Spätestens seit der erfolgreichen und gutbesuchten Ausstellungsreihe „Körperwelten“ von Gunther von Hagens, in der plastinierte menschliche Ganzkörpermodelle ausgestellt wurden, zeigte sich, welche Faszination und Anziehungskraft die Anatomie auf die Öffentlichkeit ausübt. Studierende der Veterinärmedizin lernen in den ersten zwei Jahren ihres Studiums den anatomischen Aufbau des Körpers von verschiedenen Tierarten. Das Erlernen der Anatomie ist nicht einfach. Es erfordert die Identifizierung, das Verständnis und das Auffinden anatomischer Strukturen im dreidimensionalen Raum. Das Curriculum beinhaltet daher unter anderem praktische Sektionsübungen an Tierkadavern. Dabei haben viele Studierende Schwierigkeiten, die in den Lehrbüchern in zweidimensionalen Abbildungen dargestellten Strukturen auf den dreidimensionalen Tierkörper zu übertragen. Im Rahmen eines Forschungsprojekts untersucht ein Team am Institut für Veterinär-Anatomie der Freien Universität Berlin, ob annotierte dreidimensionale Scans anatomischer Präparate eine sinnvolle Alternative zu zweidimensionalen Abbildungen darstellen. Zusätzlich sollen mittels 3D-Druck erstellte Modelle anatomischer Präparate als zeit- und raumunabhängige Lernressource untersucht werden. Die Methode der 3D-Digitalisierung und -Modellierung erschließt einen weiteren Mehrwert. Damit können die Präparate von Gurlt und Ziegler, zwei historische Sammlungen embryologischer Modelle aus dem 19. und 20. Jahrhundert, welche im Institut für Veterinär-Anatomie ausgestellt sind, digitalisiert werden. Aufgrund ihres beträchtlichen historischen Wertes und ihrer Fragilität waren sie bisher nur in Schaukästen ausgestellt, wurden aber nicht mehr aktiv in der Lehre in den Händen von Studierenden eingesetzt. In diesem Beitrag wird dargelegt, wie man diese unter Verwendung der erwähnten Techniken in die anatomische und embryologische Lehre integriert und auch mittels einer eigens erstellten Online-Plattform einer breiteren Öffentlichkeit zugänglich macht.

Der digitale Wandel hat wissenschaftlichen Sammlungen eine Fülle technischer Möglichkeiten an die Hand gegeben, Objekte zu digitalisieren, zu dokumentieren und auf vielfältige Weise nutzbar zu machen. Die sechste Auflage des „Jungen Forums für Sammlungs- und Objektforschung“ widmet sich daher den digitalen Methoden und Werkzeugen, die im Kontext einer forschenden oder auch lehrenden Befassung mit den Sammlungsdingen und ihren digitalen Abbildern zum Einsatz kommen. Dieser Band vereint elf Beiträge von Nachwuchswissenschaftler:innen aus den Bereichen Archäologie, Geschichtsdidaktik, Digital Humanities, Kunstgeschichte, Kunstpädagogik, Kunsttechnologie und Restaurierungswissenschaft, Psychologie sowie der Human- und Veterinärmedizin, die sich mit digitalen Dingwelten auseinandersetzen, indem sie jeweils unterschiedliche Fragestellungen und Methoden heranziehen. Die Reflexion über forschungsleitende methodische Aspekte objektbezogener Forschung bildet einen gemeinsamen Bezugspunkt der Beiträge.

Background Various studies from different countries indicated that veterinarians have a significantly increased risk of depression, suicidal ideation and of death by suicide. For German veterinarians a recent study has demonstrated a three times higher rate for depression, two times higher rate for suicidal ideation and a five times higher suicide risk compared to the German general population. For veterinary students, recent studies in the US and UK indicated higher mental distress. To date, empirical studies on depression, suicidal ideation and suicide risk among veterinary students in Germany were lacking so far. This study investigates depression, suicidal ideation and suicide risk of veterinary students in Germany. Methods 913 German veterinary students (14.3% response rate, 90.7% women, mean age 23.6 years) between 18 and 46 years were included and compared with representative German general population samples from 2007 (N = 1097, 55.4% women, mean age 33.9) and 2015 (N = 1033, 56.1% women, mean age 32.8) of the same age range using the depression module of the Patient Health Questionnaire (PHQ-9) and Suicide Behaviors questionnaire-Revised (SBQ-R). The general population samples were collected with the assistance of a demographic consulting company. Results The prevalence of depression among German veterinary students was 45.9% (compared to 3.2% in the general population), suicidal ideation was 19.9% (compared to 4.5% in the general population) and suicide risk was 24.0% (compared to 6.6% in the general population). Conclusion In this study, German veterinary students have a 22.1 times higher risk to be screened positive for depression, a 4 times higher risk for reporting current suicidal ideation and they are 4.2 times more likely to have an increased suicide risk compared with the general population in Germany of the same age range.

Introduction In vitro assays of angiogenesis face immense problems considering their reproducibility based on the inhomogeneous characters of endothelial cells (ECs). It is necessary to detect influencing factors, which affect the angiogenic potency of ECs. Objective This study aimed to analyse expression profiles of vimentin (VIM), triosephosphate isomerase (TPI) and adenosylmethionine synthetase isoform type–2 (MAT2A) during the whole angiogenic cascade in vitro. Furthermore, the impact of knocking down vimentin (VIM) on angiogenesis in vitro was evaluated, while monitoring TPI and MAT2A expression. Methods A long–term cultivation and angiogenic stimulation of human dermal microvascular ECs was performed. Cells were characterized via VEGFR–1 and VEGFR–2 expression and a shRNA–mediated knockdown of VIM was performed. The process of angiogenesis in vitro was quantified via morphological staging and mRNA–and protein–levels of all proteins were analysed. Results While native cells ran through the angiogenic cascade chronologically, knockdown cells only entered beginning stages of angiogenesis and died eventually. Cell cultures showing a higher VEGFR–1 expression survived exclusively and displayed an upregulation of MAT2A and TPI expression. Native cells highly expressed VIM in early stages, MAT2A mainly in the beginning and TPI during the course of angiogenesis in vitro.

The extracellular matrix (ECM) of hyaline cartilage is perfectly suited to transmit articular pressure load to the subchondral bone. Pressure is transferred by a high amount of aggrecan-based proteoglycans and collagen type II fibres in particular. After any injury, the hyaline cartilage is replaced by fibrocartilage, which is low in proteoglycans and contains collagen type I predominantly. Until now, long-term results of therapeutic procedures including cell-based therapies like autologous chondrocyte transplantation (ACT) lead to a replacement tissue meeting the composition of fibrocartilage. Therefore, it is of particular interest to discover how and to what extent isolation and in vitro cultivation of chondrocytes affect the cells and their expression of ECM components. Hyaline cartilage-derived chondrocytes were cultivated in vitro and observed microscopically over a time period of 35 days.

Literaturverzeichnis 187, Angiogenese, die Sprossung von Kapillaren aus bereits vorhandenen Blutgefäßen, ist eine Voraussetzung für Wachstum und Differenzierung von Organen und Geweben. Gleichzeitig ist Angiogenese an einer Vielzahl pathologischer Prozesse, wie beispielsweise am Wachstum und der Metastasierung von Tumoren, beteiligt. Ziel der vorliegenden Arbeit war es, eine Methode zur Quantifizierung der Angiogenese und Antiangiogenese in vitro zu etablieren, die als Ersatz- und Ergänzungsmethode zum Tierversuch alle Phasen der angiogenen Kaskade umfasst. Des weiteren sollte diese routinemäßig von verschiedenen Untersuchern und mit einem vertretbaren Zeit- und Kostenaufwand durchgeführt werden können. Hierfür wurden Endothelzellen aus dem bovinen Corpus luteum in vitro mit einem Selektivmedium zur Angiogenese stimuliert und phasenkontrastmikroskopisch untersucht. Dabei zeigten die Endothelzellen im Ablauf der Angiogenese in vitro charakteristische Veränderungen des Zellbilds in Form der Aussprossung, linearen Aneinanderreihung, Netzwerkbildung und schließlich der dreidimensionalen Organisation zu kapillarähnlichen Strukturen. Diese Phasen der Angiogenese in vitro wurden anschließend anhand des phasenkontrastmikroskopischen Zellbilds in 8 definierte Stadien eingeteilt. Der Zeitbedarf dieser Stadien wurde in Tagen gemessen und zur benötigten Gesamtzeit ins Verhältnis gesetzt. Dadurch war es im Gegensatz zu den bislang aus der Literatur bekannten in vivo- und in vitro-Modellen erstmals möglich, den zeitlichen Ablauf der Angiogenese und Antiangiogenese zu quantifizieren. Zum Nachweis der Reproduzierbarkeit der Methode zur Quantifizierung der Angiogenese wurde sowohl die Einschätzbarkeit der definierten Stadien durch unterschiedliche Personen als auch die Homogenität des Ablaufs der Angiogenese in verschiedenen Kulturschalen geprüft. Dabei zeigte sich, dass die Einschätzung der Stadien der Angiogenese in vitro durch zwei verschiedene Untersucher nur geringgradigen Abweichungen unterlag und somit durch einen einzigen Untersucher durchgeführt werden kann. Die statistische Bewertung des Ablaufs der Angiogenese in verschiedenen Kulturschalen, erfasst durch die definierten Stadien, führte zu dem Ergebnis, dass eine reproduzierbare Quantifizierung der Angiogenese in diesem in vitro- Modell mit einem relativ kleinen Stichprobenumfang erfolgen kann. Daher ist die Quantifizierung der Angiogenese und Antiangiogenese in diesem Modell nicht nur mit einem vertretbaren Zeitaufwand, sondern aufgrund der mittels der Standardausrüstung eines Zellkulturlabors durchführbaren Untersuchungen auch mit einem relativ geringen Kostenaufwand verbunden. Im Rahmen dieser Arbeit erfolgte ein umfangreicher Vergleich der bekannten in vivo-, ex vivo- und in vitro-Modelle, die zur Identifizierung und Untersuchung pro- und antiangiogener Faktoren entwickelt wurden. Die für den Einsatz eines in vitro- Modells der Angiogenese als Ersatz- und Ergänzungsmethode zum Tierversuch erforderliche Vergleichbarkeit mit der Angiogenese in vivo wurde anhand der Untersuchung verschiedener Charakteristika der Endothelzellen überprüft. Dabei zeigte die Morphometrie der sich in vitro gebildeten kapillarähnlichen Strukturen, dass, wie bei der Angiogenese in vivo, auch der Ablauf der Angiogenese in vitro mit der Zunahme der Länge, der von diesen Strukturen eingenommenen Fläche und der Anzahl ihrer Verzweigungspunkte einherging. Ebenso wiesen die ultrastrukturellen Veränderungen der Endothelzellen im Ablauf der Angiogenese in vitro Ähnlichkeiten zur Angiogenese in vivo auf. Diese beliefen sich unter anderem auf Änderungen der Anzahl verschiedener Zellorganellen, wie beispielsweise der Mitochondrien und des endoplasmatischen Retikulums. Vor allem aber war ein für die Differenzierung der Endothelzellen essentielles Merkmal, ihre Polarisierung in eine luminale und eine abluminale Seite des neugebildeten Gefäßes, zu erkennen. Allerdings deutete die Präsenz extrazellulärer Matrix im Inneren der kapillarähnlichen Strukturen auf eine konvertierte Polarität der Endothelzellen hin. Durch den immunzytochemischen Nachweis von Kollagen Typ IV, einem Bestandteil der Basalmembran, wurde zusätzlich dokumentiert, dass es sich bei der von den Endothelzellen sezernierten extrazellulären Matrix um eine der Basalmembran analoge Struktur handelte. Das Auftreten apoptotischer Endothelzellen, welches im Ablauf der Angiogenese in vivo ein für die Reifung und Lumenbildung der neuen Kapillare erforderliches Phänomen darstellt, wurde auch bei den in vitro kultivierten Endothelzellen beobachtet. Dabei unterlagen gemäß der "umgekehrten" Polarität vor allem die Endothelzellen, die den kapillarähnlichen Strukturen von außen auflagen und dadurch keinen Kontakt zur extrazellulären Matrix aufwiesen, dem programmierten Zelltod. Im Rahmen dieser Arbeit wurde auch die proangiogene Wirkung von VEGF und FGF-2 mit der etablierten Methode bestätigt und quantifiziert. Ein besonderer Befund war dabei, dass VEGF in vitro zur Bildung von Endothelzellsphäroiden führte, welche der Entstehung großlumiger Gefäße in vivo entsprechen. Auch wurde die in vivo beobachtete Stimulation der Chemokinese durch FGF-2 bzw. der Chemotaxis durch VEGF bestätigt. Die Untersuchung und Quantifizierung der Wirkung der Angiogenese-Inhibitoren Angiostatin und Suramin zeigte, dass sich die Antiangiogenese durch Angiostatin in einer "Umkehr" der Angiogenese ausdrückt. Suramin hingegen führte anfangs zu einem beschleunigten Ablauf der Angiogenese. Die anhaltende Inkubation resultierte jedoch letztlich in der Auflösung der endothelialen Strukturen und somit in der antiangiogenen Wirkung des Suramin. Das vorliegende Modell schafft eine Möglichkeit zur praktikablen Quantifizierung der Angiogenese bzw. Antiangiogenese in vitro. Es kann sowohl zur effektiven Beurteilung und Quantifizierung der Wirkung potentieller angiogener und antiangiogener Substanzen, wie auch zur Erforschung dabei ablaufender zellulärer und molekularer Regulations-mechanismen angewandt werden und bietet damit die Chance, Tierversuche in Zukunft zu ergänzen und zu ersetzen., Angiogenesis is defined as sprouting of new capillaries from pre-existing ones. Angiogenesis is a pre-requisite for growth and differentiation of organs and tissues and is involved in many pathological processes, for example growth and metastasis of tumours. Up to now numerous in vivo- and in vitro-models of angiogenesis have been developed in order to identify and analyse pro- and antiangiogenic factors. In these models, the effects of the substances tested were quantified in a few phases of angiogenesis only. The aim of the present study was to establish a method to quantify angiogenesis and antiangiogenesis in vitro in order achieve a replacement and complementary method comprising all stages of the angiogenic cascade. Furthermore, routinely accomplishment of quantitation should be possible for different investigators with a maintainable effort of time and costs. Endothelial cells derived from the bovine corpus luteum have been incubated in a selective medium, and the resulting angiogenic reaction of these cells was examined by phase contrast microscopy. In the course of in vitro angiogenesis, endothelial cells showed characteristic changes in their cellular morphology, i.e. sprouting, linear side by side arrangement and three-dimensional organisation in capillary-like structures. Subsequently, these phases of angiogenesis in vitro have been divided into eight strictly defined stages. Thus, in contrast to previously described in vivo and in vitro models, quantitation of the temporal course of angiogenesis and antiangiogenesis was established. The reproducibility of quantitation of angiogenesis was verified by examination of the defined stages by different persons and investigation of homogeneity of the course of angiogenesis in different culture dishes. Assessment of the stages of angiogenesis by two different investigators showed only minimal variances, and can therefore be established by a single investigator. The statistical evaluation of the course of angiogenesis in different culture dishes assessed by the defined stages of angiogenesis showed that a small sample size only is needed for quantitation of angiogenesis in this model. Thus, quantitation of angiogenesis and antiangiogenesis is not only time-efficient, but the method may also be performed with standard equipment of most cell culture laboratories and is thus additionally cost-efficient. Within the scope of this study, as previously described in vivo, ex vivo and in vitro-models employed for identification and testing of pro- and anti-angiogenic factors were compared in extension. In case an in vitro-model should be employed to reduce or replace animal testing, comparability to angiogenesis in vivo is essential and was established by examination of several characteristics of the cultured endothelial. The morphometry of capillary-like structures in vitro showed an increase of the area occupied, the length and the number of branching points in the course of angiogenesis. The ultrastructural changes of the endothelial cells in the course of angiogenesis in vitro showed analogies to angiogenesis in vivo, for example, a change in the number of cell organelles like mitochondria and endoplasmic reticulum occurred. Above all, an essential attribute of differentiated endothelial cells, their polarisation in the context of angiogenesis, was observable. However, the presence of extracellular matrix inside the capillary-like structures suggested a converted polarity of endothelial cells. The immunocytochemical investigation of collagen type IV demonstrated that endothelial cells secreted a characteristic component of the basal membrane. The appearance of apoptotic endothelial cells, a sign of capillary maturation in vivo, was also observed in cultured endothelial cells. According to the converted polarity of the endothelial cells, apoptosis was detected in cells at the exterior side of capillary-like structures, where cells lost contact to the extracellular matrix. In this study, proangiogenic effects of "Vascular Endothelial Growth Factor" (VEGF) and "Fibroblast Growth Factor-2" (FGF-2) were established and quantified by the newly developed method. A particularly noteworthy result was the observation that VEGF induces formation of endothelial spheroids in vitro, equivalent to large-calibre vessels in vivo. Additionally, the reports from in vivo studies of stimulation of chemokinesis induced by FGF-2, and chemotaxis induced by VEGF, respectively, were verified. Examination and quantitation of the angiogenesis-inhibitors Angiostatin and Suramin detected that Angiostatin- induced antiangiogenesis resulted in an 'inverse' angiogenesis. Suramin, on the other hand, initially resulted in increased angiogenesis. Long-term incubation ultimately resulted in disintegration of endothelial structures and thus established the antiangiogenic properties of Suramin. In conclusion, the present model allows a viable quantitation of angiogenesis and antiangiogenesis in vitro. It can be employed either in trial studies of potential angiogenic and antiangiogenic substances, respectively, or in the investigation of their essential cellular and molecular mechanisms and may thus provide an efficient method to reduce, refine and replace animal testing.

Angiogenese ist an physiologischen Vorgängen wie der Embryogenese und der Wundheilung, aber auch bei pathologischen Abläufen wie bei Neoplasien und der Makula Degeneration beteiligt. Im Bereich des Tissue Engineering ist sie ebenfalls unersetzlich und ausschlaggebend für den Erfolg einer Gewebetransplantation. In dieser Studie wurde an 40 männliche Charles Lewis-Ratten das arteriovenöse (AV-) Loop-Modell angewandt, um spontane Angiogenese unter Einfluss wachstumshemmender Faktoren in vivo zu untersuchen. Der AV-Loop wurde in einer mit Fibrin gefüllten Teflonkammer gebettet. Für die statistische Auswertung wurde der Student t-Test mit ungepaarten Stichproben angewandt und das Signifkanzniveau betrug α=0,05. Multiple Testungen wurden nach der Bonferroni-Holm-Methode angepasst. In der Anfangsphase der Studie wurde der zeitliche Verlauf der AV-Loop assoziierten Angiogenese an 16 Tieren untersucht. Es wurde ein signifikanter Anstieg der Gefäßfläche über einen Zeitraum von 5 (n=2), 10 (n=3) und 15 Tagen (n=3) beobachtet und ebenfalls eine signifikant höhere Gefäßanzahl an Tag 15 im Vergleich zu Tag 5. Acht Tiere konnten nicht in die Studie miteingeschlossen werden infolge von Thrombosierungen des Loops. Diese erwartete Verlustrate trat aufgrund Lernkurve dieses komplexen mikrochirurgischen Modells, insbesondere zu Beginn des Projektes, auf. In der zweiten Phase der Studie wurde die Neoangiogenese auf drei unterschiedliche Verfahren gehemmt. Die Implantationszeit betrug bei allen Gruppen 15 Tage. In der ersten Gruppe (n = 6) wurde ein Inhibitor des Wachstumfaktors VEGF (vascular endothelial growth factor) intravenös appliziert, nämlich der monoklonale Antikörper Bevacizumab. Hier konnte ein signifikanter Unterschied zur Kontrollgruppe (n = 6) bei der Gefäßfläche (94 432 ± 17 903 μm2 gegenüber 268 682 ± 63 575 μm2) und ebenfalls bei der Gefäßdichte (18 ± 5 Gefäße pro mm2 versus 40 ± 9 Gefäße pro mm2 in der Kontrollgruppe) gemessen werden. Dieser Befund ließ darauf schließen, dass die Neovaskularisation durch VEGF vermittelt wurde. Die direkte Bestrahlung von 2 Gy auf den venösen Graft in der zweiten Versuchsgruppe (n = 7) löste eine signifikante Verringerung von Gefäßanzahl (311 ± 73), -fläche (43 137 ± 10 225 μm2) und –dichte (15 ± 7 Gefäße pro mm2) im Vergleich zur Kontrollgruppe (776 ± 123, 268 682 ± 63 575 μm2 und 40 ± 9 Gefäße pro mm2) aus. Dieses Verfahren hatte somit starken Einfluss auf den Reifeprozess der Neoangiogenese. Bei der Kombinationsgruppe (Bevacizumab und Bestrahlung, n = 5) konnte nur bei der Gefäßfläche ein signifikant geringerer Unterschied in der Angiogenese erhoben werden. Dies ließ vermuten, dass das hier zu findende physiologische und somit geordnete Gefäßwachstum nicht auf diese hemmende Methode anspricht, wie es bei chaotischen Tumorgefäßsystemen der Fall ist und der vermutete Synergismus ausbleibt. Zusätzlich wurde im Zuge dieser Studie ein standardisiertes Auswertungsprogramm etabliert. Dabei handelt es sich um ein selbstentwickeltes Computerprogramm, das nicht nur die hier gesammelten aber auch 2-D-Aufnahmen anderer Angiogenese-Modelle benutzerunabhängig und standardisiert evaluieren kann. Zusammenfassend kann gesagt werden, dass das AV-Loop-Modell sich ausgezeichnet für die Untersuchung der Angiogenese im gesunden Gewebe eignet. It is involved in events of embryogenesis and in wound healing as well as in neoplastic growth and macula degeneration. In the field of Tissue Engineering neovascularisation plays an irreplaceable role and determines the result of the transplantation. Here, an arterio-venous loop (AV-loop) model embedded in fibrin- filled teflon chambers in 40 Charles Lewis rats was applied to conduct in vivo investigations of the physiological processes of vessel growth in healthy tissue and to understand neovascularisation under the impact of anti-angiogenic factors such as monoclonal anti-bodies and ionizing radiation (IR). For statistical analysis the unpaired t-test was applied with a significance level of α = 0,05. Multiple testing was adapted according to the Bonferroni-Holm method. At the beginning of the study the AV-loop induced angiogenesis was examined on 16 animals and consecutively characterized. A significant increase in vessel area was observed over a time frame of 5 (n=2), 10 (n=3) and 15 days (n=3). Additionally the vessel count has increased significantly at day 15 in comparison to day 5. Eight of the animals had to be excluded due to thrombosis of the loop, which was expected due to the complex microsurgical model, especially at the onset of the project. In the second phase of the study three different anti-angiogenic procedures were investigated. Time of implantation was 15 days. In group one (n = 6) the monoclonal antibody of VEGF (vascular endothelial growth factor) named Bevacizumab was applied intravenously. As a result a significant lower vessel area (94 432 ± 17 903 μm2) and density (18 ± 5 vessels per mm2) could be measured in comparison to the control group (n = 6; 268 682 ± 63 575 μm2 respectively 40 ± 9 vessels per mm2). We concluded from these results that angiogenesis was mediated by VEGF. In comparison to the controlgroup direct IR of 2 Gy led in group two (n = 7) to a significant decrease in vessel number (311 ± 73 versus 776 ± 123), area (43137±10225μm2 versus 268682±63575μm2) and density (15±7 versus 40±9 vessels per mm2). Therefore this procedure has an obvious impact on the vessel maturation. In the combined group (n = 5) of both anti-angiogenic procedures (anti- VEGF and IR) a significant decrease was only evident in the vessel area. We assumed that this physiological and accordingly organized angiogenesis does not respond to the applied inhibiting methods, as it is observed in tumor vessel growth. Additionally, an evaluation program was established with the goal of designing a user-independent and standardized computer program to measure 2-D-images of both this and other angiogenesis models. In summary the AV-loop presents a proficient model to investigate angiogenesis in healthy tissue. Being a small coverage of the joint surface, it shows a composition designed to match this function specifically. A high amount of proteoglycans and its associated water determines the elastic formability of the hyaline cartilage which allows the absorbance of pressure and shear forces. These proteoglycans, mainly based on aggrecan as core-protein, are embedded into a meshwork of collagen fibres, primarily formed of collagen type II. This composition is not to be understood as a static construct; moreover it is exposed to biophysical forces that permanently require a dynamic adaptation. This adaptation of the extracellular matrix formed by proteoglycans and collagen type II is organised by a small number of embedded chondrocytes, the cells of the hyaline cartilage. As chondrocytes are post-mitotic cells and due to the lack of vascularisation within hyaline cartilage, there is hardly any chance for regeneration of defects in order to maintain the integrity of the tissue. The resulting replacement is formed as fibrocartilage, which has not the capability to withstand the biodynamical forces within the joint. As these defects in hyaline cartilage represent a gross of the diseases of the musculoskeletal system, there is a high medical interest in the development of innovative cell-based therapies, as autologous chondrocyte transplantation (ACT) is one. With this type of therapy in vitro cultivated chondrocytes are seeded into a cartilage defect with the aim of producing hyaline cartilage. Aims of the study In the last decades the need for a detailed understanding of the biodynamics of cartilage became obvious for further development of therapies. The aim of this study was therefore to establish a cell culture system to provide an insight into the biodynamics of chondrocytes. Aside from the examination of the differentiation of in vitro cultivated chondrocytes and their synthesis of extracellular matrix as a function of the cultivation time, another aim of this study was to determine whether the application of hydrostatic pressure might have beneficial influence on the expression of extracellular matrix components by chondrocytes in vitro, in accordance with the hyaline cartilage. Material and methods Human articular chondrocytes were cultivated in vitro without the application of hydrostatic pressure in the first place. The cells were observed phase contrast microscopically and the distribution of collagen type I and II was detected immuncytochemically. In further experiments optical confluent chondrocytes were transferred to a bioreactor system applying a hydrostatic pressure of 5 or 10 bar with variable time periods of the pressure applied. Subsequently, the expression of collagen type I, collagen type II and aggrecan was investigated and quantified using qPCR and Western Blot. Chondrocytes cultivated exclusively without the application of hydrostatic pressure served as controls. In this pilot-study the samples were analysed using arithmetic mean and standard deviation to evaluate the power statistically. In addition, similar test conditions with marginal differences were pooled and the necessary sample size to meet a power of 80 % with an alpha error of 0.05 was calculated using the maximum potential standard deviation. In cases where this statistic power was obtained, an analysis of significance (\"One Way Analysis of Variance”) was carried out meeting a significance level under 0.05. Results During the cultivation of chondrocytes in vitro without hydrostatic pressure the length of the cultivation time did neither show an effect on the phase contrast microscopical morphology nor on the immuncytochemically detected distribution of collagen typ I and II. The application of increased hydrostatic pressure for 24 hours results in a 0.2-0.8-fold decrease of the expression of collagen type I and II and a 1.7-2.2-fold increase of aggrecan expression compared to the unloaded controls. This effect was more distinct with 5 bar but was accompanied by instabilities in the cell culture. This is why further investigations concentrated on the use of 10 bar pressure with subsequently shortened time period of the applied pressure. With short times of loading (1.5 and 3 hours) a pressure load of 10 bar led to a 0.8 fold decrease of the expression of collagen type I and II and showed a 1.6-2.4 fold increase of aggrecan expression. These qPCR results were supported by the protein expression of collagen type I, II and aggrecan detected in Western Blot. Conclusions A cell culture system was established to examine the effect of hydrostatic pressure on the expression of chondrocytes on the one hand, which can further be modified for the assembly of cell transplants on the other hand. Subsequently the results of this study led to a definition of cell culture conditions, stimulating the extracellular matrix production of chondrocytes towards the composition of hyaline cartilage. This was the case using a seeding density of 104 cells/cm2 and a pre-cultivation time of 6 days of normal pressure, followed by the application of 10 bar hydrostatic pressure for 1.5-3 h. With the help of this pilot-study a cell culture system was established to gain more information on biodynamics of hyaline cartilage. Moreover it is possible that this information will provide a basis for further development of cell based therapies of cartilage defects, such as ACT. Als schmaler Überzug der Gelenkflächen ist er in seinem Aufbau an diese Funktion spezifisch angepasst. Dabei bedingt der hohe Gehalt an Proteoglykanen und das an diese assoziierte Wasser die elastische Verformbarkeit des hyalinen Knorpels, die es ermöglicht, Druck- und Scherkräfte abzufedern. Die Proteoglykane, die hauptsächlich auf Aggrekan als Kernprotein basieren, sind in ein Maschenwerk kollagener Fasern eingelagert, welches im Wesentlichen durch Kollagen Typ II gebildet wird. Diese Zusammensetzung darf nicht als statisches Konstrukt verstanden werden. Vielmehr ist der hyaline Knorpel in vivo verschiedenen biophysikalischen Einflüssen ausgesetzt, die eine dynamische Anpassung erfordern. Solche Anpassungsvorgänge in Form einer Änderung der Zusammensetzung der aus kollagenen Fasern und Proteoglykanen bestehenden extrazellulären Matrix werden durch die wenigen eingelagerten Chondrozyten, die Zellen des hyalinen Knorpels, organisiert. Da die reifen Chondrozyten jedoch keine Zellteilungen aufweisen und dem hyalinen Knorpel eine Vaskularisierung fehlt, ist eine Defektregeneration kaum möglich, sodass eine Wiederherstellung der Integrität des Gewebes unterbleibt und stattdessen ein Ersatzknorpel, der Faserknorpel, gebildet wird, welcher den einwirkenden biodynamischen Belastungen jedoch nicht standhalten kann. Da die Defekte des hyalinen Knorpels einen Großteil der Erkrankungen des Bewegungsapparats darstellen und zudem die Arthrose als Langzeitfolge nach sich ziehen, besteht ein hohes medizinisches Interesse an der Entwicklung zellbasierter Therapieansätze, wie der Autologen Chondrozytentransplantation (ACT). Hierbei werden - bislang mit unterschiedlichen Erfolgen – in vitro kultivierte Chondrozyten mit dem Ziel, neuen hyalinen Knorpel zu bilden, in einen Knorpeldefekt eingebracht. Ziele der Untersuchungen In den letzten Jahrzehnten zeigte sich, dass die Entwicklung von Therapieansätzen zur Behandlung von Knorpeldefekten ein detaillierteres Verständnis des Knorpelgewebes und speziell dessen Biodynamik erfordert. Ziel dieser Arbeit war es daher, im Rahmen einer Pilotstudie, ein In-vitro-System zu etablieren, welches die Untersuchung der Biodynamik der Chondrozyten ermöglicht. Neben der Untersuchung der Morphologie der Chondrozyten und der durch sie synthetisierten extrazellulären Matrix in Abhängigkeit von der Kultivierungszeit der Zellen, wurde die Fragestellung bearbeitet, ob durch die Wirkung eines hydrostatischen Drucks günstige Effekte in Hinblick auf die Expression einer extrazellulären Matrix, wie sie im hyalinen Knorpel vorliegt, erzielt werden kann. Materialien und Methoden Eine Primärkultur humaner artikulärer Chondrozyten wurde zunächst unter Standardzellkulturbedingungen und atmosphärischem Druck kultiviert. Die Zellen wurden phasenkontrastmikroskopisch und hinsichtlich der Verteilung von Kollagen Typ I und II immunzytochemisch untersucht. In den weiteren Versuchen wurden optisch konfluente Chondrozyten in einen Bioreaktor überführt und weiter unter einem hydrostatischen Druck von 5 oder 10 bar kultiviert. Dabei wurde die Dauer der Druckeinwirkung auf die Chondrozyten variiert. Das Expressionsmuster der so kultivierten Chondrozyten wurde quantitativ in Hinblick auf Kollagen Typ I und II sowie Aggrekan mittels qPCR und Western Blot untersucht. Dabei dienten jeweils Chondrozyten, die ohne erhöhte Druckbedingungen kultiviert wurden, als Kontrollen. In dieser Pilotstudie wurden die Proben unter Berechnung der Mittelwerte und Standardabweichung hinsichtlich ihrer statistischen Power ausgewertet. Neben dieser Analyse der Einzelergebnisse wurden die Versuchsbedingungen, die kaum Unterschiede in den Ergebnissen aufwiesen, in Gruppen zusammengefasst und mit Hilfe der größtmöglichen vorhandenen Standardabweichung der Stichprobenumfang eines Versuchs errechnet, welcher die statistische Power der Ergebnisse bei einem Alpha-Fehler von 0,05 auf 80% erhöht. In den Fällen, in denen diese Power erreicht wurde, erfolgte eine Untersuchung der Unterschiede auf Signifikanz („One Way Analysis of Variance“) bei einem Signifikanzniveau < 0,05. Ergebnisse Während der In-vitro-Kultivierung der Chondrozyten unter atmosphärischem Druck zeigte die Länge der Kultivierungszeit weder einen Einfluss auf die phasenkontrastmikroskopisch untersuchte Morphologie der Zellen noch auf die immunzytochemisch detektierte Verteilung des Kollagen Typ I und II. Die Wirkung eines erhöhten hydrostatischen Drucks (5 bar, 10 bar) für 24 Stunden führte zu einer Abnahme der Expression von Kollagen Typ I und Typ II auf das 0,2-0,8-fache bei gleichzeitiger Zunahme der Expression des Aggrekan auf das 1,7-2,2-fache, verglichen mit der unbehandelten Kontrolle. Dieser Effekt war bei 5 bar ausgeprägter als bei 10 bar, führte jedoch gleichzeitig zu einer starken Instabilität des Zellkultursystems. Vor diesem Hintergrund wurde für den höheren Druck (10 bar) die Zeitdauer der Druckeinwirkung verkürzt. Hierbei konnten bei kurzzeitiger Druckeinwirkung von 10 bar (1,5 und 3 Stunden) bei Erhalt der Zellen ähnliche Effekte erzielt werden wie für die Bedingung 5 bar, 24 Stunden. Die Expression von Kollagen Typ I und Typ II sank auf das 0,8-fache, wohingegen ein Anstieg der Aggrekanexpression auf das 1,6-2,4-fache erreicht wurde. Diese Ergebnisse der qPCR konnten durch die im Western Blot für Kollagen Typ I, II und Aggrekan detektierte Proteinexpression gestützt werden. Schlussfolgerungen Im Rahmen dieser Arbeit wurde ein In-vitro-System etabliert, welches einerseits der Untersuchung des Einflusses von hydrostatischem Druck auf die Expression von Chondrozyten dienen und andererseits für die Herstellung von Zelltransplantaten weiter modifiziert werden kann. Die Ergebnisse der Untersuchungen führten zur Definition von Bedingungen für das In-vitro-System, unter denen die Expression der extrazellulären Matrix durch die Chondrozyten in Richtung der Zusammensetzung im hyalinen Knorpel stimuliert werden kann. Dies zeigte sich bei einer Aussaat der humanen Chondrozyten in einer Konzentration von 104 Zellen/cm2 und einer Vorkultivierungszeit von 6 Tagen unter Normaldruck, gefolgt von der Kultivierung unter hydrostatischem Druck von 10 bar für 1,5 bis 3 Stunden. Mit Hilfe dieser Pilotstudie wurde somit ein In-vitro-System etabliert, auf dessen Basis Untersuchungen durchgeführt werden können, die weiterführende Erkenntnisse zur Biodynamik des hyalinen Knorpels liefern und der zukünftigen Entwicklung zellbasierter Therapieansätze der Knorpeldefekte, wie der ACT, zu Gute kommen.

During early embryonic development three pairs of major veins are presented: vitelline veins, umbilical veins and cardinal veins. The paired cranial cardinal veins drain blood from the head and neck. Cranial cardinal veins give rise to the external and internal jugular veins, brachiocephalic vein and cranial vena cava. During anatomical dissection we have observed a persistent left cranial cardinal vein (left cranial vena cava) in one dog. Caudal to the right subclavia vein, a thin left cranial cardinal vein arose and branched into two branches: the internal thoracic vein and costocervical vein. This vein was much thicker than cranial vena cava. After branching, the cranial cardinal vein run caudoventrally, passed laterally the aortic arch (ventrally to the brachiocephalic trunk), pulmonary trunk and left auricle. Caudally to the left auricle this vein run to the coronary sulcus toward caudal vena cava and entered the coronary sinus. Persistent left cranial cardinal vein in dogs are rare, unlike in humans were it represents the most common congenital venous anomaly of the thoracic venous system. Considering that the anomaly has been perceived as an accidental finding during anatomical dissection, and according to anamnestic data, the dog did not have any clinical manifestation of this condition.

In order to analyze morphologic processes occurring during development of mammalian nervous system, several specific mouse strains have been constructed. This study was based on the mouse strain B6.Cg-Tg(Thy1-YFP)16Jrs/J which under the control of Thy1 gene promoter expresses yellow fluorescent protein (YFP) in all parts of neurons. This allows to precisely describing development of otherwise less visible parts of nervous cells, including axon and dendrites. During the first 10 days of embryonic development expression of YFP was not observed. First signs of expression were observed at embryonic day 12.5 (E12.5) in ventral horn of the spinal cord. During development of the embryo in the cranio-caudal direction, a stronger expression of YFP was observed in the cranial part of the spinal cord. Moreover, expression was significantly stronger in the spinal cord than in the brain. In accordance with the process of neurogenesis which primarily occurs in mid-gestation, expression of YFP in nerve cells after E12.5 become much more pronounced. Thus expression of YFP was observed in the brain, especially at the basis of the brain by E13.5. With embryo maturation, expression became stronger in the brain and by E17.5 expression was observed in the optic nerve and retina. In the newborn mouse P0 expression was observed in the olfactory bulb. Analyses of expression of Thy-YFP construct in B6.Cg-Tg(Thy1-YFP)16Jrs/J strain revealed a high rate of development of nervous system cells starting at E12.5 and increasing during later stages of gestation and reaching the first days of postnatal life.
