We report on detailed multi-wavelength observations and analysis of the very bright and long GRB 210619B, detected by the Atmosphere-Space Interactions Monitor (ASIM) installed on the International Space Station (ISS) and the Gamma-ray Burst Monitor (GBM) on-board the Fermi mission. Our main goal is to understand the radiation mechanisms and jet composition of GRB 210619B. With a measured redshift of $z$ = 1.937, we find that GRB 210619B falls within the 10 most luminous bursts observed by Fermi so far. The energy-resolved prompt emission light curve of GRB 210619B exhibits an extremely bright hard emission pulse followed by softer/longer emission pulses. The low-energy photon indices ($\alpha_{\rm pt}$) values obtained using the time-resolved spectral analysis of the burst suggest a transition between the thermal (during harder pulse) to non-thermal (during softer pulse) outflow. We examine the correlation between spectral parameters and find that both peak energy and $\alpha_{\rm pt}$ exhibit the flux tracking pattern. The late time broadband photometric dataset can be explained within the framework of the external forward shock model with $\nu_m$ $< \nu_c$ $< \nu_{x}$ (where $\nu_m$, $\nu_c$, and $\nu_{x}$ are the synchrotron peak, cooling-break, and X-ray frequencies, respectively) spectral regime supporting a rarely observed hard electron energy index ($p, Comment: 24 pages, 18 figures, 10 tables, accepted for publication in MNRAS

Creep, the time-dependent deformation of materials stressed below the yield strength, is responsible for a great number of component failures at high temperatures. Because grain boundaries (GBs) in materials usually facilitate diffusional processes in creep, eliminating GBs is a primary approach to resisting high-temperature creep in metals, such as in single-crystal superalloy turbo blades. We report a different strategy to inhibiting creep by use of stable GB networks. Plastic deformation triggered structural relaxation of high-density GBs in nanograined single-phased nickel-cobalt-chromium alloys, forming networks of stable GBs interlocked with abundant twin boundaries. The stable GB networks effectively inhibit diffusional creep processes at high temperatures. We obtained an unprecedented creep resistance, with creep rates of ~10 –7 per second under gigapascal stress at 700°C (~61% melting point), outperforming that of conventional superalloys.

Gamma-ray bursts (GRBs) are the most explosive phenomena and can be used to study the expansion of Universe. In this paper, we compile a long GRB sample for the Eiso–Ep correlation from Swift and Fermi observations. The sample contains 221 long GRBs with redshifts from 0.03 to 8.20. From the analysis of data in different redshift intervals, we find no statistically significant evidence for the redshift evolution of this correlation. Then we calibrate the correlation in six subsamples and use the calibrated one to constrain cosmological parameters. Employing a piece-wise approach, we study the redshift evolution of dark energy equation of state (EOS), and find that the EOS tends to be oscillating at low redshift, but consistent with −1 at high redshift. It hints at dynamical dark energy at 2σ confidence level at low redshift.

IntroductionIntermittent theta-burst stimulation (iTBS) is a non-invasive brain stimulation paradigm that has demonstrated promising therapeutic benefits for a variety of neuropsychiatric disorders. It has recently garnered widespread favor among researchers and clinicians, owing to its comparable potentiation effects as conventional high-frequency repetitive transcranial magnetic stimulation (rTMS), but administered in a much shorter time frame. However, there is still a lack of agreement over the optimal stimulation intensity, particularly when targeting the prefrontal regions. The objective of this study was to systematically investigate the influence of different stimulation intensities of iTBS, applied over the left dorsolateral prefrontal cortex (DLPFC), on brain activity and executive function in healthy adults.MethodsTwenty young healthy adults were enrolled in this randomized cross-over experiment. All participants received a single session iTBS over the left DLPFC at intensities of 50, 70, or 100% of their individual resting motor threshold (RMT), each on separate visits. Functional near-infrared spectroscopy (fNIRS) was used to measure changes of hemoglobin concentrations in prefrontal areas during the verbal fluency task (VFT) before and after stimulation.ResultsAfter stimulation, iTBS to the left DLPFC with 70% RMT maintained the concentration change of oxyhemoglobin (HbO) in the target area during the VFT. In contrast, 50% [t(17) = 2.203, P = 0.042, d = 0.523] and 100% iTBS [t(17) = 2.947, P = 0.009, d = 0.547] significantly decreased change of HbO concentration, indicating an inverse U-shape relationship between stimulation intensity and prefrontal hemodynamic response in healthy young adults. Notably, improved VFT performance was only observed after 70% RMT stimulation [t(17) = 2.511, P = 0.022, d = 0.592]. Moreover, a significant positive correlation was observed between task performance and the difference in HbO concentration change in the targeted area after 70% RMT stimulation (r = 0.496, P = 0.036) but not after 50 or 100% RMT stimulation.ConclusionThe linear relationship between stimulation intensity and behavioral outcomes reported in previous conventional rTMS studies may not be translated to iTBS. Instead, iTBS at 70% RMT may be more efficacious than 100% RMT.

The main scientific goal of LHAASO-WCDA is to survey gamma-ray sources with energy from 100 GeV to 30 TeV. To observe high-energy shower events, especially to measure the energy spectrum of cosmic rays from 100 TeV to 10 PeV, a dynamic range extension system (WCDA++) is designed to use a 1.5-inch PMT with a dynamic range of four orders of magnitude for each cell in WCDA-1. The dynamic range is extended by using these PMTs to measure the effective charge density in the core region of air shower events, which is an important parameter for identifying the composition of primary particles. The system has been running for more than one year. In this paper, the details of the design and performance of WCDA++ are presented.

The kilometer square array (KM2A) of the large high altitude air shower observatory (LHAASO) aims at surveying the northern γ-ray sky at energies above 10 TeV with unprecedented sensitivity. γ-ray observations have long been one of the most powerful tools for dark matter searches, as, e.g., high-energy γ rays could be produced by the decays of heavy dark matter particles. In this Letter, we present the first dark matter analysis with LHAASO-KM2A, using the first 340 days of data from 1/2-KM2A and 230 days of data from 3/4-KM2A. Several regions of interest are used to search for a signal and account for the residual cosmic-ray background after γ/hadron separation. We find no excess of dark matter signals, and thus place some of the strongest γ-ray constraints on the lifetime of heavy dark matter particles with mass between 105 and 109 GeV. Our results with LHAASO are robust, and have important implications for dark matter interpretations of the diffuse astrophysical high-energy neutrino emission.

Abnormal activation of the kynurenine and serotonin pathways of tryptophan metabolism is linked to a host of neuropsychiatric disorders. Concurrently, noninvasive brain stimulation (NIBS) techniques demonstrate high therapeutic efficacy across neuropsychiatric disorders, with indications for modulated neuroplasticity underlying such effects. We therefore conducted a scoping review with meta-analysis of eligible studies, conforming with the PRISMA statement, by searching the PubMed and Web of Science databases for clinical and preclinical studies that report the effects of NIBS on biomarkers of tryptophan metabolism. NIBS techniques reviewed were electroconvulsive therapy (ECT), transcranial magnetic stimulation (TMS), and transcranial direct current stimulation (tDCS). Of the 564 search results, 65 studies were included with publications dating back to 1971 until 2022. The Robust Bayesian Meta-Analysis on clinical studies and qualitative analysis identified general null effects by NIBS on biomarkers of tryptophan metabolism, but moderate evidence for TMS effects on elevating serum serotonin levels. We cannot interpret this as evidence for or against the effects of NIBS on these biomarkers, as there exists several confounding methodological differences in this literature. Future controlled studies are needed to elucidate the effects of NIBS on biomarkers of tryptophan metabolism, an under-investigated question with substantial implications to clinical research and practice.

V., Multipulsed GRB 190530A, detected by the Gamma-ray Burst Monitor (GBM) and Large Area Telescope onboard Fermi, is the sixth most fluent GBM burst detected so far. This paper presents the timing, spectral, and polarimetric analysis of the prompt emission observed using AstroSat and Fermi to provide insight into the prompt emission radiation mechanisms. The time-integrated spectrum shows conclusive proof of two breaks due to peak energy and a second lower energy break. Time-integrated (55.43 ± 21.30 per cent) as well as time-resolved polarization measurements, made by the Cadmium Zinc Telluride Imager (CZTI) onboard AstroSat, show a hint of high degree of polarization. The presence of a hint of high degree of polarization and the values of low energy spectral index (αpt) do not run over the synchrotron limit for the first two pulses, supporting the synchrotron origin in an ordered magnetic field. However, during the third pulse, αpt exceeds the synchrotron line of death in few bins, and a thermal signature along with the synchrotron component in the time-resolved spectra is observed. Furthermore, we also report the earliest optical observations constraining afterglow polarization using the MASTER (P < 1.3 per cent) and the redshift measurement (z = 0.9386) obtained with the 10.4 m GTC (Gran Telescopio Canarias) telescopes. The broad-band afterglow can be described with a forward shock model for an ISM (interstellar medium)-like medium with a wide jet opening angle. We introduce a new GPU-accelerated general-relativistic magneto-hydrodynamic (GR-MHD) code based on HARM which we call cuHARM. The code is written in CUDA-C and uses OpenMP to parallelize multi-GPU setups. Our code allows us to run high resolution simulations of accretion disks and the formation and structure of jets without the need of multi-node supercomputer infrastructure. A $256^3$ simulation is well within the reach of an Nvidia DGX-V100 server, with the computation being a factor about 10 times faster if only the CPU was used. We use this code to examine several disk structures all in the SANE state. We find that: (i) increasing the magnetic field, while in the SANE state does not affect the mass accretion rate; (ii) Simultaneous increase of the disk size and the magnetic field, while keeping the ratio of energies fixed, lead to the destruction of the jet once the magnetic flux through the horizon decrease below a certain limit. This demonstrates that the existence of the jet is not a linear function of the initial magnetic field strength; (iii) the structure of the jet is a weak function of the adiabatic index of the gas, with relativistic gas tend to have a wider jet., Comment: 33 pages, 16 figures, submitted to ApJ

Very energetic giant flares from galactic magnetars (peak luminosities of 1044–1047 ergs per second, lasting approximately 0.1 s) have been detected in hard X-rays and soft γ-rays4, and only one has been detected from outside our galaxy5. During such giant flares, quasi-periodic oscillations (QPOs) with low (less than 150 hertz) and high (greater than 500 hertz) frequencies have been observed6–9, but their statistical significance has been questioned10. High-frequency QPOs have been seen only during the tail phase of the flare9. Here we report the observation of two broad QPOs at approximately 2,132 hertz and 4,250 hertz in the main peak of a giant γ-ray flare11 in the direction of the NGC 253 galaxy12–17, disappearing after 3.5 milliseconds. The flare was detected on 15 April 2020 by the Atmosphere–Space Interactions Monitor instrument18,19 aboard the International Space Station, which was the only instrument that recorded the main burst phase (0.8–3.2 milliseconds) in the full energy range (50 × 103 to 40 × 106 electronvolts) without suffering from saturation effects such as deadtime and pile-up. Recently, the LHAASO Collaboration published the detection of 12 ultrahigh-energy γ-ray sources above 100 TeV, with the highest energy photon reaching 1.4 PeV. The first detection of PeV γ rays from astrophysical sources may provide a very sensitive probe of the effect of the Lorentz invariance violation (LIV), which results in decay of high-energy γ rays in the superluminal scenario and hence a sharp cutoff of the energy spectrum. Two highest energy sources are studied in this work. No signature of the existence of the LIV is found in their energy spectra, and the lower limits on the LIV energy scale are derived. Our results show that the first-order LIV energy scale should be higher than about 10^{5} times the Planck scale M_{Pl} and that the second-order LIV scale is10^{-3}M_{Pl}. Both limits improve by at least one order of magnitude the previous results.

We report the discovery of an extended very-high-energy (VHE) gamma-ray source around the location of the middle-aged (207.8 kyr) pulsar PSR J0622+3749 with the Large High-Altitude Air Shower Observatory (LHAASO). The source is detected with a significance of 8.2σ for E25 TeV assuming a Gaussian template. The best-fit location is (right ascension, declination) =(95.47°±0.11°,37.92°±0.09°), and the extension is 0.40°±0.07°. The energy spectrum can be described by a power-law spectrum with an index of -2.92±0.17_{stat}±0.02_{sys}. No clear extended multiwavelength counterpart of the LHAASO source has been found from the radio to sub-TeV bands. The LHAASO observations are consistent with the scenario that VHE electrons escaped from the pulsar, diffused in the interstellar medium, and scattered the interstellar radiation field. If interpreted as the pulsar halo scenario, the diffusion coefficient, inferred for electrons with median energies of ∼160 TeV, is consistent with those obtained from the extended halos around Geminga and Monogem and much smaller than that derived from cosmic ray secondaries. The LHAASO discovery of this source thus likely enriches the class of so-called pulsar halos and confirms that high-energy particles generally diffuse very slowly in the disturbed medium around pulsars.

We report the discovery of a UHE gamma-ray source, LHAASO J2108+5157, by analyzing the LHAASO-KM2A data of 308.33 live days. Significant excess of gamma-ray induced showers is observed in both energy bands of 25-100 TeV and $\gt$100 TeV with 9.5 sigma and 8.5 sigma, respectively. This source is not significantly favored as an extensive source with the angular extension smaller than the point-spread function of KM2A. The measured energy spectrum from 20 to 200 TeV can be approximately described by a power-law function with an index of -2.83$\pm$ 0.18stat. A harder spectrum is demanded at lower energies considering the flux upper limit set by Fermi-LAT observations. The position of the gamma-ray emission is correlated with a giant molecular cloud, which favors a hadronic origin. No obvious counterparts have been found, deeper multiwavelength observations will help to shed new light on this intriguing UHE source., Comment: 12 pages, 7 figures, accepted by ApJL

The Water Cherenkov Detector Array (WCDA) is a major component of the Large High Altitude Air Shower Array Observatory (LHAASO), a new generation cosmic-ray experiment with unprecedented sensitivity, currently under construction. The WCDA is aimed at the study of TeV $\gamma$-rays. In order to evaluate the prospects of searching for TeV $\gamma$-ray sources with the WCDA, we present in this paper a projection for the one-year sensitivity of the WCDA to TeV $\gamma$-ray sources from TeVCat (footnote: http://tevcat.uchicago.edu) using an all-sky approach. Out of 128 TeVCat sources observable to the WCDA up to a zenith angle of $45^\circ$, we estimate that 42 would be detectable for one year of observations at a median energy of 1 TeV. IntroductionRepetitive transcranial magnetic stimulation (rTMS) with theta burst stimulation (i.e. TBS) of the dorsolateral prefrontal cortex (DLPFC) is an innovative treatment for major depressive disorder (MDD). However, fewer than 50% of patients show sufficient response to this treatment; markers for response prediction are urgently needed. Research shows considerable individual variability in the brain responses to rTMS. However, whether differences in individual DLPFC modulation by rTMS can be used as a predictive marker for treatment response remains to be investigated. Here, we present a research programme that will exploit the combination of functional near-infrared spectroscopy (fNIRS) with brain stimulation. Concurrent TBS/fNIRS will allow us to systematically investigate TBS-induced modulation of blood oxygenation as a proxy for induced brain activity changes. The findings from this study will (1) elucidate the immediate effects of excitatory and inhibitory TBS on prefrontal activity in TBS treatment-naïve patients with MDD and (2) validate the potential utility of TBS-induced brain modulation at baseline for the prediction of antidepressant response to 4 weeks of daily TBS treatment.Methods and analysisOpen-label, parallel-group experiment consisting of two parts. In part 1, 70 patients and 37 healthy controls will be subjected to concurrent TBS/fNIRS. Intermittent TBS (iTBS) and continuous TBS (cTBS) will be applied on the left and right DLPFC, respectively. fNIRS data will be acquired before, during and several minutes after stimulation. In part 2, patients who participated in part 1 will receive a 4 week iTBS treatment of the left DLPFC, performed daily for 5 days per week. Psychometric evaluation will be performed periodically and at 1 month treatment follow-up. Statistical analysis will include a conventional, as well as a machine learning approach.Ethics and disseminationEthics approval was obtained from the Institutional Review Board. Different light filters affect leaf photosynthetic features and fruit quality. Consequently, selecting the appropriate covering filter for rain-shelter cultivation of peaches is a key part of successful production. We used a late-maturing peach variety ‘Xiahui 8’ to study differences in leaf photosynthetic features, chlorophyll fluorescence characteristics, and fruit quality under neutral, red, yellow, green, and blue filter, with natural light as control. The results showed that the leaf photosynthetic ability and internal quality under the neutral filter treatment were elevated compared with the control, and the appearance color was the same as the control. Leaves under neutral filter could maintain higher photosynthetic ability than other filter treatments. In addition, the fruits could also keep higher quality when treated with neutral filter. Therefore, the application of neutral filter in rain-shelter cultivation of ‘Xinhui 8’ peaches is recommended for maintaining high photosynthetic capacity and for improving fruit quality.

Under optimal conditions, most of the light energy is used to drive electron transport. However, when the light energy exceeds the capacity of photosynthesis, the overall photosynthetic efficiency drops down. The present study investigated the effects of high light on rice photooxidation-prone mutant 812HS, characterized by a mutation of leaf photooxidation 1 gene, and its wild type 812S under field conditions. Our results showed no significant difference between 812HS and 812S before exposure to high sunlight. However, during exposure to high light, shoot tips of 812HS turned yellow and their chlorophyll (Chl) content decreased. Transmission electron microscopy showed that photooxidation resulted in significant damage of chloroplast ultrastructure. It was confirmed also by inhibited photophosphorylation and reduced ATP content. The decreased coupling factor of ATP, Ca2+-ATPase and Mg2+-ATPase activities also verified these results. Further, significantly enhanced activities of antioxidative enzymes were observed during photooxidation. Malondialdehyde, hydrogen peroxide, and the superoxide generation rates also increased. Chl a fluorescence analysis found that the performance index and maximum quantum yield of PSII declined on August 4, 20 days after high-light treatment. Net photosynthetic rate also decreased and substomatal CO2 concentration increased in 812HS at the same time. In conclusion, our findings indicated that excessive energy triggered the production of toxic reactive oxygen species and promoted lipid peroxidation in 812HS plants, causing severe damage to cell membranes, degradation of photosynthetic pigments and proteins, and ultimately inhibition of photosynthesis.

Efficient photosynthesis is critical for plant survival and growth. When plant-absorbed light exceeds the overall rate of energy conversion, it will trigger photooxidation. In this study, we selected a photooxidation mutant 812HS, it was isolated from the progeny of japonica rice (Oryza sativa L.) 812S and shows leaf yellowing and hypersensitive to photooxidation. Chloroplast ultrastructure in the leaves of 812HS showed that photooxidation resulted in significant chloroplast damage compared with 812S for changes in gene expressions in response to photooxidation stress using next-generation sequencing technologies on an Illumina HiSeq 2000 platform. A total of 88508 and 88495 genes were identified from 812S and 812HS, respectively. Expressions of 1199 genes were significantly upregulated, while 1342 genes were remarkably downregulated in 812HS. These genes were notably enriched in the 21 KEGG pathways. Based on their expression patterns, several key pathways were identified to be involved in the photooxidation of 812HS. qRT-PCR analysis further confirmed the results of RNA-Seq. This study enabled us to integrate analysis of RNA-Seq in rice and offered a deeper insight into the molecular mechanisms in response to photo-oxidative stress and provided clues for further critical gene identification in the protective mechanisms against photooxidation.

Complex kinship analysis refers to a kind of special kinship analysis taken for the purpose of personal identification or other issues in civil or criminal cases because the father or （and） mother is dead, or cannot participate in the analysis for other reasons. Due to the absence of significant appraised persons in this kind of kinship analysis, grandparents, siblings or collateral relatives are required to participate in the analysis. Complex kinship analysis is widely used and the demand is increasing year by year. This paper analyzes the main types of complex kinships, the genetic markers of complex kinship analysis and their advantages and disadvantages and the calculation methods for complex kinship analysis by referring to the relevant literatures at home and abroad in recent years. The LHAASO-WFCTA experiment, which aims to observe cosmic rays in the sub-EeV range using the fluorescence technique, uses a new generation of high-performance telescopes. To ensure that the experiment has excellent detection capability associated with the measurement of the energy spectrum, the primary composition of cosmic rays, and so on, an accurate geometrical reconstruction of air-shower events is fundamental. This paper describes the development and testing of geometrical reconstruction for stereo viewed events using the WFCTA (Wide Field of view Cherenkov/Fluorescence Telescope Array) detectors. Two approaches, which take full advantage of the WFCTA detectors, are investigated. One is the stereo-angular method, which uses the pointing of triggered SiPMs in the shower trajectory, and the other is the stereo-timing method, which uses the triggering time of the fired SiPMs. The results show that both methods have good geometrical resolution; the resolution of the stereo-timing method is slightly better than the stereo-angular method because the resolution of the latter is slightly limited by the shower track length.

A sub-array of the Large High Altitude Air Shower Observatory (LHAASO), KM2A is mainly designed to observe a large fraction of the northern sky to hunt for γ-ray sources at energies above 10 TeV. Even though the detector construction is still underway, half of the KM2A array has been operating stably since the end of 2019. In this paper, we present the KM2A data analysis pipeline and the first observation of the Crab Nebula, a standard candle in very high energy γ-ray astronomy. We detect γ-ray signals from the Crab Nebula in both energy ranges of 10 100 TeV and 100 TeV with high significance, by analyzing the KM2A data of 136 live days between December 2019 and May 2020. With the observations, we test the detector performance, including angular resolution, pointing accuracy and cosmic-ray background rejection power. The energy spectrum of the Crab Nebula in the energy range 10-250 TeV fits well with a single power-law function dN/dE = (1.13 0.05 0.08 ) 10 (E/20 TeV) cm s TeV . It is consistent with previous measurements by other experiments. This opens a new window of γ-ray astronomy above 0.1 PeV through which new ultrahigh-energy γ-ray phenomena, such as cosmic PeVatrons, might be discovered.

An optimal dosage of nitrogen has long been known to play important roles in governing nitrogen-use efficiency and improving the yield of plants. However, there is limited information on the isolation and expression profiles of genes related to nitrogen metabolism in peach (Prunus persica L. Batsch). This study isolated five genes involved in nitrogen metabolism and evaluated the effects of a single foliar application of urea on levels of expression of these genes in the leaves of ‘Dazhenbaochiyue’ peach. Three new complexes [Cd(Hflu)2(NO3)](NO3) 1, [Cd(H2O)(Hflu)2(Hmalo)](NO3)∙2(H2O) 2, and [Cd(H2O)(Hflu)2(Hfum)](NO3)∙2(H2O) 3 (Hflu = [2-(2,4-difluorophenyl)-1,3-bis(1,2,4-triazol-1-yl)-propan-2-ol], H2fum = fumaric acid, H2malo = malonate acid) are synthesized by a solution diffusion method and characterized by single crystal X-ray diffraction methods, elemental analyses, IR spectroscopy as well as thermal analyses. In compound 1, Cd(II) cations are bridged by fluconazole ligands to form 2D Cd−Hflu layers, and these layers are further connected by C–H∙∙∙F hydrogen bonds to form a complicated 3D structure with the topology of (4∙6∙8)(4∙64∙84∙10)(6∙3). Compounds 2 and 3 are isostructural, and in them the Cd(II) cations are bridged by fluconazole ligands to form a 2D network, which is connected by C–H∙∙∙O hydrogen bonds to form a complicated 3D (3,4)-connected framework with the topology of (4∙82)(4∙85).

Iron is required for the Fe-S cluster assembly which occurs in chloroplasts, mitochondria, and cytosol and here we characterized 44 Fe-S cluster biosynthesis genes and investigated their expression profiles during different peach flowering stages. Quantitative real-time PCR analysis shows that the highest expression of most peach Fe-S cluster biosynthesis genes appeared in the full bloom stage. Also, the highest Fe accumulation occurred in the full bloom stage followed by beginning bloom, petal fall, and bud swell stages. Activities of nitrite reductase (NiR) and succinate dehydrogenase (SDH) were closely correlated to the flower Fe content, whereas the aconitase (ACO) activity kept steady during the whole flowering process. Moreover, shading treatment significantly reduced Fe accumulation and NiR, SDH, and ACO activities of the full blooming flowers. Seventeen Fe-S cluster biosynthesis genes were down-regulated in response to a shading treatment. In particular, plastid sulfur mobilization genes were sensitive to the shading treatment.

Constipation has a significant influence on quality of life. Patients with constipation have slow waves in their gastrointestinal smooth muscles and less faecal water contents, which are closely associated with down-regulation of the interstitial cells of Cajal (ICC) in the gastrointestinal muscles and the aquaporin protein AQP3 expressed in colon epithelial cells. Recent studies supported that patients with constipation have altered intestinal microbial structures compared with healthy controls. Intestinal dysbiosis might be one possible pathophysiological mechanism causing constipation. Bacterial strains, such as Lactobacillus spp., have shown many beneficial effects on the amelioration of constipation. However, few studies reported the structural changes of intestinal microbiota post-intervention of probiotics. In this study, a bacterial mixture was administrated to rats with loperamide-induced constipation. Effects of the bacterial mixture on small intestine transit (SIT), faecal water content, and the intestinal microbiome in rats were evaluated. Meanwhile, we investigated several factors involved in signalling pathways that regulate function of ICC and expression of AQP3 to discuss the possible underlying molecular mechanisms. Intervention of the bacterial mixture improved SIT and faecal water content in constipated rats. The up-regulation of C-kit/SP signalling pathways in ICC and AQP3 significantly contributed to improvements. These changes were closely associated with the manipulation of intestinal dysbiosis in constipated rats. Furthermore, our results revealed the important role of intestinal microbiota in affecting gut motility through regulation of serotonin biosynthesis. This monoamine neurotransmitter, secreted from enterochromaffin cells, up-regulated both substance P/neurokinin 1 receptors pathway of ICC and the expression of AQP3 in intestinal epithelial cells. Our study suggested that the disrupted microbiome in patients could be a potential therapeutic target for the improvement of constipation.

THESEUS is a space mission concept aimed at exploiting Gamma-Ray Bursts for investigating the early Universe and at providing a substantial advancement of multi-messenger and time-domain astrophysics. These goals will be achieved through a unique combination of instruments allowing GRB and X-ray transient detection over a broad field of view (more than 1sr) with 0.5¿1 arcmin localization, an energy band extending from several MeV down to 0.3¿keV and high sensitivity to transient sources in the soft X-ray domain, as well as on-board prompt (few minutes) follow-up with a 0.7¿m class IR telescope with both imaging and spectroscopic capabilities. THESEUS will be perfectly suited for addressing the main open issues in cosmology such as, e.g., star formation rate and metallicity evolution of the inter-stellar and intra-galactic medium up to redshift 10, signatures of Pop III stars, sources and physics of re-ionization, and the faint end of the galaxy luminosity function. In addition, it will provide unprecedented capability to monitor the X-ray variable sky, thus detecting, localizing, and identifying the electromagnetic counterparts to sources of gravitational radiation, which may be routinely detected in the late ¿20s/early ¿30s by next generation facilities like aLIGO/ aVirgo, eLISA, KAGRA, and Einstein Telescope. This prospective, randomized, phase III trial shows that prophylactic cranial irradiation prolongs disease-free survival, decreases the rate of cerebral metastases and does not affect quality-of-life for patients with fully resected postoperative pathologically confirmed stage IIIA-N2 non-small-cell lung cancer and high risk of cerebral metastases after adjuvant chemotherapy. Background This study compared prophylactic cranial irradiation (PCI) with observation in patients with resected stage IIIA–N2 non-small-cell lung cancer (NSCLC) and high risk of cerebral metastases after adjuvant chemotherapy. Patients and methods In this open-label, randomized, phase III trial, patients with fully resected postoperative pathologically confirmed stage IIIA–N2 NSCLC and high cerebral metastases risk without recurrence after postoperative adjuvant chemotherapy were randomly assigned to receive PCI (30 Gy in 10 fractions) or observation. The primary end point was disease-free survival (DFS). The secondary end points included the incidence of brain metastases, overall survival (OS), toxicity and quality of life. Results This trial was terminated early after the random assignment of 156 patients (81 to PCI group and 75 to control group). The PCI group had significantly lengthened DFS compared with the control group, with a median DFS of 28.5 months versus 21.2 months [hazard ratio (HR), 0.67; 95% confidence interval (CI) 0.46–0.98;P = 0.037]. PCI was associated with a decrease in risk of brain metastases (the actuarial 5-year brain metastases rate, 20.3% versus 49.9%; HR, 0.28; 95% CI 0.14–0.57;P Conclusion In patients with fully resected postoperative pathologically confirmed stage IIIA–N2 NSCLC and high risk of cerebral metastases after adjuvant chemotherapy, PCI prolongs DFS and decreases the incidence of brain metastases.

MicroRNA166 (miR166) is known to have highly conserved targets that encode proteins of the class III homeodomain-leucine zipper (HD-ZIP III) family, in a broad range of plant species. To further understand the relationship between HD-ZIP III genes and miR166, four HD-ZIP III family genes (PpHB14, PpHB15, PpHB8, and PpREV) were isolated from peach (Prunus persica) tissue and characterized. Spatio-temporal expression profiles of the genes were analyzed. Genes of the peach HD-ZIP III family were predicted to encode five conserved domains. Deduced amino acid sequences and tertiary structures of the four peach HD-ZIP III genes were highly conserved, with corresponding genes in Arabidopsis thaliana. The expression level of four targets displayed the opposite trend to that of miR166 throughout fruit development, with the exception of PpHB14 from 35 to 55 days after full bloom (DAFB). This finding indicates that miR166 may negatively regulate its four targets throughout fruit development. As for leaf and phloem, the same trend in expression level was observed between four targets and miR166 from 75 to 105 DAFB. However, the opposite trend was observed for the transcript level between four targets and miR166 from 35 to 55 DAFB. miRNA166 may negatively regulate four targets in some but not all developmental stages for a given tissue. The four genes studied were observed to have, exactly or generally, the same change tendency as individual tissue development, a finding that suggests genes of the HD-ZIP III family in peach may have complementary or cooperative functions in various tissues.

SummaryThe late-ripening peach [Prunus persica (L.) Batsch] cultivar ‘Guibao’ was chosen in order to study the effect of different bagging treatments on the appearance, skin quality, and internal quality of fruit. Diurnal variations in temperature and relative humidity (RH) in white single-layer paper bags (WPB), white single-layer polypropylene non-woven bags (WPPB), and black single-layer paper bags (BPB) were recorded for 15 d before harvest. Bagging treatments were applied 30 d after full bloom (DAFB). The WPB and WPPB treatments continued until harvest, while the BPB were removed 7 d before harvest. Observations were recorded on skin colour and fruit quality attributes such as fruit fresh weight (FW), flesh firmness, total soluble solids concentration (TSSC), colour, skin pigments, soluble sugar concentrations (SSC) and organic acid concentrations. In higher plants, the transcription factor MYB10 is an important regulator of anthocyanin biosynthesis. In order to study its role in the development of red coloration in peach leaves, the full-length MYB10 complementary DNA sequence of the red-leaf peach cultivar 'Tsukuba No. 5' (Prunus persica f. atropurpurea) was successfully cloned using reverse transcription-polymerase chain reaction. The sequence was assigned the GenBank accession No. KP315904. Bioinformatic analysis identified the complete MYB10 open reading frame, consisting of 678 bp encoding 225 amino acids. The predicted protein has a molecular weight of 26.56 kDa and a theoretical isoelectric point of 8.97. The secondary structure was found to comprise approximately 34.22% alpha helix, 15.11% extended strand, 10.67% beta turn, and 40% random coil. Subcellular analysis indicated that MYB10 may function in the cytoplasm. Assessment of the amino acid sequence suggested the presence of one serine and two threonine phosphorylation sites. Quantitative real-time polymerase chain reaction revealed that MYB10 expression positively correlated with anthocyanin content in red-leaf peach, indicating that this transcription factor plays a role in the biosynthesis of this pigment in peach trees.

How moisture limitation, P availability and fungal inoculation interact to affect plant physiology and growth is, however, poorly understood. Here, we studied the combined effects of fungal (arbuscular mycorrhizal fungi (AMF) or Penicillium spp.) inoculations and phosphorus (P) fertilization (0, 45 and 90 kg ha � 1 ) on the net rate of photosynthesis, water-use efficiency, P uptake and growth of spring wheat (Triticum aestivum var. Superb) under field conditions at two locations (Castor and Vegreville) in Alberta, Canada. Both fungal inoculation and P application increased the rate of photosynthesis. Under the same P level, AMF inoculation had a greater positive effect on the rate of photosynthesis than Penicillium inoculation. The AMF inoculation increased the instantaneous water-use efficiency (WUEi) of plants at Castor, but not at Vegreville. Leaf carbon isotope discrimination (CID, D 13 C) increased with the rate of P application but was not affected by fungal inoculations. Phosphorus concentrations of stem and seed increased with both fungal inoculation and P application irrespective of location, with AMF inoculation showing the largest effects. The interaction between P addition and fungal inoculation was significant for stem P concentration in Vegreville. Both fungal inoculation and P application increased the leaf area index (LAI), biomass production and grain yield at both locations. Under the same P level, AMF inoculation had a greater positive effect on LAI, biomass production and grain yields than Penicillium inoculation. Morphological characters such as spike length and kernels/spike were also improved by fungal inoculation and P application at both locations. We conclude that the studied sites were deficient in P availability, and both fungal inoculation and P application improved P uptake and crop productivity, while the effect of fungal inoculation on water-use efficiency was site specific.

Obesity and type 2 diabetes mellitus (T2DM) are associated with dysfunctional insulin signalling and impaired central glucose sensing. Glucose sensing neurones reside in key areas of the brain involved in glucose and energy homeostasis (e.g. ventromedial hypothalamus; VMH). We have recently shown that insulin attenuates the ability of glucose-excited (GE) neurones to sense decreased glucose. We hypothesise that this effect of insulin on VMH GE neurones is impaired during T2DM when insulin signalling is dysfunctional. To test our hypotheses, we used whole cell patch clamp recording techniques to evaluate the effects of insulin on VMH GE neurones in brain slices from wild-type and diabetic (db/db) mice. The effects of decreasing glucose from 2.5 to 0.1 mm on VMH GE neurones were similar in wild-type and db/db mice. However, decreasing glucose from 2.5 to 0.5 mm decreased the action potential frequency, membrane potential and input resistance of VMH GE neurones to a significantly greater extent in db/db versus wild-type mice. Furthermore, insulin (5 nm) blunted the effects of decreased glucose in wild-type, but not db/db mice. These differences in both glucose and insulin sensitivity between wild-type and db/db mice were completely ameliorated by the insulin sensitiser, Compound 2 (300 nm). These data are consistent with our hypothesis that impaired insulin signalling in T2DM sensitises VMH GE neurones to decreased glucose.

AMP-activated protein kinase (AMPK)-mediated cellular metabolic responses to tissue-specific and whole-body stimuli play a vital role in the control of energy homeostasis. As a cellular energy-sensing mechanism, AMPK activation stimulates glucose uptake and fat oxidation, while it suppresses lipogenesis and gluconeogenesis. The cumulative effects of AMPK activation lead to beneficial metabolic states in liver, muscle and other peripheral tissues that are critical in the pathogenesis of obesity, type 2 diabetes and related metabolic disorders. Activators of AMPK that target selected tissues hold potential as novel therapeutics for diseases in which altered energy metabolism contributes to aetiology.

Long-duration Gamma-Ray Bursts (GRBs) are an extremely rare outcome of the collapse of massive stars, and are typically found in the distant Universe. Because of its intrinsic luminosity ($L\sim 3 \times 10^{53}$ erg s$^{-1}$) and its relative proximity ($z=0.34$), GRB 130427A was a unique event that reached the highest fluence observed in the gamma-ray band. Here we present a comprehensive multiwavelength view of GRB 130427A with Swift, the 2-m Liverpool and Faulkes telescopes and by other ground-based facilities, highlighting the evolution of the burst emission from the prompt to the afterglow phase. The properties of GRB 130427A are similar to those of the most luminous, high-redshift GRBs, suggesting that a common central engine is responsible for producing GRBs in both the contemporary and the early Universe and over the full range of GRB isotropic energies., 64 pages, 11 figures; published in Science.
