Chili pepper (Capsicum frutescens L.) is a common commodity used as spice and pharmaceutical uses around the world. However, chili pepper cultivation failure often occurs due to drought exposure. The inoculation of arbuscular mycorrhizal fungi (AMF), such as Funneliformis mosseae, has the potential to induce defense against drought stress through symbiotic association with plant roots. The aim of this research was to investigate the effects of F. mosseae inoculation on the growth of chili pepper under repeated drought stress. Chili pepper plants were exposed to three drought regimes for two cycles, with one rewatering event between the cycles. The plant agronomic variables, physiological performance, and microorganism parameters were observed. The results showed that the plant height, fresh and dry shoot weight, along with fresh and dry root weight increased significantly with F. mosseae inoculation under repeated drought stress. The F. mosseae treatment also increased water relative content and decreased proline and lipid peroxidation significantly. Although drought exposure decreased the AMF root colonization rate, the total microbial activity and glomalin-related soil protein were still increased by the F. mosseae inoculation. However, F. mosseae inoculation was negatively correlated to the abundance of phosphate solubilizing microorganisms. The results suggested that F. mosseae gave positive effects on C. frutescens L. growth under repeated drought stress through induced morphological and physiological responses.

Indonesia sebagai negara kepulauan dengan berbagai tipe ekosistem merupakan sumber keragaman jenis khamir yang tinggi. Sekurang kurangnya terdapat 125 taksa khamir yang berhasil diisolasi dari alam dan pangan tradisional Indonesia, dan banyak diantaranya merupakan jenis baru. Biak biak potensial tersebut didapatkan melalui Pengembangan teknik isolasi. Mikroorganisme unggul tersebut memiliki karakter fisiologi yang dapat digunakan dalam proses produksi biofuel seperti etanol, lipid (biodiesel), enzim, surfaktan, asam organik, dan obat. Kedepan perlu dikembangkan teknologi Bioprospeksi khamir untuk pengembangan biofuel dan bioindustri, sehingga kegiatan bioprospeksi dapat dilakukan lebih cepat dan efisien, melalui penggunaan mikroorganisme terpilih yang telah divalidasi oleh taksonom. Peran taksonom sangat sentral dalam kegiatan bioprospeksi yaitu melalui rekomendasi kepada peneliti bioprospeksi tentang karakter fisiologi khusus dari isolat sesuai dengan produk yang akan dikembangkan. Namun jumlah taksonom mikroorganisme sangat terbatas. Masalah kekurangan taksnonom menjadi sangat serius karena kecepatan kerusakan lingkungan yang semakin cepat, sehingga kepunahan mikroorganisme potensial yang terdapat pada Kawasan tertentu berlangsung lebih cepat. Untuk menjamin ketersediaan sumber daya mikroorganisme, Khamir yang telah diisolasi dan diverifikasi karakter fisiologinya harus disimpan di Kultur Koleksi Mikroorganisme dengan standar manajemen yang baik. Untuk selanjutnya sumber daya mikroorganisme tersebut dapat digunakan setiap saat dan didistribusikan untuk kegiatan pembuatan produk yang bernilai ekonomi. Indonesian culture collection (InaCC) memiliki sekitar 350 biak khamir potential untuk dikembangkan pada industri Biofuel. Beberapa teknologi pendukung untuk produksi Biofuel telah dikuasai dengan baik yaitu teknologi penapisan yang mampu dengan cepat mengetahui khamir potensial sebagai pengakumulasi lipid, teknologi cepat untuk prediksi khamir penghasil etanol. Dalam hal pengembangan energi terbarukan, telah dilakukan penjajakan kerjasama nasional dengan Pertamina, dan Kerjasama internasional termasuk didalamnya pemanfaatan limbah lignoselulosa sebagai pendukung industri green biofuel. Selanjutnya perlu pengembangan metoda scale up dari pilot plan menjadi sekala industri. Kerangka pengembangan tersebut dapat dimulai dengan pembentukan zona pengembangan industri green biofuel pada kawasasn terpilih. BRIN telah menjadi inisiator Perpres tentang pengelolaan mikroorganisme yaitu melalui Peraturan Presiden No. 1/2021 tentang perlindungan kekayaan mikroorganisme Indonesia Secara garis besar Perpres ini mengatur 6 hal yaitu: (1) akses terhadap sampel yang mengandung mikroorganisme, (2) perlindungan mikroorganisme melalui mekanisme penyimpanan mikroorganisme mengikuti standard internasional, (3) pendistribusian mikroorganisme serta pemanfaataanya untuk kepentingan nasional, (4) perjanjian pengalihan material, (5) pengelolaan pangkalan data mikroorganisme, dan (6) pembinaan serta pengawasan pengelolaan mikroorganisme. PERPRES tersebut merupakan satu satunya peraturan nasional yang mengatur managemen sumber daya mikroorganisme yang memerlukan langkah langkah operasional lebih lanjut.

Symbiosis of and AMF increase soybean production, but the information on the association between these microbes in Paraserianthes falcataria (L.) I.C. Nielson or ‘sengon’ under Hg contamination is limited. We observed P. falcataria inoculated with arbuscular mycorrhiza fungi (Glomus sp.) stimulate nodule formation under Hg contamination. The study was set up in a pot experiment in the growth chamber a the Hg concentration was adjusted to 50 ppm in soil medium. Inoculation of AMF stimulates nodule formation and increases P. falcataria tolerance to Hg. We isolated several bacteria from the rhizosphere belonging to Rhizobium group and others. Understanding the ecology of soil bacteria is important for Hg bioremediation using P. falcataria.

Xylitol (C 5 H 12 O 5 ) is a non-carcinogenic polyalcoholic sugar. Xylitol is beneficial for diabetics because it can be metabolized without insulin. Corn cobs contain 30% xylose which can be fermented into xylitol by microorganisms. Xylitol can be produced by fermentation of xylose and few microorganisms. Meyerozyma caribbica is a yeast that has been proven to produce xylitol and inhibitor’s resistant. The aim of this research is to test the xylitol productivity by Meyerozyma caribbica InaCC Y67 using corn cobs hydrolyzate and the effect of the volume of fermentation media on xylitol productivity by Meyerozyma caribbica InaCC Y67. The method was carried out by culturing Meyerozyma caribbica InaCC Y67 as a starter on YPD media. Fermentation using 100 mL Erlenmeyer with the variation of fermentation volume is 10 ml and 75 ml, agitation 175 rpm and 30 o C. Parameters were measured based on the dry weight of cells, xylose and xylitol. Data were analyzed using fermentation kinetics. The results of analysis showed that the higher xylitol production was found in the fermentation volume 75 ml with an efficiency value of 7,171%. The highest xylitol production was at the 48th hour with production value of 2.050 g/L. Results from research shows that Meyerozyma caribbica InaCC Y67 can produce xylitol with corn cobs hydrolyzate. The right volume of fermentation in the fermentation process can also increase the productivity of xylitol.

Endophytic microbes are sources for the novel antibiotic. We isolated endophytic bacteria from Zingiberaceae collected from West Sulawesi, Indonesia, and investigated their anticandidal activity. Molecular identification of the isolates was done using 16S rRNA gene sequence analysis. The antimicrobial activity was tested against four bacteria and one yeast. The anticandidal compound of selected bacteria was extracted using three different solvents (chloroform, ethyl acetate, and methanol), and each fraction was tested for their anticandidal activity. Anticandidal minimum inhibitory concentration (MIC) was determined with concentration ranging from 300 to 18.75 μg/mL, and the morphology of the Candida cells after treatment was confirmed by scanning electron microscope (SEM). The identification of anticandidal compounds was conducted using GC-MS. A total of 24 isolates were collected from Zingiberaceae plants. There were 14 genera and 19 species belonging to Gammaproteobacteria (66.67%), Alphaproteobacteria (25.00%), Actinobacteria (4.17%), Bacteriodetes (4.17%), and a new record for Lelliottia aquatilis as an endophytic bacteria. One of 24 isolates identified as Pseudomonas helmanticensis isolated from Alpinia melichroa showed anticandidal activity. Ethyl acetate was the appropriate solvent to extract the anticandidal compounds. Diisooctyl phthalate was found as the most abundant compound in the extract for the anticandidal activity. An increase in extract concentration did not reduce the Candida cell number. The extract treatment showed membrane disruption of Candida albicans cells. We propose that active compounds from P. helmanticensis are potential as anticandidal sources and could be explored more for the pharmaceutical industry.

Banana rhizosphere harbors a unique diversity of microbes including fungi that play critical roles in the growth of the plant host as well as might be important for biologically controlling the fungal soil-borne pathogens particularly Fusarium oxysporum f.sp. cubense (Foc), the causing agent of devastating Panama wilt. Among other fungi, we have succeeded to isolate a Trichoderma species from rhizosphere of healthy banana. Molecular identification revealed the isolate as Trichoderma virens InaCC F1030 (being collection of Indonesian Culture Collection or InaCC). Therefore, the aim of this study was to investigate the biological control of our isolate against Foc as well as plant growth promoting ability through its ability to produce auxin (indole-3-acetic acid/IAA). Two approaches were employed to evaluate the antagonism of our isolate against Foc, through direct confrontation test and volatile organic compounds (VOCs) producing. We found that our isolate was considered as antagonistic to the Foc, but not highly antagonistic according to direct confrontation assay. It was also revealed that our isolate produces the VOCs that inhibited around 50% of the mycelial growth of the test pathogen after six to seven days of exposure. Our isolate was able to produce the IAA in axenic submerged fermentation condition particularly in the presence of the precursor L-tryptophan. IAA production ability as well as the mycelial biomass of fungus were increased approximately 17% and 120% respectively as the effect of supplementation of 0.1% of L-tryptophan. These in vitro bioassays lead us to conclude that somehow our isolate T. virens InaCC F1030 has potency to be utilized as biocontrol and biofertilizer agent.

In fermented beverages, yeasts have been exploited for many years and are well-known as alcohol producers. In Indonesian traditional beverages, however, information about microbiology and potential bioactive-compounds of rice wine produced by the local people, especially in Lombok, are limited. The present study described the compounds of traditional rice wine including yeast species and its produced compounds that have biological activity. The yeast in rice wine was isolated using three growth agar media by serial dilution, selected the yeast colonies for molecular identification, and performed gas chromatography tandem with mass spectrometry for profiling the chemical compounds of the rice wine. The result indicated that the rice wine sold without distillation still contained Saccharomyces cerevisiae as the main alcohol producer. Meanwhile, at least six bioactive-compounds such as l-(+)-Ascorbic acid 2,6-dihexadecanoate, performic acid, octadecanoic acid, sulfurous acid, tetratriacontane, and eicosane were detected and reportedly related to antimicrobial, antiviral, anticancer, and other pharmacological activities. These findings could be the first step of studies on exploring Indonesian’s local rice wine as alcohol and bioactive-compound sources for health benefits.

Recently, microorganism-based hydrogel has been attracting attention for its applications in the revegetation of marginal land and in crop production on dry land. Komagataeibacter xylinus is a bacterium that can biosynthesize bacterial cellulose, the material for microorganism-based hydrogel. Nonetheless, various factors influence the production of bacterial cellulose by K. xylinus, and one of them is the substrate nutrient used. Therefore, we investigate various types of carbon sources for bacterial cellulose synthesis by our strain collection K. xylinus InaCC B404. Five simple sugars were selected as the main carbon source, namely glucose, lactose, mannitol, xylose, and sucrose. The results showed that the type of carbon source affected bacterial cellulose production yield. Lactose was the carbon source that produced the highest yield of bacterial cellulose (8.7 g/l). Moreover, sugar consumption by the isolate was also the lowest in the lactose broth (25% w/w). However, the swelling capacity of dried natural bacterial cellulose produced was relatively similar for all carbon sources, and it was between 2.5-3.1 times per gram. Physical observation based on scanning electron microscope (SEM) micrographs showed that the appearances of bacterial cellulose derived from lactose and glucose were more rugged than other types of bacterial cellulose. On the other hand, chemical structure analyses using Fourier-Transform Infrared Spectroscopy (FTIR) revealed that the chemical composition of bacterial cellulose was relatively similar for all carbon sources. This was revealed by the presence of the characteristic bands of cellulose. This study points to the value of the use of lactose as a carbon source in the production of bacterial cellulose as a hydrogel for further application.

Red-yeasts are pigmented yeast species that could synthesize carotenoids for food supplements and pharmaceutical purposes. However, this group contains a lot of species which need to be explored thoroughly. The objective of this study was to view the β-carotene production of three indigenous red-yeasts by modifying glucose content in the growth medium and verifying their phylogenetic relationship. The glucose content modification in growth media influenced the β-carotene production of each species. Rhodosporidiobolus ruineniae (InaCCY1638/Y15Eg075) and Rhodosporidiobolus poonsokiae (InaCCY1606/Y15Kr068) produced higher β-carotene than Rhodotorula paludigena (InaCCY1527/Y15Eg004). These Rhodosporidiobolus species were able to produce higher β-carotene from 0.5 to 2% glucose content while Rhodotorula was low production in 2% glucose content. The higher producers by Rhodosporidiobolus species were clustered to Ruineniae clade and could be a potential clade for higher β-carotene production. Based on this result, using a high carotenoid producer of red-yeasts from indigenous strains is potential to be developed for β-carotene bioindustry in the future.

Trichoderma harzianum is a filamentous fungus that has been known to have biocontrol and plant growth-promoting ability. However, the propagation of this fungus particularly through solid state fermentation (SSF) and characterization of its enzyme activity as one the indicator of quality of fermentation process are still needed to be explored further. Rice grain and its derivative products have economically as well as nutrient composition features beneficial as substrates of fungal propagation through SSF. Therefore, the aim of this study was to investigate the effect of white rice, rice bran, and combination of white rice and rice bran on the activity of amylase and cellulase by Trichoderma harzianum strains under SSF. Two strains of the fungus, InaCC F116 and InaCC F89, as well as their consortium were employed as fungal inoculants. After closed fermentation in dark chamber at 30 ± 1°C for 7 days, the activity of amylolytic as well as cellulolytic enzyme was assayed. The result showed that the presence of rice bran as a substrate increased the activity of crude amylase and crude cellulase. In all substrates, the strain F116 has low activity of both enzymes. The fungal consortium improves the activity of crude enzymes in all substrates. Therefore, the amylase and cellulase activity by T. harzianum in SSF condition were strain- as well as substrate-dependent.

Marigold flower (Tagetes erecta L.) produces lutein compounds which present biological activities such as antioxidant, antiinflammatory, antimutagenicity, and immunomodulatory effects. The study was to investigate the antioxidant activity of the lutein of T. erecta L. and the effect of lutein on the activity and phagocytic capacity of macrophage cells. The antioxidant screening was carried out using diphenyl picrylhydrazyl (DPPH), 2,2′-and-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay with serial concentrations and ferric-reducing antioxidant power (FRAP) method. For the observation of activity and phagocytic capacity of peritoneal macrophages, twenty-eight mice were used and divided into seven groups each comprising four replicates, i.e., Group (I) normal controls, mice were untreated (II) a negative control, mice were induced by Staphylococcus aureus (III) positive control, mice were induced by S. aureus and treatment of meniran extract (Phyllanthus niruri). The treatment group (IV-VII) mice were induced by S. aureus and treated crude lutein, respectively: 0.15 mg, 0.30 mg, 0.60 mg, and 0.90 mg. 20 g−1 of body weight. The lutein extracted from T. erecta shows an antioxidant activity against DPPH radical with an IC50 value of 53.58 μg.ml−1, while the antioxidant activity against ABTS has an IC50 value of 72.91 μg.ml−1. The antioxidant activity test results by the FRAP method at each lutein concentrations of 10, 25, 50, and 75 ppm were obtained respectively of 33, 88, 185.5, and 288.5 μmol Fe2+/g extract. The data were analyzed using one-way ANOVA and Duncan’s multiple range test (DMRT) after. The phagocytic activity was 45.5%; 54.75%; 57.50% and 67.0%, respectively, while the phagocytic capacity values ​​were 355; 519; 611 and 767 S. aureus bacterial cells per 50 macrophage cells. The lutein from marigolds (T. erecta L.) is capable of scavenging free radicals and reducing oxidants. Lutein can increase the activity and capacity of phagocytic of peritoneum macrophage cells in mice.

The objective of the study was to isolate fungi from healthy and wilt banana in order to understand their diversity as well as their physiological characters to produce growth hormone and solubilize inorganic phosphate. The fungi were isolated from the leaves, roots, and rhizosphere of the banana. As many 13 fungal isolates were associated with wilt banana, consisted of Fusarium, Trichoderma, Bipolaris, Colletotrichum, Clonostachys, and Talaromyces genus. On the other hand, 12 fungal isolates that belong to 10 genera related to the healthy banana have been isolated, including Trichoderma, Pallidocercospora, Purpureocillium, Pallidocercospora, Mycosphaerella, Chaetomium, Neonectria, Pararamichloridium, Xylaria, and Neocordana. We found that the fungal isolates associated with wilt banana, particularly from the Fusarium genus, actively produced IAA in the presence of exogenous L-Tryptophan have high ability to solubilize insoluble phosphate. The ability of fungal isolates to solubilize insoluble phosphate was correlated with their ability to acidify the medium. The presence of arbuscular mycorrhizal in soil may affect growth and yield of chili (Capsicum annuum L.). This experiment was done to know the effect of arbuscular mycorrhizal inoculation on growth of chilli. Microwave soil sterilization was used to reduce the number of microbes in the media, enabling to observe the interaction between chili peppers and arbuscular mycorrhizal fungi. A single culture products (A) and mixed culture products (B) were used as arbuscular mycorrhizal spores. In contrast to product A, the spore counted calculation reported that product B had the most spores, with 51 spores / 50 g soil. The treatment of arbuscular mycorrhizal fungi and microwave sterilization against the height of chili plant had no significant effect, according to a two-factor ANOVA (α: 0.05) analysis of agronomic characteristics. Inoculation of mycorrhizae had a significant effect on chili plant height. Arbuscular mycorrhizal fungi inoculation and microwave sterilization had significant effect on the root length of chili plants. Arbuscular mycorrhizal fungi in single and mixed cultures could colonize roots by forming internal hyphae, vesicles, and spores. The best way to support the growth of chili plants is to use planting media that has not been sterilized and contains mycorrhizal fungi.

Lactobacillus plantarum (LAB) is a group of lactic acid bacteria that can produce exopolysaccharide (EPS). The objective of this study was to determine the effect of adding selenium chloride oxide (SeOCl2) to the increased immunomodulatory potential of EPS L. plantarum Y-1.2. The experiment was conducted in vivo using macrophage cells of peritoneal fluid of mice induced by Staphylococcus aureus. EPS was obtained by precipitation at 4 °C. Identification test using HPLC showed that crude EPS of L. plantarum Y-1.2 containing glucose monomers for value retention times 4.198 and 4.204, crude EPS approaches the retention time of glucose BP 4.234. Testing of immuno-modulatory divided into six treatment groups. Group 1-3 as the control group, a positive control (Stimuno), negative control (crude EPS 50 mg/kg BW in mice), normal control (distilled water). Group 4-6 as the test group was given crude EPS 50 mg/kg BW in mice and selenium oxide chloride doses of 0.13 µg; 0.26 µg; 0.52 µg. The test results showed a dosage of 0.52 µg selenium chloride oxide to crude EPS of 50 mg/kg BW in mice could increase the activity and phagocytic capacity amounted to 28.06% and 28.39%

A novel yeast species was isolated from leaf litter of Macropanax dispermus obtained from the Cibodas Botanical Garden, West Java, Indonesia. The two strains of the species displayed typical characteristics of the genus Citeromyces. Phylogenetic analysis based on the gene sequences of the D1/D2 domains of large subunit (LSU) rDNA, internal transcribed spacer (ITS) including 5.8S rDNA, mitochondrial small-subunit rRNA gene (MtSm), and translation elongation factor-1α (EF-1α) showed that the novel strains were clearly separated from the other four existing species of the genus Citeromyces. Therefore, the two strains were proposed to represent a novel species within the genus Citeromyces, for which the name Citeromyces cibodasensis is proposed; the type strain is NBRC 110244T (= CBS 14272T = InaCCY703T = AK 01).

The identification of yeasts or yeasts-like fungi and verify their diversity are principal aspect for bioindustry and ecosystem sustainability. Taxonomic approach provides identification tool to ensure the taxonomic position of yeasts and yeasts-like fungi which definitely set to utilization concerns. The aim of this study is to understanding the taxonomic position of yeasts and yeasts-like fungi from the distinctive of its sequences relationship. Yeasts and yeasts-like fungi strains were isolated through various culture dependent methods from natural resources samples of Karimun Besar Island, Province of Riau Islands, Indonesia. The identification process was performed through amplifying the accurate DNA-based in D1/D2 region of large subunit (26S) ribosomal DNA. As the result, a total of 85 isolates of yeasts and yeasts-like fungi were obtained with 16 closest related taxa through phylogenetic tree construction. Ascomycetous was the predominating group representing 91% of the total isolates sequences followed by Basidiomycetous (8%) and Zygomycetous (1%). The black yeasts (yeasts-like) known as Aureobasidium melanogenum was predominant species with represent to 54% of total isolates and present in particular habitat. Taxonomically, there are six isolates are represent to be novel taxa candidates which pretend to enhance genetic resources of yeasts and yeasts-like fungi especially from Indonesia. In addition, this information provides specific technique to reach specific yeasts or yeasts-like fungi species in nature by managing the sample collection and culture methods.

Citric acid demand increases steadily, and there is a need to increase productivity through selection of suitable carbon sources, and addition of substances that increase citric acids production rate. Methanol has been suggested to increase citric acid fermentation on high carbohydrate containing substances. The objective of the study was to evaluate the suitability of sweet sorghum juice for citric acids production and to verify the effect of methanol on citric acids production using Aspergillus niger InaCC F539 as inoculant. Sweet sorghum juice with the total initial reducing sugar of 11.5 % (w/v) was used as the sole carbon sources. To study the effect of total initial reducing sugar on citric acid production the initial reducing sugar was adjusted to the concentration of 30 to 75 g/L. Preliminary experiment was conducted to get the optimum methanol concentration that stimulate citric acid production. The optimum methanol concentration that stimulate citric acid production was 4% (v/v). Submerged fermentation was conducted as shake culture (125 rpm at 28 °C). Citric acids production was affected by total initial reducing sugar. Higher total initial reducing sugar produced higher citric acids. Maximum citric acid production was 18.96g/L on sweet sorghum juice with 75 g/L total initial reducing sugar. Methanol 4 % (v/v) increase citric acid production by 41.35 to 65.89 %. Juice of sweet sorghum was a good medium for citric acids production, and methanol stimulate and increase citric acid production. It is a good basis for exploring efficient and cost effective industrial scale citric acid production. Keywords: Citric acid, Methanol, Sweet sorghum, Aspergillus niger

Three novel oleaginous yeasts were isolated from soil collected around Kalimantan Island, Indonesia. Based on their morphological and biochemical characteristics and sequence typing using the D1/D2 domain of the large subunit (LSU) rRNA and translation elongation factor 1 alpha gene ( EF-1α ), the eight strains were shown to be novel species and were named Lipomyces maratuensis sp. nov. (type strain: NBRC 110264 T = InaCC Y720 T = JSAT12-2-Y011 T ; MycoBank no. MB 816185), Lipomyces tropicalis sp. nov. (type strain: NBRC 110265 T = InaCC Y730 T = JSAT12-2-Y012 T ; MycoBank no. MB 816186), and Lipomyces kalimantanensis f.a., sp. nov. (type strain: NBRC 110267 T = InaCC Y721 T = JSAT12-2-Y029 T ; MycoBank no. MB 816187). Strain NBRC 110264 exhibited high lipid production.

This study describes three novel xylose-assimilating yeasts, which were isolated from decayed wood collected from Bung Hatta Botanical Garden in West Sumatra and Cibodas Botanic Garden in West Java, or from litter from Eka Karya Bali Botanic Garden in Bali, Indonesia. Phylogenetic analysis was performed based on the sequences of the D1/D2 domains of the large ribosomal subunit (LSU), the small ribosomal subunit (SSU), the internal transcribed spacer (ITS) and elongation factor-1α (EF-1α), and the three strains were found to represent three novel species belonging to genera Barnettozyma or Wickerhamomyces. The morphological, biochemical and physiological characteristics indicated that the strains were distinct from other closely related species. Strains 13Y206T and 14Y196T belonging to the Barnettozyma clade are described as the type strains of Barnettozyma xylosiphila sp. nov. (type strain 13Y206T=NBRC 110202T=InaCC Y726T; MycoBank MB808598) and Barnettozyma xylosica sp. nov. (type strain 14Y196T=NBRC 111558T=InaCC Y1030T; MycoBank MB819485). Strain 14Y125T belonging to the Wickerhamomyces clade is described as the type strain of Wickerhamomyces xylosivorus f.a., sp. nov. (type strain 14Y125T=NBRC 111553T=InaCC Y1026T; MycoBank MB819484).

Polyol esters of fatty acids (PEFA) are amphiphilic glycolipids produced by yeast that could play a role as natural, environmentally friendly biosurfactants. We recently reported discovery of a new PEFA-secreting yeast species, Rhodotorula babjevae, a basidiomycetous yeast to display this behavior, in addition to a few other Rhodotorula yeasts reported on the 1960s. Additional yeast species within the taxonomic order Sporidiobolales were screened for secreted glycolipid production. PEFA production equal or above 1 g L−1 were detected in 19 out of 65 strains of yeast screened, belonging to 6 out of 30 yeast species tested. Four of these species were not previously known to secrete glycolipids. These results significantly increase the number of yeast species known to secrete PEFA, holding promise for expanding knowledge of PEFA synthesis and secretion mechanisms, as well as setting the groundwork towards commercialization.

Actinomycetes have been widely explored for new antibiotic production, but not many studies explore its abilities to inhibit the growth of phytopathogenic fungi and solubilize insoluble phosphate hence stimulate the growth of plants. We isolated Actinomycetes from the soil. Based on morphology, physiology, and 16S rDNA analyses, the isolate is closely related to Streptomyces sasae. The strain was able to inhibit the growth of phytopathogenic fungi Fusarium solani, and Fusarium oxysporum. S. sasae produced secondary metabolites 2-methyl-1,3-dioxolane as the major constituent. The strain assimilated variable carbon sources include L-arabinose, D-fructose, D-glucose, D-mannitol, Lactose, raffinose, L-rhamnose, and sucrose. The strain grew at pH 6.0 to 8.0, and at salinity (1-3%). Their growth was affected by the salinity level. The strain solubilized Ca-P at 1-3% salinity, but their ability to solubilize phosphate was influenced by salinity. The strain was also able to solubilize rock phosphate. Their ability to solubilize less soluble phosphate and inhibit the growth of F. solani and F. oxysporum may imply that this strain is potential for biocontrol agents. The objective of the study was to isolate microorganism that can produce growth hormone, fix atmospheric nitrogen, and solubilize inorganic phosphate that may be used for enhancing the growth of banana in marginal land. A total of 25 bacteria associated with banana that belonged to six genera were isolated from namely Acinetobacter , Enterobacter , Pantoea , Klebsiella , Rhizobium , and Cupriavidus genera. Gammaproteobacteria is the dominant class, followed by Betaproteobacteria and Alphaproteobacteria . All isolates were screened for multiple plant growth promoting traits which may play a role in banana growth, namely fixing nitrogen, solubilizing phosphate, and producing Indole Acetic Acid (IAA). Twenty-two isolates were capable to fix nitrogen, 21 isolates can solubilize insoluble phosphate, and 15 isolates produced IAA dependent of L-Tryptophan presence. Despite most of the bacteria isolates exhibited one plant growth-promoting activities, Enterobacter and Klebsiella genera showed three of plant growth promoting bacteria traits.

The objective of the study was to isolate, identify and characterize phytase producing yeast. The yeast was isolated from various resources, and the identification of yeast was conducted by the sequencing of the ITS region. Of 41 isolates, 24 isolated produced phytase. The highest phytse producing yeast was identified as Candida natalensis YEG 132, Saturnispora quitensis YEg 135, and Candida albicans YEg 226. These isolates were able to use various carbon and nitrogen sources. The maximum phytase production depends on isolates, media composition, pH and incubation temperature. Temperature affected phytase production. All isolates produce the highest phytase was at 30°C. Increased temperature to 35° C and 40°C inhibits phytase production by all strains. S. quitensis was the highest isolate inhibited by an increase of incubation temperature. The pH of cultures affects phytase production and varies depending on isolate. S. quitensis and Candida natalesis produced the highest phytase activities at pH 5.0, while C. albicans was at pH 6.0. All isolates were able to assimilate glucose, dextrose, sucrose, xylose and lactose. In general, glucose was the best carbon source for phytase activity. But each isolate has its preferential on carbon sources. S. quitensis produced the highest phytase on glucose, but not on dextrose. Each isolate has preferential on nitrogen sources. Yeast extract was the best N-sources for C. albicans, while ammonium sulfate was best for C. natalensis, S. quitensis. Ammonium nitrate was not favorable for phytase production by C. natalensis. Candida natalensis YEG 132, Saturnispora quitensis YEg 135, and Candida albicans YEg 226 was a good candidate for phytase production.

The toxic fermentation inhibitors in lignocellulosic hydrolysates pose significant problems for the production of second-generation biofuels and biochemicals. Among these inhibitors, 5-(hydroxymethyl)furfural (HMF) and furfural are specifically well known. This study investigated the furfural and 5-HMF tolerance in Candida strains, which could be used for the development of advanced generation bioethanol processes. The 10 isolates of Candida were selected based on the cell growth and bioethanol production on YPD medium containing several concentrations of furfural and 5-HMF by using spectrophotometer and HPLC. Candida parapsilosis Y80 could grow and produce bioethanol in the medium that contains furfural and 5-HMF with a concentration of 60 mM and 40 mM, respectively. Based on the results, C. parapsilosis Y80 has potential activity in the development of bioethanol fermentation.

The use of phytase as a feed supplement is getting popular. However, the production system of this enzyme should be optimized to reduce production cost. The objective of this study was to evaluate the ability of 10 fungi isolates i.e., Aspergillus niger (5 isolates), and Neurospora crassa InaCC F226 to produce phytase, and select best phytase producer for phytase production on coconut oil cake supplemented with rice brand in solid-state fermentation. A. niger Str3 and N. sitophila produced phytase of 4.6 and 3.4 unit respectively were selected for phytase production owing to its ability to produce phytase in submerging fermentation with glucose as the primary carbon sources. These potential isolates were then used for phytase production on coconut oil cake supplemented with rice brand in solid-state fermentation. The effect of inoculants type, initial moisture content, and additional carbon sources were evaluated to obtain the optimum condition for phytase production. Media contained coconut oil cake supplemented with rice brand at a ratio of 20 to 50% could be used for phytase production. Initial moisture content and incubation time affect phytase production. Optimum initial moisture content was about 60-70%. This work concludes A. niger and N. sithophyla were good inoculant for phytase production using formulated media contained coconut oil cake and rice brand in solid-state fermentation.

D-Xylitol is a five-carbon sugar alcohol, which can be obtaines from xylose of agriculutural wastes with microbial fermentation. The inhibitory chemical complexes (ICC) were founded after biomass pretreatment step. The ICC in acid hydrolysate can inhibit microbial metabolism in the medium. Several methods for detoxification; adsorption by activated charcoal, by ion exchange resin or by using enzymes. This study aims to evaluate the effect of using charcoal in xylitol fermentation by using lignocellulosic hydrolisate. In this work, detoxification process by added 1% activated charcoal to reduce the inhibitors in corn cobs hydrolysate and sugarcane trash hydrolysate. The biomass hydrolysate was fermented by Meyerozyma caribbica Y67 to produce xylitol. It is expected that detoxification can reduce concentration of inhibitors, so that microbial growth is not inhibited and more product. The results showed that M. caribbica Y67 had not significant different between pure hydrolysate and detoxified hydrolysate with activated charcoal. We suggested that M. caribbica Y67 tolerance to ICC and potential for xylitol fermentation from lignocellulosic biomasses.

The aim of the study was to determine the production of citric acid in liquid and solid media made from the juice and bagasse of sorghum plant. Sorghum has a great potential to be utilized as fermentation substrate for production of citric acid. The production of citric acid by fermentation is influenced by several factors including the type of media and used microorganisms. Fermentation to produce citric acid can be done by using mold as well as yeast. In this study, the mold isolates were Beauveria bassiana and Paelomyces lilacinus, while the yeast isolates were Candida utilis and Candida lessepsii. The fungal fermentations were conducted in two phases of medium, liquid and solid. Three formulas were prepared for liquid fermentation, namely sorghum juice only, sorghum juice supplemented with NH4NO3 and micronutrients, and sorghum juice supplemented with asparagine and micronutrients. Two formulas were prepared for solid fermentation, sulfuric acid hydrolyzed sorghum bagasse and non-hydrolyzed sorghum bagasse. The results showed that citric acid yield in solid fermentation is lower than liquid fermentation. For all formulas, the citric acid formation through liquid fermentation was higher in yeast than in molds. On the other hand, for solid fermentation, citric acid production was higher in hydrolyzed media than non-hydrolyzed media, except for yeast Candida lessepsii.

One of the rare types of pentose sugar is xylitol, which has various benefits in the field of food and medicine. Xylitol is one of the results of xylose fermentation and few microorganisms are able to produce it. Meyerozyma caribbica Y67 is one of the yeast collections of Indonesian Culture Collection (InaCC) which can produce xylitol. The production of xylitol about cell initiation and media volume gave different results for each factor. The fermentation conditions were using erlenmeyer 250 mL, agitation 150 rpm and 30ºC temperature. The parameters analyzed were dry cell weight (DCW), xylose, and xylitol. The results of this study showed that cell initiation with an optical density at 600 nm (OD600) = 5 (≍1.07x107 CFU or 3.980 g/L) had the highest efficiency in producing xylitol for 24 hours of fermentation, 51, 099%; specific growth rate (µ): 0.069. In the media volume variable, for 24 hours fermentation, the high-efficiency value of 20% volume erlenmeyer is 55, 708%; (µ): 0.082 and 48 hours fermentation is 40% volume erlenmeyer which is 71, 959%; (µ): 0.048. The research is expected to be used as a scale up recommendation for the industry.

Indole-3-acetic acid (IAA) is one of the most common plant hormones that regulate many aspects of growth and development of plants. Trichoderma harzianum, a plant growth promoting fungus, has the ability to produce IAA. However, various environmental factors influence the formation of the hormone. The objective of this study was to optimize the environmental condition for the production of IAA by Trichoderma harzianum InaCC F88 strain. The effects of L-tryptophan concentration, temperature, pH, salinity, and incubation time on IAA production and biomass were studied. The in vitro evaluations were carried out in the axenic condition of Luria-Bertani Medium. Supplementation of 1% L-tryptophan in the medium gives maximum IAA production. The most favourable initial pH and temperature for IAA production are 6.0 and 27°C, respectively. In the salinity test, the medium containing 1% NaCl yielded maximum IAA formation. After 4 days of incubation, the concentration of IAA reached equilibrium. Hence, optimum IAA production in liquid fermentation could be achieved by manipulating those factors.
