Adiponectin is believed to exert hepatoprotective effects and induces CXCL8, a chemokine that functions as a survival factor, in vascular cells. In the current study, it is demonstrated that adiponectin also induces CXCL8 expression in primary human hepatocytes but not in hepatocellular carcinoma cell lines. Knock down of the adiponectin receptor (AdipoR) 1 or AdipoR2 by small-interfering RNA indicates that AdipoR1 is involved in adiponectin-stimulated CXCL8 release. Adiponectin activates nuclear factor (NF)-[kappa]B in primary hepatocytes and pharmacological inhibition of NF-[kappa]B, the p38 mitogen-activated protein kinase, and extracellular signal-regulated kinase (ERK) 1/ERK2 reduces adiponectin-mediated CXCL8 secretion. Furthermore, adiponectin also activates STAT3 involved in interleukin (IL)-6 and leptin-mediated CXCL8 induction in primary hepatocytes. Inhibition of JAK2 by AG-490 does not abolish adiponectin-stimulated CXCL8, indicating that this kinase is not involved. Pretreatment of primary cells with 'STAT3 Inhibitor VI,' however, elevates hepatocytic CXCL8 secretion, demonstrating that STAT3 is a negative regulator of CXCL8 in these cells. In accordance with this assumption, IL-6, a well-characterized activator of STAT3, reduces hepatocytic CXCL8. Therefore, adiponectin-stimulated induction of CXCL8 seems to be tightly controlled in primary human hepatocytes, whereas neither NF-[kappa]B, STAT3, nor CXCL8 are influenced in hepatocytic cell lines. - Abstract
While the Kantian epistemology was discovered for analytical philosophy by P. F. Strawson in the middle of the last century and has inspired this philosophical discourse since then, the metaphysical features of Kantian philosophy have mostly been ignored in recent times. What is overlooked is the fact that Kant's epistemological foundation of philosophy through the Critique of Pure Reason aims precisely to establish metaphysics as a science. In order to evaluate the possibility of scientific metaphysics, the Critique of Pure Reason examines the principles of synthetic judgments a priori. She comes to the result that the metaphysical questioning about the absolute is laid out in the finite discursive reason of man as such, but cannot be satisfied with the means of speculative use of reason, but refers to the practical use of reason.

Unternehmen nutzen Techniken des strategischen Managements, um sich in zunehmend globaleren Märkten und damit in einem zunehmend komplexeren und schwierigeren, durch hohen Konkurrenzdruck und hohe Zukunftsunsicherheit geprägten wirtschaftlichen, technischen, politischen und gesellschaftlichen Umfeld zu orientieren und zu behaupten. Es gilt, eine entsprechende Strategie zu entwickeln, diesem Umfeld zu begegnen und sich selbst möglichst günstig in ihm zu positionieren; ein Alleinstellungsmerkmal, einen strategischen Wettbewerbsvorteil gegenüber anderen Marktanbietern zu erreichen; dies alles natürlich unter möglichst effizientem Einsatz und möglichst optimaler Ausschöpfung eigener Ressourcen. Die vorliegende Arbeit untersucht, inwieweit sich der Ansatz des strategischen Managements – und hier insbesondere Techniken der Strategieentwicklung – auch für den Einsatz im Bibliotheksbereich eignen. Dabei liegt der Fokus auf den beiden in der Praxis mit Abstand am weitesten verbreiteten Strategiefindungsinstrumenten: Der SWOT- und der Portfolio-Analyse. - Abstract
Given the anthropometric differences between men and women and previous evidence of sex-difference in genetic effects, we conducted a genome-wide search for sexually dimorphic associations with height, weight, body mass index, waist circumference, hip circumference, and waist-to-hip-ratio (133,723 individuals) and took forward 348 SNPs into follow-up (additional 137,052 individuals) in a total of 94 studies. Seven loci displayed significant sex-difference (FDR<5%), including four previously established (near GRB14/COBLL1, LYPLAL1/SLC30A10, VEGFA, ADAMTS9) and three novel anthropometric trait loci (near MAP3K1, HSD17B4, PPARG), all of which were genome-wide significant in women (P<5x10(-8)), but not in men. Sex-differences were apparent only for waist phenotypes, not for height, weight, BMI, or hip circumference. Moreover, we found no evidence for genetic effects with opposite directions in men versus women. The PPARG locus is of specific interest due to its role in diabetes genetics and therapy. Our results demonstrate the value of sex-specific GWAS to unravel the sexually dimorphic genetic underpinning of complex traits.

- Abstract
Men and women differ substantially regarding height, weight, and body fat. Interestingly, previous work detecting genetic effects for waist-to-hip ratio, to assess body fat distribution, has found that many of these showed sex-differences. However, systematic searches for sex-differences in genetic effects have not yet been conducted. Therefore, we undertook a genome-wide search for sexually dimorphic genetic effects for anthropometric traits including 133,723 individuals in a large meta-analysis and followed promising variants in further 137,052 individuals, including a total of 94 studies. We identified seven loci with significant sex-difference including four previously established (near GRB14/COBLL1, LYPLAL1/SLC30A10, VEGFA, ADAMTS9) and three novel anthropometric trait loci (near MAP3K1, HSD17B4, PPARG), all of which were significant in women, but not in men. Of interest is that sex-difference was only observed for waist phenotypes, but not for height or body-mass-index. We found no evidence for sex-differences with opposite effect direction for men and women. The PPARG locus is of specific interest due to its link to diabetes genetics and therapy. Our findings demonstrate the importance of investigating sex differences, which may lead to a better understanding of disease mechanisms with a potential relevance to treatment options.

Unternehmen nutzen Techniken des strategischen Managements, um sich in zunehmend globaleren Märkten und damit in einem zunehmend komplexeren und schwierigeren, durch hohen Konkurrenzdruck und hohe Zukunftsunsicherheit geprägten wirtschaftlichen, technischen, politischen und gesellschaftlichen Umfeld zu orientieren und zu behaupten. Es gilt, eine entsprechende Strategie zu entwickeln, diesem Umfeld zu begegnen und sich selbst möglichst günstig in ihm zu positionieren; ein Alleinstellungsmerkmal, einen strategischen Wettbewerbsvorteil gegenüber anderen Marktanbietern zu erreichen; dies alles natürlich unter möglichst effizientem Einsatz und möglichst optimaler Ausschöpfung eigener Ressourcen. Die vorliegende Arbeit untersucht, inwieweit sich der Ansatz des strategischen Managements – und hier insbesondere Techniken der Strategieentwicklung – auch für den Einsatz im Bibliotheksbereich eignen. Dabei liegt der Fokus auf den beiden in der Praxis mit Abstand am weitesten verbreiteten Strategiefindungsinstrumenten: Der SWOT- und der Portfolio-Analyse. Die Arbeit kommt zu einem insgesamt positiven Fazit.

- Abstract
The metabolic syndrome is defined as a cluster of disorders including visceral obesity, insulin resistance, hypertension, hypertriglyceridemia and low HDL. Patients have an increased risk to develop atherosclerotic disease characterized by excessive macrophage cholesterol deposition in the vascular wall. HDL removes excess cellular cholesterol which is subsequently transported to the liver for biliary excretion and thereby preserves cholesterol homeostasis. Circulating HDL levels are maintained by hepatic ATP-binding cassette transporter A1 (ABCA1) which also transports peripheral cell cholesterol to extracellular lipid acceptors. Lipid export activity of ABCA1 improves pancreatic -cell function and ameliorates insulin release. ABCA1 affects plasma membrane cholesterol distribution and formation of lipid rafts representing signalling platforms for diverse receptors including toll-like receptor 4 (TLR4). Pharmacological activation of ABCA1 pathways presumably progresses metabolic diseases, and current approaches demonstrate beneficial effects of small peptides mimicking ABCA1 ligands which stabilize ABCA1 and enhance lipid efflux similar to its physiological acceptor apolipoprotein A-I. Research of the last decade has resulted in the identification of several ABCA1 binding proteins influencing ABCA1 signalling, stability and activity. In the current review the proteins suggested to form a complex with ABCA1 are summarized and their up to now characterized features towards ABCA1 functions are described.
