MI/RI may be alleviated by protecting cardiomyocytes from oxidative stress. Lutein, which belongs to a class of carotenoids, has proven to be effective in cardiovascular disease treatment due to its remarkable antioxidant properties, but its application is limited due to its poor stability and low bioavailability in vivo.Methods: In this study, a delivery system was developed based on distearoyl phosphatidyl ethanolamine (DSPE)-thiol-ketone (TK)-PEG2K (polyethylene glycol 2000) (abbreviated as DTP) and PCM-SH (CWLSEAGPVVTVRALRGTGSW) to deliver lutein (abbreviated as lutein@DTPP) to damaged myocardium. First, lutein, lutein@DTP, or lutein@DTPP were injected through the tail vein once a day for 3 days and then MI/RI model rats were established by exposing rats to ischemia for 45 min and reperfusion for 6 h. We employed a range of experimental techniques including qRT-PCR, Western blotting, transmission electron microscopy, immunohistochemistry, immunofluorescence, flow cytometry, immunoprecipitation, molecular docking, and molecular dynamics simulations.Results: Lutein@DTPP exhibited good myocardial targeting and ROS-responsive release. Our data suggested that lutein@DTPP effectively suppresses ferroptosis in cardiomyocytes. Mechanistically, we observed an upregulation of mouse double minute-2 (MDM2) in the hearts of MI/RI models and cardiomyocytes exposed to hypoxia/reoxygenation (H/R) conditions. In addition, NADH-ubiquinone oxidoreductase 75 kDa Fe-S protein 1 (NDUFS1) translocation from the cytosol to the mitochondria was inhibited by MDM2 upregulation. Notably, no significant variation in the total NDUFS1 expression was observed in H/R-exposed cardiomyocytes following treatment with siMDM2. Further study indicated that lutein facilitates the translocation of NDUFS1 from the cytosol to mitochondria by directly binding and sequestering MDM2, thereby improving mitochondrial function and inhibiting ferroptosis.Conclusion: Lutein@DTPP promoted the mitochondrial translocation of NDUFS1 to restore mitochondrial function and inhibited the ferroptosis of cardiomyocytes by directly binding and sequestering MDM2. Chemically ordered face-centered cubic (fcc) phases consisting mainly of Cr, Co, and Ni were synthesized by annealing a Cr26.7Co26.7Ni26.7Si10B5P5 amorphous alloy. The annealed Cr26.7Co26.7Ni26.7Si10B5P5 sample was composed of Cr4.5BP2, Cr3 Co5Si2, and fcc phases. Synchrotron radiation anomalous X-ray scattering (AXS) measurements confirmed that the annealed Cr26.7Co26.7Ni26.7Si10B5P5 sample had superlattice reflections from an L10-, and/or L12-, and D022-type ordered structure. The diffraction intensities of the superlattice reflections observed in the AXS measurements indicated strong dependence on the incident X-ray energy. The diffraction intensity of the superlattice reflection increased at the X-ray energy near the K-edge of Cr, suggesting that Cr is mainly ordered in the L10- and D022-type structures. Microstructural analysis using scanning transmission electron microscopy (STEM) revealed crystal grains with diameters of approximately 20 nm, showing diffraction spots corresponding to superlattice reflections of 001 and 110, in addition to a two-dimensional diffraction pattern corresponding to the fcc structure. Simultaneous STEM--energy dispersive X-ray spectroscopy analysis indicated that the grains had a higher Cr concentration than the mother alloy composition of Cr26.7Co26.7Ni26.7Si10B5P5. For long-pulse negative ion–based neutral beam injection (NNBI) experiments, an automatic recording system was designed. The data processing algorithm of neutral beam injection was implemented on the basis of existing methods, and different experimental data were processed in a mode-based method according to the physical characteristics of the neutral beam injection experiment. Based on MDSplus segment storage technology, the range of recorded data was determined by intercepting the segment sequence number of the time series data. LabVIEW was used to achieve the efficient and accurate transmission of experimental processing data. A set of tests demonstrates that the NNBI auto-recording system meets the requirements of the NNBI experience in data processing accuracy (normalized mean square error [NMSE] < 0.04), data transmission accuracy (>98%), and data affect presentation. This system not only obviously improves the work efficiency, but also provides more reliable data sources for the intelligent analysis and processing of the NNBI system. [ABSTRACT FROM AUTHOR]

Background: Lead-acid battery workers are at higher risk for systemic diseases as well as oral diseases like dental caries. The aim of this study was to assess selected salivary antioxidants and their relation with dental caries among lead acid battery factory workers in comparison with non-exposed group. Materials and methods: The sample consisted of 35 subjects aged 35-45 year-old who worked in Babylon lead acid battery factory in Baghdad city and matching group that not exposed to lead were selected as a control. Dental caries severity was recorded by using DMFS index, stimulated salivary samples were collected and analyzed for the measurement of salivary antioxidants (uric acid, total protein, catalase and glutathione peroxidase enzymes). Results: The antioxidants levels (uric acid, catalase and glutathione peroxidase enzymes) were higher among the study group than the control group with non-significant difference for uric acid, highly significant difference for catalaseenzyme and significant for glutathione peroxidaseenzymes, whereas total protein level was significantly lower among the study group than the control. Regarding dental cariesseverity, DMFS values were significantly higher among study group compared to that among control group. All the correlations between salivary antioxidants and dental carries found to be weak non-significant for both groups. Conclusions: Selected salivary antioxidants were found to have little effects dental caries of the study group, although dental caries revealed higher percentage of occurrence among lead exposed workers. Therefore, special oral health preventive and educational programs are needed for them. Key words: lead exposure, lead acid battery workers, antioxidants, oral health status.
