7. Phenolic content and oxidative stability of chocolates produced with roasted and unroasted cocoa beans.

Abstract

The aim of this study was to produce chocolate using roasted (RB) and unroasted cocoa beans (URB). The effect of roasting on the total phenolic content (TPC), antioxidant activity [2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, ferric reducing antioxidant power (FRAP), and cupric ion reducing antioxidant capacity (CUPRAC) values], phenolic compounds, caffeine, oxidative stability [free fatty acid, peroxide, conjugated dienes, conjugated trienes, and thiobarbituric acid reactive substances (TBARS)], Fourier transform infrared (FTIR), and differential scanning colorimetry (DSC) analysis of both cocoa beans and chocolate samples were analyzed. According to the results, the TPC of URB (24.96 mg gallic acid equivalent (GAE)/g sample) was higher than roasted beans (21.32 mg GAE/g sample). Similar results were also seen in the TPC of chocolate samples. Although roasting did not affect the DPPH scavenging activity and caffeine content of cocoa beans, it decreased FRAP and CUPRAC values. (-)-Epicatechin and chlorogenic acid values were higher in unroasted bean and chocolate samples, but the amount of gallic acid increased with the roasting process. Free fatty acid, peroxide, conjugated dienes, conjugated trienes, and TBARS results of unroasted samples were lower than roasted ones, indicating better oxidative stability. The melting temperatures of cocoa beans changed with roasting while it was similar between chocolate samples. 11. Modeling thermal inactivation of Salmonella Enteritidis in ground chicken supplemented with a mixture of ribwort, sorrel, and yarrow extracts.

Abstract

Thermal death kinetics of Salmonella Enteritidis in ground chicken in the presence of ribwort, sorrel and yarrow extracts mixture (1%) was investigated by fitting the inactivation curves to the log–linear and Weibull models. The thermal inactivation curves were fitted well to both models (r2 ≥ 0.94). The times required for 4–log reduction in the S. Enteritidis population were shortened by 47–64% in ground chicken supplemented with the herbal extract mixture (p < 0.05). The 4–log reduction times calculated using the Weibull model were 0.13–13.6% longer than those extrapolated with the log–linear model. The z values in the ground chicken samples with and without herbal extract were 5.08°C (r2 = 0.97) and 5.57°C (r2= 0.94), respectively. The results suggest that the mixture of these extracts increased the thermal sensitivity of S. Enteritidis significantly at tested temperatures. In addition, using the Weibull model to calculate thermal death kinetics may reduce the underestimation of the thermal reduction times. Practical applications: This research aimed to investigate the effects of an herbal extract mixture consisting of ribwort, sorrel, and yarrow on the thermal resistance of Salmonella Enteritidis in ground chicken. While these plants are recognized for their antimicrobial and antioxidant properties, there are not many studies conducted to explore their effects on foodborne pathogens in the food matrix. As far as we know, this is the first study that examines the impact of these plants on the thermal resistance of S. Enteritidis in ground chicken. We determined the thermal death kinetics of S. Enteritidis in ground chicken with and without the herbal extract mixture using curve fitting to log‐linear and Weibull models. 13. One-pot approach biosynthesis of nisin-conjugated zinc oxide nanoparticles using Bacillus subtilisZBP4 with improved antibacterial activity

Abstract

Biosynthesis of nisin-conjugated zinc oxide nanoparticles (N-ZnONPs) using the supernatant of Bacillus subtilisZBP4 was demonstrated using a single-step reaction. Various amounts of nisin (5, 10, 15 mg/mL) and 8 mM ZnSO4.7H2O were reacted in the cell-free supernatant of Bacillus subtilisZBP4 at 33 °C for 48 h. Surface plasmon resonance peaks created by N-ZnONPs were measured at 341–342 nm with a UV-VIS spectrophotometer, and the band-gap energy was calculated as 3.38 eV, indicating the nano-conjugates acted as semiconductors. Transmission electron microscopy (TEM) images showed that the N-ZnONPs were pseudo-spherical with an average diameter of 23 nm. Fourier transform infrared (FTIR) spectra analysis confirmed the presence of nisin as well as other organic substances in the structure. N-ZnONPs had potent antimicrobial activity on Escherichia coliO157:H7 and Staphylococcus aureus; the minimum inhibition concentrations were 0.05 and 1 mg/mL, respectively. In addition, conjugation with nisin increased the long-term stability of nanoparticles when compared to unconjugated ZnONPs.

Abstract

Typha domingensis pollen is obtained from staminate flowers of plant that is edible. In this study, bioactive compounds from pollen were extracted using ethanol and methanol with conventional method. Further ultrasonication was applied to the samples to improve extraction efficiency. The extraction yield was higher with ethanol (26.3±0.14%) than with methanol (25.4±0.3%) and applying ultrasonication increased the yields significantly (P <0.05). Total phenolic compounds in ethanol and methanol extracts were determined as 9.83±0.48 and 9.71±0.55 mg GAE/g dry matter with the conventional method, and 11.76±0.64 and 12.74±0.37 mg GAE/g dry matter after ultrasonication, respectively. The flavonoid content with ethanol extraction was significantly higher than with methanol in both conventional and ultrasonication methods (P <0.05). Antioxidant activities using DPPH, ABTS, CUPRAC, and FRAP tests, were determined, and enhanced antioxidant capacities were observed after ultrasonication. The bioactive compounds were qualitatively analyzed using UV-VIS spectroscopy and FTIR which confirmed the presence of polyphenols. [ABSTRACT FROM AUTHOR]

21. Green synthesis of zinc oxide nanoparticles using Bacillus subtilis ZBP4 and their antibacterial potential against foodborne pathogens.

Abstract

In this study, extracellular biosynthesis of zinc oxide nanoparticles (ZnO NPs) by using the supernatant of Bacillus subtilis ZBP4 after cultivation in nutrient broth at 33 °C for 24 h was investigated. Zinc sulfate heptahydrate was used as the precursor, and the reactions were performed at 33 °C for 72 h. The effects of pH 5–9 and precursor concentration (2–10 mM) were determined and the optima were found to be pH 7.5 and 8 mM ZnSO4·7H2O, respectively. The nanoparticles were characterized by UV-VIS spectroscopy, FESEM, TEM, EDS, XRD, FTIR and zeta potential measurement. ZnO NPs appeared to be irregular spherical structures with a size range of 22–59 nm, as confirmed by FESEM and TEM. Energy-dispersive spectroscopy analysis validated the formation of ZnO NPs. X-ray diffraction analysis revealed the crystalline structure of the ZnO NPs, and they were determined to have a zeta potential of −19.0 ± 4.3 mV and a bandgap of 3.36 eV. Antibacterial activity experiments showed that ZnO NPs are effective against a broad spectrum of Gram-positive and Gram-negative food pathogens. This study provides evidence for a safe and effective method for synthesizing ZnO NPs and demonstrates their effectiveness against pathogenic bacteria. [ABSTRACT FROM AUTHOR]

Abstract

Çocukların dünyayı algılayışları yetişkinlerden farklıdır. Daha doğal, kendiliğinden hareket eder ve daha basit düşünürler. Bu davranışlar yaptıkları resimlere de yansır. Tamamen içgüdüsel olarak çizdikleri için çalışmaları incelendiğinde onlar hakkında bazı fikirlere ulaşılabilir. Birey doğduğu andan itibaren gelişim içerisindedir. Bulundukları ortama göre bedensel ve zihinsel gelişimlerine paralel olarak sanatsal faaliyetlerde de gelişme gösterirler. Çocukların çizimleri basitten karmaşığa doğru ilerler ve yaş aralıklarına göre bazı aşamalardan geçer. Karalama ile başlayan dönem, kaba motor becerilerin ilerlemesiyle bir şema haline gelir. Bir süre çocuk gördüğünü değil, bildiğini yani zihnindeki şemaları çizer. Gelişimin ilerlemesi ile çocuk çevresinde olup bitenlerin daha çok farkına varmaya başlar ve bunu resimlerine yansıtmaya başlar. Resimler artık daha detaylı ve gerçekçidir. Bu dönemde gördüklerini yansıtma kaygısı taşıdığı için ilk dönemdeki özgür tavrından uzaklaşırlar. Son gelişim dönemine gelindiğinde çocuğun imgesel düşünme yetisi artar. Duygusal değişimlerin ve cinsel kimliğin etkileri resimlerde görülmeye başlar. Çocuklar bilinçli ya da bilinçsiz yaptıkları desenler ve şekiller kağıt üzerinde anlam kazanabilir. Çizilen sembollerin anlamları dikkate alınarak yapılan yorumlar sonucunda çocuk hakkında önemli bilgilere ulaşılabilmektedir. İletişim insanoğlunun var oluşundan itibaren en temel ihtiyaçlarından biridir. İnsanlığın gelişmesiyle birlikte, kullanılan iletişim araçları da çeşitlilik göstermiştir. Children's drawings progress from simple to complex, and passt hrough somestages according to ageranges. The period that starts with scribbling becomes a schema with the progression of gross motor skills. For a while, the child draws not what he sees, but what he knows, that is, the schemas in his mind. With the progress of development, the child begins to become more aware of what is happening around himand begins to reflectthis on his paintings. Pictures are now more detailed and realistic. Since he is concerned about reflecting what he sees in thisperiod, he moves away from his free attitude in the first period. When it comes to the final developmental period, the child's imaginative thinking ability develops well, and the effects of emotional changes and sexualidentity begin to be seen in the pictures. As children draw randomly, the patterns and shapesthey make can gain meaning on paper. As a result of the comments madeby considering th emeanings of the drawn symbols, important in formation about the child can be reached. Communication is one of the most basic needs of human beings since their existence. With the development of humanity, the means of communication used have also varied. With the invention of the Internet, the process has accelerated and social networks, which have taken their final form today, have become indispensable communication and entertainment tools. Social media accounts, which are versatile, enable all activities such as video sharing, photo sharing, news reading and commenting. Why do children paint? Painting is a communication tool for children. Concepts that cannot be expressed verbally can be expressed more easily through pictures. As a result of the examinations made by the researchers, it was determined that a lot of information could be obtained about them as a result of the analysis of the children's pictures.

26. Bazı rezene popülasyonlarının (Foeniculum vulgare Mill.) ISSR markörlerine dayalı moleküler varyasyonu

Abstract

This research was conducted to determine the molecular variation between different fennel populations. in this research, six fennel populations, of which two were collected from natural flora, were used as a material. the molecular characterization of the populations was performed using six ISSR (Inter simple sequence repeat) primers. A total of 57 bands were obtained, of which 48 were evaluated as polymorphic. the average polymorphism percentage was 85%. the PIC values changed from 0.33 (ISSR-16) to 0.43 (ISSR-11) with an average of 0.37. the average RP (Resolving Power) value of the primers was found as 4.17, and the highest value was obtained from the ISSR-4 (6.33) primer. the mean EMR value was 6.78, and the highest value was obtained from the ISSR-4 (10.29) primer. the mean MI (Marker Index) value was estimated to be 2.47, with the highest value similarly observed in the ISSR-4 (3.67) primer. ISSR-4 was the most prominent primer in terms of these three parameters and the maximum total and polymorphic bands. the similarity values of the fennel populations were found to be between 0.30 and 0.95. the Mental test conducted between Jaccard's similarity matrices and the cluster result revealed the r value as 0.99, showing a very good correlation between the ISSR-based similarity matrix and the cluster. the populations of Burdur, Denizli, Isparta and Izmir, which were cultivated and traded, were genetically similar in terms of the ISSR primers examined, whereas Manisa Yunt Mountain and Odemis populations collected from natural flora were in separate groups., Bu araştırma, farklı rezene popülasyonları arasındaki moleküler varyasyonunu belirlemek amacıyla yürütülmüştür. Araştırmada materyal olarak farklı kaynaklardan sağlanan ve ikisi doğal floradan toplanmış olan 6 adet rezene popülasyonu kullanılmıştır. Popülasyonların moleküler karakterizasyonu 6 adet ISSR primeri ile belirlenmiştir. Toplam 57 bant elde edilmiş, bunlardan 48 adedi polimorfik olarak değerlendirilmiştir. Ortalama polimorfizm oranı %85 olarak bulunmuştur. PIC değerleri 0.33 (ISSR-16) ile 0.43 (ISSR-11) arasında değişim göstermiş, ortalama ise 0.37 olarak hesaplanmıştır. İncelenen ISSR primerleri bakımından ekimi ve satışı yapılan Burdur, Denizli, Isparta ve Izmir populasyonları birbirlerine genetik bakımdan oldukça benzer bulunurken doğal floradan toplanan Manisa Yuntdağı ve Ödemis popülasyonları ayrı gruplarda yer almışlardır.

32. Investigation of lentil flour utilization for the production of protease by Bacillus subtilis ZBP4.

Abstract

In the current study, use of cost effective nitrogen source in the production of protease was aimed by using Bacillus subtilis ZBP4 which is an indigenous strain capable of producing various enzymes and industrially important metabolites. Lentil flour (LF) was used for this purpose, and enzyme production from LF was compared with some other nitrogen sources including casein, yeast extract, peptone from meat, skim milk powder, whey powder, and soy bean flour. Enzyme production in 48 h with LF (863 ± 21.4 U/mL) was lower than only yeast extract (2344 ± 24 U/mL) and soy bean flour (1424 ± 67.6 U/mL). On the other hand, it was considerably higher than all of the other nitrogen sources. Effect of LF concentration (5–20 g/L) was determined by using only LF as nitrogen source and the highest enzyme production was obtained at 20 g/L concentration (1000 ± 29.2 U/mL). Effects of pH (5–9) and temperature (30–45 °C) were also determined and the optima were found as pH 8 and 30 °C. 33. Çocukluk çağı tik bozukluklarının sosyodemografik ve klinik özellikleri

Abstract

Purpose: In this study, we aimed to investigate the sociodemographic and clinical characteristics of children and adolescents diagnosed with tic disorders. Materials and Methods: The study population included 187 children with tic disorders, aged between 4 and 18 years. The data were obtained by using Yale Global Tic Severity Scale (YGTSS), Screen for Child Anxiety and Related Disorders, Schedule for Affective Disorders and Schizophrenia for School Age Children-Present and Lifetime Version (K-SADS-PL). The demographic information, history of familial diseases were recorded. Results: The study included 37 girls (mean 10.9 ± 2.5 years) and 150 boys (mean 10.9 ± 2.7 years) with a mean age of 10.9 ± 2.6 years. Of all cases, 135 were classified as Tourette Syndrome (n=135; 72,2%), 50 as motor tics (n=50; 26,7 %) and 2 as others tic disorders (n=2; 1,1%) according to the diagnostic tic disorders subtype. There was no significant difference between the two groups in terms of mean age and first symptom age according to the subtype of tic disorder. In 91% of the cases, the age of onset of tics was 12 years or younger. The rate of having at least one comorbidity, mean number of comorbidities and comorbid ADHD rates were significantly higher among boys than girls. The most common comorbid diagnoses were ADHD (n=142, 75.9%), OCD (n= 08, 57.8%) and anxiety disorder (n=57, 30.4%). Conclusion: In our study, male cases, early onset and multiple comorbid diagnoses were found to be significantly higher in childhood tic disorders., Amaç: Bu çalışmada, tik bozukluğu tanısı alan çocuk ve ergenlerin sosyodemografik verileri, klinik özellikleri ve eşlik eden diğer ruhsal bozuklukların araştırılması amaçlanmıştır. Gereç ve Yöntem: Çalışmaya, Tik Bozukluğu tanısı olan 4-18 yaş aralığında 187 olgu alındı. Ruhsal belirtilerin taranması amacı ile Okul Çağı Çocukları İçin Duygulanım Bozuklukları ve Şizofreni Görüşme Çizelgesi-Şimdi ve Yaşam boyu Şekli Türkçe uyarlaması ÇDŞG-ŞY (KSADS-PL), Tik belirti şiddeti için, Yale Genel Tik Ağırlığını Derecelendirme Ölçeği (YGTDÖ) kullanıldı. Olguların demografik bilgileri ve ailesel hastalık öyküleri kaydedildi. Bulgular: Çalışmaya, ortalama yaşı 10,9±2,6 yaş olan, 37 kız (ortalama 10,9±2,5 yaş) ve 150 erkek (ortalama 10,9±2,7 yaş) olgu alındı. Tik Bozukluğu alt tipine göre olguların 135’i (%72,2) Tourette Sendromu, %26,7’si (n=50) motor tik bozukluğu, 2’si (%1,1) bunların dışında bir tik bozukluğu tanısı aldı. Tik Bozukluğu alt tipine göre olguların ortalama yaşı, ilk belirti yaşı açısından iki grup arasında anlamlı fark yoktu. Olguların % 91’inde tiklerin başlangıç yaşı, 12 yaş ve altında idi. Kızlara göre erkekler arasında en az bir eş tanıya sahip olma oranı, ortalama eş tanı sayısı ve dikkat eksikliği hiperaktivite bozukluğu (DEHB) eş tanı oranı anlamlı biçimde daha fazlaydı. En sık görülen eş tanılar, DEHB (n=142, %75,9), obsesif kompulsif bozukluk (OKB) (n=108, %57,8) ve anksiyete bozukluğu (n=57, %30,4) idi. Sonuç: Çalışmamızda, çocukluk çağı tik bozukluklarında erkek olgu, erken başlangıç ve çoklu eş tanıların belirgin düzeyde yüksek olduğu bulunmuştur. Abstract

Extracellular protease production by a novel strain, Bacillus sp. EBTA6, has been optimized by using central composite design of response surface methodology and properties and industrial applications of crude enzyme have been investigated. Three independent variables (temperature, pH and yeast extract concentration) chosen in the experimental design were significant terms and reduced cubic model fit with the design at p < 0.0001 level. The recommended temperature, pH and yeast extract concentration were 30 °C, 8, and 15 g/L, respectively. Crude enzyme displayed activity over a wide pH and temperature ranges having the optimum at 50–60 °C and pH 8. It was quite stable at high pH values and at 50 °C. Amongst the metal ions (Mg+, Cu2+, Ca2+, Zn2+, K2+, and Sn2+), Ca2+ enhanced the activity and the others either decreased or did not change it. The enzyme activity was reduced by phenyl-methyl-sulfonyl fluoride (PMSF), and ethylene diamine tetra acetic acid (EDTA). The results revealed that the protease was serine alkaline type. Tween 20 and Tween 80 did not inhibit the enzyme, however, sodium dodecyl sulfate (SDS), reduced it by 39%. It completely removed blood stain in 20 min and coagulated milk in the presence of CaCl2. Abstract

In this study, alkaline protease enzyme production by a bacterial strain isolated from sludge samples collected from an anaerobic treatment system was investigated. According to the 16S rDNA sequence analysis, the isolate was identified as Thermoanaerobacter thermohydrosulfuricus (98.52%). Enzyme activity analyses revealed an optimum pH value of 10, an incubation time of 64 h, and a temperature of 35°C. Arabinose and casein hydrolysates were found to be the best carbon and nitrogen sources, respectively. Maximum protease activity was recorded (864.68 U/mL) when arabinose was used instead of glucose. Moreover, the addition of 1 g/L MgSO4.7H2O and 0.25 g/L Tween-80 to the medium increased the enzyme activity. Therefore, it can be concluded that T. thermohydrosulfuricus is a significant producer of alkaline protease enzymes in the culture medium.
