1. Genetic Polymorphisms in CYP2 Gene Family in Bulgarian Individuals and their Clinical Implications

Abstract

The cytochrome P450 superfamily consists of hemeproteins involved in the detoxication of different xenobiotics, including drugs. The CYP2 gene family is responsible for the metabolism of 80% of the drugs in clinical use. There are considerable interindividual and interethnic variabilities in the rate of drug metabolism as a result of genetic polymorphisms. The goal of our study was to determine the frequency of 10 genetic polymorphisms in CYP2 family genes to give light on the pharmacogenetic defects of the main CYPs, involved in drug metabolism, in Bulgarian individuals. We detected high allele frequency for CYP2D6*10 (0.27), CYP2D6*4 (0.22), and CYP2B6*9 (0.24), followed by CYP2C19*2 (0.14), CYP2C9*3 (0.11) and CYP2C9*2 (0.09). The genotype frequencies were also determined for all investigated variants. In total 47.2% of the analyzed individuals carried CYP2D6 genetic polymorphisms – 5.6% carried a single variant and 41.6% were found to have two or more such variants. Homozygotes for CYP2D6 variants were established among 14% of Bulgarian individuals. Determination of the prevailing pharmacogenetic polymorphisms of the CYPs, most responsible for drug metabolism, will lead to a lower risk of drug toxicity, increased drug efficacy, and drug dose optimization.

Abstract

Thyroid hormones and the hypothalamic-pituitary-thyroid (HPA) axis are responsible for multiple metabolic processes and psychological well-being. Hypothyroidism can induce mood changes, depressive symptoms and even major depressive disorder. This review is focused on the pathophysiology and mechanisms through which the low level of thyroid hormones may affect the brain function, causing the characteristic symptoms of depression. Key pathways of hypothyroid-associated depressive states include: morphological changes in some brain areas (mainly in the hippocampus – a reduction in its volume); a significant reduction of the cerebral blood flow (incl. hippocampus), and lowered levels of neurotrophic factors (e. g. BDNF – brain-derived neurotrophic factor), which are regulated by the thyroid hormones. An adequate and timely thyroid hormone replacement and treatment with conventional antidepressants often can reverse the psychological symptoms.

Abstract

The rising burden of depression, which will soon be the second most common cause of disability in the world, is requesting new ways to treat and prevent it. Due to high number of significant adverse drug reactions of the conventional treatment, the modern pharmaceutical industry is more often turning their focus to novel plant-based solutions. We performed literature research based on standard literature search engines – PubMed, Google Scholar, Science Direct. A standard set of keywords related to our topic e.g. “Depression”, “Mesembrine type alkaloids”, “Narcissus” was used. The review describes the classical monoamine theory of depression and connects it with the newly found biochemical, genetic and morphological alterations associated with the major depressive disorder. The purpose of this review is to highlight the most important aspects of the pathophysiology of depression and to explore the possibilities to use mesembrine-like alkaloids isolated from Narcissus cv. Hawera in its treatment. We describe their effect on brain biochemistry and possible future investigations.

Surprisingly, very few studies testing the effect of the KD in corneal kindling test were published. The aim of this study was to evaluate the effect of the KD on the seizure activity in corneal kindling mouse model.

Abstract

The FAMU experiment at RIKEN-RAL is a muonic atom experiment with the aim to determine the Zemach radius of the proton by measuring the 1s hyperfine splitting in muonic hydrogen. The activity of the FAMU Collaboration in the years 2015-2023 enabled the final optimisation of the detector-target setup as well as the gas working condition in terms of temperature, pressure and gas mixture composition. The experiment has started its data taking in July 2023. In this letter, we recall the main facts concerning the g-factor of positronium and we show how the value of the g-factor of the positronium is important. Taking it better into consideration may provide a solution to the reported discrepancy between QED theory and experiments concerning the hyperfine splitting of the fundamental level of the positronium. Abstract

Data from different studies correlating the serum 25(OH)D levels with the metabolic and glycemic parameters in type 2 diabetes patients are still varying. The objective if this study was to describe the correlation between serum 25(OH)D levels and some metabolic parameters in Bulgarian type 2 diabetes patients on oral antidiabetic drugs. One hundred type 2 diabetes patients participated - 56 men and 44 women. The mean age and diabetes duration of the women was 59.0 and 9.8 years, of the men - 58.0 and 7.7 years respectively. Complete patient history was taken and physical examination was performed (body weight and height, waist circumference). Body composition was measured on a leg-to-leg body impedance analyzer (TBF-215, Tanita Corp., Tokyo, Japan). Serum levels of vitamin D were measured by electro-hemi-luminescent detection as 25-(ОН) D Total (ECLIA, Elecsys 2010, Roche Diagnostics, Switzerland). Glycated hemoglobin A1c was measured on a NycoCard reader (Alere™). Total, HDL-cholesterol (direct) and triglycerides were analyzed on a Cobas Integra 400+ analyzer. Correlation analysis was performed on a SPSS 13.0 for Windows platform and included 10 curves. The data were first analyzed for the group as a whole and then separately for men and women as well as in the different vitamin D tertiles. The mean serum 25-OH-vitamin D levels were 23.8 ± 12.1 nmol/l in women and 33.3 ± 20.0 nmol/l in men. We were unable to find any statistically significant correlation between serum 25(OH) vitaminand the serum lipids (cholesterol profile and triglycerides). On the contrary, there was a weak correlation with the glycated hemoglobin A1c (cubic model, R2 = 0.178, p = 0.05) and the BMI (inverse model, R2 = 0.101, p = 0.038). The sub-analyses (men versus women or according to tertiles of vitamin D) did not produce any additional information. The influence of vitamin D on the parameters of the metabolic control in type 2 diabetes is very weak on an individual level. It might be only demonstrated in large epidemiological surveys.

Abstract

We report the first experimental determination of the collision-energy dependence of the muon transfer rate from the ground state of muonic hydrogen to oxygen at near-thermal energies. A sharp increase by nearly an order of magnitude in the energy range 0 - 70 meV was found that is not observed in other gases. The results set a reliable reference for quantum-mechanical calculations of low-energy processes with exotic atoms, and provide firm ground for the measurement of the hyperfine splitting in muonic hydrogen and the determination of the Zemach radius of the proton by the FAMU collaboration., Comment: 30 pages, 10 figures

12. Forbidden 'ortho'--'para' electric dipole transitions in H$_2^+$ ion

Abstract

We present the first systematic calculation of the electric dipole forbidden transitions in the homonuclear molecular ion H$_2^+$. We get that the transition rate from the ground "ortho" $(v\!=\!0,N\!=\!1,J\!=\!1/2)$ state to the ground "para" $(v\!=\!0,N\!=\!0,J\!=\!1/2)$ state is $4.9\!\times\!10^{-14}$ s$^{-1}$ that corresponds to the lifetime of $6.4\!\times\!10^5$ yr. The Einstein coefficient $A_{nn'}$ for the $(v\!=\!1,N\!=\!1,J\!=\!1/2)\!\to\!(v\!=\!0,N\!=\!0,J\!=\!1/2)$ transition is equal to $0.12\!\times\!10^{-9}$ s$^{-1}$, which is of "comparable" order with the values for quadrupole transitions in H$_2^+$. Abstract

The first measurement of the temperature dependence of the muon transfer rate from muonic hydrogen to oxygen was performed by the FAMU collaboration in 2016. The results provide evidence that the transfer rate rises with the temperature in the range 104-300 K. This paper presents the results of the experiment done in 2018 to extend the measurements towards lower (70 K) and higher (336 K) temperatures. The 2018 results confirm the temperature dependence of the muon transfer rate observed in 2016 and sets firm ground for comparison with the theoretical predictions.

Abstract

Molecular hydrogen ions are of metrological relevance due to the possibility of precise theoretical evaluation of their spectrum and of external-field-induced shifts. In homonuclear molecular ions the electric dipole $E1$ transitions are strongly suppressed, and of primary laser spectroscopy interest is the electric quadrupole ($E2$) transition spectrum. In continuation of previous work on the H$_2^+$ ion, we report here the results of the calculations of the hyperfine structure of the laser-induced electric quadrupole transitions between a large set of ro-vibrational states of D$_2^+$; the inaccuracies of previous evaluations have been corrected. The effects of the laser polarization are studied in detail. We show that the electric quadrupole moment of the deuteron can in principle be determined with low fractional uncertainty $(\simeq1\times10^{-4})$ by comparing the results presented here with future data from precision spectroscopy of D$_2^+$., Comment: 19 pages, 4 figures. Abstract

We report the first measurement of the temperature dependence of muon transfer rate from $\mu$p atoms to oxygen between 100 and 300 K. Data were obtained from the X-ray spectra of delayed events in gaseous target H$_2$/O$_2$ exposed to a muon beam. Based on the data, we determined the muon transfer energy dependence up to 0.1 eV, showing an 8-fold increase in contrast with the predictions of constant rate in the low energy limit. Abstract

The main goal of the FAMU experiment is the measurement of the hyperfine splitting (hfs) in the 1S state of muonic hydrogen $\Delta E_{hfs}(\mu^-p)1S$. The physical process behind this experiment is the following: $\mu p$ are formed in a mixture of hydrogen and a higher-Z gas. When absorbing a photon at resonance-energy $\Delta E_{hfs}\approx0.182$~eV, in subsequent collisions with the surrounding $H_2$ molecules, the $\mu p$ is quickly de-excited and accelerated by $\sim2/3$ of the excitation energy. The observable is the time distribution of the K-lines X-rays emitted from the $\mu Z$ formed by muon transfer $(\mu p) +Z \rightarrow (\mu Z)^*+p$, a reaction whose rate depends on the $\mu p$ kinetic energy. The maximal response, to the tuned laser wavelength, of the time distribution of X-ray from K-lines of the $(\mu Z)^*$ cascade indicate the resonance. During the preparatory phase of the FAMU experiment, several measurements have been performed both to validate the methodology and to prepare the best configuration of target and detectors for the spectroscopic measurement. Abstract

The high precision measurement of the hyperfine splitting of the muonic-hydrogen atom ground state with pulsed and intense muon beam requires careful technological choices both in the construction of a gas target and of the detectors. In June 2014, the pressurized gas target of the FAMU experiment was exposed to the low energy pulsed muon beam at the RIKEN RAL muon facility. The objectives of the test were the characterization of the target, the hodoscope and the X-ray detectors. The apparatus consisted of a beam hodoscope and X-rays detectors made with high purity Germanium and Lanthanum Bromide crystals. Abstract

The FAMU experiment aims to measure for the first time the hyperfine splitting of the muonic hydrogen ground state. From this measurement the proton Zemach radius can be derived and this will shed light on the determination of the proton charge radius. In this paper, we describe the scientific goal, the method and the detailed preparatory work. This includes the outcome of preliminary measurements, subsequent refined simulations and the evaluation of the expected results. The experimental setup being built for the measurement of the hyperfine splitting to be performed at the RAL laboratory muon facility is also described.

Abstract

We report the first measurement of the temperature dependence of muon transfer rate from muonic hydrogen atoms to oxygen between 100 and 300 K. Data were obtained from the X-ray spectra of delayed events in a gaseous target, made of a H2/O2 mixture, exposed to a muon beam. This work sets constraints on theoretical models of muon transfer and is of fundamental importance for the measurement of the hyperfine splitting of muonic hydrogen ground state as proposed by the FAMU collaboration.

Abstract

The aim of the FAMU (Fisica degli Atomi Muonici) experiment is to realize the first measurement of the hyperfine splitting (hfs) in the 1S state of muonic hydrogen Delta E-1S(hfs), by using the RIKEN-RAL intense pulsed muon beam and a high-energy mid-infrared tunable laser. This requires a detailed study of the muon transfer mechanism at different temperatures and hence at different epithermal states of the muonic system. The experimental setup involves a cryogenic pressurized gas target and a detection system based on silicon photomultipliers-fiber beam hodoscopes and high purity Germanium detectors and Cerium doped Lanthanium Bromide crystals, for X-rays detection at energies around 100 keV.Simulation, construction and detector performances of the FAMU apparatus at RAL are reported in this paper.

Abstract

The FAMU experiment aims to accurately measure the hyperfine splitting of the ground state of the muonic hydrogen atom. A measurement of the transfer rate of muons from hydrogen to heavier gases is necessary for this purpose. In June 2014, within a preliminary experiment, a pressurized gas-target was exposed to the pulsed low-energy muon beam at the RIKEN RAL muon facility (Rutherford Appleton Laboratory, UK). The main goal of the test was the characterization of both the noise induced by the pulsed beam and the X-ray detectors. The apparatus, to some extent rudimental, has served admirably to this task. Technical results have been published that prove the validity of the choices made and pave the way for the next steps. This paper presents the results of physical relevance of measurements of the muon transfer rate to carbon dioxide, oxygen, and argon from non-thermalized excited µp atoms. The analysis methodology and the approach to the systematics errors are useful for the subsequent study of the transfer rate as function of the kinetic energy of the µp currently under way.
