Phytoremediation represents a potentially effective and environmentally friendly technology to remediate arsenic (As) in mine waste soils. However, soil amendments are often required to improve phytoremediation due to depleted nutrients in mine waste. This study aims to assess the effect of biosolids biochar, applied at different rates (0%, 5%, and 10%) on As phytoremediation using three plant species: Poa labillardieri, Rytidosperma caespitosum, and Juncus pauciflorus. This study was conducted in a replicated greenhouse pot study using soil from an abandoned mine site. Dry plant biomass, As concentration in plants and soil, and soil microbial abundance were investigated. Juncus pauciflorus produced eight times more root and shoot biomass than R. caespitosum in soils amended with 10% biochar. The highest As uptake was also observed in J. pauciflorus grown in soils amended with 10% biochar (7.10 mg/plant), while R. caespitosum had the lowest As uptake in soils without biochar (0.16 mg/plant). In soils amended with 10% biochar, the total bacterial community decreased to approximately 8.50 log10 copies/g, compared to the initial soil (9.05 log10 copies/g), while the number of gene copies of the nifH gene increased, suggesting the importance of nitrogen-fixing bacteria to promote plant growth. Taguchi analysis confirmed that plant species was the key factor for As phytoremediation, followed by biochar application dose. This study showed that J. pauciflorus and the addition of 10% biochar was the best treatment for remediating As-contaminated mine waste, offering the potential for use commercially. Moreover, the utilisation of biochar derived from biosolids as a soil amendment for enhancing phytoremediation represents good circular economy practice to manage excessive biosolids production. Human-driven habitat loss is recognized as the greatest cause of the biodiversity crisis, yet to date we lack robust, spatially explicit metrics quantifying the impacts of anthropogenic changes in habitat extent on species' extinctions. Existing metrics either fail to consider species identity or focus solely on recent habitat losses. The persistence score approach developed by Durán et al. (Durán et al. 2020 Methods Ecol. Evol. 11, 910–921 (doi:10.1111/2041-210X.13427) represented an important development by combining species' ecologies and land-cover data while considering the cumulative and non-linear impact of past habitat loss on species' probability of extinction. However, it is computationally demanding, limiting its global use and application. Here we couple the persistence score approach with high-performance computing to generate global maps of what we term the LIFE (Land-cover change Impacts on Future Extinctions) metric for 30 875 species of terrestrial vertebrates at 1 arc-min resolution (3.4 km2 at the equator). These maps provide quantitative estimates, for the first time, of the marginal changes in the expected number of extinctions (both increases and decreases) caused by converting remaining natural vegetation to agriculture, and restoring farmland to natural habitat. We demonstrate statistically that this approach integrates information on species richness, endemism and past habitat loss. Our resulting maps can be used at scales from 0.5–1000 km2 and offer unprecedented opportunities to estimate the impact on extinctions of diverse actions that affect change in land cover, from individual dietary choices through to global protected area development. Food production does more damage to wild species than any other sector of human activity, yet how best to limit its growing impact is greatly contested. Reviewing progress to date in interventions that encourage less damaging diets or cut food loss and waste, we conclude that both are essential but far from sufficient. In terms of production, field studies from five continents quantifying the population-level impacts of land sharing, land sparing, intermediate and mixed approaches for almost 2000 individually assessed species show that implementing high-yield farming to spare natural habitats consistently outperforms land sharing, particularly for species of highest conservation concern. Sparing also offers considerable potential for mitigating climate change. Delivering land sparing nevertheless raises several important challenges—in particular, identifying and promoting higher yielding farm systems that are less environmentally harmful than current industrial agriculture, and devising mechanisms to limit rebound effects and instead tie yield gains to habitat conservation. Progress will depend on conservationists forging novel collaborations with the agriculture sector. While this may be challenging, we suggest that without it there is no realistic prospect of slowing biodiversity loss. Chlorinated and fluorinated nitrophenols (HNCs) are widely used in agriculture and industry, with a global market valued at USD $25 billion, one which is expected to grow by 5% by 2030. However, these compounds pose significant environmental risks; they are classified as toxic by the International Agency for Research on Cancer (IARC). Existing treatment methods include advanced oxidation, adsorption, and bioremediation, though to date, there has been only limited research on fungal remediation of these halogenated pollutants. This study aims to explore a sustainable approach by using fungi's potential to degrade HNCs in minimal media. Ten fungi were selected through literature screening; Caldariomyces fumago and Curvularia sp. were highly effective, degrading over 50% of 2-chloro-4-nitrophenol (2C4NP) and 80% of 5-fluoro-2-nitrophenol (5F2NP) within 24 and 48 h, respectively. Additionally, five strains showed degradation potential for fluorinated compounds. Further studies revealed C. fumago could degrade up to 1 mM of chlorinated compounds and 12 mM of fluorinated compounds, far exceeding any known environmental concentrations of HNCs; importantly, ecotoxicology tests demonstrated reductions in toxicity of 77% and 85%, respectively. This work highlights fungi's underexplored ability to degrade toxic HNCs, offering a sustainable mycoremediation strategy and positioning mycology as a critical tool for future environmental remediation efforts. Aim: To evaluate the implementation of a mixed virtual and in‐person brief intervention for young people, aged 12–25 years, presenting to a large urban mental health service in crisis with suicidal ideation and/or self‐harm. Methods: A pragmatic, real‐world evaluation was conducted on the Youth Brief Intervention Service between June 2021 (inception) and October 2022. Service users were offered four sessions over an approximate one‐month period. Sessions focused on distress tolerance, safety plans and support systems. Implementation outcomes related to service uptake, retention, fidelity of the model and service user experience. Effectiveness outcomes were measured pre‐post and included mental health‐related hospital service utilization (primary outcome), functioning, mental health status, self‐harm, suicidal ideation and quality of life. Results: Of the 136 young people referred to the Youth Brief Intervention Service, 99 were accepted with 17 disengaging before the first session. Eighty percent of people who commenced, completed the package of care. Young persons' and parent/carers experience of service was high (97% and 88%, respectively). Mental health‐related emergency department presentations and inpatient days decreased from 3 months pre‐intake to 3 months post‐intake (42 vs. 7 presentations, X2 = 25.3, p <.001; 11 vs. 0 inpatient days, X2 = 9.1, p =.01). There were significant improvements in mental health status, days engaging in self‐harm, general health and functioning and quality of life. Conclusions: The Youth Brief Intervention Service is feasible, acceptable, subjectively beneficial and coincided with less mental health‐related emergency department presentations and inpatient days, and improved mental health status and behaviour. Purpose To determine if high lung shunt fraction (LSF) is an independent prognostic indicator of poor survival in patients who undergo yttrium 90 radioembolization for unresectable liver-dominant metastatic colorectal cancer. Materials and Methods Retrospective data were analyzed from 606 patients (62% men; mean age, 62 years) who underwent radioembolization to treat liver metastases from colorectal adenocarcinoma between July 2002 and December 2011 at 11 U.S. centers. Institutional review board exemptions were granted prior to the collection of data at each site. Overall survival was estimated by using Kaplan-Meier survival and univariate Cox proportional hazards models to examine the effect of LSF on survival and to compare this to other potential prognostic indicators. Multivariate analysis was also performed to determine whether LSF is an independent risk factor for poor survival. Results LSF higher than 10% was predictive of significantly decreased survival (median, 6.9 months vs 10.0 months; hazard ratio, 1.60; P < .001) and demonstrated a mild but significant correlation to serum carcinoembryonic antigen levels and tumor-to-liver volume ratio (Pearson correlation coefficients, 0.105 and 0.113, respectively; P < .05). A progressive decrease in survival was observed as LSF increased from less than 5% to more than 20% (P < .05). LSF did not correlate with the presence of extrahepatic metastases or prior administration of bevacizumab. Conclusion Increased LSF is an independent prognostic indicator of worse survival in patients undergoing radioembolization for liver-dominant metastatic colorectal adenocarcinoma. High LSF correlates poorly to other potential markers of tumor size, such as tumor-to-liver volume ratio or serum carcinoembryonic antigen level, and does not correlate to the presence of extrahepatic metastases. © RSNA, 2016 Online supplemental material is available for this article.

Reverse-transcription-loop-mediated isothermal amplification (RT-LAMP) has frequently been proposed as an enabling technology for simplified diagnostic tests for RNA viruses. However, common detection techniques used for LAMP and RT-LAMP have drawbacks, including poor discrimination capability, inability to multiplex targets, high rates of false positives, and (in some cases) the requirement of opening reaction tubes postamplification. Here, we present a simple technique that allows closed-tube, target-specific detection, based on inclusion of a dye-labeled primer that is incorporated into a target-specific amplicon if the target is present. A short, complementary quencher hybridizes to unincorporated primer upon cooling down at the end of the reaction, thereby quenching fluorescence of any unincorporated primer. Our technique, which we term QUASR (for quenching of unincorporated amplification signal reporters, read "quasar"), does not significantly reduce the amplification efficiency or sensitivity of RT-LAMP. Equipped with a simple LED excitation source and a colored plastic gel filter, the naked eye or a camera can easily discriminate between positive and negative QUASR reactions, which produce a difference in signal of approximately 10:1 without background subtraction. We demonstrate that QUASR detection is compatible with complex sample matrices such as human blood, using a novel LAMP primer set for bacteriophage MS2 (a model RNA virus particle). Furthermore, we demonstrate single-tube duplex detection of West Nile virus (WNV) and chikungunya virus (CHIKV) RNA.

Topics include the challenges of mitigating leakage, the impact of market-driven leakage on biodiversity, and the need for better incorporation of leakage considerations into global conservation strategies.
