Ovarian cancer (OvCa) is a leading cause of mortality among gynecological malignancies and usually manifests as intraperitoneal spheroids that generate metastases, ascites, and an immunosuppressive tumor microenvironment. In this study, we explore the immunomodulatory properties of cowpea mosaic virus (CPMV) as an adjuvant immunotherapeutic agent using an in vitro model of OvCa peritoneal spheroids. Previous findings highlighted the potent efficacy of intratumoral CPMV against OvCa in mouse tumor models. Leveraging the precision control over material deposition and cell patterning afforded by digital-light-processing (DLP) based bioprinting, we constructed OvCa-macrophage spheroids to mimic peritoneal spheroids using gelatin methacrylate (GelMA), a collagen-derived photopolymerizable biomaterial to mimic the extracellular matrix. Following CPMV treatment, bioprinted spheroids exhibited inhibited OvCa progression mediated by macrophage activation. Our analysis indicates that CPMV regulates and activates macrophage to both induce OvCa cell killing and restore normal cell-cell junctions. This study deepened our understanding of the mechanism of CPMV intratumoral immunotherapy in the setting of OvCa. This study also highlights the potential of studying immunotherapies using high throughput tissue models via DLP bioprinting.

INTRODUCTION: Despite numerous investigations into the impact of drugs/probiotics on the gut microbiota composition in Familial Mediterranean Fever (FMF) patients, the question as to whether there exists a significant bacterial diversity(ies) independent of the placebo effect that can be reliably considered in clinical and nutritional trials remains unresolved. METHODS: This study represents the in augural analysis of the placebos influence on the gut microbiota of both healthy individuals and FMF afflicted men, utilizing previously collected data from PhyloChip™ DNA microarray experiments. A total of 15 healthy and 15 FMF male volunteers, aged 18 to 50, participated in this partially randomized placebo trial, which is accessible through the GEO Series accession number GSE111835. RESULTS AND DISCUSSION: Key findings from current investigations include i. the anticipated divergence in gut bacteria resistance to placebo between healthy and FMF individuals, ii. the minor impact of placebo on gut bacterial diversities in healthy individuals, with Enterobacteriaceae diversities identified as placebo-resistant among healthy gut bacteria, and iii. the comprehensive influence of placebo on all bacterial phyla in the gut microbiome of FMF patients, extending to nearly all bacterial genera, except for the resilience of gut Akkermansia muciniphila spp. to placebo in FMF patients. This study underscores the susceptibility of Faecalibacterium, Blautia, and Clostridium genera to placebo. Consequently, this investigation holds significance for the proper design of placebo-controlled trials and establishes a foundation for further exploration of the gut-brain axis. Furthermore, it contributes valuable insights to discussions regarding proposals for probiotic therapies, particularly focusing on Faecalibacterium spp., Blautia spp., and Clostridium spp.

Neonatal septicaemia is an intricate systemic affliction induced in newborns. A multitude of fungi, viruses, and bacteria elicit this condition. Conventional methodologies such as radiological imaging, microbial testing, blood culture, and inflammatory markers suffer from an array of constraints for prompt detection of infection. This present study illustrates a cutting-edge smart electrochemical biosensing system. The fabricated system was meticulously engineered to discern the concentration of C-reactive protein (CRP). CRP is an irreplaceable prognostic indicator for neonatal sepsis. This work explicates the fabrication of a bioreceptor sensing platform (BioRe Sense) on a screen-printed miniaturized electrode. The surface of the electrode underwent modification with molybdenum disulfide nanosheets (MoS2NSs). Further, the interface was designed with molecularly imprinted polymer (MIP) targeting CRP. An electrochemical technique carefully examined the electrode at each modification stage. The created sensing interface can detect a wide concentration range from 0.01 nM to 100 nM, with a lower detection limit of 0.01 nM.

BackgroundIt is known that the gut microbiome of a healthy person affects the process of COVID-19 after getting infected with SARS-CoV-2 virus. It is also believed that colchicine can alleviate the severity of COVID-19.ObjectiveCurrent investigations aimed to evaluate the associations between the baseline gut microbiota composition of healthy and Familial Mediterranean fever (FMF) - carrier Armenian men populations, and the severity of the COVID-19 disease after their infection with the SARS-CoV-2. The study has a purpose of answering three core questions: i. Do the characteristics of gut microbiome of Armenians affect the course of COVID-19 severity? ii. How does the COVID-19 disease course on go for FMF patients who have been taking colchicine as a medication over the years after getting infected with SARS-CoV-2? iii. Is there an initial gut micribiota structure pattern for non-FMF and FMF patients in the cases when COVID-19 appears in mild form?MethodsThe gut microbiota composition in non-FMF and FMF patients before the first infection (mild and moderate course of COVID-19) was considered. COVID-19 was diagnosed by SARS-CoV-2 nucleic acid RT-PCR in nasopharyngeal swab and/or sputum.ResultsThe number of patients with male FMF with mild COVID-19 was approximately two times higher than that of non-FMF male subjects with COVID-19. In addition, an association of COVID-19 disease severity with the baseline gut Prevotella, Clostridium hiranonis, Eubacterium biforme, Veillonellaceae, Coprococcus, and Blautia diversities in the non-FMF and FMF populations were revealed by us, which can be used as risk/prognostic factor for the severity of COVID-19.

To study the diversity of agromizids (leaf miners) in the South Caucasus, the species external morphological features, their host plants, the development of larvae and pupae and to assess the degree of crop infestation by leaf‐miners.The study was carried out within the framework of the scs 21‐002 scientific program “Leaf miners as pests of agricultural plants (Diptera: Agromyzidae): the latest control measures”. Type samples were fixed in 75 % ethyl alcohol and deposited in the Agrarian Scientific Research Laboratory of the Scientific Centre of Zoology and Hydroecology. Species were identified by examining the male genitalia. The degree of damage of the host–plant was determined based on intensity indicators.As a result of this research, a new species of the Calycomyza genus was discovered in the South Caucasus fauna. Larvae of C. jucunda (Wulp, 1867) were found on the leaves of Beta vulgaris, a host plant mentioned for the first time here. As a Nearctic species, it was also first discovered in the southern Caucasus.The study of the male genitalia indicates expansion of the biogeographical distribution of Calycomyza species, as most of them are found in the Nearctic and Neotropical regions. In terms of intensity, C. jucunda is classified as a less dangerous species for agricultural crops.

Blended hybrid mathematics instruction offers the flexibility of online learning and the engagement of face-to-face interaction. However, research on student participation in these learning environments needs to be improved. This study investigated the dynamics of student participation, focusing on four key dimensions: conceptual understanding, mathematical communication, problem-solving, and critical thinking and analysis. A survey of 158 BS Mathematics students revealed that online learners tended to exhibit a higher level of conceptual understanding. At the same time, face-to-face interactions enhance problem-solving skills and stimulate more remarkable development of critical thinking and analytical abilities. Mathematical communication skills were somewhat constrained in both settings, with face-to-face students displaying slightly higher participation. These findings underscore the importance of considering the diverse dimensions of student participation when delivering online and hybrid mathematics courses. Educators should provide opportunities for students to hone and cultivate essential skills across online and face-to-face contexts, thus enriching their learning experience.

Genus Cornus L. (Cornaceae) includes, according to various sources, from 4 to about 60 species, growing mainly in temperate regions of the northern hemisphere. Cornus mas L. is the most widely used species in the genus, from which numerous semi-cultivated and cultivated forms and sorts were obtained. The wild C. mas is of Western Asian origin, distributed in the mountains of the Caucasus, Asia Minor, and Southern Europe, and the genotype of the species shows reliable polymorphism. The purpose of this study was to present a detailed palynomorphological characteristic of the species C. mas of the Armenian flora using light microscopy (LM) and scanning electron microscopy (SEM), to clarify the disagreements noted in the literature regarding to the pollen aperture type for the species C. mas, as well as to conduct a comparative palynomorphological analysis of the genus Cornus as a whole (based on the literary data). The data obtained are consistent with the opinion of a number of authors about the presence in the species C. mas of the quite rare compound pollen aperture type. This compound aperture type is formed by a combination of the colpus with a peculiar endoaperture complex, consisting of a small oblong os and paracolpi, located on both sides of the colpi. The os small, elliptical, reaching the paracolpi, forming together butterfly-like or H-shape endexine thinnings. In our opinion, both the os and the paracolpi arose due to the thinning or absence of the internal layers of the exine. Exine 1.3–1.5 μm, columellae separate, thick, capitate. Exine ornamentation verrucate (LM); exine ornamentation densely verrucate with rare perforations, verrucae varying in size (SEM). An extensive analysis of the literature data on pollen of 42 species of the genus Cornus revealed the significant differences in the characteristics of the type of apertures not only at the interspecific, but also at the intraspecific level. In terms of size, pollen grains are predominantly large or of medium size, rarely small. The large pollen grains were noted for C. alba L., C. alsophila W. W. Sm., C. alternifolia L.f., C. amomum Mill., C. asperifolia Michx., C. controversa Hemsl., C. coreana Wangerin, C. drummondii C. A. Mey., C. excelsa Kunth, C. glabrata Benth., C. hemsleyi C. K. Schneid. & Wangerin, C. monbeigii Hemsl., C. paucinervis Hance, C. poliophylla C. K. Schneid. & Wangerin, C. rugosa Lam., C. sanguinea L., C. sericea L., C. stolonifera Michx., C. walteri Wangerin, C. wilsoniana Wangerin. Medium pollen size is typical for C. boliviana J. F. Macbr., C. brachypoda C. A. Mey., C. canadensis L., C. capitata Wall., C. chinensis Wangerin, C. controversa, C. disciflora Moc. & Sessé ex DC., C. floccosa Wangerin, C. florida Hook., C. foemina Mill., C. hongkongensis Hemsl., C. kousa Buerger, C. longipetiolata Hay., C. macrophylla Wall., C. mas L., C. nuttallii Audubon, C. oblonga Wall., C. peruviana J. The largest pollen grains were noted for the species C. monbergii (P x E = 81.0 x 48.5 μm) and the smallest ones for C. officinalis (P x E = 20.0 x 21.0 μm). Exine ornamentation as a rule is represented by a number of primitive types, namely, scabrate, rugulate, verrucate, granulate, echinate, etc., sometimes with perforations.

Pterygium is an ocular surface disorder with high prevalence that can lead to vision impairment. As a pathological outgrowth of conjunctiva, pterygium involves neovascularization and chronic inflammation. Here, we developed a 3D multicellular in vitro pterygium model using a digital light processing (DLP)-based 3D bioprinting platform with human conjunctival stem cells (hCjSCs). A novel feeder-free culture system was adopted and efficiently expanded the primary hCjSCs with homogeneity, stemness and differentiation potency. The DLP-based 3D bioprinting method was able to fabricate hydrogel scaffolds that support the viability and biological integrity of the encapsulated hCjSCs. The bioprinted 3D pterygium model consisted of hCjSCs, immune cells, and vascular cells to recapitulate the disease microenvironment. Transcriptomic analysis using RNA sequencing (RNA-seq) identified a distinct profile correlated to inflammation response, angiogenesis, and epithelial mesenchymal transition in the bioprinted 3D pterygium model. In addition, the pterygium signatures and disease relevance of the bioprinted model were validated with the public RNA-seq data from patient-derived pterygium tissues. By integrating the stem cell technology with 3D bioprinting, this is the first reported 3D in vitro disease model for pterygium that can be utilized for future studies towards personalized medicine and drug screening.

Limbal stem cell deficiency and corneal disorders are among the top global threats for human vision. Emerging therapies that integrate stem cell transplantation with engineered hydrogel scaffolds for biological and mechanical support are becoming a rising trend in the field. However, methods for high-throughput fabrication of hydrogel scaffolds, as well as knowledge of the interaction between limbal stem/progenitor cells (LSCs) and the surrounding extracellular matrix (ECM) are still much needed. Here, we employed digital light processing (DLP)-based bioprinting to fabricate hydrogel scaffolds encapsulating primary LSCs and studied the ECM-dependent LSC phenotypes. The DLP-based bioprinting with gelatin methacrylate (GelMA) or hyaluronic acid glycidyl methacrylate (HAGM) generated microscale hydrogel scaffolds that could support the viability of the encapsulated primary rabbit LSCs (rbLSCs) in culture. Immunocytochemistry and transcriptional analysis showed that the encapsulated rbLSCs remained active in GelMA-based scaffolds while exhibited quiescence in the HAGM-based scaffolds. The primary human LSCs encapsulated within bioprinted scaffolds showed consistent ECM-dependent active/quiescent statuses. Based on these results, we have developed a novel bioprinted dual ECM Yin-Yang model encapsulating LSCs to support both active and quiescent statues. Our findings provide valuable insights towards stem cell therapies and regenerative medicine for corneal reconstruction.

Ocular surface diseases including conjunctival disorders are multifactorial progressive conditions that can severely affect vision and quality of life. In recent years, stem cell therapies based on conjunctival stem cells (CjSCs) have become a potential solution for treating ocular surface diseases. However, neither an efficient culture of CjSCs nor the development of a minimally invasive ocular surface CjSC transplantation therapy has been reported. Here, we developed a robust in vitro expansion method for primary rabbit-derived CjSCs and applied digital light processing (DLP)-based bioprinting to produce CjSC-loaded hydrogel micro-constructs for injectable delivery. Expansion medium containing small molecule cocktail generated fast dividing and highly homogenous CjSCs for more than 10 passages in feeder-free culture. Bioprinted hydrogel micro-constructs with tunable mechanical properties enabled the 3D culture of CjSCs while supporting viability, stem cell phenotype, and differentiation potency into conjunctival goblet cells. These hydrogel micro-constructs were well-suited for scalable dynamic suspension culture of CjSCs and were successfully delivered to the bulbar conjunctival epithelium via minimally invasive subconjunctival injection. This work integrates novel cell culture strategies with bioprinting to develop a clinically relevant injectable-delivery approach for CjSCs towards the stem cell therapies for the treatment of ocular surface diseases.

Progestins are widely used for the treatment of gynecologic disorders and alone, or combined with an estrogen, are used as contraceptives. While their potencies, efficacies and side effects vary due to differences in structures, doses and routes of administration, little is known about their effects on the endometrial transcriptome in the presence or absence of estrogen. Herein, we assessed the transcriptome and pathways induced by progesterone (P4) and the three most commonly used synthetic progestins, medroxyprogesterone acetate (MPA), levonorgestrel (LNG), and norethindrone acetate (NETA), on human endometrial stromal fibroblasts (eSF), key players in endometrial physiology and reproductive success. While there were similar transcriptional responses, each progestin induced unique genes and biofunctions, consistent with their structural similarities to progesterone (P4 and MPA) or testosterone (LNG and NETA), involving cellular proliferation, migration and invasion. Addition of estradiol (E2) to each progestin influenced the number of differentially expressed genes and biofunctions in P4 and MPA, while LNG and NETA signatures were more independent of E2. Together, these data suggest different mechanisms of action for different progestins, with progestin-specific altered signatures when combined with E2. Further investigation is warranted for a personalized approach in different gynecologic disorders, for contraception, and minimizing side effects associated with their use.

Enrollment management (EM) has been a focus of higher education since the 1970s. There is a large base of empirical research on EM, a coordinated effort to support undergraduate students from admission to graduation that has been widely researched. However, there is limited academic research on graduate enrollment management (GEM). What is missing is grounding GEM in literature and viewing the graduate student lifecycle with a cohesive lens. This article provides an academic foundation for GEM while discussing future research areas and encouraging GEM professionals to adopt a scholar-practitioner lens. The authors lay the foundation for the GEM concept, synthesize related literature, and discuss future research directions.

This research addresses the imperative challenge of a lightweight design for an Unmanned Aerial Vehicle (UAV) chassis to enhance the thrust-to-weight and power-to-weight ratios, crucial for optimal flight performance, focused on developing an intriguing lightweight yet robust quadcopter chassis. Advanced generative design techniques, integrated with topology optimization, using Autodesk Fusion 360 software (v. 16.5. 0.2083), 3D-printing methods and lightweight materials like Polylactic Acid (P.L.A.), Acrylonitrile Butadiene Styrene (A.B.S.), and Nylon 6/6 play a significant role in achieving the desired balance between structural integrity and weight reduction. The study showcases successful outcomes, presenting quadcopter chassis designs that significantly improve structural efficiency and overall performance metrics. The findings contribute to aerial robotics and hold promise for precision agriculture applications with relevant performed simulations, emphasizing the importance of tailored design methodologies for other engineering domains. In conclusion, this research provides a foundational step toward advancing drone technology, with weight reductions of almost 50%, P/W and T/W ratios increment of 6.08% and 6.75%, respectively, at least an 11.8% increment in Factor of Safety, at least a 70% reduction in stress values and reduced manufacturing time from its comparative DJI F450 drone, demonstrating the critical role of innovative design approaches in optimizing operational efficiency for targeted applications.

PurposeMesenchymal stem cells (MSCs) have been extensively studied for their capacity to enhance wound healing and represent a promising research field for generating cell therapies for corneal scars. In the present study, we investigated MSCs from different tissues and their potential to differentiate toward corneal keratocytes.MethodsAdipose-derived stem cells, bone marrow MSCs, umbilical cord stem cells, and corneal stromal stem cells (CSSCs) were characterized by their expression of surface markers CD105, CD90, and CD73, and their multilineage differentiation capacity into adipocytes, osteoblasts, and chondrocytes. MSCs were also evaluated for their potential to differentiate toward keratocytes, and for upregulation of the anti-inflammatory protein TNFα-stimulated gene-6 (TNFAIP6) after simulation by IFN-γ and TNF-α.ResultsKeratocyte lineage induction was achieved in all MSCs as indicated by the upregulated expression of keratocyte markers, including keratocan, lumican, and carbohydrate sulfotransferase. TNFAIP6 response to inflammatory stimulation was observed only in CSSCs; increasing by 3-fold compared with the control (P < 0.05).ConclusionsBased on our findings, CSSCs appeared to have the greatest differentiation potential toward the keratocyte lineage and the greatest anti-inflammatory properties in vitro.

Postpneumonectomy syndrome is a rare complication of a pneumonectomy. Patients may experience dyspnea, stridor, recurrent pulmonary infections, or dysphagia due to rotation and shift of the mediastinum. The current intervention of choice involves the placement of a tissue expander in the empty hemithorax to realign the mediastinum. Because this treatment can present with intraoperative anesthetic challenges and requires close monitoring, we present this case to highlight specific concerns that may need to be addressed including difficulties ventilating, complete airway collapse, hemodynamic instability, and pain control perioperatively.

PURPOSE:The study aims to test the hypothesis that platelet-rich plasma (PRP) stimulates cellular processes involved in endometrial regeneration relevant to clinical management of poor endometrial growth or intrauterine scarring. Based on the concept of «surveillance capitalism» and research on the digital transformation of public administration, the authors analyzes the structure and logic of functioning of the «smart city» model using the example of Moscow. Based on the material of street protests, the political effects of the use of digital infrastructure by the city authorities, in particular, camera systems with face recognition technologies, are examined. The study of the Russian situation correlates with the latest decisions of the United Nations (UN) Human Rights Council and the European Union’s initiatives to control remote biometric recognition technologies.

Intestinal microorganisms play a crucial role in health and disease. The disruption of host-microbiota homeostasis has been reported to occur not only during disease development but also as a result of medication. Familial Mediterranean fever (FMF) is an inflammatory genetic disease characterized by elevated systemic reactivity against the commensal gut microbiota and high levels of Candida albicans in the gut. This study's major objective was to investigate the effects of commercial probiotic Narine on the relative abundance of gut bacteria (specifically, enterobacteria, lactobacilli, Staphylococcus aureus, and enterococci) of C. albicans carrier and non-carrier FMF patients in remission. Our main finding indicates that the probiotic reduces numbers of C. albicans and abundance of enterobacteria in male and female patients of C. albicans carriers and non-carriers. It has pivotal effect on Enterococcus faecalis: increase in male non-carriers and decrease in female ones regardless of C. albicans status. No effect was seen for Lactobacillus and S. aureus. Our data suggest that M694V/V726A pyrin inflammasome mutations leading to FMF disease may contribute to gender-specific differences in microbial community structure in FMF patients. The study's secondary objective was to elucidate the gender-specific differences in the gut's microbial community of FMF patients. The tendency was detected for higher counts of enterobacteria in female FMF subjects. However, the small number of patients of these groups preclude from conclusive statements, pointing at the need for additional investigations with appropriate for statistical analysis groups of subjects involved in the study.
