2. Response to neoadjuvant chemotherapy in HER2 non-overexpressing breast cancer subtypes

Background Pathologic complete response (pCR) is a surrogate marker of a better relapse-free (RFS) and overall survival (OS), particularly in HER2 overexpressing and triple-negative breast cancer (TNBC) subtypes. Our aim is to assess the impact of HER2 expression level on pCR, in patients with luminal or TNBC treated with neoadjuvant chemotherapy (NAC). Methods We conducted a retrospective analysis in luminal and TN non-metastatic breast cancer patients, addressed to cancer center Georges Francois Leclerc in Dijon for NAC. Hormone receptor and HER2 expression were centrally assessed according to CAP/ASCO criteria. The pathologic response was dichotomized in pCR and non-pCR and evaluated according to Chevalier classification. Results Between 2007 and 2018, 761 patients (pts) were recorded, of which 263 (34.5%) had TNBC and 498 (65.5%) luminal-HER2 negative BC. Among TNBC, 189 pts (70.8%), 54 pts (20.2%) and 24 pts (8.9%) had respectively score 0, 1 and 2, and negative FISH. In luminal tumors these results were respectively 246 (49.3%), 181 (36.3%) and 21 (4.2%). Median age, size of primary tumour and lymph node involvement were similar in both subpopulations. Regarding NAC, 20.9% received only taxanes, 22.1% only anthracyclines and 55.1% both agents. The combination was more often recommended in TNBC 74.2% vs 59.3% (p = 0.03). pCR rate in the primary tumour and lymph nodes was 29% (157 pts) in the whole cohort. This rate varied according to HER2 expression (0, 1+, 2+) from respectively 48.6% (51pts), 36.4% (8pts) and 28.6% (4pts) (p = 0.256) in TNBC and from 8.5% (7pts), 5.3% (5pts) and 4.1% (2pts) (p = 0.532) among luminal tumors. Both OS and RFS were associated with a good Chevalier pathologic response (p Conclusions No correlation between pCR and the level of HER 2 expression was found neither in luminal nor in TNBC patients. In TNBC patients, differences in terms of pCR were noticed between the 3 HER expression groups. The predictive role of HER2 expression should be further explored in early localized TNBC in a larger population. 4. Determining adolescent health information needs: A survey in Geneva, 2022.

- Abstract
Purpose: When adolescents feel that their opinions are valued and respected, they are more likely to seek and use health services. Our aim was to identify what health information adolescents in Geneva, Switzerland, want and how much they are willing to participate in health promotion, linking it to international recommendations., Method: Data were collected from 970 adolescents (aged 12-15 years), between September and November 2022, using a classroom-based survey., Results: Adolescents expressed their preferences: the study showed that the topics of sleep, self-perception, biology, mental health, and physical activity received the highest interest. Girls were more interested than boys in the issues of sleep, mental health, violence, and social life. Adolescents' increasing interest in sexuality issues as they get older is inversely proportional to their interest in other issues such as pandemics, medical research, and the environment, which they are more interested in during early adolescence. Everyone over two felt it was important to promote health in their social environment. And more than a third of teens are open to participating in such activities., Discussion: Adolescents are open to various health subjects, except for pandemics. Age dependent interest highlights the importance of tailoring intervention programs. Adolescents consider health promotion important and are willing to participate, as recommended by the World Health Organization. By involving them as experts in their adolescence, we have taken an essential step. 5. Imbalance of TH17/TREG cells in Tunisian patients with systemic sclerosis.

- Abstract
Fibrosis is a pathological manifestation in which connective tissue replaces normal one. It can affect many tissues from the skin to internal organs such as the lungs. Manifestations of pulmonary involvement can be pulmonary arterial hypertension or pulmonary fibrosis. The latter one is currently the leading cause of death in various autoimmune diseases, including systemic sclerosis. Our study group consists of 50 patients with systemic sclerosis: 24 with limited cutaneous form and 26 with diffuse cutaneous form. This cohort was compared to 50 healthy controls (age and sex matched); our aim is to explore the distribution of TH17 cells (TH17) as well as regulatory T cells (TREG) and study their correlation with the disease's progress. Our results show an increase for IL17A in patients compared to controls and that this increase is correlated with a specific clinical involvement: Pulmonary fibrosis. This correlation suggests a crucial role of IL17A in fibrosis especially in systemic sclerosis. In addition, we have shown that the percentages of TH17 cells are higher in patients; however, the percentages of TREG cells are similar between patients and controls. A study of TREG cell activity showed that TREG lost suppressive activity by inactivating the FOXP3 transcription factor. This proves that despite their presence, TREG does not adequately carry out their regulatory activity. Finally, we analyzed the correlation between TH17/TREG and clinical damage; the results show a positive correlation with pulmonary involvement proving the role of TH17/TREG balance in induced fibrosis in systemic sclerosis. No significative difference was observed, for all the parameters, between the two different forms of the disease. - Abstract
Background: External quality assessments (EQAs) for the molecular detection of human respiratory syncytial virus (RSV) are necessary to ensure the standardisation of reliable results. The Phase II, 2019-2020 World Health Organization (WHO) RSV EQA included 28 laboratories in 26 countries. The EQA panel evaluated performance in the molecular detection and subtyping of RSV-A and RSV-B. This manuscript describes the preparation, distribution, and analysis of the 2019-2020 WHO RSV EQA., Methods: Panel isolates underwent whole genome sequencing and in silico primer matching. The final panel included nine contemporary, one historical virus and two negative controls. The EQA panel was manufactured and distributed by the UK National External Quality Assessment Service (UK NEQAS). National laboratories used WHO reference assays developed by the United States Centers for Disease Control and Prevention, an RSV subtyping assay developed by the Victorian Infectious Diseases Reference Laboratory (Australia), or other in-house or commercial assays already in use at their laboratories., Results: An in silico analysis of isolates showed a good match to assay primer/probes. The panel was distributed to 28 laboratories. Isolates were correctly identified in 98% of samples for detection and 99.6% for subtyping., Conclusions: The WHO RSV EQA 2019-2020 showed that laboratories performed at high standards. Updating the composition of RSV molecular EQAs with contemporary strains to ensure representation of circulating strains, and ensuring primer matching with EQA panel viruses, is advantageous in assessing diagnostic competencies of laboratories. Solieria filiformis has been reported to have molecules with various biological activities. In this study we used environmentally friendly extraction methods, such as enzyme-assisted extraction (EAE), as a first step to obtain bioactive compounds from this species. Five combinations of protease (PRO) and carbohydrase (AMG) were utilized (1:0, 0:1, 2:1, 1:1, 1:2 PRO:AMG) to obtain Water Soluble Enzymatic Hydrolysates (WSEHs). Extraction yields, biochemical and structural characterization, as well as in vitro activity against Herpes simplex virus type 1 (HSV-1) and antioxidant capacities were determined. All PRO:AMG combinations significantly improved yields. EAE yielded heterogeneous extracts rich in iota-carrageenan and phenols, as confirmed by FTIR spectra. The highest antiherpetic activity (EC
50 4.5 ± 0.4 μg mL-1 ) was found in the WSEHs obtained under 2:1 PRO:AMG. At this combination high antioxidant capacity was also obtained for ABTS (2,2'-Azino-Bis-3-ethylbenzoThiazoline-6-Sulfonic acid) radical scavenging activity and Ferric Reducing Antioxidant Power (FRAP). These could probably play a synergistic role associated to the strong antiviral activity obtained. These results suggest that 2:1 PRO:AMG could be effective in promoting the hydrolytic breakdown of high MW polysaccharides, contributing to the improvement of WSEHs bioactivity. Objectives: Studies highlight the beneficial effects of social and leisure activities on the well-being of aging people. Our aims were: to investigate activity profiles among older adults living in their own homes, to highlight potential changes in profiles over a period of five years and to examine effects of differences between these profiles on well-being variables., Methods: Three waves were considered in this longitudinal study, with 550, 410 and 374 people (76, 79, 82 years on average), respectively. We used cluster analysis to obtain activity profiles of social and leisure activities, ANOVAs with age and external variables (satisfaction with life, time occupation satisfaction, self-esteem and depression) to explore differences between cluster groups, and chi-square tests of association to explore whether the same individual belonged to the same activity profile over time., Results: The number of clusters decreased strongly from the first wave to the other two waves (n = 7 to n = 2 and 3). Two main types of activity differentiated the profiles: social activities (voluntary work and associations) and recreational activities outside the home/at home. The activities showed no systematic or strong effects on external variables, nevertheless for the oldest subjects when comparing the less active to the more active (recreational profile and social profile) the latter expressed a greater satisfaction of time occupation and self-esteem, and lower scores of depressive symptoms., Conclusions: Such activities could be an indicator of a certain independence (physical and mental availability) which plays a strong role in the well-being of older adults.

- Abstract
The development of metrology of iron-55 in low-level radioactivity in environmental solid matrices was realised for conducting radioecological studies. A protocol was developed based on the adaptation of existing methods for the purification of iron-55 with selective chromatographic resin, which was then measured with liquid scintillation. The loss attached treatment chemical steps were quantified with elemental iron by Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES). The tests were used to define the iron retention capacity of selective chromatographic resin, a key element in chemical treatment, and test sample size needed to reach the detection limit of 30 Bq kg(-1) dry. The solid samples were analysed with the developed protocol. The activities obtained from iron-55 were below the detection limit of 30 Bq kg(-1) dry., (Copyright © 2014 Elsevier Ltd. Bemisia tabaci-transmitted geminiviruses are one of the major threats on cassava and vegetable crops in Africa. However, to date, few studies are available on the diversity of B. tabaci and their associated endosymbionts in Africa. More than 28 species have been described in the complex of B. tabaci cryptic species; among them, 2 are invasive pests worldwide: MED and MEAM1. In order to assess the species diversity of B. tabaci in vegetable crops in Senegal, several samplings in different localities, hosts and seasons were collected and analyzed with nuclear (microsatellite) and mitochondrial (COI) markers. The bacterial endosymbiont community was also studied for each sample. Two species were detected: MED Q1 and MEAM1 B. Patterns of MED Q1 (dominance on most of the samples and sites, highest nuclear and mitochondrial diversity and broader secondary endosymbiont community: Hamiltonella, Cardinium, Wolbachia and Rickettsia), point toward a predominant resident begomovirus vector group for MED Q1 on market gardening crops. Furthermore, the lower prevalence of the second species MEAM1 B, its lower nuclear and mitochondrial diversity and a narrower secondary endosymbiont community (Hamiltonella/Rickettsia), indicate that this genetic group is exotic and results from a recent invasion in this area., (© 2014 Institute of Zoology, Chinese Academy of Sciences.)

- Abstract
In order to assess the performance of Bayesian individualization of busulfan (BU) dosage regimens, veno-occlusive disease (VOD) rate was monitored for paediatric patients undergoing allogeneic bone marrow transplantation (BMT). Consecutive patients undergoing allogeneic BMT with BU as conditioning regimen during 5-years period (January 2000-February 2006) were reviewed. VOD was a major outcome variable. Preconditioning risk of VOD was estimated for each patient using a scoring system that included type of transplant, recipient CMV-positive status and total parenteral nutrition (TPN) provided pretransplantation. A risk-adjusted cumulative sum method was used to compare observed versus predicted outcome by assigning a risk score, based on log-likelihood ratios, to each patient. These cumulative scores were sequentially plotted with preset control limits for 'signalling' where results were substantially different than expected (doubling or halving of odds ratio). Sixty-six children received BMT after oral busulfan-based conditioning regimen with median age 3.9 years, 63.6% of male. Median preconditioning risk of VOD was 0.34 ranging from 0.23 to 0.84. Observed VOD rate was 16.7% (n = 11) which was 60.7% (17) fewer than the expected number estimated by the risk score. The resulting risk-adjusted score for each patient was plotted sequentially. This plot adopted early a negative slope, crossing the lower control limit twice, after 27 and 66 patients, indicating improved results compared to those expected. Bayesian individualization of oral busulfan dosage regimens is useful to reduce the VOD rate in children undergoing allogeneic BMT., (Copyright 2007 John Wiley & Sons, Ltd.)
