Abstract

Variants in RAC3, encoding a small GTPase RAC3 which is critical for the regulation of actin cytoskeleton and intracellular signal transduction, are associated with a rare neurodevelopmental disorder with structural brain anomalies and facial dysmorphism. We investigated a cohort of 10 unrelated participants presenting with global psychomotor delay, hypotonia, behavioural disturbances, stereotyped movements, dysmorphic features, seizures and musculoskeletal abnormalities. MRI of brain revealed a complex pattern of variable brain malformations, including callosal abnormalities, white matter thinning, grey matter heterotopia, polymicrogyria/dysgyria, brainstem anomalies and cerebellar dysplasia. These patients harboured eight distinct de novo RAC3 variants, including six novel variants (NM_005052.3): c.34G > C p.G12R, c.179G > A p.G60D, c.186_188delGGA p.E62del, c.187G > A p.D63N, c.191A > G p.Y64C and c.348G > C p.K116N. We then examined the pathophysiological significance of these novel and previously reported pathogenic variants p.P29L, p.P34R, p.A59G, p.Q61L and p.E62K. In vitro analyses revealed that all tested RAC3 variants were biochemically and biologically active to variable extent, and exhibited a spectrum of different affinities to downstream effectors including p21-activated kinase 1. We then focused on the four variants p.Q61L, p.E62del, p.D63N and p.Y64C in the Switch II region, which is essential for the biochemical activity of small GTPases and also a variation hot spot common to other Rho family genes, RAC1 and CDC42. Acute expression of the four variants in embryonic mouse brain using in utero electroporation caused defects in cortical neuron morphology and migration ending up with cluster formation during corticogenesis. Notably, defective migration by p.E62del, p.D63N and p.Y64C were rescued by a dominant negative version of p21-activated kinase 1. Our results indicate that RAC3 variants result in morphological and functional defects in cortical neurons during brain development through variant-specific mechanisms, eventually leading to heterogeneous neurodevelopmental phenotypes. Scala et al. identify six de novo variants in RAC3, which encodes a small GTPase, in 10 unrelated subjects with neurodevelopmental phenotypes. In vivo and in vitro analyses in mice reveal that RAC3 variants cause morpho-functional defects in cortical neurons through variant-specific mechanisms, disrupting corticogenesis.

Contains fulltext : 252015.pdf (Publisher’s version ) (Closed access), Robinow syndrome is characterized by a triad of craniofacial dysmorphisms, disproportionate-limb short stature, and genital hypoplasia. A significant degree of phenotypic variability seems to correlate with different genes/loci. Disturbances of the noncanonical WNT-pathway have been identified as the main cause of the syndrome. Biallelic variants in ROR2 cause an autosomal recessive form of the syndrome with distinctive skeletal findings. Twenty-two patients with a clinical diagnosis of autosomal recessive Robinow syndrome were screened for variants in ROR2 using multiple molecular approaches. We identified 25 putatively pathogenic ROR2 variants, 16 novel, including single nucleotide variants and exonic deletions. Detailed phenotypic analyses revealed that all subjects presented with a prominent forehead, hypertelorism, short nose, abnormality of the nasal tip, brachydactyly, mesomelic limb shortening, short stature, and genital hypoplasia in male patients. A total of 19 clinical features were present in more than 75% of the subjects, thus pointing to an overall uniformity of the phenotype. Disease-causing variants in ROR2, contribute to a clinically recognizable autosomal recessive trait phenotype with multiple skeletal defects. A comprehensive quantitative clinical evaluation of this cohort delineated the phenotypic spectrum of ROR2-related Robinow syndrome. The identification of exonic deletion variant alleles further supports the contention of a loss-of-function mechanism in the etiology of the syndrome.

Here, we synthesized exfoliated polymer clay nanocomposites (PCNs) by in situ polymerization of oil based vinyl macromonomer with styrene in the presence of clay. In order to facilitate the clay dispersion mixing was carried out by sonication besides classical mixing. The polymer clay nanocomposites were characterized by Fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), X-ray diffraction spectroscopy (XRD) and thermal gravimetric analysis (TGA). The samples prepared by mixing in sonication bath gave lower char yield compared to the samples of classical mixing.

Abstract

This study aimed to determine optimal extraction parameters by using response surface methodology, examining the effect of microencapsulation on crocin retention, and evaluating the temperature and pH stability of crocin microcapsules. The effect of process variables on the crocin yield was successfully determined by quadratic models (R-2>0.99 and adj R-2>0.98). All variables significantly affected the crocin yield (P0.05). The thermal and pH stability of the crocin was determined at different temperatures (60, 70, 80, and 90 degrees C) and pH levels (2, 4.5, and 6). Abstract

Context: Primary ovarian insufficiency (POI) encompasses a spectrum of premature menopause, including both primary and secondary amenorrhea. For 75% to 90% of individuals with hyper-gonadotropic hypogonadism presenting as POI, the molecular etiology is unknown. Common etiologies include chromosomal abnormalities, environmental factors, and congenital disorders affecting ovarian development and function, as well as syndromic and nonsyndromic single gene disorders suggesting POI represents a complex trait.Objective: To characterize the contribution of known disease genes to POI and identify molecular etiologies and biological underpinnings of POI.Design, Setting, and Participants: We applied exome sequencing (ES) and family-based genomics to 42 affected female individuals from 36 unrelated Turkish families, including 31 with reported parental consanguinity.Results: This analysis identified likely damaging, potentially contributing variants and molecular diagnoses in 16 families (44%), including 11 families with likely damaging variants in known genes and five families with predicted deleterious variants in disease genes (IGSF10, MND1, MRPS22, and SOHLH1) not previously associated with POI. Of the 16 families, 2 (13%) had evidence for potentially pathogenic variants at more than one locus. Absence of heterozygosity consistent with identity-by-descent mediated recessive disease burden contributes to molecular diagnosis in 15 of 16 (94%) families. GeneMatcher allowed identification of additional families from diverse genetic backgrounds.Conclusions: ES analysis of a POI cohort further characterized locus heterogeneity, reaffirmed the association of genes integral to meiotic recombination, demonstrated the likely contribution of genes involved in hypothalamic development, and documented multilocus pathogenic variation suggesting the potential for oligogenic inheritance contributing to the development of POI.

Abstract

This study was conducted to determine the occurrence, population growth, infestation and damage caused by Olive fly in olive orchards of Mardin province during 2016-2017. Pheremone traps, yellow sticky traps and beat sheet sampling were used to monitor population development and other parameters. It was observed that the first adults emerged at the end of July or beginning of August and the pest continued emerging until end of November. The pest occurred in 42.5% of the monitored orchards, with very low infestation (1.7-3.81%) and damage rate (2.4%-4.2%). The highest damage and infestation were recorded in Derik-1 area, while the lowest damage and infestation was noted in Dara orchard. The peak population development was observed during October in all orchards. Although population development varied in different years depending on temperature and humidity, Olive fly constitutes two peaks per year. The first peak appeared in September, while the latter in October. Overall, the pest had low population pressure in the monitored orchards in Mardin province. However, September and October are very critical for producers; therefore, management strategies should be opted against the pest during these months. Since olive fruits enter maturity phases during this period, these are susceptible to oviposition by the pest. Pest population could be significantly high, unless pest controlled during September and October. Yellow sticky traps combined with pheromone traps were 2-3 times more effective than Mcphail traps combined with 2% diammonium phosphate. Therefore, yellow sticky traps combined with pheromone traps will be beneficial against the pest as an effective biotechnical control strategy. [ABSTRACT FROM AUTHOR]

Abstract

The parasitoid species, Necremnus cosmopterix Ribeset Bernardo (Hymenoptera: Eulophidae) was first recorded in Turkey in 2014 as a larval parasitoid of tomato leaf-miner, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae). N. cosmopterix recovered from T. absoluta larvae in Southeast Anatolia Region, Diyarbakir province of Turkey. Morphological characters were discussed.

Abstract

Background and aim: Although the IgA anti-tissue transglutaminase test (IgA anti-tTG) has been recommended as the first step in the diagnosis of celiac disease (CD), there are controversial data about the real accuracy of the test in clinical practice. Therefore we evaluated the sensitivity and specificity of the IgA anti-tTG in a group of patients who were suspected of having CD. Material and methods: The study was performed at Van Training and Research Hospital, Van-Turkey. Details of patients in whom the IgA anti-tTG was requested from January 2009 to April 2012 were obtained from databases. Duplicate requests were excluded. Histopathologic examination of duodenal biopsies and serologic evaluations were compared. Results: A total of 1614 IgA anti-tTG were requested from different patients. In all, 49.6% of requests were in females and 29.8% from children under the age of sixteen. A total of 192 (11.9%) requests were found to be positive. Duodenal biopsies were performed to 61 (31.8%) of seropositive patients. The overall sensitivity and specificity of IgA anti-tTG were 93.3% and 9.5%. Conclusion: Our data have revealed that clinicians should be aware of solely relying on the results of the IgA anti-tTG test could result in unnecessary diagnostic procedures and treatments. Item does not contain fulltext, Robinow syndrome is a rare congenital disorder characterized by mesomelic limb shortening, genital hypoplasia, and distinctive facial features. Recent reports have identified, in individuals with dominant Robinow syndrome, a specific type of variant characterized by being uniformly located in the penultimate exon of DVL1 and resulting in a -1 frameshift allele with a premature termination codon that escapes nonsense-mediated decay. Here, we studied a cohort of individuals who had been clinically diagnosed with Robinow syndrome but who had not received a molecular diagnosis from variant studies of DVL1, WNT5A, and ROR2. Because of the uniform location of frameshift variants in DVL1-mediated Robinow syndrome and the functional redundancy of DVL1, DVL2, and DVL3, we elected to pursue direct Sanger sequencing of the penultimate exon of DVL1 and its paralogs DVL2 and DVL3 to search for potential disease-associated variants. Remarkably, targeted sequencing identified five unrelated individuals harboring heterozygous, de novo frameshift variants in DVL3, including two splice acceptor mutations and three 1 bp deletions. Similar to the variants observed in DVL1-mediated Robinow syndrome, all variants in DVL3 result in a -1 frameshift, indicating that these highly specific alterations might be a common cause of dominant Robinow syndrome. Here, we review the current knowledge of these peculiar variant alleles in DVL1- and DVL3-mediated Robinow syndrome and further elucidate the phenotypic features present in subjects with DVL1 and DVL3 frameshift mutations.

23. A statistical evaluation on spelling level of students in the primary education in Amasya

Abstract

This article has been prepared on spelling levels of students in the primary education in Amasya. In the study, the data obtained through the statistical study was used consisting of 450 students chosen randomly in the 6th, 7th and 8th classes of 15 primary schools in the city center of Amasya. These data have been presented in the article in detail in the form of tables and graphics. The observed spelling mistakes in the article are "using capital letters, lengthening, consonant harmony, writing conjunctions of 'ki' and 'de', adding question suffix and spelling of some special words". The obtained data indicated that the average success level of the students participated in the research is % 68.33. If we take into account importance of spelling rules by the point of their writings, we understand that these data mark an important problem: Spelling levels of the students in the second step of primary education in Amasya is lower than the expected level.

Abstract

Under this AFOSR GameChanger we carried out groundbreaking research by developing methods and materials to realize small and conformal structurally integrated antennas for unmanned airborne vehicles (UAVs). We are proud to report several groundbreaking developments in all areas. Among them we (a) developed a conformal, load-bearing antenna technology by embroidering e-fibers on polymer composites, (b) Characterized performance of the e-fibers in terms of core and specific strength for load-bearing applications, (c) Developed a new principles DFT method for systematic search of ferromagnetic, low conductivity nanowires, (d) Developed a strategy for increasing permittivity and permeability proportionately using magnetic nanowires combined with conventional high contrast materials for antenna miniaturization, (e) demonstrated theoretical feasibility of laminate EM materials that alter wave propagation characteristics, critical to future miniaturization efforts, (f) Developed a multi-time scaling model and algorithm for coupling mechanical and EM solutions in real time, critical to multiphysics coupled EM-Structural analysis, (g) Extended our new domain decomposition algorithm to 3-D space and characterized more complex/intricate details on large platforms such as e-fibers embroidered on UAV platforms. In addition to this report, our ground breaking work under this program has been published in over 120 journal papers and over 70 conference summaries., The original document contains color images.

Abstract

Surveys for determining alternative host plants of Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) in the Southeast Anatolia Region of Turkey were carried out in open fields of tomato growing areas between the years of 2011 and 2012. Plants of four different families including Solanacaeae, Amaranthaceae, Asteraceae and Poaceae have been identified as host plants. From cultivated members of the Solanaceae family; Solanum lycopersicum L. (tomato), Solanum melongena L. (aubergine), and Capsicum annuum L. (pepper), uncultivated Solanaceae; Solanum nigrum L, (black nightshade), S. woronowii Pojark. and Physalis angulata L. (cutleaf groundcherry), from the other non-cultivated weeds around tomato fields Amaranthus viridis L. (slender amaranth), belonging to the Amaranthaceae, Sorghum halepense L. (Pers.) (johnson grass) belonging to Poaceae and Xanthium strumarium L. (coclebur) belonging to Asteraceae family were detected as host plants of T. absoluta. Among them, S. woronowii, P. angulata, X. strumarium, A. viridis and S. halepense are the first records as hosts of T. absoluta in Turkey. [ABSTRACT FROM AUTHOR]

Abstract

Objective: Influenza viruses have retained their importance throughout history by creating pandemics since the segmental structure of the RNA undergo frequent genetic changes. The purpose of this study was to investigate the pandemic influenza A (H1N1) cases detected our region.Material and Method: Totally 570 patients were registered with pre-diagnosis of Pandemic Influenza A (H1N1) infection in five different hospitals located in Van region between 15 October 2009 - 15 January 2010. After nasopharyngeal/throat swab samples were taken from patients, they were sent to Refik Saydam Hygiene Center (RSHC) in accordance with the rules of biosafety, and specimens were investigated for Pandemic Influenza A(H1N1) 2009 by using Real-Time PCR method.Results: Pandemic Influenza A (H1N1) was determined by RSHC as positive for 220 patients. 36 of these patients were in intensive care conditions, 85 were hospitalized and 16 passed away. 109 patients, who were diagnosed with Pandemic Influenza A (H1N1), were within range of 6-25 years. All patients in this age range were treated and discharged. The most common symptoms of patients who were hospitalized were defined as cough and fever.
