, to detect atrazine. A comprehensive analysis through biochemical, biophysical and bioinformatics approaches was conducted to determine the interaction between the Ha006a protein and the herbicide atrazine. These experimental findings elucidated the potential of leveraging the inherent pesticide sequestration mechanism of the Ha006a enzyme for sensor fabrication. Numerous optimizations were undertaken to ensure the precision, reproducibility and convenient storage of the resulting electrochemical sensor, Ha006a/MCPE. This biosensor exhibited exceptional performance in detecting atrazine, demonstrating outstanding selectivity with a lower limit of detection of 5.4 µM. The developed biosensor has emerged as a reliable and cost-effective green tool for the detection of atrazine from diverse environmental samples. The Ha006a-based biosensor fabrication has expanded the possibilities for the efficient integration of insect enzymes as analytical tools, paving the way for the design of cost-effective biosensors capable of detecting and quantifying pesticides.

Short-duration and early-bulking potato varieties are well-suited for commercial cultivation in the subtropical Indo-Gangetic plains of India. To maximize seed tuber yield, prevent late-season exposure to aphid vectors, and facilitate the timely planting of wheat crops during the rabi season, it is essential to cut the haulms (vines) of seed potatoes at the earliest possible time. A study was conducted to standardize the optimal vine-killing date for two popular potato varieties in the north-western plains of India, Kufri Pukhraj and Kufri Jyoti, by examining variations in seed yield across different vine-killing dates and assessing the incidence of aphid vectors transmitting potato viruses. Tubers were planted on 15 October for two consecutive seasons, and haulms were cut at 70, 80, and 90 days after planting (DAP), with continuous monitoring of aphid populations. Results showed that total tuber yield and tuber numbers increased with delayed vine-killing. Similarly, the yield and number of oversized tubers significantly rose as the vine-killing date was extended. However, seed-size and undersized tuber yields were largely unaffected, while the number of undersized tubers decreased between 70 and 90 DAP. Further, the gain in total tuber yield and yield of over-sized tubers occurred mainly during 70–80 DAP for Kufri Pukhraj in contrast to Kufri Jyoti where maximum increase occurred between 80 and 90 DAP. Therefore, it is concluded that the optimum time of vine killing for maximum seed tuber yield is directly linked to the tuber growth cycle of individual variety. The period of maximum incidence of aphid vectors varied between the two years. Based on the incidence of Myzus persicae, the seed crops in north-western plains of India should not be extended beyond last week of December although real time monitoring of the aphid incidence is recommended.

Coronavirus disease 2019–related transplant hepatic pseudoaneurysms have not been reported but can be life-threatening. They can be either solitary or multiple and can grow rapidly within weeks. They should be classified as mycotic and treated on an emergent basis. Both stenting of the vessel and coil embolization can potentially be viable treatment options of coronavirus disease 2019–related pseudoaneurysms.

We present a case of 70-year-old male who had a very difficult to treat pancreatico-cutaneous fistula following surgery which we treated with combination of silver nitrate, betadine iodine and glue. Silver nitrate and betadine acted in combination to prevent infection while also causing sclerosis of the cavity and the fistula along with the glue.

The tick-borne intra-cellular parasite, Theileria annulata, causes mortality in bovines. Schizont of the parasite in lymphocytes divides continuously stimulating the clonal expansion of lymphoid cells in lymph nodes leading to the neoplasticlike lymphoid cell proliferation and sometimes may also cause lymphoma, especially in chronic conditions. Two cattle, with a history of fever and respiratory distress, were presented to the Large Animal Unit, Teaching Veterinary Clinical Complex, GADVASU. On clinical examination, the animals had a high fever (106-107ºF), anemia, icteric mucus membranes, and prescapular lymph node enlargement. Whole blood and fine needle aspirate from the lymph node were collected. A blood smear examination revealed anemia, leucopenia with relative lymphocytosis, and thrombocytopenia. Further, there was the presence of schizonts (Koch blue bodies) in the lymphocytes and piroplasms of T. annulata in the red blood cells. Lymph node aspirate examination revealed the feature that mimics lymphoma by the presence of a monomorphic population of lymphoblasts and with all stages of mitotic figures along with lymphoglandular bodies. Numerous schizonts were also recorded in the lymphoblasts during the examination of lymph node aspirate.

The present field experiment was conducted during winter (rabi) seasons of 2018–19 and 2019–20 at ICAR-Central Potato Research Institute, Regional Station Jalandhar, Punjab to find out the suitable planting geometry and dehaulming date for enhancing the seed potato (Solanum tuberosum L.) yield and profitability under north-western plains of India. According to the study planting geometry and dehaulming date strongly influenced the seed size tuber yield and quality. Among the different planting geometries, paired row bed planting significantly increased the yield and number of seed size tubers when dehaulming was done after 80–90 days. Maximum gross income, net returns and benefit:cost ratio was also observed under paired row bed planting over the conventional planting geometry. Paired row bed planting increased the tuber yield by 21.7% and benefit:cost ratio by 18.6% over the control. Hence, it is concluded from the experiment that paired row bed planting is found suitable for maximizing the seed size tuber numbers, yield as well as getting the maximum economic returns.

Introduction: This study investigated the role of diallyl disulfide (DADS) against glycerol-induced nephrotoxicity in rats. Moreover, the role of peroxisome proliferator activated receptor-γ (PPAR-γ) in DADS-mediated renoprotection has been explored. Materials and Methods: Male Wistar albino rats were challenged with glycerol (50% w/v, 8 mL/kg intramuscular) to induce nephrotoxicity. Kidney injury was quantified by measuring serum creatinine, creatinine clearance, urea, potassium, fractional excretion of sodium, and microproteinuria in rats. Renal oxidative stress was measured in terms of thiobarbituric acid reactive substances, superoxide anion generation, and reduced glutathione levels. Hematoxylin–eosin (H&E) and periodic acid Schiff staining of renal samples was done to show histological changes. Glycerol-induced muscle damage was quantified by assaying creatine kinase (CK) levels in rat serum. Results: Administration of glycerol resulted in muscle damage as reflected by significant rise in CK levels in rats. Glycerol intoxication led kidney damage was reflected by significant change in renal biochemical parameters, renal oxidative stress and histological changes in rat kidneys. Administration of DADS attenuated glycerol-induced renal damage. Notably, pretreatment with bisphenol A diglycidyl ether, a PPAR-γ antagonist, abolished DADS renoprotection in rats. Conclusion: We conclude that DADS affords protection against glycerol-induced renal damage in rats. Moreover, PPAR-γ plays a key role in DADS-mediated renoprotective effect.

The allelic variations in a diversity panel of 353 potato accessions, including 256 accessions belonging to Solanum tuberosum sub spp. tuberosum, 49 accessions belonging to Solanum tuberosum sub spp. andigena, and 48 Indian potato varieties were analysed using 25 simple sequence repeat (SSR) markers. The SSR allelic profiles revealed high levels of polymorphism and distinctness among the accessions studied. A total of 343 alleles of 25 SSR markers were observed in the diversity panel of 353 highly diverse tetraploid potato accessions. The number of alleles produced per SSR varied from 8 for the marker STM1053 to 25 for the marker STIKA. The polymorphic information content (PIC) ranged from 0.66 (STG0010) to 0.93 (STM1106) with an average of 0.82. The cluster analysis using the SSR allelic profiles of 353 accessions divided the population into five major groups. The association mapping for late blight resistance identified six markers with the general linear model (GLM), and out of these six markers significance of three markers was reconfirmed with the mixed linear model (MLM). The findings of this study suggest that SSRs are the appropriate markers for evaluating genetic diversity and population structure within different potato germplasm collections. A significant diversity across the tetraploid potato accessions was observed. Moreover, the markers identified in this study could be useful in marker-assisted selection (MAS) breeding in potato for late blight resistance (LBR).

Huanglongbing (HLB, aka citrus greening), one of the most devastating diseases of citrus, has wreaked havoc on the global citrus industry in recent decades. The culprit behind such a gloomy scenario is the phloem-limited bacteria “Candidatus Liberibacter asiaticus” (CLas), which are transmitted via psyllid. To date, there are no effective long-termcommercialized control measures for HLB, making it increasingly difficult to prevent the disease spread. To combat HLB effectively, introduction of multipronged management strategies towards controlling CLas population within the phloem system is deemed necessary. This article presents a comprehensive review of up-to-date scientific information about HLB, including currently available management practices and unprecedented challenges associated with the disease control. Additionally, a triangular disease management approach has been introduced targeting pathogen, host, and vector. Pathogen-targeting approaches include (i) inhibition of important proteins of CLas, (ii) use of the most efficient antimicrobial or immunity-inducing compounds to suppress the growth of CLas, and (iii) use of tools to suppress or kill the CLas. Approaches for targeting the host include (i) improvement of the host immune system, (ii) effective use of transgenic variety to build the host’s resistance against CLas, and (iii) induction of systemic acquired resistance. Strategies for targeting the vector include (i) chemical and biological control and (ii) eradication of HLB-affected trees. Finally, a hypothetical model for integrated disease management has been discussed to mitigate the HLB pandemic.

Here, we have demonstrated that on modification of linear polyethylenimine (lPEI, LP) with amphiphilic 3-bromopropyltetramethylguanidinium (PTMG) linker, the transfection efficiency exhibited by the modified polymers decreased while cell viability improved. A series of LP-PTMG polymers was synthesized by the reaction of varying amounts of 3-bromopropyl tetramethylguanidinium linker with lPEI (25 kDa). These modified polymers interacted efficiently with pDNA and formed nanosized complexes as shown by dynamic light scattering analysis. The size of the complexes in the series LP-PTMG/pDNA was observed in the range of ∼178–205 nm. The interaction of modified polymers with plasmid DNA was stronger than linear PEI as evidenced by heparin release assay which showed ∼83% pDNA release from LP-PTMG-3/pDNA complexes in comparison to ∼95% in lPEI/pDNA complexes on treatment with same amount of heparin suggesting the formation of self-assembled structures in modified polymers. The transfection studies in HeLa and Chinese hamster ovary cells showed a decrease in transfection efficiency of LP-PTMG polymers, the reason for this may be strong binding of modified polymers with pDNA due to accumulation of charge on the surface. This finding showed the significance of optimum binding of polymer and DNA to form polyplexes as well as release of DNA from the polyplexes.

Potato cyst nematode (Globodera spp.) is one of the serious pests of potato worldwide including India. Infested seed tubers and soil are the primary means of spread of cysts to non-infested areas, whereas air, water, human, animals, field equipments, composts, gunny bags are other means of spread. Therefore, a study was carried out with different concentrations of NaOCl under in vitro and in vivo condition to see its decontamination effect on potato cyst nematode (PCN). The results revealed that potato tubers harvested from the PCN infested fields can be completely decontaminated by giving dip treatment in NaOCl (2%) solution for 30 min. and the treated tubers sprouts normally after two months of storage.

Self-assembly is the process of association of individual units of a material into highly arranged/ordered structures/patterns. It imparts unique properties to both inorganic and organic structures, so generated, via non-covalent interactions. Currently, self-assembled nanomaterials are finding a wide variety of applications in the area of nanotechnology, imaging techniques, biosensors, biomedical sciences, etc., due to its simplicity, spontaneity, scalability, versatility, and inexpensiveness. Self-assembly of amphiphiles into nanostructures (micelles, vesicles, and hydrogels) happens due to various physical interactions. Recent advancements in the area of drug delivery have opened up newer avenues to develop novel drug delivery systems (DDSs) and self-assembled nanostructures have shown their tremendous potential to be used as facile and efficient materials for this purpose. The main objective of the projected review is to provide readers a concise and straightforward knowledge of basic concepts of supramolecular self-assembly process and how these highly functionalized and efficient nanomaterials can be useful in biomedical applications. Approaches for the self-assembly have been discussed for the fabrication of nanostructures. Advantages and limitations of these systems along with the parameters that are to be taken into consideration while designing a therapeutic delivery vehicle have also been outlined. In this review, various macro- and small-molecule-based systems have been elaborated. Besides, a section on DNA nanostructures as intelligent materials for future applications is also included.

Abstract Auxin levels are tightly regulated within the plant cell, and its storage in the isolated cavity of proteins is a measure adopted by cells to maintain the availability of auxin. We report the first crystal structure of Wrightia tinctoria 11S globulin (WTG) in complex with Indole-3-acetic acid (IAA), an auxin, at 1.7 Å resolution. WTG hexamers assemble as a result of the stacking interaction between the hydrophobic surfaces of two trimers, leaving space for the binding of charged ligands. The bound auxin is stabilized by non-covalent interactions, contributed by four chains in each cavity. The presence of bound ligand was confirmed by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) and high-resolution mass spectrometry (HRMS). Here, we hypothesize that the cleavage of globulins by endopeptidases leads to the movement of the hydrophilic loop region from the surface to the periphery, leaving space for the binding of auxin, and promotes hexamer formation. As the process of germination proceeds, there is a change in the pH, which induces the dissociation of the hexamer and the release of auxin. The compact hexameric assembly ensures the long-term, stable storage of the hormone. This suggests a role for globulin as a novel player in auxin homeostasis.

Portal vein thrombosis is common in cirrhotic patients and results in increased morbidity and mortality. Transjugular intrahepatic portosystemic shunt (TIPS) creation is a well-established therapy for refractory variceal bleeding and refractory ascites in patients who do not tolerate repeated large volume paracentesis. Experience and technical improvements have led to improved TIPS outcomes that have encouraged an expanded application. Complete portal vein thrombosis has come a long way from being a contraindication to an indication for TIPS procedure. As experience and technology have evolved, the ultrasound guidance transvenous access of portal vein from the hepatic vein help in overall higher success rate of performing the TIPS procedure and reducing the procedure-related complications.

The development of technology and limitation of numerous ventricular assist devices has increased usage of the total artificial heart (TAH) for mechanical circulatory support. A primary important feature of TAH is that it is one of the few mechanical circulatory support devices that provides biventricular support. As the number of patients with TAH rises, spreading awareness about the device in the interventional radiology community is important.

Huanglongbing (HLB) or citrus greening is highly destructive disease that is affecting the citrus industry worldwide and it has killed millions of citrus plants globally. HLB is caused by the phloem limited, Gram negative, non-culturable, alpha-proteobacterium, 'Candidatus Liberibacter asiaticus'. Currently, polymerase chain reaction (PCR) and real time PCR have been the gold standard techniques used for detection of 'Ca. L. asiaticus'. These diagnostic methods are expensive, require well equipped laboratories, not user-friendly and not suitable for on-site detection of the pathogen. In this study, a sensitive, reliable, quick and low cost recombinase polymerase based isothermal amplification combined with lateral flow assay (HLB-RPA-LFA) technique has been developed as a diagnostic tool for detection of 'Ca. L. asiaticus'. The assay was standardized by designing the specific primer pair and probe based on the conserved 16S rRNA gene of 'Ca. L. asiaticus'. The assay was optimized for temperature and reaction time by using purified DNA and crude plant extracts and the best HLB-RPA-LFA was achieved at the isothermal temperature of 38°C for 20 to 30 min. The efficacy and sensitivity of the assay was carried out by using field grown, HLB-infected, HLB-doubtful and healthy citrus cultivars including mandarin, sweet orange cv. mosambi, and acid lime. The HLB-RPA-LFA did not show cross-reactivity with other citrus pathogens and is simple, cost-effective, rapid, user-friendly and sensitive. Thus, the HLB-RPA-LFA method has great potential to provide an improved diagnostic tool for detection of 'Ca. L. asiaticus' for the farmers, nurserymen, disease surveyors, mobile plant pathology laboratories, bud-wood certification and quarantine programs.

Huanglongbing (HLB, also known as citrus greening) is considered to be the most devastating disease that has significantly damaged the citrus industry globally. HLB is caused by the Candidatus Liberibacter asiaticus (CLas), the fastidious phloem-restricted gram-negative bacterium, vectored by the asian citrus psyllid. To date, there is no effective control available against CLas. To alleviate the effects of HLB on the industry and protect citrus farmers, there is an urgent need to identify or develop inhibitor molecules to suppress or eradicate CLas from infected citrus plant. In this paper, we demonstrate for the first time an in planta efficacy of two antimicrobial compounds against CLas viz. 2S albumin (a plant based protein; ~12.5 kDa), Nano-Zinc Oxide (Nano-ZnO; ~ 4.0 nm diameter) and their combinations. Aqueous formulations of these compounds were trunk-injected to HLB affected Mosambi plants (Citrus sinensis) grafted on 3-year old rough lemon (C. jambhiri) rootstock with known CLas titer maintained inside an insect-free screen house. The effective concentration of 2S albumin (330 ppm) coupled with the Nano-ZnO (330 ppm) at 1:1 ratio was used. The dynamics of CLas pathogen load of treated Mosambi plants was assessed using TaqMan-qPCR assay every 30 days after treatment (DAT) and monitored till 120 days. We observed that 2S albumin-Nano-ZnO formulation performed the best among all the treatments decreasing CLas population by 96.2%, 97.6%, 95.6%, and 97% of the initial bacterial load (per 12.5 ng of genomic DNA) at 30, 60, 90, and 120 DAT, respectively. Our studies demonstrated the potency of 2S albumin-Nano-ZnO formulation as an antimicrobial treatment for suppressing CLas in planta and could potentially be developed as a novel anti CLas therapeutics to mitigate the HLB severity affecting the citrus industry worldwide.

Introduction: Macro vascular disease is a major cause of death in diabetic individuals. Because many diabetic individuals have multiple risk factors for atherosclerosis, the relative risks of Ischemic Heart Disease (IHD) and Cerebro-Vascular Disease (CVD) are 2-4 fold and 2-3 fold higher respectively, than the risk in non-diabetic subjects. Several studies indicate that postprandial metabolic derangements, most notably hyperglycemia and hypertriglyceridemia are important cardiovascular risk factors since they induce oxidative stress and endothelial dysfunction. Measurement of Carotid Intima Media Thickness (CIMT) by ultrasonography is a non-invasive and quantitative method of evaluating early atherosclerotic changes in the vasculature. It has been shown that an increase in CIMT is associated with an increased risk of IHD or CVD. Aim: The present study was undertaken to find the correlation of postprandial hypertriglyceridemia with carotid intima media thickness in North Indian patients with type II Diabetes Mellitus. Materials and Methods: The present study was done amongst 50 North Indian patients, between the age group of 35-75 years suffering from Type II Diabetes Mellitus for less than 1 year duration. All routine investigations and special investigations like Lipid Profile (Fasting)-total Cholesterol, HDL, LDL, Triglycerides, VLDL, Lipid profile {Postprandial (4 hours after a standard meal)} - Triglycerides, B- mode ultrasonography of common carotid artery were done in all the patients. An attempt was made to find the correlation between postprandial hypertriglyceridemia with carotid intima media thickness. Results: Both PPTG and FTG showed a strong correlation with CIMT, the correlation coefficients being 0.879 and 0.764 respectively. If the subjects with normal FTG were taken to calculate correlation between PPTG and CIMT, correlation coefficient was 0.848 and if only the subjects with raised FTG were taken then the correlation coefficient was 0.735; suggesting a stronger correlation of PPTG with CIMT at normal fasting triglyceride levels. Conclusion: Both, increased postprandial triglyceride levels and fasting triglyceride levels are risk factors for increasing carotid intima media thickness in patients with Type II Diabetes Mellitus.
