The coronavirus disease 2019 (COVID-19), caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), turned into a global pandemic, and there remains an urgent demand for specific/targeted drugs for the disease. The 3C-like protease (3CLpro) is a promising target for developing anti-coronavirus drugs. Schisandra sphenanthera fruit is a well-known traditional Chinese medicine (TCM) with good antiviral activity. This study found that the ethanolic extract displayed a significant inhibitory effect against SARS-CoV-2 3CLpro. Forty-four compounds were identified in this extract using ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS). Combining molecular docking and in vitro experiments, we found that two epimeric 7,8-secolignans, rel-(1S,2R)-1-(3,4-dimethoxyphenyl)-2-methyl-3-oxobutyl-3,4-dimethoxybenzoate (2) and rel-(1S,2S)-1-(3,4-dimethoxyphenyl)-2-methyl-3-oxobutyl-3,4-dimethoxybenzoate (4), potently inhibited 3CLpro with IC50 values of 4.88 ± 0.60 μM and 4.75 ± 0.34 μM, respectively. Moreover, in vivo and in vitro experiments indicated that compounds 2 and 4 were potent in regulating the inflammatory response and preventing lung injury. Our findings indicate that compounds 2 and 4 may emerge as promising SARS-CoV-2 inhibitors via 3CLpro inhibition and anti-inflammatory mechanisms.

Non-alcoholic fatty liver disease (NAFLD), characterized by hepatic steatosis, is a common metabolic liver disease worldwide. Currently, satisfactory drugs for NAFLD treatment remain lacking. Obesity and diabetes are the leading causes of NAFLD, and compounds with anti-obesity and anti-diabetic activities are considered suitable candidates for treating NAFLD. In this study, biochemical and histological assays revealed that a natural lignan schisanhenol (SAL) effectively decreased lipid accumulation and improved hepatic steatosis in free fatty acid (FFA)-treated HepG2 cells and high-fat diet (HFD)-induced NAFLD mice. Further, molecular analyses, microRNA (miRNA)-seq, and bioinformatics analyses revealed that SAL may improve NAFLD by targeting the miR-802/adenosine monophosphate-activated protein kinase (AMPK) pathway. Liver-specific overexpression of miR-802 in NAFLD mice significantly impaired SAL-mediated liver protection and decreased the protein levels of phosphorylated (p)-AMPK and PRKAB1. Dual-luciferase assay analysis further confirmed that miR-802 inhibits hepatic AMPK expression by binding to the 3' untranslated region of mouse Prkab1 or human PRKAA1. Additionally, genetic silencing of PRKAA1 blocked SAL-induced AMPK pathway activation in FFA-treated HepG2 cells. The results demonstrate that SAL is an effective drug candidate for treating NAFLD through regulating miR-802/AMPK-mediated lipid metabolism.

This study explored the effect of raw material morphology on the properties of bamboo–plastic composites produced using hot-pressing. To provide a reference for reducing the production cost and improve the product properties of the composites, polylactic acid (PLA)-based bamboo–plastic composites were prepared using bamboo chips with a shaved morphology (BS) and fiber morphology (BF) and PLA as the matrix material, via hot-pressing. The properties of the bamboo–plastic composites formed with BS and BF chips were studied and compared with those of composites with conventional granular morphology (BM) and powder morphology (BP). The results showed that when the content of the bamboo chips was at 50% (the same below), the mechanical properties of the BF/PLA composites were remarkably better than those of the other PLA-based composites. However, the BF/PLA composites showed a high degree of hydrophilicity, with a water contact angle of 70.0° and a water absorption of 10.8% at 288 h. More holes could be seen in the BF/PLA composites using a scanning electron microscope. Among the four types of PLA-based composites, better melt fluidity was found only in the BF/PLA composites, and the melting index was 65.3 g/min.

This study extracted Bambusa blumeana fiber using alkaline boiling and then applied double-roll pressing in order to develop it, explore the applications of its various parts, and improve its utilization potential. The results showed that the outer bamboo fibers are finer and straighter than the inner and middle fibers, and the fracture mode of the bamboo fibers is brittle. From the inside to the outside, the tensile strength of the fiber bundle gradually increases from the top to the bottom. Moreover, the tensile strength of the outside bamboo fiber is twice that of the inside, reaching a maximum of 982 MPa. The surface of the interior bamboo fiber is relatively smooth and can be used in textile and decorative fields. Compared with internal and central bamboo fiber, the outer fiber has higher thermal stability and higher crystallinity, which makes it more advantageous in the process of strengthening composite materials. Studying the structures of fibers from different parts of Bambusa blumeana can provide substantial scientific support for the differential applications of bamboo fibers.

The melamine-formaldehyde (MF) resin adhesive was modified by graphene oxide (GO), the chemical structure, wettability, bonding performance, tensile properties, curing performance and thermal properties of the modified resin were analyzed, and the toughening mechanism was also discussed in this study. The results showed that: (1) The MF resin with a high molar ratio possessed stable methylene ether bonds, which could easily generate parallel folding in space to form a π-π stacking supramolecular self-assembly special structure, with the potential of enhancing the toughness of molecular structures. (2) GO contained a large number of oxygen-containing reactive functional groups, which could further lower the curing temperature of the MF resin. A dense cross-linked network structure improved the thermal stability of the resin. (3) The bonding strength and toughness of the resin were significantly improved when the content of GO was 0.1 wt%. However, due to the large specific surface area and the intense π-π interaction between sheets, GO was easy to agglomerate, and the properties of the resin with GO content of 0.4 wt% degraded sharply. (4) The crystallinity of the MF resin modified by GO decreased, and the surface energy and plastic deformation energy increased due to the increased fracture crack path and fracture surface of the resin, which was the macro-reason for the improvement of toughness. (5) The strong π-π interaction between GO sheets and π-π accumulation between triazine rings were like parallel “springs” in the molecular structure of the resin, which might be the internal reason for the improvement of toughness. In addition, it was also proved that this special structure could limit the activity of hydroxymethyl and the release of free formaldehyde in the resin.

Phenylpropenes, such as isoeugenol and eugenol, are produced as defend compounds, floral attractants, and flavor constituents by phenylpropene synthases belonging to the PIP reductase family. Moreover, isoeugenol is proposed to be involved in the biosynthesis of dibenzocyclooctadiene lignans, the main active compounds of Schisandra chinensis (Turcz.) Baill. fruits (SCF). S. chinensis, a woody vine plant, is widely used for its medicinal, horticultural, edible, and economic values. In this study, nine ScPIP genes were identified and characterized from the transcriptome datasets of SCF. The expression profiles revealed that ScPIP genes were differentially expressed during different developmental stages of SCF. Three ScPIPs were selected and cloned as candidate genes encoding phenylpropene synthases according to phylogenetic analysis. ScPIP1 was proved to function as isoeugenol synthase (IGS) and designated as ScIGS1 through in vivo functional characterization in Escherichia coli. Subcellular localization analysis demonstrated that ScIGS1 was localized in both the cytoplasm and nucleus. The three-dimensional (3D) model of ScIGS1 was obtained using homology modeling. Site-directed mutagenesis experiments revealed that the substitution of residues at positions 110 and 113 impacted the product specificity of ScIGS1 and the mutation of Lys157 to Ala abolishing catalytic function. Moreover, the kcat values of mutants were lower than that of ScIGS1 using a deep learning approach. In conclusion, this study provides a basis for further research on PIP reductases and the biosynthetic pathway of dibenzocyclooctadiene lignans.

Background: Schisandra chinensis fruit is a well-known traditional Chinese medicine (TCM), whose extract has a potent inhibitory effect on the severe acute respiratory syndrome-coronavirus-2 (SARS‑CoV‑2) 3-chymotrypsin-like protease (3CLpro) and papain-like protease (PLpro). Purpose: This work aims to find the active components from the fruit of S. chinensis against SARS‑CoV‑2 3CLpro and PLpro. Materials and methods: The chemical constituents of the fruit of S. chinensis were retrieved based on the electronic databases, such as Web of Science, PubMed, Medline Plus, and CNKI. Molecular docking was used to screen the active components against SARS‑CoV‑2 3CLpro and PLpro. Potential hit compounds were further evaluated by enzymatic activity assay. Furthermore, the anti-inflammatory activities of the active compounds were further explored using the phorbol-12-myristate-13-acetate (PMA)-induced THP1 cells model. Results: In this work, we retrieved 75 components of S. chinensis fruit, including 62 dibenzocyclooctadiene-type lignans, 3 diarylbutane-type lignans, 2 tetrahydrofuran-type lignans, and 8 nortriterpenoids. Combining molecular docking study and in vitro experiments, we found that pregomisin (63), meso‑dihydroguaiaretic acid (64), and nordihydroguaiaretic acid (65) could potently inhibit 3CLpro with IC50 values of 3.07 ± 0.38, 4.12 ± 0.38, and 6.06 ± 0.62 μM, respectively, and inhibit PLpro with IC50 values of 5.23 ± 0.33, 4.24 ± 0.46, and 16.28 ± 0.54 μM, respectively. Interestingly, compounds 63, 64, and 65 also have potent activities of regulating the inflammatory response in vitro. Conclusion: Our results suggest that compounds 63, 64, and 65 may be promising SARS-CoV-2 3CLpro and PLpro inhibitors and anti-inflammatory.

Developing evidence-based uses of herbal medicines and natural product-based drug discovery are two core aims of ethnopharmacology. This requires an understanding of the medicinal plants and the traditional medical knowledge associated with them which is a basis for cross-cultural comparison. The botanical drugs of traditional medical systems are still not understood well, even for well-known and widely respected traditions like Ayurveda. In this study, a quantitative ethnobotanical analysis was performed on the single botanical drugs included in the Ayurvedic Pharmacopoeia of India (API), presenting an overview on the medicinal plants of Ayurveda from perspectives of plant systematics and medical ethnobotany. Part-I of API includes 621 single botanical drugs, which are sourced from 393 species (323 genera in 115 families). Of these, 96 species yield two or more drugs, together accounting for 238 drugs. Taking the traditional concepts, biomedical uses and the pragmatic disease classification into account, therapeutic uses of these botanical drugs are sorted into 20 categories, which meet primary health demands. The therapeutic uses of the drugs sourced from the same species may differ considerably, but 30 of the 238 drugs are used in highly similar way. The comparative phylogenetic analysis identifies 172 species with high potential for specific therapeutic uses. This medical ethnobotanical assessment for the first time provides a comprehensive understanding on the single botanical drugs in API from the perspective of medical botany using an “etic” (scientist-oriented) approach. This study also highlights the importance of quantitative ethnobotanic methods in understanding traditional medical knowledge.

Abstract The present quality control method of Chinese medicinal materials (CMM) has obvious deficiency, which cannot be compatible with the multi-target and multi-component characteristics and production process of CMM. Plant metabolomics with a huge impetus to comprehensively characterize the metabolites and clarify the complexity and integrity of CMM, has been widely used in the research of CMM. This article comprehensively reviewed the application of plant metabolomics in the quality control of CMM. It introduced the concept, technique, and application examples, discussed the prospects, limitations, improvements of plant metabolomics. MS and NMR, as important techniques for plant metabolomics, are mainly highlighted in the case references. The purpose of this article is to clarify the advantage of plants metabolomics for promoting the optimization of the CMM quality control system and proposing a system approach to realize the overall quality control of CMM based on plant metabolomics combined with multidisciplinary method.

Abstract Background The stems of Kadsura interior A. C. Smith are used as traditional Chinese medicine (TCM) Kadsurae Caulis, with the traditional efficacy of tonifying and invigorating the blood, therefore being favored to treat blood deficiency (BD) widely. However, the stems of K. interior and its closely related species are morphologically similar and they may readily be misused as Kadsurae Caulis, thus likely to exert negative effects on clinical efficacy and clinical medication safety. Methods Firstly, blood tonic efficacies of the stems of K. interior (KIS) and its closely related species were compared using BD mouse model induced by 1-acetyl-2-phenylhydrazine (APH) and cyclophosphamide (CTX). Secondly, the chemical constituents from the stems of K. interior and its closely related species were evaluated and compared using a plant metabolomics approach. Plant metabolomics in this study aims at discovering differential metabolites and comprehensively assessing the chemical constituents by combining state-of-the-art high-resolution UPLC-Q/TOF–MS/MS technique and multivariate data analysis. Finally, based on the pharmacological data and the chemical constituents in UPLC-Q/TOF–MS fingerprints, the potential blood tonic active markers were screened by the spectrum-effect relationship analysis and quantified by UPLC-UV-DAD. Results The ethanol extract of the stems of K. interior significantly increased the levels of hematocrit (HCT), hemoglobin (HGB), and red blood cells (RBC) in BD mice. The Schisandra chinensis is an important edible plant, and previous phytochemical research focused on the S. chinensis fruit (SF) due to its long history as traditional Chinese medicine. Schisandra chinensis fruit was used as an astringent tonic to astringe the lungs and the kidneys, replenish energy, promote the production of body fluids, tonify the kidney, and induce sedation. The components of S. chinensis, such as its stems (SS), leaves (SL), and roots (SR), have drawn little attention regarding their metabolites and bioactivities. In this study, a strategy of combining a chemical database with the Progenesis QI informatics platform was applied to characterize the metabolites. A total of 332 compounds were tentatively identified, including lignans, triterpenoids, flavonoids, tannins, and other compound classes. Heatmap and principal component analysis (PCA) showed remarkable differences in different parts of the plants. By multiple orthogonal partial least-squares discriminant analyses (OPLS-DA), 76 compounds were identified as potential marker compounds that differentiate these different plant parts. Based on the variable influence on the projection score from OPLS-DA, the active substances including gomisin D, schisandrol B, schisantherin C, kadsuranin, and kadlongilactone F supported the fact that the biological activity of the roots was higher than that of the fruit. These substances can be used as marker compounds in the plant roots, which likely contribute to their antioxidant and anti-inflammatory activities. The plant roots could be a new medicinal source that exhibits better activity than that of traditional medicinal parts, which makes them worth exploring.

Objective: Schisandrae Sphenantherae Fructus (SSF) is a traditional Chinese medicine used to treat coughs and pulmonary inflammatory diseases. However, the pharmacodynamic material basis and mechanisms for SSF in asthma treatment remain unclear. This study aims to screen the anti-asthmatic fraction and verify the pharmacodynamic material basis, predict the potential mechanism, and verify the interaction ability between compounds and core targets.Methods: First, three fractions from SSF were compared in terms of composition, comparison, and anti-asthmatic effects. Then, the ultra-performance liquid chromatography-quadrupole/time-of-flight-mass spectrometry/mass spectrometry (UPLC-Q/TOF-MS/MS) strategy was used to identify the compounds from the active fraction, and the anti-asthmatic efficacy of the active fraction was further studied by the ovalbumin (OVA)-induced asthma murine model. Finally, network pharmacology and molecular methods were used to study the relationships between active compounds, core targets, and key pathways of PEF in asthma treatments.Results: The petroleum ether fraction (PEF) of SSF showed better effects and could significantly diminish lung inflammation and mitigate the level of serum immunoglobulin E (IgE), interleukin (IL)-4, IL-5, IL-6, IL-13, and IL-17 in mice. A total of 26 compounds from the PEF were identified, among which the main compounds are lignans and triterpenes. Moreover, 21 active compounds, 129 overlap-ping targets, and 10 pathways were screened by network pharmacology tools. The top five core targets may play a great role in asthma treatment. Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis suggested that the PEF can treat asthma by acting on multiple asthma pathological processes, including the IL-17 signaling pathway, T helper (Th) 17 cell differentiation, and the calcium signaling pathway. Molecular docking was performed to evaluate the interactions of the protein–ligand binding, and most docked complexes had a good binding ability.Conclusion: The present results might contribute to exploring the active compounds with anti-asthmatic activity.

Atherosclerosis is one of the main complications of diabetes mellitus, involving a variety of pathogenic factors. Endothelial dysfunction, inflammation, and oxidative stress are hallmarks of diabetes mellitus and atherosclerosis. Although the ability of diabetes to promote atherosclerosis has been demonstrated, a deeper understanding of the underlying biological mechanisms is critical to identifying new targets. NLRP3 plays an important role in both diabetes and atherosclerosis. While the diversity of its activation modes is one of the underlying causes of complex effects in the progression of diabetes and atherosclerosis, it also provides many new insights for targeted interventions in metabolic diseases.

Most plants of Kadsura have economic value and medicinal application. Among them, K. interior and its closely related species have been demonstrated to have definite efficacy. However, the taxonomy and phylogenetic relationship of Kadsura in terms of morphology and commonly used gene regions remain controversial, which adversely affects its rational application. In this study, a total of 107 individuals of K. interior, K. heteroclita, K. longipedunculata, K. oblongifolia, and K. coccinea were studied from the perspectives of genetic diversity, phylogeny, and ecology via single nucleotide polymorphisms (SNPs) developed through restriction site-associated DNA sequencing (RAD-seq). Based on these SNPs, the genetic diversity, phylogenetic reconstruction, and population genetic structure were analyzed. Subsequently, divergence time estimation and differentiation scenario simulation were performed. Meanwhile, according to the species distribution records and bioclimatic variables, the Last Glacial Maximum and current potential distributions of five species were constructed, and the main ecological factors affecting the distribution of different species were extracted. The FST calculated showed that there was a moderate degree of differentiation among K. heteroclita, K. longipedunculata, and K. oblongifolia, and there was a high degree of genetic differentiation between K. interior and the above species. The phylogenetic tree indicated that each of the species was monophyletic. The results of population genetic structure and divergence scenario simulation and D-statistics showed that there were admixture and gene flow among K. heteroclita, K. longipedunculata, and K. oblongifolia. The results of ecological niche modeling indicated that the distribution areas and the bioclimatic variables affecting the distribution of K. interior and its related species were different. This study explored the differences in the genetic divergence and geographical distribution patterns of K. interior and its related species, clarifying the uniqueness of K. interior compared to its relatives and providing a reference for their rational application in the future.

Myocardial ischemia/reperfusion (MI/R) injury is a serious threat to human health. Hydroxysafflor yellow A (HSYA), the main water-soluble ingredient extracted from Carthami flos (Carthamus tinctorius L.), has therapeutic potential for treating MI/R injury. However, the mechanisms of HSYA−mediated protection from MI/R injury are incompletely understood. In the present study, we investigated the effects and the underlying mechanisms of HSYA during MI/R. Adult Sprague-Dawley rats were subjected to left anterior descending artery ligation for 30 min followed by 24 h of reperfusion with or without HSYA treatment. The protective effect of HSYA was detected by 2,3,5-triphenyl tetrazolium chloride (TTC) staining, hematoxylin eosin (HE) staining, and myocardial enzymes detections. Serum levels of inflammatory factors such as TNF-α, interleukin (IL)-1β, and IL-18, were detected using ELISA kits. The expression of NLRP3 and other related proteins in the myocardium was detected by western blot and immunohistochemistry. The expression of autophagy-related proteins, including Atg5, BECN1, P62, and LC3B, was detected by western blot to evaluate the effect of HSYA on autophagy. Results showed that HSYA decreased the myocardial infarct size and attenuated the cardiac dysfunction in rats after I/R. In addition, HSYA inhibited myocardial apoptosis compared with the I/R group, decreased the levels of inflammatory cytokines in rat serum, reduced NLRP3 inflammasome expression, and induced autophagy. Mechanistically, our results demonstrated that HSYA can activate AMPK to improve autophagy and inhibit NLRP3 inflammasome by inhibiting the mTOR pathway. This work provides strong data supporting for the clinical applications of HSYA in MI/R injury.

Carthamus tinctorius L. (safflower), an economic crop and herb, has been extensively studied for its diverse chemical constituents and pharmacological effects, but the mechanism of safflower pigments (SP) leading to different colors of florets has not been clarified. In the present study, we compared the contents of SP in two varieties of safflower with white and red florets, named Xinhonghua No. 7 (WXHH) and Yunhong No. 2 (RYH). The results showed the contents of SP in RYH were higher than WXHH. To investigate genes related to SP, we obtained six cDNA libraries of florets from the two varieties by transcriptome sequencing. A total of 225,008 unigenes were assembled and 40 unigenes related to safflower pigment biosynthesis were annotated, including 7 unigenes of phenylalanine ammonia-lyase (PAL), 20 unigenes of 4-coumarate-CoA ligase (4CL), 1 unigene of trans-cinnamate 4-monooxygenase (C4H), 7 unigenes of chalcone synthase (CHS), 4 unigenes of chalcone isomerase (CHI), and 1 unigene of flavanone 3-hydroxylase (F3H). Based on expression levels we selected 16 differentially expressed unigenes (DEGs) and tested them using reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR), which was consistent with the sequencing results. Consequently, we speculated that in WXHH, 3 PALs, 3 4CLs, 1 C4H, 1 CHS, and 1 CHI, which were down-regulated, and 1 F3H, which was up-regulated, may play a key role in the formation of white florets.

The stems and roots of Kadsura species have been used as the folk medicine in Traditional Chinese medicine (TCM) and have good traditional efficacy and medicinal application with a long history. Among these species, K. coccinea, K. heteroclita and K. longipedunculata are the most widely distributed species in the regions of south and southwest China. Owing to their similar appearance, the crude drugs are often confusedly used by some folk doctors, even some pharmaceutical factories. To discriminate the crude drugs, haplotype analysis based on cpDNA markers and ITS was firstly employed in this study. Generic delimitation, interspecific interrelationships, and the identification of medicinal materials between K. longipedunculata and K. heteroclita remained unresolved by the existing molecular fragments. The original plant could be identified through the morphological character of flower, fruit and leaf. However, in most situation collectors have no chance to find out these characters due to lack of reproductive organs, and have no experience with the minor difference and transitional variation of leaf morphology. The chemical characterization show that the chemometric of chemical composition owned higher resolution to discriminate three herbs of Kadsura species. In conclusion, this integrative approach involving molecular phylogeny, morphology and chemical characterization could be applied for authentication of the Kadusra. Our study suggests the use of this comprehensive approach for accurate characterization of this closely related taxa as well as identifying the source plant and confused herbs of TCM.

Viscum coloratum (Viscaceae) is a kind of semiparasitic shrub used as medicinal plant. The complete chloroplast (cp) genome of V. coloratum was sequenced and characterized in this study. Length of total cp genome is 128,744 bp, which is constructed by four typical regions including a large single-copy region (LSC, 73,684 bp), a small single-copy region (SSC, 8630 bp) and a pair of inverted-repeat regions (IRs, 23,215 bp). 100 unique genes are predicted including 68 protein-coding genes, 28 tRNA genes and four rRNA genes. Phylogenetic analysis were performed based on 30 shared genes of 12 species in Santalales using maximum likelihood method. The results showed a close relationship between V. coloratum and V. album. The complete cp genome of V. coloratum would provide valuable genetic resources for further study on phylogeny, population genetics and identification in Viscum.

Fritillaria delavayi has been widely used as a traditional Chinese medicine (TCM) to treat respiratory diseases for thousands of years. In this study, the complete chloroplast genome of F. delavayi was assembled. The circular genome is 151,938 bp in size, which is comprised of one large single-copy (LSC) region of 81,757 bp and one small single-copy (SSC) region of 17,537 bp and separated by a pair of inverted repeat (IR) regions of 26,322 bp. A total of 112 unique genes (78 protein-coding, 30 tRNA, and 4 rRNA) are predicted and 19 of them are duplicated in IR regions. The overall GC content is 37.0% while the GC content of the LSC, SSC, and IR regions are 34.8, 30.5, and 42.5%, separately. Phylogenetic analysis indicated that F. delavayi was closely related to F. cirrhosa.

The antihyperlipidaemic effect and potential mechanisms of Schisandra chinensis fruit extract were investigated in hamsters fed with a high-fat diet. Treating with SF extract significantly decreased the serum levels of total cholesterol, triacylglycerol and low-density lipoprotein cholesterol, alleviated insulin resistance, attenuated inflammation and declined liver lipid accumulation. The expression and activities of key lipid-metabolic regulators were altered to the lipid-lowering side. Screening of different fractions of SF extract for lipid-lowering activities revealed that the lignan-rich fraction was the most effective. Eight abundant lignans were isolated from the major active fraction of SF extract, in which schisandrols A, B and schisandrins B, C showed significant lipid-lowering effects in HepG2 cells, and schisandrin C was potent in decreasing the blood lipids in Triton WR-1339 induced hyperlipidaemic mice. Overall, these results indicated that the extract and lignans in SF were effective regulators of lipogenesis and could be useful for treating hyperlipidaemia.

Phellodendri Amurensis Cortex (PAC) is a well-known herbal medicine in China with complex components, but the previous research has mostly focused on its alkaloids analysis. For the first time, a simpler and more efficient method was proposed in this paper to simultaneously determine the content of three different kinds of compounds—phenolic acids, alkaloids and limonoids—in PAC. The phenolic acids included 3-O-feruloylquinic acid, 4-O-feruloylquinic acid and syringin. The alkaloids include magnoflorine, phellodendrine, jatrorrhizine, palmatine and berberine, while the limonoids include obaculactone and obacunone. An approach combining multi-wavelength and HPLC-DAD was used in this study due to the great difference in maximum absorption wavelength of the various components. Four wavelengths at 215, 275, 280 and 310 nm, respectively, were chosen for monitoring. It has been indicated through appropriate tests that this approach is of high accuracy, good repeatability and stability and provides a scientific basis for the quality assessment of PAC and associated derivatives. In addition, the chromatographic fingerprints method combined with multivariate statistical analysis chosen in this study was proved to be effective and reasonable for an accurate classification of 33 batches of samples collected from different locations.

With both a full collection of native vascular plant distributions and a full checklist of source plants of the Chinese Materia Medica (CMM), the Uygur Medicine (UM), and the Kazak Medicine (KM) for the Xinjiang region, we defined medicinal plant: vascular plant ratios (simplified as medicinal plant ratios hereafter) as the value of medicinal plant richness divided by vascular plant richness. We aimed to find whether the ratios are constant or change in different environments, which environmental variables determine medicinal plant ratios, and whether the ratios are more influenced by human or by natural environments. Finally, suggestions for medicinal plant conservation were addressed. We found that (1) medicinal plant ratios were not constant, and they were high in the Tarim Basin which was largely covered by desert, while they were relatively low in mountainous areas, especially in the Tianshan Mountains where the general species richness was high; (2) medicinal plant ratios were not significantly influenced by human activities, indicated by human population density distributions, but they were highly correlated with plant species richness and climate, i.e. ratios decreased with plant species richness and MAP, and were related quadratically with MAT; (3) CMM ratio and UM ratio were more influenced by plant richness than by climate, while KM ratio was more influenced by climate. We concluded that the percentages of plants used as medicines were not influenced by distances from human settlements, but were determined by species richness or climate. We suggest that (1), in general, the medicinal plant ratio could be a complementary indicator for medicinal plant conservation planning and (2), for the region of Xinjiang, not only high diversity areas, but also some extreme environments should be considered as compensation for a better protection of medicinal plants.

Species pool hypothesis is broadly known and frequently tested in various regions and vegetation types. However it has not been tested in the arid Xinjiang region of China due to lack of data. Here with systematic data from references and field survey, we comprehensively examined species pool hypothesis in this region. Took species richness in 0.1° × 0.1° grid cells as regional species richness (RSR) which were obtained from the distribution maps of vascular plant species, and took species diversity of 190 and 103 plots in forest and grassland biomes across Xinjiang as local species richness (LSR), together with the digitalized soil pH and climate data, we tested the species pool hypothesis in this region. We found that: (1) the average RSR was higher in mountains than that in basins and it was negatively correlated with soil pH in mountains while positively correlated with soil pH in basins in Xinjiang; (2) RSR showed a positive correlation with mean annual precipitation (MAP) while showed a hump-shaped pattern with mean annual temperature (MAT); and the changing patterns of LSR were different for forest and grassland along the geographical and climate gradients; (3) LSR of forest was more affected by RSR than by climate, while on the contrary, LSR of grassland was more affected by climate than by RSR. Our results validated the species pool hypothesis in revealing that RSR had a significant role in shaping LSR patterns in addition to climate. We concluded that the relative effects of climate vs. RSR on LSR differed markedly between the forest and grassland communities across Xinjiang. Our results also showed that RSR revealed a contrasting relationship with soil pH in mountains and in basins, which might reflect differences in evolutionary processes of various habitats. In summary, our research systematically analyzed the correlation of species richness in regional and local scales in Xinjiang which provides more insights into the understanding of species pool hypothesis.

Melamine-dialdehyde starch resins used for wood surface finishes have been developed. The reaction of melamine with dialdehyde starch has been shown to occur by FTIR and MALDI ToF spectrometry, with several oligomer species due to the reaction of the two materials being identified, and the resin thermal stability was studied by thermogravimetric analysis. The resins were prepared by two different procedures when it was realized that dialdehyde starch is sensitive to too high a temperature for prolonged times. The melamine-dialdehyde starch resins were applied on particleboard supports as a direct liquid surface finish and a resin-impregnated paper. The surface finishes were tested for adhesion by the cross-cut test, their initial sessile drop contact angle, and the contact angle evolution as a function of time. The best results were obtained by the resins catalyzed by 2% ammonium sulfate and applied to the support surface as a resin-impregnated paper hot pressed for 3 min at 200°C, although the results at 180°C also looked promising. Screw connection is a type most commonly applied to timber structures. As important commercial tree species in China, Masson pine and Chinese fir have the potential to prepare wood structures. In this study, the effects of the diameter of the self-tapping screw and the guiding bores on the nail holding performance on different sections of Masson pine and Chinese fir dimension lumbers were mainly explored. The results showed that: (1) The nail holding strength of the tangential section was the maximum, followed by that of the radial section, and that of the cross section was the minimum. (2) The nail holding strength of Masson pine was higher than that of Chinese fir. (3) The nail holding strength grew with the increase in the diameter of self-tapping screws, but a large diameter would lead to plastic cracking of the timber, thus further affecting the nail holding strength. Masson pine and Chinese fir reached the maximum nail holding strength when the diameter of self-tapping screws was 3.5 mm. (4) Under a large diameter of screws, prefabricated guiding bores could mitigate timber cracking and improve its nail holding strength. (5) Prefabricated guiding bores were more necessary for the screw connection of Masson pine. The results obtained could provide a scientific basis for the screw connection design of Masson pine and Chinese fir timber structures. [ABSTRACT FROM AUTHOR]
