Abstract

The Brain Imaging Data Structure (BIDS) is a community-driven standard for the organization of data and metadata from a growing range of neuroscience modalities. This paper is meant as a history of how the standard has developed and grown over time. We outline the principles behind the project, the mechanisms by which it has been extended, and some of the challenges being addressed as it evolves. We also discuss the lessons learned through the project, with the aim of enabling researchers in other domains to learn from the success of BIDS.

Abstract

As the global health crisis unfolded, many academic conferences moved online in 2020. This move has been hailed as a positive step towards inclusivity in its attenuation of economic, physical, and legal barriers and effectively enabled many individuals from groups that have traditionally been underrepresented to join and participate. A number of studies have outlined how moving online made it possible to gather a more global community and has increased opportunities for individuals with various constraints, e.g., caregiving responsibilities. Yet, the mere existence of online conferences is no guarantee that everyone can attend and participate meaningfully. In fact, many elements of an online conference are still significant barriers to truly diverse participation: the tools used can be inaccessible for some individuals; the scheduling choices can favour some geographical locations; the set-up of the conference can provide more visibility to well-established researchers and reduce opportunities for early-career researchers. While acknowledging the benefits of an online setting, especially for individuals who have traditionally been underrepresented or excluded, we recognize that fostering social justice requires inclusivity to actively be centered in every aspect of online conference design. Here, we draw from the literature and from our own experiences to identify practices that purposefully encourage a diverse community to attend, participate in, and lead online conferences. Reflecting on how to design more inclusive online events is especially important as multiple scientific organizations have announced that they will continue offering an online version of their event when in-person conferences can resume.

Abstract

Only a tiny fraction of the data and metadata produced by an fMRI study is finally conveyed to the community. This lack of transparency not only hinders the reproducibility of neuroimaging results but also impairs future meta-analyses. In this work we introduce NIDM-Results, a format specification providing a machine-readable description of neuroimaging statistical results along with key image data summarising the experiment. NIDM-Results provides a unified representation of mass univariate analyses including a level of detail consistent with available best practices. This standardized representation allows authors to relay methods and results in a platform-independent regularized format that is not tied to a particular neuroimaging software package. Tools are available to export NIDM-Result graphs and associated files from the widely used SPM and FSL software packages, and the NeuroVault repository can import NIDM-Results archives. Abstract

The development of magnetic resonance imaging (MRI) techniques has defined modern neuroimaging. Since its inception, tens of thousands of studies using techniques such as functional MRI and diffusion weighted imaging have allowed for the non-invasive study of the brain. Despite the fact that MRI is routinely used to obtain data for neuroscience research, there has been no widely adopted standard for organizing and describing the data collected in an imaging experiment. This renders sharing and reusing data (within or between labs) difficult if not impossible and unnecessarily complicates the application of automatic pipelines and quality assurance protocols. To solve this problem, we have developed the Brain Imaging Data Structure (BIDS), a standard for organizing and describing MRI datasets. The BIDS standard uses file formats compatible with existing software, unifies the majority of practices already common in the field, and captures the metadata necessary for most common data processing operations.

Abstract

The Brain Imaging Data Structure (BIDS) is a community-driven standard for the organization of data and metadata from a growing range of neuroscience modalities. This paper is meant as a history of how the standard has developed and grown over time. We outline the principles behind the project, the mechanisms by which it has been extended, and some of the challenges being addressed as it evolves. We also discuss the lessons learned through the project, with the aim of enabling researchers in other domains to learn from the success of BIDS.

Abstract

Cardiovascular ageing contributes to cognitive impairment. However, the unique and synergistic contributions of multiple cardiovascular factors to cognitive function remain unclear because they are often condensed into a single composite score or examined in isolation. We hypothesized that vascular risk factors, electrocardiographic features and blood pressure indices reveal multiple latent vascular factors, with independent contributions to cognition. In a population-based deep-phenotyping study (n = 708, age 18–88), path analysis revealed three latent vascular factors dissociating the autonomic nervous system response from two components of blood pressure. These three factors made unique and additive contributions to the variability in crystallized and fluid intelligence. The discrepancy in fluid relative to crystallized intelligence, indicative of cognitive decline, was associated with a latent vascular factor predominantly expressing pulse pressure. This suggests that higher pulse pressure is associated with cognitive decline from expected performance. The effect was stronger in older adults. Controlling pulse pressure may help to preserve cognition, particularly in older adults. Our findings highlight the need to better understand the multifactorial nature of vascular aging.

Abstract

Low-intensity transcranial electrical stimulation (tES), including alternating or direct current stimulation, applies weak electrical stimulation to modulate the activity of brain circuits. Integration of tES with concurrent functional MRI (fMRI) allows for the mapping of neural activity during neuromodulation, supporting causal studies of both brain function and tES effects. Methodological aspects of tES-fMRI studies underpin the results, and reporting them in appropriate detail is required for reproducibility and interpretability. Despite the growing number of published reports, there are no consensus-based checklists for disclosing methodological details of concurrent tES-fMRI studies. The objective of this work was to develop a consensus-based checklist of reporting standards for concurrent tES-fMRI studies to support methodological rigor, transparency and reproducibility (ContES checklist). A two-phase Delphi consensus process was conducted by a steering committee (SC) of 13 members and 49 expert panelists through the International Network of the tES-fMRI Consortium. The process began with a circulation of a preliminary checklist of essential items and additional recommendations, developed by the SC on the basis of a systematic review of 57 concurrent tES-fMRI studies. Contributors were then invited to suggest revisions or additions to the initial checklist. After the revision phase, contributors rated the importance of the 17 essential items and 42 additional recommendations in the final checklist. The state of methodological transparency within the 57 reviewed concurrent tES-fMRI studies was then assessed by using the checklist. Experts refined the checklist through the revision and rating phases, leading to a checklist with three categories of essential items and additional recommendations: (i) technological factors, (ii) safety and noise tests and (iii) methodological factors. 31. Decoding frequency-specific modulation of fMRI network connectivity: a tACS study

Earlier studies emphasized regional specificity in cognition and behaviour, while recent studies emphasize the role of connectivity and how it leads to a coordinated interplay between brain regions. Non-invasive brain stimulation, such as transcranial Alternating Current Stimulation (tACS), has been shown to modulate functional connectivity to shape cognitive function. However, it is not clear that different stimulation frequencies will impact brain dynamics differently and that this will be readily measured using imaging approaches that can verify the effects of stimulation. We propose multivariate pattern analyses (MVPA) to demonstrate whether tACS has differentiable effects on regional BOLD activity and connectivity within the brain networks which allow us to decode the stimulation frequency. Using a randomised epoch-based design, 18 participants underwent four runs of concurrent tACS-fMRI while performing a match-to-sample task. During the task, participants received dual-channel HD-tACS targeting the frontoparietal network (FPN) in the right hemisphere at four pre-specified frequencies (5, 10, 20, 60 Hz). FMRI data were pre-processed and analysed within the Automatic Analysis pipeline, which implemented MVPA decoding using The Decoding Toolbox. We tested the decoder accuracy for both the magnitude of functional activity and the effective connectivity as estimated using psychophysiological interaction (PPI) analysis at voxel, region, and network levels, summarised within the seven networks of the Schaefer-based parcellation. Univariate analyses confirmed that the task successfully engaged the FPN along with the visual cortex and the basal ganglia network. Decoding based on the PPI connectivity pattern was the most successful in decoding across stimulation conditions; especially within and between the "Dorsal Attention" and "Control" networks, which involve frontoparietal brain regions. Our study demonstrates that functional connectivity within a task-relevant network is sensitive to specific stimulation frequencies. Our findings provide support for the state-dependency of brain stimulation and pave the way for future closed-loop neuromodulation studies.

Abstract

The locus coeruleus (LC), the origin of noradrenergic modulation of cognitive and behavioral function, may play an important role healthy ageing and in neurodegenerative conditions. We investigated the functional significance of age-related differences in mean normalized LC signal intensity values (LC-CR) in magnetization-transfer (MT) images from the Cambridge Centre for Ageing and Neuroscience (Cam-CAN) cohort - an open-access, population-based dataset. Using structural equation modelling, we tested the pre-registered hypothesis that putatively noradrenergic (NA)-dependent functions would be more strongly associated with LC-CR in older versus younger adults. A unidimensional model (within which LC-CR related to a single factor representing all cognitive and behavioral measures) was a better fit with the data than the a priori two-factor model (within which LC-CR related to separate NA-dependent and NA-independent factors). Our findings support the concept that age-related reduction of LC structural integrity is associated with impaired cognitive and behavioral function.

41. Early functional brain network changes in Mild Cognitive Impairment

Poster number: 1152.Introduction:Alzheimer's disease is the leading neurodegenerative disorder both in terms of its scale and its socio-economic impact, therefore there is an increasing international effort for more accurate early diagnosis. Although a growing number of neuroimaging reports suggests that disruption of extensive brain networks rather than dysfunction of one or more brain areas are behind the symptoms of Alzheimer's disease and other dementias1, the underlying effects of early mild cognitive impairment on functional brain network topology remain poorly understood. Hence we used graph theoretical analysis to evaluate the functional network properties in early mild cognitive impairment.Methods:Resting state functional images were obtained from ADNI2 study2 together with the corresponding T1-weighted MPRAGE and fieldmap images for 24 subjects classified as normal controls (CN, n=12) or with early mild cognitive impairment (EMCI, n=12) at the initial ADNI2 visit, as well as 12 and 24 months later. Images were processed using a novel pipeline automatic analysis3. Workflow consisted of combined motion and distortion correction, as well as slice timing correction. Images were normalised using DARTEL. Time series have been extracted from atlas based brain regions for the construction of brain connectivity matrices and to compute network-based statistics.Results:We investigated the interaction between factors GROUP (NC, EMCI) and TIME (initial, year1, year2) to identify longitudinal changes specific for the ECMI group. Our results demonstrate that early mild cognitive impairment reorganizes the resting brain networks with the earliest changes affecting the hubs of the default mode network and leading to a decrease in local information transfer in the prefrontal cortex, as indicated by local efficiency accompanied by similar decreases in the hippocampus. Conclusions:These findings underscore the utility of graph-based network analysis to identify longitudinal functional network connectivity changes for early diagnosis using resting state in early Mild cognitive impairment.

Abstract

As the global health crisis unfolded, many academic conferences moved online in 2020. This move has been hailed as a positive step towards inclusivity in its attenuation of economic, physical, and legal barriers and effectively enabled many individuals from groups that have traditionally been underrepresented to join and participate. A number of studies have outlined how moving online made it possible to gather a more global community and has increased opportunities for individuals with various constraints, e.g., caregiving responsibilities. Yet, the mere existence of online conferences is no guarantee that everyone can attend and participate meaningfully. In fact, many elements of an online conference are still significant barriers to truly diverse participation: the tools used can be inaccessible for some individuals; the scheduling choices can favour some geographical locations; the set-up of the conference can provide more visibility to well-established researchers and reduce opportunities for early-career researchers. While acknowledging the benefits of an online setting, especially for individuals who have traditionally been underrepresented or excluded, we recognize that fostering social justice requires inclusivity to actively be centered in every aspect of online conference design. Here, we draw from the literature and from our own experiences to identify practices that purposefully encourage a diverse community to attend, participate in, and lead online conferences. Reflecting on how to design more inclusive online events is especially important as multiple scientific organizations have announced that they will continue offering an online version of their event when in-person conferences can resume.
