- Abstract
Alterations in the X‐linked recessive DMD gene cause dystrophinopathies with a broad clinical spectrum most commonly ranging from Duchenne muscular dystrophy (DMD) or Becker muscular dystrophy (BMD) to cardiomyopathy or intellectual disability. Carrier females are commonly unaffected but may show signs of dystrophinopathies. In addition, few asymptomatic male carriers with elevated creatine kinase levels have been described possibly related to deletions around exon 48. We now further support this assumed genotype–phenotype correlation by reporting an attenuated phenotype in a three‐generation family with a deletion of exon 48 of the DMD gene with clinically unaffected carrier males and females. We confirmed deep intronic breakpoints in this family by genome sequencing, but such data are not available for published cases. Therefore, further observations are needed to clarify genotype–phenotype correlation in this region, since few reports also describe predicted in‐frame copy number changes affecting this region in association with classical signs of dystrophinopathies.

8. Value of data meets IT security – assessing IT security risks in data-driven value chains

- Abstract
Digitalization forces manufacturing companies to shift towards customer-oriented, highly data-driven forms of value creation. This results in a changing IT security risk landscape as data becomes an attractive target for adversaries leading to an increasing number of attacks. In order to successfully protect data, it is essential that it is assessed in an integrated manner. Although IT security and data-based value creation have been studied by large research bodies, the existing literature fails to provide guidance on IT security risk analysis in data-based value chains. To contribute to the closure of this research gap, we propose a modeling approach which allocates different data types to value activities and analyses the data types in relation to the properties of relevant IT security risks. The evaluation, conducted with industry experts, reveals that it is not only a company’s primary assets that are of concern but also less important data types subject to significant levels of exposure that bear considerable IT security risks.

9. New Approaches for Value-based IT Innovation Management

IT and IT innovations are now considered as one of the most relevant value drivers from an economic and social perspective and an important factor for business success. Thereby, creation and adoption of IT innovations can lead to manifold benefits for a company, such as higher expected cash flows due to the first mover advantage. However, they are also associated with various challenges, such as a high uncertainty about their future development. This doctoral thesis contributes to the field of (IT) innovation management by providing new approaches that support the management of the creation and adoption of IT innovations following the principles of a value-based management (VBM). Related to creation of IT innovations, research papers P1 and P2 focus on improvement of IT innovation projects’ value contribution and investigate how companies can evaluate their projects from an ex ante perspective to optimize the degree of openness (P1), as well as to optimize the team design (P2). Related to adoption of IT innovations, research paper P3 focuses on improvement of the investment strategy and investigates how to allocate a strategic IT innovation budget to IT innovations with different maturity to balance their risk and return potentials. Based on the example of Big Data Analytics, P4 addresses the need for carefully planning and structuring the adoption of IT innovations to ensure the realization of their value contribution in the sense of VBM, whereas P5 points out the importance of evaluating the changes arising through this adoption, for example by assessing the associated IT security risks.

11. Toward an optimal degree of openness in IT innovation projects

- Abstract
In spite of substantial interest in open innovation (OI), both research and practice lack methods that support companies in managing their IT innovation projects (ITIPs) relative to OI. We contribute to the closure of this gap by providing an approach for an ex ante financial evaluation of OI application, which involves developing a theoretical model that determines the optimal degree of openness in ITIPs. Based on our model, we examine relevant causal relationships by analyzing the influence of a company's ability to manage OI and the probability of success in OI application on the theoretical optimum. We find that the optimal openness level is linked with the company's ability to manage OI, which can incorporate organizational, cultural, and technological maturity. To increase the value contribution of OI, companies should focus on a steady improvement in managing OI. The results provide both an indicator for practical decision‐making and a starting point for future research.

Die Radiojodtherapie mit I131 wird bei Schilddrüsenautonomien seit über 60 Jahren mit guten Behandlungserfolgen angewandt, einige Parameter der Durchführung haben sich jedoch in den letzten 15 Jahren verändert. Diese Arbeit untersucht zwei Patientenkollektive, die im Jahr 1998 (n=80) und 2008 (n=94) wegen einer funktionellen Autonomie radiojodtherapiert und über zwei Jahre nachverfolgt wurden. In beiden Jahren wurden ähnliche Erfolgsraten von >90% bei der Beseitigung der Hyperthyreose erzielt. 2008 hatte sich die Radiojodtherapie vereinfacht und es wurden geringere Dosen an I131 verabreicht. Radioiodine therapy with I131 for thyroid autonomy has been established for over 60 years. Over the last 15 years though, some characteristics have changed. This restrospective dissertation compares two groups of patients who were treated with radioactive iodine for thyroid autonomy in 1998 and 2008 and could be followed for at least two years. In both years, success rates of over 90% were reached for eradication of thyroid hyperfunction. In 2008, radioiodine therapy had become simpler and less radiation was used to treat autonomous thyroid tissue.
