1. Intra-islet glucagon signalling regulates beta-cell connectivity, first-phase insulin secretion and glucose homoeostasis

Abstract: Objective: Type 2 diabetes (T2D) is characterised by the loss of first-phase insulin secretion. We studied mice with β-cell selective loss of the glucagon receptor (Gcgrfl/fl X Ins-1Cre), to investigate the role of intra-islet glucagon receptor (GCGR) signalling on pan-islet [Ca2+]I activity and insulin secretion. Methods: Metabolic profiling was conducted on Gcgrβ-cell−/− and littermate controls. Crossing with GCaMP6f (STOP flox) animals further allowed for β-cell specific expression of a fluorescent calcium indicator. These islets were functionally imaged in vitro and in vivo. Wild-type mice were transplanted with islets expressing GCaMP6f in β-cells into the anterior eye chamber and placed on a high fat diet. Part of the cohort received a glucagon analogue (GCG-analogue) for 40 days and the control group were fed to achieve weight matching. Calcium imaging was performed regularly during the development of hyperglycaemia and in response to GCG-analogue treatment. Results: Gcgrβ-cell−/− mice exhibited higher glucose levels following intraperitoneal glucose challenge (control 12.7 mmol/L ± 0.6 vs. Gcgrβ-cell−/− 15.4 mmol/L ± 0.0 at 15 min, p = 0.002); fasting glycaemia was not different to controls. In vitro, Gcgrβ-cell−/− islets showed profound loss of pan-islet [Ca2+]I waves in response to glucose which was only partially rescued in vivo. Diet induced obesity and hyperglycaemia also resulted in a loss of co-ordinated [Ca2+]I waves in transplanted islets. This was reversed with GCG-analogue treatment, independently of weight-loss (n = 8). Conclusion: These data provide novel evidence for the role of intra-islet GCGR signalling in sustaining synchronised [Ca2+]I waves and support a possible therapeutic role for glucagonergic agents to restore the insulin secretory capacity lost in T2D.

Abstract: The IceCube Neutrino Observatory instruments about 1 km3 of deep, glacial ice at the geographic South Pole. It uses 5160 photomultipliers to detect Cherenkov light emitted by charged relativistic particles. An unexpected light propagation effect observed by the experiment is an anisotropic attenuation, which is aligned with the local flow direction of the ice. We examine birefringent light propagation through the polycrystalline ice microstructure as a possible explanation for this effect. The predictions of a first-principles model developed for this purpose, in particular curved light trajectories resulting from asymmetric diffusion, provide a qualitatively good match to the main features of the data. This in turn allows us to deduce ice crystal properties. Since the wavelength of the detected light is short compared to the crystal size, these crystal properties include not only the crystal orientation fabric, but also the average crystal size and shape, as a function of depth. By adding small empirical corrections to this first-principles model, a quantitatively accurate description of the optical properties of the IceCube glacial ice is obtained. In this paper, we present the experimental signature of ice optical anisotropy observed in IceCube light-emitting diode (LED) calibration data, the theory and parameterization of the birefringence effect, the fitting procedures of these parameterizations to experimental data, and the inferred crystal properties.

Abstract: The Module-0 Demonstrator is a single-phase 600 kg liquid argon time projection chamber operated as a prototype for the DUNE liquid argon near detector. Based on the ArgonCube design concept, Module-0 features a novel 80k-channel pixelated charge readout and advanced high-coverage photon detection system. In this paper, we present an analysis of an eight-day data set consisting of 25 million cosmic ray events collected in the spring of 2021. We use this sample to demonstrate the imaging performance of the charge and light readout systems as well as the signal correlations between the two. We also report argon purity and detector uniformity measurements and provide comparisons to detector simulations.

Abstract: Abstract The Pandora Software Development Kit and algorithm libraries provide pattern-recognition logic essential to the reconstruction of particle interactions in liquid argon time projection chamber detectors. Pandora is the primary event reconstruction software used at ProtoDUNE-SP, a prototype for the Deep Underground Neutrino Experiment far detector. ProtoDUNE-SP, located at CERN, is exposed to a charged-particle test beam. This paper gives an overview of the Pandora reconstruction algorithms and how they have been tailored for use at ProtoDUNE-SP. In complex events with numerous cosmic-ray and beam background particles, the simulated reconstruction and identification efficiency for triggered test-beam particles is above 80% for the majority of particle type and beam momentum combinations. Specifically, simulated 1 GeV/c charged pions and protons are correctly reconstructed and identified with efficiencies of 86.1 $$\pm 0.6$$ ± 0.6 % and 84.1 $$\pm 0.6$$ ± 0.6 %, respectively. The efficiencies measured for test-beam data are shown to be within 5% of those predicted by the simulation.

Abstract: Abstract Liquid argon time projection chamber detector technology provides high spatial and calorimetric resolutions on the charged particles traversing liquid argon. As a result, the technology has been used in a number of recent neutrino experiments, and is the technology of choice for the Deep Underground Neutrino Experiment (DUNE). In order to perform high precision measurements of neutrinos in the detector, final state particles need to be effectively identified, and their energy accurately reconstructed. This article proposes an algorithm based on a convolutional neural network to perform the classification of energy deposits and reconstructed particles as track-like or arising from electromagnetic cascades. Results from testing the algorithm on experimental data from ProtoDUNE-SP, a prototype of the DUNE far detector, are presented. The network identifies track- and shower-like particles, as well as Michel electrons, with high efficiency. The performance of the algorithm is consistent between experimental data and simulation.

8. Inter-annual variability patterns of reef cryptobiota in the central Red Sea across a shelf gradient

Abstract: Abstract The combination of molecular tools, standard surveying techniques, and long-term monitoring programs are relevant to understanding environmental and ecological changes in coral reef communities. Here we studied temporal variability in cryptobenthic coral reef communities across the continental shelf in the central Red Sea spanning 6 years (three sampling periods: 2013–2019) and including the 2015 mass bleaching event. We used a combination of molecular tools (barcoding and metabarcoding) to assess communities on Autonomous Reef Monitoring Structures (ARMS) as a standardized sampling approach. Community composition associated with ARMS for both methodologies (barcoding and metabarcoding) was statistically different across reefs (shelf position) and time periods. The partition of beta diversity showed a higher turnover and lower nestedness between pre-bleaching and post-bleaching samples than between the two post-bleaching periods, revealing a community shift from the bleaching event. However, a slight return to the pre-bleaching community composition was observed in 2019 suggesting a recovery trajectory. Given the predictions of decreasing time between bleaching events, it is concerning that cryptobenthic communities may not fully recover and communities with new characteristics will emerge. We observed a high turnover among reefs for all time periods, implying a homogenization of the cryptobiome did not occur across the cross shelf following the 2015 bleaching event. It is possible that dispersal limitations and the distinct environmental and benthic structures present across the shelf maintained the heterogeneity in communities among reefs. This study has to the best of our knowledge presented for the first time a temporal aspect into the analysis of ARMS cryptobenthic coral reef communities and encompasses a bleaching event. We show that these structures can detect cryptic changes associated with reef degradation and provides support for these being used as long-term monitoring tools.

9. Myocarditis following COVID-19 mRNA vaccinations: Twin and sibling case series

Abstract: ImportanceMyocarditis and myopericarditis are well described adverse events of special interest (AESI) following COVID-19 vaccinations. Whilst the aetiology is still being investigated; there is evidence that genetic predisposition may be a risk factor for the development of myocarditis. Furthermore, hormones are thought to contribute to sex-specific differences in myocarditis, skewed toward a larger risk in adolescent males. Objective: This unique sibling case series may help highlight potential mechanisms and prognostic factors in the development of myocarditis following COVID-19 vaccination in adolescent males. In this context, twin and familial studies provide a unique epidemiological perspective to investigate the interplay between genetic predisposition and other factors. Participants: Observational case series of all siblings reported to SAEFVIC11 SAEFVIC: Surveillance of Adverse Events Following Vaccination in the Community. with chest pain following COVID-19 vaccinations in Victoria, Australia. Exposure: mRNA vaccination (Comirnaty BNT162b2 COVID-19 (Pfizer-BioNTech) and Spikevax mRNA-1273 (Moderna). Findings: Our case series comprises 6 young males; two sets of monozygotic twins and one set of fraternal brothers following reports of chest pain associated with COVID-19 mRNA vaccination. Five patients were diagnosed with myocarditis as per Brighton Collaboration Criteria (Level 2). The remaining sibling, who did not have myocarditis, was subsequently diagnosed with pubertal delay. Conclusions: Understanding the genetic and hormonal risk factors and aetiology for myocarditis associated with COVID-19 vaccines is paramount. Further evaluation of specific genetic targets or biomarkers is required to understand the implications of population vaccine policy, particularly for adolescent and young adult males at highest risk for this AESI.

10. AUTOMATED RECOGNITION OF PERMAFROST DISTURBANCES USING HIGH-SPATIAL RESOLUTION SATELLITE IMAGERY AND DEEP LEARNING MODELS

Abstract: The accelerated warming conditions of the high Arctic have intensified the extensive thawing of permafrost. Retrogressive thaw slumps (RTSs) are considered as the most active landforms in the Arctic permafrost. An increase in RTSs has been observed in the Arctic in recent decades. Continuous monitoring of RTSs is important to understand climate change-driven disturbances in the region. Manual detection of these landforms is extremely difficult as they occur over exceptionally large areas. Only very few studies have explored the utility of very high spatial resolution (VHSR) commercial satellite imagery in the automated mapping of RTSs. We have developed deep learning (DL) convolution neural net (CNN) based workflow to automatically detect RTSs from VHRS satellite imagery. This study systematically compared the performance of different DLCNN model architectures and varying backbones. Our candidate CNN models include: DeepLabV3+, UNet, UNet++, Multi-scale Attention Net (MA-Net), and Pyramid Attention Network (PAN) with ResNet50, ResNet101 and ResNet152 backbones. The RTS modeling experiment was conducted on Banks Island and Ellesmere Island in Canada. The UNet++ model demonstrated the highest accuracy (F1 score of 87%) with the ResNet50 backbone at the expense of training and inferencing time. PAN, DeepLabV3, MaNet, and UNet, models reported mediocre F1 scores of 72%, 75%, 80%, and 81% respectively. Our findings unravel the performances of different DLCNNs in imagery-enabled RTS mapping and provide useful insights on operationalizing the mapping application across the Arctic.

Abstract: Abstract DUNE is a dual-site experiment for long-baseline neutrino oscillation studies, neutrino astrophysics and nucleon decay searches. ProtoDUNE Dual Phase (DP) is a 6 $$\times $$ × 6 $$\times $$ × 6 m $$^3$$ 3 liquid argon time-projection-chamber (LArTPC) that recorded cosmic-muon data at the CERN Neutrino Platform in 2019–2020 as a prototype of the DUNE Far Detector. Charged particles propagating through the LArTPC produce ionization and scintillation light. The scintillation light signal in these detectors can provide the trigger for non-beam events. In addition, it adds precise timing capabilities and improves the calorimetry measurements. In ProtoDUNE-DP, scintillation and electroluminescence light produced by cosmic muons in the LArTPC is collected by photomultiplier tubes placed up to 7 m away from the ionizing track. In this paper, the ProtoDUNE-DP photon detection system performance is evaluated with a particular focus on the different wavelength shifters, such as PEN and TPB, and the use of Xe-doped LAr, considering its future use in giant LArTPCs. The scintillation light production and propagation processes are analyzed and a comparison of simulation to data is performed, improving understanding of the liquid argon properties.

Abstract: Lakes in permafrost regions are dynamic landscape components and play an important role for climate change feedbacks. Lake processes such as mineralization and flocculation of dissolved organic carbon (DOC), one of the main carbon fractions in lakes, contribute to the greenhouse effect and are part of the global carbon cycle. These processes are in the focus of climate research, but studies so far are limited to specific study regions. In our synthesis, we analyzed 2167 water samples from 1833 lakes across the Arctic in permafrost regions of Alaska, Canada, Greenland, and Siberia to provide first pan-Arctic insights for linkages between DOC concentrations and the environment. Using published data and unpublished datasets from the author team, we report regional DOC differences linked to latitude, permafrost zones, ecoregions, geology, near-surface soil organic carbon contents, and ground ice classification of each lake region. The lake DOC concentrations in our dataset range from 0 to 1130 mg L−1 (10.8 mg L−1 median DOC concentration). Regarding the permafrost regions of our synthesis, we found median lake DOC concentrations of 12.4 mg L−1 (Siberia), 12.3 mg L−1 (Alaska), 10.3 mg L−1 (Greenland), and 4.5 mg L−1 (Canada). Our synthesis shows a significant relationship between lake DOC concentration and lake ecoregion. We found higher lake DOC concentrations at boreal permafrost sites compared to tundra sites. We found significantly higher DOC concentrations in lakes in regions with ice-rich syngenetic permafrost deposits (yedoma) compared to non-yedoma lakes and a weak but significant relationship between soil organic carbon content and lake DOC concentration as well as between ground ice content and lake DOC. Our pan-Arctic dataset shows that the DOC concentration of a lake depends on its environmental properties, especially on permafrost extent and ecoregion, as well as vegetation, which is the most important driver of lake DOC in this study. This new dataset will be fundamental to quantify a pan-Arctic lake DOC pool for estimations of the impact of lake DOC on the global carbon cycle and climate change.

Abstract: Abstract The deep underground neutrino experiment (DUNE), a 40-kton underground liquid argon time projection chamber experiment, will be sensitive to the electron-neutrino flavor component of the burst of neutrinos expected from the next Galactic core-collapse supernova. Such an observation will bring unique insight into the astrophysics of core collapse as well as into the properties of neutrinos. The general capabilities of DUNE for neutrino detection in the relevant few- to few-tens-of-MeV neutrino energy range will be described. As an example, DUNE’s ability to constrain the $$\nu _e$$ ν e spectral parameters of the neutrino burst will be considered.

Abstract: Abstract The Deep Underground Neutrino Experiment (DUNE) will be a powerful tool for a variety of physics topics. The high-intensity proton beams provide a large neutrino flux, sampled by a near detector system consisting of a combination of capable precision detectors, and by the massive far detector system located deep underground. This configuration sets up DUNE as a machine for discovery, as it enables opportunities not only to perform precision neutrino measurements that may uncover deviations from the present three-flavor mixing paradigm, but also to discover new particles and unveil new interactions and symmetries beyond those predicted in the Standard Model (SM). Of the many potential beyond the Standard Model (BSM) topics DUNE will probe, this paper presents a selection of studies quantifying DUNE’s sensitivities to sterile neutrino mixing, heavy neutral leptons, non-standard interactions, CPT symmetry violation, Lorentz invariance violation, neutrino trident production, dark matter from both beam induced and cosmogenic sources, baryon number violation, and other new physics topics that complement those at high-energy colliders and significantly extend the present reach.

Abstract: Abstract Background Symptoms arising from vestibular system dysfunction are observed in 49–59% of people with Multiple Sclerosis (MS). Symptoms may include vertigo, dizziness and/or imbalance. These impact on functional ability, contribute to falls and significant health and social care costs. In people with MS, vestibular dysfunction can be due to peripheral pathology that may include Benign Paroxysmal Positional Vertigo (BPPV), as well as central or combined pathology. Vestibular symptoms may be treated with vestibular rehabilitation (VR), and with repositioning manoeuvres in the case of BPPV. However, there is a paucity of evidence about the rate and degree of symptom recovery with VR for people with MS and vestibulopathy. In addition, given the multiplicity of symptoms and underpinning vestibular pathologies often seen in people with MS, a customised VR approach may be more clinically appropriate and cost effective than generic booklet-based approaches. Likewise, BPPV should be identified and treated appropriately. Methods/ design People with MS and symptoms of vertigo, dizziness and/or imbalance will be screened for central and/or peripheral vestibulopathy and/or BPPV. Following consent, people with BPPV will be treated with re-positioning manoeuvres over 1–3 sessions and followed up at 6 and 12 months to assess for any re-occurrence of BPPV. People with central and/or peripheral vestibulopathy will be entered into a randomised controlled trial (RCT). Trial participants will be randomly allocated (1:1) to either a 12-week generic booklet-based home programme with telephone support or a 12-week VR programme consisting of customised treatment including 12 face-to-face sessions and a home exercise programme. Customised or booklet-based interventions will start 2 weeks after randomisation and all trial participants will be followed up 14 and 26 weeks from randomisation. The primary clinical outcome is the Dizziness Handicap Inventory at 26 weeks and the primary economic endpoint is quality-adjusted life-years. A range of secondary outcomes associated with vestibular function will be used. Discussion If customised VR is demonstrated to be clinically and cost-effective compared to generic booklet-based VR this will inform practice guidelines and the development of training packages for therapists in the diagnosis and treatment of vestibulopathy in people with MS. Abstract: Abstract The sensitivity of the Deep Underground Neutrino Experiment (DUNE) to neutrino oscillation is determined, based on a full simulation, reconstruction, and event selection of the far detector and a full simulation and parameterized analysis of the near detector. Detailed uncertainties due to the flux prediction, neutrino interaction model, and detector effects are included. DUNE will resolve the neutrino mass ordering to a precision of 5 $$\sigma $$ σ , for all $$\delta _{\mathrm{CP}}$$ δ CP values, after 2 years of running with the nominal detector design and beam configuration. It has the potential to observe charge-parity violation in the neutrino sector to a precision of 3 $$\sigma $$ σ (5 $$\sigma $$ σ ) after an exposure of 5 (10) years, for 50% of all $$\delta _{\mathrm{CP}}$$ δ CP values. It will also make precise measurements of other parameters governing long-baseline neutrino oscillation, and after an exposure of 15 years will achieve a similar sensitivity to $$\sin ^{2} 2\theta _{13}$$ sin 2 2 θ 13 to current reactor experiments.

21. Temporal evolution of temperatures in the Red Sea and the Gulf of Aden based on in situ observations (1958–2017)

Abstract: The Red Sea holds one of the most diverse marine ecosystems in the world, although fragile and vulnerable to ocean warming. Several studies have analysed the spatio-temporal evolution of temperature in the Red Sea using satellite data, thus focusing only on the surface layer and covering the last ∼30 years. To better understand the long-term variability and trends of temperature in the whole water column, we produce a 3-D gridded temperature product (TEMPERSEA) for the period 1958–2017, based on a large number of in situ observations, covering the Red Sea and the Gulf of Aden. After a specific quality control, a mapping algorithm based on optimal interpolation have been applied to homogenize the data. Also, an estimate of the uncertainties of the product has been generated. The calibration of the algorithm and the uncertainty computation has been done through sensitivity experiments based on synthetic data from a realistic numerical simulation. TEMPERSEA has been compared to satellite observations of sea surface temperature for the period 1981–2017, showing good agreement especially in those periods when a reasonable number of observations were available. Also, very good agreement has been found between air temperatures and reconstructed sea temperatures in the upper 100 m for the whole period 1958–2017, enhancing confidence in the quality of the product. The product has been used to characterize the spatio-temporal variability of the temperature field in the Red Sea and the Gulf of Aden at different timescales (seasonal, interannual and multidecadal). Clear differences have been found between the two regions suggesting that the Red Sea variability is mainly driven by air–sea interactions, while in the Gulf of Aden the lateral advection of water plays a relevant role. Regarding long-term evolution, our results show only positive trends above 40 m depth, with maximum trends of 0.045 + 0.016 ∘C decade−1 at 15 m, and the largest negative trends at 125 m (-0.072+0.011 ∘C decade−1). Multidecadal variations have a strong impact on the trend computation and restricting them to the last 30–40 years of data can bias high the trend estimates.

Abstract: The Deep Underground Neutrino Experiment (DUNE) is an international, world-class experiment aimed at exploring fundamental questions about the universe that are at the forefront of astrophysics and particle physics research. DUNE will study questions pertaining to the preponderance of matter over antimatter in the early universe, the dynamics of supernovae, the subtleties of neutrino interaction physics, and a number of beyond the Standard Model topics accessible in a powerful neutrino beam. A critical component of the DUNE physics program involves the study of changes in a powerful beam of neutrinos, i.e., neutrino oscillations, as the neutrinos propagate a long distance. The experiment consists of a near detector, sited close to the source of the beam, and a far detector, sited along the beam at a large distance. This document, the DUNE Near Detector Conceptual Design Report (CDR), describes the design of the DUNE near detector and the science program that drives the design and technology choices. The goals and requirements underlying the design, along with projected performance are given. It serves as a starting point for a more detailed design that will be described in future documents.

Abstract: Ponds and lakes are abundant in Arctic permafrost lowlands. They play an important role in Arctic wetland ecosystems by regulating carbon, water, and energy fluxes and providing freshwater habitats. However, ponds, i.e., waterbodies with surface areas smaller than 1. 0 × 104 m2, have not been inventoried on global and regional scales. The Permafrost Region Pond and Lake (PeRL) database presents the results of a circum-Arctic effort to map ponds and lakes from modern (2002–2013) high-resolution aerial and satellite imagery with a resolution of 5 m or better. The database also includes historical imagery from 1948 to 1965 with a resolution of 6 m or better. PeRL includes 69 maps covering a wide range of environmental conditions from tundra to boreal regions and from continuous to discontinuous permafrost zones. Waterbody maps are linked to regional permafrost landscape maps which provide information on permafrost extent, ground ice volume, geology, and lithology. This paper describes waterbody classification and accuracy, and presents statistics of waterbody distribution for each site. Maps of permafrost landscapes in Alaska, Canada, and Russia are used to extrapolate waterbody statistics from the site level to regional landscape units. PeRL presents pond and lake estimates for a total area of 1. 4 × 106 km2 across the Arctic, about 17 % of the Arctic lowland ( 6 m2 down to 1. 0 × 102 m2 contributed up to 21 % to the total water fraction. Waterbody density ranged from 1. 0 × 10 to 9. 4 × 101 km−2. Ponds are the dominant waterbody type by number in all landscapes representing 45–99 % of the total waterbody number. The implementation of PeRL size distributions in land surface models will greatly improve the investigation and projection of surface inundation and carbon fluxes in permafrost lowlands. 27. Repeat-induced point mutations driving Parastagonospora nodorum genomic diversity are balanced by selection against non-synonymous mutations

Abstract: Abstract Parastagonospora nodorum is necrotrophic fungal pathogen of wheat with significant genomic resources. Population-level pangenome data for 173 isolates, of which 156 were from Western Australia (WA) and 17 were international, were examined for overall genomic diversity and effector gene content. A heterothallic core population occurred across all regions of WA, with asexually-reproducing clonal clusters in dryer northern regions. High potential for SNP diversity in the form of repeat-induced point mutation (RIP)-like transitions, was observed across the genome, suggesting widespread ‘RIP-leakage’ from transposon-rich repetitive sequences into non-repetitive regions. The strong potential for RIP-like mutations was balanced by negative selection against non-synonymous SNPs, that was observed within protein-coding regions. Protein isoform profiles of known effector loci (SnToxA, SnTox1, SnTox3, SnTox267, and SnTox5) indicated low-levels of non-synonymous and high-levels of silent RIP-like mutations. Effector predictions identified 186 candidate secreted predicted effector proteins (CSEPs), 69 of which had functional annotations and included confirmed effectors. Pangenome-based effector isoform profiles across WA were distinct from global isolates and were conserved relative to population structure, and may enable new approaches for monitoring crop disease pathotypes.

Abstract: Many phytopathogenic bacteria require a type III secretion system (TTSS) to activate effector-triggered immunity (ETI). We identified a calcium-binding protein, EfhXXfa, in the citrus pathogen Xanthomonas citri subsp. aurantifolii that does not require a TTSS to activate reactive oxygen species (ROS) and elicit a hypersensitive reaction (HR) in tomato leaves following infection. Purified, recombinant EfhXXfa was shown to bind two moles of calcium per mole of protein, whereas mutation of the first of two EF-hands did not bind calcium. EfhXXfa expression was determined to be inducible in hrp-inducing medium. Additionally, growth of X. perforans transconjugants with and without the efhXXfa gene in hrp-inducing medium differed in intracellular calcium concentration; the transconjugant without efhXXfa yielded higher cell pellet masses and higher increased intracellular calcium concentrations relative to cells expressing EfhXXfa. An EfhXXfa homolog, EfhXXe, present in the pepper pathogen X. euvesicatoria, triggered ROS production and an HR in tomato leaves when expressed in the tomato pathogen X. perforans and is a host-limiting factor. Interestingly, all tested X. perforans and X. euvesicatoria strains pathogenic on tomato contain a stop codon immediately upstream of the first EF-hand domain in the efhXXe gene, whereas most X. euvesicatoria strains pathogenic on pepper do not. Abstract: Abstract Background This study examines the impact of SARS-CoV-2 (i.e., coronavirus, COVID, COVID-19) using data from a measurement-based care (MBC) system utilized in an outpatient psychiatric clinic providing telemedicine care. A novel Patient Rated Outcome Measure (PROM), the COVID-19 Events Checklist (CEC) was administered in a hospital system based ambulatory clinic beginning April 2020 to track COVID-19-19’s impact on patients’ mental, emotional, and health-related behaviors during the pandemic. The study (1) provides descriptive CEC data, and (2) compares CEC results with PROMs evaluating anxiety (Generalized Anxiety Disorder-7; GAD-7), depression (Patient Health Questionnaire; PHQ-9), and psychological distress (Brief Adjustment Scale-6; BASE-6). Methods This retrospective observational study included patient intake data collected from April 2020 to March 2021. Patient (N = 842) reports on the CEC’s five domain questions were aggregated to calculate average reports of COVID-19 related impacts at intake over the initial 12 months of the pandemic. Trends in COVID-19 related impacts were examined, and non-aggregated scores on the PHQ-9, GAD-7, and BASE-6 were compared to primary dichotomous (yes/no) CEC survey questions via Wilcoxon rand sum testing. Results Results capture the relationship between COVID-19 exposure, COVID-19- related sequelae and behaviors, and psychological symptom severity. Specifically, Wilcoxon rank-sum tests indicate that social determinants of health (SDOH), negative mental health impacts, and positive coping skill use were significantly associated with psychological symptomatology including overall psychological functioning via the BASE-6, anxiety via the GAD-7, and depressive symptoms via the PHQ-9. 31. Improving Academic Performance through a School-Based Intervention Targeting Academic Executive Functions -- A Pilot Study

Abstract: Background: Academic challenges such as losing/not turning in assignments, misplacing materials, and inefficient studying are common in middle-school students with autism spectrum disorder (ASD) without intellectual disability. Deficits in organization, planning, prioritizing, memory/materials management, and studying skills [i.e. academic executive functioning (EF) deficits] contribute to these challenges. Objectives: To assess the feasibility, satisfaction, and initial efficacy of the school-based version of the Achieving Independence and Mastery in School (AIMS) intervention in a proof-of-concept trial with 6 students with ASD. Methods: 6 middle-schoolers with ASD without ID participated in AIMS. Parents and teachers rated academic EFs and functioning. Results: Results suggest high feasibility, youth satisfaction, and improved EF skills and academic behaviors by parent and teacher report. Conclusion: These promising results support further intervention development and suggest that academic EF skills are malleable in students with ASD.

32. Multichannel, triaxial, neutron time-of-flight diagnostic for experiments at the Z facility

Abstract: Neutron bang times t_{bang} and mean neutron speeds u[over ¯] have been measured at the Z facility for a series of D_{2}-filled targets, in magnetized liner inertial fusion experiments. Measurements were made by a novel neutron time-of-flight (nTOF) diagnostic, adapted for use at this facility, and consisted of detecting the neutron times in flight at seven independent scintillator–photomultiplier tube detectors (channels), located on three noncoplanar lines of sight, with distances to the neutron source varying between 690 and 2510 cm. The nTOF signals were analyzed by identifying fiducials on the detector traces to quantify the time in flight to each distance, using a nonrelativistic model for a uniformly thermalized, Maxwellian plasma distribution. The measured neutron arrival times were then linearly regressed on distance with the bang time and mean speed estimated from the fit parameters. A particular shot, 2584, is analyzed here to illustrate the method and the issues encountered in these measurements. On this particular shot, six usable channel traces were obtained. The standard errors of the parameter fits were as follows: t_{bang}=3102.95±0.97 ns (standard error) with six nTOF traces on the system clock and u[over ¯]=2.1524±0.0032 cm/ns (standard error), from which the mean, nonrelativistic, kinetic energy E[over ¯] of the neutrons was 2.4216±0.0144 MeV (standard error). The estimates of u[over ¯] and E[over ¯] here agree within 1% of the published values for the D(d,n)^{3}He reaction. Hence, these measurements are consistent with the production of a thermalized, Maxwellian D-D fusion plasma in this experiment. The source duration was estimated to be 3.25±0.84 ns (standard error) from six pulse-width measurements.

Abstract: Agencies from CEOS (Committee on Earth Observation Satellites) have traditionally focused their efforts on the response phase. Rapid urbanization and increased severity of weather events has led to growing economic and human losses from disasters, requiring international organisations to act now in all disaster risk management (DRM) phases, especially through improved disaster risk reduction policies and programmes. As part of this effort, CEOS agencies have initiated a series of actions aimed at fostering the use of Earth observation (EO) data to support disaster risk reduction and at raising the awareness of policy and decision-makers and major stakeholders of the benefits of using satellite EO in all phases of DRM. CEOS is developing a long-term vision for sustainable application of satellite EO to all phases of DRM. CEOS is collaborating with regional representatives of the DRM user community, on a multi-hazard project involving three thematic pilots (floods, seismic hazards and volcanoes) and a Recovery Observatory that supports resilient recovery from one major disaster. These pilot activities are meant as trail blazers that demonstrate the potential offered by satellite EO for comprehensive DRM. In the framework of the 2015 3rd World Conference on Disaster Risk Reduction (WCDRR), the CEOS space agencies intend to partner with major stakeholders, including UN organizations, the Group on Earth Observations (GEO), international relief agencies, leading development banks, and leading regional DRM organisations, to define and implement a 15-year plan of actions (2015- 2030) that responds to high-level Post-2015 Framework for Disaster Risk Reduction priorities. This plan of action will take into account lessons learned from the CEOS pilot activities.

Abstract: ABSTRACT The widespread use of antimicrobials that target bacterial pathogens has driven evolution of resistance, compromising the efficacy of these bactericides. Understanding the emergence and spread of resistance genes via mobile genetic elements is crucial for combating antimicrobial resistance. Copper resistance (CuR) in Xanthomonas euvesicatoria pv. perforans has severely affected the efficacy of copper-based bactericides for controlling bacterial leaf spot disease of tomato and pepper. Here, we investigated the evolutionary pathways of CuR acquisition and dissemination in X. euvesicatoria pv. perforans using an extensive collection of strains. We determined that chromosomally encoded CuR predominates over plasmid-borne CuR in multiple distinct phylogenetic groups of X. euvesicatoria pv. perforans. Our analysis revealed a single site of chromosomal integration by a CuR genomic island, although the genomic island showed sequence variation among phylogenetic groups. While chromosomal CuR was more prevalent, strains with plasmid-borne resistance conferred greater copper tolerance. Additionally, we identified strains carrying two copies of CuR genes, on plasmid and chromosome, that exhibited increased copper tolerance. Strains of X. euvesicatoria pv. perforans from the USA shared identical CuR gene sequences whether on plasmids or chromosome while different alleles were found in strains from other countries. In contrast to X. euvesicatoria pv. perforans, plasmid-borne CuR predominated in closely related pathovar, X. euvesicatoria pv. euvesicatoria. Overall, these findings contribute to a better understanding of the evolution and persistence of CuR in X. euvesicatoria pv. perforans and its closest relatives.IMPORTANCEThe emergence of antimicrobial resistance is a significant threat to agricultural production as it reduces the efficacy of various antimicrobials including copper-based bactericides that are widely used to control plant diseases. The challenge of increasing antimicrobial resistance entering a production system necessitates a deeper understanding of the dynamics and mechanisms by which pathogens acquire resistance. As a result of this research, we have identified different mechanisms of copper resistance acquisition as well as levels of copper resistance in a devastating plant pathogen, X. euvesicatoria pv. perforans. The evolution and dissemination of copper resistance in strains through plasmid or chromosomally integrated genomic island or both presents barriers to current management approaches, where growers rely heavily on copper-based bactericides to manage disease outbreaks. This knowledge is crucial when considering the continued use of existing antimicrobials or adopting alternative antimicrobials in efforts to implement enhanced antimicrobial stewardship strategies in agriculture.

Abstract: Underactuation as a method of driving prosthetic hands has a long history. The pragmatic requirements of such a device to be light enough to be worn and used regularly have meant that any multi degree of freedom prosthetic hand must have fewer actuators than the usable degrees of freedom. Aesthetics ensures that while the hand needs five fingers, five actuators have considerable mass, and only in recent years has it even been possible to construct a practical anthropomorphic hand with five motors. Thus there is an important trade off as to which fingers are driven, and which joints on which fingers are actuated, and how the forces are distributed to create a functional device. This paper outlines some of the historical solutions created for this problem and includes those designs of recent years that are now beginning to be used in the commercial environment. Abstract: Coxsackievirus A6 (CVA6) has become increasingly clinically relevant as a cause of Hand, Foot and Mouth Disease (HFMD) globally since 2008. However, most laboratories do not routinely determine the enteroviral type of positive samples. The non-pharmaceutical measures introduced to curb transmission during the COVID-19 pandemic may also have perturbed CVA6 epidemiology. We thus aimed to determine the prevalence, clinical presentation and genetic relationship of CVA6 across three complete epidemic seasons: one pre-SARS-CoV-2 emergence and two post-SARS-CoV-2 emergence in our regional healthcare setting. Surplus diagnostic nucleic acid from diagnosed enteroviral positives diagnosed between September and December of 2018 and between May 2021 and April of 2023 was subject to VP1 gene sequencing to determine the CVA6 cases and interrogate their phylogenetic relationship. The confirmed CVA6 cases were also retrospectively clinically audited. CVA6 infections were identified in 33 and 69 individuals pre- and post-pandemic, respectively, with cases peaking in November of 2018 and 2022, but in October of 2021. HFMD was the primary diagnosis in 85.5% of the post-pandemic cases, but only 69.7% of the pre-pandemic cases, where respiratory and neurological symptoms (45.5% and 12.1%, respectively) were significantly elevated. A complete VP1 sequence was retrieved for 94% of the CVA6 cases, revealing that studied infections were genetically diverse and suggestive of multiple local and international transmission chains. CVA6 presented a significant clinical burden in our regional U.K. hospital setting both pre- and post-pandemic and was subject to dynamic clinical and genetic epidemiology.

Abstract: A rapid situation analysis was conducted in Kibaha and Ngorongoro districts in Tanzania to map resources as well as analysing emergency preparedness to infectious diseases in animal (domestic and wild) and human populations. Kibaha was chosen as a district close to a commercial city (Dar es Salaam) while Ngorongoro represented a remote, border district with high interactions between humans, domestic and wild animals. In this study, data on resources and personnel as well as emergency preparedness were collected from all wards (n = 22), human health facilities (n = 40) and livestock facilities in the two districts using interview checklists and questionnaires. Descriptive statistics for resources were calculated and mapped by district. Kibaha district had a higher human population density, more health workers, better equipped health facilities and better communication and transport systems. On the other hand, Ngorongoro had a higher population of livestock and more animal health facilities but a poorer ratio of animal health workers to livestock. The average ratio of health personnel to population in catchment areas of the health facilities was 1:147 (range of 1:17−1:1200). The ratio of personnel to human population was significantly higher in Kibaha (1:95) than in Ngorongoro (1:203) district (p = 0 < 0.001). Considering the limited resources available to both human and animal health sectors and their different strengths and weaknesses there are opportunities for greater collaboration and resource-sharing between human and animal health for improved surveillance and emergency-preparedness.
