The occupational environment may affect one’s psychophysical health by leveraging both external workplace stressors and individual psychological responses. We developed a comprehensive questionnaire to assess occupational stress among postgraduate medical trainees, investigating both situational and personal aspects. Exploratory factor analysis was used to identify the constructs captured by the questionnaire, and reliability was assessed by estimating Cronbach’s alpha. Construct-specific scores were computed, and their correlation with established pre-validated scales (criterion validation) was assessed. Four factors—“stress”, “coping”, “empathy”, and “trauma”—explained 50% of data variability and demonstrated satisfactory overall internal consistency (Cronbach’s alpha = 0.76). Significant correlations were found between the “stress” score and the “emotional exhaustion” component of the Maslach Burnout Inventory (MBI) (r = −0.76), the “coping” score and the “positive attitudes” component of the Coping Orientation to Problems Experienced Inventory (COPE) (r = 0.46), and the “empathy” score with the “empathic concern” (r = 0.52), “fantasy” (r = 0.41), and “perspective taking” (r = 0.45) components of the Interpersonal Reactivity Index (IRI). No significant differences in scores were found in terms of gender or medical specialization. This study suggests that the SCOPE questionnaire may be a promising tool for assessing workplace stress and psychological responses among medical residents.

The rapid technological development that geomatics has been experiencing in recent years is leading to increasing ease, productivity and reliability of three-dimensional surveys, with portable laser scanner systems based on Simultaneous Localization and Mapping (SLAM) technology, gradually replacing traditional techniques in certain applications. Although the performance of such systems in terms of point cloud accuracy and noise level has been deeply investigated in the literature, there are fewer works about the evaluation of their use for surface reconstruction, cartographic production, and as-built Building Information Model (BIM) creation. The objective of this study is to assess the suitability of SLAM devices for surface modeling in an urban/architectural environment. To this end, analyses are carried out on the datasets acquired by three commercial portable laser scanners in the context of a benchmark organized in 2023 by the Italian Society of Photogrammetry and Topography (SIFET). In addition to the conventional point cloud assessment, we propose a comparison between the reconstructed mesh and a ground-truth model, employing a model-to-model methodology. The outcomes are promising, with the average distance between models ranging from 0.2 to 1.4 cm. However, the surfaces modeled from the terrestrial laser scanning point cloud show a level of detail that is still unmatched by SLAM systems.

Nowadays, portable Mobile Mapping Systems (MMSs) and robotic mapping platforms leveraging on Simultaneous Localization and Mapping (SLAM) methods are gaining increasing attention for architectural and construction surveying, representing an efficient solution for geometric data acquisition for scan-to-BIM purposes. However, the applicability of standard modelling workflows and the accuracy of Building Information Models (BIM) that can be obtained from SLAM-based point clouds is still an open question. In this paper, we propose a preliminary evaluation on the feasibility of extracting as-built BIM from (i) a point cloud acquired with a commercial portable MMS, and (ii) a point cloud obtained through an open-source SLAM algorithm, surveying the environment with an autonomous mobile robotic platform. In both cases, the main structural elements of the test site are accurately generated, thus showing promising results. On the other hand, the experiment highlights also the need for SLAM systems capable of providing less noisy point clouds, in order to capture and model architectural details.

In recent years, portable Mobile Mapping Systems (MMSs) are emerging as valuable survey instruments for fast and efficient mapping of both internal and external environments. The aim of this work is to assess the performance of a commercial handheld MMS, Gexcel HERON Lite, in two different outdoor applications. The first is the mapping of a large building, which represents a standard use-case scenario of this technology. Through the second case study, that consists in the survey of a torrent reach, we investigate instead the applicability of the handheld MMS for natural environment monitoring, a field in which portable systems are not yet widely employed. Quantitative and qualitative assessment is presented, comparing the point clouds obtained from the HERON Lite system against reference models provided by traditional techniques (i.e., Terrestrial Laser Scanning and Photogrammetry).

High-Resolution Topography (HRT) data sets are becoming increasingly available, improving our ability and opportunities to monitor geomorphic changes through multi-temporal Digital Terrain Models (DTMs). The use of repeated topographic surveys enables inferring the sediment dynamics of hazardous geomorphic processes such as floods, debris flows, and landslides, and allows us to derive important information on the risks often associated with these processes. The topographic surveying platforms, georeferencing systems, and processing tools have seen important developments in the last two decades, in particular Light Detection And Ranging (LiDAR) technology used in Airborne Laser Scanning (ALS) and Terrestrial Laser Scanning (TLS). Moreover, HRT data, produced through these techniques, changed a lot in terms of point cloud density, accuracy and precision over time. Therefore, old “legacy” data sets and recent surveys can often show comparison problems, especially when multi-temporal data are not homogeneous in terms of quality and uncertainties. In this context, data co-registration should be used to guarantee the coherence among multi-temporal surveys, minimizing, on stable areas, the distance between corresponding points acquired at different epochs. Although several studies highlight that this process is fundamental to properly compare multi-temporal DTMs, it is often not addressed in LiDAR post-processing workflows. In this paper we focus on the alignment of multi-temporal surveys in a topographically complex and rugged environment as the Moscardo debris-flow catchment (Eastern Italian Alps), testing various co-registration methods to align multi-temporal ALS point clouds (i.e. years 2003, 2009 and 2013) and the derived DTMs. In particular, we tested the pairwise registration with manual correspondences, the Iterative Closest Point (ICP) algorithm and a mathematical model that allows aligning simultaneously a generic number of point clouds, the so-called Generalized Procrustes Analysis (GPA), also in its GPA-ICP variant. Then, to correct the possible small inaccuracies generated from the gridding interpolation process, a custom-developed DTM co-registration tool (GRD-CoReg) was used to align gridded data. Both alignment phases (i.e. at point cloud and DTM level) proved to be fundamental and allowed us to obtain proper and reliable DTMs of Difference (DoDs), useful to quantify the debris mobilized and to detect the spatial and temporal patterns of catchment-scale erosion and deposition. The consistency of DoDs data was verified through the comparison between the erosion estimate of DoDs and the volumes of debris-flow events measured by the monitoring station close to the Moscardo torrent catchment outlet. The GPA-ICP algorithm followed by the GRD-CoReg tool proved to be the most effective solution for improving DoDs results with a decrease of systematic trend due to vertical and horizontal uncertainties between surveys, especially at steep slopes. The net volume difference (i.e. the sediment output from the catchment) of the 2003–2013 period changed from 3,237,896 m3 to 135,902 m3 in DoDs obtained from not co-registered and co-registered DTMs. The volume of debris flows measured at the catchment outlet during the same time interval amounts to 169,660 m3. The comparison with debris-flow volume measures at the monitoring station shows, therefore, that the DTMs obtained from the co-registration processes generate more reliable DoDs than those obtained from the raw DTMs (without the alignment).

Early warning systems for debris flows are low cost measures for mitigating this kind of hazard. The early warning systems provide a timely alert for upcoming events in order to take protective measures, such as closing railways-roads, evacuating people from the threatened areas, and put rescue forces into readiness. These systems usually are sensor-based, and the alert time is the interval between the timing of the first detachment of debris flow by a sensor and its arrival into the threatened area. At the purpose of increasing the alert time, we propose an early warning system based on a model-cascade: nowcasting, hydrological- and triggering models. Nowcasting anticipates rainfall pattern that is transformed into runoff by the hydrological model. The triggering model estimates the volume of sediments that the runoff can entrain, and compares it with a critical threshold. If this is exceeded the alert is launched. The proposed early warning system is tested against the available data of the Rovina di Cancia (Northeast Italy) site.

Recent advances in novel immune strategies, particularly chimeric antigen receptor (CAR)-bearing T-cells, have shown limited efficacy against glioblastoma (GBM) in clinical trials. We currently have an incomplete understanding of how these emerging therapies integrate with the current standard of care, specifically radiation therapy (RT). Additionally, there is an insufficient number of preclinical studies monitoring these therapies with high spatiotemporal resolution. To address these limitations, we report the first longitudinal fluorescence-based intravital microscopy imaging of CAR T-cells within an orthotopic GBM preclinical model to illustrate the necessity of RT for complete therapeutic response. Additionally, we detail the first usage of murine-derived CAR T-cells targeting the disialoganglioside GD2 in an immunocompetent tumor model. Cell culture assays demonstrated substantial GD2 CAR T-cell-mediated killing of murine GBM cell lines SB28 and GL26 induced to overexpress GD2. Complete antitumor response in advanced syngeneic orthotopic models of GBM was achieved only when a single intravenous dose of GD2 CAR T-cells was following either sub-lethal whole-body irradiation or focal RT. Intravital microscopy imaging successfully visualized CAR T-cell homing and T-cell mediated apoptosis of tumor cells in real-time within the tumor stroma. Findings indicate that RT allows for rapid CAR T-cell extravasation from the vasculature and expansion within the tumor microenvironment, leading to a more robust and lasting immunologic response. These exciting results highlight potential opportunities to improve intravenous adoptive T-cell administration in the treatment of GBM through concurrent RT. Additionally, they emphasize the need for advancements in immunotherapeutic homing to and extravasation through the tumor microenvironment.

Structure-from-Motion (SfM) photogrammetry is increasingly employed in geomorphological applications for change detection, but repeatability and reproducibility of this methodology are still insufficiently documented. This work aims to evaluate the influence of different survey acquisition and processing conditions, including the camera used for image collection, the number of Ground Control Points (GCPs) employed during Bundle Adjustment, GCP coordinate precision and Unmanned Aerial Vehicle flight mode. The investigation was carried out over three fluvial study areas characterized by distinct morphology, performing multiple flights consecutively and assessing possible differences among the resulting 3D models. We evaluated both residuals on check points and discrepancies between dense point clouds. Analyzing these metrics, we noticed high repeatability (Root Mean Square of signed cloud-to-cloud distances less than 2.1 cm) for surveys carried out under the same conditions. By varying the camera used, instead, contrasting results were obtained that appear to depend on the study site characteristics. In particular, lower reproducibility was highlighted for the surveys involving an area characterized by flat topography and homogeneous texturing. Moreover, this study confirms the importance of the number of GCPs entering in the processing workflow, with different impact depending on the camera used for the survey.
