A 2–yr production study was conducted to evaluate 3 systems of growing stocker calves under rotational stocking. One group of steers was stocked on pasture from early April to mid August (springstocked steers = SSS), and another group of steers was stocked from early July to late October (fall–stocked steers = FSS). Steers were stratified by BW (n = 72, BW=229±llkgforSSS;n=72,BW=248±18 kg for FSS) and randomly assigned to 1 of 3 treatments. The 3 treatments were 1) rotationally stocked only (control; CON), steers rotated to a new paddock as forage availability dropped below acceptable levels in the occupied paddock; 2) rotationally stocked with distillers grains (DIST); this was the same as CON except steers were supplemented with varying amounts of distillers dried grains with solubles (DDGS) based on forage nutritive value; and 3) rotationally stocked with round–bale silage (SIL); excess forage in spring was harvested and stored as round–bale silage and fed back as needed. Total BW gain/ha over the entire grazing season did not differ between DIST and SIL (P = 0.09) steers, but both were greater thall CON (P < 0.01). Total BW gain/ha for DIST, SIL, and CON was 459 (SEM = 11.5), 402 (SEM = 31.0), and 276 (SEM = 26.2) kg, respectively. For ADG, there was a group x year (P < 0.01) and group x treatment (P = 0.02) interaction. Steer ADG for SSS did not differ between SIL and DIST (P = 0.51), but was greater than the CON (P = 0.01). The ADG for SSS was 0.79 (SEM = 0.04), 0.81 (SEM = 0.05), and 0.62 (SEM = 0.05) kg for DIST, SIL, and CON, respectively. For the FSS, ADG for all 3 treatments was different (P = 0.02). The FSS ADG was 0.72 (SEM 0.03), 0.53 (SEM = 0.04), and 0.29 (SEM = 0.04) kg for DIST, SIL, and CON, respectively. The only treatment with equivalent (P = 0.07) ADG between early and late–stocked steers (SSS vs. FSS) was DIST. Adjusting the amount of DDGS supplemented to steers based on forage nutritive value resulted in consistent BW gains throughout the grazing study, whereas steers in the SIL and CON treatments gained less BW during the latter portion of the season. Controlling forage maturity by removal in the SIL treatment resulted in total BW gains/ha that were not different than the DIST treatment. [ABSTRACT FROM AUTHOR]

The article reports on interaction of formaldehyde with soil humic substances (HS). HS are major components of soil organic matter. They are structurally complex, containing a wide variety of functional groups, depending on such factors as their age and the parent material from which they are derived. Formaldehyde is present in the environment both as a result of natural processes and from man-made sources and is considered an environmental contaminant due to detection at levels higher than background concentrations. Gel filtration chromatography provides a simple and reliable method of directly separating HS into fractions based on differences in their molecular sizes as they pass through a column packed with a gel matrix with a specific pore size range.

Gender-dependent metabolic variation in Han Wistar rats (n=25 male and n=25 female) was investigated using (1)H nuclear magnetic resonance (NMR) spectroscopy of urine coupled with chemometric methods. Statistically discriminatory regions of the spectra for male and female rats were identified and biomarker characterization was achieved by the further application of solid-phase extraction chromatography with NMR detection and high-performance liquid chromatography mass spectrometry. A novel discriminating molecule was identified as the sulfate conjugate of m-hydroxyphenylpropionic acid, which was excreted in higher concentrations by male rats. Other gender-related metabolite differences in the urine profiles included higher levels of trimethylamine-N-oxide, N,N'-dimethylglycine, m-hydroxyphenylpropionic acid, N-acetylglycoprotein, and cholate in samples from female animals. These studies emphasize the utility of multicomponent metabolic profiling for investigating physiological and genetic variation in experimental animals that may be of relevance to their use as models of toxicity and disease.

Directly coupled HPLC-NMR-MS was used to characterize two major metabolites of 5-trifluoromethylpyridone (2-hydroxy-5-trifluoromethylpyridine), a model compound for herbicides, after it had been dosed into hydroponically grown maize plants. The combination of NMR and MS data allowed the identification of both of these metabolites, namely, the N-glucoside and O-malonylglucoside conjugates of the parent pyridone. This work demonstrates the efficiency and the potential application of HPLC-NMR-MS to the investigation of the metabolism of agrochemicals. The work also indicates that combination of the use of hydroponically grown plants and directly coupled HPLC-NMR-MS allows rapid identification of metabolites with little sample preparation.

Reconstructed ion chromatograms have been used to identify relevant high performance liquid chromatography (HPLC) peaks in a directly coupled high performance liquid chromatography/nuclear magnetic resonance spectroscopy/mass spectrometry (HPLC/NMR/MS) experiment. This has been applied to a study of the metabolism of a model compound, 5-nitropyridone (2-hydroxy-5-nitropyridine), in maize plants grown hydroponically. By monitoring the on-flow reconstructed ion chromatogram corresponding to the 5-nitropyridone fragment at m/z 143, and additional molecular ions corresponding to metabolites identified as products from similar compounds, relevant peaks were identified rapidly for subsequent stopped-flow 1H NMR spectroscopic analysis. The combination of coupled HPLC/NMR/MS enabled the direct identification of three metabolites, namely the N-glucoside, N-malonylglucoside, and O-malonylglucoside. This work demonstrates the power of HPLC/NMR/MS for the structural elucidation of xenobiotic metabolites in complex biological matrices (such as plant material) with minimal sample preparation. In particular, using mass spectrometry for the initial identification of relevant HPLC peaks allows the analysis of complex samples without the necessity for other spectroscopic markers, such as 19F NMR signal for fluorinated compounds or UV spectroscopy for molecules with strong UV chromophores.

Direct coupling of NMR spectroscopic detection with both capillary zone electrophoresis (CZE) and capillary electrochromatography (CEC) was applied to the separation of metabolites of the drug paracetamol in an extract of human urine. Continuous-flow CZE-NMR and CEC-NMR allowed the detection of the major metabolites, the glucuronide and sulfate conjugates of the drug and the endogenous material hippurate. Identification of these substances was achieved by examination of individual rows of the NMR chromatogram and this also gave estimates of the detection limits. For CEC-NMR, spectra were also obtained in the stopped-flow mode including a two-dimensional TOCSY NMR experiment which afforded confirmatory evidence for paracetamol glucuronide. Characterisation of drug metabolites using NMR spectroscopy is therefore possible with nanolitre sample volumes.

Measuring procedures used for hydrogel contact lenses are sufficiently difficult to make identification of a given soft lens annoyingly time-consuming and sometimes impossible. Etching a number or letter on the lens when it is received from the laboratory allows the practitioner to tell whether the proper lens is being worn, even while the lens is still on the patient's eye. A sharp pointed tool on a dry hydrogel lens produces this identifying number.

Our ability to manipulate ourselves and the objects which surround us in our object space depends upon reasonably precise localization of the perceived images of these objects in our visual space. To be functionally sensible, this premise suggests that a "center of projection" must be located somewhere on the subject's body. Two studies which were done in an effort to illuminate an earlier study in regard to the center of projection are reported. One study used an "optical gun" which forced the subject to "shoot" at a perceived target. A second study used a focused beam headlamp to indicate the perceived direction of a perceived target. The perceived direction of the targets was altered by changes in the accommodative stimulus. Since the subject was aware only of the perceived direction of the image and since, in addition, he could not see the results of his efforts to localize the target, the location of a fixed center of projection could not be demonstrated.
