Adolescence and emerging adulthood is likely a sensitive period for the neural effects of stress due to increasing life stress, onset of stress-related disorders, and continued gray matter (GM) development. In adults, stress is associated with GM differences in the medial prefrontal cortex (mPFC), hippocampus, and amygdala, but little is known about these relations, and whether they differ by gender, during adolescence and emerging adulthood. Further, it is unknown whether dependent (self-generated) and independent (fateful) stressors have distinct associations with GM, as each have distinct relations with internalizing psychopathology. We tested relations between recent dependent and independent stressor frequency (ALEQ-R) and GM structure using MRI in a priori regions of interest (mPFC, amygdala, and hippocampus) and across the cortex in youth from the Denver/Boulder metro area ages 14-22 (N = 144). Across both genders, no effects passed multiple comparison correction (FDR q > 0.05). However, there were significant differences between male and female youth (FDR q < 0.05), with opposite relations between dependent stressor frequency and cortical GM thickness in the salience network and emotion regulation regions and with surface area in default mode network regions. These results motivate future investigations of gender differences in neural mechanisms of stress generation and reactivity.

This paper reviews three studies investigating the relationship between brain regions involved in executive control and those involved in reading comprehension in typically-developing teens. In the first study, three regions of posterior left lateral prefrontal cortex (i.e., precentral gyrus, inferior frontal junction, inferior frontal gyrus) were consistently activated across three task domains: reading comprehension, numerical estimation, and executive processes in working memory, suggesting these regions exert domain-general executive control. In a second study, brain activity in the precentral gyrus during reading comprehension was found to be associated with individual differences in reading achievement. A subsequent investigation indicated that while all three prefrontal regions exhibit greater connectivity to the angular gyrus during reading comprehension compared to symbol identification, each also exhibits unique connectivity to distinct posterior regions implicated in reading processes. Hence, reading difficulties during adolescence may arise, in part, from difficulties in executive control over processing in language-related brain regions.

AimTo examine individual variability between perceived physical features and hormones of pubertal maturation in 9-10-year-old children as a function of sociodemographic characteristics.MethodsCross-sectional metrics of puberty were utilized from the baseline assessment of the Adolescent Brain Cognitive Development (ABCD) Study-a multi-site sample of 9-10 year-olds (n = 11,875)-and included perceived physical features via the pubertal development scale (PDS) and child salivary hormone levels (dehydroepiandrosterone and testosterone in all, and estradiol in females). Multi-level models examined the relationships among sociodemographic measures, physical features, and hormone levels. A group factor analysis (GFA) was implemented to extract latent variables of pubertal maturation that integrated both measures of perceived physical features and hormone levels.ResultsPDS summary scores indicated more males (70%) than females (31%) were prepubertal. Perceived physical features and hormone levels were significantly associated with child's weight status and income, such that more mature scores were observed among children that were overweight/obese or from households with low-income. Results from the GFA identified two latent factors that described individual differences in pubertal maturation among both females and males, with factor 1 driven by higher hormone levels, and factor 2 driven by perceived physical maturation. The correspondence between latent factor 1 scores (hormones) and latent factor 2 scores (perceived physical maturation) revealed synchronous and asynchronous relationships between hormones and concomitant physical features in this large young adolescent sample.ConclusionsSociodemographic measures were associated with both objective hormone and self-report physical measures of pubertal maturation in a large, diverse sample of 9-10 year-olds. The latent variables of pubertal maturation described a complex interplay between perceived physical changes and hormone levels that hallmark sexual maturation, which future studies can examine in relation to trajectories of brain maturation, risk/resilience to substance use, and other mental health outcomes.

The Adolescent Brain Cognitive Development (ABCD) Study is an ongoing, nationwide study of the effects of environmental influences on behavioral and brain development in adolescents. The main objective of the study is to recruit and assess over eleven thousand 9-10-year-olds and follow them over the course of 10 years to characterize normative brain and cognitive development, the many factors that influence brain development, and the effects of those factors on mental health and other outcomes. The study employs state-of-the-art multimodal brain imaging, cognitive and clinical assessments, bioassays, and careful assessment of substance use, environment, psychopathological symptoms, and social functioning. The data is a resource of unprecedented scale and depth for studying typical and atypical development. The aim of this manuscript is to describe the baseline neuroimaging processing and subject-level analysis methods used by ABCD. Processing and analyses include modality-specific corrections for distortions and motion, brain segmentation and cortical surface reconstruction derived from structural magnetic resonance imaging (sMRI), analysis of brain microstructure using diffusion MRI (dMRI), task-related analysis of functional MRI (fMRI), and functional connectivity analysis of resting-state fMRI. This manuscript serves as a methodological reference for users of publicly shared neuroimaging data from the ABCD Study.

The ABCD study is recruiting and following the brain development and health of over 10,000 9-10 year olds through adolescence. The imaging component of the study was developed by the ABCD Data Analysis and Informatics Center (DAIC) and the ABCD Imaging Acquisition Workgroup. Imaging methods and assessments were selected, optimized and harmonized across all 21 sites to measure brain structure and function relevant to adolescent development and addiction. This article provides an overview of the imaging procedures of the ABCD study, the basis for their selection and preliminary quality assurance and results that provide evidence for the feasibility and age-appropriateness of procedures and generalizability of findings to the existent literature.

The ability to enact cognitive control under changing environmental demands is commonly studied using set-shifting paradigms. While the control processes required for task set reconfiguration (switch costs) have been studied extensively, less research has focused on the control required during task repetition in blocks containing multiple tasks as compared to those containing a single task (mixing costs). We investigated how individual differences in mixing costs related to other executive functions (EFs) in a large sample (N = 749) of young adults. Individual differences in mixing costs across three different set-shifting paradigms loaded significantly onto a mixing cost latent variable. This Mixing Cost factor moderately correlated with a Common EF factor capturing variance shared across nine EF tasks designed to tap response inhibition, working memory updating, and mental set-shifting. It did not correlate with Updating-Specific and Shifting-Specific factors. Results indicate that the additional cognitive control required during mixed-task block repeat trials relies on general executive processes, as well as unique abilities distinct from both set-shifting and working memory updating.

Individuals vary greatly in their ability to select one item or response when presented with a multitude of options. Here we investigate the neural underpinnings of these individual differences. Using magnetic resonance spectroscopy, we found that the balance of inhibitory versus excitatory neurotransmitters in pFC predicts the ability to select among task-relevant options in two language production tasks. The greater an individual's concentration of GABA relative to glutamate in the lateral pFC, the more quickly he or she could select a relevant word from among competing options. This outcome is consistent with our computational modeling of this task [Snyder, H. R., Hutchison, N., Nyhus, E., Curran, T., Banich, M. T., O'Reilly, R. C., et al. Neural inhibition enables selection during language processing. Proceedings of the National Academy of Sciences, U.S.A., 107, 16483-16488, 2010], which predicts that greater net inhibition in pFC increases the efficiency of resolving competition among task-relevant options. Moreover, the association with the GABA/glutamate ratio was specific to selection and was not observed for executive function ability in general. These findings are the first to link the balance of excitatory and inhibitory neural transmission in pFC to specific aspects of executive function.

People must constantly select among potential thoughts and actions in the face of competition from (a) multiple task-relevant options (underdetermined competition) and (b) strongly dominant options that are not appropriate in the current context (prepotent competition). These types of competition are ubiquitous during language production. In this work, we investigate the neural mechanisms that allow individuals to effectively manage these cognitive control demands and to quickly choose words with few errors. Using fMRI, we directly contrast underdetermined and prepotent competition within the same task (verb generation) for the first time, allowing localization of the neural substrates supporting the resolution of these two types of competition. Using a neural network model, we investigate the possible mechanisms by which these brain regions support selection. Together, our findings demonstrate that all competition is not alike: resolving prepotent competition and resolving underdetermined competition rely on partly dissociable neural substrates and mechanisms. Specifically, activation of left ventrolateral pFC is specific to resolving underdetermined competition between multiple appropriate responses, most likely via competitive lateral inhibition. In contrast, activation of left dorsolateral pFC is sensitive to both underdetermined competition and prepotent competition from response options that are inappropriate in the current context. This region likely provides top-down support for task-relevant responses, which enables them to out-compete prepotent responses in the selection process that occurs in left ventrolateral pFC.

Recently, multi-voxel pattern analysis has verified that information can be removed from working memory (WM) via three distinct operations replacement, suppression, or clearing compared to information being maintained (Kim et al., 2020). While univariate analyses and classifier importance maps in Kim et al. (2020) identified brain regions that contribute to these operations, they did not elucidate whether these regions represent the operations similarly or uniquely. Using Leiden-community-detection on a sample of 55 humans (17 male), we identified four brain networks, each of which has a unique configuration of multi-voxel activity patterns by which it represents these WM operations. The visual network (VN) shows similar multi-voxel patterns for maintain and replace, which are highly dissimilar from suppress and clear, suggesting this network differentiates whether an item is held in WM or not. The somatomotor network (SMN) shows a distinct multi-voxel pattern for clear relative to the other operations, indicating the uniqueness of this operation. The default mode network (DMN) has distinct patterns for suppress and clear, but these two operations are more similar to each other than to maintain and replace, a pattern intermediate to that of the VN and SMN. The frontoparietal control network (FPCN) displays distinct multi-voxel patterns for each of the four operations, suggesting that this network likely plays an important role in implementing theseWMoperations. These results indicate that the operations involved in removing information fromWMcan be performed in parallel by distinct brain networks, each of which has a particular configuration by which they represent these operations. The serotonin transporter gene (5-HTTLPR) influences emotional reactivity and attentional bias toward or away from emotional stimuli, and has been implicated in psychopathological states, such as depression and anxiety disorder. The short allele is associated with increased reactivity and attention toward negatively-valenced emotional information, whereas the long allele is associated with increased reactivity and attention toward positively-valenced emotional information. The neural basis for individual differences in the ability to exert cognitive control over these bottom-up biases in emotional reactivity and attention is unknown, an issue investigated in the present study. Healthy adult participants were divided into two groups, either homozygous carriers of the 5-HTTLPR long allele or homozygous carriers of the short allele, and underwent functional magnetic resonance imaging (fMRI) while completing an Emotional Stroop-like task that varied in the congruency of task-relevant and task-irrelevant information and the emotional valence of the task-irrelevant information. Behaviorally, participants demonstrated the classic "Stroop effect" (responses were slower for incongruent than congruent trials), which did not differ by 5-HTTLPR genotype. However, fMRI results revealed that genotype influenced the degree to which neural systems were engaged depending on the valence of the conflicting task-irrelevant information. While the "Long" group recruited prefrontal control regions and superior temporal sulcus during conflict when the task-irrelevant information was positively-valenced, the "Short" group recruited these regions during conflict when the task-irrelevant information was negatively-valenced. Thus, participants successfully engaged cognitive control to overcome conflict in an emotional context using similar neural circuitry, but the engagement of this circuitry depended on emotional valence and 5-HTTLPR status. These results suggest that the interplay between emotion and cognition is modulated, in part, by a genetic polymorphism that influences serotonin neurotransmission.

The inhibition of unwanted behaviors is considered an effortful and controlled ability. However, inhibition also requires the detection of contexts indicating that old behaviors may be inappropriate--in other words, inhibition requires the ability to monitor context in the service of goals, which we refer to as context-monitoring. Using behavioral, neuroimaging, electrophysiological and computational approaches, we tested whether motoric stopping per se is the cognitively-controlled process supporting response inhibition, or whether context-monitoring may fill this role. Our results demonstrate that inhibition does not require control mechanisms beyond those involved in context-monitoring, and that such control mechanisms are the same regardless of stopping demands. These results challenge dominant accounts of inhibitory control, which posit that motoric stopping is the cognitively-controlled process of response inhibition, and clarify emerging debates on the frontal substrates of response inhibition by replacing the centrality of controlled mechanisms for motoric stopping with context-monitoring.
