Factor analysis has previously identified four independent factors that characterize the insulin resistance syndrome in women, interpreted as 1) weight/waist, 2) lipids, 3) insulin/glucose, and 4) systolic and diastolic blood pressure. Because it is not known whether similar factors emerge for men, or for diabetics, factor analysis was used to investigate the clustering of features characterizing the insulin resistance syndrome using data from 3,159 elderly (71-93 years) Japanese-American men participating in the fourth examination of the Honolulu Heart Program during 1991-1993. Consistent with previous results, factor analysis reduced eight risk factors (insulin, glucose, systolic blood pressure, diastolic blood pressure, triglycerides, high-density lipoprotein cholesterol, weight, and waist circumference) to four uncorrelated factors that explained 78.2% and 74.7% of the variance in nondiabetics (n = 2,760) and diabetics (n = 399), respectively. These factors were interpreted as 1) weight/waist, 2) blood pressure, 3) lipids, and 4) insulin/glucose. Modest differences in the associations between fasting insulin and factors 1, 3, and 4 were noted for diabetics. These consistently identified composite factors may represent markers for underlying pathophysiologic mechanisms of the insulin resistance syndrome and risk of non-insulin-dependent diabetes mellitus.

The value of research articles is increasingly contingent on complex data analysis results which substantiate their claims. Compared to data production, data analysis more readily lends itself to a higher standard of transparency and repeated operator-independent execution. This higher standard can be approached via fully reexecutable research outputs, which contain the entire instruction set for automatic end-to-end generation of an entire article from the earliest feasible provenance point. In this study, we make use of a peer-reviewed neuroimaging article which provides complete but fragile reexecution instructions, as a starting point to draft a new reexecution system which is both robust and portable. We render this system modular as a core design aspect, so that reexecutable article code, data, and environment specifications could potentially be substituted or adapted. In conjunction with this system, which forms the demonstrative product of this study, we detail the core challenges with full article reexecution and specify a number of best practices which permitted us to mitigate them. We further show how the capabilities of our system can subsequently be used to provide reproducibility assessments, both via simple statistical metrics and by visually highlighting divergent elements for human inspection. We argue that fully reexecutable articles are thus a feasible best practice, which can greatly enhance the understanding of data analysis variability and the trust in results. Lastly, we comment at length on the outlook for reexecutable research outputs and encourage re-use and derivation of the system produced herein.

The Oxford English Dictionary defines corruption as dishonest or illegal behaviour, especially of people in authority. Education is arguably one of the most important aspects of life as with such a lot of doors are opened for individuals, which are not offered to those without. Generation after generation the power and importance of education is only further highlighted, thus more and more children are pursuing higher education. As such, the number of students enrolled in schooling worldwide has been on an upwards trajectory, with an increase of over 150 million students since 2000 for secondary school and around 82 million since 2000 for primary school (Statista, 2022). Furthermore, the completion rate of primary school was close to 90% in 2019 and 76% for secondary school (Statista, 2022). Education can provide individuals with so many options and aid in their life path by increasing the aspects of stability in life, financial stability, equality between individuals worldwide, self-dependence, confidence, provides safety, and helps one reach their goals (Nair, 2022). In Canada itself, more than 1.4 million students are enrolled in a Canadian University, and 2.2 million students are enrolled in some form of postsecondary education (Universities Canada, 2018).

Many larger museums and archives have begun to implement a centralized approach to digitization of collections by creating Digitization Coordinator positions. This new effort has initiated a singular vision for digitization that incorporates priorities, workflows, and resources to greatly improve the efficiency and throughput of digitization in collections. Smaller institutions are now starting to see the benefit of creating a more structured cross-disciplinary approach to digitization, allowing for better awareness and resourcing of digitization needs.The workshop brought together natural sciences digitization professionals from the USA and EU, highlighting lessons learned and best practices to realize the benefits of a coordinated approach including advocacy for digitization, accelerating digitization efficiency and, ultimately, increasing digital collections access and usability to address societal challenges, such as biodiversity decline. Insights, lessons learned and initial thoughts on best practices are described, and the supporting workshop resources are shared so that others can benefit.

Canada’s history with the indigenous community is a long and complex. History which spills over to current events in society and impacts the Indigenously community greatly till this day. This country has a concerning Indigenous Education Gap—a disparity in educational achievement between Indigenous and non-Indigenous people. The Indigenous Education Gap is widening and rising quickly across Canada. Bridging the education gap is thus a critical component of any plan for enhancing the prosperity, health, and well-being of Canada's indigenous population, as well as eliminating marginalization. Education is one of the many concerns that Indigenous people in Canada must tackle. The acts of previous Canadian administrations, both implicit and explicit, have resulted in a considerable educational disparity between Indigenous and non-Indigenous pupils. The western system of education is inherently exclusive, and its fundamental educational process is based on privilege. Resolving the enormous and ongoing inequalities in student educational learning results must be based on Indigenous notions of education, rather than a standard western/colonial approach

Abstract Psilocybin and lysergic acid diethylamide (LSD) experiences can range from very positive to highly challenging (e.g., fear, grief, and paranoia). These challenging experiences contribute to hesitancy toward psychedelic-assisted psychotherapy among health care providers and patients. Co-use of 3,4-Methylenedioxy methamphetamine (MDMA) with psilocybin/LSD anecdotally reduces challenging experiences and enhances positive experiences associated with psilocybin/LSD. However, limited research has investigated the acute effects of co-use of MDMA and psilocybin/LSD. In a prospective convenience sample (N = 698) of individuals with plans to use psilocybin/LSD, we examined whether co-use of MDMA with psilocybin/LSD (n = 27) is associated with differences in challenging or positive experiences. Challenging experiences were measured using the Challenging Experiences Questionnaire and positive experiences were measured using the Mystical Experience Questionnaire and single-item measures of self-compassion, compassion, love, and gratitude. Potentially confounding variables were identified and included as covariates. Relative to psilocybin/LSD alone, co-use of psilocybin/LSD with a self-reported low (but not medium–high) dose of MDMA was associated with significantly less intense total challenging experiences, grief, and fear, as well as increased self-compassion, love and gratitude. Co-use of psilocybin/LSD and MDMA was not associated with differences in mystical-type experiences or compassion. Findings suggest co-use of MDMA with psilocybin/LSD may buffer against some aspects of challenging experiences and enhance certain positive experiences. Limitations include use of a convenience sample, small sample size, and non-experimental design. Additional studies (including controlled dose–response studies) that examine the effects and safety of co-administering MDMA with psilocybin/LSD (in healthy controls and clinical samples) are warranted and may assist the development of personalized treatments.

Enzyme immobilization offers a number of advantages that improve biocatalysis; however, finding a proper way to immobilize enzymes is often a challenging task. Implanting enzymes in metal–organic frameworks (MOFs) via co-crystallization, also known as biomineralization, provides enhanced reusability and stability with minimal perturbation and substrate selectivity to the enzyme. Currently, there are limited metal–ligand combinations with a proper protocol guiding the experimental procedures. We have recently explored 10 combinations that allow custom immobilization of enzymes according to enzyme stability and activity in different metals/ligands. Here, as a follow-up of that work, we present a protocol for how to carry out custom immobilization of enzymes using the available combinations of metal ions and ligands. Detailed procedures to prepare metal ions, ligands, and enzymes for their co-crystallization, together with characterization and assessment, are discussed. Precautions for each experimental step and result analysis are highlighted as well. Proteolysis is a critical biochemical process yet a challenging field to study experimentally due to the self-degradation of a protease and the complex, dynamic degradation steps of a substrate. Mass spectrometry (MS) is the traditional way for proteolytic studies, yet it is challenging when time-resolved, step-by-step details of the degradation process are needed. We recently found a way to resolve the cleavage site, preference/selectivity of cleavage regions, and proteolytic kinetics by combining site-directed spin labeling (SDSL) of protein substrate, time-resolved two-dimensional (2D) electron paramagnetic resonance (EPR) spectroscopy, protease immobilization via metal–organic materials (MOMs), and MS. The method has been demonstrated on a model substrate and protease, yet there is a lack of details on the practical operations to carry out our strategy. Thus, this protocol summarizes the key steps and considerations when carrying out the EPR/MS study on proteolytic processes, which can be generalized to study other protein/polypeptide substrates in proteolysis. Details for the experimental operation and cautions of each step are reported with figures illustrating the concepts. This protocol provides an effective approach to understanding the proteolytic process with the advantages of offering time-resolved, residue-level resolution of structural basis underlying the process. Such information is important for revealing the cleavage site and proteolytic mechanisms of unknown proteases. At least 200 billion black soldier fly (Hermetia illucens) larvae (BSFL) are reared each year as food and feed, and the insect farming industry is projected to grow rapidly. Despite interest by consumers, producers, and legislators, no empirical evidence exists to guide producers in practicing humane – or instantaneous – slaughter for these novel mini-livestock. BSFL may be slaughtered via freezing, boiling, grinding, or other methods; however standard operating procedures (SOPs) and equipment design may affect the likelihood of instantaneous death using these methods. We tested how larval body size and particle size plate hole diameter affect the likelihood of instantaneous death for black soldier fly larvae that are slaughtered using a standard meat grinder. Larval body size did not affect the likelihood of instantaneous death for larvae that are 106–175 mg in mass. However, particle size plate hole diameter had a significant effect on the likelihood of instantaneous death, with only 54% of larvae experiencing an instant death when using the largest particle size plate (12-mm hole diameter) compared to 84% using the smallest particle size plate (2.55 mm). However, a higher percentage of instantaneous death (up to 99%) could be achieved by reducing the proportion of larvae that become stuck in the machine. We conclude by outlining specific recommendations to support producers in achieving a 99% instantaneous death rate through specific SOPs to be used with similarly designed machines. We also develop a protocol for producers that wish to test their own grinding SOPs.

Abstract The use of gradient descent methods for optimizing k-eigenvalue nuclear systems has been shown to be useful in the past, but the use of k-eigenvalue gradients have proved computationally challenging due to their stochastic nature. ADAM is a gradient descent method that accounts for gradients with a stochastic nature. This analysis uses challenge problems constructed to verify if ADAM is a suitable tool to optimize k-eigenvalue nuclear systems. ADAM is able to successfully optimize nuclear systems using the gradients of k-eigenvalue problems despite their stochastic nature and uncertainty. Furthermore, it is clearly demonstrated that low-compute time, high-variance estimates of the gradient lead to better performance in the optimization challenge problems tested here.

Objective: Our objective is to develop a functional precision medicine platform designed to directly identify tailored drug regimens that target individual patient cancer cells and give benefit to the same donors by supporting clinical decision-making. We demonstrate our ex vivo drug sensitivity screening platform for precision medicine using Leukaemia and Multiple Myeloma samples from a South African patient cohort as proof of concept. Methodology: Through collaboration with Chris Hani Baragwanath and Donald Gordon Hospitals, Johannesburg, South Africa, we performed patient sample collections of n=80. Collected patient samples include Acute myeloid leukaemia (AML) (n=7), Chronic lymphocytic leukaemia (CLL) (n=4), Chronic myeloid leukaemia (CML) (n= 30), Multiple Myeloma (n=40) and health donor (n=5). For each patient sample, peripheral blood mononuclear cell (PBMC) isolation was performed and cryopreseved in liquid nitrogen. Results: Our preliminary demographic analysis results show that we can group patients based on diagnosis, staging, exclusion and inclusion criteria. From our demographic analysis, we have also identified highly frequent chemotherapy drugs used in the cohort. Further, we can identify the most frequent chemotherapy drugs given as medication to the patient cohort. We then selected 30 drugs that are relevant for leukemia and multiple myeloma for Ex vivo drug sensitivity screening test. Conclusion: Using our results we will then select effective drugs for monotherapy and also drug combinations. Selected drug combinations will then be validated on patient samples using our ex vivo drug sensitivity test. These results will be analyzed using our statistical capabilities and developed as a packaged product of preclinical information for precision clinical trials. Thus, we are progressing our cutting-edge translational platform from technology readiness level (TRL4) to TRL6 on blood cancer.

Abstract Premise We recognized the need for a customized imaging protocol for plant specimens at the time of collection for the purpose of three‐dimensional (3D) modeling, as well as the lack of a broadly applicable photogrammetry protocol that encompasses the heterogeneity of plant specimen geometries and the challenges introduced by processes such as wilting. Methods and Results We developed an equipment list and set of detailed protocols describing how to capture images of plant specimens in the field prior to their deformation (e.g., with pressing) and how to produce a 3D model from the image sets in Agisoft Metashape Professional. Conclusions The equipment list and protocols represent a foundation on which additional improvements can be made for specimen geometries outside of the range of the six types considered, and an easy entry into photogrammetry for those who have not previously used it.

Interferometric synthetic aperture radar (InSAR) processing techniques have been widely used to derive surface deformation or retrieve terrain elevation. Over the development of the past few decades, most research has mainly focused on its application, new techniques for improved accuracy, or the investigation of a particular error source and its correction method. Therefore, a thorough discussion about each error source and its influence on InSAR-derived products is rarely addressed. Additionally, InSAR is a challenging topic for beginners to learn due to the intricate mathematics and the necessary signal processing knowledge required to grasp the core concepts. This results in the fact that existing papers about InSAR are easy to understand for those with a technical background but difficult for those without. To cope with the two issues, this paper aims to provide an organized, comprehensive, and easily understandable review of the InSAR error budget. In order to assist readers of various backgrounds in comprehending the concepts, we describe the error sources in plain language, use the most fundamental math, offer clear examples, and exhibit numerical and visual comparisons. In this paper, InSAR-related errors are categorized as intrinsic height errors or location-induced errors. Intrinsic height errors are further divided into two subcategories (i.e., systematic and random error). These errors can result in an incorrect number of phase fringes and introduce unwanted phase noise into the output interferograms, respectively. Location-induced errors are the projection errors caused by the slant-ranging attribute of the SAR systems and include foreshortening, layover, and shadow effects. The main focus of this work is on systematic and random error, as well as their effects on InSAR-derived topographic and deformation products. Furthermore, because the effects of systematic and random errors are greatly dependent on radar wavelengths, different bands are utilized for comparison, including L-band, S-band, C-band, and X-band scenarios. As examples, we used the parameters of the upcoming NISAR operation to represent L-band and S-band, ERS-1 and Sentinel-1 to represent C-band, and TerraSAR-X to represent X-band. This paper seeks to bridge this knowledge gap by presenting an approachable exploration of InSAR error sources and their implications. This robust and accessible analysis of the InSAR error budget is especially pertinent as more SAR data products are made available (e.g., NISAR, ICEYE, Capella, Umbra, etc.) and the SAR user-base continues to expand. Finally, a commentary is offered to explore the error sources that were not included in this work, as well as to present our thoughts and conclusions.

The relationship between new greenspaces and gentrification is an important one for urbanization. Here the authors show a positive relationship for at least one decade between greening in the 1990s–2000s and gentrification that occurred between 2000–2016 in 17 of 28 studied cities in North America and Europe.

This article describes the process of designing and running a new course on Experimental Archaeology and Experiential History at a small liberal arts college in central Minnesota. We discuss the general methodological and pedagogical goals for the course, a representative three-week sequence of readings and labs based on the lives of shepherds, and the pedagogical and digital infrastructure we have developed. We hope this paper leads to discussion and collaboration with people teaching similar courses, and also helps people get their own courses of this type started.

ObjectiveFocal intimal flaps (FIF) are a variety of defects of the aorta that result in a short, flap-like projection into the lumen, and are often encountered in asymptomatic patients undergoing computed tomography angiography (CTA) surveillance for aortic aneurysm, but the natural history and clinical significance of such lesions has not yet been studied.MethodsWe retrospectively identified patients with an asymptomatic FIF and available imaging follow-up (>1 year). FIF was defined as flap-like intimal irregularity < 4 cm in length involving the thoracic aorta (TA), abdominal aorta (AA) or common iliac arteries (CIA). FIF characteristics included length and circumferential extent as well as the presence and size (width and depth) of associated penetrating aortic ulcers (PAUs). Patient characteristics, adverse events and history of surgical repair was determined by chart review. FIFs and associated PAUs were assessed for progression by comparing baseline and follow-up CTA studies.ResultsA total of 84 FIFs were identified in 77 patients. Average age was 69.2 ± 10.1 years, and 81% were male (81%). Common co-morbidities included: hypertension (78%), hyperlipidemia (68%), smoking (60%), coronary artery disease (41%), aortic aneurysm (34%), type II diabetes mellitus (27%) and prior cardiovascular surgery (25%). FIFs were most commonly located in the abdominal aorta (n = 50, 60%). Nearly all FIFs were associated with local atherosclerotic plaque (93%). Mean follow-up interval was 3.5 ± 2.6 years (259 cumulative follow-up years). Change in FIF length and local aortic diameter over follow-up were 0.7 ± 2.3 mm and 0.8 ± 1.1 mm, respectively. Nearly half (47%) of FIFs were associated with penetrating aortic ulcers (PAU) with baseline depth of 7.3 mm (IQR: 6.1–10.2) and change in depth of 0.5 ± 1.4 mm. Only 12% of FIFs and 0% of associated PAUs demonstrated growth (≥3 mm) at follow-up. No acute pathology developed in the location of FIFs and no aortic interventions were performed specifically to treat FIFs.ConclusionFocal intimal flaps identified in asymptomatic patients with aortic disease were co-localized with atherosclerotic plaque and PAUs, and demonstrated indolent behavior, not leading to significant growth or acute aortic events, supporting a conservative management approach.

On-demand microtransit services are frequently seen as an important tool in supporting first and last mile operations surrounding fixed route high frequency transit facilities, but questions remain surrounding who will use these novel services and for what purposes. In November 2019, the Utah Transit Authority launched an on-demand microtransit service in south Salt Lake County in partnership with a private mobility operator. This paper reports the results of an expressed preferences survey of 130 transit riders in the microtransit service area that was collected before and immediately after the service launched. There is not a clear relationship between current transit access mode and expressed willingness to use microtransit, although some responses from new riders indicate the novel service competes most directly with commercial transportation network company operations. The survey responses also reveal younger passengers express a more than expected willingness to use microtransit, middle-aged passengers a less than expected willingness, and older passengers neutral or no expressed opinion. The results suggest additional relationships between household size and transit use frequency, but further research is necessary. The effect of other user characteristics, including income and automobile availability, is less statistically clear and requires further research.

ObjectiveTo evaluate the evidence for altered cortical and spinal cord functions in individuals with patellofemoral pain (PFP).MethodsWe conducted a comprehensive search of databases to appraise and analyze the studies published prior to December 10, 2021 that examined spinal reflex excitability measured using Hoffmann reflex (H-reflex) amplitudes, corticospinal excitability measured using transcranial magnetic stimulation (TMS)-elicited motor evoked potential (MEP) amplitudes, motor threshold (MT), or stimulus-response (SR) curves, cortical reorganization assessed using TMS cortical mapping or structural magnetic resonance imaging (MRI), or functional changes of the brain assessed using functional MRI (fMRI) in individuals with PFP.ResultsEight studies were eligible for analyses. While an earlier study showed that pain had no effect on the H-reflex amplitude of the quadriceps muscle, more recent evidence reported a decrease in vastus medialis (VM) H-reflex amplitude in participants with PFP. VM H-reflex amplitude was correlated with pain, chronicity, physical function, and isometric knee extensor torque production in participants with PFP. Altered corticospinal excitability was reported in participants with PFP, observed as increased MT in the VM and vastus lateralis (VL) muscles. In addition, cortical reorganization has been observed, where decreased number of cortical peaks, shifts and reduced volumes, and increased overlap of motor cortex representations for the VM, VL, and rectus femoris (RF) muscles were reported in participants with PFP.ConclusionThere is emerging evidence on altered cortical and spinal cord functions in individuals with PFP, however, solid conclusions cannot be drawn due to limited literature available. Abstract In this commentary, we strive to illustrate common misconceptions of the dopamine fasting fad that has become popular among wellness enthusiasts and purported by health gurus. Here, we review the proposed Dopamine fasting technique for managing behavioral addictions as proposed by California psychiatrist Dr. Cameron Sepah. We first summarize correct and incorrect interpretations of what Dopamine fasting involves. Next, we contextualize the role of dopamine as it relates to behavioral modification interventions for addiction. Particularly, we discuss the role of dopamine in behavioral addiction and the effectiveness of cognitive behavioral therapy (CBT) techniques for various addictions which are the basis of the proposed dopamine fasting technique. While we see potential for dopamine fasting to offer significant benefits to individuals, we highlight the limitation of the self‐guided aspect of dopamine fasting, which could pose physical and emotional harm to individuals if the guideline is misinterpreted or misused as the sole treatment for severe disorders which require clinician input. Future studies should aim to assess not only the scientific efficacy of dopamine fasting as a potential treatment approach for behavioral addiction, but also the needs and well‐being of individuals who seek self‐directed treatment from popular media trends.

In 2020, we are facing unprecedented times, and as some form of lockdown continues with no signs of ending feelings of hopelessness are completely natural and understandable. Unprecedented times does not mean that these current issues and struggles have never been faced by humanity before, however. The Spanish Flu which took place after World War One and the Black Death that was rampant in Asia and Europe in the 14th century quickly come to mind as examples of past pandemics, but these are only two examples of devastating diseases throughout human history. The Plague of Athens that was raging during the beginning of the Peloponnesian War in 430 BCE is another such example. Though removed from our current situation by many centuries, its symptoms and the effects it had on the population of Athens have been meticulously recorded by the general and historian Thucydides, giving us the opportunity to compare his account to our own experiences today. The disease may be different, and the image he portrays may be more violent and desperate than our own, but nonetheless we can see similarities in how these two separate societies have reacted to unforeseen hardships. In this comparison, we can come to understand at once our own good fortune at going through a pandemic with the support of modern technology and medicine as well as how universal our reactions are to this type of suffering, thereby making it natural rather than shameful. Humanity has faced a great deal of diversity before, and COVID-19 will likely prove to be no different.

Introduction Reduced neuromuscular control due to altered neurophysiological functions of the central nervous system has been suggested to cause movement deficits in individuals with patellofemoral pain (PFP). However, the underlying neurophysiological measures of brain and spinal cord in this population remain to be poorly understood. The purpose of this systematic review is to evaluate the evidence for altered cortical and spinal cord functions in individuals with PFP.Methods and analysis The protocol for conducting the review was prepared using the Preferred Reporting Items for Systematic Review and Meta-Analysis Protocols guidelines. We will systematically search the literature that examines cortical and spinal cord functions in individuals with PFP, aged 18–45 years. The studies for cross-sectional, prospective, longitudinal, case–control and randomised control trial designs will be included from the following databases: PubMed (MEDLINE), EMBASE and Web of Science. Only studies published in English prior to 1 February 2021 will be included. The risk of bias and quality assessment will be performed using National Institutes of Health’s Quality Assessment Tool for Observational Cohort and Cross-Sectional Studies. We will conduct meta-analysis of the data where appropriate. Narrative synthesis will be taken if a meta-analysis is not possible.Ethics and dissemination This is a systematic review from the existing literature and does not require ethical approval. The results of this study will be published in a peer-reviewed journal in the field of rehabilitation medicine, sports/orthopaedic medicine or neurology, regardless of the outcome.PROSPERO registration number CRD42020212128.

Understanding the Tibetan Plateau’s role in environmental change has gained increasing scientific attention in light of warming and changes in land management. We examine changes in greenness over the Tibetan Plateau using the Normalized Difference Vegetation Index (NDVI) from the Global Inventory Monitoring and Modeling Study (GIMMS3g) to identify significant changes over the entire plateau, six ecoregions, and protected areas based on a multiyear time series of July imagery from 1982 to 2015. We also test whether there have been changes in human populations in protected areas. There has been relatively little change in mean NDVI over the Tibetan Plateau or ecoregions, however, there were significant changes at the pixel level. There are sixty-nine protected areas on the Tibetan Plateau; sixty-two protected areas had no significant change in mean NDVI and seven protected areas experienced a significant increase in NDVI. There has been an increase in population within protected areas from 2000 to 2015; however, mean populations significantly increased in two protected areas and significantly decreased in four protected areas. Results suggest a slow greening of the Tibetan Plateau, ecoregions, and protected areas, with a more rapid greening in northern Tibet at the pixel level. Most protected areas are experiencing minor changes in NDVI independent of human population.

Injuries involving the anterior cruciate ligament are among the most common athletic injuries, and are the most common involving the knee. The anterior cruciate ligament is a key translational and rotational stabilizer of the knee joint during pivoting and cutting activities. Traditionally, surgical intervention in the form of anterior cruciate ligament reconstruction has been recommended for those who sustain an anterior cruciate ligament rupture and wish to remain active and return to sport. The intra-articular environment of the anterior cruciate ligament makes achieving successful healing following repair challenging. Historically, results following repair were poor, and anterior cruciate ligament reconstruction emerged as the gold-standard for treatment. While earlier literature reported high rates of return to play, the results of more recent studies with longer follow-up have suggested that anterior cruciate ligament reconstruction may not be as successful as once thought: fewer athletes are able to return to sport at their preinjury level, and many still go on to develop osteoarthritis of the knee at a relatively younger age. The four principles of tissue engineering (cells, growth factors, scaffolds, and mechanical stimuli) combined in various methods of bioaugmentation have been increasingly explored in an effort to improve outcomes following surgical treatment of anterior cruciate ligament injuries. Newer technologies have also led to the re-emergence of anterior cruciate ligament repair as an option for select patients. The different biological challenges associated with anterior cruciate ligament repair and reconstruction each present unique opportunities for targeted bioaugmentation strategies that may eventually lead to better outcomes with better return-to-play rates and fewer revisions.

Although cities globally are increasingly mobilizing re-naturing projects to address diverse urban socio-environmental and health challenges, there is mounting evidence that these interventions may also be linked to the phenomenon known as green gentrification. However, to date the empirical evidence on the relationship between greenspaces and gentrification regarding associations with different greenspace types remains scarce. This study focused on 28 mid-sized cities in North America and Western Europe. We assessed improved access to different types of greenspace (i.e. total area of parks, gardens, nature preserves, recreational areas or greenways [i] added before the 2000s or [ii] added before the 2010s) and gentrification processes (including [i] gentrification for the 2000s; [ii] gentrification for the 2010s; [iii] gentrification throughout the decades of the 2000s and 2010s) in each small geographical unit of each city. To estimate the associations, we developed a Bayesian hierarchical spatial model for each city and gentrification time period (i.e. a maximum of three models per city). More than half of our models showed that parks—together with other factors such as proximity to the city center—are positively associated with gentrification processes, particularly in the US context, except in historically Black disinvested postindustrial cities with lots of vacant land. We also find than in half of our models newly designated nature preserves are negatively associated with gentrification processes, particularly when considering gentrification throughout the 2000s and the 2010s and in the US. Meanwhile, for new gardens, recreational spaces and greenways, our research shows mixed results (some positive, some negative and some no effect associations). Considering the environmental and health benefits of urban re-naturing projects, cities should keep investing in improving park access while simultaneously implementing anti-displacement and inclusive green policies.

Of the approximately 6700 lakes and reservoirs larger than 1 km2 in the Contiguous United States (CONUS), only ~430 (~6%) are actively gaged by the United States Geological Survey (USGS) or their partners and are available for download through the National Water Information System database. Remote sensing analysis provides a means to fill in these data gaps in order to glean a better understanding of the spatiotemporal water level changes across the CONUS. This study takes advantage of two-plus years of NASA’s ICESat-2 (IS-2) ATLAS photon data (ATL03 products) in order to derive water level changes for ~6200 overlapping lakes and reservoirs (>1 km2) in the CONUS. Interactive visualizations of large spatial datasets are becoming more commonplace as data volumes for new Earth observing sensors have markedly increased in recent years. We present such a visualization created from an automated cluster computing workflow that utilizes tens of billions of ATLAS photons which derives water level changes for all of the overlapping lakes and reservoirs in the CONUS. Furthermore, users of this interactive website can download segmented and clustered IS-2 ATL03 photons for each individual waterbody so that they may run their own analysis. We examine ~19,000 IS-2 derived water level changes that are spatially and temporally coincident with water level changes from USGS gages and find high agreement with our results as compared to the in situ gage data. The mean squared error (MSE) and the mean absolute error (MAE) between these two products are 1 cm and 6 cm, respectively.

Addressing seasonal water uncertainties and increased power generation demand has sparked a global rise in large-scale hydropower projects. To this end, the Blue Nile impoundment behind the Grand Ethiopian Renaissance Dam (GERD) will encompass an areal extent of ~1763.3 km2 and hold ~67.37 Gt (km3) of water with maximum seasonal load changes of ~27.93 (41% of total)—~36.46 Gt (54% of total) during projected operational scenarios. Five different digital surface models (DSMs) are compared to spatially overlapping spaceborne altimeter products and hydrologic loads for the GERD are derived from the DSM with the least absolute elevation difference. The elastic responses to several filling and operational strategies for the GERD are modeled using a spherically symmetric, non-rotating, elastic, and isotropic (SNREI) Earth model. The maximum vertical and horizontal flexural responses from the full GERD impoundment are estimated to be 11.99 and 1.99 cm, regardless of the full impoundment period length. The vertical and horizontal displacements from the highest amplitude seasonal reservoir operational scenarios are 38–55% and 34–48% of the full deformation, respectively. The timing and rate of reservoir inflow and outflow affects the hydrologic load density on the Earth’s surface, and, as such, affects not only the total elastic response but also the distance that the deformation extends from the reservoir’s body. The magnitudes of the hydrologic-induced deformation are directly related to the size and timing of reservoir fluxes, and an increased knowledge of the extent and magnitude of this deformation provides meaningful information to stakeholders to better understand the effects from many different impoundment and operational strategies.

We report the design of a targeted resequencing panel for monogenic dyslipidemias, LipidSeq, for the purpose of replacing Sanger sequencing in the clinical detection of dyslipidemia-causing variants. We also evaluate the performance of the LipidSeq approach versus Sanger sequencing in 84 patients with a range of phenotypes including extreme blood lipid concentrations as well as additional dyslipidemias and related metabolic disorders. The panel performs well, with high concordance (95.2%) in samples with known mutations based on Sanger sequencing and a high detection rate (57.9%) of mutations likely to be causative for disease in samples not previously sequenced. Clinical implementation of LipidSeq has the potential to aid in the molecular diagnosis of patients with monogenic dyslipidemias with a high degree of speed and accuracy and at lower cost than either Sanger sequencing or whole exome sequencing. Furthermore, LipidSeq will help to provide a more focused picture of monogenic and polygenic contributors that underlie dyslipidemia while excluding the discovery of incidental pathogenic clinically actionable variants in nonmetabolism-related genes, such as oncogenes, that would otherwise be identified by a whole exome approach, thus minimizing potential ethical issues.

Turbulent plasma edge density fluctuations can broaden a traversing microwave beam degrading its quality. This can be a problem for scenarios relying on a high spatial localization of the deposition of injected microwave power, like controlling MHD instabilities. Here we present numerical estimations of the scattering of a microwave beam by density fluctuations over a large parameter range, including extrapolations to ITER. Two codes are used, the full-wave code IPF-FDMC and the wave kinetic equation solver WKBeam. A successful comparison between beam broadening obtained from DIII-D experiments and corresponding full-wave simulations is shown.

The Slumgullion landslide, located in southwestern Colorado, has been active since the early 1700s and current data suggests that the most active portion of the slide creeps at a rate of ~1.5⁻2.0 cm/day. Accurate deformation measurement techniques are vital to the understanding of persistent, yet slow-moving landslides like the Slumgullion. The factors that affect slope movements at the Slumgullion are on-time scales that are well suited towards a remotely sensed approach to constrain the 12 different kinematic units that make up the persistent creeping landslide. We derive a time series of motion vectors (magnitude and direction) using subpixel offset techniques from very high resolution TerraSAR-X Staring Spotlight ascending/descending data as well as from a novel high-resolution amalgamation of airborne lidar and unmanned aerial systems (UAS) Structure from Motion (SfM) digital surface model (DSM) hillshades. Deformation rates calculated from the spaceborne and airborne datasets show high agreement (mean difference of ~0.9 mm/day), further highlighting the potential for the monitoring of ongoing mass wasting events utilizing unmanned aircraft systems (UAS) We compare pixel offset results from an 11-day synthetic aperture radar (SAR) pair acquired in July of 2016 with motion vectors from a coincident low-cost L1 only Global Navigation Satellite System (GNSS) field campaign in order to verify the remotely sensed results and to derive the accuracy of the azimuth and range offsets. We find that the average azimuth and range pixel offset accuracies utilizing the methods herein are on the order of 1/18 and 1/20 of their along-track and slant range focused ground pixel spacing values of 16.8 cm and 45.5 cm, respectively. We utilize the SAR offset time series to add a twelfth kinematic unit to the previously established set of eleven unique regions at the site of an established minislide within the main landslide itself. Lastly, we compare the calculated rates and direction from all spaceborne- and airborne-derived motion vectors for each of the established kinematic zones within the active portion of the landslide. These comparisons show an overall increased magnitude and across-track component (i.e., more westerly angles of motion) for the descending SAR data as compared to their ascending counterparts. The processing techniques and subsequent results herein provide for an improved knowledge of the Slumgullion landslide’s kinematics and this increased knowledge has implications for the advancement of measurement techniques and the understanding of globally distributed creeping landslides.

This paper describes and illustrates five major clusters of related tasks (herein referred to as task clusters) that are common to efficient and effective practices in the digitization of biological specimen data and media. Examples of these clusters come from the observation of diverse digitization processes. The staff of iDigBio (The U.S. National Science Foundation’s National Resource for Advancing Digitization of Biological Collections) visited active biological and paleontological collections digitization programs for the purpose of documenting and assessing current digitization practices and tools. These observations identified five task clusters that comprise the digitization process leading up to data publication: (1) pre-digitization curation and staging, (2) specimen image capture, (3) specimen image processing, (4) electronic data capture, and (5) georeferencing locality descriptions. While not all institutions are completing each of these task clusters for each specimen, these clusters describe a composite picture of digitization of biological and paleontological specimens across the programs that were observed. We describe these clusters, three workflow patterns that dominate the implemention of these clusters, and offer a set of workflow recommendations for digitization programs.

Several new protective systems are being installed on the DIII D tokamak to increase the safety margins for plasma operations with injected ECH power at densities approaching cutoff. Inadvertent overdense operation has previously resulted in reflection of an rf beam back into a launcher causing extensive arcing and melt damage on one waveguide line. Damage to microwave diagnostics, which are located on the same side of the tokamak as the ECH launchers, also has occurred. Developing a reliable microwave based interlock to protect the many vulnerable systems in DIII-D has proved to be difficult. Therefore, multiple protective steps have been taken to reduce the risk of damage in the future. Among these is a density interlock generated by the plasma control system, with setpoint determined by the ECH operators based on rf beam trajectories and plasma parameters. Also installed are enhanced video monitoring of the launchers, and an ambient light monitor on each of the waveguide systems, along with a Langmuir probe at the mouth of each launcher. Versatile rf monitors, measuring forward and reflected power in addition to the mode content of the rf beams, have been installed as the last miter bends in each waveguide line. As these systems are characterized, they are being incorporated in the interlock chains, which enable the ECH injection permits. The diagnostics most susceptible to damage from the ECH waves have also been fitted with a variety of protective devices including stripline filters, thin resonant notch filters tuned to the 110 GHz injected microwave frequency, blazed grating filters and shutters. Calculations of rf beam trajectories in the plasmas are performed using the TORAY ray tracing code with input from kinetic profile diagnostics. Using these calculations, strike points for refracted beams on the vacuum vessel are calculated, which allows evaluation of the risk of damage to sensitive diagnostics and hardware.

In this methodological review, we conducted a quantitative content analysis of a random sample of 107 education-related doctoral dissertations published in Proquest Dissertations and Theses database in 2011. Seven raters coded each article in terms of page lengths (overall and within each chapter), research method (qualitative, quantitative, or mixed-methods), author gender, and university characteristics (online or traditional). We found that the median education dissertation length was 161 pages long, but those page lengths differed between research methods. The median page lengths of qualitative, mixed method, and quantitative dissertations were 210, 187, and 147 respectively. The median page length of education dissertations from online universities was 44 pages shorter than education dissertations from their traditional counterparts. Contrary to previous research, we found no statistically significant relationship between gender and methods choice.
