We present a detailed chronological reconstruction of the 2011 eruption of Puyehue- Cordón Caulle volcano (Chile) based on information derived from newspapers, scientific reports and satellite images. Chronology of associated volcanic processes and their local and regional effects (i.e. precursory activity, tephra fallout, lahars, pyroclastic density currents, lava flows) are also presented. The eruption had a severe impact on the ecosystem and on various economic sectors, including aviation, tourism, agriculture, and fishing industry. Urban areas and critical infrastructures, such as airports, hospitals and roads, were also impacted. The concentration of PM10 (Particulate Matter ≤10 µm) was measured during and after the eruption, showing that maximum safety threshold levels of daily and annual exposures were surpassed in several occasions. Probabilistic analysis of atmospheric and eruptive conditions have shown that the main direction of dispersal is directly towards east of the volcano and that the climactic phase of the eruption, dispersed toward south-east, has a probability of occurrence within 1%. The management of the crisis, including evacuation of people, is discussed, as well as the comparison with the impact associated with other recent eruptions located in similar areas and having similar characteristics (i.e. Quizapu, Hudson, and Chaitén volcanoes). This comparison shows that the regions downwind and very close to the erupting volcanoes suffered very similar problems, without a clear relation with the in tensity of the eruption (e.g. health problems, damage to vegetation, death of animals, roof collapse, air traffic disruptions, road closure, lahars and flooding). This suggests that a detailed collection of impact data can be largely beneficial for the development of plans for the management of an eruptive crisis and the mitigation of associated risk of the Andean region. We report on the optical characterization of site-controlled InP/GaInP quantum dots (QDs). Spatially resolved low temperature cathodoluminescence proves the long-range ordering of the buried emitters, revealing a yield of ∼90% of optically active, positioned QDs and a strong suppression of emitters on interstitial positions. The emission of single QDs shows a pronounced degree of linear polarization along the [0,-1,1] crystal axis with an average degree of polarization of 94%. Photon correlation measurements of the emission from a single QD indicate the single-photon character of the exciton and biexciton emission lines as well as the cascaded nature of the photon pair. Splice-switching oligonucleotides (SSOs) can restore protein functionality in pathologies and are promising tools for manipulating the RNA-splicing machinery. Delivery vectors can considerably improve SSO functionality in vivo and allow dose reduction, thereby addressing the challenges of RNA-targeted therapeutics. Here, we report a biocompatible SSO nanocarrier, based on redox-responsive disulfide cross-linked low-molecular-weight linear polyethylenimine (cLPEI), for overcoming multiple biological barriers from subcellular compartments to en-route serum stability and finally in vivo delivery challenges. Intracellularly responsive cross-links of cLPEI significantly accelerated the endosomal escape and offered efficient SSO release to the cell's nucleus, thereby leading to high splice correction in vitro. In vivo performance of cLPEI-SSOs was investigated in a novel transgenic mouse model for splice correction, spatiotemporal tracking of SSO delivery in wild-type mice, and biodistribution in a colorectal cancer peritoneal metastasis model. A single intravenous application of 5 mg kg -1 cLPEI-SSOs induced splice correction in liver, lung, kidney, and bladder, giving functional protein, which was validated by RT-PCR. Near-infrared (NIR) fluorescence imaging and X-ray computed tomography revealed improved organ retention and reduced renal excretion of SSOs. NIR microscopy demonstrated the accumulation of SSOs in angiogenic tumors within the pancreas. Successful nuclear delivery of SSOs was observed in the hepatocytes. Thus, cLPEI nanocarriers resulted in highly efficient splice correction in vivo, highlighting the critical role of the enhanced SSO bioavailability.

Spatial exploration is a complex behavior that can be used to gain information about developmental processes, personality traits, or mental disorders. Typically, this is done by analyzing movement throughout an unknown environment. However, in human research, until now there has been no overview on how to analyze movement trajectories with regard to exploration. In the current paper, we provide a discussion of the most common movement measures currently used in human research on spatial exploration, and suggest new indices to capture the efficiency of exploration. We additionally analyzed a large dataset (n = 409) of human participants exploring a novel virtual environment to investigate whether movement measures could be assigned to meaningful higher-order components. Hierarchical clustering of the different measures revealed three different components of exploration (exploratory behavior, spatial shape, and exploration efficiency) that in part replicate components of spatial exploratory behavior identified in animal studies. A validation of our analysis on a second dataset (n = 102) indicated that two of these clusters are stable across different contexts as well as participant samples. For the exploration efficiency cluster, our validation showed that it can be further differentiated into a goal-directed versus a general, area-directed component. In humans, defects in leucine catabolism cause a variety of inborn errors in metabolism. Here, we use Caenorhabditis elegans to investigate the impact of mutations in mccc-1, an enzyme that functions in leucine breakdown. Through untargeted metabolomic and transcriptomic analyses we find extensive metabolic rewiring that helps to detoxify leucine breakdown intermediates via conversion into previously undescribed metabolites and to synthesize mevalonate, an essential metabolite. We also find that the leucine breakdown product 3,3-hydroxymethylbutyrate (HMB), commonly used as a human muscle-building supplement, is toxic to C. elegans and that bacteria modulate this toxicity. Unbiased genetic screens revealed interactions between the host and microbe, where components of bacterial pyrimidine biosynthesis mitigate HMB toxicity. Finally, upregulated ketone body metabolism genes in mccc-1 mutants provide an alternative route for biosynthesis of the mevalonate precursor 3-hydroxy-3-methylglutaryl-CoA. Autophagy serves as a stress response pathway by mediating the degradation of cellular material within lysosomes. In autophagy, this material is encapsulated in double-membrane vesicles termed autophagosomes, which form from precursors referred to as phagophores. Phagophores grow by lipid influx from the endoplasmic reticulum into Atg9-positive compartments and local lipid synthesis provides lipids for their expansion. How phagophore nucleation and expansion are coordinated with lipid synthesis is unclear. Here, we show that Faa1, an enzyme activating fatty acids, is recruited to Atg9 vesicles by directly binding to negatively charged membranes with a preference for phosphoinositides such as PI3P and PI4P. We define the membrane-binding surface of Faa1 and show that its direct interaction with the membrane is required for its recruitment to phagophores. Furthermore, the physiological localization of Faa1 is key for its efficient catalysis and promotes phagophore expansion. The famous Wigner's friend experiment considers an observer-the friend-and a superobserver-Wigner-who treats the friend as a quantum system and her interaction with other quantum systems as unitary dynamics. This is at odds with the friend describing this interaction via collapse dynamics, if she interacts with the quantum system in a way that she would consider a measurement. These different descriptions constitute the Wigner's friend paradox. Extended Wigner's friend experiments combine the original thought experiment with non-locality setups. This allows for deriving local friendliness inequalities, similar to Bell's theorem, which can be violated for certain extended Wigner's friend scenarios. A Wigner's friend paradox and the violation of local friendliness inequalities require that no classical record exists, which reveals the result the friend observed during her measurement. Otherwise, Wigner agrees with his friend's description and no local friendliness inequality can be violated. In this article, I introduce classical communication between Wigner and his friend and discuss its effects on the simple as well as extended Wigner's friend experiments. By controlling the properties of a (quasi) classical communication channel between Wigner and the friend, one can regulate how much outcome information about the friend's measurement is revealed. This gives a smooth transition between the paradoxical description and the possibility of violating local friendliness inequalities, on the one hand, and the effectively collapsed case, on the other hand.
