The Alice Gold and Silver Mining Company was a mining operation in Walkerville, Montana. Its headquarters were in Salt Lake City, Utah. The records consist primarily of letterpress volumes of outgoing correspondence from Walkerville mine superintendents to Salt Lake City company officials. Also included are organizational materials, financial records, productions records, and legal files., This collection is composed primarily of interoffice correspondence and financial records. The former includes letters between Joseph R. Walker and Matthew H. Walker at the Salt Lake City company offices and the company superintendents at the mine in Walkerville. These letters discuss daily operations, national mining issues, equipment used, labor relations, assay results, dealings with Butte business interests, etc. (1877-1940). In addition, the collection contains court papers, legal documents, and organizational materials. (Manuscript Collection 57).

International audience; The principles behind the touchstone have never been investigated in a scientific manner, even though there are many technical and historical accounts of its effectiveness. In this article I investigate whether and to what extent the content of Au-Ag-Cu ternary alloys can be visually ascertained with a touchstone without acid. I show that it is very likely to be true as the touchstone solves several problems (illumination, surface state and curvature, etc.) and makes it possible to carry out a luminance comparison. A colorimetric approach confirms the high level of accuracy attained.; Même si les études techniques et historiques abondent, les principes de la pierre de touche n’ont jamais été abordés de façon scientifique. Cet article étudie si et dans quelle mesure la composition des alliages ternaires Au-Ag-Cu peut être visuellement déterminée à la pierre de touche uniquement et sans l’aide d’acides forts. Il apparaît que la pierre de touche permet de surmonter les obstacles à une comparaison de la luminance des alliages (état et géométrie de la surface, éclairage…). Green and sensitive spectrofluorometric methods have been developed and validated for the determination of timolol maleate (TML)/hydrochlorothiazide (HCT) and amiloride hydrochloride (AMH)/hydrochlorothiazide in tablets. The proposed spectrofluorometric procedures were found to be linear in the range of 4–12, 5–35 and 0.025–0.2 mg L−1 for HCT, TML and AMH, resp. The excitation and emission wavelengths for HCT, TML and AMH at room temperature were 270 and 375, 295 and 435, 330 and 415 nm, resp. The methods were validated with respect to ICH guidelines. The AMH showed higher sensitivity with lower values of LOD and LOQ values compared to HCT and TML. The proposed methods were applied to two pharmaceutical formulations; the method for HCT and AMH has proven as reliable assaying method, whereas the method for TML, when combined with HCT, is applicable to screening semi-quantitative analyses.

Chagas disease is a chronic infection caused by the parasite Trypanosoma cruzi (T. cruzi). The development of the parasite in the human heart muscle or digestive tract causes severe damage, resulting in organ failure. The current treatments, benznidazole and nifurtimox, have a limited efficacy, show significant side effects, and increasing resistance. Post-translational modifications (PTMs) play a crucial role in the life cycle of protozoan parasites. Bromodomains (BDs) are readers of lysine acetylation, one of the PTMs found on histone tails. The function of BDs remains uncharacterised in T. cruzi. The life cycle of the parasite is complex and is thought to rely heavily on epigenetic control and therefore our main interest is to validate the T. cruzi BDs as therapeutic targets. Optimisation of expression and purification protocols of the protein BD-containing factors (BDFs) in E. coli (Chapter 1) was followed by the development of a waterLOGSY- based assay to screen acetyl-lysine mimic containing compounds (Figure 0.1). The binding of the hits was further characterised by 1H NMR ligand-observed protein titration to obtain KD. Excitingly, the human BD and kinase ligand, BI-2536 (8), was shown to bind to TcBDF3 with a KD value between 25 and 55 μM. The binding was confirmed to be 14.6±3.0 μM (n=3) using ITC. BI-2536 (8) is to date the highest affinity ligand identified and was chosen as a lead structure for further development (Chapter 2). Based on this initial work, eight functional probes were designed and synthesised to enable the development of higher-throughput assays for T. cruzi BDs (Chapter 3). The 19F NMR reporter molecules, photo-activatable diazirines, and fluorescent probes were assessed against TcBDF3 to establish novel assays (Chapter 4). Firstly, the fluorinated probes were used as 19F NMR reporter molecules to observe their binding and displacement to and from TcBDF3. Secondly, a protein LCMS assay was established to characterise binding and subsequent cross-linking of the diazirine probes. Finally, fluorescent probes were validated by fluorescence polarisation. Satisfactorily, one probe binds to TcBDF3, with a KD value of 3 μM. Displacement of the fluorescent probe by the parent compound BI-2536 (8) results in a dose-dependent response, however, a full curve and an IC50 value could not be obtained due to assay interference at high ligand concentrations. Further validation is required, using higher affinity ligands to show that the fluorescence polarisation displacement can generate IC50 values. With GSK collaborators, 556 diazirines of the GSK PhaBits library were screened against TcBDF3 and 5-1, identifying one and six new binding fragments, respectively. Additionally, a DNA-encoded library of ca. 1.9 trillion encodings were screened against TcBDF2 and 5- 1, giving 82 and 134 hit clusters, respectively, to be investigated for binding. The low hit rates for both screening campaigns highlights that the T. cruzi BDs are challenging targets. In this work we report the first production of soluble T. cruzi BD proteins and their study using NMR assays such as waterLOGSY and NMR titration. Critically, this initial work led to the development of functional probes to provide a tool box to study TcBDF3 at higher throughput with diverse and orthogonal assays. The novel biophysical assays established here can be used to screen efficiently for binding compounds and opens up the possibility of rapid SAR. Identified ligands will enable validation of the bromodomains of T. cruzi as targets for Chagas disease for the first time. This will have a wide range of implications in the use of epigenetics targets for treatments of parasites more broadly.

Contrastaria, an independent business unit belonging to Imprensa Nacional Casa da Moeda, is responsible for certifying and monitoring precious metal items with in the Portuguese Jewellery and Watch Industry. In Portugal, with some exceptions, all precious metals artefacts are required to be hall marked before being introduced in the market, guaranteeing the product's authenticity and securing the market's credibility. The Consulting Project addresses the challenge of "Fostering the Jewellery and Watch Sector through the Lens of the Portuguese Assay Office with a Shared Value Approach ".The Thesis was carried out using a problem-solving driven approach, where the Team sought to validate several before hand-created hypotheses. The validation process consisted of an in-depth internal and external analysis, based on primary–interviews and surveys–and secondary data–papers, reports, and frameworks. Accordingly, meaningful in sights were ga thered, guiding the formulation of impact ful recommendations that will increase the awareness of the Portuguese population regarding Contrastaria's Mission and Hallmarks, generate internal processes' efficiency, and protect online consumers of precious metal artefacts. Consequently, in a more holistic view, the Portuguese Assay Office's recognition will be enhanced, customer satisfaction improved, online demand and supply for precious metals jewellery strengthened, and the online Imitation Jewellery Market weakened. There fore, successfully solving the challenge proposed by Contrastaria, adding value to the institution and placing it in the industry's core. The individual work developed focuses on the Jewellery and Watch Industry analysis, by the comprehension of the (1) Precious Metals Value Chain and the (2) STEEP framework. The (1) Precious Metals Value Chain represents every stage a precious metal artefact goes through, allowing a deeper understanding of each industry player and the irrespective pain points and strengths. The (2) STEEP framework, on the other hand, evaluates all the external forces that impact Contrastaria’s operations, high lighting potential market opportunities oreventhreats with in the industry. Accordingly, the Jewellery and Watch Sector has been strongly affected by the pandemic crisis. How ever, almost every player is recovering and re-establishing pre-pandemic levels. The industry’s current trends rely on the adoption of sustainable processes and techniques, the Imitation Jewellery Market’s growth, and the continuous increase of online demand. All things considered, the Team’s recommendations will tackle or exploit such tendencies.

[Resumen] La investigación en fútbol femenino ha aumentado de manera notable en los últimos años. A pesar de ello, en el ámbito del rendimiento táctico colectivo las publicaciones aún son escasas. El objetivo de este estudio fue analizar y describir cómo se produjeron las acciones ofensivas dinámicas en las selecciones ganadoras y perdedoras en la Copa Mundial Femenina de la FIFA Francia 2019 y establecer diferencias entre ambos grupos. Desde una metodología observacional, se elaboró un instrumento de observación ad hoc y se analizaron 1883 acciones ofensivas que tuvieron lugar en la fase final del campeonato. Se realizaron dos tipos de pruebas estadísticas para comprobar la existencia de diferencias entre grupos. Se aplicó la prueba chicuadrado para los criterios de tipo cualitativo y la prueba U de Mann Whitney para los criterios de tipo continuo con un nivel de significación p < .05. Se demostró la existencia de diferencias significativas para los siguientes criterios: temporalidad, resultado temporal, tiempo total de posesión, tiempo de posesión en campo rival, organización defensiva rival, zona de inicio en amplitud y resultado de la acción. Las selecciones que ganaron los partidos mostraron una mayor capacidad de desarrollar sus ataques en los primeros momentos de la primera y la segunda parte, así como una mayor calidad colectiva para mantener la posesión de balón en campo rival y finalizar sus acciones con éxito. Estos resultados pueden ser utilizados con el objetivo de implementar estrategias de entrenamiento y competición que permitan aumentar el rendimiento en fútbol femenino de élite. [Abstract] Research into women's football has increased dramatically in recent years. Despite this, in the field of collective tactical performance, publications are still scarce. The objective of this study was to analyse and describe how dynamic offensive actions came about in the winning and losing teams in the 2019 FIFA Women's World Cup in France and to establish differences between the two groups. From an observational methodology, an ad hoc observation instrument was developed and 1,883 offensive actions that took place in the final phase of the championship were analysed. Two types of statistical tests were carried out to verify the existence of differences between groups. The chi-square test was applied for the qualitative type criteria and the Mann Whitney U test for the continuous type criteria with a significance level of p < .05. The existence of significant differences was demonstrated for the following criteria: temporality, match status, total time in possession, time in possession in opponent’s half, opponent’s defensive organisation, wide start area and result of the action. The teams that won the matches showed a greater ability to develop their attacks in the first moments of the first and second halves, as well as a greater collective quality to maintain possession of the ball in the opponent's half and to finish their actions successfully. These results can be used with the aim of implementing training and competition strategies to increase performance in elite women's football. En el presente trabajo ahondaremos sobre la estructura organizativa de la Lliga Reumatologica Catalana una asociación de pacientes que busca agrupar a aquellos pacientes que padezcan alguna patología reumática, actualmente en el ámbito catalán. Nos centramos en observar las fortalezas y debilidades de esta organización en el panorama organizativo actual y como podría mejorar su posición para ser capaz de cumplir la misión de la organización de forma satisfactoria.

En este trabajo se caracterizó química y mineralógicamente el material de alimentación, concentrado y las colas de la planta de procesamiento de Llallagua-Catavi para valorar la posibilidad de un mejor aprovechamiento del estalo de estos recursos al igual que algún otro subproducto que pudiera presentarse con el fin de mejorar la producción de la explotación. Las técnicas utilizadas han sido fundamentalmente el análisis químico y caracterización mineralógica mediante difracción de rayos X, microscopía electrónica de barrido y mineralogía cuantitativa automatizada (MLA). El material que entra a la planta son principalmente las colas de un procesamiento anterior y se divide en arenas o lamas finas. También se incluyen los desmontes, que es el material estéril extraído durante la explotación para llegar a la mineralización. In this work, the feed material, concentrate and the tailings of the Llallagua-Catavi processing plant were chemically and mineralogically characterized to assess the possibility of a better recovery of tin, as well as any other by-product that could contain in order to improve the production of the exploitation. The techniques used have been chemical analysis and mineralogical characterization by X-ray diffraction, scanning electron microscopy and automated quantitative mineralogy (MLA).

Premi Emili Viader 2021 La mina de San Pablo, que se encuentra a 30km al sureste de Oruro (Bolivia), es una explotación de minería artesanal donde su producción principal se centra en la casiterita como mena de estaño para luego venderlo a plantas de procesamiento en Oruro. El objetivo de este trabajo es estudiar la mineralogía y la química mineral del depósito de San Pablo, incluido en el distrito minero de Santa Fe. Los objetivos específicos son la determinación de la mineralogía y la química-mineral, buscar presencia de Indio, y estudiar los isotopos de azufre el origen del depósito mineral. The San Pablo mine, located 30km south-east of Oruro (Bolivia), consists in an artisanal exploitation where the main production is cassiterite as tin ore to later on sell it to processing plants in Oruro. The objective of this work is to study the mineralogy and mineral chemistry of the San Pablo deposit, included in the Santa Fe mining district. To this end the specific objectives were to determine the mineralogy and chemical composition of the ore minerals, to search for the presence of Indium, and to do sulphur isotopes to contribute to the determination of the origin of the deposit. Objectives To characterize the clonal complexity in Mycobacterium tuberculosis (MTB) infections considering factors that help maximize the detection of coexisting strains/variants. Methods Genotypic analysis by Mycobacterial Interspersed Repetitive-Unit–Variable-Number Tandem-Repeats (MIRU-VNTR) was performed directly on 70 biopsy specimens from two or more different tissues involving 28 tuberculosis cases diagnosed post-mortem in Mozambique, a country with a high tuberculosis burden. Results Genotypic data from isolates collected from two or more tissues were obtained for 23 of the 28 cases (82.1%), allowing the analysis of within-patient diversity. MIRU-VNTR analysis revealed clonal diversity in ten cases (35.7%). Five cases showed allelic differences in three or more loci, suggesting mixed infection with two different strains. In half of the cases showing within-host diversity, one of the specimens associated with clonal heterogeneity was brain tissue. Conclusions Direct MTB genotyping from post-mortem tissue samples revealed a frequent within-host Mycobacterium tuberculosis diversity, including mixed and polyclonal infections. Most of this diversity would have been overlooked if only standard analysis of respiratory specimens had been performed.

Serotonin (5-HT) is a hormone and neurotransmitter that modulates neural activity as well as a wide range of other physiological processes including cardiovascular function, bowel motility, and platelet aggregation. 5-HT synthesis is catalyzed by tryptophan hydroxylase (TPH) which exists as two distinct isoforms; TPH1 and TPH2, which are responsible for peripheral and central 5-HT, respectively. Due to the implication of 5-HT in a number of pathologies, including depression, anxiety, autism, sexual dysfunction, irritable bowel syndrome, inflammatory bowel disease, and carcinoid syndrome, there has been a growing interest in finding modulators of these enzymes in recent years. We thus performed high-throughput screening (HTS) using a fluorescence-based thermal shift assay (DSF) to search the Prestwick Chemical Library containing 1,280 compounds, mostly FDA-approved drugs, for TPH1 binders. We here report the identification of omeprazole, a proton pump inhibitor, as an inhibitor of TPH1 and TPH2 with low micromolar potency and high selectivity over the other aromatic amino acid hydroxylases. The S-enantiomer of omeprazole, esomeprazole, has recently also been described as an inhibitor of monoamine oxidase-A (MAO-A), the main enzyme responsible for 5-HT degradation, albeit with lower potency compared to the effect on TPH1 and TPH2. In order to investigate the net effect of simultaneous inhibition of TPH and MAO-A in vivo, we administered high-dose (100 mg/kg) omeprazole to CD-1 mice for 4 days, after which the animals were subjected to the tail suspension test. Finally, central (whole brain) and peripheral (serum) 5-HT content was measured using liquid chromatography-mass spectrometry (LC-MS). Omeprazole treatment significantly increased 5-HT concentrations, both in brain and in serum, and reduced the time spent immobile in the tail suspension test relative to vehicle control. Thus, the MAO-A inhibition afforded by high-dose omeprazole appears to overcome the opposing effect on 5-HT produced by inhibition of TPH1 and TPH2. Further modification of proton pump inhibitor scaffolds may yield more selective modulators of 5-HT metabolism.

Objective: to analyze the student’s progression in the acquisition of specific and transversal competences in relation to the competence dimensions. Method: the cross-sectional descriptive study was carried out in the clinical practice subjects included in the Nursing Degree. We included 323 students and we contemplated the development of competences through an ad-hoc questionnaire with 4 dimensions: delivery and care management, therapeutic communication, professional development and care management. Results: the academic results between the practice of the second and third year showed an improvement in care provision and therapeutic communication skills (Clinical Placements I: 12%-29%; Clinical Placements II: 32%-47%) and worsened in professional development and care management (Clinical Placements I: 44%-38%; Clinical Placements II: 44%-26%). Conclusion: the correlations between these two years were high in all the dimensions analyzed. The evaluation of competence progression in the context of clinical practice in nursing university studies allows us to optimize these practices to the maximum and establish professional profiles with a greater degree of adaptation to the professional future.

We organized 10Kin1day, a pop-up scientific event with the goal to bring together neuroimaging groups from around the world to jointly analyze 10,000+ existing MRI connectivity datasets during a 3-day workshop. In this report, we describe the motivation and principles of 10Kin1day, together with a public release of 8,000+ MRI connectome maps of the human brain. Ongoing grand-scale projects like the European Human Brain Project (1), the US Brain Initiative (2), the Human Connectome Project (3), the Chinese Brainnetome (4) and exciting world-wide neuroimaging collaborations such as ENIGMA (5) herald the new era of big neuroscience. In conjunction with these major undertakings, there is an emerging trend for bottom-up initiatives, starting with small-scale projects built upon existing collaborations and infrastructures. As described by Mainen et al. (6), these initiatives are centralized around self-organized groups of researchers working on the same challenges and sharing interests and specialized expertise. Aim The oncological risk/benefit trade-off for laparoscopy in rectal cancer is controversial. Our aim was to compare laparoscopic vs open surgery for resection of rectal cancer, using unselected data from the public healthcare system of Catalonia (Spain). Methods This was a multicentre retrospective cohort study of all patients who had surgery with curative intent for primary rectal cancer at Catalonian public hospitals from 2011 to 2012. We obtained follow-up data for up to 5 years. To minimize the differences between the two groups, we performed propensity score matching on baseline patient characteristics. We used multivariate Cox proportional hazards regression analyses to assess locoregional relapse at 2 years and death at 2 and 5 years. Results Of 1513 patients with Stage I-III rectal cancer, 933 (61.7%) had laparoscopy (conversion rate 13.2%). After applying our propensity score matching strategy (2:1), 842 laparoscopy patients were matched to 517 open surgery patients. Multivariate Cox analysis of death at 2 years [hazard ratio (HR) 0.65, 95% CI 0.48, 0.87; P = 0.004] and 5 years (HR 0.61, 95% CI 0.5, 0.75; P < 0.001) and of local relapse at 2 years (HR 0.44, 95% CI 0.27, 0.72; P = 0.001) showed laparoscopy to be an independent protective factor compared with open surgery. Conclusions Laparoscopy results in lower locoregional relapse and long-term mortality in rectal cancer in unselected patients with all-risk groups included. Studies using long-term follow-up of cohorts and unselected data can provide information on clinically relevant outcomes to supplement randomized controlled trials.

[eng]What are the relations between public policy analysis and the Law? How can linking these research fields contribute to the development of institutional capacities in the policymaking process? How can a teaching experience emphasize this issue? How can multiple public policy methodologies approach the Law? This paper tries to answer those questions. In Brazil, the formalistic legal logic separates both research fields (Law and Public Policy Analysis), creating obstacles that obstruct better governance in all three government branches. From the methodological point of view, this paper explores the experience of graduate students at the University of Brasilia Law School (FD/UnB), incorporating their professional experience related to the public policy cycle. The case study we explore here is the Law and Public Policy Analysis course ministered in the second half of 2015 and 2016. We present how the discipline was developed and how the students perceived the topic in their research. We connected the literature to the students’ perceptions. It seems that linking legal models to the public policy cycle tend to improve the integration and performance of policy stakeholders and the Lawyers., [spa]¿Cuáles son las relaciones entre el análisis de políticas públicas y el derecho? ¿Cómo puede la vinculación de estos campos de investigación contribuir al desarrollo de las capacidades institucionales en el proceso de formulación de políticas? ¿Cómo puede una experiencia de enseñanza enfatizar este tema? ¿Cómo pueden múltiples metodologías de políticas públicas abordar la ley? Este artículo intenta responder a estas preguntas. En Brasil, la lógica legal formalista separa ambos campos de investigación (Derecho y Análisis de Políticas Públicas), creando obstáculos que obstruyen una mejor gobernabilidad en las tres ramas del gobierno. Desde el punto de vista metodológico, este trabajo explora la experiencia de los estudiantes de postgrado de la Facultad de Derecho de la Universidad de Brasilia (FD/UnB), incorporando su experiencia profesional relacionada con el ciclo de políticas públicas. El estudio que aquí exploramos es el curso de Análisis de Leyes y Políticas Públicas impartido en la segunda mitad de 2015 y 2016. Se presenta cómo se desarrolló la disciplina y cómo los estudiantes percibieron el tema en su investigación. Conectamos la literatura con las percepciones de los estudiantes. L'estudi que aquí explorem és el curs d'Anàlisi de Lleis i Polítiques Públiques im partit a la segona meitat de 2015 i 2016. Es presenta com es va desenvolupar la disciplina i com els estudiants van percebre el tema en la seva recerca. Connectem la literatura amb les percepcions dels estudiants. Pel que sembla, la vinculació dels models jurídics amb el cicle de les polítiques públiques tendeix a millorar la integració i l'acompliment de les parts interessades en les polítiques i dels advocats.

Victory is the ultimate aim in soccer and therefore when a team wins an elite European or world championship, attempts will invariably be made to emulate the winning team’s style of play. In this study, we performed an in‐depth analysis of play by the Spanish soccer team during the 2012 UEFA European Championship, where it was crowned champion. Using observational methodology and T‐pattern analysis, we identified hidden patterns of play that ended in a goal for the Spanish team. A generalizability coefficient (e2) of 0.986 demonstrated that the offensive patterns detected are robust and highly generalizable. These patterns were formed by technical actions consisting of ball control and pass, with alternations between short and long passes, in the central area of the rival pitch, with use of both wings to achieve width of play and prioritization of width over depth of play. We also found patterns showing that goals and shots at goal were made on a ball delivered from the opposite direction to the shot and were not preceded by a technical action.

The modelling of high pressure grinding rolls is described by the population balance model, a mass balance which includes several functions that are related to the mineral characteristics, material kinetics and operative conditions of the device. The breakage distribution function is one of these functions and refers to the way in which the daughter particles are generated by the process of comminution. The piston-die press is presented as a methodology to determine the breakage distribution function of two different materials, from the mechanical response point of view: altered granite and a cal-silicate material. The aim is to determine the relation between the operative conditions and the mineral characteristics in order to explain and predict the breakage function parameters. The materials were characterised using XRD and single compression strength tests. The altered granite is a brittle material, which generates more fines under single compression conditions compared to bed compression conditions, mainly due to the mineral composition and the response of the material to the breakage action. The cal-silicate material shows a normal trend in its breakage behaviour. As is expected, the mineralogical characterisation is a useful tool to predict the values of the parameters of the breakage distribution function.
