Background Evolocumab is a monoclonal antibody that inhibits proprotein convertase subtilisin–kexin type 9 (PCSK9) and lowers low-density lipoprotein (LDL) cholesterol levels by approximately 60%. Whether it prevents cardiovascular events is uncertain. Methods We conducted a randomized, double-blind, placebo-controlled trial involving 27,564 patients with atherosclerotic cardiovascular disease and LDL cholesterol levels of 70 mg per deciliter (1.8 mmol per liter) or higher who were receiving statin therapy. Patients were randomly assigned to receive evolocumab (either 140 mg every 2 weeks or 420 mg monthly) or matching placebo as subcutaneous injections. The primary efficacy end point was the composite of cardiovascular death, myocardial infarction, stroke, hospitalization for unstable angina, or coronary revascularization. The key secondary efficacy end point was the composite of cardiovascular death, myocardial infarction, or stroke. The median duration of follow-up was 2.2 years. Purpose: The aim of the present work was to validate and optimize adiabatic T1ρ and T2ρ mapping for in vivo measurements of articular cartilage at 3 Tesla (T). Methods: Phantom and in vivo experiments were systematically performed on a 3T clinical system to evaluate the sequences using hyperbolic secant HS1 and HS4 pulses. R1ρ and R2ρ relaxation rates were studied as a function of agarose and chondroitin sulfate concentration and pulse duration. Optimal in vivo protocol was determined by imaging the articular cartilage of two volunteers and varying the sequence parameters, and successively applied in eight additional subjects. Reproducibility was assessed in phantoms and in vivo. Results: Relaxation rates depended on agarose and chondroitin sulfate concentration. The sequences were able to generate relaxation time maps with pulse lengths of 8 and 6 ms for HS1 and HS4, respectively. In vivo findings were in good agreement with the phantoms. The implemented adiabatic T1ρ and T2ρ sequences demonstrated regional variation in relaxation time maps of femorotibial cartilage. Reproducibility in phantoms and in vivo was good to excellent for both adiabatic T1ρ and T2ρ. Conclusions: The findings indicate that sequences are suitable for quantitative in vivo assessment of articular cartilage at 3 T.

Item does not contain fulltext, The degree to which genetic factors influence brain connectivity is beginning to be understood. Large-scale efforts are underway to map the profile of genetic effects in various brain regions. The NIH-funded Human Connectome Project (HCP) is providing data valuable for analyzing the degree of genetic influence underlying brain connectivity revealed by state-of-the-art neuroimaging methods. We calculated the heritability of the fractional anisotropy (FA) measure derived from diffusion tensor imaging (DTI) reconstruction in 481 HCP subjects (194/287 M/F) consisting of 57/60 pairs of mono- and dizygotic twins, and 246 siblings. FA measurements were derived using (Enhancing NeuroImaging Genetics through Meta-Analysis) ENIGMA DTI protocols and heritability estimates were calculated using the SOLAR-Eclipse imaging genetic analysis package. We compared heritability estimates derived from HCP data to those publicly available through the ENIGMA-DTI consortium, which were pooled together from five-family based studies across the US, Europe, and Australia. FA measurements from the HCP cohort for eleven major white matter tracts were highly heritable (h(2)=0.53-0.90, p<10(-5)), and were significantly correlated with the joint-analytical estimates from the ENIGMA cohort on the tract and voxel-wise levels. The similarity in regional heritability suggests that the additive genetic contribution to white matter microstructure is consistent across populations and imaging acquisition parameters. It also suggests that the overarching genetic influence provides an opportunity to define a common genetic search space for future gene-discovery studies. Uniquely, the measurements of additive genetic contribution performed in this study can be repeated using online genetic analysis tools provided by the HCP ConnectomeDB web application.

To examine the effects of the reconstruction algorithm of magnitude images from multichannel diffusion MRI on fiber orientation estimation.It is well established that the method used to combine signals from different coil elements in multichannel MRI can have an impact on the properties of the reconstructed magnitude image. Using a root-sum-of-squares approach results in a magnitude signal that follows an effective noncentral-χ distribution. As a result, the noise floor, the minimum measurable in the absence of any true signal, is elevated. This is particularly relevant for diffusion-weighted MRI, where the signal attenuation is of interest.In this study, we illustrate problems that such image reconstruction characteristics may cause in the estimation of fiber orientations, both for model-based and model-free approaches, when modern 32-channel coils are used. We further propose an alternative image reconstruction method that is based on sensitivity encoding (SENSE) and preserves the Rician nature of the single-channel, magnitude MR signal. We show that for the same k-space data, root-sum-of-squares can cause excessive overfitting and reduced precision in orientation estimation compared with the SENSE-based approach.These results highlight the importance of choosing the appropriate image reconstruction method for tractography studies that use multichannel receiver coils for diffusion MRI acquisition.

US$9.24 billion of this funding was specifically directed towards renewable energy-related activities, significantly increasing federal support for photovoltaic solar research and development, manufacturing capacity expansion, and deployment. But over US$100B was directed more broadly to the converging nexus of renewable energy, energy efficiency, green-house gas abatement, and smart grid sectors, collectively referred to as the “green economy.” In this paper we analyze the mechanisms by which this unprecedented federal investment in the energy economy of the future has been distributed both directly through Departments of Energy and Commerce R&D and economic development initiatives at the national level, and channeled by the Department of Labor into state-managed job-development programs. We discuss the economic development theories underlying this strategy, as well as its potential impact on the PV industry in the US.

- Abstract
BACKGROUND: The Whitehall II (WHII) study of British civil servants provides a unique source of longitudinal data to investigate key factors hypothesized to affect brain health and cognitive ageing. This paper introduces the multi-modal magnetic resonance imaging (MRI) protocol and cognitive assessment designed to investigate brain health in a random sample of 800 members of the WHII study. METHODS/DESIGN: A total of 6035 civil servants participated in the WHII Phase 11 clinical examination in 2012-2013. A random sample of these participants was included in a sub-study comprising an MRI brain scan, a detailed clinical and cognitive assessment, and collection of blood and buccal mucosal samples for the characterisation of immune function and associated measures. Data collection for this sub-study started in 2012 and will be completed by 2016. The participants, for whom social and health records have been collected since 1985, were between 60-85 years of age at the time the MRI study started. Here, we describe the pre-specified clinical and cognitive assessment protocols, the state-of-the-art MRI sequences and latest pipelines for analyses of this sub-study. DISCUSSION: The integration of cutting-edge MRI techniques, clinical and cognitive tests in combination with retrospective data on social, behavioural and biological variables during the preceding 25 years from a well-established longitudinal epidemiological study (WHII cohort) will provide a unique opportunity to examine brain structure and function in relation to age-related diseases and the modifiable and non-modifiable factors affecting resilience against and vulnerability to adverse brain changes.

46. Achromatopsia with amblyopia

- Abstract
39 patients suffering from congenital achromatopsia were investigated by various methods, including electroretinography, during both light and dark adaptation. This condition was typically expressed by the presence of photophobia, low visual acuity, nystagmus and various degrees of ametropia. The fundus was normal in 9 cases. Most of the other cases showed various macular or foveal abnormalities. 24 patients showed complete absence of color vision, and practically all displayed the ‘scotopic line’ in the Farnsworth Panel D-15 test. The electroretinogram (ERG) was almost always extinct in light. In the dark, in most patients the ERG displayed the scotopic mechanism solely, but some ERGs indicated subnormal photopic components either at the beginning or during all dark adaptation. This presence of photopic activity in the ERG of achromats was verified by 2 additional experiments. In one, the recovery of the positive wave of four achromats was compared on a percentage scale with that of four normal subjects and found to be similar, although the slightly faster course in achromats indicates less photopic activity than in normals. In the second, the positive amplitudes of the ERGs of 12 achromats with purely scotopic ERGs were recorded at completed dark adaptation as function of increasing stimulus intensities, all above the photopic threshold, and compared with those of 16 normal subjects. The amplitudes increased linearly with the 1.2 log intensity range in both groups, though the slope of the curve of the achromats was 1/4 that of the normals. In another experiment, the positive wave of the ERG, as elicited by light over 5 log units in the scotopic range, was found in an achromat to be of very similar shape as that of a normal, indicating scotopic acitivity to be similar in both subjects. The fact that, nevertheless, photopic components were not demonstrable in most ERGs, despite present photopic activity, can be explained by the relatively insensitive electrical method coupled with the subnormality of the retinal photopic mechanism in every achromat.

47. The ocular dominance of cortical neurons in cats developed with divergent and convergent squint

- Abstract
In five normal cats, 70.7% of the neurons recorded from the visual cortex responded to binocular stimulation; only 27.2% of the neurons in 12 cats raised with convergent squint and 63.6% of neurons in four cats with divergent squint displayed binocularity. In cats with convergent squint, 15% of the neurons showed normal receptive fields through one eye and nonconnected scattered receptive fields through the nondominant eye; however, movement detection, velocity discrimination and unidirectionality were retained. Furthermore, 65.2% of the monocular neurons were found to react through the normal eye and the rest through the squinting eye.

48. Receptive fields and firing patterns of sustained cells in the cat visual cortex

- Abstract
Ten out of 82 simple, complex and hypercomplex cells in areas 17 and 18 of the cat visual cortex responded with either rhythmic or nonrhythmic sustained firing patterns to movement stimulation. Sustained (X −) cells tended to show multimodal average response histograms with a dominant excitatory peak indicating organizational subdivision of their receptive fields. The dimensions of the excitatory centers ranged from 1.9 to 8.8°. Ocular dominance, orientation, directional selectivity and velocity discrimination were the same as known in regular cortical neurons. The differences between the responses to stationary and moving stimuli suggest that cells in the visual cortex can show either sustained or transient behavior, depending on the stimulus type.

50. C<INF>6</INF><SUP>-</SUP> Carbon Cluster Anion: An Infrared Absorption and Resonance Raman Isotopic Study

- Abstract
The Fourier transform infrared and resonance Raman spectra of the C
6 - carbon cluster anion has been measured in argon matrices. Fundamental symmetric stretching modes have been observed at 2086, 1775, and 634 cm-1, together with the overtone of a symmetric bending mode at 467 cm-1. The major asymmetric stretching mode lies at 1936.7 cm-1. Theoretical calculations utilizing Hartree−Fock (HF/6-31G*), Moller−Plesset perturbation (MP2/6-31G*), density functional (B3LYP/6-31G*), and quadratic configuration interaction QCISD/6-31G*, including triples QCISD(T)/6-31G*, support the assignment of these bands to the C6 - cluster and indicate that it is linear with an acetylenic-like structure. Frequency shifts determined from infrared and resonance Raman studies of 12C/13C isotopically substituted C6 - are generally in excellent agreement with the calculated isotopic frequency shifts, although not all isotopomeric bands were observed. The combination of resonance Raman and Fourier transform infrared spectroscopies with theoretical computations is shown to be a powerful approach for the study of carbon clusters and their anions.- Published
