ObjectiveTo analyze from a pathological perspective the differences between intermittent and persistent hydronephrosis in children with uretero-pelvic junction obstruction.Methods23 children who underwent unilateral dismembered pyeloplasty (Anderson-Hynes operation) for intermittent hydronephrosis from September 2017 to March 2024 were included in the observation group. They were compared with a control group consisting of 23 children with persistent hydronephrosis matched for age, gender, and affected side. All children had the narrowed segment surgically excised during the operation, while other obstructive causes (such as polyps, crossing vessels, or tumor compression) were excluded. The specimens were analyzed for muscle and collagen content using Masson's trichrome staining, and the collagen-to-muscle ratio (CMR) was calculated. The number of Cajal-like cells was quantified with c-kit immunohistochemical staining. Understanding the ignition and dynamic processes for the combustion of hydrate is crucial for efficient energy utilization. Through reactive force field molecular dynamics simulations, we studied the high-temperature decomposition and combustion processes of methane hydrates in a pure oxygen environment. We found that at an ignition temperature of 2800 K, hydrates decomposed from the interface to the interior, but the layer-by-layer manner was no longer strictly satisfied. At the beginning of combustion, water molecules reacted first to generate OH•, followed by methane oxidation. The combustion pathway of methane is C H 4 → CH 3 • → C H 3 O • → C H 2 O → H C • O → HCO O • → CO (C O 2 ). During the combustion process, a liquid water layer was formed between melted methane and oxygen, which hindered the reaction's progress. When there is no heat resistance, oxygen will transform into radicals such as OH• and O•, which have faster diffusion rates, allowing oxygen to conveniently cross the mass transfer barrier of the liquid water layer and participate in the combustion process. Increasing the amount of OH• may cause a surge in the reaction. On the other hand, when significant heat resistance exists, OH• is difficult to react with low-temperature hydrate components, but it can transform into O• to trigger the oxidation of methane. The H• generated has a sufficient lifetime to contact high-temperature oxygen molecules, converting oxygen into radicals that easily cross the water layer to achieve mass transfer. Therefore, finding ways to convert oxygen into various radicals is the key to solving the incomplete combustion of hydrates. We describe here the results from the testing of a small molecule first-in-class apolipoprotein E4 (ApoE4)-targeted sirtuin1 (SirT1) enhancer, A03, that increases the levels of the neuroprotective enzyme SirT1 while not affecting levels of neurotoxic sirtuin 2 (SirT2) in vitro in ApoE4-transfected cells. A03 was identified by high-throughput screening (HTS) and found to be orally bioavailable and brain penetrant. In vivo, A03 treatment increased SirT1 levels in the hippocampus of 5XFAD-ApoE4 (E4FAD) Alzheimer's disease (AD) model mice and elicited cognitive improvement while inducing no observed toxicity. We were able to resolve the enantiomers of A03 and show using in vitro models that the L-enantiomer was more potent than the corresponding D-enantiomer in increasing SirT1 levels. ApoE4 expression has been shown to decrease the level of the NAD-dependent deacetylase and major longevity determinant SirT1 in brain tissue and serum of AD patients as compared to normal controls. A deficiency in SirT1 level has been recently implicated in increased tau acetylation, a dominant post-translational modification and key pathological event in AD and tauopathies. Therefore, as a novel approach to therapeutic development for AD, we targeted identification of compounds that enhance and normalize brain SirT1 levels.

• Intercropping intercepted more light than sole peanut but less than sole maize.• Maize light use efficiency (LUE) increased with plant density in the intercropping.• Intercropping did not affect LUE of maize but increased peanut LUE.Intercropping increases crop yields by optimizing light interception and/or use efficiency. Although intercropping combinations and metrics have been reported, the effects of plant density on light use are not well documented. Here, we examined the light interception and use efficiency in maize-peanut intercropping with different maize plant densities in two row configurations in semiarid dryland agriculture over a two-year period. The field experiment comprised four cropping systems, i.e. monocropped maize, monocropped peanut, maize-peanut intercropping with two rows of maize and four rows of peanut, intercropping with four rows of maize and four rows of peanut, and three maize plant densities (3.0, 4.5 and 6.0 plants m−1 row) in both monocropped and intercropping maize. The mean total light interception in intercropping across years and densities was 779 MJ·m−2, 5.5% higher than in monocropped peanut (737 MJ·m−2) and 7.6% lower than in monocropped maize (843 MJ·m−2). Increasing maize density increased light interception in monocropped maize but did not affect the total light interception in the intercrops. Across years the LUE of maize was 2.9 g·MJ−1 and was not affected by cropping system but increased with maize plant density. The LUE of peanut was enhanced in intercropping, especially in a wetter year. The yield advantage of maize-peanut intercropping resulted mainly from the LUE of peanut. These results will help to optimize agronomic management and system design and provide evidence for system level light use efficiency in intercropping.

Background and purpose: Liver fibrosis is a critical risk factor for the progression from chronic liver injury to hepatocellular carcinoma. Clinically, there is a lack of therapeutic drugs for liver fibrosis. Previous studies have confirmed that GL‐V9, a newly synthesized flavonoid derivative, exhibits anti‐inflammatory activity, but whether it has anti‐hepatic fibrosis actions remains unclear. This study aimed to investigate the anti‐fibrotic activities and potential mechanisms of GL‐V9. Experimental approach: Bile duct ligation (BDL) and carbon tetrachloride (CCl4) challenges were used to assess the anti‐fibrotic effects of GL‐V9 in vivo. Mouse primary hepatic stellate cells (pHSCs) and the human HSC line LX2 also served as a liver fibrosis model in vitro. Cellular functions and molecular mechanism were analysed using senescence‐associated beta‐galactosidase staining, real‐time PCR, western blotting, immunofluorescence, and co‐immunoprecipitation. Key results: GL‐V9 attenuated hepatic histopathological injury and collagen accumulation, as well as decreasing the expression of fibrotic genes in vivo. GL‐V9 promoted senescence and inhibited the expression of fibrogenic genes in HSCs in vitro. Mechanistic studies revealed that GL‐V9 induced senescence by upregulating GATA4 expression in HSCs. Further studies confirmed that GL‐V9 stabilized GATA4 by promoting autophagic degradation of P62. Conclusion and implications: GL‐V9 exerted potent anti‐fibrotic effects both in vivo and in vitro by stabilizing GATA4, thereby promoting the senescence of HSCs, and by avoiding its activation and ultimately inhibiting liver fibrosis. This action indicated that the flavonoid GL‐V9 is a potential therapeutic candidate for the treatment of liver fibrosis. [ABSTRACT FROM AUTHOR]
