Summary: Background: Insulin resistance (IR) is an important risk factor for multiple chronic diseases, increasing mortality and reducing life expectancy. The associations between emerging surrogates for IR, triglyceride-glucose index (TyG) and TyG-related indicators, with all-cause mortality and life expectancy in middle-aged and older patients in primary care are unclear. Methods: This study originated from the Polish primary care cohort LIPIDOGRAM2015, including patients aged ≥45 years. Baseline fasting triglycerides and fasting glucose were used to derive TyG. Other TyG-related indicators included TyG-adjusted body mass index (TyG-BMI), TyG-adjusted waist circumference (TyG-WC), TyG-adjusted waist-to-hip, and TyG-adjusted waist-to-height. In this longitudinal analysis, we assessed associations between TyG-related indicators with total all-cause mortality, premature (age at death ≤75 years) all-cause mortality and years of life lost (YLL). Findings: We included 10,688 patients (mean age 61.8 ± 9.3 years; 63.5% female). Cumulative total and premature all-cause mortality were 7.2% and 4.6%, respectively, during 5.7 years (IQR 5.6–5.7) of follow-up. Lowest (Q1) and highest quartile (Q4) of TyG-BMI and TyG-WC were associated with total all-cause mortality (second quartile [Q2]: reference; TyG-BMI: Q1: aHR 1.33, 95% CI 1.07–1.65, Q4: aHR 1.28, 95% CI 1.03–1.58; TyG-WC: Q1: aHR 1.44, 95% CI 1.14–1.82, Q4: aHR 1.29, 95% CI 1.04–1.59), similar results for premature all-cause mortality. Within age 45–80 years, compared with Q2 and third quartile, YLL were 4.49 and 5.46 years for TyG-BMI Q1 and Q4, respectively, 3.24 and 5.31 years for TyG-WC Q1 and Q4, respectively. Interpretation: TyG-BMI and TyG-WC demonstrated a U-shaped association with total and premature all-cause mortality. Low and high levels of TyG-BMI and TyG-WC were associated with reduced life expectancy. Despite the relatively short follow-up period, significant associations were still observed, but longer follow-up studies are required to further explore these relationships. The effect of water bath cooking (WBC), oven convection roasting (OCR), grilling (G), pan frying (PF) on selected physical properties of goose meat was compared in this study. A measurement of cooking loss, texture, color parameters, and sensory evaluation was carried out. The experimental material covered 96 breast muscles cut from carcasses of 17-week-old “Polish oat geese.” The kind of goose meat (with and without skin) and the type of heat treatment affected cooking loss, shear force (SF), and rheological parameters (hardness, cohesiveness, gumminess, and chewiness). The water bath–cooked and pan-fried samples for both kinds of meat were characterized by lower cooking loss than other ones. Goose meat with skin and subcutaneous fat showed higher cooking loss and lower SF value, hardness, gumminess, and chewiness than that without skin for all methods. The water bath–cooked samples were characterized by the lowest SF value, hardness, and chewiness for both kinds of meat. They had the highest value of L∗ parameter and were characterized by a lighter color among others, too. Pan-fried meat showed the highest value of a∗ and lowest of ho parameters; the color of these samples was redder. Moreover, the lower C values of oven convection-roasted and grilled samples showed that they were brighter. The relevance of the study lies in the investigation of the influence of the autonomic nervous system tone on the content of lipids in the body of cows, which is important to consider in order to ensure the high productivity of the dairy herd and balance diets in animal feeding. The purpose of the study was to determine the effect of the tone of the ganglion nervous system on the content of cholesterol and lipoproteins of various densities in the blood of dairy cows of the Ukrainian Black-and-White breed. To determine heart rate variability, an electrocardiograph was used with subsequent measurement of the main indicators according to the Baevsky stress index, which included determining the mode, its amplitude, variation range, autonomous equilibrium index, autonomous rhythm indicator, and stress index. Three experimental groups were formed from experimental cows: normotonics, vagotonics, and sympathotonics. The content of lipoproteins in blood plasma was determined using a Sinnova BS-3000 biochemical analyser (China). According to the results of biochemical analysis, differences in the obtained indicators were established among experimental groups of cows with different tones of the autonomic nervous system. This is due to the fact that activation of the sympathetic nerve branch leads to changes in liver function, which affects the production of glucose in the body. This can lead to increased cholesterol production. In addition, an increased tone of the sympathetic branch can stimulate the activity of enzymes responsible for the synthesis of cholesterol, and lead to a decrease in the level of hormones that regulate its synthesis and transport in the body. In particular, vagotonic animals had the highest blood content of total cholesterol and high-density lipoproteins. Sympathotonics showed the lowest blood values for total cholesterol and high-density lipoproteins. Normotonics occupied an intermediate place among the experimental groups of cows. A detailed study of this issue reveals the features of the influence of the autonomic nervous system tone on the indicators of lipid metabolism in the blood of cows, which is important to consider in production to effectively solve issues related to maintaining their productivity and improving the quality of milk

Breeding of hybrids between wild boars and domestic pigs is in the consumer interest because of the need to ensure food security and diversification via widening the genetic basis of animals reared for meat. To expand the knowledge about their nutritional requirements, this study aimed to investigate hindgut fermentation in these animals. Caecal and colon cultures were incubated for 12 h in vitro with or without wheat bran as a supplementary substrate. Short-chain fatty acids, ammonia, methane, and total gas production were determined. The total concentrations of short-chain fatty acids in unincubated caecal and colon samples were 93.1 and 115 mmol/kg, respectively. The short-chain fatty acid profile in fresh hindgut contents was characterized by a high molar proportion of acetate (74.8-75.0 mol%), followed by propionate (18.2-18.5 mol%) and butyrate (5.4-5.5 mol%). The presence of wheat bran lowered acetate and increased butyrate, propionate, and valerate molar proportions. The ammonia level remained low (1.3-2.43 mmol/kg) regardless of the addition of the substrate. The relatively low pH and ammonia concentration in wild boar/pig hybrids may be caused by the low level of crude protein in diet of these animals. The rate of methanogenesis increased during the fermentation simultaneously with an increase in the production of gases after wheat bran addition. Methane production in the caecal and colon samples incubated with the substrate reached 15.6 and 16.1 mmol/kg, respectively. The hindgut fermentation pattern in wild boar/pig hybrids generally resembled that described earlier in domestic pigs, although some observed dissimilarities may be caused by distinct microbial activity.

Over the past few decades, the advancements in the perinatal and neonatal intensive care have led to a significant survival of premature infants. However neurodevelopmental outcome still remains the topical issues of concern. Developmental care is an approach that is aimed to reduce the mismatches between extra- and intra-uterine environments, decrease the stress of preterm newborns in neonative intensive care units, and thus promote optimal neurobehavioral development of the infant. The Newborn Individualized Developmental Care and Assessment Program (NIDCAP) model was developed as a clinical framework for the implementation of developmental care. The model focuses on detailed reading of each individual infant’s behavioral cues. By observing the child during the routine manipulation performance (before, during and after) and a detailed description of his/her behavioral responses, a professional can assess the ability of the infant’s immature nervous system to tolerate the environment and care manipulations. Such evaluation will enable to determine the adequacy of environmental conditions and care manipulations to baby’s opportunities and needs, with their subsequent correction and adaptation. NIDCAP’s aim is to support the child in its increasing tolerance to stimuli and to minimize stressful events and manipulation. With the help of NIDCAP approaches in neonatal care, medical staff study how to read infants’ behavior, hear their voice and understand them.

The aim of this study was to evaluate the effect of the baical skullcap root (Scutellaria baicalensis radix) on the performance, chemical composition of the muscles and sensory characteristics of the meat of broiler chickens. 120 one-day old Hubbard Hi-Y broiler hybrids were assigned to four groups of 30 each and fed either a basal diet (control) or a basal diet supplemented with 0.5%, 1.0% or 1.5% ground root of skullcap. The body weight, weight gain, feed consumption and feed : gain ratio were calculated after 42 days. The addition of the ground baical skullcap root to the fodder had no effect on the quality or chemical composition of the breast and leg muscles of the broiler chickens. However, it positively influenced some of the performance characteristics of the chickens.

Stiffness modulus represents one of the most important parameters for the mechanical characterization of asphalt mixtures (AMs). At the same time, it is a crucial input parameter in the process of designing flexible pavements. In the present study, two selected mixtures were thoroughly investigated in an experimental trial carried out by means of a four-point bending test (4PBT) apparatus. The mixtures were prepared using spilite aggregate, a conventional 50/70 penetration grade bitumen, and limestone filler. Their stiffness moduli (SM) were determined while samples were exposed to 11 loading frequencies (from 0.1 to 50 Hz) and 4 testing temperatures (from 0 to 30 °C). The SM values ranged from 1222 to 24,133 MPa. Observations were recorded and used to develop a machine learning (ML) model. The main scope was the prediction of the stiffness moduli based on the volumetric properties and testing conditions of the corresponding mixtures, which would provide the advantage of reducing the laboratory efforts required to determine them. Two of the main soft computing techniques were investigated to accomplish this task, namely decision trees with the Categorical Boosting algorithm and artificial neural networks. The outcomes suggest that both ML methodologies achieved very good results, with Categorical Boosting showing better performance (MAPE = 3.41% and R2 = 0.9968) and resulting in more accurate and reliable predictions in terms of the six goodness-of-fit metrics that were implemented.

Fryderyka Chopina w Warszawie. Badaniem ankietowym objęci zostali studenci Uniwersytetu Jagiellońskiego i Politechniki Krakowskiej.

Construction of modern and durable asphalt and cement pavements requires high quality materials and suitable technologies that take into account sustainability concerns which are related to the environmental protection, mitigation and compensation for road construction effects on surface water and groundwater, soil, air, wildlife, landscape, vibration and noise. The objectives of this paper are to identify possible development directions of materials and technologies in road construction in the time perspective of approximately 30 years. In order to achieve that goal a nationwide Delphi survey with 150 invited experts was deployed. The study concluded that binding materials with improved viscoelastic range – and often with specific modifications – would continue to play a leading role. Furthermore, technologies that enable monitoring the state of road pavement condition in a continuous manner would be used to a greater range. Introduction of sensors into the pavement network would lead to the construction of “smart” roads while spreading of nanomaterial technology would improve the durability and reliability of road pavement construction.

Environment conservation and diminishing natural resources caused an increase in popularity of the application of renewable bio-origin resources for the construction of road pavement. Currently, there are known additions of bio-origin materials for bitumen modification. Such material is also used as a flux additive for bitumen or as a rejuvenator once working with reclaimed asphalt pavement (RAP). This paper presents research dealing with asphalt mixtures with RAP modified with a bio-agent of rapeseed origin. The main idea of the conducted research was to apply more RAP content directly to the batch mix plant without extra RAP heating. The RAP used in this study was milled from a base asphalt layer; the addition of RAP stiffens new asphalt mixtures. A bio-agent, due to its fluxing action, was used to support the asphalt mixing process and to decrease the over-stiffening of the mixture caused by RAP addition. This research includes bitumen and mixture tests. For the bitumen study, three different bitumens (35/50, 50/70, and 70/100) were tested in a dynamic shear rheometer (DSR) for complex modulus G* and for phase angle |δ| in the temperature range 0–100 °C. The reference mixture and mixtures with 2.5% bio-agent were tested to assess the influence of RAP and the bio-agent addition on the asphalt mixture properties. Low temperature behavior (TSRST), stiffness, and fatigue resistance (4PB) were tested. Based on the bitumen test, it was determined that even a low rate of bio-agent (2.5%) beneficially changes bitumen properties at a low temperature; moreover, polymerization processes occurring in the second stage of the process improves bitumen properties at a high operational temperature. The research with these asphalt mixtures demonstrates that the bio-origin flux acts as a rejuvenator and allows for an application of 30% cold RAP. Thermal cracking resistance of the mixture with RAP and 2.5% bio-agent improved. The bio-agent removes unfavorable stiffening of RAP and increases the fatigue resistance of the asphalt mixture.

Pot marigold is an ornamental plant of great importance in pharmacy and cosmetology. However, there is limited published information on the diversity of the species at both the morphological and genetic levels. This paper aimed to determine the morphological and genetic diversity of selected marigold varieties. The research focused on varieties with different flower colours, and the most important morphological features of flowers and plant height were analysed. Genetic diversity analyses were performed using start codon target (SCoT) markers. The correlation coefficients between the analysed morphological features of the studied marigold varieties showed a significant positive relationship between plant height, the number of inflorescences per plant and inflorescence diameter. Genetic analyses grouped the studied varieties according to the colour of their flowers. The results obtained in our work indicate that, despite the variability observed in morphological features, the gene pool of the analysed marigold varieties is limited. This information can be valuable for marigold breeders, particularly for selecting varieties suitable for crossbreeding in breeding programs. Purpose: Developing innovative precision and personalized cancer therapeutics is essential to enhance cancer survivability, particularly for prevalent cancer types such as colorectal cancer. This study aims to demonstrate various approaches for discovering new targets for precision therapies using artificial intelligence (AI) on a Polish cohort of colorectal cancer patients., Methods: We analyzed 71 patients with histopathologically confirmed advanced resectional colorectal adenocarcinoma. Whole exome sequencing was performed on tumor and peripheral blood samples, while RNA sequencing (RNAseq) was conducted on tumor samples. We employed three approaches to identify potential targets for personalized and precision therapies. First, using our in-house neoantigen calling pipeline, ARDentify, combined with an AI-based model trained on immunopeptidomics mass spectrometry data (ARDisplay), we identified neoepitopes in the cohort. Second, based on recurrent mutations found in our patient cohort, we selected corresponding cancer cell lines and utilized knock-out gene dependency scores to identify synthetic lethality genes. Third, an AI-based model trained on cancer cell line data was employed to identify cell lines with genomic profiles similar to selected patients. Copy number variants and recurrent single nucleotide variants in these cell lines, along with gene dependency data, were used to find personalized synthetic lethality pairs., Results: We identified approximately 8,700 unique neoepitopes, but none were shared by more than two patients, indicating limited potential for shared neoantigenic targets across our cohort. Additionally, we identified three synthetic lethality pairs: the well-known APC-CTNNB1 and BRAF-DUSP4 pairs, along with the recently described APC-TCF7L2 pair, which could be significant for patients with APC and BRAF variants. Furthermore, by leveraging the identification of similar cancer cell lines, we uncovered a potential gene pair, VPS4A and VPS4B, with therapeutic implications., Conclusion: Our study highlights three distinct approaches for identifying potential therapeutic targets in cancer patients. Each approach yielded valuable insights into our cohort, underscoring the relevance and utility of these methodologies in the development of precision and personalized cancer therapies. Importantly, we developed a novel AI model that aligns tumors with representative cell lines using RNAseq and methylation data. Background: Heart failure (HF) is characterized by significant mortality in both adults and children. Characteristics of pediatric HF are feeding problems, poor weight gain, exercise intolerance, or dyspnea. These changes are often accompanied by endocrine disorders. The main causes of HF are congenital heart defects (CHD), cardiomyopathies, arrhythmias, myocarditis, or heart failure secondary to oncological treatment. Heart transplantation (HTx) is the method of choice for treatment of end-stage HF in pediatric patients., Aims: This article aimed to summarize the single-center experience in heart transplantation in children., Methods: Between 1988 and 2021 in the Silesian Center for Heart Diseases in Zabrze, 122 pediatric cardiac transplantations were performed. In the group of recipients with failing Fontan circulation, HTx was performed in 5 children. The study group was evaluated for the postoperative course: rejection episodes depending on the medical treatment scheme, coinfections, and mortality., Results: One-, 5-, and 10-year survival rates between 1988 and 2001 were 53%, 53%, and 50%, respectively. One-, 5-, and 10-year survival rates between 2002 and 2011 were 97%, 90%, and 87%, respectively; between 2012 and 2021 (1-year of follow-up), the survival rate was 92%. The main cause of mortality both in early and late periods after transplantation was graft failure., Conclusions: Cardiac transplantation in children remains the main method of treatment for endstage heart failure. Our results at both early and long-term posttransplant periods are comparable to those obtained in the most experienced foreign centers.

The aim of this study was to examine the influence of the level (1, 3, and 5 mg I/kg) and source of iodine (KI, Ca(IO 3 ) 2 , and KIO 3 ) on thyroid hormone and immunoglobulin concentrations in the blood serum of laying hens alongside a histological picture of the thyroid. In the first, birds were fed grain-soybean meal mixtures, and in the second, two kinds of diets based on corn-soybean or corn-soybean-rapeseed meal were applied. In the experiments, we determined the levels of the blood serum thyroid hormones fT 3 and fT 4 , as well as the morphological structure of the thyroid gland. In the second experiment, the concentration of immunoglobulins in blood serum was assayed. In both experiments, no influence of iodine source on thyroid hormone concentration was observed. However, increasing the iodine level in the full mixture and adding rapeseed meal in both experiments caused an increase in fT 3 concentration. Increasing I-addition in both experiments led to a decrease in thyroid gland follicle diameter. Rapeseed meal inclusion (at a level of 10%) to the complete hen mixture led to an increase in thyroid gland follicle diameter. Applying KIO 3 as an iodine source in both experiments caused a decrease in the thyroid gland height of follicle epithelial cells. Immunoglobulin concentrations in the serum were not affected by experimental factors. The results suggest that the methodologies of studies on the bioavailability of minerals and the corresponding analytical methods require unification. The lack of such standardization makes it impossible to engage in a satisfactory discussion of the results and exchange experiences.

Arnica montana L. is one of Europe's endemic endangered medicinal plants, with diverse biological activities commonly used in medicine, pharmacy, and cosmetics. Its flower heads are a rich source of raw material, with antibacterial, antifungal, antiseptic, anti-inflammatory, antiradical, antioxidant, and antitumor properties. The objective of the present study was (i) to characterize the chemical composition of flower heads of A. montana plants cultivated under nitrogen fertilization, (ii) to identify the impact of the nitrogen fertilization and extraction method (water, ethanol) on the antioxidant activity of extracts, and (iii) to determine the role of different nitrogen doses applied during plant cultivation and different extraction methods in the anticancer activity of the extracts through analysis of apoptosis and autophagy induction in HT29, HeLa, and SW620 cell lines. The present study shows that nitrogen is a crucial determinant of the chemical composition of arnica flower heads and the antioxidant and anticancer activity of the analyzed extracts. Nitrogen fertilization can modify the composition of pharmacologically active substances (sesquiterpene lactones, flavonoids, essential oil) in Arnicae flos . The content of sesquiterpene lactones, flavonoids, and essential oil increased with the increase in the nitrogen doses to 60 kg N ha -1 by 0.66%, 1.45%, and 0.27%, respectively. A further increase in the nitrogen dose resulted in a decrease in the content of the analyzed secondary metabolites. Varied levels of nitrogen application can be regarded as a relevant way to modify the chemical composition of arnica flower heads and to increase the anticancer activity, which was confirmed by the increase in the level of apoptosis with the increase in fertilization to a level of 60 kg N ha -1 . The fertilization of arnica plants with low doses of nitrogen (30 and 60 kg N ha -1 ) significantly increased the LOX inhibition ability of the ethanol extracts. The present study is the first report on the anticancer activity of A. montana water extracts, with emphasis on the role of water as a solvent. In further studies of factors modifying the quality of Arnicae flos , attention should be paid to the simultaneous use of nitrogen and other microelements to achieve synergistic results and to the possibility of a more frequent use of water as a solvent in studies on the biological activity of A. montana extracts.

Human bodily mechanisms and functions produce low-frequency vibrations. Our ability to perceive these vibrations is limited by our range of hearing. However, in-ear infrasonic hemodynography (IH) can measure low-frequency vibrations (<20 Hz) created by vital organs as an acoustic waveform. This is captured using a technology that can be embedded into wearable devices such as in-ear headphones. IH can acquire sound signals that travel within arteries, fluids, bones, and muscles in proximity to the ear canal, allowing for measurements of an individual's unique audiome. We describe the heart rate and heart rhythm results obtained in time-series analysis of the in-ear IH data taken simultaneously with ECG recordings in two dedicated clinical studies. We demonstrate a high correlation (r = 0.99) between IH and ECG acquired interbeat interval and heart rate measurements and show that IH can continuously monitor physiological changes in heart rate induced by various breathing exercises. We also show that IH can differentiate between atrial fibrillation and sinus rhythm with performance similar to ECG. The results represent a demonstration of IH capabilities to deliver accurate heart rate and heart rhythm measurements comparable to ECG, in a wearable form factor. The development of IH shows promise for monitoring acoustic imprints of the human body that will enable new real-time applications in cardiovascular health that are continuous and noninvasive., (© 2022. The effect of fortification of wheat rolls with 1-5% Greek oregano on the profile of phytochemicals, their changes during processing and simulated digestion (with special emphasis on rosmarinic acid), and in vitro protein and starch digestibility was assessed. Additionally, the main functional properties, e.g. the basic technological quality (baking loss, total volume, specific volume, density, porosity), rosmarinic acid distribution, and consumer acceptance of the fortified products were investigated. Rosmarinic acid content (HPLC analysis), total phenolic content, and in vitro antioxidant activities (ABTS, DPPH, FRAP) were determined. These parameters were analyzed during the preparation, processing, and simulated digestion (at the gastric and intestinal phase) of rolls. Potential bioaccessibility was calculated based on the digestion results. The fortification significantly improved rosmarinic acid content, total phenolic content, and antioxidant activity of the rolls. The potential bioaccessibility of phytochemicals was relatively good, especially at the intestinal phase. Nonetheless, higher doses of the supplement negatively affected the nutrient digestibility and the functional properties of the rolls. Regarding nutraceutical potential, fortified rolls seem to be a good food vehicle for Greek oregano phytochemicals, however, simultaneous optimization of nutritional and functional parameters seems to be challenging due to the multiple and often unpredictable factors affecting the final quality of fortified products.

This study is designed to determine the efficacy of Cerebrolysin treatment as an add-on therapy to mechanical thrombectomy (MT) in reducing global disability in subjects with acute ischemic stroke (AIS). We have planned a single center, prospective, open-label, single-arm study with a 12-month follow-up of 50 patients with moderate to severe AIS, with a small established infarct core and with good collateral circulation who achieve significant reperfusion following MT and who receive additional Cerebrolysin within 8 h of stroke onset compared to 50 historical controls treated with MT alone, matched for age, clinical severity, occlusion location, baseline perfusion lesion volume, onset to reperfusion time, and use of iv thrombolytic therapy. The primary outcome measure will be the overall proportion of subjects receiving Cerebrolysin compared to the control group experiencing a favorable functional outcome (by modified Rankin Scale 0-2) at 90 days, following stroke onset. The secondary objectives are to determine the efficacy of Cerebrolysin as compared to the control group in reducing the risk of symptomatic secondary hemorrhagic transformation, improving neurological outcomes (NIHSS 0-2 at day 7, day 30, and 90), reducing mortality rates (over the 90-day and 12 months study period), and improving: activities of daily living (by Barthel Index), health-related quality of life (EQ-5D-5L) assessed at day 30, 90, and at 12 months. The other measures of efficacy in the Cerebrolysin group will include: assessment of final stroke volume and penumbral salvage (measured by CT/CTP at 30 days) and its change compared to baseline volume, changes over time in language function (by the 15-item Boston Naming Test), hemispatial neglect (by line bisection test), global cognitive function (by The Montreal Cognitive Assessment), and depression (by Hamilton Depression Rating Scale) between day 30 and day 90 assessments). Cystic fibrosis is an autosomal progressive disease affecting the lung, pancreas, and liver. Some patients develop end-stage respiratory and liver failure. For such patients, combined lung-liver transplantation remains the only therapeutic option. Background: Adherence to health behaviors and to immunosuppressant medications are variables among heart transplant recipients with potential life-threatening outcomes. It is understood that patients with psychosocial barriers are at increased risk of poor outcomes due to various factors. Health locus of control includes any activity that can be done to prevent health problems. Health locus of control is defined as individual beliefs based on past experiences in health issues and having internal and or external control. The literature suggests that patients with a strong internal locus of control have a sense of responsibility for their health behaviors., Purpose: To investigate the effect of the health locus of control on the level of self-reported health behaviors among organ transplant recipients., Materials and Methods: A cross-sectional design was used for this study. The study group comprises 222 individuals after heart, kidney, lung, and liver transplantation. The following standardized tools were used: The Multidimensional Health Locus of Control, The Health Behavior Inventory, Morisky Medication-Taking Adherence Scale 4-item. Data were analyzed using IBM SPSS. The level of significance for the statistical test was set at 0.05., Results: Powerful Others scored the highest among the 3 Health Locus of Control scales in heart, kidney, and liver recipients. Participants paid little attention to Healthy eating habits; however, the total score of the Health Behavior Inventory was high (91.68-94.40). There was a difference between the kind of transplant and center for 4 aspects of health behavior. Higher scores of The Multidimensional Health Locus of Control were associated with higher scores of Health Behaviors and level of adherence. Greater Powerful Others and Internal Health Locus of control predict higher health behaviors explaining 42 % of the variance., Conclusions: The intensification of declared health behaviors is high. However, in educating posttransplant patients, attention should be paid to strengthening proper healthy eating habits. Associations between health locus of control dimensions and health behavior have been shown among transplant recipients. Background: the Bonebridge hearing implant is an active transcutaneous bone conduction implant suitable for various types of hearing loss. It was first launched in 2012 as the BCI 601, with a newer internal part (BCI 602) released in 2019. With the new size and shape, the BCI 602 can be used in patients previously excluded due to insufficient anatomical conditions, especially in patients with congenital defects of the outer and middle ear., Objectives: the purpose of this study is to evaluate the objective and subjective benefits of the new Bonebridge BCI 602 in children who have hearing impairment due to conductive or mixed hearing loss. Safety and effectiveness of the device was assessed., Methods: the study group included 22 children aged 8-18 years (mean age 14.7 years) who had either conductive or mixed hearing loss. All patients were implanted unilaterally with the new Bonebridge BCI 602 implant. Pure tone audiometry, speech recognition tests (in quiet and noise), and free-field audiometry were performed before and after implantation. Word recognition scores were evaluated using the Demenko and Pruszewicz Polish Monosyllabic Word Test, and speech reception thresholds in noise were assessed using the Polish Sentence Matrix Test. The subjective assessment of benefits was carried outusing the APHAB (Abbreviated Profile of Hearing Aid Benefit) questionnaire., Results: after implantation of the Bonebridge BCI 602 all patients showed a statistically significant improvement in hearing and speech understanding. The mean word recognition score (WRS) changed from 12.1% before implantation to 87.3% after 6 months. Mean speech reception threshold (SRT) before implantation was +4.79 dB SNR and improved to -1.29 dB SNR after 6 months. All patients showed stable postoperative results. The APHAB questionnaire showed that difficulties in hearing decreased after implantation, with a statistically significant improvement in global score. Pre-operative scores (M = 35.7) were significantly worse than post-operative scores at 6 months (M = 25.7)., Conclusions: the present study confirms that the Bonebridge BCI 602 is an innovative and effective solution, especially for patients with conductive and mixed hearing loss due to anatomical ear defects. The Bonebridge BCI 602 system provides valuable and stable audiological and surgical benefits. Subjective assessment also confirms the effectiveness of the BCI 602. The BCI 602 offers the same amplification as the BCI601, but with a smaller size. The smaller dimensions make it an effective treatment option for a wider group of patients, especially children with congenital defects of the outer and middle ear.

Context: Trigonella foenum-graecum L. (Fabaceae) has many therapeutic properties and anticancer potential., Objective: The cytotoxic activities of standardized extracts and a fraction from fenugreek seeds and their compounds (sapogenins, flavone C-glycosides, alkaloid trigonelline) against human cancer SKOV-3, HeLa and MOLT-4 cells were evaluated., Materials and Methods: Fenugreek seeds were extracted with 70% methanol (A) or water (B). Furthermore, the seeds were purified with petroleum ether and chloroform and next extracted with methanol to obtain fraction (C). The quantitative analysis of saponins and flavonoids in the extracts was done with HPLC methods. The extracts (5-120 µg/mL) and compounds (1-50 µg/mL) were tested on the cells by MTT assay and RTCA system. The effect of a fraction on ROS production, mitochondrial membrane potential and caspase-3/7 activity in HeLa and SKOV-3 cells was also evaluated by flow cytometry., Results: The strongest cytotoxic activity on cancer cells showed the fraction C (IC 50 was 3.91 ± 0.03 for HeLa, 3.97 ± 0.07 for SKOV-3, and 7.75 ± 0.37 for MOLT-4) with the highest content of steroidal saponins (163.18 ± 11.03 μg/mg) and flavone C-glycosides (820.18 ± 0.05 μg/mg). The fraction significantly increased ROS production (up to four times higher than in keratinocytes as control) and caspases activity in the cells. The examined flavonoids did not exhibit the cytotoxic activity in contrast to yamogenin, tigogenin, and diosgenin., Conclusions: The obtained results complement the data on the cytotoxic activity of Foenugraeci Semen and synergistic effect of flavonoids and saponins complex contained in the plant.

BACKGROUND The aim of this study was to assess the effectiveness of bone conduction hearing aids in children under 2 years old who have congenital microtia and atresia. MATERIAL AND METHODS This prospective study involved 42 children under 2 years old with congenital microtia and atresia who were divided into 2 groups: 21 with unilateral defect and 21 with bilateral defect. All children were provided with bone conduction hearing aids on a softband. Air and bone auditory thresholds were assessed by auditory brainstem responses (ABRs). The LittlEARS questionnaire was used to evaluate auditory development at baseline and after 6 months of hearing aids use. Behavioral observation audiometry (BOA) was used to assess auditory thresholds and compare aided and unaided hearing. RESULTS After 6 months of hearing aid use, the total score of the LittlEARS questionnaire in children with unilateral defect was 24±5.60, while children with bilateral defect achieved a result of 26.29±6.17. Hearing thresholds in both groups with bone conduction hearing aids improved significantly and approached the normal level. CONCLUSIONS Our results confirm that bone conduction hearing aids provide an effective method of auditory rehabilitation for children with conductive and mixed hearing loss caused by microtia and atresia. Using bone conduction hearing aids in such children is crucial for proper hearing, speech, and language development.
