Depression is a mental disorder that affects 300 million people of all ages worldwide, but fewer than half of those with the condition receive adequate treatment. In addition, the high pharmacological refractoriness (affecting 30%-50% of patients) and toxicity of some classical antidepressants support the pursuit of new therapies. People with this condition show depressed mood, loss of pleasure, high levels of oxidative stress, and accelerated biological aging (decreased telomere length and expression of the telomerase reverse transcriptase (TERT), the enzyme responsible for telomere maintenance). Because of the close relationship between depression and oxidative stress, nutraceuticals with antioxidant properties are excellent candidates for therapy. This study represents the first investigation of the possible antidepressant and antiaging effects of commercial samples of clarified açaí (Euterpe oleracea) juice (EO). This fruit is rich in antioxidants and widely consumed. In this study, mice were treated with saline or EO (10 μL/g, oral) for 4 days and then with saline or lipopolysaccharide (0.5 mg/kg, i.p.) to induce depressive-like behavior. Only four doses of EO were enough to abolish the despair-like and anhedonia behaviors and alterations observed in electromyographic measurements. The antidepression effect of EO was similar to that of imipramine and associated with antioxidant and antiaging effects (preventing lipid peroxidation and increasing TERT mRNA expression, respectively) in three major brain regions involved in depression (hippocampus, striatum, and prefrontal cortex). Additionally, EO significantly protected hippocampal cells, preventing neuronal loss associated with the depressive-like state and nitrite level increases (an indirect marker of nitric oxide production). Moreover, EO alone significantly increased TERT mRNA expression, revealing for the first time a potent antiaging action in the brain that suggests neuroprotection against long-term age-related consequences.

Human exposure to mercury is a serious problem of public health in Amazon. As in other vulnerable populations throughout the world, Amazonian riverine populations are chronically exposed to this metal and some symptoms of mercury intoxication were already detected in these populations. However, studies on the genetic susceptibility to mercury toxicity in the Amazon are scarce, and they tested a limited number of individuals. In this context, apolipoprotein E gene (APOE) is a key element with a well-established association among their alleles and the neurodegenerative consequences of mercury intoxication. However, no studies have addressed APOE genotyping in Amazonian exposed populations. Additionally, epidemiological studies with APOE genotyping in Amazon have been restricted to indigenous populations. Therefore, this work analyzed for the first time the genotypic and allelic profiles of APOE in Amazonian riverine populations chronically exposed to mercury. Eight hundred and twenty three individuals were enrolled in our study donating blood (794) and/or hair (757). APOE genotyping was analyzed by real-time PCR. Total mercury and mercury species were quantified by ICP-MS and GC-pyro-AFS, respectively. Genomic ancestry markers were evaluated by multiplex-PCR reaction, separated by capillary electrophoresis on the ABI 3130 Genetic Analyzer instrument and analyzed on GeneMapper ID v3.2. The 3 and 3/3 were the most frequent allele and genotype, respectively, followed by 4 allele and 3/4 genotype. Only 2/2 genotype was not found, suggesting that the absence of this genotype is a generalized phenomenon in Amazon. Also, our data supported an association between the presence of APOE4 and the Amerindian origin in these populations. Fifty-nine individuals were identified at maximum risk with levels of mercury above 10 μg/g and the presence of APOE4. Interestingly, among individuals with high mercury content, APOE4-carriers had high mercury levels than APOE2-carriers, pointing to a different heavy metal accumulation according to the APOE allele. These data suggest that APOE4, in addition to a possible pharmacodynamic effect, may influence pharmacokinetically the mercury exposure causing its higher accumulation and leading to worse deleterious consequences. Our results may aid in the development of prevention strategies and health policy decision-making regarding these at-risk vulnerable populations. Assim como em outras populações vulneráveis ​​em todo o mundo, populações ribeirinhas amazônicas estão cronicamente expostas a esse metal e alguns sintomas de intoxicação por mercúrio já foram detectados nessas populações. No entanto, estudos sobre a suscetibilidade genética à toxicidade do mercúrio na Amazônia são escassos e testaram um número limitado de indivíduos. Nesse contexto, o gene da apolipoproteína E (APOE) é um elemento chave com uma associação bem estabelecida entre seus alelos e as consequências neurodegenerativas da intoxicação por mercúrio. No entanto, nenhum estudo abordou a genotipagem da APOE em populações expostas na Amazônia. Além disso, estudos epidemiológicos com genotipagem APOE na Amazônia têm sido restritos a populações indígenas. Portanto, este trabalho analisou pela primeira vez os perfis genotípicos e alélicos de APOE em populações ribeirinhas amazônicas expostas cronicamente ao mercúrio. Oitocentos e vinte e três indivíduos foram incluídos em nosso estudo doando sangue (794) e/ou cabelo (757). A genotipagem da APOE foi analisada por PCR em tempo real. O mercúrio total e as espécies de mercúrio foram quantificados por ICP-MS e GC-piro-AFS, respectivamente. Os marcadores de ancestralidade genômica foram avaliados por reação multiplex-PCR, separados por eletroforese capilar no instrumento ABI 3130 Genetic Analyzer e analisados ​​no GeneMapper ID v3.2. O 3 e 3/3 foram os alelos e genótipos mais frequentes, respetivamente, seguidos do alelo 4 e do genótipo 3/4. Apenas o genótipo 2/2 não foi encontrado, sugerindo que a ausência desse genótipo é um fenômeno generalizado na Amazônia. Além disso, nossos dados apoiaram uma associação entre a presença de APOE4 e a origem ameríndia nessas populações. Cinquenta e nove indivíduos foram identificados em risco máximo com níveis de mercúrio acima de 10 μg/ge presença de APOE4. Curiosamente, entre os indivíduos com alto teor de mercúrio, os portadores de APOE4 apresentaram níveis de mercúrio mais elevados do que os portadores de APOE2, apontando para um acúmulo diferente de metais pesados ​​de acordo com o alelo APOE. Esses dados sugerem que a APOE4, além de um possível efeito farmacodinâmico, pode influenciar farmacocinéticamente a exposição ao mercúrio causando seu maior acúmulo e levando a piores consequências deletérias. Nossos resultados podem auxiliar no desenvolvimento de estratégias de prevenção e tomada de decisão de políticas de saúde em relação a essas populações vulneráveis ​​em risco.

Per un buon funzionamento di tali strutture, è necessario che le stazioni siano inquadrate nel sistema di riferimento delle orbite dei satelliti GNSS. Come noto tale sistema di riferimento si materializza sulla superficie terrestre mediante un frame che attribuisce sia coordinate che velocità ad osservatori geodetici (stazioni GPS, etc..) permanenti appartenenti a circuiti internazionali (IGS, EUREF, etc..). Le coordinate di un qualsiasi punto sulla superficie terreste cambiano con continuità anche se il punto non subisce l’azione di eventi locali (frane, terremoti, etc.). Questo comporta la necessità di eseguire periodicamente l’inquadramento della rete istituita nel frame delle stazioni che definiscono il Sistema Geodetico assunto. Tali calcoli richiedono l’uso di programmi di natura scientifica capaci di gestire sia le ricezioni che dati ancillari anch’essi prodotti dai centri di gestione dei sistemi geodetici internazionali. Attualmente i principali software che vengono utilizzati sono il codice Bernese, Gamit-Globk e Gipsy/Oasis II. Tali software possono basarsi o sul classico approccio differenziato con il calcolo di basi GNSS o sull’approccio non differenziato noto con l’acronimo PPP (Precise Point Positioning). Il lavoro fornisce alcune considerazioni sulla elaborazione dei dati di tali reti inquadrate nei sistemi di riferimento globali, utilizzando differenti software scientifici ed utilizzando differenti approcci.

This paper show the main results obtained by some experimentations performed with the aim to improve the trajectory of the vehicle during GPS outages using GPS virtual positions. As well known the navigation system is generally constituted by one or more geodetic GNSS receivers and other sensors such as Inertial Measurement Units (IMU) and Odometers (ODO). The observations acquired by these sensors can be fused using Kalman Filter applied with different approaches (Tightly or Loosely Coupled); when all the navigation systems work well, the final accuracy can be at centimeters level. As well known in historical city centers the sky visibility can be reduced for the presence of high buildings closed to the streets. Working in wide areas the solution obtained by the data fusion between GPS and IMU sensors can be assumed as good and can constitute the reference trajectory useful to compare other solutions obtained in different ways. Starting from this reference solution other solution can be obtained performing a GPS outage simulation obtained cutting GPS solution for a defined window time. The new GPS file has been combined again with the IMU into the Kalman filter obtaining a solution in presence of a GPS outage. Starting from this last solution and disposing of the original file of GPS positions (without GPS outages), some simulation has been performed reintroducing some original GPS solutions into the file characterized by GPS outage and reprocessing the data into the Kalman filter. The reintroduction of the GPS positions simulates the correction potentially derived by topographic survey. In order to evaluate the influence of this approach in terms of accuracy and cost/benefit ratio, many tests using the three test areas has been performed and the obtained results are presented.

Neurotoxicity induced by methylmercury (MeHg) increases the formation of reactive radicals and accelerates free radical reactions. This review summarizes recent findings in the MeHg-induced formation of free radicals and the role of oxidative stress in its neurotoxicity. Oxidative stress on CNS can produce damage by several interacting mechanisms, including mitochondrial damage with increase in intracellular free Ca(2+), activation and inhibition of enzymes, release of excitatory amino acids, metallothioneins expression, and microtubule disassembly. The nature of antioxidants is discussed and it is suggested that antioxidant enzymes and others antioxidants molecules may protect the central nervous system against neurotoxicity caused by MeHg.

Negli ultimi anni in Italia sono nate e stanno consolidandosi reti di Stazioni Permanenti GNSS Regionali per il posizionamento in tempo reale (reti NRTK). Per un buon funzionamento di tali strutture, è necessario che le stazioni siano inquadrate nel sistema di riferimento delle orbite dei satelliti GNSS. Come noto tale sistema di riferimento si materializza sulla superficie terrestre mediante un frame che attribuisce sia coordinate che velocità ad osservatori geodetici (stazioni GPS, etc..) permanenti appartenenti a circuiti internazionali (IGS, EUREF, etc..). Dunque, le coordinate di un qualsiasi punto sulla superficie terreste cambiano con continuità anche se il punto non subisce l’azione di eventi locali (frane, terremoti, etc.). Altro aspetto di natura differente dal primo consiste nel problema legato all’esigenza che reti adiacenti o addirittura sovrapposte (in grado di fornire un servizio di posizionamento di precisione in tempo reale nelle medesime aree) consentano all’utenza di poter lavorare su un sistema di riferimento comune. Il lavoro che si intende presentare fornisce alcuni risultati, ancora preliminari, su aspetti legati all’elaborazione dei dati di tali reti inquadrate nei sistemi di riferimento globali, utilizzando differenti software scientifici. Ci si pone dunque il problema di quali siano le procedure più idonee alla determinazione di tali coordinate e quali siano le differenze in termini di calcolo nell’utilizzo di differenti codici di calcolo.

Riassunto In questo lavoro viene descritta un'esperienza di rilievo ed elaborazione dati acquisiti, nell'ambito di una rete sperimentale, con l'acquisizione congiunta dalle costellazioni dei sistemi GPS e GLONASS e l'integrazione delle osservabili ai fini dell'elaborazione delle rete in modalità statica. Tale rete sarà oggetto di future sperimentazioni con metodologie Network-based Realt Time Kinematic (NRTK). Saranno brevemente descritte le peculiarità del sistema GLONASS, gli accorgimenti da seguire ai fini dell'integrazione delle osservazioni e discussi i risultati ottenuti nella elaborazione dei dati effettuata nella doppia modalità, ovvero utilizzando esclusivamente osservazioni GPS e GPS-GLONASS congiunte.
