Cardiac point-of-care ultrasound (POCUS) is a technology increasingly leveraged at the bedside by pediatric critical care and neonatology providers to identify real-time hemodynamic pathophysiology. We present a framework for (1) identifying the scope of cardiac POCUS within the clinical practice setting, (2) standardizing views for protocolized hemodynamic assessment relevant to pediatric critical illness and (3) integrating POCUS findings for therapeutic guidance. Within the review, we also discuss practical strengths and limitations to image acquisition and interpretation within the varied cardiac POCUS views. Finally, we explore unique considerations within the neonatal population. Conclusion: Cardiac POCUS is a technology and tool that reveals important real-time information at the bedside of the critically ill child and infant. Understanding strengths and limitations of cardiac POCUS views and protocolizing an approach to answer focused clinical questions provides a framework for training and translation to clinical care. Aim: Spatial sampling bias (SSB) is a feature of opportunistically sampled species records. Species distribution models (SDMs) built using these data (i.e. presence‐background models) can produce biased predictions of suitability across geographic space, confounding species occurrence with the distribution of sampling effort. A wide range of SSB correction methods have been developed but simulations suggest effects on predictive performance are highly variable. Here, we aim to identify the SSB correction methods that have the highest likelihood of improving true predictive performance and evaluation strategies that provide a reliable indicator of model performance when independent test data are unavailable. Methods: A meta‐analysis was used to evaluate the performance of SSB correction methods in studies where there were direct comparisons between corrected and uncorrected SDMs. A simulation model was then developed to test evaluation strategies against a known truth using four common SSB correction methods. Results: Effect sizes from published studies suggest some support for small positive effects of SSB correction on predictive performance when assessed using independent test data, but this was not evident using internal cross‐validation and no single method stood out as consistently effective. Simulations support these findings and show that evaluation using internal test data was generally a poor indicator of the true effect of SSB correction. Methods that adjust models relative to a known driver of SSB produced the largest performance gains, but were also the most inconsistent. Main Conclusions: Correcting SSB in presence‐background SDMs without independent test data to evaluate the effect on model performance requires careful implementation. We recommend clearer documentation of SSB correction effects on SDMs, presenting results from models with and without correction, evaluating effects of different assumptions of SSB implementation on predictions, as well as greater efforts to collect independent test datasets to validate model predictions. Students formulate questions on weather and climate while watching the movie, and the course is then designed from the student questions. Inquiry about weather has become integral at a nearby elementary school: second graders conduct daily weather briefings when studying their unit on weather. As needed, the instructor may augment the student-led weather briefing afterward with additional information, especially in weather situations that can extend learning with new concepts. Protected areas (PAs) are a core component in global efforts to prevent further declines in biodiversity. We examine whether the United Kingdom's PA portfolio has reduced local extinctions of breeding birds and consider how the characteristics of different PA designations may have affected conservation outcomes. We use breeding bird atlas data to calculate the proportion of species in each 10 × 10‐km grid cell confirmed as breeding in 1988–1991 but not in 2008–2011. We find that the protection offered by PAs is little to no different from that of the wider landscape and, depending on the type of PA, attribute this to PAs being either too small or not managed for biodiversity outcomes. To meet the ambitious new global targets for expanding PA coverage set at the 15th UN Biodiversity Conference, all countries need to monitor and ensure PA effectiveness; high PA coverage does not guarantee that biodiversity is protected. Much previous international research has demonstrated links between general school attainment and active engagement with music. The research reported here compared the change in examination outcomes in English and mathematics in national examinations at ages 11 and 16 of instrumentalists and non-instrumentalists. Data from 701 pupils showed statistically significant differences in examination outcomes between instrumentalists and non-instrumentalists, which was also related to the duration of learning and the instrument played. Instrumentalists had greater change scores in mathematics, but not English, although instrumentalists of lower socio-economic status showed greater change in both subjects. Multiple sclerosis (MS) is characterized by the chronic inflammatory destruction of myelinated axons in the central nervous system. Several ideas have been put forward to clarify the roles of the peripheral immune system and neurodegenerative events in such destruction. Yet, none of the resulting models appears to be consistent with all the experimental evidence. They also do not answer the question of why MS is exclusively seen in humans, how Epstein-Barr virus contributes to its development but does not immediately trigger it, and why optic neuritis is such a frequent early manifestation in MS. Here we describe a scenario for the development of MS that unifies existing experimental evidence as well as answers the above questions. We propose that all manifestations of MS are caused by a series of unfortunate events that usually unfold over a longer period of time after a primary EBV infection and involve periodic weakening of the blood–brain barrier, antibody-mediated CNS disturbances, accumulation of the oligodendrocyte stress protein αB-crystallin and self-sustaining inflammatory damage. Here we describe a scenario for the development of multiple sclerosis (MS) that unifies existing experimental evidence as well as answers the above questions. We propose a hypothesis that manifestations of MS are caused by a series of unfortunate events that usually unfold over a longer period of time after a primary EBV infection and involve a periodic weakening of the blood-brain barrier, antibody-mediated CNS disturbances, accumulation of the oligodendrocyte stress protein aB-crystallin and self-sustaining inflammatory damage. This article examines how deaths related to mental health in England and Wales are investigated and the extent to which lessons are learned in their aftermath. It uses two concepts from academic literature to discuss organisational responses to these deaths: organisational learning, and organised irresponsibility. Organisational learning stresses the importance of learning lessons from data; in contrast, Beck's concept of organised irresponsibility states that organisational lesson learning is impeded by the fragmented and risk-averse nature of public institutions. The article considers 210 organisational responses to Reports to Prevent Future Deaths (PFDs) issued by Coroners. PFDs are sent to any organisation Coroners believe could act to prevent future deaths. The article identifies three findings: Firstly, organisations tend to produce generic responses rather than addressing specific issues raised by Coroners. Second, organisations tend to cite existing policies as responses to Coroners despite those policies not preventing specific deaths. Third, institutions seek to displace blame onto other organisations in attempting to avoid accepting responsibility for the death. The article adds to the canon of knowledge on deaths in healthcare, and in the care of the state by identifying significant structural weaknesses that impede organisational lesson learning about preventable deaths. Cardiac surgery is quite common in the United States. Outcomes after cardiac surgery are not only dependent on how the surgery went and how the anesthesia care was provided intraoperatively but also on the optimal management in the postoperative critical care setting. It is of paramount importance that the cardiac intensivist has a comprehensive understanding of cardiopulmonary physiology and the sequelae of cardiopulmonary bypass. Most preventable deaths after cardiac surgery have been linked to postoperative problems in the intensive care unit (ICU).1,2 Failure to recognize and rescue a patient from potentially reversible complications is a cause of perioperative morbidity and mortality. Patients who undergo cardiac surgery often present with multiple rapidly changing clinical problems; they are initially unstable with extremely fluid and dynamic clinical status. Postoperative care of these patients requires knowledge of general fundamental concepts of patient care as well as concepts unique to this set of patients. The initial management of these patients as they return from the operating room is critical, because clinical errors at this time can have far-reaching implications. The initial management should begin even before the patient arrives in the cardiovascular intensive care unit (CVICU). It is vital that the cardiac intensivist reviews the chart and notes the type of surgery, indications, preoperative hemodynamic data, comorbid conditions, medications, and allergies. Upon the patient’s arrival in the CVICU, a careful systematic assessment of the patient begins with obtaining a comprehensive handoff from the surgical and anesthesia team. The cardiac intensivist should ascertain what procedure was done in the operating room and inquire as to any intraoperative events that might impact the patient’s postoperative course. Then, they should physically examine the patient as part of this initial evaluation. During the initial assessment, the intensivist should avoid focusing on any one issue and attempt to get a global picture of the patient’s clinical status. A thorough knowledge of the specific monitoring and drug delivery lines is imperative, as is knowledge of where the drains are placed. Aim: Organisms primarily influence ecosystems through their functional traits when they are physically active. Following the nocturnal bottleneck, the expansion of mammals into the daytime expanded mammalian functional diversity (FD), however there is also notable overlap in trait space across diel niches leading to redundant FD. We explore how the unique and redundant contribution of each diel niche varies spatially and in relation to natural variation in light and temperature. Location: Global. Taxon: Extant mammals. Methods: Based on five major functional traits (body mass, litter size, diet breadth, foraging strata, habitat breadth) for 5033 extant terrestrial mammals, we determine biogeographical variation in nocturnal, crepuscular, cathemeral and diurnal FD. We calculate the proportion of mammalian FD that is unique to each diel niche, and the proportion that is redundant across the daily cycle. Results: The diversification of mammals into the daytime resulted in the creation of new FD (28.5% of FD is not nocturnal; Lower Quartile 17.3%; Upper Quartile 38.2%). Most of this expansion occurred at higher latitudes where uniquely cathemeral FD dominates (>55°N, 41.1% of mammalian FD; Lower Quartile 33.3%, Upper Quartile 53.6%), associated with fewer hours of biologically useful moonlight and daylight. Where there are more hours of biologically useful daylight, unique diurnal FD is common. However, more than half of non‐nocturnal FD is redundant, increasing ecosystem stability as different species carry out similar functions at different times of day, and suggesting that many mammals have not diversified far from their nocturnal ancestors. Main conclusions: Over much of the land surface more than a half of FD only occurs at night, underscoring the importance of nocturnal mammals for ecosystems. Understanding diel variation in FD not only informs on community structure and ecosystem function but also on ecosystem functional persistence in the Anthropocene, with pressures at night being particularly concerning. Accurate accounting of emissions and removals of CO 2 is critical for the planning and verification of emission reduction targets in support of the Paris Agreement. Here, we present a pilot dataset of country-specific net carbon exchange (NCE; fossil plus terrestrial ecosystem fluxes) and terrestrial carbon stock changes aimed at informing countries' carbon budgets. These estimates are based on "top-down" NCE outputs from the v10 Orbiting Carbon Observatory (OCO-2) modeling intercomparison project (MIP), wherein an ensemble of inverse modeling groups conducted standardized experiments assimilating OCO-2 column-averaged dry-air mole fraction (XCO2) retrievals (ACOS v10), in situ CO 2 measurements or combinations of these data. The v10 OCO-2 MIP NCE estimates are combined with "bottom-up" estimates of fossil fuel emissions and lateral carbon fluxes to estimate changes in terrestrial carbon stocks, which are impacted by anthropogenic and natural drivers. These flux and stock change estimates are reported annually (2015–2020) as both a global 1 ∘ × 1 ∘ gridded dataset and a country-level dataset and are available for download from the Committee on Earth Observation Satellites' (CEOS) website: 10.48588/npf6-sw92. Across the v10 OCO-2 MIP experiments, we obtain increases in the ensemble median terrestrial carbon stocks of 3.29–4.58 PgCO2yr-1 (0.90–1.25 PgCyr-1). This is a result of broad increases in terrestrial carbon stocks across the northern extratropics, while the tropics generally have stock losses but with considerable regional variability and differences between v10 OCO-2 MIP experiments. We discuss the state of the science for tracking emissions and removals using top-down methods, including current limitations and future developments towards top-down monitoring and verification systems. Aim: There is a convergence between the peak age of onset of mental illness, offending behaviour and associated risk of crime victimization. This study compared self‐reported childhood maltreatment and mental health outcomes among a cohort of help‐seeking young people who disclosed either a prior criminal charge, crime victimization, or both, to those who did not report any such experiences. The outcomes may inform the development of a diversion option for young people at a first or early encounter with police. Methods: The Transitions Study (N = 802; mean age = 18.3 years; 66% female) baseline and 12‐month follow‐up data were analysed in relation to childhood maltreatment (Childhood Trauma Questionnaire) and changes in mental health (Kessler Psychological Distress Scale and Social and Occupational Functioning Assessment Scale) among young people attending headspace centres in Australia. Outcomes were compared between those young people reporting a prior criminal charge and/or crime victimization and those who did not. Results: Young people who reported a prior criminal charge or crime victimization reported higher frequency and severity of childhood maltreatment, than those who did not. Both groups reported a reduction in psychological distress following youth mental health service access at 12‐month follow‐up, however functional improvement was only evident for young people who did not report a prior criminal charge or crime victimization. Conclusions: Higher rates of reported childhood maltreatment and reduced psychological distress following service access supports trialling a diversion option to a trauma‐informed community‐based primary mental health service for young people following an early encounter with police. Young people who experience crime victimisation report similar barriers to accessing victim-support and mental health services as young people report for mental health help-seeking. Waiting for repeat offending to trigger a psychosocial or mental health response means more crime, and the likely need for more intensive mental health support. Increasing access could contribute to improved outcomes more broadly, including improved youth mental health, reduced exposure to victimisation and the reorientation of potential offending trajectories to divert young people from future justice involvement. Aim: Spatial sampling biases in biodiversity data arise because of complex interactions between geography, species characteristics and human behaviour, including preferences for or against particular species or habitats; biases are therefore not necessarily independent of the environmental niches of species. We evaluate when correlations between spatial sampling biases and environmental niches are likely to affect species distribution models (SDMs) developed both with and without attempts to correct these biases. Innovation: A virtual species and virtual ecologist framework was used to simulate biodiversity data with either no spatial sampling bias or biases that were correlated (positively or negatively) with one of the environmental variables used to define the environmental niches of the species. The environmental variables used to define the species niche were simulated with spatial autocorrelation operating at multiple spatial scales. Virtual samples were then used to model species distributions, with models evaluated based on their ability to rank the suitability of sites correctly. Main conclusions: Correlations between spatial sampling bias and environmental niches frequently reduced the rank correlation of model predictions, but the relative importance of these effects varied with species type (greater decline in rank correlation as the environmental niche broadens) and data type (models built using detection/non‐detection data were less affected than those using detection‐only data). Bias‐correction effectiveness varied depending on the structure of the spatial bias but was also highly variable across methods and dependent on data type. The implications of these results are that spatial sampling bias is a greater concern for SDMs where: (1) the distribution of effort is non‐random with respect to an environmental gradient thought to be correlated with a species' distribution; (2) the species being modelled has a broad environmental niche; and (3) the data for modelling contain only information on detections (i.e., presence only). Although there is an ever-increasing number of disease-modifying treatments for relapsing multiple sclerosis (MS), few appear to influence coronavirus disease 2019 (COVID-19) severity. There is concern about the use of anti-CD20-depleting monoclonal antibodies, due to the apparent increased risk of severe disease following severe acute respiratory syndrome corona virus two (SARS-CoV-2) infection and inhibition of protective anti-COVID-19 vaccine responses. These antibodies are given as maintenance infusions/injections and cause persistent depletion of CD20+ B cells, notably memory B-cell populations that may be instrumental in the control of relapsing MS. However, they also continuously deplete immature and mature/naïve B cells that form the precursors for infection-protective antibody responses, thus blunting vaccine responses. Seroconversion and maintained SARS-CoV-2 neutralizing antibody levels provide protection from COVID-19. However, it is evident that poor seroconversion occurs in the majority of individuals following initial and booster COVID-19 vaccinations, based on standard 6 monthly dosing intervals. Seroconversion may be optimized in the anti-CD20-treated population by vaccinating prior to treatment onset or using extended/delayed interval dosing (3–6 month extension to dosing interval) in those established on therapy, with B-cell monitoring until (1–3%) B-cell repopulation occurs prior to vaccination. Some people will take more than a year to replete and therefore protection may depend on either the vaccine-induced T-cell responses that typically occur or may require prophylactic, or rapid post-infection therapeutic, antibody or small-molecule antiviral treatment to optimize protection against COVID-19. Further studies are warranted to demonstrate the safety and efficacy of such approaches and whether or not immunity wanes prematurely as has been observed in the other populations. We review emerging evidence indicating that differential rates of B-cell subset repopulation following CD20-depleting antibody treatments can be exploited, to maintain control of autoimmunity whilst allowing SARS-CoV-2 vaccination-induced seroconversion. Coastal marine ecosystems are integral in providing numerous ecosystem functions and services. However, intense and chronic anthropogenic stressors often impact marine ecosystems surrounding highly urbanized, densely populated coastal megacities. The associated reduction of water quality and biodiversity of these ecosystems potentially erodes ecosystem functions and provision of services. While there is general consensus that greater biodiversity supports a multitude of ecosystem functions, evidence from marine ecosystems in real-world settings is lacking. In this study, we evaluated the influence of water quality on benthic and fish assemblages, as well as quantified six key ecosystem functions in coastal waters of Hong Kong. Hong Kong's marine ecosystems are marred with long-term damage that is directly linked with rapid economic growth and anthropogenic activities. We found that water quality significantly affected community composition and clustered assemblages into separate groups related to strength of impact: high impact sites were generally dominated by bivalves and small, opportunistic organisms such as damselfish (Pomacentrinae) and cardinalfish (Apogonidae); low impact sites were dominated by macroalgae, bryozoa, and commercially important piscivores such as groupers (Epinephelinae) and seabreams (Sparidae). Ecosystem function tests showed that low impact sites were much more productive than high impact sites, with 15-times greater macroalgal cover and 5-times higher fish biomass. On the other hand, herbivory and predation rates were context-dependent due to underlying biodiversity effects such as selection, complementarity, and competition. Eutrophication increased decomposition and reduced carbon sequestration, but excessive nutrient input could have suppressed decomposer activities via hypoxia. Therefore, we show that in high impact sites where top-down control is removed, resilient and opportunistic organisms such as cardinalfishes and sea urchins may expand to fulfill functional roles, effectively maintaining ecosystem functions such as predation and herbivory in degraded habitats. Contrary to general consensus, this study demonstrates the merit in conserving even highly impacted marine habitats due to their persistent ecosystem functionality despite chronic water quality issues and shifts in benthic and fish assemblages. Design of a steel catenary riser requires the use of connection hardware to decouple the large bending moments induced by the host floater at the hang-off location. Reliability of this connection hardware is essential, particularly in applications involving high pressure and high temperature fluids. One option for this connection hardware is the metallic tapered stress joint. Titanium (Ti) Grade 29 has been identified as an attractive material candidate for demanding stress joint applications due to its "high strength, low weight, superior fatigue performance and innate corrosion resistance".2 Titanium stress joints for deepwater applications are typically not fabricated as a single piece due to titanium ingot volume limitations, thus making an intermediate girth weld necessary to satisfy length requirements. As with steel, the potential effect of hydrogen embrittlement induced by cathodic and galvanic potentials must be assessed to ensure long-term weld integrity. This paper describes testing from a joint industry project (JIP) conducted to qualify titanium stress joint (TSJ) welds for ultra-deepwater applications under harsh service and environmental conditions. Corrosion-fatigue crack growth rate (CFCGR) results for Ti Grade 29 flat welding-groove weld (1G/PA) gas tungsten arc welding (GTAW) specimens in seawater under cathodic potential and sour brine under galvanic potential are presented and compared to vendor recommended design curves. The objective was to summarize data on coronavirus disease 2019 (COVID-19) testing uptake, social determinants of health, and patient satisfaction with mobile health clinic services within underserved minority and low-income communities. This COVID-19 pilot project was conducted during June and July 2020 in low-income residential neighborhoods in West Baltimore, Maryland. Quantitative data were collected and assessed cross-sectionally. Demographically, 85% of the patients identified as Black or African American (n = 265) and 58.2% as female (n = 184). The COVID-19 test was administered by the registered nurse to 78.2% (n = 288) of the patients. More than 90% of patients confirmed high levels of satisfaction with the services they received from the community mobile health clinic. Social determinants were assessed and females reported significantly worse health literacy than their male counterparts (P < 0.05). Study findings suggest that the community mobile health clinic model was effective in attracting hard-to-reach and marginalized individuals, who otherwise may have gone untested or undiagnosed. This care delivery model can be one solution to disparities by improving access to COVID-19 testing and primary care for communities with higher vulnerability to COVID-19 complications. The performance of cover songs in popular music has long been a subject of critical discussion and debate due to the artistic, social, cultural, and commercial issues that covers raise. In non-Western societies, most popular songs covered by artists are Anglo-American, a situation which implicitly privileges Western music and reinforces both the "west and the rest" trope and the cultural imperialism thesis. Taking American amateur artists and their online videos performing Filipino popular music as case studies, this article examines how social media platforms facilitate and problematize center-periphery relations in popular music through a diffusion of cultural products "from the rest to the west." Moreover, we show that more than the promise of audience reach, the phenomenon reflects how these cover artists embody cultural and social situatedness in Filipino culture. Aim: Microclimate information is often crucial for understanding ecological patterns and processes, including under climate change, but is typically absent from ecological and biogeographic studies owing to difficulties in obtaining microclimate data. Recent advances in microclimate modelling, however, suggest that microclimate conditions can now be predicted anywhere at any time using hybrid physically and empirically based models. Here, we test these methods across a sparsely vegetated and topographically diverse sub‐Antarctic island ecosystem (Macquarie Island). Innovation: Microclimate predictions were generated at a height of 4 cm above the surface on a 100 × 100 m elevation grid across the island for the snow‐free season (Oct–Mar), with models driven by either climate reanalysis data (CRA) or CRA data augmented with meteorological observations from the island's automatic weather station (AWS+CRA). These models were compared with predictions from a simple lapse rate model (LR), where an elevational adjustment was applied to hourly temperature measurements from the AWS. Prediction errors tended to be lower for AWS+CRA‐driven models, particularly when compared to the CRA‐driven models. The AWS+CRA and LR models had similar prediction errors averaged across the season for Tmin and Tmean, but prediction errors for Tmax were much smaller for the former. The within‐site correlation between observed and predicted daily Tmean was on average >0.8 in all months for AWS+CRA predictions and >0.7 in all months for LR predictions, but consistently lower for CRA predictions. Main conclusions: Prediction of microclimate conditions at ecologically relevant spatial and temporal scales is now possible using hybrid models, and these often provide added value over lapse rate models, particularly for daily extremes and when driven by in situ meteorological observations. These advances will help add the microclimate dimension to ecological and biogeographic studies and aid delivery of climate change‐resilient conservation planning in climate change‐exposed ecosystems. Fatty acids (FAs) are the main components of lipids in corals. We examined FAs profiles from five symbiotic coral species belonging to five different genera (Acropora, Pavona, Turbinaria, Favites, and Platygyra) and four different families (Acroporidae, Agariciidae, Dendrophyllidae, Faviidae). We separated symbionts from the coral host tissue to investigate the interaction of FA between symbionts and host tissue. After separation, we used FA profiles, in particular specific FAs (e.g. 16:0, 18:0, 18:3n-3, 20:5n-3, 22:6n-3) and their ratios (EPA:DHA, PUFA:SFA) as biomarkers (i.e. signature lipids) to examine chemotaxonomy and trophic level (autotrophy vs. heterotrophy) of each coral species. Gas chromatography–mass spectrometry (GC–MS) was performed to identify and quantify FA. For quantification, the dry weight of total lipids was used to normalize FA concentration (μg mg−1). We found that (1) the five different coral species showed define species-specific FA profiles; (2) certain FAs were valuable biomarkers to determine relative trophic strategies (i.e. autotrophy and/or heterotrophy; (3) the application of FA ratios to define trophic level requires caution in research application and data interpretation. Considering the limitations of FA ratios determined herein, we suggest it to be more appropriate to examine response to environmental change within species. Going forward, our study provides important FA baseline data that builds the foundation for future investigations on the impact of environmental changes related to nutrition and metabolism in symbiotic corals. Multiple sclerosis is the major demyelinating autoimmune disease of the central nervous system. Relapsing MS can be treated by a number of approved monoclonal antibodies that currently target: CD20, CD25 (withdrawn), CD49d and CD52. These all target potentially pathogenic memory B cell subsets and perhaps functionally inhibit pathogenic T cell function. These consist of chimeric, humanized and fully human antibodies. However, despite humanization it is evident that all of these monoclonal antibodies can induce binding and neutralizing antibodies ranging from < 1% to over 80% within a year of treatment. Importantly, it is evident that monitoring these allow prediction of future treatment-failure in some individuals and treatment cessation and switching therefore potentially limiting disease breakthrough and disability accumulation. In response to the COVID-19 pandemic and the need to avoid hospitals, shortened infusion times and extended dose intervals have been implemented, importantly, subcutaneous delivery of alternative treatments or formulations have been developed to allow for home treatment. Asthma is a major global disease affecting adults and children, which can lead to hospitalization and death due to breathing difficulties. Although targeted monoclonal antibody therapies have revolutionized treatment of severe asthma, some patients still fail to respond. Here we critically evaluate the literature on biologic therapy failure in asthma patients with particular reference to anti-drug antibody production, and subsequent loss of response, as the potential primary cause of drug failure in asthma patients. Encouragingly, asthma in most cases responds to treatment, including the use of an increasing number of biologic drugs in moderate to severe disease. This includes monoclonal antibody inhibitors of immunoglobulin E and cytokines, including interleukin 4, 5, or 13 and thymic stromal lymphopoietin. These limit mast cell and eosinophil activity that cause the symptomatic small airways obstruction and exacerbations. Despite humanization of the antibodies, it is evident that benralizumab; dupilumab; mepolizumab; omalizumab; reslizumab and tezepelumab all induce anti-drug antibodies to some extent. These can contribute to adverse events including infusion reactions, serum sickness, anaphylaxis and potentially disease activity due to loss of therapeutic function. Monitoring anti-drug antibodies (ADA) may allow prediction of future treatment-failure in some individuals allowing treatment cessation and switching therefore potentially limiting disease breakthrough. A Variant of Uncertain Significance (VUS) is a difference in the DNA sequence with uncertain consequences for gene function. A VUS in a hereditary cancer gene should not change medical care, yet some patients undergo medical procedures based on their VUS result, highlighting the unmet educational needs among patients and healthcare providers. To address this need, we developed, evaluated, and refined novel educational materials to explain that while VUS results do not change medical care, it remains important to share any personal or family history of cancer with family members given that their personal and family medical history can guide their cancer risk management. We began by reviewing the prior literature and transcripts from interviews with six individuals with a VUS result to identify content and design considerations to incorporate into educational materials. We then gathered feedback to improve materials via a focus group of multidisciplinary experts and multiple rounds of semi-structured interviews with individuals with a VUS result. Themes for how to improve content, visuals, and usefulness were used to refine the materials. In the final round of interviews with an additional 10 individuals with a VUS result, materials were described as relatable, useful, factual, and easy to navigate, and also increased their understanding of cancer gene VUS results. This article examines the experiences of family members when a loved one dies after police contact in the United States. It uses qualitative data from semistructured interviews with the bereaved families of 43 U.S. citizens who died after police contact and considers their experiences as covictims of homicide. It examines how they experience grief in the aftermath of such a death and considers Doka's concept of disenfranchised grief in evaluating how social norms affect their grieving process. It argues that individuals affected by deaths after police contact are often unable to grieve in a way that is socially legitimized. The article finds that disenfranchised grief has a racial dimension with regard to deaths after police contact with non-White families being deeply affected by it due to their position within society, the context in which their loved one died, and in terms of how the deceased was socially constructed. Dracula (Browning, 1931) introduces the general parameters of the classical screen vampire narrative of the 1930s and 1940s; a drawing-room murder mystery. In this investigative 'drama of proof', the murderer, already known to the audience, is a vampire who has inserted himself between a young engaged couple, seeking the woman for himself. The classical vampire protagonist is an unreflecting figure driven by a craving for both the young woman of his desire and for human blood as sustenance. The inevitable death of the vampire represents a triumph of 'life', marriage understood as the basic unit by which community reproduces itself, playing out a Freudian (and romantic) opposition between the erotic instinct and the aggressive instinct or death drive. The classical screen vampire narrative does not remain static. In aqueous solution, polar groups make hydrogen bonds with water, and hence burial of such groups in the interior of a protein is unfavorable unless the loss of hydrogen bonds with water is compensated by formation of new ones with other protein groups. For this reason, buried "unsatisfied" polar groups making no hydrogen bonds are very rare in proteins. Efficiently representing the energetic cost of unsatisfied hydrogen bonds with a pairwise-decomposable energy term during protein design is challenging since whether or not a group is satisfied depends on all of its neighbors. Here we describe a method for assigning a pairwise-decomposable energy to sidechain rotamers such that following combinatorial sidechain packing, buried unsaturated polar atoms are penalized. The penalty can be any quadratic function of the number of unsatisfied polar groups, and can be computed very rapidly. We show that inclusion of this term in Rosetta sidechain packing calculations substantially reduces the number of buried unsatisfied polar groups. Author summary: We present an algorithm that fits into existing protein design software that allows researchers to penalize unsatisfied polar atoms in protein structures during design. These polar atoms usually make hydrogen-bonds to other polar atoms or water molecules and the absence of such interactions leaves them unsatisfied energetically. Penalizing this condition is tricky because protein design software only looks at pairs of amino acids when considering which amino acids to choose. Current approaches to solve this problem use additive approaches where satisfaction or unsatisfaction is approximated on a continuous scale; however, in reality, satisfaction or unsatisfaction is an all-or-none condition. The simplest all-or-none method is to penalize polar atoms for simply existing and then to give a bonus any time they are satisfied. This fails when two different amino acids satisfy the same atom; the pairwise nature of the protein design software will double count the satisfaction bonus. Here we show that by anticipating the situation where two amino acids satisfy the same polar atom, we can apply a penalty to the two amino acids in advance and assume the polar atom will be there.
