3. Inhibition of Aflatoxin B1 Production by Procyanidins Present in Annona muricata and Uncaria tomentosa Aqueous Extracts.

Abstract

Aflatoxin B1 (AFB1), primarily produced by Aspergillus flavus and A. parasiticus, is the most dangerous mycotoxin for humans and contaminates a variety of crops. To limit fungal growth and aflatoxin production in food and feed, research has been increasingly focusing on alternatives to pesticides. Studies show that some aqueous plant extracts with strong antioxidant properties could significantly impact AFB1 production, representing an eco-friendly and sustainable method to protect crops. The present study demonstrates that aqueous extracts of Anonna muricata (AM) and Uncaria tomentosa (UT) inhibit AFB1 synthesis in a dose-dependent manner with a half-maximal inhibitory concentration of 0.25 and 0.28 mg dry matter per milliliter of culture medium, respectively. This effect correlates with the presence of polyphenols and, more precisely, with condensed tannins. It is also related to the subsequent antioxidant activity of both extracts. A bio-guided fractionation followed by high-performance liquid chromatography and mass spectrometry analysis of the active fractions identifies procyanidins and, more precisely, catechin (5.3% w/w for AM and 5.4% w/w for UT) and epicatechin (10.6% w/w for AM and 25.7% w/w for UT) as the major components in both extracts. The analysis of how pure standards of these molecules affect AFB1 production demonstrates that catechin plays an essential role in the inhibition observed for both plant extracts, since the pure standard inhibits 45% of AFB1 synthesis at a concentration close to that of the extracts. Abstract

Aflatoxins are carcinogenic mycotoxins that may contaminate many crops and more especially maize. To protect consumers from these contaminants, many countries set up low regulatory thresholds of few µg/kg. The control of food requires time-consuming analysis for which sampling is a key step. It would therefore of key sanitary and economic relevance to develop rapid, sensitive and accurate methods that could even be applied on line at harvest, to identify batches to be excluded as soon as possible. In this study, we analyzed more than 500 maize samples taken at harvest during 3 years for their aflatoxin contamination using HPLC-MS. Among them, only 7% were contaminated but sometimes at levels largely exceeding European regulations. We demonstrate that Near InfraRed Spectroscopy (NIRS) could be of great help to classify cereal samples according to their level of aflatoxin contamination (below or higher than E.U. regulation). To build the model, all AF contaminated samples as well as an equivalent number of AF free samples were used. NIRS performance was not sufficient to quantify the toxins with adequate precision. However, its ability to discriminate naturally contaminated maize samples according to their level of contamination with aflatoxins in relation to European regulations using a quadratic PCA-DA model was excellent. Abstract

Microbial contamination of the surface of building materials and subsequent release of microbial particles into the air can significantly affect indoor air quality. Avoiding the development or, at least, reducing the quantity of microorganisms growing on building materials is a key point to reduce health risks for building occupiers. In that context, the antimicrobial activity of TiO2, ZnO and Au/ZnO was assessed by measuring log reductions of Escherichia coli and Aspergillus niger populations both in the dark and under a light intensity close to real-life conditions. The bactericidal activities (≥2.3 log reduction) of tested products were stronger than their fungicidal activities (≤1.4 log reduction) after 2 h of contact. Different parameters including concentration of photocatalyst, intensity of light (dark vs. 5 W/m2 UV-A), and duration of contact between photocatalyst and microbial cells and spores were investigated. Results of this study confirmed bactericidal activities of TiO2, ZnO and AuZnO on E. coli and brought new insight on their fungicidal activity on the spores of A. niger. They also confirmed the greatest antimicrobial efficiency of ZnO compared to TiO2 and its increased photocatalytic activity when decorated with Au, leading to the highest log reductions detected after 2 h of contact for both tested microorganisms (4 and 1.4 for E. coli and A. niger, respectively). Abstract

International audience; The objective of grain storage control is to ensure a secure supply of food industries both in quantity and quality between two harvests. Despite efforts to monitor and control the environment, there are still risks of material and/or quality loss that can reach 50% of the total value of the harvest in developing countries, and between 5 and 10% in industrialized countries. In industrialized countries, economic losses are more associated with deterioration of the sanitary and technological quality of grain than with losses of material. Control of this quality is a real challenge, especially during the storage phase. Among other factors, fungal contamination and subsequent mycotoxins production present serious problems in terms of diminishing grain quality and health risks for consumers. It has been shown that moulds emit Volatile Organic Compounds (VOCs) when they colonize a substrate, these compounds being detectable before the infestations are visible. The fungal contamination Index (FCI) was initially developed by CSTB to monitor early fungal developments in different indoor environments. This study aims to test the feasibility to use VOCs emission and especially FCI to detect fungal growth on maize grains during storage. First results allowed the characterization of specific VOC profile of maize, with more than 100 different VOC emitted during grain storage, in laboratory conditions. Furthermore, results clearly demonstrated that the development of fungi following artificial contamination or development of mycoflora initially present on grains led to the emission of a different pattern of VOCs.

Abstract

The aim of this study was to compare the zootechnical and health impact, and residues in edible tissues ofdeoxynivalenol (DON), fumonisins (FBs) and zearalenone (ZEN) in single and multi-contamination in broilerfor a chronic exposure (0 to 35 days of age). The mycotoxins were obtained by culturing toxinogenic fungalstrains and incorporated into the experimental feeds to obtain a DON diet (5 mg / kg), a FBs diet (20 mg / kg),ZEN diet (0.5 mg / kg) and a multi-contaminated diet (5 mg/kg DON + 20 mg/kg FBs+ 0.5 mg/kg ZEN),whereas the control diet did not contain mycotoxins. These feeds were individually given to 70 animals allocatedto the 5 groups. Exposure to mycotoxins did not significantly impact the zootechnical performance of broiler. Atday 35, blood analysis revealed no significant difference among groups on routine plasma biochemistry andvarious markers of oxidative stress, except an elevation of sphinganine/sphingosine ratio (plasma and liver) foranimals exposed to FBs compared to unexposed, without difference between single or multi-contaminatedgroups. The search for residues revealed higher levels of FB1 and ZEN (free and conjugated forms) in liver thanin muscle, and no interactive effect of multi-contamination on the persistence of these contaminants.; L’objectif était de comparer l’impact zootechnique et sanitaire du déoxynivalénol (DON), des fumonisines (FBs)et de la zéaralénone (ZEA) et d’évaluer leur niveau de persistance à l’état résiduel dans les produits comestibles,suite à une mono ou multi-contamination chez le poulet de chair lors d’une exposition chronique (J0-J35). Lesmycotoxines ont été obtenues par culture de souches fongiques toxinogènes et incorporées aux alimentsexpérimentaux pour obtenir un lot DON (5 mg/kg), un lot FBs (20 mg/kg), un lot ZEA (0,5 mg/kg) et un lotmulti-contaminé (5 mg/kg DON + 20 mg/kg FBs+ 0,5 mg/kg ZEA) ; l’aliment du lot témoin ne contenait pas demycotoxines. Ces aliments ont été individuellement distribués à 70 animaux répartis entre les 5 lots.L’exposition aux mycotoxines n’a pas significativement altéré les performances zootechniques des poulets. AJ35, l’analyse de sang n’a révélé aucune différence significative entre lots sur la biochimie plasmatique deroutine et différents marqueurs de stress oxydatif. Seule une élévation significative du rapportsphinganine/sphingosine chez les animaux exposés aux FBs par rapport aux animaux non exposés a été observée dans le plasma, sans différence entre poulets recevant l’aliment mono- ou multi-contaminé. La recherche de résidus a révélé des teneurs plus élevées en FB1 et ZEA (formes libres et conjuguées) dans le foie que dans les muscles, et l’absence d’effet interactif de la multi-contamination sur la persistance de ces composés.

Abstract

Mycotoxins are secondary metabolites produced by molds. The presence of these contaminants in animal feed isa major problem because mycotoxins have various adverse effects on health, affecting in particular certainfunctions such as reproduction. The objective of our approach is to evaluate the impact of the main fusariotoxins,in vivo and in vitro, on the testicular functions of broiler chickens. For the in vivo test, 5 groups of 12 broilers(Ross PM3) received the control or experimentally contaminated diet from hatch to slaughter (35 days of age).The diets were contaminated in: deoxynivalenol (DON, 5 mg / kg), fumonisins (FBs, 20 mg / kg), zearalenone(ZEA, 0.5 mg / kg) in single contamination or containing the 3 mycotoxins. The mycotoxins required for theexperiments were obtained by culturing toxigenic fungal strains.Analysis of the testis growth and histological examination has shown that the proliferative activity of the testis isincreased in animals exposed to FBs compared to the control group. On the other hand, no difference was foundin markers of cell death (caspase 3 activity and TUNEL immunostaining), of inflammation (IL6, IL1β andIFNɣ), or in oxidative stress (antioxidant capacity and catalase activity). In vitro experiments using purifiedmycotoxins diluted in a solvent (DMSO) were tested on chicken testicular cell cultures after 96 h of exposure.The results confirmed the in vivo data showed no effect at low dose. However, high doses (FB1: 100 nM, ZEA:30 μM, DON 1 μM) have an impact on cell viability.In conclusion, in vitro, only high doses can lead to testicular lesions but in vivo the maximum concentrationsacceptable did not induce any significant effect on the testes of 35 days-old chicken.; Les mycotoxines sont des métabolites secondaires produits par certaines moisissures. La présence de cescontaminants dans les aliments destinés aux animaux est un problème majeur, car les mycotoxines peuvent avoir des effets néfastes variés sur leur santé, en impactant notamment certaines fonctions telle que la reproduction. L’objectif de notre démarche est d’évaluer l’impact des principales fusariotoxines, in vivo et in vitro sur les fonctions testiculaires du poulet de chair. Pour l’essai in vivo, 5 groupes de 12 poulets de chair (Ross PM3) ont reçu de la naissance à l’abattage (35 jours d’âge) les aliments suivants : un aliment témoin sain ou un des 4 aliments expérimentalement contaminés avec du déoxynivalénol (DON, 5mg/kg), des fumonisines (FBs, 20 mg/kg), de la zéaralénone (ZEA, 0,5 mg/kg) en contamination unique ou contenant les 3 mycotoxines. Les mycotoxines nécessaires aux expérimentations ont été obtenues par culture de souches fongiques toxinogènes. L’analyse de la croissance et l’examen histologique du testicule ont montré que l’activité proliférative du testicule, est augmentée chez les animaux exposés aux FBs, en comparaison du groupe témoin. Par contre, aucune différence sur les marqueurs de mortalité cellulaire (activité caspase 3 et immunomarquage TUNEL), d’inflammation (IL6, IL1β et IFNɣ), ou de stress oxydatif (capacité antioxydante et activité de la catalase) n’a été révélée entre les lots. Des mycotoxines purifiées et diluées dans un solvant (DMSO) ont été testés sur des cultures testiculaires de poulets après 96h d’exposition. Les résultats ont confirmé les données in vivo montrant aucun effet à faible dose et un effet négatif des doses élevées (FB1: 100 nM ; ZEA : 30 μM, DON 1μM). En conclusion, in vitro, seules des fortes doses peuvent conduire à des lésions testiculaires mais in vivo les doses maximales autorisées n’induisent pas d’effet négatif notable sur les testicules de poulets élevés jusqu’à 35j.

Abstract

The aim of this study was to compare zootechnical and health impact of deoxynivalenol (DON), fumonisins(FBs) and zearalenone (ZEN) and to assess their residual persistence level in edible tissues, following a mono ormulti-contamination of ducks during chronic exposure in the gavage phase. Abstract

Couscous “Lemzeiet” was made from fermented wheat using a traditional process specific to the region of Constantine (North East of Algeria). This study involved the valorisation of traditional fermentation techniques of wheat in an underground silo called “Matmor” and manufacture of couscous “Lemzeiet”. The first part of this study consisted in a survey conducted in the city of Constantine. The 229 participants were from the east of Algeria: Constantine (191), Sigus (22) and Mila (16). The age of the individuals ranged between 20 and 87 years. Thirty-six of them were represented by men, who practice the fermentation of wheat naturally in the “Matmor”. Fifty-eight were represented by women who manufacture the couscous “Lemzeiet” by a process which we have reproduced experimentally in order to assess the technological and nutritional quality of couscous. It involved the use of a mixture of fine semolina (“dkak”) of fermented wheat (70%) and white couscous (30%) for forming the grains, a mixture of eggs (50 %) and “smen” (50%) for the finishing step, and a series of sieves with openings of 410 -2532 μm, for calibration and sieving the couscous. The experimentally manufactured couscous “Lemzeiet” was composed (%) of three different granulometric sizes fractions (μm) as follows: [1523-2532]: 50, 11%;] 1140-1523]: 29, 80%;] 899-1140]: 20, 51%. The grains were homogeneous with a good aggregation, rounded and smooth and not sticky after steaming. The analysis of the chemical composition showed significant changes in lipid, proteins and ash of manufactured couscous “Lemzeiet”.

Abstract

International audience; Mycotoxins are toxic secondary metabolites produced by many fungal species. They are frequent contaminants of many foods and feeds and especially cereals. More than 100 countries set up regulations for these natural contaminants to prevent toxicity in animals and humans. Many analytical methods are developed to assess contamination. Most of the official methods rely on liquid chromatography however the use of immunological tools allowed the development of rapid methods for screening purpose. Mass spectrometry is the method of choice for multi-mycotoxin analysis.

National audience; Les fumonisines (FBs) sont des mycotoxines qui contaminent principalement le maïs. Chez le canard, une altération de la croissance et des pertes de productivité ont été observées lors d’expositions voisines des valeurs maximales recommandées en FBs dans l’aliment volaille. Ces niveaux n’ont pas les mêmes conséquences chez la dinde. Les teneurs en Sphinganine (Sa) sont de bons marqueurs d’exposition aux FBs dans toutes les espèces. Le métabolisme des sphingolipides du canard est également plus sensible aux FBs que la dinde. L’objectif de cette étude est de déterminer si les différences de toxicité constatées entre canards et dindes lors d’exposition aux FBs sont liées à des différences d’effets sur le métabolisme des sphingolipides, principalement sur la phosphorylation de la Sa et son excrétion cellulaire. Des FBs ont été administrées par gavage a des canards et des dindes recevant 0 (groupe témoin) ou 10 mg de FB1+FB2/kg de poids vif. Cinq animaux par lot ont été sacrifiés à 1, 4, 8, 15 et 22 jours d’exposition, le sérum et le foie ont été prélevés afin de mesurer la Sa dans ces organes ainsi que ses formes phosphorylées. Cette étude a confirmé la plus grande sensibilité des canards à la FB1. Lors de l’exposition une diminution de la capacité de phosphorylation de la Sa et une diminution de l’excrétion des bases sphingoides est observée dans les deux espèces.
