Zanthoxylum leprieurii is a medicinal plant widely studied due to its great phytochemical diversity, especially its acrinonic alkaloids, which have shown to be promising anticancer candidates. The aim of this work was to promote the isolation of acridonic alkaloids from fruits of Z. leprieurii and carried out absorption and fluorescence spectroscopy studies with calf thymus DNA and BSA. Five acridone alkaloids have been isolated, including the first description of 3-desmethoxy arborinine ( 2 ). In the study of interaction with biomacromolecules it was observed that all compounds show interaction with calf thymus DNA and BSA. Compound 2 promoted the bigger increase in BSA fluorescence (3.01%) with a lower fluorescence quenching constant (Ksv = 0.13 × 10 4 ). Taken together, these results reaffirm the great phytochemical diversity of Z. leprieurii , and show that acridonic alkaloids have an affinity with both DNA and BSA, therefore providing clues to their mechanisms of action related to their anticancer activities.

Recent studies on the distribution of microplastics in aquatic sediments have deployed different methods and devices for density separation of microplastics from sediments. However, instrument specific limitations have been noted, including their high cost, difficulty in handling, or/and the potential for elevated contamination risk due to their plastic composition. This study improves existing sediment microplastic separation techniques by modifying the commonly used conical shape glass separating funnels. The modification consists in connecting a silicone tube at the base of the funnel, whose opening and closure was manually controlled by a Mohr clamp. This adjustment made to the funnels have effectively mitigated critical clogging problems frequently encountered in density separation units. An experiment was conducted using sand-based sediment spiked with polyamide fragments to validate this method modification. Following a complete extraction protocol with the modification of separating funnels, the microplastic extraction efficiency from sediments was high with a 90% recovery rate. Based on these promising results, future studies should consider naturally diverse substrates, as recovery efficiency may be sediment-dependent. Human African Trypanosomiasis (HAT) is an infectious disease caused by protozoan parasites belonging to the Trypanosoma genus. In sub-Saharan Africa, there is a significant threat as many people are at risk of infection. Despite this, HAT is classified as a neglected tropical disease. Over the last few years, several studies have reported the existence of a wide diversity of trypanosome species circulating in African animals. Thus, domestic and wild animals could be reservoirs of potentially dangerous trypanosomes for human populations. However, very little is known about the role of domestic animals in maintaining the transmission cycle of human trypanosomes in central Africa, especially in Gabon, where serious cases of infection are recorded each year, sometimes leading to hospitalization or death of patients. Komo-Mondah, located within Estuaries (Gabonese province), stays the most active HAT disease focus in Gabon, with a mean of 20 cases per year. In this study, we evaluated the diversity and prevalence of trypanosomes circulating in domestic animals using the Polymerase Chain Reaction (PCR) technique. We found that 19.34% (53/274) of the domestic animals we studied were infected with trypanosomes. The infection rates varied among taxa, with 23.21% (13/56) of dogs, 16.10% (19/118) of goats, and 21.00% (21/100) of sheep infected. In addition, we have observed a global mixed rate of infections of 20.75% (11/53) among infected individuals. Molecular analyses revealed that at least six Trypanosome species circulate in domestic animals in Gabon ( T. congolense , T. simiae , T. simiae Tsavo, T. theileri , T. vivax , T. brucei (including T. brucei brucei , and T. brucei gambiense )). In conclusion, our study showed that domestic animals constitute important potential reservoirs for trypanosome parasites, including T. brucei gambiense , which is responsible for HAT.

Background: We have previously demonstrated that a mixture of Curcuminoids extract, hydrolyzed COllagen and green Tea extract (CCOT) inhibited inflammatory and catabolic mediator's synthesis by bovine and human chondrocytes. A randomly allocated, double-blind, prospective, placebo-controlled study was performed to evaluate the efficacy of a diet containing this CCOT mixture on dogs with naturally occurring osteoarthritis (OA). Therefore, 42 owner's dogs with OA were randomly assigned to receive for 3 months an experimental diet (control) or the same diet supplemented with CCOT., Results: Ground reaction forces did not show statistical differences between groups. After 3 months of feeding, there was a significant reduction of pain at manipulation in the CCOT group, but not in the control group. The evolution for pain at manipulation depended on the diet. The three other parameters evaluated by veterinary subjective assessment (lameness, pain at palpation and joint mobility) did not show statistical differences. Concerning owner subjective assessment, pain severity score worsened in the control group but remained stable in CCOT group. The evolution for pain severity depended on the diet. No statistical difference was found for pain interference, except for the ability to rise to standing from lying down, which was significantly improved in the CCOT compared to the control group. Serum OA biomarkers did not show statistical differences., Conclusions: Objective variables measured, such as ground reaction forces and OA biomarkers, did not show statistical differences. However, indicators of pain appeared reduced in dogs receiving CCOT mixture for 3 months. The difference of evolution between groups suggests that a greater number of dogs may be necessary to reach a stronger effect on other parameters.

Objectives: To describe the complications, short and long-term outcome and owner satisfaction of dogs with cranial cruciate ligament rupture treated with a recently described new osteotomy for the modified Maquet technique (N-MMT)., Materials and Methods: Medical records and radiographs of 82 dogs (84 stifles) were reviewed. Details regarding short-term outcome and complications were recorded from the medical records. Long-term follow-up information was obtained by telephone interview. Historical data and complications were statistically analysed., Results: Major complications occurred in 34/84 stifles. Intra-operative complications occurred in 26/84 stifles, all of which were fissures or fractures of the cortical hinge. Twenty-one of these fractures or fissures were repaired with a figure-of-eight wire. The second most common major complication was late meniscal tears in 3/84 stifles. One dog sustained a complete tibial fracture. Non-displaced fracture of the cortical hinge was the most common postoperative minor complication, which occurred in 5/84 stifles. The median preoperative lameness score was 3 out of 6. Final in-hospital re-evaluation of limb function was available in 58 dogs. The median lameness score at that time was 0 out of 6. Development of osteotomy related complications was not associated with a significant change in the postoperative lameness score., Clinical Significance: Subjectively assessed clinical outcome with the N-MMT was good to excellent in this cohort of dogs. However, a high rate of intra- and postoperative complications of the N-MMT procedure was also present in these dogs.

Objective: To describe 2 minimally invasive approaches to the spinal canal for treatment of intervertebral disc disease and compare their efficacy to conventional hemilaminectomy., Study Design: Experimental; randomized, controlled design., Animals: Canine cadavers (n = 10; 5 small and 5 large dogs)., Methods: Barium-impregnated agarose gel (BA-gel) was injected into the spinal canal at 3 intervertebral spaces of the thoracolumbar spine in each cadaver. Sites were randomly assigned to 1 of 3 approaches: conventional (standard) hemilaminectomy (SH), endoscopic foraminotomy (EF), or foraminotomy via an illuminated port (FP). Computed tomographic scans were performed before and after the procedures. Procedures were compared for duration, bone window size, incision length, complications and percentage of BA-gel removed via repeated measures ANOVA., Results: The incisions created during EF and FP were similar and smaller to that of a SH. The duration of EF was prolonged compared to FP and SH. The size of the vertebral window created was greater after SH in large dogs, while no difference was found between procedures in small dogs. The amount of simulated disc material removed from the spinal canal did not differ between procedures, regardless of the size of the dog., Conclusions: The two minimally invasive approaches were feasible in small and large dogs. Both techniques allowed similar removal of simulated disc material and may decrease soft tissue morbidity compared to SH., (© Copyright 2014 by The American College of Veterinary Surgeons.)

Objectives: To evaluate the mechanical properties of the distal cortical hinge associated with a new osteotomy design for the Modified Maquet Technique (MMT)., Study Design: Ex vivo mechanical study., Methods: The osteotomy was started 10 mm caudal to the tibial tuberosity and extended over 150% of the length of the tibial crest; it was slightly curved distally to stay at a distance of 2 to 4 mm from the cranial cortex, according to the body weight. Ninety-six tibiae were tested in advancement, and 60 tibiae were axially loaded perpendicular to the tibial plateau, until failure of the crest. Desired advancement was measured using the common tangent method in 60 tibiae. Angle of opening, thickness, and area of the cortical hinge were recorded., Results: Desired advancement of 6 mm, 9 mm, 12 mm and 15 mm was recorded in 16, 12, 18 and 14 tibiae respectively. Mean maximal advancement in these bones was 15.6 ± 6.4 mm, 20.8 ± 5.2 mm, 21.3 ± 5.2 mm and 22.7 ± 5.2 mm respectively. The desired advancement was reached in all but one tibia. Advancement was mainly influenced by the angle of opening and the stiffness of the cortical hinge. Mean ultimate load to failure was 6.12 ± 2.4 times the body weight. It was significantly associated with the body weight, thickness, and area of the cortical hinge., Clinical Relevance: Mean maximal advancement was higher than clinically required without occurrence of fissure or fracture. Ultimate load to failure and maximal advancement could be predicted using calculated formulae.
