We explored the role that exogenous and endogenous competitors for attention play in infants' abilities to encode and retain information over a 6-month period. Sixty-six children visited the laboratory at 15 months, and 32 returned for a second visit at 21 months. Children observed models of conventional- relation and enabling-relation action sequences. Half the children were distracted by a "Mister Monkey" mechanical toy during the conventional-relation sequence, while the other half was distracted during the enabling-relation sequence. The Early Childhood Behavior Questionnaire indexed endogenous factors at both ages. Immediate postmodel production of target actions indexed encoding efficiency, and 6-month production of target actions indexed long-term recall. The exogenous distracter impacted encoding efficiency (i.e., immediate recall), but not long-term recall. Endogenous factors (i.e., temperament) were primarily associated with long-term recall. Of special interest was our finding that endogenous factors, especially surgency, moderated the effect of the exogenous distracter. It appears that when learning conventional-relation sequences in the presence of exogenous distracters, surgency mobilizes attentional resources toward the learning objective; however, when learning enabling-relation sequences under the same conditions, surgency either boosts the saliency of the distracters or boosts children's susceptibility to them.

The development of a novel VHF radar designed to measure winds and temperatures in the planetary boundary layer is described. The radar operates at 54.1 MHz and is compact and easily transportable. The antenna system consists of 12 Yagis grouped into three subarrays arranged in the form of an equilateral triangle. Transmission takes place on the whole array, and reception takes place on the three subarrays, with winds measured by the spaced antenna technique over a height range between 300 and 3000 m. Results from field trials conducted in southern Australia in a variety of meteorological conditions are presented. Comparisons with high-resolution radiosondes launched from the radar site show excellent agreement, with rms differences between radiosonde and radar wind components being about 1.5 m s−1. Observations carried out in rain show that echoes from precipitation are clearly distinguishable from clear-air echoes. Unlike UHF radars, this means that vertical air velocities can be measured during precipitation, and the evolution of drop-size distributions can be studied down to low altitudes. It is shown that temperatures derived from a radio acoustic sounding system are measured up to heights near 2 km, depending on background wind conditions.

CEM-C7, a human leukemic CD4+ T-lymphocyte cell line and three of its subclones, CEM-4R4, CEM-3R43, and ICR-27, previously cultured in a medium supplemented with 5 to 10% fetal bovine serum, have been adapted to serum-free media. The best medium of those tested was RPMI 1640 supplemented with 5 micrograms/ml each transferrin and insulin + 5 ng/ml sodium selenite +/- 0.1% bovine serum albumin. While growing either with or without albumin, the several clonal lines of CEM cells displayed growth similar to serum-supplemented cultures. Cell proliferation of CEM-C7 cells cultured in both serum-free media has been sustained for 3 mo. with culture doubling times of about 25 h for both serum-supplemented and serum-free cultures (viability greater than or equal to 90%). Cell morphology remained essentially the same in serum-free or serum containing media. The expression of CD4, a marker for T-derived lymphoid cells, was not significantly different in serum-free medium. When grown in serum-free medium, CEM-C7 cells exhibited increased steroid responsiveness as evidenced by increased glucocorticoid receptor binding sites, increased induction of glutamine synthetase, and cell lysis at lower concentrations of steroid. Receptor mutant subclones of CEM-C7, which are proven to be completely unresponsive to micromolar concentrations of dexamethasone when grown in serum-supplemented medium, become partially sensitive to the hormone after growth in defined medium. The increased sensitivity of CEM-C7 cells and its subclones to dexamethasone in serum-free medium returned to previous levels when these cells were recultured in serum-containing medium. Our results suggest that substances in serum influence steroid effects on these cells and that the molecular details of glucocorticoid hormone action may be pursued more precisely in a clearly defined culture medium.

Urban community and jail.Evaluate outcome and process of an extensive tuberculosis contact investigation, including completion of treatment of latent TB infection (TLTBI).Between April 2000 and September 2001, 18 epidemiologically-linked tuberculosis cases were identified; 15 were culture-confirmed, all with a matching 14-band DNA fingerprint pattern. The source case had cavitary pulmonary disease and had been incarcerated 4 months prior to diagnosis. Sixty-six of 67 (99%) community contacts and 221/344 (64%) jail contacts were evaluated. The presumed new infection rate was 56% for community contacts (11 cases, 25 tuberculin skin test [TST] positive) and 20% for jail contacts (6 cases, 32 TST converters). Screening results for 113 (33%) jail contacts were obtained in the jail TST registry upon rearrest. All identified cases completed treatment. Of 22 community contacts initiating TLTBI, 11 completed (44% of infected, 50% of initiators). Of 32 infected jail contacts, 12 initiated TLTBI (all who remained incarcerated), and 10 completed (31% of infected, 83% of initiators). None of 20 additional in-fected jail contacts, all of whose TST conversions were identified with re-arrest data, were subsequently located. Two additional related cases have been identified as of October 2003.Close health department/corrections collaboration facilitated this extensive contact investigation, which identified high Mycobacterium tuberculosis transmission rates and controlled the outbreak. Numerous contacts were identified and screened, but rates of treatment completion for infected contacts were low. Novel strategies are needed to maximize the number of infected contacts who are not only identified and evaluated, but completely treated.

Nurses can play an important role as effective change agents in helping health care agencies and institutions recognize the need to move to more family-centered models, understand the benefits of a family-centered approach, and develop strategies for implementing family-centered care. The Institute for Family-Centered Care has identified a number of areas in which many institutions wish to make changes and has been developing guidance materials that explore possibilities, highlight hospitals that have incorporated changes, and provide practical strategies and useful materials to facilitate change. Topics covered in these guidance resources include the following: Developing Family-Centered Vision, Mission, and Philosophy of Care Statements; Developing and Sustaining a Patient and Family Advisory Council; Collaborative Design Planning; Creating Children's Advisory Councils; Creating and Enhancing Patient and Family Resource Centers; Creating Patient and Family Faculty Programs; Working with Seniors and their Families to Enhance Geriatric Care; and Changing Visiting Practices--Families as Partners.

Adopting a family-centered approach to care requires a profound cultural change--from an emphasis on technology and science to one of sharing and collaboration among staff, patients, and family. Organizations that make the shift find that, despite the hard work, everyone ends up happier and healthier.

The purpose of this technical note is to herein presents an approach for estimating the suspended-sediment source from cutterhead, hopper, and clamshell dredges. The approach involves modification of an existing method developed from limited field data. These estimates are needed to provide input to a numerical model called SSFATE (Suspended Sediment FATE) that is being developed under the Dredging Operations and Environmental Research (DOER) Program.

Glucocorticoids (GCs) induce apoptosis in lymphoid cells that contain functional GC receptors (GRs). However, GC resistance often is seen in cells with demonstrable GRs; one such line is CEM-C1. We have tested the hypothesis that positive interactions between GC and cyclic AMP (cAMP) regulate GC actions in CEM clones. Treatment of both GC-resistant CEM-C1 [resistant to 1 microM dexamethasone (Dex)] and the sensitive sister clone, CEM-C7 (approximately 65% cell death with 20 nM Dex, approximately 99% death with 1 microM Dex), with aor = 20 microM concentration of the protein kinase A activator, forskolin, had no significant effect on cell viability. Cotreatment with Dex and forskolin resulted in a strong synergistic death response, with only approximately 10% CEM-C1 cells surviving treatment with 1 microM Dex and 20 microM forskolin. This death was blocked by the GR antagonist RU 38486. However, the extent of apoptosis did not correlate with the amount of GR protein or binding activity in either C7 or C1 cells. As reported previously, Dex-evoked cell death was associated with suppression of c-Myc in C7 cells. In CEM-C1 cells, Dex alone did not affect c-Myc; however, Dex plus forskolin suppressed c-Myc levels. To evaluate mechanisms of Dex-forskolin synergism, fresh subclones of CEM-C7 (clone 14) and CEM-C1 (clone 15) were isolated, to ensure purity of phenotype. In these, forskolin (with or without Dex) caused a similar increase in cAMP (approximately 300-fold) and phospho-cAMP-responsive element binding protein (approximately 4-5-fold) levels, whereas total cAMP-responsive element binding protein expression was not affected. GR transcription function, as tested from a GR-responsive 330-bp mouse mammary tumor virus promoter-luciferase reporter construct, was induced 8- and 4-fold by 1 microM Dex treatment of CEM-C7-14 and CEM-C1-15 cells, respectively. Forskolin (10 microM) significantly potentiated Dex response in CEM-C1-15 cells (13.5-fold) but had only a modest effect (1.5-fold) in CEM-C7-14 cells. These studies suggest that sensitization of CEM-C1 cells by cross-talk between GR and protein kinase A pathways may occur via cooperative effects on GR-mediated gene transcription.

The content and accuracy of radiographic interpretations by radiologists was assessed to determine the reports' ability to provide sufficient information necessary to make clinical treatment decisions. A retrospective review was performed of 371 radiographic studies (21 1 consecutive patients) ana their reports which had been generated by three Board-certified radiologists. Data were collected regarding fracture assessment and description as well as the description and assessment of orthopedic implants. These descriptions were categorized by their ability to be used clinically (precise) or not (general). Fracture descriptions were considered complete for 85% of reports, while their assessment of alignment and displacement (necessary to determine fracture care) was complete on only 9%. Orthopedic implants were described precisely on 12% of reports with 7% of the descriptions in error. The effect and position of orthopedic implants were described precisely for only 27% and 25% of cases, respectively, while implant stability was assessed precisely in only 4% of cases. For 61 % of preoperative studies, the radiologist's report was not available until after the surgical procedure had already been performed. For all variables considered, an average of 3% of descriptions contained an error. Radiologists' reports of radiographs of these patients undergoing orthopedic procedures did not contain sufficient descriptive information to be used clinically, were not promptly available, and contained an error for 3% of variables studied. The attending orthopedic surgeon has traditionally interpreted such radiographs and should continue to do so to provide patients with more immediate and complete clinical evaluation and management

Previously we have shown that glucocorticoid sensitivity could be restored to a clone of glucocorticoid-resistant leukemic T cells by transfecting them with an expression vector for the glucocorticoid receptor. Furthermore, transfection with plasmids expressing fragments of the receptor containing the DNA-binding domain resulted in constitutive loss of cells. In this paper, we report the results of transfecting both types of constructs into lines of glucocorticoid-resistant human leukemic cells of T cell, B cell, and myeloid origin. In all the lymphoid lines tested, transfection of the holoreceptor gene resulted in appearance of steroid-dependent cell death. In the same lines, transfection of glucocorticoid receptor fragments expressing amino acids 1-465* (465 residues of the normal sequence plus a novel 21 amino acid C-terminus) or expressing only 398-465* caused cell death without the addition of steroids. The amount of cell loss following transfection of these constitutively lethal fragments was in the same range as that following transfection of the holo glucocorticoid receptor plus administration of glucocorticoid. However, the cell loss due to the constitutively active fragments occurred more rapidly. Neither of the myeloid lines tested were sensitive to any of the transfected constructs, with or without added steroid.

With increases in computing power, researchers all over the world are developing and applying three-dimensional hydrodynamic models. The development of one such model called CH3D-WES and its application on Chesapeake Bay are presented. The basic equations solved, boundary conditions employed, and numerical solution techniques utilized, along with the parameterization of the vertical turbulence which is crucial when computing flows in partially stratified water bodies, are discussed. Finally, model results from application to the September 1983 data set on Chesapeake Bay are presented and compared to field data. This data set contains a major wind mixing event that was aided by a cooling of the surface waters of the bay.

To evaluate possible involvement of a paracrine/autocrine inhibitory growth factor in myeloma cell growth, we studied the expression and actions of two forms of transforming growth factor beta (TGF-beta 1 and TGF-beta 2) on two closely related myeloma cell lines, OPM-1 and OPM-2. Earlier studies showed that both cell lines contain glucocorticoid receptors, but only OPM-2 cells are growth inhibited by dexamethasone (Dex). We found that OPM-2 growth was inhibited by TGF-beta, with TGF-beta 1 exerting a greater effect than TGF-beta 2, and Dex plus TGF-beta 1 acting synergistically. In OPM-1 (Dex insensitive), TGF-beta mRNA was not expressed, whereas it was induced by Dex in OPM-2. It was also possible to block partially the growth inhibition of Dex in OPM-2 cells by the addition of anti-TGF-beta 1 antibodies. These data suggest that the glucocorticoid effect(s) on myeloma cells may be mediated at least in part through modulation of internal and/or external levels of TGF-beta 1.

Porcine embryos at Day 13 (Day 0 = first day of oestrus) were collected surgically and embryonic discs were isolated microsurgically. The discs were washed and cultured in Dulbecco's modified Eagle's medium without serum, with either 14C-leucine alone or 14C-leucine plus insulin-like growth factor-I (IGF-I) (100 ng/ml) at 37 degrees C for 48 h in 5% CO2 in air. After incubation, discs were morphologically evaluated, frozen in liquid nitrogen and stored at -70 degrees C. No statistical differences in morphology were observed between embryonic discs cultured in medium with IGF-I and those cultured in medium alone (control). Although more radioactivity was incorporated by embryonic discs in the presence of IGF-I than by those cultured in medium without the growth factor, the difference between the two groups was not significant. From two-dimensional gel electrophoresis, it was observed that IGF-I selectively stimulated the synthesis of four new proteins with Mr of 24,000, 70,000, 77,000 and 95,000, respectively and pI between 5.5 and 6.5. At least 90% of the other proteins in the gels was synthesized in greater amount by embryonic discs cultured in the presence of IGF-I than in the controls. These results show that IGF-I can stimulate protein synthesis in pig embryonic discs cultured in vitro and suggest that this growth factor may play an important role in regulating early development.

The cardiovascular actions of three concentrations of desflurane (formerly I-653), a new inhalation anesthetic, were examined in 12 unmedicated normocapnic, normothermic male volunteers. We compared the effects of 0.83, 1.24, and 1.66 MAC desflurane with measurements obtained while the same men were conscious. Desflurane caused a dose-dependent increase in right-heart filling pressure and a decrease in systemic vascular resistance and mean systemic arterial blood pressure. As measured by echocardiography, left ventricular end-diastolic area did not change except for a small increase at 1.66 MAC desflurane, and systolic wall stress was less at all concentrations of desflurane than during the conscious state. Desflurane did not change cardiac index or left ventricular ejection fraction. Heart rate did not change at 0.83 MAC, but progressively increased with deeper desflurane anesthesia. Stroke volume index was less at all concentrations of desflurane than while the men were conscious, but desflurane did not alter the velocity of ventricular circumferential fiber shortening. Mixed venous blood PO2 and oxyhemoglobin saturation were higher during all concentrations of desflurane anesthesia than during the conscious state. No volunteer developed a metabolic acidosis. We conclude that desflurane with controlled ventilation and constant PaCO2 causes cardiovascular depression, as indicated by the increased cardiac filling pressure and decreased stroke volume index and by no change in the velocity of circumferential fiber shortening in the presence of decreased systolic wall stress. However, cardiac output is well maintained, and heart rate does not increase at light levels of anesthesia. The cardiovascular actions of 0.83 and 1.66 MAC desflurane were also reexamined in 6 of the 12 men during the seventh hour of anesthesia. Prolonged desflurane anesthesia resulted in lesser cardiovascular depression than was evidenced during the first 90 min. The measures of cardiac filling (central venous pressure and left ventricular end-diastolic cross-sectional area) did not differ between the early and late periods of anesthesia. Systemic vascular resistance decreased further during the late period, but systolic wall stress did not differ between the two time periods. During the seventh hour of desflurane anesthesia, heart rate and cardiac index were higher at both anesthetic concentrations than during the first 90 min of anesthesia. Left ventricular ejection fraction and velocity of fiber shortening did not change with duration of desflurane anesthesia. Oxygen consumption, oxygen transport, the ratio of the two, mixed venous PO2, and mixed venous oxyhemoglobin saturation (SO2) increased late in the anesthetic in comparison with the first 90 min.

Desflurane, formerly known as I-653 (CF2H-O-CFH-CF3), is a new inhalation anesthetic derived by fluorine substitution for the alpha-ethyl chlorine of isoflurane (CF2H-O-CClH-CF3). The lower solubility and increased stability of desflurane provided by the C-F bond lessen biotransformation to potentially hepatotoxic metabolites. Repeated administration of desflurane to rats, with or without induced hepatic enzymes, does not result in evidence of hepatic injury. In the recent study we extended the tests for liver cell injury to another species, the pig. Our test included prolonged exposure to desflurane or isoflurane, both in the absence and presence of commonly used adjuvants. We measured plasma alanine aminotransferase activity in eight young female swine anesthetized in random order with desflurane (0.8-1.6 MAC) and isoflurane (0.7-1.4 MAC), for a total dose of about 5.5 MAC-hours of each anesthetic, 3-8 days apart. Plasma alanine aminotransferase activities remained in the normal range, and were not significantly greater over baseline values in samples drawn immediately after, 4 h after, or 3-8 days after (mean +/- SD, 6.1 +/- 2.1) the administration of either anesthetic was discontinued after the first study with either desflurane or isoflurane. Five additional pigs were given a mean total dose of 9.7 MAC-hours of desflurane or isoflurane in conjunction with succinylcholine, N2O, fentanyl, naloxone, atracurium, thiopental, edrophonium, and atropine. No changes in plasma alanine aminotransferase activity were detected in blood samples drawn at termination of the anesthesia, 24 h later, and 4-7 days later (mean +/- SD, 5.8 +/- 1.3).(ABSTRACT TRUNCATED AT 250 WORDS)

Pig embryos at the 1- or 2-cell stage (before the 'block' to development in vitro) were cultured in 8 different media derived from Krebs'-Ringer-bicarbonate medium. A 2 x 2 x 2 factorial arrangement was used for the treatments, with glucose, glutamine and phosphate being the major effects tested. Embryos were obtained from sows approximately 44-48 h after the observation of oestrus, with the majority being at the 1-cell stage. Embryos from each female were randomly assigned to each treatment. After in-vitro culture, all embryos were scored for the stage of development attained and stained to determine final cell number. Significant effects were evident due to female, glucose, glutamine, a phosphate x glucose interaction and a glutamine x glucose interaction. None of the media components tested was inhibitory to embryo development. The greatest development (45-60% morula or blastocyst) was achieved with glucose and glutamine (both alone and in combination) in the media, demonstrating that an amino acid can serve as the sole energy source for complete preimplantation embryonic development in vitro.

Ten healthy unpremedicated male volunteers were exposed to an average inspired concentration of 3.6% desflurane (I-653) in oxygen for 89 (SD 17) min. Standard haematological and biochemical tests were performed before (control) and immediately after exposure and at 4, 24, 72 and 192 h. Liver function was assessed at these times by determining total and unconjugated (indirect) bilirubin, and plasma concentrations of aspartate amino transferase, alanine amino transferase, gamma-glutamyl transpeptidase and alkaline phosphatase. Renal function was assessed by examining the urine microscopically for leucocytes, red blood cells, casts and crystals at these times, and creatinine clearance and urine concentrating ability were determined before and 24 and 72 h after exposure. Two additional and sensitive tests of renal integrity were performed before and 4 and 24 h after anaesthesia: measurement of urinary retinol-binding protein and beta-N-acetyl-D-glucosaminidase. Serum inorganic fluoride was determined immediately before and 1, 4, 24, 48, 72 and 192 h after anaesthesia. Urinary inorganic and organic fluoride concentration and excretion rate were determined before and 24, 48, and 72 h after anaesthesia. There were no significant changes in any measured haematological or biochemical variable (other than an increase in total white cell count from 5.9 (1.3) to 8.3 (1.1) x 10(9) litre-1 immediately after exposure) nor in liver or renal function tests. There was no increase in serum or urinary inorganic fluoride or urinary non-volatile organic fluoride concentrations.

Body image is the constantly changing mental picture of one's body. The examination of children's drawings provides an insight into cognitive and psychosocial functioning, which are important aspects in their development of self-concept.

Graduates with a B.S. or graduate degree from the Department of Animal Science (1986 to 1996) of Texas AM University were surveyed by mail to gather opinions on the curriculum and satisfaction with their degrees. The survey contained five Likert questions (1 = excellent, 4 = poor) regarding satisfaction with major, educational results, personal growth, professional development, and skills important for employment. Multiple-choice questions (n = 9) addressed education, background, job status, and gender. Surveys (n = 2,192) were sent to students, and 470 responded. The undergraduate respondents (n = 396) comprised 71.2% (n = 282) nontransfer students and 28.8% (n = 114) transfer students. Transfer students reported a mean grade point ratio (GPR) of 3.05 +/- 0.43 at Texas AM University in comparison to a nontransfer student mean GPR of 3.03 +/- 0.45 for hours completed at Texas AM University (P = 0.83). The results for time from graduation to full-time employment for transfer students were similar to those of nontransfer students (P = 0.95). Differences in current annual salary between transfer students and nontransfer students were apparent (P0.0001). Transfer students and nontransfer students reported similarly that courses within the department were valuable (P = 0.95) and not redundant (P = 0.08). The two groups similarly rated contributions to critical thinking (P = 0.59) and speaking (P = 0.38). Undergraduate respondents (n = 392) were 48.9% (n = 192) male and 51.1% (n = 200) female. Males and females reported a mean GPR of 3.02 +/- 0.46 and 3.07 +/- 0.43, respectively (P = 0.35). Differences in job search time (P0.0001), starting salary (P = 0.0004), and current salary (P0.0001) were apparent between genders; females were paid less and searched longer before finding their first job. Males and females responded similarly that courses were valuable (P = 0.16) and were taught effectively (P = 0.37) and that teaching assistants made positive contributions (P = 0.43). Females felt a lesser contribution to critical thinking ability, leadership ability, and technical expertise. Results suggest the continuance of current educational practices, but changes to specific issues should be initiated.

Wet brewers grains (WBG) were evaluated as a feed source for growing Angus heifers (201 kg, 313 d of age) during a 112-d wintering trial. Effects on growth, feed efficiency, serum Se and first breeding performance were examined. Three replications of four heifers each were fed the following diets: (1) corn silage with no protein supplement (CSNS); (2) corn silage plus 33.8% of the diet dry matter (DM) from WBG (CSWBG), and (3) WBG, corn and fescue hay at 62.2, 10.8 and 26.1% of diet DM, respectively (WBGCH). Average daily gains for heifers fed CSNS, CSWBG and WBGCH were .50, .73 and .56 kg/head (P less than .05), and DM:gain ratios were 9.52, 6.34 and 6.13 (P less than .05), respectively. Serum Se concentrations were higher (P less than .01) in heifers fed diets containing WBG than in heifers fed CSNS. Serum Se concentrations were also influenced by sampling time (P less than .01) and the diet x sampling time interaction (P less than .05). At the end of the trial, serum Se concentrations were .050, .085 and .105 ppm (P less than .01), respectively, for heifers on the three dietary treatments. Following a 66-d breeding period, 12 of 12 heifers fed CSNS and WBGCH diets and 10 of 12 heifers fed CSWBG were diagnosed pregnant and subsequently calved.

Correlated responses in male reproductive traits were determined at 4, 6 and 8 weeks of age in lines of mice selected for large litter size (L+), large 6-week body weight (W+), large litter size and small body weight (L+W-) and small litter size and large body weight (L-W+), and in an unselected control (K). Concentration of serum testosterone and weights of testes, seminal vesicles, epididymides and adrenal glands increased with age. Line differences in testosterone concentration were not detected. L+ and W+ males exhibited positive correlated responses in testes, epididymides and seminal vesicle weights. Testis weight adjusted for body weight was significantly larger for L+ than controls and approached significance for Wf . Realized genetic correlation betestis weight and litter size was 0.60± 0.04, and the realized partial genetic correlation holding body weight constant was 0.42. Therefore, pleiotropic loci, acting viathe hypothalamic-pituitary axis, affect testis weight and litter size independently of body weight. Additionally, genes influencing overall growth have a pleiotropic effect on testis weight and litter size in mice; the realized genetic correlations of body weight with testis weight and with litter size were 0.60 ± 0.03 and 0.52 ± 0.10. Testis weight increased in both L+W- and L-W+ males. The positive correlated response in L+W- may have resulted from changes in frequency of genes controlling reproductive processes; whereas, in L-W+ it could have been the result of changes in the frequency of genes associated with body weight.

Metabolic acidosis and renal solute load in relation to the protein intake of low birth weight Malaysian neonates. Thirty-one low birth weight Malaysian neonates were divided into two groups for the purpose of comparing two different dilutions of modified cow's milk. One dilution supplied 2.4 g protein/100 kcal (16.9 g/l) and the other 3.3 g/100 kcal (22.5 g/l). The two milks were iso-caloric at 69 kcal/100 ml. Clinical observation and weekly estimations of acid-base status, plasma sodium, chloride, potassium, urea and osmolality and of urine osmolality were made during the first three weeks of life. Metabolic acidosis was observed in both groups of infants, being significantly more severe in the higher protein group. Most acidotic babies remained asymptomatic. Plasma urea and plasma and urine osmolalities were all significantly higher in the higher protein group. No case of hyperosmolality occurred. Caution is recommended when feeding cow's milk of protein density greater than 2.4 g/100 kcal to low birth weight infants.

A study was conducted with 24 crossbred boars (77.5 +/- 2.8 days of age) to determine the effects of low Se status on various spermatozoal characteristics and on Se concentration in semen, serum and primary and accessory reproductive tissues. All boars were fed a low Se diet (cornstarch and Torula yeast) ad libitum. Twelve boars were injected every 14 +/- 1 days with sodium selenite (.33 mg Se/kg body weight) and 12 served as saline-treated controls (low Se status). At 210 +/- 5 days of age, six boars in each group were slaughtered, and serum and various tissues were collected and assayed for Se. Treated boars had higher concentrations of Se in the serum (P less than .001), kidney (P less than .001), liver (P less than .001), heart (P less than .001), skeletal muscle (P less than .01), testis (P less than .01), epididymis (P less than .05), seminal vesicle (P less than .01), bulbourethral gland (P less than .001) and prostate (P less than .001) tissues. Starting at 230 +/- 4 days of age, semen samples were collected from the remaining boars at 4- to 6-day intervals until a total of four ejaculates had been obtained from all but two boars. There were no significant treatment differences in semen volume, percentage normal spermatozoa, percentage viability or spermatozoa concentration/milliliter; however, for combined semen Se data, treated boars had more Se than control boars in the whole semen (.165 vs .07 ppm, respectively), spermatozoa (.418 vs .199 micrograms/10(9) spermatozoa, respectively) and seminal plasma (.03 vs .007 ppm, respectively). The boars were castrated around 250 days of age, and no differences in testis length, diameter, weight and spermatozoal concentration were found between groups. Additionally, there were no apparent differences in daily gain, daily feed consumed and the feed to gain ratio between control and treated boars. Although concentrations of Se in serum, semen and reproductive tissues were much lower in control boars than in treated boars, no apparent impairment of sperm morphology or viability resulted from low Se status.

Fluctuations in peripheral concentrations of total unconjugated estrogens (F) and the response of luteinizing hormone (LH), testosterone (T), and total glucocorticoids (CC) to administration of exogenous estradiol-17#{216}(E2 ) were studied in the male bovine in two experiments. In Experiment 1 hourly blood samples were collected via jugular cannula from four puberal Angus bulls during 24 h preinfusion, 12 h infusion and 12 h postinfusion periods. Episodic fluctuations in peripheral concentrations of E, LII, T, and CC were observed during the 24 h preinfusion period; however, the only significant temporal relationship observed was between concentrations of LH at 1 h and concentrations of T at the subsequent hour (LAG- LH-T-CORR, r = 0.42). Infusion of E2 at the rate of 41.7 mg/h resulted in acute increases in circulating concentrations and areas beneath hormone profile curves for LH, T, and CC. Oligomeric, J-chain-containing immunoglobulins were observed to be transferred selectively from serum into colostrum. These studies suggest that, in the case of the mammary gland secretion, a significant role for extraglandular synthesis of IgA merits consideration. Thus, for example, colostrum may contain antibodies synthesized locally as well as antibodies synthesized in the much larger lymphoid tissues such as the gut lamina propria.

A new mutant causing jaundice in mice is reported. Allelism tests indicate that it is not allelic with known hemolytic anemia mutants in mice [hemolytic anemia (sphha), jaundiced (ja), normoblastic anemia (nb), and spherocytosis (sph and sph2Be)]. The jaundiced condition appears in young mice at about 24 hours postpartum and is due to a major increase in unconjugated bilirubin in serum compared to normal controls. Adult mutant mice are not jaundiced and bilirubin levels do not differ from normal mice. Adult male mutants have reduced testes size and no viable sperm. Female mutants are fertile but overall maternal performance is reduced as indicated by increased mortality and reduced growth rate of pups. Genetic tests indicate that a single autosomal recessive gene is responsible for the condition. We propose that the mutant be designated hyper-unconjugated bilirubinemia, with the gene symbol hub.

We evaluated the effects of [d-Ala2,Me,Phe4,Met(0)ol]-enkephalin (FK 33-824) and morphine, opioid receptor agonists, on concentrations of LH, cortisol and GH during the follicular phase in heifers. During three trials, oestrous cycles of Angus heifers were synchronized by two injections of prostaglandin F2α (PGF2α) resulting in 17 induced follicular phases over an 80-day period. Treatments were administered 24 h after the second injection of PGF2α. Blood samples were collected at 15-min intervals from 3 h before until 5 h after i.v. administration of 2 ml physiological saline (trials 1, 2 and 3), 0·5 mg morphine/kg (trials 1 and 2) or 1·8 μg FK 33-824/kg (trials 1 and 2) or 6·7 μg FK 33-824/kg (trial 3). Administration of both doses of FK 33-824 and morphine inhibited episodic release of LH for approximately 60 min. The concentration of cortisol was increased (PP>0·5) by morphine or saline. The serum concentration of GH was increased (P Journal of Endocrinology (1989) 121, 11–17

Temporal relationships among serum concentrations of corticosteroids (CS), luteinizing hormone (LH), progesterone (P), and testosterone (T) were examined in ten bulls exposed to the stress of electroejaculation (EE). Concentrations of CS and P increased concomitantly among bulls to peak levels by 15 min post-EE and then declined to pre-EE levels between 2 and 4 hr after EE. Total number of LH and T peaks decreased from 24 and 18, respectively, during the 12-hr pre-EE period to 11 and 14 during the 12-h post-EE period. Mean concentration of LH decreased after EE until 4 hr post-EE when episodic LH secretion resumed and concentrations of CS and P had declined to pre-EE levels. Similarly, mean concentration of T declined after EE and then increased 6-8 hr post-EE in response to resumption of LH secretion. The temporal associations of elevated concentrations of CS and P with basal LH and T before EE, absence of T response to endogenous LH peaks, and the temporary absence of episodic LH and T peaks after EE, suggest that endogenous adrenal steroids may modulate secretion of T in the bull.

SummaryOf the 16 group A streptococcal strains tested, only 9 exhibiting type-specificity and production of hyaluronic acid possessed capacity to induce aerosol infections in mice. Conversely, the remaining 7 strains which lacked either one or the other of these attributes failed to demonstrate infectivity by aerosol challenge. A statistically significant relationship between HA production and mouse infectivity was observed.

The physical stability and low blood solubility of the new inhaled anesthetic, I-653, imply that this agent produces limited or no toxic effects. To test this possibility, its effects were compared with those of other volatile agents on hepatic, renal, and pulmonary specimens taken from enzyme-induced, hypoxic rats. Male Sprague-Dawley rats were pretreated with phenobarbital and exposed for 1 hr to 12% O2 and no anesthesia (control) or to 1.2 MAC of one of three anesthetics: I-653, isoflurane, or halothane. Liver, kidney, and lung specimens were taken 24 hr after exposure concluded. The livers of all rats given halothane (n = 6) had swelling (20.5 +/- 5.7% of a lobule [mean +/- SD]) and centrilobular necrosis (6.7 +/- 2.1% of a lobule). Isoflurane produced only slight injury (6.7 +/- 3.5% swelling and 1.0 +/- 0.0% necrosis in 3 of 15 rats). No hepatic injury occurred in control rats (n = 20) or in those given I-653 (n = 16). Pulmonary and renal injury were not evident with any agent. Results suggest that I-653 has equivalent or less effect on hepatic, renal, and pulmonary integrity than currently available inhaled anesthetics.

To determine whether brief periods of hypoxia could produce hepatic injury, we pretreated Sprague-Dawley rats with phenobarbital, deprived them of food for 24 hr, and then exposed them to various hypoxic mixtures of nitrogen and oxygen for various lengths of time. Rats exposed to 6% oxygen for 15 or more minutes had centrilobular injury, the severity of which was directly related to the length of exposure (r = 0.71). Extrapolation of these data indicated that no injury would occur if exposures were less than 5 min. Experiments using lower concentrations of oxygen did not reveal hepatic injury, probably because death of the animals occurred before the appearance of detectable injury of the liver. Feeding animals before imposition of hypoxia markedly decreased the risk of hepatic injury. Whether patients who are deprived of food preoperatively and whose liver enzymes are induced incur an increased risk of hepatic injury from brief periods of hypoxia remains to be determined.

Nitrous oxide administration may limit DNA synthesis by inactivating methionine synthetase, and may thus hamper the repair of an injured organ such as the liver. To test this possibility, we pretreated rats with phenobarbital and exposed them to 0.3 MAC halothane in 9% oxygen for 46 min, followed immediately and again 24 hr later by 70% nitrous oxide (0.25 MAC) at an FIO2 of 0.30 for 2 hr. The results from this group were compared with an anesthetic control group in which 0.35% isoflurane (0.25 MAC) was substituted for the nitrous oxide. Additional groups were given a third exposure to nitrous oxide or isoflurane 48 hr after the halothane exposure. All rats were killed 24 hr after their last anesthetic exposure. A second (nonanesthetic) control group of phenobarbital-pretreated rats received 0.3 MAC halothane in 9% oxygen for 46 min and no anesthetic thereafter. They were killed 24, 48, or 72 hr later. Histologic changes in the livers of rats did not differ among the groups given nitrous oxide, isoflurane, or no additional anesthetic after exposure to halothane alone. Thus neither nitrous oxide nor isoflurane appears to hinder the repair of hepatic injury produced by halothane in the hypoxic rat model.

The interactions of glucocorticoids with their receptors somehow determine the cellular responses seen. The high potency glucocorticoid cortivazol differs from the usual glucocorticoids in two ways, structurally and in binding to receptors. Cortivazol contains a phenylpyrazol fused at carbon atoms 2 and 3 to the A ring of the cyclophenathrene, replacing the supposedly essential 3-keto,4,5-double bond pattern of glucocorticoids. Cortivazol binds to the glucocorticoid receptor in the cytosol from CEM C7 cells (a human acute lymphoblastic leukemia line) in a fashion consistent with interaction with at least two sites. Standard glucocorticoids show only one-site binding. In mutant leukemia cells derived from CEM C7, resistant to kill by 10(-6) M dexamethasone and deficient in standard glucocorticoid binding sites, cortivazol still finds a binding site and kills the cells. In wild-type leukemia cells, the binding sites of cortivazol, those with both higher (Kd approximately 5 x 10(-10) M) and lower (Kd approximately 1 x 10(-8] affinity appear to be on forms of the glucocorticoid receptor itself, and not on two different classes of molecules.

Paternal half-sib heritability and genetic correlation estimates were obtained utilizing data from 578 Hereford bulls from 66 sires. Bulls were maintained in three lines (weaning weight, postweaning gain and control) of an ongoing selection project. Growth performance traits studied were adjusted 205-d weaning weight, 365-d weight, individual feed efficiency, sonoray fat thickness and postweaning gain. Heritability estimates for these traits were .15 +/- .17, .33 +/- .19, .46 +/- .21, .28 +/- .18 and .52 +/- .21, respectively. Scrotal measurements taken were circumference of both testes and length and diameter of right testis at 205 and 365 d of age. Heritability estimates were .08 +/- .20, .07 +/- .20 and .28 +/- .24 at 205 d, and .44 +/- .24, .16 +/- .21 and .40 +/- .24 at 365 d, respectively. Excised testes traits, circumference, right testis length, diameter and weight, total sperm in the testes and sperm/gram of testes had heritability estimates of .21 +/- .26, .19 +/- .26, .02 +/- .24, .63 +/- .27, .14 +/- .21 and -.13 +/- .18, respectively. Genetic correlations of scrotal measurements at 205 d with scrotal measurements at 365 d and excised testes traits were negative. Scrotal measurements at 365 d had high positive genetic correlations with excised testes size, weight and total sperm. These relationships suggest that selection for increase of scrotal size at 365 d should increase testes size and weight and sperm numbers. Genetic correlations of weights and gain with scrotal measurements at 365 d and excised testes characteristics were moderate to high, in a favorable direction. Genetic correlations of testes traits with feed efficiency were essentially zero, while those with fat thickness were moderately positive. These results suggest that increasing testes size should not adversely affect growth performance traits except through the reduction in selection intensity.

We previously demonstrated that upper abdominal surgery on rats pretreated with phenobarbital and anesthetized with halothane caused centrilobular necrosis of the liver, which may be secondary to hepatic hypoxia induced by vasoconstriction. This study examined the possibility that celiac plexus blockade might decrease the degree of injury seen in the surgical model. Rats, pretreated with phenobarbital, were anesthetized with halothane, enflurane, isoflurane, or fentanyl with 100% oxygen. At the start of the hepatic artery dissection, the celiac plexus was blocked by injection of bupivacaine. Subsequently, the rat livers were evaluated for presence and degree of centrilobular necrosis. Animals anesthetized with halothane or fentanyl had a significantly greater incidence of centrilobular necrosis than controls (rats pretreated with phenobarbital who received no anesthesia or surgery). Hepatic injury in rats anesthetized with isoflurane or enflurane did not differ from that in controls. We conclude that celiac plexus blockade with bupivacaine provides no protection against hepatic necrosis in this model.

Oviductal fluid (OVF) was harvested chronically from 5 sows beginning on Day 1 of the estrous cycle (Day 0 of estrous cycle = day of detected estrus) and used for embryo culture (Day 3 OVF only). Two experiments were conducted to investigate in vitro development of 1-cell and 2-cell porcine embryos in a modified Kreb's Ringer bicarbonate medium (culture medium, CM), early luteal phase OVF or CM supplemented with OVF (CM-OVF, 25% OVF v/v in CM) with or without transfer to fresh CM. In Experiment 1, 1-cell and 2-cell embryos were harvested from sows (n = 7) approximately 44 h after detected estrus. In Experiment 2, 1-cell embryos were collected from 5 sows treated with altrenogest and gonadotropins, approximately 50 h after injection of human chorionic gonadotropin. The volume of OVF (ml) declined progressively throughout the 4 days of collection (24 h, 8.44 +/- 0.28; 48 h, 6.88 +/- 1.78; 72 h, 4.96 +/- 0.35; 96 h, 4.64 +/- 0.25 after onset of estrus; p less than .01). In rats pretreated with phenobarbital breathing 10% oxygen, subanesthetic doses of halothane, isoflurane, enflurane, thiopental, and fentanyl caused hepatic injury. Because hypoxia per se can produce such injury, we hypothesized that the anesthetic-induced injury resulted from increased hypoxemia secondary to respiratory depression. Male Sprague-Dawley rats were pretreated with phenobarbital; half of the rats were fed and the other half were deprived of food for the 24 h before study. Isoflurane anesthesia was given for the placement of a catheter into the femoral artery. After 1 h of recovery, the rats were exposed to 10% oxygen. Control samples were obtained and halothane, isoflurane, enflurane, thiopental, or fentanyl was administered. Rats given food had higher PaCO2 and lower pH values than starved rats. Also, arterial oxygen saturation (SaO2) tended to be lower in rats given food. At concentrations of 0.15-0.2 MAC or higher, halothane, isoflurane, and enflurane slightly increased PaCO2 values relative to values for a control group exposed only to hypoxia. However, SaO2 and PaO2 did not show significant drug-induced changes. Fentanyl transiently decreased PaO2 and SaO2. Thiopental caused no changes. Thus, we conclude that subanesthetic doses of anesthetics may depress the ventilatory response to hypoxia but that this depression is inconsistent and appears to be too small to cause hepatic damage.

To determine the relative washin and washout characteristics of isoflurane, enflurane, halothane, and methoxyflurane, we administered all four anesthetics simultaneously (total = 1.1 MAC) to nine healthy patients for 2 hr. Concentrations of anesthetics in end-tidal gases were measured during washin and for 5-9 days during washout. Multiexponential (multicompartment) models were fit to the washin and washout curves using least-squares analysis. Slowly equilibrating compartments could only be identified during washout. For 27 of the 36 data sets, five-compartment models fit the washout curves significantly better than four-compartment models. The time constant for our first compartment is consistent with that predicted for washout of the lungs. Time constants for the second, third, and fifth compartments were consistent with current data for blood flows and solubilities of vessel-rich, muscle, and fat tissue groups, respectively. The fourth compartment has a time constant that lies between the time constants predicted for muscle and fat.
