Abstract

The sex determination process in cucurbits involves the control of stamen or carpel development during the specification of male or female flowers from a bisexual floral meristem, a function coordinated by ethylene. A gain-of-function mutation in the miR164 -binding site of CpCUC2B , ortholog of the Arabidopsis transcription factor gene CUC2 , not only produced ectopic floral meristems and organs, but also suppressed the development of carpels and promoted the development of stamens. The cuc2b mutation induced the transcription of CpCUC2B in the apical shoots of plants after female flowering but repressed other CUC genes regulated by miR164 , suggesting a conserved functional redundancy of these genes in the development of squash flowers. The synergistic androecious phenotype of the double mutant between cuc2b and etr2b , an ethylene-insensitive mutation that enhances the production of male flowers, demonstrated that CpCUC2B arrests the development of carpels independently of ethylene and CpWIP1B. The transcriptional regulation of CpCUC1 , CpCUC2 , and ethylene genes in cuc2b and ethylene mutants also confirms this conclusion. However, the epistasis of cuc2b over aco1a , a mutation that suppresses stamen arrest in female flowers, and the down-regulation of CpACS27A in cuc2b female apical shoots, indicated that CpCUC2B promotes stamen development by suppressing the late ethylene production. Abstract

Treball Final de Grau en Psicologia. Codi: PS1048. Curs acadèmic: 2016/2017 El propósito de este trabajo es examinar la relación entre la práctica físico deportiva y los constructos de autoconcepto, bienestar psicológico y afrontamiento. Participaron 118 personas de distintas nacionalidades con un rango de edad comprendido entre 18 y 55 años. Los cuestionarios utilizados para recoger los datos de los constructos mencionados son el Cuestionario de Autoconcepto físico (CAF), la Escala de Bienestar Psicológico de Ryff (PWBS) y el Inventario de Estrategias de Afrontamiento (CSI). El estudio sigue un diseño transversal de tipo correlacional en el que se utiliza la encuesta para recoger los datos necesarios. Los análisis indican diferencias tanto en el género como en el tipo de ejercicio, siendo el bienestar psicológico la variable influida por el tipo de ejercicio y el estilo de afrontamiento por el género, el autoconcepto presenta influencia tanto del tipo de ejercicio como del género. Las relaciones encontradas entre los constructos indican una fuerte correlación entre autoconcepto y bienestar, bienestar y afrontamiento adecuado, junto con correlaciones negativas del autoconcepto, y el bienestar con el afrontamiento inadecuado. Estos resultados indican el papel protector del autoconcepto y el bienestar frente a los estilos inadecuados y el fomento de los estilos adecuados de forma directa por parte del bienestar y de forma indirecta por el autoconcepto. The aim of this study is to examine the relationships between experience in physical sports activity and various self-evaluations such as self-concept, psychological well-being and coping. Participants are 118 from different nationality, aged between 18 and 55 years. The instruments assess to evaluate all the variables are the Physical Self-Concept Questionnaire (CAF), the Psychological Well-Being Scales (PWBS) and the Coping Strategies Inventory (CSI). The study has a cross-sectional, correlational design, in which surveys are used to collect data. The statistical analysis indicate differences in gender and in the type of exercise, being psychological well-being the variable influenced by the type of exercise and coping by gender, self-concept is influenced by both the type of exercise and gender. Relationships found between the variables indicate a strong correlation between self-concept and well-being, well-being and suitable coping, along with negative correlations of self-concept and well-being with unsuitable coping. These results indicate a protective role of self-concept and well-being against unsuitable styles and the promotion of appropriate styles directly influenced by well-being and indirectly by self-concept.

Abstract

The bachelor thesis exposed here attempts to be part of the “Design for All” community, where age, gender, capabilities and culture do not matter. This project is guided by its design rules and methodology. The aim of the project is to develop a functional system which will allow students from San Rafael School to easily recognize themselves, as well as the room they are entering. The system is composed by an open source small motherboard which will be properly programmed to recognize every student going into a room inside the school. This task is carried out by scanning Low Energy Bluetooth beacons which are held by the students. The system then will launch a custom voice message welcoming them by their own name and the name of the room they are entering. The system will also recognize the rest of Bluetooth Low Energy devices around, but only the students will be recognized and get their custom voice message. To do that, the motherboard will be adapted with a small speaker and a modest power supply. El presente Trabajo Fin de Grado pretende formar parte de la comunidad “Diseño para Todos”, en la que edad, género, capacidades y cultura no importan. Este proyecto está guiado por sus doctrinas de diseño y metodología. El propósito de este proyecto es desarrollar un sistema funcional que permitirá a los alumnos del colegio San Rafael reconocer la estancia en la que entran, además de reconocerse a sí mismos. El Sistema está compuesto por una pequeña placa base de licencia de código abierto que será programada adecuadamente para reconocer a cada alumno que entre a una estancia dentro del colegio. Esta tarea será llevaba a cabo escaneando balizas emisoras de Bluetooth de baja energía, portadas por los alumnos. El sistema reproducirá un mensaje de voz personalizado dándoles la bienvenida con su propio nombre y la estancia a la que acceden. El sistema reconocerá de igual forma cualquier dispositivo Bluetooth de baja energía cercano, sin embargo, solo los estudiantes reconocidos obtendrán el mensaje de voz. 23. The transcription factor CpMYB62 controls the genetic network that leads to the determination of female flowers in Cucurbita pepo

Abstract

In monoecious species, female flowering constitutes the developmental process that determines the onset and production of fruit and is therefore closely related to crop yield. This article presents the identification and phenotypic and molecular characterization of myb62, an ethylmethane sulfonate loss-of-function mutation that completely blocks the female floral transition, converting all female flowers into male flowers. BSA-seq analysis coupled with WGS showed that myb62corresponds to a C>T transition in the coding region of the gene CpMYB62, generating a premature stop codon and a truncated transcription factor without its N-terminal effector domain. The myb62phenotype was partially rescued by exogenous ethylene application, indicating that the function of CpMYB62is mediated by ethylene. Different evidence supports this conclusion: first, the reduced ethylene production of the mutant, and second, the male flower productive phenotype of the double mutant between myb62and the ethylene-insensitive mutant etr2b, which demonstrated that myb62is epistatic over etr2b. Furthermore, transcriptomic analysis of WT and myb62apical shoots confirmed that CpMYB62regulates master sex-determining genes, upregulating those encoding the ethylene biosynthesis enzymes CpACO2Band CpACS27Aand those encoding for transcription factors that promote the development of carpels(CpCRC), but downregulating those involved in the arrest of carpels (CpWIP1), In the gene network controlling sex determination in cucurbits, CpMYB62 occupies the most upstream position, activating ethylene and other sex determining genes involved in female flower determination in Cucurbitapepo.

Abstract

Endophytic fungi are microorganisms that live inside plant tissues without causing disease symptoms. In most cases, their relationship with the host plant is considered beneficial because the endophyte is able to produce bioactive metabolites that the host can use as defense mechanisms, and this allows survival of both organisms. Recent studies have demonstrated that the endophytic fungus confers protection to the host plant against attack by pathogens and herbivores. Since they have the ability to benefit the host plant, it is reasonable to think that the diverse bioactive compounds produced by the endophyte might exhibit potential applications in agriculture and medicine, and also serve as a source of inspiration for the discovery of new and more active molecules. In this paper, some aspects regarding endophytic fungi are described: definition, discovery, classification, interactions with their host plants, ecological role, selected examples of bioactive metabolites, host selection strategy, and methodological strategies for their isolation. Finally, we show some recent results of our research, which demonstrate that endophytic fungi are promising sources of new bioactive secondary metabolites., Los hongos endófitos habitan en las plantas sin causar síntomas aparentes de enfermedad. La estrecha relación que existe entre el endófito y su planta hospedera se considera de gran importancia, ya que el hongo es capaz de producir metabolitos bioactivos, así como modificar los mecanismos de defensa de su hospedera, permitiendo e incrementando la sobrevivencia de ambos organismos. Estudios recientes demuestran la enorme capacidad que tienen los hongos endófitos para producir compuestos activos que le confieren protección a su hospedera contra el ataque de patógenos y herbívoros, constituyendo una nueva vía para la obtención de diversos precursores o moléculas novedosas de utilidad en la agricultura y en la medicina. En este trabajo se describen diferentes aspectos relacionados con los hongos endófitos, abarcando su definición, descubrimiento, clasificación, interacción con su hospedera, papel ecológico, ejemplos de metabolitos secundarios bioactivos, así como estrategias metodológicas para su obtención. Por último, se presentan algunos resultados recientes, de estudios realizados en nuestro grupo de trabajo mostrando que los hongos endófitos son una fuente potencial de metabolitos secundarios bioactivos.

Abstract

The worldwide cultivated Cucurbita pepo L. is one of the most diverse species in the plant kingdom. In this study, chilling tolerance over a wide range of cultivars was characterized to discover the allelic variants to improving the postharvest quality of the immature fruit during cold storage. For this purpose, fruits from 126 accessions of worldwide origin have been evaluated for weight loss and chilling injury after 3, 7 and 14 days of cold storage, classifying them into tolerant, partially tolerant, and sensitive accessions. To verify this classification, antioxidant capacity and lipid peroxidation (MDA) of contrasting accessions (tolerant vs. sensitive) were assessed. The antioxidant capacity significantly decreased during cold storage in the sensitive accessions, while it was maintained in tolerant accessions. Additionally, the sensitive accessions presented a higher accumulation of MDA during this period. Finally, a GWAS analysis using GBS data available in CuGenDBv2, combined with weight loss percentage data, led to the identification of a candidate QTL located on chromosome 17 that regulates postharvest cold tolerance in zucchini. The region contains four SNPs whose alternative alleles were significantly associated with lower weight loss percentage and chilling injury indices during cold storage. Two SNPs are located in the 3' UTR region of the gene CpERS1, a gene involved in ethylene perception. The other two SNPs generate missense mutations in the coding region of a Pectin methyl esterase inhibitor gene (CpPMI). The role of this QTL and these variants in chilling tolerance is discussed., (© 2024 The Author(s).
